Ribulose bisphosphate carboxylase (EC 4.1.1.39 [EC] ) activity wasvery low in tomato leaf extracts unless prepared in the presenceof Mg2+, and polyclar AT. With young leaves, but not with fully-expanded leaves, the RuBP carboxylase activityextracted was increased by prolonged illumination of the leaves(2 h). The main effect of the light treatment was to increasethe specific activity of the enzyme but there was also a smallincrease in RuBP carboxylase protein. Tomato leaf RuBP carboxylasein extracts had specific activities in the range 0.2–0–6µmol CO2 min–1 mg–-1 total protein extracted,or 0.5–1.2 µmol CO2 min–1 mg–1 RuBPcarboxylase, and an apparent Km (CO2) at 20 ?C of 9.3 ? 1.2µM (using a of 6.407). The biphasic reaction course, fallover, of carboxyla-tion catalysedby ribulose 1,5-bisphosphate carboxylase/ox-ygenase (RuBisCO)has been known as a characteristic of the enzyme from higherland plants. Fallover consists of hysteresis in the reactionseen during the initial several minutes and a very slow suicideinhibition by inhibitors formed from the substrate ribulose-l,5-bisphosphate(RuBP). This study examined the relationship between occurrenceof fallover and non-catalytic RuBP-binding sites, and the putativehysteresis-inducible sites (Lys-21 and Lys-30S of the largesubunit in spinach RuBisCO) amongst RuBisCOs of a wide varietyof photosynthetic organisms. Fallover could be detected by followingthe course of the carboxylase reaction at 1 mM RuBP and thenon-catalytic binding sites by alleviation of fallover at 5mM RuBP. RuBisCO from Euglena gracilis showed the same linearreaction course at both RuBP concentrations, indicating an associationbetween an absence of fallover and an absence of the non-catalyticbinding sites. This was supported by the results of an equilibriumbinding assay for this enzyme with a transition state analogue.Green macroalgae and non-green algae contained the plant-type,fallover enzyme. RuBisCOs from Conjugatae, Closterium ehrenbergii,Gona-tozygon monotaenium and Netrium digitus, showed a muchsmaller decrease in activity at 1 mM RuBP than the spinach enzymeand the reaction courses of these enzymes at 5 mM RuBP werealmost linear. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. This study was done to explore an enzymatic mechanism for thephotosynthetic carbon reduction cycle whereby the rate of synthesisof ribulose 1,5-bisphosphate (RuBP) could be changed while thelevels of intermediates other than 3-phosphoglycerate and RuBPwere kept constant. Chloroplast aldolase was purified to homogeneityfrom spinach leaves. When the enzyme was assayed in the directionof fructose 1,6-bisphosphate synthesis in the presence of theconcentrations of the substrates reported in vivo, the activitywas severely inhibited by physiological concentrations of RuBP.The aldolase reaction proceeded with a sequential mechanism.The Km for dihydroxyacetone phosphate and D-glyceraldehyde 3-phosphatewere 0.45 mM and 40 µM, respectively. The activity wascompetitively inhibited by RuBP with respect to dihydroxyacetonephosphate. The KI was 0.78 mM. The maximum activity of aldolasein spinach leaves was calculated as 1,360µmol (mg Chl)–1h–1 An equation to express the reaction for the synthesisof fructose 1,6-bisphosphate by aldolase was constructed topredict the metabolic rate of this reaction in vivo. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. A stable freeze-dried powder was prepared of partly purifiedribulose bisphosphate carboxylase from wheat leaves. As withpreparations from other leaves it is necessary to incubate theenzyme with Mg2$ and CO2 to achieve maximum activity. At 25°C this activity was 0.75 IU mg–1 protein for a preparationactivated at 50 °C for 10 min; the Km for CO2 was 15 µM. The time for reactivation of enzyme that had been inactivatedthrough the absence of CO2 and Mg2$ was influenced by the lengthof the inactivating treatment. After a short inactivation periodthe enzyme was reactivated within a few minutes, whereas aftera longer period several hours were needed. Enzyme in the latterstate had some properties in common with enzyme inactivatedby lower temperatures but in the presence of CO2 and Mg2$. Theenzyme kinetic characteristics are similarly affected by bothkinds of inactivation; the maximum velocity is decreased butthe affinity for CO2 is not affected. Reactivation following a long inactivating treatment becomesmore dependent on Mg2$ concentration as the temperature is increasedfrom 0 to 20 °C.  相似文献

The effect of photosynthetic photon flux density (PPFD) on carboxylationefficiency, estimated as the initial slope (IS) of net CO2 assimilationrate versus intercellular CO2 partial pressure response curve,as well as on ribulose-1, 5-bisphosphate carboxylase (Rubisco)activation was measured in Trifolium subterraneum L. leavesunder field conditions. The relationship between IS and PPFDfits a logarithmic curve. Rubisco activation accounts for theIS increase only up to a PPFD of 550 µmol photons m-2s-1. Further IS increase, between 550 and 1000 µmol photonsm-2 s-1, could be related to a higher ribulose fcwphosphate(RuBP) availability. The slow, but sustained IS increase above1000 µmol photons m-2 s-1 could be explained by the mesophyllCO2 diffusion barriers associated with the high chlorophylland protein content in field developed leaves. The rate of photosynthesis by the freshwater alga Lemanea mamillosais proportional to CO2 concentration, virtually to the pointof saturation, and inversely proportional to the radius of thethallus. By contrast, the CO2 response curve of very thin slicesof the thallus is a rectangular hyperbola with a (lower) halfsaturation concentration of 10 mmol m–3. For the intactplant, the kinetics of CO2 fixation are strongly masked by internalCO2 transport limitations, although the maximum rate of photosynthesisis probably determined by the rate of supply of ribulose bisphosphate(RuBP). The flow of water over the alga becomes turbulent atwater velocities greater than about 90 mm s–1 and thethallus stretches significantly at higher water velocities.In its natural habitat, therefore, the external unstirred layerwill be thin (< 10 µm) and the thallus will be stretched,leading to rapid external and increased internal rates of CO2transport from the bulk solution. The estimated maximum rateof CO2 transport is commensurate with the maximum rate of photosynthesis(i.e. the rate of supply of RuBP). The Km(CO2) ancl Vmax of ribulose 1,5-bisphosphate (RuBP) carboxylaseand its protein ratio to total soluble protein from Oryza speciesincluding cultivars (25 varieties) and wild types (11 species,21 strains) were surveyed. Their variabilities among cultivarsof O. sativa were very small. The averages of the Km(CO2) andVmax values and the ratio of carboxylase to soluble protein,and their standard errors were 10.2?1.0µM, 1.72?0.13units.mg–1(pH 8.0 and 25?C) and 52?2%, respectively. However, some differencesseemed to exist based on genome constitution in the Oryza genus.RuBP carboxylases from the species with the AgAg genome, O.graberrima and O. breviligulate, exhibited low Km(CO2) values(8.0?0.8 µM). High Vmax was associated with the CC genome,O. eichingeri and O. officinalis (2.08?0.15 units.mg–1).A higher ratio of RuBP carboxylase protein to soluble proteinwas found for the AA genome, O. sativa and O. perennis. The mechanism of the regulation of the activation of ribulose1,5-bisphosphate carboxylase/ oxygenase (RuBisCO) by inorganicphosphate (Pi) in the presence of limiting concentrations ofCO2 was explored. The activation state of RuBisCO increasedsigmoidally following a biphasic kinetics against the concentrationof Pi in the activation mixture with an intermediary plateauat 2 to 3 mM Pi when the enzyme was activated for 30 min. Theintermediary plateau could not be seen when the preincubationtime was 10 min and the activation was completed at 10 mM Pi.RuBisCO from Euglena also showed a quite similar activationkinetics. The activation was not due to the contaminating CO2included in the stock Pi solution or in the activation buffercontaining the enzyme. The experiments with 2-carboxyarabinitol1,5-bisphosphate showed that the Pi stimulated activation wasdue to the promotion of binding of the activator CO2 to theactivation sites. It was also found that Pi increased the affinityof RuBisCO for the activator CO2 5.4-fold accompanied by a decreaseof the half-saturating concentration of CO2 to 1.6 µMat 20 mM MgCl2. The activity of the enzyme ribulose bisphosphate carboxylase(RuBPCase) was estimated after rapidly extracting it from intactwheat leaves pretreated under different light and CO2 levels.No HCO3 was added to the extraction buffer since it isshown to inhibit RuBPCase. The activity increased as light intensityor CO2 concentration during pretreatment was increased. Enzymeactivity increased as temperature during pretreatment was decreased.Light activation did not affect the affinity of RuBPCase forCO2. A Km of 30 µM CO2 under air level O2 was determined.CO2, light and temperature are three main limiting factors ofphotosynthesis. It seems that the activity of RuBPCase is regulatedby these factors according to the requirements for CO2 fixation.  相似文献

The time-dependent, slow inhibition of ribulose bisphosphatecarboxylase (RuBisCO) in the absence of ribulose bisphosphatewas dependent on the concentrations of RuBisCO and xylulosebisphosphate (XuBP). When incubated with excess XuBP, RuBisCOlost its activity gradually with incubation time. When RuBisCOof the concentration of 1.5 µM was incubated with 20 µMXuBP, the activity was inhibited for the initial 10 minutes,after which the activity recovered gradually with time. Therecovery was because XuBP in the incubation mixture was carboxy-latedto form 3-phosphoglycerate. The concentration of XuBP half-saturatingthe XuBP-carb-oxylation reaction of RuBisCO was 12 to 15 µM.The initial inhibition and the subsequent recovery of the activitywere due to the elimination from and re-binding to RuBisCO,respectively, of the activator CO2. An enzyme assay was developed to measure the initial and Mg2+–CO2activated forms of Ribulose 1,5-bisphosphate Carboxylase/Oxygenase(Rubisco) in rose leaves. The assay was verified by co-extractionof the leaflets with partially purified spinach Rubisco andthrough correlation with net photosynthetic rates of individualleaflets (r2=0.7324). Changes in activities were measured asa function of depth of leaves in the canopy for two cultivarsof greenhouse hybrid tea roses. Initial Rubisco activity declinedwith increasing canopy depth for both cultivars. The activatedform of the enzyme, however, remained constant with canopy depthfor cv. Red Success; but increased with canopy depth, then declinedafter mid-canopy in the cv. Royalty. Rubisco activities werealso measured in the cv. Red Success grown in CO2 enriched environments(100 mm3 dm–3) at three humidity levels. The activitieswere not significantly affected by humidity treatment. A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

Seeds of apple cv. Golden Delicious were germinated and cultivatedin the greenhouse until the third leaf emerged. Respirationofgerminating seeds or photosynthesis of the first leaves wasmeasured by infra-red gas analysis and porometry, respectively.To study the role of phosphoenolpyruvate carboxylase (PEPC),the dominant carboxylase in the carbon economy, its CO2 refixationpotentialwas related to the amount of CO2 lost in respiration. With arange of 0.2 (dry seeds) to 18 (cotyledons) µmol CO2 h–1g–1 PEPC activity resembled or exceeded the amount ofC02 lost in respiration before the third leaf developed. Itis concludedthat PEPC largely contributes to economize the carbonmetabolism of apple seedlings before they become photosyntheticallycompetent. Limitation of photosynthesis and light activation of ribulose,1,5-bisphosphate carboxylase (RuBPCO) were examined in the 5thleaf of seedlings of red clover (Trifolium pratense L. cv. Renova)for 5 d following an increase in photosynthetic photon fluxdensity (PPFD) from 200 to 550µmol quanta m–2 s–1.Net photosynthesis and its stimulation at 2.0 kPa O2 initialactivity of rapidly extracted RuBPCO, standard activity of RuBPCOafter incubation of the extracts in the presence of CO2, Mg2+,and inorganic phosphate and contents of soluble protein, starch,soluble sugars, and various photosynthetic metabolites weredetermined. Photosynthesis decreased and starch content increased.No decrease in photosynthesis was found if, when PPFD was increased,all leaves except the investigated 5th leaf were removed, suggestingthat the decrease in photosynthesis was due to accumulated carbohydrates.The stimulation of photosynthesis at 2.0 kPa O2 did not decreaseand the ratio of the total foliar steady-state contents of triosephosphate to 3-phosphoglycerate increased suggesting that thedecrease in photosynthesis was not due to limiting inorganicphosphate in chloroplasts. Intercellular CO2 partial pressureand RuBP content were not decreased. Nevertheless, the ratioof photosynthesis to initial RuBPCO activity decreased, suggestingthat the catalysis per active RuBPCO site was decreased. Theincrease in PPFD in the growth cabinet and the PPFD at whichleaves were preconditioned for 1 h, affected not only initialactivity but also the standard activity of RuBPCO. The resultssuggest that a varying proportion of RuBPCO was bound to membranesand was contained in the insoluble fraction of the extracts.A comparison of photosynthesis with extracted RuBPCO activitysuggested that membrane bound RuBPCO did not contribute to photosyntheticCO2 fixation and that the binding and release to and from membranesmodulated actual RuBPCO activity in vivo. Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site. Rates of photosynthesis of four submerged stream macrophyteswere examined under varying pH and composition of inorganiccarbon species. Callitriche stagnatis and Sparganium simplexused only CO2 for photosynthesis. Potamogeton crispus and P.pectinatus used HCO3 in addition to CO2, but with much lowerefficiency. The photosynthetic rates at air equilibrium anda total inorganic carbon concentration of 5.0 mM were 2–3times lower than maximum rates at CO2 saturation for the HCO3users and 10–14 times lower for the CO2 users. The CO2compensation point of entire plants of Callitriche (2.5 µM)and Sparganium (6.0µM) was well below the equilibriumconcentration (15 µM). and the low saturation points (250–500µM) also pointed to efficient use of CO2. Callitricheand Sparganium compete successfully with HCO3 users inhardwater streams, which have a higher exchange and generationcapacity of CO2 than stagnant and more soft waters. Rates ofphotosynthesis of Potamogeton crispus and P. pectinatus decreasedat high pH. Depending on the two alternative hypotheses forHCO3use, this decline can be explained by CO3––inhibition of HCO3 uptake or by increasing capacity tobuffer H+efflux from the plant. Habitats subject to high pH,e. g. small ponds with dense vegetation, may have a strong selectionfor efficient mechanisms of HCO3 use. Key words: Photosynthesis, Macrophytes, Carbon-source  相似文献
