The spiral structure of the cyanobacterium Arthrospira (Spirulina) platensis (Nordst.) Gomont was previously found to be altered by solar ultraviolet radiation (UVR, 280–400 nm). However, how photosynthetic active radiation (PAR, 400–700 nm) and UVR interact in regulating this morphological change remains unknown. Here, we show that the spiral structure of A. platensis (D-0083) was compressed under PAR alone at 30°C, but that at 20°C, the spirals compressed only when exposed to PAR with added UVR, and that UVR alone (the PAR was filtered out) did not tighten the spiral structure, although its presence accelerated morphological regulation by PAR. Their helix pitch decreased linearly as the cells received increased PAR doses, and was reversible when they were transferred back to low PAR levels. SDS-PAGE analysis showed that a 52.0 kDa periplasmic protein was more abundant in tighter filaments, which may have been responsible for the spiral compression. This spiral change together with the increased abundance of the protein made the cells more resistant to high PAR as well as UVR, resulting in a higher photochemical yield.  相似文献

To study the impact of solar UV radiation (UVR) (280 to 400 nm) on the filamentous cyanobacterium Arthrospira (Spirulina) platensis, we examined the morphological changes and photosynthetic performance using an indoor-grown strain (which had not been exposed to sunlight for decades) and an outdoor-grown strain (which had been grown under sunlight for decades) while they were cultured with three solar radiation treatments: PAB (photosynthetically active radiation [PAR] plus UVR; 280 to 700 nm), PA (PAR plus UV-A; 320 to 700 nm), and P (PAR only; 400 to 700 nm). Solar UVR broke the spiral filaments of A. platensis exposed to full solar radiation in short-term low-cell-density cultures. This breakage was observed after 2 h for the indoor strain but after 4 to 6 h for the outdoor strain. Filament breakage also occurred in the cultures exposed to PAR alone; however, the extent of breakage was less than that observed for filaments exposed to full solar radiation. The spiral filaments broke and compressed when high-cell-density cultures were exposed to full solar radiation during long-term experiments. When UV-B was screened off, the filaments initially broke, but they elongated and became loosely arranged later (i.e., there were fewer spirals per unit of filament length). When UVR was filtered out, the spiral structure hardly broke or became looser. Photosynthetic O2 evolution in the presence of UVR was significantly suppressed in the indoor strain compared to the outdoor strain. UVR-induced inhibition increased with exposure time, and it was significantly lower in the outdoor strain. The concentration of UV-absorbing compounds was low in both strains, and there was no significant change in the amount regardless of the radiation treatment, suggesting that these compounds were not effectively used as protection against solar UVR. Self-shading, on the other hand, produced by compression of the spirals over adaptive time scales, seems to play an important role in protecting this species against deleterious UVR. Our findings suggest that the increase in UV-B irradiance due to ozone depletion not only might affect photosynthesis but also might alter the morphological development of filamentous cyanobacteria during acclimation or over adaptive time scales.  相似文献

To test the effects of photosynthetic active radiation (PAR, 400–700 nm) and ultraviolet radiation (UVR, 280–400 nm) on phototaxis and photosynthesis of free swimming microalgae, experiments were performed with Tetraselmis subcordiformis (Wille) Butcher under a solar simulator. In particular, we evaluated the effects of different PAR levels and radiation regimes (i.e., PAR only and PAR+UVR) on those two processes. We found that the cells preferred to move to a particular area (e.g., receiving 100 W m?2 PAR) with little photochemical suppression or inhibition of carbon fixation. Adding UV-A to high PAR decreased its swimming capacity and photosynthetic capability, and further adding UV-B led to more inhibition. The suppression of the moving capability of T. subcordiformis was reversible but the cells exposed to PAR combined with UVR needed longer time intervals to recover their motility as compared with those irradiated only with PAR. Based on the above results, we postulate that in nature, the motile capability and photosynthesis of free swimming the green microalga might be impaired by enhanced solar UVR. On the other hand, the cells can reduce the damage caused by high irradiances (and even get the optimum light level for photosynthesis) by a behavioral swimming response.  相似文献

It is well known that light and nutrients are essential to plants; however, there are few investigations in which these have been studied in combination on macroalgae, especially when solar ultraviolet radiation (UVR) is concerned. We cultured the red alga Gracilaria lemaneiformis (Bory) at different nitrate concentrations and light levels with or without UVR for 24 days. The results showed that nitrate supply markedly enhanced the growth and photosynthesis, increased the absorptivity of UV‐absorbing compounds (UVACs), and decreased photoinhibition in the presence of UVR. The thalli that received photosynthetically active radiation (PAR) treatment exhibited higher growth rates than those that received PAR + UVR at ambient or enhanced nitrate concentrations. However, under PAR + UVR treatment, the absorptivity of UVACs was higher than that of PAR and fluctuated with light levels. UVR was found to reduce the maximal net photosynthetic rate, apparent photosynthetic efficiency and light‐saturating irradiance while increasing the dark respiration rate, and inducing higher inhibition of growth and photosynthesis under high light versus under low light. Ultraviolet B significantly induced the synthesis of UVACs but led to higher inhibition on growth and photosynthesis than ultraviolet A.  相似文献

Natural levels of solar UVR were shown to break and alter the spiral structure of Arthrospira (Spirulina) platensis (Nordst.) Gomont during winter. However, this phenomenon was not observed during summer at temperatures of ~30°C. Since little has been documented on the interactive effects of solar UV radiation (UVR; 280–400 nm) and temperature on cyanobacteria, the morphology, photosynthesis, and DNA damage of A. platensis were examined using two radiation treatments (PAR [400–700 nm] and PAB [PAR + UV‐A + UV‐B: 280–700]), three temperatures (15, 22, and 30°C), and three biomass concentrations (100, 160, and 240 mg dwt [dry weight] · L?1). UVR caused a breakage of the spiral structure at 15°C and 22°C, but not at 30°C. High PAR levels also induced a significant breakage at 15°C and 22°C, but only at low biomass densities, and to lesser extent when compared with the PAB treatment. A. platensis was able to alter its spiral structure by increasing helix tightness at the highest temperature tested. The photochemical efficiency was depressed to undetectable levels at 15°C but was relatively high at 30°C even under the treatment with UVR in 8 h. At 30°C, UVR led to 93%–97% less DNA damage when compared with 15°C after 8 h of exposure. UV‐absorbing compounds were determined as negligible at all light and temperature combinations. The possible mechanisms for the temperature‐dependent effects of UVR on this organism are discussed in this paper.  相似文献

During October to December 2003 we carried out experiments to assess the impact of high solar radiation levels (as normally occurring in a tropical region of Southern China) on the cyanobacteria Nostoc sphaeroides and Arthrospira (Spirulina) platensis. Two types of experiments were done: a) Short-term (i.e., 20 min) oxygen production of samples exposed to two radiation treatments (i.e., PAR+UVR—280–700 nm, and PAR only—400–700 nm, PAB and P treatments, respectively), and b) Long-term (i.e., 12 days) evaluation of photosynthetic quantum yield (Y) of samples exposed to three radiation treatments (i.e., PAB; PA (PAR+UV-A, 320–700 nm) and P treatments, respectively). N. sphaeroides was resistant to UVR, with no significant differences (P>0.05) in oxygen production within 20 min of exposure, but with a slight inhibition of Y within hours. A fast recovery of Y was observed after one day even in samples exposed to full solar radiation. A. platensis, on the other hand, was very sensitive to solar radiation (mainly to UV-B), as determined by oxygen production and Y measurements. A. platensis had a circadian rhythm of photosynthetic inhibition, and during the first six days of exposure to solar radiation, it varied between 80 and 100% at local noon, but cells recovered significantly during afternoon hours. There was a significant decrease in photosynthetic inhibition after the first week of exposure with values less than 50% at local noon in samples receiving full solar radiation. Samples exposed to PA and P treatments recovered much faster (within 2–3 days), and there were no significant differences in Y between the three radiation treatments when irradiance was low (late afternoon to early morning). Long-term acclimation seems to be important in A. platensis to cope with high UVR levels however, it is not attained through the synthesis of UV-absorbing compounds but it seems to be mostly related to adaptive morphological changes.  相似文献

Background

Many coral reef organisms are photosynthetic or have evolved in tight symbiosis with photosynthetic symbionts. As such, the tissues of reef organisms are often exposed to intense solar radiation in clear tropical waters and have adapted to trap and harness photosynthetically active radiation (PAR). High levels of ultraviolet radiation (UVR) associated with sunlight, however, represent a potential problem in terms of tissue damage.

Methodology/Principal Findings

By measuring UVR and PAR reflectance from intact and ground bare coral skeletons we show that the property of calcium carbonate skeletons to absorb downwelling UVR to a significant extent, while reflecting PAR back to the overlying tissue, has biological advantages. We placed cnidarians on top of bare skeletons and a UVR reflective substrate and showed that under ambient UVR levels, UVR transmitted through the tissues of cnidarians placed on top of bare skeletons were four times lower compared to their counterparts placed on a UVR reflective white substrate. In accordance with the lower levels of UVR measured in cnidarians on top of coral skeletons, a similar drop in UVR damage to their DNA was detected. The skeletons emitted absorbed UVR as yellow fluorescence, which allows for safe dissipation of the otherwise harmful radiation.

Conclusions/Significance

Our study presents a novel defensive role for coral skeletons and reveals that the strong UVR absorbance by the skeleton can contribute to the ability of corals, and potentially other calcifiers, to thrive under UVR levels that are detrimental to most marine life.  相似文献

Previous study has shown that Porphyra conchocelis is sensitive to high levels of PAR (400–700 nm) as well as ultraviolet radiation (UVR: 280–400 nm), resulting in high inhibition of photosynthesis. However, little is known about whether the inner covering layer of the shell, in which the conchocelis lives, may provide protection against solar UVR. Our study indicates that the covering calcareous matrix is about 0.06 mm thick, transmitting 63, 47, and 28% of PAR, ultraviolet radiation A (UVA: 315–400 nm), and ultraviolet radiation B (UVB: 280–315 nm), respectively. We used a shading layer that simulated the above transmissions, and the effective quantum yield of PSII and photosynthetic carbon fixation in the conchocelis increased to greater extents in the presence of UVA or UVB. Attenuation of UVA by 19% and UVB by 37% due to the shading layer increased the PSII yield by 44%–77% and photosynthetic carbon fixation by about 60%. Our study clearly shows that the photosynthetic machinery of Porphyra haitanensis T. J. Chang et B. F. Zheng conchocelis was efficiently protected from harmful UVR by the covering calcareous matrix.  相似文献

以浮游硅藻假微型海链藻(Thalassiosira pseudonana(Hustedt)Hasle et Heimdal CCMP 1335)为材料,研究不同混合速率下,随辐射水平增加,UV辐射和可见光PAR对其光系统Ⅱ功能的影响。结果显示,混合速率慢时,随着PAR及UV辐射水平的增加,假微型海链藻PSⅡ的光化学效率(F_v/F_m)持续受到抑制,光合效率α和相对最大电子传递速率rETR_(max)下降。尤其是UV辐射存在时,PSⅡ反应中心D1蛋白含量下降,有活性的PSⅡ反应中心数量减少,单位反应中心吸收(ABS/RC)和耗散(DI_0/RC)的能量增加。混合速率快时,PAR辐射下PSⅡ光化学活性相比混合速率慢时升高,D1蛋白含量增加;而UV辐射存在下各光合参数表现出与混合速率慢时类似的变化趋势。研究结果表明水体混合速率的加快可缓解高水平可见光导致的光抑制,而对UV辐射的抑制效应并未产生显著改变。  相似文献

In vivo chlorophyll fluorescence analysis reflecting the photosystem II functionality was investigated in the cyanobacterium Anabaena variabilis PCC 7937 under simulated solar radiation in a combination with various cut-off filters (WG 280, WG 295, WG 305, WG 320, WG 335, WG 345, and GG 400) to assess the effects of photosynthetically active radiation (PAR), ultraviolet-A (UV-A), and ultraviolet-B (UV-B) radiations on photosynthesis. The photosynthetic activity (PA) was severely inhibited immediately after 10 min of exposure to high PAR, UV-A, and UV-B radiations compared with low PAR grown control samples. After 1 h of exposure, PA of 17.5 ± 2.9% was detected in the high PAR exposed samples compared with the control, while only a trace or no PA was observed in the presence of ultraviolet radiation (UVR). A recovery of PA was recorded after 2 h of the exposure, which continued for next 4, 8, 12, and 24 h. After 24 h of the exposure, PA of 57.5 ± 1.9%, 36.1 ± 11.7%, 23.5 ± 3.3%, 22.3 ± 5.2%, 20.8 ± 6.7%, 13.2 ± 6.6%, and 21.6 ± 9.5% was observed compared with the control sample in 400, 345, 335, 320, 305, 295, and 280 nm cut-off filters-covered samples, respectively. The relative electron transport rate, measured after 24 h exposure, showed also a disturbance in electron transfer between the two photosystems under the high PAR and UVR treatments relative to the control samples, suggesting the inhibition of photosynthesis. This study suggests that both high PAR and UVR inhibited the photosynthetic performance of A. variabilis PCC 7937 by damaging the photosynthetic apparatus, however, photoprotective mechanisms evolved by the organism allowed an immediate repair of ecologically important machinery, and enabled its survival.  相似文献

Solar ultraviolet radiation (UVR, 280–400 nm) is known to affect macroalgal physiology negatively, while nutrient availability may affect UV‐absorbing compounds (UVACs) and sensitivity to UVR. However, little is known about the interactive effects of UVR and nitrate availability on macroalgal growth and photosynthesis. We investigated the growth and photosynthesis of the red alga Gracilaria lemaneiformis (Bory) Grev. at different levels of nitrate (natural or enriched nitrate levels of 41 or 300 and 600 μM) under different solar radiation treatments with or without UVR. Nitrate‐enrichment enhanced the growth, resulted in higher concentrations of UVACs, and led to negligible photoinhibition of photosynthesis even at noon in the presence of UVR. Net photosynthesis during the noon period was severely inhibited by both ultraviolet‐A radiation (UVA) and ultraviolet‐B radiation (UVB) in the thalli grown in seawater without enriched nitrate. The absorptivity of UVACs changed in response to changes in the PAR dose when the thalli were shifted back and forth from solar radiation to indoor low light, and exposure to UVR significantly induced the synthesis of UVACs. The thalli exposed to PAR alone exhibited higher growth rates than those that received PAR + UVA or PAR + UVA + UVB at the ambient or enriched nitrate concentrations. UVR inhibited growth approximately five times as much as it inhibited photosynthesis within a range of 60–120 μg UVACs · g?1 (fwt) when the thalli were grown under nitrate‐enriched conditions. Such differential inhibition implies that other metabolic processes are more sensitive to solar UVR than photosynthesis.  相似文献

Species of the genus Ulva (Chlorophyta) are regarded as opportunistic organisms, which efficiently adjust their metabolism to the prevailing environmental conditions. In this study, changes in chlorophyll‐a fluorescence‐based photoinhibition of photosynthesis, electron transport rates, photosynthetic pigments, lipid peroxidation, total phenolic compounds, and antioxidant metabolism were investigated during a diurnal cycle of natural solar radiation in summer (for 12 h) under two treatments: photosynthetically active radiation (PAR: 400–700 nm) and PAR+ ultraviolet (UV) radiation (280–700 nm). In the presence of PAR alone, Ulva rigida showed dynamic photoinhibition, and photosynthetic parameters and pigment concentrations decreased with the intensification of the radiation. On the other hand, under PAR+UV conditions a substantial decline up to 43% was detected and an incomplete fluorescence recovery, also, P‐I curve values remained low in relation to the initial condition. The phenolic compounds increased their concentration only in UV radiation treatments without showing a correlation with the antioxidant activity. The enzimatic activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX) increased over 2‐fold respect at initial values during the onset of light intensity. In contrast, catalase (CAT) increased its activity rapidly in response to the radiation stress to reach maxima at 10 a.m. and decreasing during solar. The present study suggests that U. rigida is capable of acclimating to natural radiation stress relies on a concerted action of various physiological mechanisms that act at different times of the day and under different levels of environmental stress.  相似文献

We have studied the effects of nitrate supply under photosynthetic active radiation (PAR) plus ultraviolet radiation (UVR) exposure on photosynthetic pigments (chlorophyll a and carotenoids), photoprotective UV screen mycosporine-like amino acids (MAAs), and photosynthetic parameters, including the maximum quantum yield (F v/F m) and electron transport rate (ETR) on the red agarophyte Gracilaria tenuistipitata. Apical tips of G. tenuistipitata were cultivated under ten different concentrations of NO3 for 7 days. It has been shown that G. tenuistipitata cultured under laboratory conditions has the ability to accumulate high amounts of MAAs following a nitrate concentration-dependent manner under PAR + UVR. Two MAAs were identified, shinorine and porphyra-334. The relative concentration of the first increased under high concentrations of nitrate, while the second one decreased. The presence of antheraxanthin is reported for the first time in this macroalgae, which also contains zeaxanthin, lutein, and β-carotene. The accumulation of pigments, photoprotective compounds, and photosynthetic parameters of G. tenuistipitata is directly related to N availability. All variables decreased under low N supplies and reached constant maximum values with supplements higher than 0.5 mM NO3. Our results suggest a high potential to acclimation and photoprotection against stress factors (including high PAR and UVR) directly related to N availability for G. tenuistipitata.  相似文献

Zn availability in the ocean has been suggested to limit primary production by affecting CO2 acquisition processes for photosynthesis, therefore influencing the global carbon cycle. Also, UV radiation (UVR, 280–400 nm) is known to affect primary production in different ways. It remains to be ascertained whether Zn availability and UVR can act synergistically, antagonistically, or independently on oceanic primary production. We cultured the cosmopolitan diatom Skeletonema costatum (Grev.) Cleve under different radiation treatments with or without UVR (only photosynthetically active radiation), at 0, 3, and 10 pmol · L?1 Zn2+. Specific growth rate, photosynthetic carbon assimilation, external carbonic anhydrase (eCA) activity, and estimated cell abundance increased with increasing concentrations of Zn2+ from 0 to 3 and 10 pmol · L?1, irrespective of the radiation treatment. Higher eCA activity was observed in the cells grown at the high level of Zn2+ in the presence of UVR. An approximately linear relationship between μ and the daily dose of PAR was observed at 3 and 10 pmol · L?1 Zn2+ concentrations. However, the dependency of μ on the daily PAR dose disappeared when the cells were grown in the presence of UVR, which overall depressed both μ and photosynthetic carbon assimilation. The inhibitory effect of UVR was inversely related to Zn2+ concentrations. The ultraviolet‐B (UVB)‐related inhibition of growth and photosynthesis decreased with time, reflecting a faster acclimation of the cells to UVR at replete Zn2+ levels. Overall, growth in the presence of higher Zn2+ concentrations reduced the sensitivity to UV radiation in Skeletonema costatum.  相似文献

Hatching success was examined under exposure to solar ultraviolet radiation (UVR) using filters to give three different light conditions [C1: UV‐B, UV‐A and photosynthetically active radiation (PAR), C2: UV‐A and PAR, C3: PAR] in red Pagrus major and black Acanthopagrus schlegeli sea bream. Global warming and ozone depletion, and the resulting increase of ultraviolet radiation (UVR), have far-reaching impacts on biota, especially affecting the algae that form the basis of the food webs in aquatic ecosystems. The aim of the present study was to investigate the interactive effects of temperature and UVR by comparing the photosynthetic responses of similar taxa of Chlorella from Antarctic (Chlorella UMACC 237), temperate (Chlorella vulgaris UMACC 248) and tropical (Chlorella vulgaris UMACC 001) environments. The cultures were exposed to three different treatments: photosynthetically active radiation (PAR; 400–700 nm), PAR plus ultraviolet-A (320–400 nm) radiation (PAR + UV-A) and PAR plus UV-A and ultraviolet-B (280–320 nm) radiation (PAR + UV-A + UV-B) for one hour in incubators set at different temperatures. The Antarctic Chlorella was exposed to 4, 14 and 20°C. The temperate Chlorella was exposed to 11, 18 and 25°C while the tropical Chlorella was exposed to 24, 28 and 30°C. A pulse-amplitude modulated (PAM) fluorometer was used to assess the photosynthetic response of microalgae. Parameters such as the photoadaptive index (Ek) and light harvesting efficiency (α) were determined from rapid light curves. The damage (k) and repair (r) rates were calculated from the decrease in ΦPSIIeff over time during exposure response curves where cells were exposed to the various combinations of PAR and UVR, and fitting the data to the Kok model. The results showed that UV-A caused much lower inhibition than UV-B in photosynthesis in all Chlorella isolates. The three isolates of Chlorella from different regions showed different trends in their photosynthesis responses under the combined effects of UVR (PAR + UV-A + UV-B) and temperature. In accordance with the noted strain-specific characteristics, we can conclude that the repair (r) mechanisms at higher temperatures were not sufficient to overcome damage caused by UVR in the Antarctic Chlorella strain, suggesting negative effects of global climate change on microalgae inhabiting (circum-) polar regions. For temperate and tropical strains of Chlorella, damage from UVR was independent of temperature but the repair constant increased with increasing temperature, implying an improved ability of these strains to recover from UVR stress under global warming.  相似文献

发状念珠藻(Nostoc flagelliforme Bornet & Flahault)是一种重要的陆生经济蓝藻,室内培育出的原植体如何适应阳光辐射的问题尚需探讨。为此,作者将室内水培发菜置于阳光下培养,测定了其生长、有效光化学效率(F/Fm΄)和色素的变化。结果表明,较高的可见光(PAR,395-700 nm)和紫外辐射(UVR,280-395 nm)均导致水培发菜的F/Fm΄下降。第1天中午,PAR和UVR分别使F/Fm΄下降了54%和13%;傍晚,F/Fm΄有部分恢复。UVR对发菜适应阳光2d后的生长无负面作用。发菜在适应全阳光辐射期间,紫外吸收物质(Scytonemin和Mycosporine-like amino acids)含量不断增加,9d后,分别增加了124倍和9倍。这些紫外吸收物质的增加对发菜细胞降低光抑制,适应阳光辐射,起到了重要作用。本研究的结果可为水培发菜室外培养方法的建立提供一定的理论依据。  相似文献

At Helgoland, in the North Sea, growth of the high sublittoral brown macroalga Dictyota dichotoma (Hudson) Lamoroux was examined in October (the time of tetraspore release) in an outdoor tank by exposing 2-day-old germlings to four solar radiation treatments achieved with different filter materials or an additional artificial light source: photosynthetically active radiation (PAR; 395–700 nm), PAR plus ultraviolet (UV)-A (320–700 nm), full solar spectrum, or solar spectrum plus artificial UV radiation (UVR). Based on length measurements over a period of 3 weeks, the growth rate in germlings strongly decreased in conditions with UVR compared to PAR: by 14% under PAR+UV-A, by 31% under the full solar spectrum and by 65% with additional UVR. Although growth rates of germlings under UVR were reduced mainly in the first week, the plants did not regain the size of the untreated plants even after 9 weeks. Regardless of the exposure, no defects in morphology or anatomy including the exposed apical meristem were detected, except for a reduction in cell division rates perhaps due to additional cost for photoprotective or repair mechanisms. Depending on the actual position of D. dichotoma plants in the natural habitat, individuals in high positions receive substantial amounts of the more harmful UV-B while those lower down might only receive UV-A during part of the day, thus the effect of UV-B on the growth of D. dichotoma will depend on its position in the field. The effects of tidal variation of the light climate and the implications of our results for the zonation of D. dichotoma are discussed. The brown macroalga Laminaria saccharina (L.) J. V. Lamour. was grown in large outdoor tanks at 50% ambient solar radiation for 3–4 weeks in July and August of 2000, 2001, and 2002, in either ambient or nitrogen (N)–enriched seawater and in either ambient light [PAR + ultraviolet radiation (UVR)] or ambient light minus UVR. Growth, N‐content, photosynthetic pigments, and RUBISCO content increased in N‐enriched seawater, indicating N‐limitation. UVR inhibited growth, but this inhibition was ameliorated by N‐enrichment. The response of growth to UVR could not be explained by changes in respiration and photosynthesis. Gross light‐saturated photosynthesis (Pmax) remained unaffected by UVR but was significantly higher under N‐enrichment, as was dark respiration (Rd). UVR had no effect on pigments or N content. However, RUBISCO contents were low in the presence of UVR, reflecting the overall change in soluble cellular protein. Overall, our data indicate that the response to UVR in L. saccharina depends on other environmental factors, such as N, and these effects need to be considered when evaluating the response of macroalgae to increased UVR.  相似文献
