In this article we explore the evolutionary history of a functional complex at the molecular level in plethodontid salamanders. The complex consists of a proteinaceous courtship pheromone, a pheromone-producing gland on the male's chin, and a set of behaviors for delivering the pheromone to the female. Long-term evolutionary stasis is the defining feature of this complex at both the morphological and behavioral levels. However, our previous assessment of the pheromone gene, plethodontid receptivity factor (PRF), revealed rapid evolution at the molecular level despite stasis at higher levels of organization. Analysis of a second pheromone gene, sodefrin precursor-like factor (SPF), now indicates that evolutionary decoupling in this complex is pervasive. The evolutionary profiles of SPF and PRF are remarkably similar in that: (a) both genes exhibit high levels of sequence diversity both within and across taxa, (b) genetic diversity has been driven by strong positive selection, and (c) the genes have evolved heterogeneously in different salamander lineages. The composition of the pheromone signal as a whole, however, has experienced an extraordinary evolutionary transition. Whereas SPF has been retained throughout the 100 MY radiation of salamanders, PRF has only recently been recruited to a pheromone function (27 million years ago). When SPF and PRF coexist in the same clade, they show contrasting patterns of evolution. When one shows rapid evolution driven by positive selection, the other shows neutral divergence restrained by purifying selection. In one clade, the origin and subsequent rapid evolution of PRF appear to have interfered with the evolution and persistence of SPF, leading to a pattern of evolutionary replacement. Overall, these two pheromone genes provide a revealing window on the dynamics that drive the evolution of multiple traits in a signaling complex.  相似文献

Sexual communication in plethodontid salamanders is mediated by a proteinaceous pheromone that a male delivers to a female during courtship, boosting her receptivity. The pheromone consists of three proteins from three unrelated protein families. These proteins are among a small group of pheromones known to affect female receptivity in vertebrates. Previously, we showed that the genes of two of these proteins (PRF and SPF) are prone to incessant evolution driven by positive selection, presumably as a consequence of coevolution with female receptors. In this report, we focus on the evolution of the third pheromone protein gene family, plethodontid modulating factor (PMF), to determine whether it shows the same pattern of diversification. We used RT-PCR in mental gland cDNA to survey PMF sequences from three genera of plethodontid salamanders (27 spp.) to measure rates of evolution, level of gene diversification, modes of selection, and types of amino acid substitution. Like PRF and SPF, PMF is produced by a multigene family characterized by gene duplication and high levels of polymorphism. PMF evolution is rapid, incessant, and driven by positive selection. PMF is more extreme in these dimensions than both PRF and SPF. Nestled within this extraordinary variation, however, is a signature of purifying selection, acting to preserve important structural and biochemical features of the PMF protein (i.e., secretion signal, cysteine residues, and pI). Although a pattern of persistent diversification exists at the molecular level, the morphological and behavioral aspects of the pheromone delivery system show evolutionary stasis over millions of years.  相似文献

Functionally equivalent genes may evolve heterogeneously across closely related taxa as a consequence of lineage-specific selective pressures. Such disparate evolutionary modes are especially prevalent in genes that encode postcopulatory reproductive proteins, presumably as a result of sexual selection. We might therefore expect genes that mediate reproduction prior to insemination to evolve in a similar manner. Plethodontid receptivity factor (PRF), a proteinaceous salamander pheromone produced by the male, increases female receptivity during courtship interactions. To test for lineage-specific differences in PRF's evolution, we intensively sampled PRF genes across the eastern Plethodon phylogeny (27 spp.; 34 populations) to compare gene diversification, rates of evolution, modes of selection, and types of amino acid substitution. Our analyses indicate that PRF evolutionary dynamics vary considerably from lineage to lineage. Underlying this heterogeneity, however, are two well-defined transitions in evolutionary mode. The first mode is representative of a typical protein profile, wherein neutral divergence and purifying selection are the dominant features. The second mode is characterized by incessant, cyclical evolution driven by positive selection. In this mode, the positively selected sites are bound by a limited assortment of acceptable amino acids that appear to evolve independently of other sites, resulting in a tremendous number of unique PRF alleles. Several of these selected sites are implicated in receptor binding. These sites are apparently involved in a molecular tango in which the male signal and female receptors coevolve within a confined molecular space. PRF's lineage-specific evolutionary dynamics, in combination with evidence of a molecular tango, highlight the molecular action of sexual selection on a chemical signal that is used during courtship.  相似文献

Pheromones are important chemical signals for many vertebrates, particularly during reproductive interactions. In the terrestrial salamander Plethodon shermani, a male delivers proteinaceous pheromones to the female as part of their ritualistic courtship behavior. These pheromones increase the female's receptivity to mating, as shown by a reduction in courtship duration. One pheromone component in particular is plethodontid receptivity factor (PRF), a 22-kDa protein with multiple isoforms. This protein alone can act as a courtship pheromone that causes the female to be more receptive. We used a bacterial expression system to synthesize a single recombinant isoform of PRF. The recombinant protein was identical to the native PRF, based on mass spectrometry, circular dichroism spectra, and a behavioral bioassay that tested the effects of recombinant PRF (rPRF) on female receptivity (21% reduction in courtship duration). The rPRF appears to mimic the activity of a mixture of PRF isoforms, as well as a mixture of multiple different proteins that comprise the male courtship gland extract. Pheromones that are peptides have been characterized for some vertebrates; to date, however, rPRF is one of only 2 synthesized vertebrate proteins to retain full biological activity.  相似文献

In this study, I take a new approach to modeling the evolutionary constraint of protein sequence, introducing the stabilizing selection of protein function into the nearly-neutral theory. In other words, protein function under stabilizing selection generates the evolutionary conservation at the sequence level. With the help of random mutational effects of nucleotides on protein function, I have derived the distribution of selection coefficient among sites, called the S-distribution whose parameters have clear biological interpretations. Moreover, I have studied the inverse relationship between the evolutionary rate and the effective population size, showing that the number of molecular phenotypes of protein function, i.e., independent components in the fitness of the organism, may play a key role for the molecular clock under the nearly-neutral theory. These results are helpful for having a better understanding of the underlying evolutionary mechanism of protein sequences, as well as human disease-related mutations.  相似文献

Despite the considerable amount of interest in phylogeny reconstruction, patterns of homoplasy in morphological and behavioral data have received only limited attention to date, whereas the patterns of homoplasy in molecular data are relatively well understood. First, because the number of alternative molecular character states is strictly limited (particularly for nucleotide sequence data), higher rates of substitution generate higher levels of homoplasy. Second, depending on the relative proportions of constrained and unconstrained sites, each molecular data set has a time frame of applicability outside of which resolution becomes ambiguous. There is good evidence to suggest that numbers of alternative character states for morphological and even behavioral data may be similarly limited and that higher rates of evolution are often linked to higher rates of homoplasy. Like molecular data sets, morphological and behavioral data sets contain rapidly evolving characters as well as more conservative elements. Morphologies and behaviors related to sexual recognition and reproduction show low levels of intraspecific variation, but high levels of lability between species, making them crucial for species identification but often poor as markers of relationship at greater time depths. The organization theory of speciation derived by Carson is a model based on genome dynamics, and it predicts exactly this window of applicability for characters related to sexual reproduction. Nonsexual characters related to environmental adaptation should be applicable at greater phylogenetic depths. A better understanding of patterns of homoplasy enables a more sophisticated approach to the assessment of the relative reliabilities of alternative tree topologies.  相似文献

The effects of interaction of artificial directional and stabilizing selection were studied using a recessive mutation of Drosophila melanogaster radius incompletus which causes interruption of the second longitudinal wing vein (L2). The character under directional selection was the length of the proximal segment of L2; stabilizing selection was conducted for a complex of morphometric wing characters. Three experimental designs in three replications were used: 1) directional and stabilizing selection; 2) directional selection; 3) unselected controls. The dynamics and variation of the character under directional selection was shown to be different in the stabilized and unstabilized lines. Coefficient of variation which increased in the unstabilized lines remained at the same level in the stabilized lines and in the controls. The results are in agreement with those of the previous experiment, where directional selection was conducted for a behavioral trait, which suggests general character of the relationships observed.  相似文献

The extent and pattern of protein and DNA polymorphisms are discussed with emphasis on the mechanism of maintenance of the polymorphisms. Statistical studies suggest that a large proportion of genetic variability at the molecular level is maintained by a mutation-drift balance. At some loci, such as those for histocompatibility in mammals, however, a form of overdominant selection seems to be involved. In the presence of overdominant selection, polymorphic alleles may be maintained for tens of millions of years, so that the number of nucleotide differences between alleles is often very large, as in the case of self-incompatibility alleles in plants. There are also an increasing number of examples in which an adaptive change of a morphological or physiological character is caused by a single nucleotide substitution. Nevertheless, these mutations seem to be a small proportion of the total nucleotide changes that contribute to genetic variability and evolution. Although there are many examples of frequency-dependent selection, this form of selection is apparently unimportant for the maintenance of genetic variability except in some special cases. Observations on the evolutionary change of DNA suggest that the driving force of evolution is mutation rather than selection.  相似文献

Empirical studies show that lineages typically exhibit long periods of evolutionary stasis and that relative levels of within‐species trait covariance often correlate with the extent of between‐species trait divergence. These observations have been interpreted by some as evidence of genetic constraints persisting for long periods of time. However, an alternative explanation is that both intra‐ and interspecific variation are shaped by the features of the adaptive landscape (e.g., stabilizing selection). Employing a genus of insects that are diverse with respect to a suite of secondary sex traits, we related data describing nonlinear phenotypic (sexual) selection to intraspecific trait covariances and macroevolutionary divergence. We found support for two key predictions (1) that intraspecific trait covariation would be aligned with stabilizing selection and (2) that there would be restricted macroevolutionary divergence in the direction of stabilizing selection. The observed alignment of all three matrices offers a point of caution in interpreting standing variability as metrics of evolutionary constraint. Our results also illustrate the power of sexual selection for determining variation observed at both short and long timescales and account for the apparently slow evolution of some secondary sex characters in this lineage.  相似文献

It is widely believed that behavior is more evolutionarily labile and/or more difficult to characterize than morphology, and thus that behavioral characters are not as useful as morphological characters for estimating phylogenetic relationships. To examine the relative utility of behavior and morphology for estimating phylogeny, we compared levels of homoplasy for morphological and behavioral characters that have been used in systematic studies. In an analysis of 22 data sets that contained both morphological and behavioral characters we found no significant difference between mean consistency indices (CIs, which measure homoplasy) within data sets for the two types of characters. In a second analysis we compared overall CIs for 8 data sets comprised entirely of behavioral characters with overall CIs for 32 morphological data sets and found no significant difference between the two types of data sets. For both analyses, 95% confidence limits on the difference between the two types of characters indicate that, even if given the benefit of the doubt, morphological characters could not have substantially higher mean CIs than behavioral characters. These results do not support the idea that behavioral characters are less useful than morphological characters for the estimation of phylogeny.  相似文献

Summary The extent of correlation was estimated between isozyme genotypes and the four widely segregating characters — leaf segment W/L ratio, stigma exsertion, fruit weight, and seed weight — in the first backcross of F1 Lycopersicon esculentum x Solanum pennellii to the former parent. The inbred parents differ in their alleles at the 12 tested isozymic loci, which are known to mark a minimum of eight of the twelve tomato chromosomes. Based on the isozyme data, a mean heterozygosity value, ¯H, was calculated which estimates the proportion of pennillii alleles in each individual. Correlations between mean heterozygosity and observed levels of each quantitative trait were highly significant and positive or negative as expected from the relative parental values. Plants with the lowest mean heterozygosity — i.e., closest to the esculentum zymotype also had mean values closest to those of this parent amongst the whole backcross population for each of the quantitative traits.Bivariate and multiple regression analysis was used to evaluate the ability of isozymes vs diagnostic morphological characters to estimate the portion of recurrent parent genes carried in each backcross individual. The results suggest that isozyme data gives better estimates than single diagnostic morphological characters and approach the level obtained by combinations of three morphological traits. Since electrophoretic determinations are made on small seedlings, selection at that stage can effect great savings of space and effort by greatly deminishing the size of the population needed at maturity. As such, isozyme selection would precede morphological selection but not replace it, thus the predictive value of these biochemical markers as well as diagnostic morphological characters could be obtained.  相似文献

Mating success of male grasshoppers, Chorthippus brunneus, has previously been shown to be highest for intermediate, and lowest for extreme individuals with respect to at least one parameter of the male calling song and probably also one morphological character. The heritabilities of these and other song and morphological characters have been estimated in an experiment utilizing the North Carolina I design. Heritability estimates for song parameters are all low (<0·28). Estimates for morphological characters are higher, particularly for features of the stridulatory file, despite the fact that it was just these characters that were shown to be under stabilizing sexual selection. A stabilizing selection which is made in cattle according to phenotypic characters affects simultaneously a genetic character--the level of individual heterozygosity++. As a result animals with mean (modal) level of heterozygosity++ have the least probability of elimination. Similarly, the driving selection in pigs and horses leads to a change in both phenotypic characters and genetic character, the level of individual heterozygosity++. The distribution of genetic polymorphisms in a population contains information about evolutionary processes. The Poisson random field (PRF) model uses the polymorphism frequency spectrum to infer the mutation rate and the strength of directional selection. The PRF model relies on an infinite-sites approximation that is reasonable for most eukaryotic populations, but that becomes problematic when is large ( greater, similar 0.05). Here, we show that at large mutation rates characteristic of microbes and viruses the infinite-sites approximation of the PRF model induces systematic biases that lead it to underestimate negative selection pressures and mutation rates and erroneously infer positive selection. We introduce two new methods that extend our ability to infer selection pressures and mutation rates at large : a finite-site modification of the PRF model and a new technique based on diffusion theory. Our methods can be used to infer not only a "weighted average" of selection pressures acting on a gene sequence, but also the distribution of selection pressures across sites. We evaluate the accuracy of our methods, as well that of the original PRF approach, by comparison with Wright-Fisher simulations.  相似文献

Population size as a measure of population fitness was assayed in laboratory lines of Drosophila melanogaster subjected to directional selection for positive photoactivity and stabilizing selection for a complex of metric wing characters. Three experimental designs were used: directional and stabilizing selection (DS); directional selection (D); unselected controls (C). Population size increased under all three experimental designs. The population size of DS and C lines as well as the rate of increase of the DS lines were higher as compared to the D lines. The results suggest that stabilizing selection maintains population fitness at the higher level.  相似文献

The present study evaluates the evolutionary framework of the Old World fruitbats based on the cytochrome b and 16S rRNA mitochondrial gene sequences from a wide range of taxa. Phylogenetic analyses indicated that morphology-based subfamilies and most suprageneric groups are nonnatural assemblages. They also support the existence of an endemic African clade of fruitbats. The discrepancy between the evolutionary relationships yielded by molecular and morphological data sets may be, at least in part, explained by the recurrent retention of primitive morphology (Rousettus-like) across different lineages. The maintenance of primitive characters in different groups of flying foxes, as well as morphological convergence in nectar-feeding bats and possibly also in short-muzzle bats, may have led to high levels of homoplasy, resulting in misleading taxonomic arrangements. This may be particularly so with respect to high taxonomic levels based on morphological characters.  相似文献

Yeast decode pheromone gradients to locate mating partners, providing a model for chemotropism. How yeast polarize toward a single partner in crowded environments is unclear. Initially, cells often polarize in unproductive directions, but then they relocate the polarity site until two partners’ polarity sites align, whereupon the cells “commit” to each other by stabilizing polarity to promote fusion. Here we address the role of the early mobile polarity sites. We found that commitment by either partner failed if just one partner was defective in generating, orienting, or stabilizing its mobile polarity sites. Mobile polarity sites were enriched for pheromone receptors and G proteins, and we suggest that such sites engage in an exploratory search of the local pheromone landscape, stabilizing only when they detect elevated pheromone levels. Mobile polarity sites were also enriched for pheromone secretion factors, and simulations suggest that only focal secretion at polarity sites would produce high pheromone concentrations at the partner’s polarity site, triggering commitment.  相似文献

Our understanding of balancing selection is currently becoming greatly clarified by new sequence data being gathered from genes in which polymorphisms are known to be maintained by selection. The data can be interpreted in conjunction with results from population genetics models that include recombination between selected sites and nearby neutral marker variants. This understanding is making possible tests for balancing selection using molecular evolutionary approaches. Such tests do not necessarily require knowledge of the functional types of the different alleles at a locus, but such information, as well as information about the geographic distribution of alleles and markers near the genes, can potentially help towards understanding what form of balancing selection is acting, and how long alleles have been maintained.  相似文献
