There is an increasing need to develop field immobilization techniques that allow researchers to handle safely swift foxes (Vulpes velox) with minimal risk of stress or injury. We immobilized captive swift foxes to determine the safety and effectiveness of ketamine hydrochloride and xylazine hydrochloride at different dosages. We attempted to determine appropriate dosages to immobilize swift foxes for an adequate field-handling period based on three anesthesia intervals (induction period, immobilization period, and recovery period) and physiologic responses (rectal temperature, respiration rate, and heart rate). Between October 1998-July 1999, we conducted four trials, evaluating three different dosage ratios of ketamine and xylazine (2.27:1.2, 5.68:1.2, and 11.4:1.2 mg/kg ketamine:mg/kg xylazine, respectively), followed by a fourth trial with a higher dosage at the median ratio (11.4 mg/kg ketamine: 2.4 mg/kg xylazine). We found little difference in induction and recovery periods among trials 1-3, but immobilization time increased with increasing dosage (P < 0.08). Both the immobilization period and recovery period increased in trial 4 compared with trials 1-3 (P < or = 0.03). There was a high variation in responses of individual foxes across trials, making it difficult to identify an appropriate dosage for field handling. Heart rate and respiration rates were depressed but all physiologic measures remained within normal parameters established for domestic canids. We recommend a dosage ratio of 10 mg/kg ketamine to 1 mg/kg xylazine to immobilize swift foxes for field handling. 相似文献

long range dispersal is rare. At Gough Island, southern Atlantic Ocean, we observed a breeding adult male elephant seal during
September 2009, which had been tagged on its natal beach at Marion Island, southern Indian Ocean, in November 1998. The individual
was resighted only once on Marion Island, 6 months after tagging. This 3,860 km movement represents dispersal (and likely
gene flow) between distinct populations from different elephant seal geographical provinces. Given the polygynous breeding
system of this species, the presence of this single male may have a disproportionate genetic effect on the small number of

Immobilization and anaesthesia of adult male southern elephant seals (Mirounga leonina) is potentially risky for animals and scientists. A tiletamine/zolazepam injection is considered the most appropriate drug combination for field application in this species. Since appropriate dosages are difficult to assess due to uncertainties in weight estimation, we used photogrammetry-derived weight estimates to ensure precise post hoc calculations of dosages. We report on 15 intramuscular tiletamine/zolazepam immobilizations of post-moult males of the upper weight class at King George Island/Isla 25 de Mayo, in April 2010. Initial injections were made using blowpipe syringes. Mean tiletamine/zolazepam combined dosages of 0.71 mg kg?1 (SD ± 0.16) ranged between 0.46 and 1.01 mg kg?1. In four cases, ketamine was added in dosages between 0.96 and 2.61 mg kg?1. Mean induction period was 23 min (± 15), and the mean duration of the procedures from first injection to release of the animals required 96 min (± 51). Four seals exhibited periods of apnoea, and one case of an extended, repetitive, and potentially critical apnoea (> 25 and 8 min) required intervention in order to successfully re-initiate spontaneous respiration. All procedures resulted in proper immobilizations allowing for the deployment of the satellite tags on the seals’ heads. The fact that even substantial deviations between the initial weight estimates and the photogrammetry-derived weight estimates had no apparent effect on the course of the immobilization underlines the drugs’ wide safety margin in this species. 相似文献

We compare genetic (both nuclear and mitochondrial) and morphometric measures between two putative populations of southern elephant seal (Mirounga leonina), and interpret the results in the context of data from mark-recapture and satellite-telemetric studies. One population is on the Argentine mainland, while the other is 2,400 km away on South Georgia island. We found pronounced differentiation at the mitochondrial DNA (mtDNA) control region that was distinct from the pattern of variation seen among island rookeries. Some morphometric characters and seven out of ten nuclear-DNA markers also showed differentiation between the island and mainland sites. Diversity at nuclear markers was high in both populations but mtDNA diversity was low in the mainland population, suggesting a founder event and little subsequent immigration of females. Morphological differences may suggest different selective environments at the two sites. 相似文献

Genetic diversity provides the raw material for populations to respond to changing environmental conditions. The evolution of diversity within populations is based on the accumulation of mutations and their retention or loss through selection and genetic drift, while migration can also introduce new variation. However, the extent to which population growth and sustained large population size can lead to rapid and significant increases in diversity has not been widely investigated. Here, we assess this empirically by applying approximate Bayesian computation to a novel ancient DNA dataset that spans the life of a southern elephant seal (Mirounga leonina) population, from initial founding approximately 7000 years ago to eventual extinction within the past millennium. We find that rapid population growth and sustained large population size can explain substantial increases in population genetic diversity over a period of several hundred generations, subsequently lost when the population went to extinction. Results suggest that the impact of diversity introduced through migration was relatively minor. We thus demonstrate, by examining genetic diversity across the life of a population, that environmental change could generate the raw material for adaptive evolution over a very short evolutionary time scale through rapid establishment of a large, stable population. 相似文献

The ability of air-breathing marine predators to forage successfully depends on their ability to remain submerged. This is in turn related to their total O(2) stores and the rate at which these stores are used up while submerged. Body size was positively related to dive duration in a sample of 34 adult female southern elephant seals from Macquarie Island. However, there was no relationship between body size and dive depth. This indicates that smaller seals, with smaller total O(2) stores, make shorter dives than larger individuals but operate at similar depths, resulting in less time being spent at depth. Nine adult female elephant seals were also equipped with velocity time depth recorders. In eight of these seals, a plot of swimming speed against dive duration revealed a cloud of points with a clear upper boundary. This boundary could be described using regression analysis and gave a significant negative relationship in most cases. These results indicate that metabolic rate varies with activity levels, as indicated by swimming speed, and that there are quantifiable limits to the distance that a seal can travel on a dive of a given swimming speed. However, the seals rarely dive to these physiological limits, and the majority of their dives are well within their aerobic capacity. The southern elephant seal population at Macquarie Island has undergone a serious decline since regular surveys were commenced in 1949. Approximately 2,900 cows and 250 bulls were counted in the isthmus study area during the 1985 breeding season. Comparisons with 20 other counts made in the same area between 1949 and 1984 showed that, although there was considerable fluctuation between the years, the number of seals ashore has dropped at an average rate of 2.1% per year, resulting in a net decrease of approximately 50% for both males and females. This is similar to the ratio of decline of elephant seal populations in the southern Indian Ocean. The census information from all major elephant seal populations was reviewed and it was concluded that there may be a common factor, or group of factors, acting to reduce the Macquarie Island and Indian Ocean populations, while the populations of the South Atlantic seem to be stable. Several potential explanations for these observations are advanced, but it is concluded that a greater knowledge of the ecology of the seals is needed if the declines are to be understood. 相似文献

Chromosome analyses were carried out on bone marrow cells from three elephant seal pups (Mirounga leonina), a species characterized by a 2n = 34 chromosome number. Arm ratios for the different chromosomes were calculated to facilitate the classification of chromosomes into groups. The karyotype of the southern elephant seal differs from other reported 34-chromosome phocids in the morphology of the Y chromosome and in that of one of the autosomal chromosome pairs. 相似文献

We examined the cortisol responses to chemical and physical restraint stress in southern elephant seal Mirounga leonina females and their pups at three stages during lactation. In anaesthetised females the serum cortisol levels changed moderately during the 45-min sampling period following restraint, with average peaks at 23 min after anaesthetic administration. Overall, cortisol was relatively low 2 days postpartum and increased throughout lactation. In physically restrained pups serum cortisol increased rapidly after capture; the response was milder at age 2 days than at 11 days and 21 days. Levels were higher in female pups than in males. In order to test whether cortisol levels and/or responses became chronically (i.e. days to weeks) altered due to restraint, we compared the cortisol response at a late stage of lactation between three groups of mother-pup pairs previously given different levels of chemical (mothers) or physical (pups) restraint stress: control (not handled previously), moderate treatment (previously handled twice), and high treatment (previously handled 3-4 times). Pups of the three treatment groups showed similar adrenocortical responses suggesting no chronic effect of repeated physical restraint, despite the clear acute effects. Mothers of the control and moderate treatment groups showed similar cortisol responses; however, mothers of the high treatment group showed significantly attenuated responses. This indicated that elephant seals tolerated moderate degrees of handling disturbance; however, repeated (3-4) chemical immobilisations in lactating females may reduce their adrenocortical responsiveness for a period of days or weeks. 相似文献

Summary A time-depth-temperature recorder provided a continuous record of diving by a female southern elephant seal in relation to water temperature for 27 days (1939 dives) after completion of moult. Mean maximum dive depth was 391±2.6 m and the overall maximum was 775 m. Dives lasted on average 17.5±0.09 min. Most dives showed a rapid descent to the discontinuity between the cold surface water and warmer deep water. Consequently the seal spent 57% of its time while diving at a depth of 200–400 m when it may have been foraging. This strongly suggests that the seal was exploiting a food source at the discontinuity between vertically stratified water masses. The water temperature data also indicated that the seal was diving in waters south of the Antarctic Polar Front and at some distance from the northern edge of the pack ice. The seal spent 88% of its time under water. Normal surface intervals between dives lasted an average of 2.1 ± 0.1 min whereas 16 extended surface intervals (>10 min duration) lasted 32.7±4.6 min. Dives were deeper during the day than at night and all but one extended surface interval occurred at night. The pattern of dives was similar to records from northern elephant seals but this is the first study to show how diving behaviour relates to water temperature. 相似文献

Serum clinical chemistry parameters were examined in lactating southern elephant seal Mirounga leonina mothers and their pups from the declining Macquarie Island population. There were significant changes in serum values from 2 to 21 days postpartum in both nursing mothers (increase: inorganic phosphate; decrease: creatinine, potassium, chloride, cholesterol, total protein, albumin, globulin, aspartate aminotransferase, creatine kinase) and suckling pups (increase: inorganic phosphate, globulin, cholesterol; decrease: albumin, alkaline phosphatase, gammaglutamyl transferase; increase followed by decrease: triglycerides, iron). We found no evidence that changes were due to chronic stress effects caused by repeated chemical immobilisations (mothers) or physical restraint (pups): at late lactation, clinical chemistry values were similar for mother-pup pairs of a control group (not handled previously), moderate treatment group (previously handled twice) and high treatment group (previously handled three to four times). We were not able to detect differences in clinical chemistry values between mother-pup pairs distributed over two areas differing in the frequency of human visits. The clinical chemistry values presented here can serve as reference ranges to allow future comparison with other southern elephant seal populations to investigate factors, e.g. food limitation, suspected to be involved in population declines. 相似文献

Genes of the major histocompatibility complex (MHC) are highly polymorphic in most terrestrial mammal populations so far studied. Exceptions to this are typically populations that lack genome-wide diversity. Here I show that two populations of the southern elephant seal (Mirounga leonina) have low DNA restriction fragment length polymorphism at MHC loci when compared with terrestrial mammals. Limited studies on MHC polymorphism in two cetacean species suggest this is a feature of marine mammal populations in general. MHC polymorphism is thought to be maintained by balancing selection, and several types of disease-based and reproductive-based mechanisms have been proposed. For the three marine mammal species examined, the low MHC polymorphism cannot be explained by low genome-wide diversity, or by any reproductive-based selection pressure. It can, however, be explained by diminished exposure to pathogenic selection pressure compared with terrestrial mammals. Reduced exposure to pathogens would also mean that marine mammal populations may be susceptible to occasional pathogen-induced mass mortalities. 相似文献
