The effects of sperm freezing concentration (40 × 106 mL−1 vs. 400 × 106 mL−1), straw size (0.25 mL vs. 0.5 mL) and freezing method (liquid nitrogen vapour in a Styrofoam® box vs. programmable freezing machine) were evaluated in a 2 × 2 × 2 factorial experimental design using 3 split ejaculates from each of 4 stallions. Immediately after thawing, the total motility and forward progressive motility of spermatozoa frozen at a concentration of 40 × 106 mL−1 was higher than for spermatozoa frozen at 400 × 106 mL−1. No significant differences were observed in the semen parameters assessed after cryopreservation in either 0.25 or 0.5 mL straws. However, the programmable freezer provided a more consistent and reliable freezing rate than liquid nitrogen vapour. We conclude that an effective protocol for the cryopreservation of stallion spermatozoa at low concentrations would include concentrations of 40 × 106 mL−1 in 0.25 mL straws using a programmable freezer. This freezing protocol would be suitable for emerging sperm technologies such as sex-preselection of stallion spermatozoa as the sorting process yields only low numbers of spermatozoa in a small volume available for either immediate insemination or cryopreservation.  相似文献

The aims of the present study were to evaluate if seasonality in semen characteristics and plasma testosterone concentrations exist in Markhoz male goats. Ten Markhoz (Angora) bucks were housed and fed according to standard recognized practices. During the observation period, semen was collected monthly with the aid of an electro-ejaculator and examined microscopically immediately after collection. Physical parameters of semen and the semen index were recorded. Blood samples were also taken monthly throughout the observation period and the plasma testosterone concentration monitored. Bucks demonstrated a higher semen quality (P < 0.05) in autumn and summer (semen index of 965 × 106 and 752 × 106 ml−1, respectively), compared to spring and winter (semen index of 606 × 106 and 512 × 106, respectively). This coincided with a higher (P < 0.05) plasma testosterone concentration in autumn and summer (8.1 and 10.1 ng ml−1, respectively), compared to that obtained in spring (3.0 ng ml−1) and winter (2.5 ng ml−1). During autumn and summer, the ejaculate volume (average of 1.2 and 1.0 ml), sperm output (1159 × 106 and 1005 × 106 sperm ml−1), sperm mass motility (4.2 and 4.3), sperm progressive motility (83.9 and 82.0%) and percentage live sperm (90.7 and 88.2%, respectively) of the bucks were higher (P < 0.05) than in the spring (0.6 ml, 880 × 106 sperm ml−1, 3.3, 71.5% and 80.2%) and winter (0.7 ml, 863 × 106 sperm ml−1, 4.0, 71.5% and 84.9%, respectively). During autumn and summer, the percentage of sperm abnormalities (5.0 and 9.2%) was significantly lower than that in spring (12.9%) and winter (11.2%). The semen pH was slightly alkaline being significantly (P < 0.05) lower in the autumn (7.1) than in spring (7.3). Data showed season of the year to influence all semen parameters evaluated—indicating that optimal buck performance may be obtained in late summer and autumn. It can thus be said that Markhoz bucks have distinct seasonal spermatogenic activity, with poorer semen characteristics being recorded during winter and spring. This may be a critical obstacle when implementing an intensive breeding system of three kidding seasons in 2 years, with natural mating being implemented.  相似文献

Sperm vitrification has been recently developed, but fertility trials have not been performed yet in equine species. In this study, a new warming technique for vitrified donkey semen was developed and the uterine inflammatory response and fertility were compared to conventional freezing. In Experiment 1, sperm was vitrified in straws and warmed in 3 ml of extender or in a water bath at: 37 °C/30 s; 43 °C/10 s; and 60 °C/5 s. Sperm motility, plasma and acrosome membranes and DNA integrity were compared between treatments. In Experiment 2, jennies were inseminated twice (500 × 106 sperm) in the uterine body either with vitrified or frozen semen (2 cycles/jenny). Pregnancy rates and the uterine inflammatory response (polymorphonuclear neutrophil concentration; PMN) were evaluated after artificial insemination (AI). No differences between warming in extender/water bath were found and 43 °C/10 s was better than lower temperatures in terms of total (53.8 ± 13.2%) and progressive sperm motility (41.4 ± 11.4%). No differences in PMN concentration (× 103 PMN/ml) were found between vitrified (276.8 ± 171.6) or frozen (309.7 ± 250.7) semen after AI. However, PMN decreased faster (P < 0.05) using vitrified semen. Pregnancy rates were greater for vitrified (22%) than frozen semen (10%) but not statistically different. In conclusion, donkey sperm vitrified in straws could be directly warmed in a water bath at 43 °C/10 s, reducing the uterine inflammatory response obtained after AI and promoting positive pregnancy outcomes. These findings confirm the possibility to use vitrified semen as an alternative for AI in jennies.  相似文献

The aim of this study was to analyze the main influences on the quality and quantity of boar semen. A total of 230,705 records of semen collections were utilised to estimate statistics of semen traits of 2712 boars belonging to the following breeds: Czech Meat Pig, Duroc, Hampshire, Landrace, Large White, Czech Large White and Pietrain, and various crosses of these breeds. The evaluation was based on semen volume (VO), concentration of spermatozoa (CO), progressive motion of spermatozoa (MO), abnormal spermatozoa (AB), total number of spermatozoa (NOT) and corrected number of spermatozoa (NOC). The breeds differed significantly for all examined traits. The maximal differences between the breeds were 95 ml for VO, 109 × 103 mm−3 for CO, 9% for MO, 1.6% for AB, 24 × 109 for NOT and 19 × 109 for NOC. The maximal heterosis effect reached 12% for VO, 17% for CO, 4% for MO, −14% for AB, and 8% for NOT and NOC. The results demonstrate that the year-season effect has a clear effect on semen quality. The lowest values of semen traits were observed in summer while the highest values were found in autumn and winter. Age of boar was found to have a strong impact on sperm output. Sperm output tended to increase up to a boars’ age of 3.5 years. An acceptable level of semen volume occurred after a sexual pause of 3 days and the pool of spermatozoa was restoring after 5–7 days and fully after 10–11 days.  相似文献

Experiments were conducted to develop a suitable protocol for cryopreservation of spotted skunk semen. Semen was collected by electroejaculation of captive male skunks (n = 16) from late January through late November. In the first experiment, fresh semen was diluted in either TEST (n = 10), TRIS (n = 9), or BF5F (n = 7) extenders and maintained at 4°C for 16 hr. Sperm motility in these extenders was not significantly different before cooling (P = 0.71), but samples diluted with BF5F exhibited significantly lower sperm motility than the other extenders at all time points after cooling (P < 0.05). In the second experiment, fresh semen was diluted in TEST containing either 3, 5, or 10% DMSO or 3, 5, or 10% glycerol as a cryopreservative. These samples were cooled to 4°C and frozen in 0.25 ml French straws on dry ice. Some samples containing 5% DMSO or 5% glycerol (n = 4), were also frozen on dry ice as pellets. Frozen samples were maintained in liquid nitrogen. Fresh samples had significantly greater sperm motility in dimethyl sulfoxide (DMSO) than in glycerol (P < 0.05), while frozen and thawed samples had the highest motility in 5 or 10% DMSO or 10% glycerol. Samples frozen in French straws had significantly greater sperm motility after freezing and thawing than those frozen by the pellet method (P < 0.05). Optimum cryoprotection was achieved with the TEST extender containing 5 or 10% DMSO, when used in conjunction with French straws. © 1992 Wiley-Liss, Inc.  相似文献

In the 2004/2005 breeding season, the fertility of sex-sorted (SS) and non-sorted (NS) frozen stallion spermatozoa from two Hannovarian stallions was compared. A hysteroscopic insemination technique [Morris, L.H., Tiplady, C., Allen, W.R., 2003a. Pregnancy rates in mares after a single fixed time hysteroscopic insemination of low numbers of frozen–thawed spermatozoa onto the uterotubal junction. Equine Vet. J. 35, 197–201] was used to deposit low doses (6, 13 or 25 × 106 frozen–thawed SS or NS spermatozoa) onto the utero-tubal junction at 32 or 38 h after the administration of Chorulon (2500 IU, Intervet). Fertility was low, with one pregnancy (13 × 106 spermatozoa, 500 μL) obtained after artificial insemination with frozen SS spermatozoa (n = 29 cycles) which resulted in the birth of a filly. Two pregnancies were obtained in mares inseminated with 6 × 106 NS spermatozoa in 250 μL (n = 31 cycles). Mares failing to conceive on two experimental cycles were allocated to the conventional insemination group. Insemination with >500 × 106 motile NS frozen–thawed spermatozoa, yielded satisfactory per cycle conception rates (35.5%, 22/62) for both stallions combined and was within the values of their normal fertility as quoted by the stud's records. This suggests that the quality of the frozen semen was acceptable and that the freezing processes yielded viable spermatozoa capable of fertilisation. The poor fertility after hysteroscopic insemination with low doses of sex-sorted or non-sorted spermatozoa from the same stallions may be directly attributable to the low dose insemination conditions with frozen–thawed rather than sex-sorted spermatozoa.  相似文献

Activated sludge from a wastewater treatment plant consists of a consortium of microbes that utilize various organic molecules including persistent organic pollutants for their survival. Phenolic compounds and their derivatives along with dibenzofuran (DBF) are found as dominating pollutants in distillery waste. The acclimatization process leads to selective enrichment of the microbial community; and in this study, we report the acclimatizing effect of phenol on improving the treatment efficiency of two different distillery sludges—sludge from conventional aeration tanks (CAT), and from an extended aeration tank (EAT). The adaptation-dependent performance of activated biomass was studied by monitoring the increase in colony-forming units (CFUs) on mineral media and the utilization pattern for phenol (300×103 and 530×103 CFU for CAT and EAT sludge, respectively) and DBF (260×103 and 430×103 CFU for CAT and EAT sludge, respectively). The study showed that the acclimatization process remarkably improved the performance sludge for treatment of distillery wastewater. There was an improvement in chemical oxygen demand (COD) removal efficiency from 19% (unacclimatized sludge) to 31% in the case of acclimatized sludge (raw wastewater), which improved further to 82% and 87% with dilution of wastewater by 10 times (0.1×) and by 50 times (0.02×), respectively. Highest growth yields were observed with 0.1× wastewater (0.324 and 0.308 g g−1 d−1 for CAT and EAT sludges, respectively), while lower values are reported for the remaining two forms of wastewater. The study proposes that acclimatization step could be included as part of a treatment plant where the activated biomass could be intermittently metabolically charged by exposing it to selected molecules to increase treatment efficiency.  相似文献

The effect of refrigerated and frozen storage on the viability of Vibrio vulnificus was evaluated using cell suspensions (1 × 108 CFU/ml). Ethidium bromide monoazide (EMA) was utilized to selectively allow real-time (Rti) PCR amplification of target DNA from viable but not dead cells. Bacterial survivors from the EMA Rti-PCR were evaluated by comparison with the plate count assay following different temperature exposures (− 20 and 4 °C) every 24 h for 72 h. The log CFU values from the EMA Rti-PCR assays were erroneously higher than that from plate counts. DNA amplification was not completely suppressed by EMA treatment of low temperature destroyed cells suggesting that membrane damage was not sufficient to allow effective EMA penetration into the cells. The optimal concentration of sodium deoxycholate (SD) was also determined to enhance discrimination of viable and dead cells following exposure of cells to low temperatures. The use of 0.01% or less of SD did not inhibit the Rti-PCR amplification derived from viable bacterial cells. A rapid decrease of the log CFU was observed with cell suspensions subjected to frozen storage and a slow decline in the log CFU occurred at 4 °C. The combination of SD and EMA treatments applied to cells of V. vulnificus held at − 20 °C and 4 °C resulted in a high level of correlation between the log of CFU (plate counts) and the log of the number of viable cells determined from SD+EMA Rti-PCR.  相似文献

Laccase-catalyzed oxidation of phenolic compounds in organic media   总被引:1,自引:0,他引:1  
Rhus vernificera laccase-catalyzed oxidation of phenolic compounds, i.e., (+)-catechin, (−)-epicatechin and catechol, was carried out in selected organic solvents to search for the favorable reaction medium. The investigation on reaction parameters showed that optimal laccase activity was obtained in hexane at 30 °C, pH 7.75 for the oxidation of (+)-catechin as well as for (−)-epicatechin, and in toluene at 35 °C, pH 7.25 for the oxidation of catechol. Ea and Q10 values of the biocatalysis in the reaction media of the larger log p solvents like isooctane and hexane were relatively higher than those in the reaction media of lower log p solvents like toluene and dichloromethane. Maximum laccase activity in the organic media was found with 6.5% of buffer as co-solvent. A wider range of 0–28 μg protein/ml in hexane than that of 0–16.7 μg protein/ml in aqueous medium was observed for the linear increasing conversion of (+)-catechin. The kinetic studies revealed that in the presence of isooctane, hexane, toluene and dichloromethane, the Km values were 0.77, 0.97, 0.53 and 2.9 mmol/L for the substrate of (+)-catechin; 0.43, 0.34, 0.14 and 3.4 mmol/L for (−)-epicatechin; 2.9, 1.8, 0.61 and 1.1 mmol/L for catechol, respectively, while the corresponding Vmax values were 2.1 × 10−2, 2.3 × 10−2, 0.65 × 10−2 and 0.71 × 10−2 δA/μg protein min); 1.8 × 10−2, 0.88 × 10−2, 0.19 × 10−2 and 1.0 × 10−2 δA/μg protein min); 0.48 × 10−2, 0.59 × 10−2, 0.67 × 10−2 and 0.54 × 10−2 δA/μg protein min), respectively. FT-IR indicated the formation of probable dimer from (+)-catechin in organic solvent. These results suggest that this laccase has higher catalytic oxidation capacity of phenolic compounds in suitable organic media and favorite oligomers could be obtained.  相似文献

Failure of cervical insemination with cryopreserved semen is hindering implementation of AI in sheep in field condition. Here the effect of equilibration time and catalase on post-thaw qualities of ram semen was investigated. Pooled semen was diluted (800 × 106 sperm mL−1) with a TES-Tris-fructose extender with 6% glycerol, 15% egg yolk and supplemented with 0, 50, 100 and 200 U mL−1 catalase and packaged into 0.25 mL straws. In experiment 1, straws were equilibrated at 5 °C either for 3 h in a cold cabinet (E3) or for 10 (E10) and 22 h (E22) inside a refrigerator. In experiment 2, all straws were equilibrated for 22 h inside refrigerator. Straws were frozen at −25 °C min−1 up to −125 °C using a cell freezer and finally plunged into liquid nitrogen. The post-thaw total and rapid motility were higher (P < 0.05) in E22 compared to E3 and E10. Sperm kinetics was comparable between E3 and E22, but lower in E10. Similarly, acrosome integrity, functional membrane integrity, percent high cholesterol (mCHO) and live-high mitochondrial membrane potential (MMP) were higher (P < 0.05) while live-high intracellular calcium and acrosome-reacted sperm were lower in E22 compared to E3 and E10. The percent rapid motile, high mCHO and live-high MMP were significantly (P < 0.05) lower in catalase-treated samples compared to the control, while the membrane integrity was comparable within the groups. In conclusion, pre-freezing equilibration for 22 h compared to 3 or 10 h resulted in higher post-thaw sperm functions while catalase had negative impact on cryopreservation of ram semen.  相似文献

This study investigated the effects of dietary Bifidobacterium bifidum (BFD) and mannan-oligosaccharide (MOS), as a synbiotic, on the production performance, gut microbiology, serum biochemistry, antioxidant profile and health indices of broiler chicken. Six dietary treatments were T1 (negative control), T2 (positive control-20 mg antibiotic BMD kg−1 diet; BMD: bacitracin methylene disalicylate), T3 (0·1% MOS + 106 CFU BFD per g feed), T4 (0·1% MOS + 107 CFU BFD per g feed), T5 (0·2% MOS + 106 CFU BFD per g feed) and T6 (0·2% MOS + 107 CFU BFD per g feed). Significantly (P < 0·01) better growth performance and efficiency was observed in birds supplemented with 0·2% MOS along with 106 CFU BFD per g of feed compared to BMD and control birds. Supplementation with 0·2% MOS along with either 106 or 107 CFU BFD per g feed reduced (P < 0·01) the gut coliform, Escherichia coli, total plate count, and Clostridium perfringens count and increased the Lactobacillus and Bifidobacterium count. Significantly (P < 0·01) higher serum and liver antioxidant enzyme pool, serum HDL cholesterol and lower serum glucose, triglyceride, total cholesterol, cardiac risk ratio, atherogenic coefficient and atherogenic index of plasma were observed in birds supplemented with 0·2% MOS along with 106 CFU BFD per g of feed compared to control or BMD supplemented birds. Better production performance, gut microbial composition, serum biochemistry, antioxidant profile and health indices were depicted by broiler chicken supplemented with 0·2% MOS and 106 CFU BFD per g of feed.  相似文献

We investigated the intestinal microflora of coastal fish including Takifugu niphobles using both culture techniques and library cloning. As a result, the numbers of bacteria appeared on agar media were 1.0 × 104 to 1.4 × 109 CFU/g (colony forming units/gram), whereas those of total bacteria stained with 4′,6-diamidino-2-phenylindole were 4.7 × 1010 to 1.9 × 1011 cells/gram, irrespective of different fish species. In addition, the culture technique showed that the intestinal microflora in all specimens was mainly composed of the genus Vibrio. In contrast, the direct count method showed that spirochaetes with length of 2.5-4.5 μm were present in the intestinal contents of T. niphobles at high densities, whereas such bacteria could not be detected in those of other fish species. Library cloning yielded the sequences of 16S rRNA genes that were divided into seven taxonomic categories of bacteria including Actinobacteria, Bacilli, Clostridia, Gammaproteobacteria, Mollicutes, Spirochaetes and an unclassified bacterial group. These results demonstrate that the molecular diversity of the intestinal bacteria in T. niphobles based on the clone library method reflects the direct observation by fluorescence microscopy to some extent.  相似文献

Aims: The purposes of this study were to evaluate the efficacy of high pressure to inactivate Escherichia coli O157:H7 in ground beef at ambient and subzero treatment temperatures and to study the fate of surviving bacteria postprocess and during frozen storage. Methods and Results: Fresh ground beef was inoculated with a five‐strain cocktail of E. coli O157:H7 vacuum‐packaged, pressure‐treated at 400 MPa for 10 min at ?5 or 20°C and stored at ?20 or 4°C for 5–30 days. A 3‐log CFU g?1 reduction of E. coli O157:H7 in the initial inoculum of 1 × 106 CFU g?1 was observed immediately after pressure treatment at 20°C. During frozen storage, levels of E. coli O157:H7 declined to <1 × 102 CFU g?1 after 5 days. The physiological status of the surviving E. coli was affected by high pressure, sensitizing the cells to pH levels 3 and 4, bile salts at 5% and 10% and mild cooking temperatures of 55–65°C. Conclusions: High‐pressure processing (HPP) reduced E. coli O157:H7 in ground beef by 3 log CFU g?1 and caused substantial sublethal injury resulting in further log reductions of bacteria during frozen storage. Significance and Impact of the Study: HPP treatment of packaged ground beef has potential in the meat industry for postprocess control of pathogens such as E. coli O157:H7 with enhanced safety of the product.  相似文献

Collaborative research was conducted at the USDA-ARS Subtropical Agricultural Research Center in southern Texas to assess the microbial control potential of Beauveria bassiana and Paecilomyces fumosoroseus against Bemisia whiteflies. Laboratory assays demonstrated the capacity of both pathogens to infect Bemisia argentifolii nymphs on excised hibiscus leaves incubated at relative humidities as low as 25% at 23 ± 2°C (ca. 35% infection by B. bassiana and P. fumosoroseus resulted from applications of 0.6–1.4 × 103 conidia/mm2 of leaf surface). In small-scale field trials using portable air-assist sprayers, applications at a high rate of 5 × 1013 conidia in 180 liters water/ha produced conidial densities of ca. 1–2.5 × 103 conidia/mm2 on the lower surfaces of cucurbit leaves. Multiple applications of one isolate of P. fumosoroseus and four isolates of B. bassiana made at this rate at 4- to 5-day intervals provided >90% control of large (third- and fourth-instar) nymphs on cucumbers and cantaloupe melons. The same rate applied at 7-day intervals also provided >90% control in zucchini squash, and a one-fourth rate (1.25 × 1013 conidia/ha) applied at 4- to 5-day intervals reduced numbers of large nymphs by >85% in cantaloupe melons. In contrast to the high efficacy of the fungal applications against nymphs, effects against adult whiteflies were minimal. The results indicated that both B. bassiana and P. fumosoroseus have strong potential for microbial control of nymphal whiteflies infesting cucurbit crops.  相似文献

Six moorings were deployed at different locations in the deep submarine canyons along the south–west margin of Crete, providing a total of eight sediment-trap time series from June 2005 to May 2006. Within this dataset, we analyzed the record from intact coccospheres, which represent the signal of export production from the coccolithophore community. The most abundant species at all stations during the whole investigated period were E. huxleyi and A. robusta, followed by S. pulchra HET, G. flabellatus, H. carteri, F. profunda, S. pulchra HOL oblonga, while the rest of the species represented ≤ 1% of the assemblage. Overall the assemblage composition was comparable at all stations, with slight variations mostly related to the different preservation of coccosphere integrity at the different collection depths. The consistent pattern of seasonal variation in species distribution and total coccolithophore export allowed us to define the occurrence of three main periods: a) March to June, with high overall coccosphere flux (up to 4.3 × 105–3.4 × 106 coccospheres m− 2 day− 1), increased abundance of E. huxleyi and subordinate H. carteri s.s., Umbilicosphaera spp. and S. pulchra; b) June to November, with high but gradually decreasing total coccosphere flux (up to 7 × 105–1.4 × 106 coccospheres m− 2 day− 1) and high relative abundance of the deep photic zone species A. robusta, F. profunda, G. flabellatus as well as S. pulchra and Coronosphaera spp., R. clavigera, U. tenuis, D. tubifera and holococcolithophores; c) November to February, with low overall export fluxes (3.5–9 × 104 coccospheres m− 2 day− 1) and high relative abundance of A. robusta, S. pulchra and Syracosphaera spp. These three periods correspond to the seasonal changes in sea surface temperature, surface mixed layer depth and rainfall and are associated with varying total surface primary production, as detected through remote sensing in the surface waters.  相似文献

As part of a 3-fold approach to select potential mycoinsecticides for whitefly control, we evaluated infectivity, thermal requirements, and toxicogenic activity of the entomopathogenic fungus Beauveria bassiana (Ascomycota: Clavicipitaceae) under laboratory conditions. Twenty-five native B. bassiana isolates and a commercially available mycoinsecticide (based on B. bassiana) were evaluated for virulence to fourth instar nymphs of sweetpotato whitefly, Bemisia tabaci, and greenhouse whitefly, Trialeurodes vaporariorum, at a concentration of 1 × 107 conidia/ml. All isolates were pathogenic for both whitefly species, whereas mortality rates varied from 3 to 85%. A second series of bioassays was conducted on 10 selected isolates using four 10-fold concentrations ranging from 1 × 105 to 1 × 108 conidia/ml. Median lethal concentrations (LC50) of the four most virulent isolates varied from 1.1 × 105 to 6.2 × 106 conidia/ml and average survival time (AST) of treated nymphs from 5.9 to 7.4 days. T. vaporariorum were significantly more susceptible to all B. bassiana isolates than B. tabaci. The thermal biology of the eight most virulent isolates to both whitefly species was investigated at six temperatures (10–35 °C). The colony radial growth rate was estimated from the slope of the linear regression of colony radius on time and data were then fitted to a modified generalized β function that accounted for 90.5–99.3% of the data variance. Optimum temperatures for extension rate ranged from 23.1 to 27.1 °C, whereas maximum temperatures for fungal growth varied from 31.8 to 36.6 °C. On the basis of their virulence and thermal requirements, three isolates showed promise as candidates for whitefly management in Mediterranean greenhouses. Whilst in vitro production of macromolecular compounds toxic to Galleria mellonella larvae was not a requisite for virulence, ASTs of larvae injected with Sephadex G-25 fractions from candidate isolates ranged from 1.4 to 3.7 days compared with 5–6 days for non-toxic G-25 fractions. In addition, proteinase K treatment significantly reduced their toxic activity suggesting that they were proteins and revealing the potential of these isolates to be further improved through biotechnology to kill the pest more quickly.  相似文献

This study determined the pathogenicity and virulence of Beauveria bassiana and Metarhizium anisopliae to eggs of the chinch bug Blissus antillus (Hemiptera: Lygaeidae). Eggs were inoculated under laboratory conditions by immersion in concentrations of 1 × 104 and 5 × 106 conidia/ml. Inoculated eggs were kept under controlled conditions. Evaluations were carried out daily for 20 days. M. anisopliae isolates were highly virulent to eggs, even at 1 × 104 conidia/ml. All B. bassiana isolates tested were considered to be of low virulence or avirulent. The most virulent isolate tested was ESALQ 818 (M. anisopliae), which caused 96.7% infection, when eggs were immersed in suspensions of 1 × 104 conidia/ml. Conidial production on infected eggs was observed to be highest for M. anisopliae isolate CG144, with a mean value of 11.6 × 105 conidia/ml/egg. Infection of Blissus eggs oviposited on plant stems was greater when M. anisopliae isolate CG144 was formulated in mineral oil (63.5% mortality) than when formulated in Tween 80 (27.1% mortality).  相似文献

Cryopreservation has become an integral component of any cell transplantation technique helping to overcome the issues associated with known spatial and temporal barriers between donor and recipient. The aim of this study was to develop a protocol for large quantity cryopreservation of bovine testicular germ cells. The impact of 3 different packaging methods (5 ml semen straw, 20 ml freezing bag and 1.5 ml cryovial) and varying cell densities (3 × 106, 9 × 106, or 18 × 106 cells/ml) on the survival of testis germ cells was examined. Cells processed in 5 ml semen straws had a significantly higher viability (70.7 ± 1.2%, P < 0.05) compared to those cells in 20 ml freezing bags (46.7 ± 0.1%) or 1.5 ml cryovials (46.3 ± 2.2%). For 5 ml straws, a 20 min cooling prior to cryopreservation resulted in a higher post thaw viability (73.2 ± 0.6%) than a 10 min cooling (56.0 ± 2.2%), while the density of the cell suspension did not impact on post thaw viability. Thus cryopreservation of testicular germ cells in 5 ml straws at a density between 3 × 106 and 18 × 106 cells/ml in liquid nitrogen vapour for 20 min cooling appears to be a simple and practical way to preserve cells. Subsequent testing of frozen/thawed cells exhibited viable cultures and retained the ability to proliferate. The freezing protocol does not preferentially preserve type A spermatogonia. However, the cell surface properties of somatic cells appear to be affected by the freezing procedure and therefore the frozen/thawed cells are less suitable for enriching type A spermatogonia by differential plating.  相似文献

Seasonal changes in plasma testosterone concentration and semen quality were evaluated in yak bulls throughout a 1-year period. Blood samples were collected every week from adult yak bulls (n = 15). These blood samples were analyzed for testosterone using a highly sensitive enzyme-linked immunoassay. Ejaculates were collected from five representative bulls each week. Ejaculate volume, progressive motility, live sperm count and sperm concentrations were determined. Mean testosterone in plasma was 1.03 ± 0.25 ng/ml. Concentrations of testosterone changed throughout the year (P < 0.05) and were found to be highest during the winter. It was also higher during the autumn than in summer and spring (P < 0.05). Mean ejaculate volume, progressive motility, live sperm count and spermatozoa concentration were 2.7 ± 0.3 ml, 72.8 ± 1.4%, 82.3 ± 0.9% and 968 ± 233 × 106 ml−1, respectively. Ejaculate volume and sperm concentration were higher (P < 0.05) in autumn than in other seasons. To conclude, a highly sensitive EIA for testosterone was developed and validated for yak plasma. Seasonal changes in semen quality were associated with changes in the concentration of testosterone in plasma from yak bulls.  相似文献

Dietary chitosans with different molecular weight Mw and the degree of deacetylation DDA (high molecular weight chitosan HCS with Mw 7.60 × 105 and DDA 85.5%, middle molecular weight chitosan MCS with Mw 3.27 × 104 and DDA 85.2%, chito-oligomer COS with Mw 0.99 × 103 and DDA 85.7% and water-soluble chitosan WSC with Mw 3.91 × 104 and DDA 52.6%) were used at the 1.05% level to feed mice for 90 days. Afterwards no pathological symptoms, clinical signs or deaths were observed. The body weight of mice in chitosan group and control group showed no significant difference. Although HCS, COS and WSC had no significant effect on the level of Fe, Zn and Cu in the tested mice’s liver, spleen, heart and kidney, MCS significantly increased the level of Fe, Zn and Cu in liver. Therefore dietary ingestion of chitosan did not depress the level of Fe, Zn and Cu in mice.  相似文献
