The androgenic glands (AG) of male decapod crustaceans produce insulin-like androgenic gland (IAG) hormone that controls male sex differentiation, growth and behavior. Functions of the AG are inhibited by gonad-inhibiting hormone originating from X-organ-sinus gland complex in the eyestalk. The AG, and its interaction with the eyestalk, had not been studied in the blue swimmer crab, Portunus pelagicus, so we investigated the AG structure, and then changes of the AG and IAG-producing cells following eyestalk ablation. The AG of P. pelagicus is a small endrocrine organ ensheathed in a connective tissue and attached to the distal part of spermatic duct and ejaculatory bulb. The gland is composed of several lobules, each containing two major cell types. Type I cells are located near the periphery of each lobule, and distinguished as small globular cells of 5-7 μm in diameter, with nuclei containing mostly heterochromatin. Type II cells are 13-15 μm in diameter, with nuclei containing mostly euchromatin and prominent nucleoli. Both cell types were immunoreactive with anti-IAG. Following bilateral eyestalk ablation, the AG underwent hypertrophy, and at day 8 had increased approximately 3-fold in size. The percentage of type I cells had increased more than twice compared with controls, while type II cells showed a corresponding decrease.  相似文献

Analogy to Insecta suggests that JH(s) may play important roles in the development and reproduction of Crustacea. It has recently been shown by Laufer et al. (1987), using LC and GC/MS that the mandibular organ of adult crabs synthesized methyl farnesoate, the unepoxidated from of JH III, and that synthesis varied according to the sex, as a function of eyestalk ablation, and stage of vitellogenesis. In experiments reported here, we found that eyestalk removal from juvenile spider crabs, Libinia emarginata, resulted in a two-fold increase in the rate of MF synthesis by MOs in vitro. Furthermore, 2 h incubations of MOs from eyestalk ablated animals with eyestalk extracts inhibited the rate of synthesis of MF by about 60%. These results suggest that MF may be a gonad stimulating hormone (GSH) reported by other workers. Secretion by the MO is inhibited by an eyestalk factor (MO-IH), and MO-IH may be similar or identical to the gonad inhibiting hormone (GIH) reported by others.  相似文献

  • 2.2. In constant salinity, crabs showed a persistent respiratory rhythm, with a maximum oxygen consumption during the day. Under these conditions, ablation significantly increased the respiratory rate but not the rhythm.
  • 3.3. In variable salinities, the highest respiratory rates occurred in salinities of 11 and 16%. during the night. In these crabs, ablation of eyestalks and subsequent injection of eyestalk extracts did not alter the respiration rate rhythm.

To investigate the mechanism of sex determination of crabs, amplified fragment-length polymorphism DNA analysis was applied to identify sex-specific markers for Portunus trituberculatus. Eleven pairs of primers were used to amplify DNA isolated from male and female crabs. A total of 481 and 499 fragments were amplified for females and males, respectively, and the ratios of polymorphic fragments were 20.59–65.52% and 27.27–62.22%, respectively. Distribution difference values between female and male groups for 10 polymorphisms were greater than 40%; Cluster analysis revealed that Nei's genetic distances for 57 polymorphisms clustered according to sex. The results indicate that a difference at the DNA level exists between female and male crabs and provide data for further study on sex determination.  相似文献

Heart rate and arterial haemolymph flow rates were measured in freshly trapped Dungeness crabs, Cancer magister, using a pulsed-Doppler flowmeter. In the laboratory, freshly collected subtidal crabs exhibited endogenous rhythms in both cardiac function and haemolymph flow through one or more arterial systems, of both tidal and diurnal periodicity. The strongest tidal rhythms were recorded in the sternal and paired anterolateral arteries. These endogenous rhythms of selective tissue perfusion are related to an underlying locomotor activity, but may also be involved with hormonal transport or feeding. Changes in both heart rate and stroke volume were responsible for the increases in haemolymph flow rates. These rhythms were not entrained by aerial exposure, since confinement of arhythmic crabs in intertidal cages did not re-entrain an endogenous tidal rhythm. Endogenous locomotory rhythms are known to be controlled by neurohormones released in cycles from the sinus gland on the eyestalk. These hormones may also control the endogenous cardiovascular rhythms, since these were abolished after eyestalk ablation in freshly collected Cancer magister. These results suggest that hormones synthesized and released by the X-organ/sinus gland complex may, together with pericardial hormones, play a role in modulation of crustacean cardiovascular function.  相似文献

Testicular cell culture of crab, Scylla serrata (Forskal) was used to study the effects of White spot syndrome virus (WSSV). We are showing the susceptibility of cell culture of crabs to WSSV. The proliferating cell culture of testes were maintained for more than 4 months in a medium prepared from L15 and crab saline supplemented with epidermal growth factor. The cell cultures inoculated with different concentrations of virus showed distinct cytopathic effects such as change in cell appearance, shrinkage and cell lysis. WSSV infection of cultured cells was confirmed by Nested PCR technique. The incorporation of viral DNA in cultured cells was shown by RAPD profile generated using 10-mer primers. The controls that were not exposed to WSSV did not show cytopathic effects. This work shows the usefulness of proliferating testicular cell culture for studying WSSV infection using molecular tools. Thus, this report gains significance as it opens new vistas for diagnostics and drugs for WSSV.  相似文献

A disease caused by a parasitic dinoflagellate of the genus Hematodinium was identified in red, Paralithodes camtschaticus, and blue, Paralithodes platypus, king crabs from the north-east region of the Sea of Okhotsk, Russia, during annual stock surveys. No carapace color change was observed even in heavily infected crabs, but diseased crabs possessed creamy-yellow hemolymph, which was visible through the arthrodial membranes of the abdomen and appendages. Several stages of the parasite’s life history, including trophonts, plasmodia, sporonts and macrodinospores, were observed in tissues of infected king crabs. Numerous parasite cells were observed in the lumina of the myocardium, the gills, the connective tissue of antennal glands and the sinuses of nerve ganglia, eyestalks and gastrointestinal tract of king crabs with gross signs of infection. Based on sequencing of the 18S rDNA, it appears that the Hematodinium sp. found in red and blue king crabs is identical or closely related to Hematodinium sp. isolated from crabs of the genera Chionoecetes and Lithodes. Observed prevalences were 0.33% in sublegal male red king crabs, 0.18% in female red king crabs, 0.34% in sublegal male blue king crabs and 0.31% in female blue king crabs.  相似文献

The hormonal control of oogenesis has been investigated in the freshwater prawn, Macrobrachium kistnensis. Experiments using bilateral eyestalk ablation and injection of eyestalk extract supported the presence of a gonad inhibiting factor in the eyestalk of female prawns. Injections of brain and thoracic ganglion extract in normal female prawns and in those subjected to bilateral eyestalk ablation revealed the presence of a gonad stimulating factor in the central nervous tissue.  相似文献

The hypothesis is tested that methionine-enkephalin, a hormone produced in and released from eyestalk of crustaceans, produces hyperglycemia indirectly by stimulating the release of hyperglycemic hormone from the eyestalks. Injection of methionine-enkephalin leads to hyperglycemia and hyperglucosemia in the estuarine crab Scylla serrata in a dose-dependent manner. Decreases in total carbohydrate (TCHO) and glycogen levels of hepatopancreas and muscle with an increase in phosphorylase activity were also observed in intact crabs after methionine-enkephalin injection. Eyestalk ablation depressed hemolymph glucose (19%) and TCHO levels (22%), with an elevation of levels of TCHO and glycogen of hepatopancreas and muscle. Tissue phosphorylase activity decreased significantly during bilateral eyestalk ablation. Administration of methionine-enkephalin into eyestalkless crabs caused no significant alterations in these parameters when compared to eyestalk ablated crabs. These results support the hypothesis that methionine-enkephalin produces hyperglycemia in crustaceans by triggering release of hyperglycemic hormone from the eyestalks.  相似文献

Two new species of benthopelagic copepods of the genus Stephos T. Scott, 1892, belonging to the family Stephidae G.O. Sars, 1902, are described based on specimens collected in the stagnant water flooding the burrows excavated by ocypodid crabs in two intertidal mud-flats, and from near-bottom shallow waters in Korea, respectively. They can be easily diagnosed based on the ornamentation of both the female genital double-somite and genital operculum; the morphology of the distal segment of the male right P5; the presence/absence of a tiny pointed process on the distomedial angle of second segment of female P5; and the condition (seta or spine) of the lateral armature element on the distal segment of female fifth legs, among other features. This is one of the few cases reported of calanoid copepods living as commensals of other invertebrates, and raises to six the number of members of the genus reported from Asia. This is also the first record of the family Stephidae in Korea.  相似文献

Previous studies have shown that injection of extracellular products (ECP) of Pseudoalteromononas atlantica isolated from shell disease-infected edible crabs (Cancer pagurus) into healthy crabs causes rapid death. In this study we examined the nature of the active lethal factor(s) in ECP. Injection of ECP into crabs caused a rapid decline in the total number of circulating hemocytes (blood cells), and the crabs died within 60 to 90 min. The individuals that died showed eyestalk retraction, limb paralysis, and lack of antennal sensitivity, suggesting that the active factor(s) targeted the nervous system. Histopathological investigations showed that affected crabs had large aggregates of hemocytes in the gills, and there was destruction of the tubules in the hepatopancreas. The active factor in ECP was not sensitive to heat treatment (100°C for 30 min) and proteinase K digestion. As lipopolysaccharide (LPS) was a potential candidate for the lethal factor, it was purified from whole P. atlantica bacteria or ECP and subsequently injected into crabs. These crabs had all of the external symptoms observed previously with ECP, such as limb paralysis and eyestalk retraction, and they died within 90 min after challenge, although no significant decline in the number of circulating hemocytes was observed. Similarly, in vitro incubation of hemocytes with purified LPS (1 to 20 μg) from P. atlantica did not result in the clumping reaction observed with ECP but did result in a degranulation reaction and eventual cell lysis. Injection of crabs with Escherichia coli or Pseudomonas aeruginosa LPS (1 μg g of body weight−1) did not cause any of the characteristic symptoms observed following exposure to P. atlantica LPS. No mortality of crabs followed the injection of E. coli LPS, but P. aeruginosa LPS caused ca. 80% mortality at 2 h after injection. Overall, these results show that the main virulence factor of P. atlantica for edible crabs is LPS either alone or in combination with other heat-stable factors.  相似文献

Summary

To overcome the problem of getting crustaceans to reproduce in captivity, eyestalk ablation or X-organ sinus gland removal is commonly utilized in commercially important species such as shrimp. We have investigated the effect of unilateral and bilateral eyestalk ablation on methyl farnesoate (MF) production by mandibular organs (MOs) and on ovarian maturation in female spider crabs Libinia emarginata, a useful model since these animals are in a terminal molt and are devoid of a functional Y-organ. Non-reproductive, over-wintering female L. emarginata were induced to be reproductive by feeding and increasing the holding temperature to stimulate the endocrine system. In addition, we removed X-organ sinus glands by eyestalk ablation either unilaterally (UEA) or bilaterally (BEA) to further stimulate MF synthesis by MOs. Endogenous MF in the hemolymph was extracted and quantified by means of HPLC and in some cases by GC/MS. Oocyte growth and egg quality were studied simultaneously to determine how they were related to MF levels found during vitellogenesis. The initial MF concentration in unablated controls was low, 0.31 ng/ml of hemolymph, and this increased (p<0.05) to about 1 ng/ml by 2 weeks, remaining at about that level for the remainder of the experiment. Eyestalk ablation significantly stimulated MF concentrations by week 1 to nearly 2 and 3.5ng/ml in the UEA (p <0.01) and BEA (p <0.001) animals, respectively. Oocytes appeared to respond to increased MF levels, as ovarian maturation was initiated from the point at which MF increased (p <0.05). Thereafter, the rate of oocyte growth was directly correlated with the extent of elevation of MF. The gonado-somatic index [(GSI) = gonad weight/body weight × 100] of controls at the start was about 1.5 and increased to 6.5 by week 4. Mature oocytes were reached at a GSI around 7. Oocyte maturation was accomplished at week 2 in BEA, week 3 in UEA, and later than week 4 in controls. After maturation, oocytes started to degrade in some ablated animals, particularly in the bilaterally ablated ones where the highest MF concentrations were observed. These data indicate that MF elevations are required for stimulating ovarian maturation in Crustacea. MF appears to accelerate gonad development during the vitellogenic process, but may be deleterious at high concentrations. These results have a significant and important application and implications for aquaculture.  相似文献

Calcium–calmodulin dependent protein kinase I is a component of a calmodulin-dependent protein kinase cascade and involved in many physiological processes. The full-length cDNA of calcium–calmodulin dependent protein kinase I (MnCaMKI) was cloned from the freshwater prawn Macrobrachium nipponense and its expression pattern during the molt cycle and after eyestalk ablation is described. The full-length cDNA of MnCaMKI is 3262 bp in length and has an open reading frame (ORF) of 1038 bp, encoding a 345 amino acid protein. The expression of MnCaMKI in three examined tissues was upregulated in the premolt stage of the molt cycle. Its expression was induced after eyestalk ablation (ESA): the highest expression level was reached 1 day after ESA in hepatopancreas, and 3 days after ESA in muscle. By dsRNA-mediated RNA interference assay, expression of MnCaMKI and ecydone receptor gene (MnEcR) was significantly decreased in prawns treated by injection of dsMnCaMKI, while expression of these two genes was also significantly decreased in prawns treated by injection of dsMnEcR, demonstrating a close correlation between the expression of these two genes. These results suggest that CaMKI in M. nipponense is involved in molting.  相似文献

Although conspicuous courtship displays are an effective way of attracting the attention of receptive females, they could provide valuable information to rival males on the location of these females. In fiddler crabs, males that see a receptive female wave their single, greatly enlarged claw in a highly conspicuous courtship display. We test whether other males use this courtship display to alert them to the presence of receptive females that they cannot directly see. We show that male fiddler crabs (Uca mjoebergi) eavesdrop on the courtship displays of nearby males to detect mate-searching females. This allows males to begin waving before a female becomes visible. Furthermore, males appear to adjust their waving according to the information available: eavesdropping males wave 12 times faster than non-courting males but only 1.7 times slower than males in full visual contact with the female.  相似文献

Biochemical analyses of total protein, lipid, carbohydrate, DNA, amino acid, and length, width, and dry weight measurements are reported for different stages of Romanomermis culicivorax cultured in the mosquito, Culex pipiens. The Bradford technique for assaying total protein was the most sensitive and reliable biochemical technique tested for assaying in vivo growth of R. culicivorax. Increases in total protein, lipid, carbohydrate, and dry weight during growth from preparasite to postparasite were greater than 6,900-fold for females and 2,300-fold for males. DNA increased 650-fold and 233-fold during development to female and male postparasites, respectively. The proportions of amino acids for preparasites were significantly different (P ≤ 0.01) from female and male postparasites for all amino acids tested, except methionine and tyrosine. Female and male postparasites were similar in protein, lipid, carbohydrate, DNA, and most amino acid proportions, but were significantly different in relative concentrations of serine, glycine, and alanine (P ≤ 0.01). Preliminary results suggest that the use of amino acid ratios from female postparasites improves the in vitro culture performance of R. culicivorax.  相似文献
