草鱼出血病是草鱼种的一种严重疾病,通常在水温25—30℃时发病,其症状表现为全身性组织器官充血。其病原悬液能通过赛氏滤器(Seitz,EKS石棉滤板),能在草鱼种体内及草鱼鳍单层细胞中传代繁殖,在草鱼肌肉单层细胞和性腺单层细胞中感染并出现细胞病变。病原悬液对脂溶剂乙醚敏感,耐热性强,对四环素类药物不敏感。试验结果,我们认为草鱼出血病的病原是病毒。  相似文献

稀有鮈鲫对草鱼出血病病毒敏感性的初步研究   总被引:5,自引:0,他引:5  
本文报道稀有鲫对草鱼出血病病毒(GCHV)的敏感性。GCHV是草鱼出血病的病原,用GCHV人工感好1-6月龄的稀有鲫,在水温22-32℃时能导致稀有鲫出现出血病症状。在水温28℃时,病鱼在ld内死亡,潜伏期为5d,发病高峰期在感染后第6-8d。GCHV能在稀有鲫体内传代,并诱导80%以上的稀行鲫患病死亡。将人工感染GCHV的稀有鲫病鱼组织超薄切片,电镜观察,发现在肠道、脾脏、肾脏等组织中存在大小与形态和GCHV相似的病毒颗粒。从稀有鲫出血病病鱼组织中纯化病毒,免疫电镜观察,发现病毒颗粒能被GCHV的特异性抗体聚集成团。由上可知,稀有鲫出血病是由GCHV感染所致,稀有鲫对GCHV是敏感的,可以作为草鱼抗出血病育种的模型。  相似文献

在病鱼肾组织中发现有病毒颗粒,在头肾组织中也看到类似的颗粒。病毒呈球形或六角形,直径为60—78毫微米,平均为69毫微米;颗粒中央有一电子密度较高的核心,其直径平均为32毫微米,核心周围包有外膜,宽20毫微米左右。此外,还有一种无外膜的、电子密度均匀的病毒颗粒,直径为46—60毫微米,平均为52毫微米,这种颗粒出现在细胞核和胞质包涵体内。所观察到的病毒颗粒均出现在肾的造血组织内,但红血球和颗粒白血球中没有发现。肾小管上皮细胞正常,也未观察到病毒颗粒。在正常鱼的肾组织中没有发现与病鱼标本中类似的病毒颗粒。因此可以认为,我们人工感染致病的草鱼肾组织中所观察到的病毒颗粒,是草鱼出血病的病原,暂名为草鱼疱疹病毒。  相似文献

用草鱼出血病病鱼分离出的草鱼出血病病毒(Grass carp hemorrhage virus,GCHV)感染其它常见鱼并用ELISA方法检查感染鱼组织提取液,结果表明:青鱼、鲢鱼、布氏鳌条对GCHV抗体呈阳性反应;鲤鱼、鳙鱼、鲫鱼、团头鲂、泥鳅则呈阴性反应。综合感染鱼发病症状及死亡特征,初步认为:青鱼对GCHV是易感的,GCHV能在鲢鱼、布氏蟹条体内增值,但毒力较低,鳙鱼、鲫鱼、团头鲂、鲤鱼、泥鳅能抗GCHV感染。  相似文献

草鱼出血病     
草鱼Ctenopharyngodon idella是我国重要的淡水鱼类养殖品种,但养殖过程极易发生出血病导致草鱼大量死亡,给草鱼养殖业带来巨大损失。我国在20世纪70年代开始进行病原的研究,1980年发现病毒颗粒并确认为草鱼出血病病原,1991年国际病毒分类委员会将其命名为草鱼呼肠孤病毒(Grass carp reovirus,GCRV)。一直以来除呼肠孤病毒感染草鱼引起出血病外,未见有病毒和细菌混合感染的报道。本刊2009年8期介绍了邓国  相似文献

草鱼出血病病毒感染鱼肾(Clk)单层细胞后,产生明显的细胞病变,其繁殖周期为2—3天。病毒增殖的上升期在感染后的12—48小时。28℃下,当感染复数(Multiplicityof Infection简称MOI)为0.05PFU/cell时,病毒滴价可达1.78×10~(11)PFU/ml,不同温度(25℃、28℃、30℃)下通气培养,测定病毒滴价略有差异。感染病毒的细胞培养液经ELISA检测和电子显微镜观察均为阳性结果,并且能感染健康草鱼。  相似文献

本文采用α-[~(32)p]ATP标记物在鱼肾细胞系(CIK)系统中,对草鱼出血病病毒(Grass carp hemorrhage virus,GCHV)基因组进行了体内转录的研究。通过放线菌素D抑制宿主细胞基因组的转录活动,从感染病毒细胞中分离出病毒的mRNA,分别采用液相杂交和Nortbern blot方法检查病毒mRNA的转录情况。试验结果表明,GCHV含有内源性转录酶,其基因组的转录活动是在病毒感染细胞后4小时开始,由早期基因所转录,8—10小时获得晚期基因的转录产物。这些mRNA的大小、数目大体上与病毒基因组一致。  相似文献

从患出血病的牙鲆 (Paralichthysolivaceus)的肝、脾、肾组织中分离到 3个菌株。经人工感染证实PHK致病性强 ,为病原菌 ,经鉴定为沙鱼弧菌 (Vibriocarchariae)。药敏试验结果显示 ,该菌株对红霉素、庆大霉素、先锋噻肟、菌必治、链霉素、氟嗪酸、多粘菌素B、头孢西丁、环丙沙星、头孢克肟、先锋必敏感 ;对萘啶酸、头孢噻吩、氯霉素、四环素、先锋Ⅴ、痢特灵、青霉素G、先锋孟多、氨苄青霉素、强力霉素等不敏感。  相似文献

Invasion of Edwardsiella ictaluri into cultured mammalian, fish and enzymatically harvested catfish enteric epithelial cells is described. Gentamicin survival assays were used to demonstrate the ability of this catfish pathogen to invade IEC-6 (origin: rat small intestinal epithelium), Henle 407 (origin: human embryonic intestinal epithelium), fathead minnow (FHM, minnow epithelial cells) and trypsin/pepsin-harvested channel catfish enteric epithelial cells. Invasion of all cell types occurred within 2 h of contact at 26 degrees C, in contrast to Escherichia coli DH5 alpha, which did not invade cells tested. Eight Edwardsiella ictaluri isolates from diseased catfish and the ATCC (American Type Culture Collection) strain were evaluated for invasion efficiency using FHM cells. All isolates were invasive, but at differing efficiencies. Invasion blocking assays using chemical blocking agents were performed on a single isolate (LA 89-9) using IEC-6 epithelial cells. Preincubation of IEC-6 cells with cytochalasin D (microfilament depolymerizer) and monodansylcadaverine (blocks receptor-mediated endocytosis) significantly reduced invasion by E. ictaluri, whereas exposure to colchicine (microtubule depolymerizer) had no effect on bacterial internalization. Results indicate that actin polymerization and receptor-mediated endocytosis are involved in uptake of E. ictaluri by IEC-6 epithelial cells. Invasion trials using freshly harvested cells from the intestine of the natural host, Ictalurus punctatus, show that invasion occurs, but at a low efficiency. This is possibly due to loss of outer membrane receptors during enzymatic cell harvest. This study provides the first documentation of the invasion of cultured mammalian and fish cells by E. ictaluri, and identifies possible mechanisms used for intracellular access. Additionally, the study describes several functional in vitro invasion models using commercially available cell lines as well as cells from the natural host (channel catfish, I. punctatus).  相似文献

倪氏复口吸虫是我国淡水鱼类的一种常见的危害严重的寄生虫。利用稀有鮈鲫作为寄主,研究了倪氏复口吸虫在其体内的移行途径,并观察了复口吸虫的感染是否在寄主耳石上留下标记轮。倪氏复口吸虫尾蚴从稀有鮈鲫体表的各个部位侵入,但主要集中在头部,侵入体内的尾蚴通过肌肉和结缔组织直接进入眼球。感染倪氏复口吸虫的稀有鮈鲫耳石表面微结构发生变化,部分鱼耳石上出现明显的标记轮。  相似文献

Osmotic fragilities and erythrocyte sedimentation rates (ESR) were measured in healthy, diseased, treated and recovered trout, Salmo gairdneri . Bacterial infection with Aeromonas and Streptococcus resulted in more fragile erythrocytes and raised ESR values. Treatment with antibacterial agents improved the values of both parameters. Possible reasons for such changes are discussed. Caution in the use of apparently healthy fish for the unqualified interpretation of haematological results is expressed.  相似文献

借助细胞培养和电镜技术,揭示了鳜球形病毒(Siniperca chuatsi spherical virus,SCSV)与鳜弹状病毒(Siniperca chuatsi rhabdovirus,SCRV)在草鱼鳍细胞(Grass carp fins,GCF)中共感染的现象。在筛选到敏感鱼类细胞系和建立了 鳜病毒体外增殖系统的基础上,取息典型病毒感染出血症的鳜组织,制备组织悬液,接种到GCF细胞中传代培养, 在攻毒后间隔不同时间收集细胞,对攻毒细胞的超薄切片进行电镜观察。揭示两种形态的鳜病毒可在同一个GCF 细胞中增殖,并描述和分析了病毒复制引起感染细胞的超微病变。本研究结果有助于阐明鱼类重要病毒病害的发 生过程及致病机理。  相似文献

ISG15 is one of the most strongly induced genes upon viral infection, interferon (IFN) stimulation, and lipopolysaccharide (LPS) stimulation, and only one copy has been found in mammals so far. Here two fish ISG15 genes, termed CaISG15-1 and CaISG15-2, have been cloned and sequenced from UV-inactivated GCHV (grass carp haemorrhagic virus)-infected and IFN-produced CAB cells (crucian carp Carassius auratus blastulae embryonic cells) by suppression subtractive hybridization. The full-length cDNA sequences of two crucian carp ISG15 encode a 155-amino-acid protein and a 161-amino-acid protein, both of which show 78.9% identity overall and possess the characteristic structures of mammalian ISG15 proteins including two tandem ubiquitin-like domains and the C-terminal canonical LRLRGG motif. In CAB cells treated with different stimuli including active virus, UV-inactivated GCHV and IFN containing supernatant (ICS), the expression of both CaISG15-1 and CaISG15-2 was upregulated but displayed different kinetics. Poly I:C and LPS were also able to induce an increase in mRNA for both genes. In CAB cells responsive to active GCHV, UV-inactivated GCHV, CAB ICS, Poly I:C and LPS, CaISG15-1 was upregulated more significantly than CaISG15-2. These results suggest that there are two ISG15 homologues in crucian carp, both of which might play distinct roles in innate immunity against viral and bacterial infection.  相似文献

利用稀有鲫个体小、性成熟时间短、在实验室控制条件下容易饲养等特点 ,将其作为一种毒性实验材料 ,从形态学和组织学水平对生活污水可能产生的毒性效应进行了初步研究。研究结果表明 ,在早期生命阶段 ,高浓度的生活污水对稀有鲫有急性毒性效应。受排污口污水暴露的稀有鲫鱼卵根本无法孵化 ;对处于性腺发育期稀有鲫的暴露则引起肝脏细胞水平上的毒性效应 ,且随污水浓度的增高 ,对鱼体性发育的阻碍效应也越强 ,而整个生命期即使仅暴露于低浓度的生活污水中 ,其个体发育和性发育也均受到影响 ,肝细胞也表现出强烈的毒性效应  相似文献

抗草鱼出血病病毒转基因稀有鮈鲫的初步研究   总被引:2,自引:1,他引:1  
研究采用草鱼H1基因启动子,以草鱼呼肠孤病毒(Grass carp reovirus,GCRV)外衣壳蛋白VP7基因为靶基因,以增强型绿色荧光蛋白(eGFP)为报告基因,构建了3个小发卡RNA(shRNA)表达载体pH1siGCRV(x)-CMVeGFP。CIK细胞感染实验表明,pH1siGCRV2-CMVeGFP具有较高的病毒抑制作用。通过显微注射将pH1siGCRV2-CMVeGFP导入稀有鮈鲫(Gobiocypris rarus)受精卵,获得转基因稀有鮈鲫P0代群体。转基因稀有鮈鲫攻毒实验显示,转基因稀有鮈鲫死亡率为30%,抗草鱼出血病能力显著提高。进一步的实时荧光定量PCR检测证实,转基因稀有鮈鲫脾脏、后肠和肝脏中GCRV的含量显著低于对照鱼,并随着时间的延续逐渐减少,转基因稀有鮈鲫体内GCRV的复制受到有效抑制。研究为抗草鱼出血病转基因鱼育种奠定重要基础。    相似文献

The pH, pCO2 and pO2 values for diseased fish did not differ significantly from those values for healthy fish, but fish treated with terramycin showed a highly significant rise in pCO2 with a corresponding fall in pO2. Haematocrit values, as well as haemoglobin and total plasma protein concentrations were significantly lower in diseased fish while treated fish showed a significant rise in total plasma protein. Glucose concentrations fell very significantly in diseased fish and showed a significant increase in treated fish.
The plasma electrolytes, sodium, potassium and chloride, were significantly reduced in diseased fish and potassium levels also fell in treated fish. Diseased fish showed very significantly reduced erythrocyte counts. Of the three parameters MCV, MCH and MCHC, only the values for MCH in diseased fish differed significantly from those for healthy fish.  相似文献

The association of DNA-containing viruses with diseases of marine and anadromous fish is reviewed. One section of the review describes those diseases with a proven viral etiology. Available information on the physical, chemical, and biological properties of the viruses is included. Another section deals with those diseases where a viral etiology is suspected but not established. The primary evidence associating viruses with many of these diseases is the observation of virus particles in electron micrographs of thin sections of tissue samples from diseased fish. Finally, the possible role of pollutants, and other stress factors, in predisposing fish to viral infection is discussed as are the problems associated with studying diseases of wild fish populations.  相似文献
