The in vivo effects of (+/-) baclofen (10(-6)-10(-4) M), muscimol (10(-5)-10(-4) M), and (+) bicuculline (10(-4)-10(-3) M) applications on evoked potentials in the olfactory bulb (OB) and primordial hippoccamp (PH) were studied in frogs. Baclofen was found to decrease drastically postsynaptic components of OB orthodromic potential and to slightly increase OB antidromic potential. Muscimol decreased only the second component of OB orthodromic potential and OB antidromic potential. Baclofen and muscimol decreased all the components of PH orthodromic potential, but for the first positive one. All the effects were reversible and dose-dependent. Bicuculline antagonized muscimol effects, without affecting baclofen effects in both structures under study. The results suggest the presence of two types of GABA-receptors in OB and PH of frogs. GAGAB-receptors are shown to be located on the primary olfactory afferents and to be responsible for presynaptic inhibition in OB.  相似文献

Summary The densities and diameters of intramembranous particles in olfactory and nasal respiratory structures of frog, ox, rat and dog have been compared using the freeze-fracture technique. Dendritic endings and the various segments of the cilia of the olfactory receptor cells of a given species have identical particle densities (700–1,800 particles/m2 in P-and 100–600 in E-faces). Densities in P-faces of respiratory cilia are about 1/3 of those in the olfactory cilia. E-face particle densities of these respiratory cilia are often higher than P-face densities. Microvillus P-face densities range from 700–2,000 (respiratory cell microvilli) to 1,800–3,400 particles/m2 (olfactory supporting and Bowman's gland microvilli). Microvillus E-faces show no conspicuous mutual differences. Literature comparisons showed that odour concentrations at threshold are considerably lower (105–1010 times) than the concentrations of olfactory receptor ending intramembranous particles (5 M–30 M) expressed in the same units.Relative differences in particle distributions of the various cell structures studied are usually species-independent. Absolute values vary considerably with the species. Relative P-face particle densities of the supporting cell microvilli tend to correlate with those of dendritic ending structures. Particle diameters are usually similar for corresponding structures and fracture faces in the four species. Apical structures of supporting and Bowman's gland cells in rat and dog show rod-shaped particle aggregates in their P-and pits in their E-faces. Neither sex-dependency nor an influence related to physiological treatments on the particle distributions could be demonstrated.  相似文献

Goldfish are ideal vertebrates for the study of regeneration within the central nervous system. The present behavioural and neuroanatomical investigations after bilateral transection of the entire olfactory tracts of either lateral or medial subtracts have been designed (1) to examine the relationship between morphological changes and changes in the perception of spontaneously preferred chemosensory stimuli, (2) to investigate the animals' ability to qualitatively discriminate amino acids in olfactory concentrations (below taste threshold, 10-6–10-8 M), one of which had been rewarded preoperatively (specific regeneration), and (3) to examine the discriminative ability for amino acids at concentrations above taste threshold (> 10-5 M) in intact sham-operated, and in operated specimens at various time intervals before functional regeneration. Within 10–14 days after bilateral transection of the lateral olfactory tracts, specific regeneration was observed. After bilateral transection of the medial olfactory tracts, no immediate behavioural change was recorded for 1 week. Thereafter, goldfish behaviour became unstable and dropped to the chance level for 3–4 weeks. Subsequent to this time the goldfish returned to the preoperative level. Following bilateral crushing of the olfactory tracts and after total tractotomy, a specific regeneration was observed after 4 weeks and 6–8 weeks, respectively, post op. A specific heat shock puff (II-48 C) has been induced in isolated salivary glands of D. hydei by pyridoxine (vitamin B6) at concentrations from 10–7 to 10–2 M. The puff begins to form within 5 min and continues to increase in size up to 2 h. Under conditions of puff induction the -amanitin sensitive synthesis of a 40 KD protein is induced.  相似文献

Vitamin B6 induces in vivo as well as in vitro the appearance of a puff at region 2–48C in Drosophila hydei. At concentrations of 10–2 M or lower, region 2–48C is the only region responding to vitamin B6 provided that oligomycin is present in the incubation medium. Pyridoxal phosphate and pyridoxamine phosphate supplied to medium containing oligomycin induce upon incubation of salivary glands a larger puff at 2–48C. — Puff 2–48C produces large quantities of a unique RNP-product; globular 140–220 Å particles which aggregate to stable complexes of 0.1–0.2 in diameter. Upon continuous in vitro incubation with vitamin B6, puff 2–48C becomes loaded with these aggregates which have never been observed in any other puff of Drosophila hydei.  相似文献

The effect of L-arginine on the Na+,K+-ATPase activity in rat aorta endothelium was studied at its physiological concentrations in the range of 10–6-10–3 M. The enzyme activity was 35.5% increased by low concentrations of L-arginine (10–5 M) and its activity was 32.3-37.1% decreased at the L-arginine concentrations of 10–4-10–3 M. A similar inhibition (by 34.5-42.8%) was also found in the presence of a NO-donor nitroglycerol (10–4-10–3 M). An optical isomer of L-arginine, D-arginine, at the concentrations of 10–5 M also increased the enzyme activity by 37.1%, but its inhibiting effect was much less pronounced and was 15.7% at the D-arginine concentration of 10–3 M. An inhibitor of NO-synthase, L-NAME (NG-nitroarginine, methyl ester), failed to inhibit Na+,K+-ATPase. However, the presence of L-NAME abolished the inhibition of Na+,K+-ATPase by high concentrations of L-arginine. Thus, the effect of L-arginine on the endothelial Na+-pump depended on its concentration, and it is suggested that the enzyme inhibition by high concentrations of L-arginine should be associated with activation of the endogenous synthesis of NO.  相似文献

Intracellular recordings were made from the major neurites of local interneurons in the moth antennal lobe. Antennal nerve stimulation evoked 3 patterns of postsynaptic activity: (i) a short-latency compound excitatory postsynaptic potential that, based on electrical stimulation of the antennal nerve and stimulation of the antenna with odors, represents a monosynaptic input from olfactory afferent axons (71 out of 86 neurons), (ii) a delayed activation of firing in response to both electrical- and odor-driven input (11 neurons), and (iii) a delayed membrane hyperpolarization in response to antennal nerve input (4 neurons).Simultaneous intracellular recordings from a local interneuron with short-latency responses and a projection (output) neuron revealed unidirectional synaptic interactions between these two cell types. In 20% of the 30 pairs studied, spontaneous and current-induced spiking activity in a local interneuron correlated with hyperpolarization and suppression of firing in a projection neuron. No evidence for recurrent or feedback inhibition of projection neurons was found. Furthermore, suppression of firing in an inhibitory local interneuron led to an increase in firing in the normally quiescent projection neuron, suggesting that a disinhibitory pathway may mediate excitation in projection neurons. This is the first direct evidence of an inhibitory role for local interneurons in olfactory information processing in insects. Crayfish glutamic acid decarboxylase (GAD), like the homologous enzymes from other species, is inhibited by carbonyl-trapping agents (e.g. aminooxyacetic acid; AOAA) and sulfhydryl reagents (e.g. 5,5-dithiobis-(2-nitrobenzoic acid); DTNB). It also is inhibited by the product GABA, many anions (e.g. SCN and Cl), and some cations (e.g. Zn+2). The inhibition by AOAA, but not that by DTNB, was prevented by increasing the concentration of the pyridoxal phosphate (PLP) coenzyme. GABA blocked the effects of PLP on enzyme activity. The inhibition by AOAA, DTNB, GABA, and chloride all were competitive with substrate. The effect of GABA occurs at physiological concentrations and may contribute to the regulation of GAD activity in vivo. The quantitative effect of anions is dependent on the cation with which they are administered. ATP stimulated GAD activity in homogenates prepared with potassium phosphate or Tris-acetate buffer, even when no exogenous PLP was provided.  相似文献

Background

Niemann-Pick type C disease (NPC) is a rare autosomal recessive lipid storage disease characterized by progressive neurodegeneration. As only a few studies have been conducted on the impact of NPC on sensory systems, we used a mutant mouse model (NPC1−/−) to examine the effects of this disorder to morphologically distinct regions of the olfactory system, namely the olfactory epithelium (OE) and olfactory bulb (OB).

Methodology/Principal findings

For structural and functional analysis immunohistochemistry, electron microscopy, western blotting, and electrophysiology have been applied. For histochemistry and western blotting, we used antibodies against a series of neuronal and glia marker proteins, as well as macrophage markers.NPC1−/− animals present myelin-like lysosomal deposits in virtually all types of cells of the peripheral and central olfactory system. Especially supporting cells of the OE and central glia cells are affected, resulting in pronounced astrocytosis and microgliosis in the OB and other olfactory cortices. Up-regulation of Galectin-3, Cathepsin D and GFAP in the cortical layers of the OB underlines the critical role and location of the OB as a possible entrance gate for noxious substances. Unmyelinated olfactory afferents of the lamina propria seem less affected than ensheathing cells. Supporting the structural findings, electro-olfactometry of the olfactory mucosa suggests that NPC1−/− animals exhibit olfactory and trigeminal deficits.

Conclusions/Significance

Our data demonstrate a pronounced neurodegeneration and glia activation in the olfactory system of NPC1−/−, which is accompanied by sensory deficits.  相似文献

Summary Extracellular levels of cholecystokinin (CCK), dopamine (DA), glutamate (Glu) and aspartate (Asp) were simultaneously monitored in the frontoparietal cortex and the striatum of halothane-anaesthetized rats using in vivo microdialysis. Under basal conditions, cortical and striatal CCK levels were 3.11 ± 0.39 pM and 2.76 ± 0.15 pM, respectively. Local KCl (10–1 M) and bicuculline (10–4 M) co-application in cortex or striatum increased the CCK levels 18-fold and 26-fold, respectively. The DA level in striatum was 3.78 ± 0.28 nM and the local perfusion with KCl + bicuculline led to a 45-fold increase. The cortical and striatal outputs of Glu were of the order of 2 · 10–6 M and Asp levels were around 6 · 10–7 M. Local stimulation with KCl (10–1 M) and bicuculline (10–4 M) caused a small increase (2 fold) in cortical and striatal levels of Glu and Asp. The addition of KCl (10–1 M) and bicuculline (10–4 M) to the cortical perfusion medium did not modify CCK, DA or Glu concentrations in striatum. These results demonstrate that CCK, DA, Glu and Asp may be simultaneously monitored in vivo and support the idea that their extracellular levels recovered in the microdialysis perfusates could be derived from neuronal pools.  相似文献

In research on -aminobutyric acid (GABA) used at different concentrations on the amplitude of EPSP within populations (PEPSP), as recorded from dentrites in isolated hippocampal slices, GABA induced a dose-dependent reversible reduction in PEPSP amplitude with no noticeable signs of desensitization. Highest sensitivity to GABA was shown by PEPSP in hippocampal zone CA1 (threshold concentration: 3×10–5–2×10–4 M; (concentration at which the effect equal to 1/2 of maximum occurs) IC50: 5×10–4–1×10–3 M). The effects of GABA on PEPSP were not blocked by bicuculline, picrotoxin, or penicillin. Action of GABA on dendritic antidromic population spike (DAPS — postynaptic effects) were slightly diminished by these blockers. Baclofen inhibited PEPSP more powerfully than GABA (threshold concentration: 1×10–6 M: IC50: 3×10–6 M), although it only produced a minor reduction in DAPS amplitude even at high concentrations. It is concluded that the inhibitory effect of GABA on PEPSP in hippocampal zone CA1 may be put down mainly to its presynaptic action mediated by GABAB receptors on axonal terminals of Schaffer collaterals.A. Summary Transmitter receptor ion channels from previously identified rabbit olfactory bulb neurons were studied by using a thin slice preparation in combination with patch-clamp measurements. PG cells, which closely resembled previously described periglomerular interneurons in their morphology, responded to microapplication of GABA, acetylcholine, norepinephrine and glycine with the activation of distinct ionic currents. JG cells, which belong either to the class of short axon cells or external tufted cells, never showed GABA responses. In mitral cells ionic currents activated by GABA, acetylcholine, norepinephrine and glutamate could be elicited. Further measurements of GABA-activated currents of PG cells were made and indicated that these cells expressed two different types of GABA receptors: one which showed fast desensitization with a decay time constant of about 5 s, and one which slowly desensitized with a decay time constant of about 20–30 s. Both types were completely inhibited by bicuculline methiodide (50 M). GABA receptors were not blocked by Zn2+ (0.1 mM). From the dose-response relationship of the peak GABA-activated currents, an apparent dissociation constant of 50 M was derived. From single channel measurements in excised outside-out patches, a single channel conductance of GABA-activated Cl currents of 24 pS was obtained during continuous application of the agonist. The effect of vanadate on the phosphorylation of synaptosomal membrane proteins prepared from rat cerebral cortex was studied. Vanadate concentrations of 10–6, 10–5, and 10–4 M increased the endogenous phosphorylation activity by 25%, 37%, and 75%, respectively. Increasing the ATP concentration in the assay medium from 50 to 500 M did not influence the above effect. A commercial preparation of the purified protein kinase was stimulated 40% by 10–3 M vanadate. Calcium-calmodulin dependent activity was stimulated only 20% by 10–5 M vanadate. The effect was not enhanced by further increasing vanadate concentration. Addition of calcium ions (above 50 M) suppressed the vanadate effect, while an inhibition was observed at high Ca2+ concentration (2.5 mM). Below 50 M calcium ions stimulated phosphorylation activity in the absence of vanadate and did not affect the stimulatory action of vanadate. Cyclic AMP-dependent endogenous phosphorylation was also stimulated by vanadate. Activation by cAMP could not be observed at vanadate concentrations above 10–6 M. Possible mechanisms of the vanadate effect are discussed.  相似文献

Summary Freeze-fracture data on antennal olfactory and labellar gustatory sensilla of the blowfly Calliphora vicina were compared with those of vertebrate olfactory organs.Insect antennal and vertebrate olfactory axons have similar diameters and show vesicular expansions; insect labellar axons are on average twice as thick and show no vesicular expansions. Vertebrate olfactory and insect labellar and antennal axons display similar intramembranous particle densities. Antennal axons show particle arrangements, resembling tight-junctions. The few extremely thick axons found in labella and antennae show particle arrangements resembling gap-junctions.In regions, proximal to the pores in the insect sensillar hairs, P-faces of olfactory and gustatory cilia show about 200 particles/m2. The most proximal and distal portions of the sensory cilia, necklaces and regions in the vicinity of the hair pores respectively, were only encountered in antennal sensilla. P-faces of the ciliary membranes underneath these pores display 1,000–1,200 particles/ m2 in unbranched and branched cilia. These values agree with values found in vertebrate olfactory cilia. It is suggested that these high particle densities are related to entities involved in chemoreceptive activities.Accessory cell micropliae have P-face densities of 2,000–3,000 particles/ m2, values similar to those found in vertebrate supportive cell microvilli. The membranes of the accessory cells display septate-junctions in areas where these cells overlap themselves, each other and in places where they adhere to the exoskeleton or the basement membrane.  相似文献

Summary Superfusion of synaptosomes prepared from rat olfactory bulb revealed constant basal release of endogenous taurine (Tau), aspartate (Asp), glutamate (Glu) and-aminobutyrate (GABA): their release rates were 110.4 ± 13.0, 30.3 ± 6.7, 93.7 ± 13.1, and 53.3 ± 8.8 pmol/min/mg protein, respectively. The depolarizing-stimulation with 30mM KCl evoked 1.17-, 2.18-, 2.55- and 1.53-fold increases, respectively. Tau release was calcium-independent. However, the perfusion of synaptosomes with Tau (10µM) inhibited the evoked increase in GABA release by 63% without changing basal release, although it did not affect release of Asp and Glu. Phaclofen (10µM, a GABAB receptor antagonist), but not bicuculline (10µM, a GABAA receptor antagonist), counteracted the Tau-induced reduction in GABA release. These data suggest that Tau may be abundantly released from nerve endings of rat olfactory bulb and that it may regulate GABA release through the activation of presynaptic GABAB autoreceptors.  相似文献

Membranes prepared from the retinal pigment epithelium of several species possess a specific site which binds [3H]strychnine. This binding has a somewhat lower affinity and a much greater density than the corresponding interaction in the hindbrain or neural retina. Binding is not greatly altered in the presence of 10–3 M glycine,l-alanine, -alanine, taurine, or serine. Thus, the receptor does not resemble the classical glycine receptor of the hindbrain and spinal cord. This new type of binding site appears to be confined to the pigment epithelial layer of the retina.  相似文献

To understand avian olfaction, it is important to characterize the peripheral olfactory system of a representative bird species. This study determined the functional properties of olfactory receptor neurons of the chicken olfactory epithelium. Individual neurons were acutely isolated from embryonic day-18 to newborn chicks by dissection and enzymatic dissociation. We tested single olfactory neurons with behaviorally relevant odorant mixtures and measured their responses using ratiometric calcium imaging; techniques used in this study were identical to those used in other studies of olfaction in other vertebrate species. Chick olfactory neurons displayed properties similar to those found in other vertebrates: they responded to odorant stimuli with either decreases or increases in intracellular calcium, calcium increases were mediated by a calcium influx, and responses were reversibly inhibited by 100 M Lcis–diltiazem, 1 mM Neomycin, and 20 M U73122, which are biochemical inhibitors of second messenger signaling. In addition, some cells showed a complex pattern of responses, with different odorant mixtures eliciting increases or decreases in calcium in the same cell. It appears that there are common features of odorant signaling shared by a variety of vertebrate species, as well as features that may be peculiar to chickens.  相似文献

The interaction of various hormones and regucalcin on (Ca2+–Mg2+)-ATPase activity in rat liver plasma membranes was investigated. The presence of epinephrine (10–6–10–4 M), and insulin (10–8–10 M) in the reaction mixture produced a significant increase in (Ca2+–Mg2+)-ATPase activity, while the enzyme activity was decreased significantly by calcitonin, (3×10–8–3×10–6 M). These hormonal effects, except for calcitonin, were clearly inhibited by the presence of vanadate (10–4 M) which can inhibit the Ca2+-dependent phosphorylation of enzyme. Meanwhile, regucalcin (0.25 and 0.50 M), isolated from rat liver cytosol, elevated significantly (Ca2+–Mg2+)-ATPase activity in the plasma membranes, although this elevation was not inhibited by vanadate (10–4 M). the epinephrine (10–5 M) or phenylephrine (10–4 M)-induced increase in (Ca2+–Mg2+)-ATPase activity was disappeared in the presence of regucalcin; in this case the effect of regucalcin was also weakened. However, the inhibitory effect of calcitonin (3×10–6 M) was not weakened by the presence of regucalcin (0.5 M). Moreover, GTP (10–5 and 10–4 M)-induced increase in (Ca2+–Mg2+)-ATPase activity was not seen in the presence of regucalcin (0.25 M). The present finding suggests that the activating mechanism of regucalcin on (Ca2+–Mg2+)-ATPase is not involved on GTP-binding protein which modulates the receptor-mediated hormonal effect in rat liver plasma membranes.  相似文献

Intracellular responses of granule cells and secondary neurons of the carp olfactory bulb to electrical stimulation of the olfactory nerve and olfactory tract were investigated. Synaptic responses of granule cells to both types of stimuli consisted of an early and late EPSP and IPSP. Comparison of responses of the secondary and granule neurons indicated that the granule cells are interneurons of postsynaptic inhibition of secondary neurons. The results suggest that dendro-dendritic and recurrent collateral pathways exist for the activation of granule cells and that inhibitory synapses are located on those dendrites of the secondary neurons that are in contact with endings of olfactory nerve fibers.M. Rhodopseudomonas acidophila strain 7050 can satisfy all its nitrogen and carbon requirements from l-alanine. Addition of 100 M methionine sulfoximine to alanine grown cultures had no effect on growth rate indicating that deamination of alanine via alanine dehydrogenase and re-assimilation of the released NH 4 + by glutamine synthetase/glutamate synthase was an insignificant route of nitrogen transfer in this bacterium. Determination of aminotransferase activities in cell-free extracts failed to demonstrate the presence of direct routes from alanine to either aspartate or glutamate. The only active aminotransferase involving l-alanine was the alanine-glyoxylate enzyme (114–167 nmol·min–1·mg–1 protein) which produced glycine as end-product. The amino group of glycine was further transaminated to yield aspartate via a glycineoxaloacetate aminotransferase (117–136 nmol·min–1 ·mg–1 protein). No activity was observed when 2-oxoglutarate was substituted for oxaloacetate. The formation of glutamate from aspartate was catalysed by aspartate-2-oxoglutarate aminotransferase (85–107 nmol·min–1·mg–1 protein).
