运用致病毒素筛选抗稻瘟病细胞突变体   总被引:29,自引:0,他引:29

本文研究报道了抗稻瘟病细胞突变体筛选方法、程序与结果,并对有关问题进行了讨论。毒素对水稻成熟胚愈伤组织的诱导与分化有明显的抑制作用,抑制程度随毒素浓度提高和处理时间延长而加强,但抗感品种之间差异显著。以含毒素的诱导培养基或诱导与分化培养基均含毒素的筛选处理效果较好,而且简便易行。5年来接种筛选了29个品种25000多个外植体,在已鉴定的349个R_1个体中,有183株表现抗病。经多次接种鉴定与选择,已获得12个高抗稻瘟病的株系和1个抗病丰产品系,并对部分抗病突变体的抗性基因进行了遗传分析。  相似文献

 A recombinant inbred line derived from a cross between CO39 and ‘Moroberekan’, RIL276, was found to be resistant to lineage 44 isolates of Pyricularia grisea in the Philippines. One hundred F2 individuals were obtained from a backcross of RIL276 and CO39. Phenotypic analysis showed that RIL276 carries a single locus, tentatively named Pi44(t), conferring complete resistance to lineage 44 isolates of P. grisea. RFLP probes, STS primers and AFLP markers were applied to identify DNA markers linked to Pi44(t). Neither RFLP nor STS-PCR analysis gave rise to DNA markers linked to the locus. Using bulk segregant AFLP analysis, however, two dominant AFLP markers (AF348 and AF349) linked to Pi44(t) were identified. AF349 and AF348 were located at 3.3±1.5 cM and 11±3.5 cM from Pi44(t), respectively. These markers were mapped on chromosome 11 using an F2 population derived from a cross between ‘Labelle’ and ‘Black Gora’. The location of AF348 on chromosome 11 was confirmed using another F2 mapping population derived from IR40931-26-3-3-5/ PI543851. DNA products at the loci linked to Pi44(t) were amplified from RIL276, ‘Labelle’ and PI543851 using the same primer pairs used to amplify AF349 and AF348. Sequence analysis of these bands showed 100% identity between lines. Transfer of a grapevine stilbene synthase gene to rice (Oryza sativa L.)   总被引:17,自引:0,他引:17  
A gene derived from grapevine (Vitis vinifera) coding for stilbene synthase has been transferred into protoplasts of the commercially important japonica rice cultivar Nipponbare using PEG-mediated direct gene transfer. Transgenic plants were regenerated from calli selected on kanamycin. Southern blot analysis of genomic DNA isolated from regenerants and progeny plants demonstrated that the stilbene synthase gene is stably integrated in the genome of transgenic rice plants and inherited in the offspring. The transient formation of stilbene-synthase-specific mRNA shortly after inoculation with the fungus of the rice blast Pyricularia oryzae has demonstrated that the grapevine stilbene synthase promoter is also active in monocotyledonous plants. 以ZA_(15)和ZB_(11)等2个致病小种对6个抗稻瘟病体细胞突变体进行了抗性遗传分析。结果表明,86-S1、88-86、88-42和88-40等4个突变体对ZA_15、ZB_(11)小种的抗性分别由1个显性基因控制,同时这2个抗性基因还存在紧密的连锁关系。88-127和88-145对ZA_15的抗病性受2个重复显性基因控制;而对ZB_(11)的抗性则分别受2个互补显性基因和1个显性基因控制。等位性测定表明,86-S1、88-42和8B-86等3个突变体具有的抗性基因是等位的,可能是Pi-(?)或与之等位的抗性基因。  相似文献

冈46B(G46B)是水稻生产应用中的一个农艺性状十分优良的保持系,其主要的缺陷是稻瘟病抗性较弱,通过对地谷,BL-1,Pi-4号等三个分别含抗病基因Pi-d(t)^1、Pi-b、Pi-tα^2的稻瘟病抗性材料与G46B聚合杂交,并利用抗病基因连锁的分子标记对杂交后代进行辅助选择,在聚合杂交的F2代及B1C1代群体中共获得了15株含Pi-d(t)^1、Pi-b、Pi-tα^2等三个抗稻瘟病基因的材料,其可能的基因型分别为:三基因杂合体Pi-d(t)^1pi-d(t)^1,Pi-bpi-b/Pi-tα^2 pi-tα^2 4株,双基因杂合体10株,其中Pi-d(t)^1Pi-d(t)^1/Pi-bpi-b/Pi-tα^2pi-tα^2 6株,Pi-d(t)^1pi-d(t)^1/Pi-bpi-b/Pi-tα^2Pi-tα^2 3株,Pi-d(t)^1pi-d(t)^1,Pi-bPi-6,Pi-tα^2 pi-tα^2 1株,双基因纯合体Pi-d(t)^1Pi-d(t)^1/Pi-bpi-b/Pi-tα^2Pi-tα^2仅1株,这一研究结果为进一步改良G46B的稻瘟病抗性奠定了基础,同时这一研究结果表明利用分子标记可快速、有效地实现多个抗病基因的聚合,大大提高水稻抗病育种的效率。  相似文献

文章通过对所构建的水稻突变体库进行大规模筛选,获得一个稳定遗传的矮秆突变体,与野生型日本晴相比,该突变体表现为植株矮化、叶片卷曲、分蘖减少和不育等性状,命名为dtl1(dwarf and twist leaf 1)。dtl1属于nl型矮秆,激素检测表明,矮秆性状与赤霉素和油菜素内酯无关。遗传分析显示,突变性状受单一隐性核基因控制。利用dtl1与籼稻品种Taichung Native 1杂交构建F2群体,将该突变基因DTL1定位于水稻第10染色体长臂2个SSR标记RM25923和RM6673之间约70.4 kb区域内,并与InDel标记Z10-29共分离,在该区域预测有13个候选基因,但未见调控水稻株高相关基因的报道,因此,认为DTL1基因是一个新的控制水稻株高的基因。  相似文献

Field resistance is defined as the resistance that allows effective control of a parasite under natural field conditions and is durable when exposed to new races of that parasite. To identify the genes for field resistance to rice blast, quantitative trait loci (QTLs) conferring field resistance to rice blast in Japanese upland rice were detected and mapped using RFLP and SSR markers. QTL analysis was carried out in F4 progeny lines from the cross between Nipponbare (moderately susceptible, lowland) and Owarihatamochi (resistant, upland). Two QTLs were detected on chromosome 4 and one QTL was detected on each of chromosomes 9 and 12. The phenotypic variation explained by each QTL ranged from 7.9 to 45.7% and the four QTLs explained 66.3% of the total phenotypic variation. Backcrossed progeny lines were developed to transfer the QTL with largest effect using the susceptible cultivar Aichiasahi as a recurrent parent. Among 82 F3 lines derived from the backcross, resistance segregated in the expected ratio of resistant 1 : heterozygous 2 : susceptible 1. The average score for blast resistance measured in the field was 4.2 ± 0.67, 7.5 ± 0.51and 8.2 ± 0.66, for resistant, heterozygous and susceptible groups, respectively. The resistance gene, designated pi21, was mapped on chromosome 4 as a single recessive gene between RFLP marker loci G271 and G317 at a distance of 5.0 cM and 8.5 cM, respectively. The relationship to previously reported major genes and QTLs conferring resistance to blasts, and the significance of marker-assisted selection to improve field resistance, are discussed. To understand the development of rice leaf blades,we identified a new rolled-leaf mutant,w32,from indica cultivar IR64 through EMS mutagenesis. The mutant showed a stable rolled-leaf phenotype throughout the life cycle. Two F2 populations were developed by crossing w32 to cultivar IR24 and PA64. Genetic analysis showed that the rolled-leaf phenotype was controlled by a single recessive gene. A rice semidwarfing gene, sd-1, known as the "green revolution gene," was isolated by positional cloning and revealed to encode gibberellin 20-oxidase, the key enzyme in the gibberellin biosynthesis pathway. Analysis of 3477 segregants using several PCR-based marker technologies, including cleaved amplified polymorphic sequence, derived-CAPS, and single nucleotide polymorphisms revealed 1 ORF in a 6-kb candidate interval. Normal-type rice cultivars have an identical sequence in this region, consisting of 3 exons (558, 318, and 291 bp) and 2 introns (105 and 1471 bp). Dee-Geo-Woo-Gen-type sd-1 mutants have a 383-bp deletion from the genome (278-bp deletion from the expressed sequence), from the middle of exon 1 to upstream of exon 2, including a 105-bp intron, resulting in a frame-shift that produces a termination codon after the deletion site. The radiation-induced sd-1 mutant Calrose 76 has a 1-bp substitution in exon 2, causing an amino acid substitution (Leu [CTC] to Phe [TTC]). Expression analysis suggests the existence of at least one more locus of gibberellin 20-oxidase which may prevent severe dwarfism from developing in sd-1 mutants.  相似文献

02428h是从半矮秆材料02428体细胞培养后代中发现的隐性高秆突变体, 其株高性状由1对长穗颈基因eui和1对半矮秆基因sd-1共同控制。以02428h与半矮秆材料南京11杂交的F2为作图群体, 利用Gramene公布的SSR标记和根据NCBI中的BAC序列自己新开发的SSR标记, 将eui基因定位在第5染色体上的RM3673和RM0012之间, 两侧遗传距离分别为0.3 cM和1.0 cM, 为该基因的分子标记辅助选择奠定了基础。  相似文献

综述了近年来国际上研究水稻化感作用的新进展,比较分析了当前常用于室内评价水稻化感作用潜力的几种生物测试法的优缺点,指出了琼脂迟播共培法是较为理想的室内生物测试法并已广泛应用于化感作用研究中。在此基础上,分析了水稻化感作用的数量遗传特性及其QTL定位的研究现状;阐明了水稻化感作用的遗传多样性及其分子生态特性;并就当前普遍关注的焦点问题:逆境条件(如低氮或高伴生杂草密度胁迫)常引起水稻化感作用潜力增大的生理过程与分子机制作了阐述。结合近年来应用差异蛋白组学和生物信息学的研究实例,阐明了逆境引起水稻化感作用增强与其酚类合成代谢相关酶蛋白表达丰度增加,萜类合成代谢相关酶蛋白表达丰度下降有关。就究竟什么是水稻的化感物质及其作用方式等问题作了分析与讨论.指出水稻的化感抑草效应是其众多化感物质综合作用的结果,应重视区分化感物质对靶标杂草的原生作用和进入土壤生态系统中经生物转化后的次生作用。根据当前植物化感作用研究的发展趋势,阐明了进一步研究水稻化感作用的焦点问题,提出了水稻化感作用的根际生物学特性与分子生态学机制,是未来国际上竞相角逐的重点研究领域,并认为以现代系统生物学理论为指导,运用基因组学、蛋白质组学和代谢组学等技术方法,是揭示这一分子生态学过程与机制的重要技术选择和优先研究领域。  相似文献

A rice mutant,G069, characteristic of few tiller numbers, was found in anther culture progeny from theF 1 hybrid between anindica-japonica cross, Gui630×02428. The mutant has another two major features: delayed tillering development and yellowing apex and margin on the mature leaves. As a donor parent,G069 was further backcrossed with the recurrent parent,02428, for two turns to develop aBC 2F2 population. Genetic analysis in theBC 2F2 population showed that the traits of few-tillering and yellowing apex and margin on the mature leaves were controlled by one recessive gene. A pool of equally mixed genomic DNA, from few-tillering individual plants inBC 2F2, was constructed to screen polymorphism with simple sequence repeat (SSR) markers in comparison with the02428 genome. One SSR marker and three restriction fragment length polymorphism (RFLP) markers were found possibly linked with the recessive gene. By using these markers, the gene of few-tillering was mapped on chromosome 2 between RFLP marker C424 and S13984 with a genetic distance of 2.4 cM and 0.6 cM, respectively. Rice blast, caused by Magnaporthe oryzae (synonym: Pyricularia oryzae), severely reduces rice production and grain quality. The molecular mechanism of rice resistance to M. oryzae is not fully understood. In this study, we identified a chaperone DnaJ protein, OsDjA6, which is involved in basal resistance to M. oryzae in rice. The OsDjA6 protein is distributed in the entire rice cell. The expression of OsDjA6 is significantly induced in rice after infection with a compatible isolate. Silencing of OsDjA6 in transgenic rice enhances resistance to M. oryzae and also results in an increased burst of reactive oxygen species after flg22 and chitin treatments. In addition, the expression levels of WRKY45, NPR1 and PR5 are increased in OsDjA6 RNAi plants, indicating that OsDjA6 may mediate resistance by affecting the salicylic acid pathway. Finally, we found that OsDjA6 interacts directly with the E3 ligase OsZFP1 in vitro and in vivo. These results suggest that the DnaJ protein OsDjA6 negatively regulates rice innate immunity, probably via the ubiquitination proteasome degradation pathway.  相似文献

Bacillus thuringiensis Cry1Ba endotoxin, which was shown to exhibit a tenfold lower lethal concentration 50 (LC50) than Cry1Ac in a Striped Stem Borer (SSB) diet incorporation assay. The 1.950-bp synthetic cry1B gene, possessing an overall GC content of 58 %, was cloned under the control of the maize ubiquitin promoter first intron and first exon regions. The resulting vector, designated as pUbi-cry1B, was transferred to two commercial Mediterranean cultivars of rice, Ariete and Senia, using microprojectile acceleration-mediated transformation. Thirty-two and 47 T0 events were generated in cvs. Ariete and Senia, respectively. Southern blot and immunoblot analyses allowed the identification of 7 Senia and 1 Ariete events harbouring both an intact gene cassette and expressing Cry1B at a level ranging from 0.01% to 0.4% of the total soluble proteins. Three Senia and 1 Ariete events were found to be protected against second instar SSB larvae in whole plant feeding assays, exhibiting 90–100% mortality 7 days after infestation. Spatial and temporal variation in transgene expression was further examined in resistant event 64 of cv. Ariete. Stable accumulation of Cry1B, representing 0.4% of the total soluble proteins, was observed over the T2 to T4 generations in leaf tissue 20, 40, 70 and 90 days after germination in both young and old leaves and in internodes. Ariete event 64 was found to be fully protected from attacks of third and fourth instar SSB larvae over subsequent generations. A novel zebra mutant, zebra-15, derived from the restorer line JinhuilO (Oryza sativa L. ssp. indica) treated by EMS, displayed a distinctive zebra leaf from seedling stage to jointing stage. Its chlorophyll content decreased (55.4%) and the ratio of Chla/Chlb increased (90.2%) significantly in the yellow part of the zebra-15, compared with the wild type. Net photosynthetic rate and fluorescence kinetic parameters showed that the decrease of chlorophyll content significantly influenced the photosynthetic efficiency of the mutant. Genetic analysis of F2 segregation populations derived from the cross of XinonglA and zebra-15 indicated that the zebra leaf trait is controlled by a single recessive nuclear gene. Ninety-eight out of four hundred and eighty pairs of SSR markers showed the diversity between the XinonglA and the zebra-15, their F2 population was then used for gene mapping. Zebra-15 (Z-15) gene was primarily restricted on the short arm of chromosome 5 by 150 F2 recessive individuals, 19.6 cM from marker RM3322 and 6.0 cM from marker RM6082. Thirty-six SSR markers were newly designed in the restricted location, and the Z-15 was finally located between markers nSSR516 and nSSR502 with the physical region 258 kb by using 1,054 F2 recessive individuals.  相似文献

Ac/Ds插入突变是水稻基因功能鉴定的有力工具之一。文章从水稻中花11 Ds-T-DNA转化纯合体与Ac-T-DNA 转化纯合体的杂交群体中筛选到一个淡绿叶突变体。该突变体在三叶期由绿苗转为淡绿叶苗, 自然光照下突变体迅速焦枯, 但是在弱光照条件下, 突变体能缓慢生长至开花结实; 突变体光合作用特性研究表明该突变是典型的光抑制突变体。遗传分析表明该突变为Ds插入导致的隐性突变。  相似文献
