Intracellular interactions between bacteria and host cells are widespread in nature. In this review, the similarity between the infection processes of bacteria in plant and animal cells will be addressed. As paradigms, we selected the symbiosis between rhizobia and leguminous plants, and the survival of intracellular pathogenic bacteria in animal cells. The rhizobial symbiosis with leguminous plants is a model system for the study of plant-bacterium interactions. Through this interaction, the bacteria are released in a vacuole-like structure, called the symbiosome. The molecular processes, which lead to a functional symbiosome, are far from known. However, membrane fusion processes, and therefore also Ca2+, are crucial to establish this highly specialized organelle-like structure. A homologous system is the infection by certain bacterial pathogens of animal cells. These bacteria enter their host via phagocytosis and avoid the fusion with lysosomes, resulting in a membrane-bound vacuole in which the pathogens survive. The origin and maturation of this phagosome depends on Ca2+-signaling processes in the host cell and on proteins that regulate membrane fusion processes, such as SNAREs, Rab proteins, synaptotagmins and calmodulin. The aim of this review is to compare the endosymbiosis in leguminous plants with the surviving pathogens in animal host cells with a focus on Ca2+-signaling and membrane fusion-related processes. For both systems, the interaction starts with a bacterial entry of the host cell. It will be demonstrated that in both cases Ca2+ is a crucial second messenger. However, more emphasis will be put on the comparison of the later stages of infection, i.e., the formation of specialized bacteria-containing vacuoles. From structural, functional, and proteomic data, it is clear that phagosomes and symbiosomes are more related to each other than originally assumed. Proteins such as V-ATPases, calreticulin, phosphatidylinositol-3-kinase, Rab proteins, and SNAREs are present in both the phagosome and the symbiosome membrane, indicating that common cellular processes are used for building these intracellular organelles. 相似文献

Worldwide conservation goals to protect biodiversity emphasize the need to rethink which objectives are most suitable for different landscapes. Comparing two different Swedish farming landscapes, we used survey data on birds and vascular plants to test whether landscapes with large, intensively managed farms had lower richness and diversity of the two taxa than landscapes with less intensively managed small farms, and if they differed in species composition. Landscapes with large intensively managed farms did not have lower richness than smaller low intensively managed farms. The landscape types were also similar in that they had few red listed species, normally targeted in conservation. Differences in species composition demonstrate that by having both types of agricultural landscapes regional diversity is increased, which is seldom captured in the objectives for agro-environmental policies. Thus we argue that focus on species richness or red listed species would miss the actual diversity found in the two landscape types. Biodiversity conservation, especially in production landscapes, would therefore benefit from a hierarchy of local to regional objectives with explicit targets in terms of which aspects of biodiversity to focus on. 相似文献

Marine fish production is now being carried out afteralmost two decades of research. The anticipated introduction of newspecies and its expansion to the Mediterranean shoreline will help in leading the increased maricultureproduction. Two marine fish hatcheries that operatetoday in Israel produce 7 million fingerlings a year.Traditionally, aquaculture in Israel raises fish ininland freshwater ponds and irrigation reservoirs. Inaddition, Lake Kinneret, the only freshwater lake inIsrael, is stocked yearly with juvenile fish raised inlocal hatcheries (tilapia) or imported fromMediterranean countries (mugil). While culture offreshwater teleost species (carp) was introduced morethan fifty years ago, mariculture started on acommercial scale less than 5 years ago. The limitedsupply of freshwater will accelerate the futureculture of marine species.The bottleneck of almost all marine finfish productionlies in obtaining adequate numbers of fingerlings, dueto their high mortality at early life stages. Theproduction is hindered by inadequate supply of food toearly larval stages which require live food.Development of technologies in Israel for masscultivation of food chain organisms including algae,rotifers and brine shrimp followed their developmentin other parts of the world, most notably thoseachieved in Japan. The local commercial scaleproduction of rotifers relies on several batch orsemi-continuous cultures in conical or flatbottomrectangular containers that supply daily 0.6-4billion rotifers in each hatchery. Originally arelatively large local Brachionus plicatilisstrain was used, but later smaller B.rotundiformis strains were introduced, resulting ina mixture of undefined strains. The incorporation ofalgae (Nannochloropsis sp.) generated in highyield raceways contributes to the reliability ofrotifer cultures. Algae are supplied directly from theraceways or centrifuged and stored as a frozen pasteuntil required in the hatchery. The current dependablesupply of live cultures reduces the need for preservedstocks of rotifers, either as resting eggs or keptalive at low temperatures. To the fish grower,rotifers are live food capsules that deliver essentialnutrients (e.g. long chain unsaturated fatty acids)for growth and survival of fish larvae. A greenhouse experlment was performed In order to Investigate the effects of dlfferent levels of water stress on leaf water potentlal (ψw), stomatal resistance (rs), protein content and chlorophyll (Chl) content of tomato plants (Lycoperslcon esculentum Mill. cv. Nlkita). Water stress was Induced by addlng polyethylene glycol (PEG 6 000) to the nutrlent solution to reduce the osmotlc potential (ψs). We Investlgated the behavlor of antl-oxldant enzymes, such as catalase (CAT) and superoxide dlsmutase (SOD), durlng the development of water stress. Moderate and severe water stress (i.e. ψs= -0.51 and -1.22 MPa, respectlvely) caused a decrease In ψw for all treated (water-stressed) plants compared with control plants, wlth the reductlon belng more pronounced for severely stressed plants. In addltion, rs was slgnlflcantly affected by the Induced water stress and a decrease in leaf soluble protelns and Chl content was observed. Whereas CAT actlvlty remained constant, SOD actlvlty was increased in water-stressed plants compared wlth unstressed plants. These results Indicate the possible role of SOD as an anti-oxidant protector system for plants under water stress condltlons. Background
Acute alveolar hypoxia causes pulmonary vasoconstriction (HPV) which serves to match lung perfusion to ventilation. The underlying mechanisms are not fully resolved yet. The major vascular segment contributing to HPV, the intra-acinar artery, is mostly located in that part of the lung that cannot be selectively reached by the presently available techniques, e.g. hemodynamic studies of isolated perfused lungs, recordings from dissected proximal arterial segments or analysis of subpleural vessels. The aim of the present study was to establish a model which allows the investigation of HPV and its underlying mechanisms in small intra-acinar arteries.Methods
Intra-acinar arteries of the mouse lung were studied in 200 μm thick precision-cut lung slices (PCLS). The organisation of the muscle coat of these vessels was characterized by α-smooth muscle actin immunohistochemistry. Basic features of intra-acinar HPV were characterized, and then the impact of reactive oxygen species (ROS) scavengers, inhibitors of the respiratory chain and Krebs cycle metabolites was analysed.Results
Intra-acinar arteries are equipped with a discontinuous spiral of α-smooth muscle actin-immunoreactive cells. They exhibit a monophasic HPV (medium gassed with 1% O2) that started to fade after 40 min and was lost after 80 min. This HPV, but not vasoconstriction induced by the thromboxane analogue U46619, was effectively blocked by nitro blue tetrazolium and diphenyleniodonium, indicating the involvement of ROS and flavoproteins. Inhibition of mitochondrial complexes II (3-nitropropionic acid, thenoyltrifluoroacetone) and III (antimycin A) specifically interfered with HPV, whereas blockade of complex IV (sodium azide) unspecifically inhibited both HPV and U46619-induced constriction. Succinate blocked HPV whereas fumarate had minor effects on vasoconstriction.Conclusion
This study establishes the first model for investigation of basic characteristics of HPV directly in intra-acinar murine pulmonary vessels. The data are consistent with a critical involvement of ROS, flavoproteins, and of mitochondrial complexes II and III in intra-acinar HPV. In view of the lack of specificity of any of the classical inhibitors used in such types of experiments, validation awaits the use of appropriate knockout strains and siRNA interference, for which the present model represents a well-suited approach. 相似文献8.
The aim of the current survey was to investigate the complementary self-treatment of the common cold and flu with medicinal plants among pharmacy customers in Estonia. A multiple-choice questionnaire listing 10 plants and posing questions on the perceived characteristics of cold and flu, the effectiveness of plants, help-seeking behaviour, self-treatment and sources of information, was distributed to a sample of participants in two medium size pharmacies. The participants were pharmacy customers: 150 in Tallinn (mostly Russian speaking) and 150 in Kuressaare (mostly Estonian speaking). The mean number of plants used by participants was 4.1. Of the respondents, 69% self-treated the common cold and flu and 28% consulted with a general practitioner. In general, medicinal plants were considered effective in the treatment of the above-mentioned illnesses and 56% of the respondents had used exclusively medicinal plants or their combination with OTC medicines and other means of folk medicine for treatment. The use of medicinal plants increased with age and was more frequent among female than male respondents. Among Estonian-speaking customers lime flowers, blackcurrant and camomile were more frequently used, and among Russian speaking customers raspberry and lemon fruits. Regardless of some statistically significant differences in preferred species among different age, education, sex and nationality groups, the general attitude towards medicinal plants for self-treatment of the common cold and flu in Estonia was very favourable. 相似文献

Metastasis is the major cause of death in cancer patients. Whereas colorectal cancer (CRC) incidence increases with age, metastatic spread seems to decline. Furthermore, the epidemiology of CRC is changing. There is an increase in CRC incidence in the young, presenting at an advanced stage with higher likelihood of synchronous or metachronous metastases, and a decline in CRC incidence and metastatic spread in the oldest-old. Emerging data suggest that age-related changes with regard to tumor biology (e.g. genomic instability), the tumor microenvironment (e.g. inflammaging) and the immune system (e.g. immunosenescence), complemented by interaction between the genome and exposome might contribute to the observed metastatic patterns. As aging is a key prognostic factor, this highlights the need for further studies investigating age-related patterns and underlying mechanisms of tumor growth and dissemination. Eventually, this might allow for better risk stratification, refinement of screening strategies and follow-up care as well as therapies tailored to reflect patient age and that might possibly target responsible biomarkers in a precision medicine approach. This review aims to discuss the influence of aging on metastatic spread in colorectal cancer and elucidate underlying mechanisms responsible for the observed metastatic patterns. 相似文献

Ceropegia thwaitesii Hook (Asclepiadaceae), an endemic plant species, due to habitat destruction and over exploitation has a very restricted distribution in the Western Ghats of Tamil Nadu, India. The present wrok aimed to determine the chemical composition, the total phenolic (TPC), flavonoid (TFC) and tannin content (TEC), and to assess the antioxidant properties of various extracts of in vivo plants (IVP) and in vitro regenerated plants (IRP) of C. thwaitesii. Some phenolic compounds like gallic acid, cathechol, vanillin and salicylic acid were identified and quantified by HPLC. All the extracts possessed relevant radical scavenging activity on DPPH, Superoxide radical scavenging activity, and Nitric oxide radicals as well as total antioxidant ability. DPPH assay of in vitro methanol stems extracts and ethanol leaves extracts revealed the best antioxidant properties with important IC50 values of 0.248?±?0.45?µg/mL and 0.397?±?0.67?µg/mL, respectively, whereas in vivo chloroform stems extracts showed a lower antioxidant activity (IC50 of 10.99?±?0.24?µg/mL). The IRP methanol extracts of stem and leaves had good inhibitory activity against all tested microorganisms in a dose-dependent manner. These results suggested that in vitro raised plants of C. thwaitesii are an excellent source of antioxidant compounds to be exploited on an industrial level as food additive. 相似文献

Physical activity and sports have repeatedly been reported to be associated with telomere length. We studied the association of different types of sports across different stages of life on relative leukocyte telomere length (rLTL) in advanced age.815 participants (397 men) from the Berlin Aging Study II aged over 61 years were included in the analysis. rLTL was measured by real time PCR and physical activity was determined retrospectively by questionnaire, assessing type and duration of sports in the past as well as currently. Five separate multiple linear regression models adjusted for various control variables were performed. 67.3% of participants exercised currently, whereas 19.4% performed sports only between the age of 20 and 30. rLTL was higher in subjects who stated to exercise currently (N = 456), and in subjects who engaged in endurance (N = 138) or intensive activity sports (N = 32). Current physical activity was positively associated with rLTL in the risk factor adjusted regression model (β = 0.26, p < 0.001) and practicing sports for a minimum of 10 years preceding the assessment had a significant effect on rLTL (β = 0.39, p = 0.011). The highest impact was seen for intensive activity sports (β = 0.79, p < 0.001) and physical activity since at least 42 years (β = 0.47, p = 0.001). However, physical activity only between 20 and 30 years of age did not affect rLTL in old age when compared to no sports at all (β = -0.16, p = 0.21). Physical activity is clearly associated with longer rLTL. The effect is seen with longer periods of physical activity (at least 10 years), with intensive sports activities having the greatest impact on rLTL. Our data suggest that regular physical activity for at least 10 years is necessary to achieve a sustained effect on rLTL. 相似文献

Background
Hepatitis E virus (HEV) infects a range of species, including humans, pigs, wild boars and deer. Zoonotic transmission may contribute to the high HEV seroprevalence in the human population of many countries. A novel divergent HEV from moose (Alces alces) in Sweden was recently identified by partial genome sequencing. Since only one strain was found, its classification within the HEV family, prevalence in moose and zoonotic potential was unclear. We therefore investigated samples from 231 moose in seven Swedish counties for HEV, and sequenced a near complete moose HEV genome. Phylogenetic analysis to classify this virus within the family Hepeviridae and to explore potential host specific determinants was performed.Methods and Findings
The HEV prevalence of moose was determined by PCR (marker for active infection) and serological assays (marker of past infection) of sera and 51 fecal samples from 231 Swedish moose. Markers of active and past infection were found in 67 (29%) animals, while 34 (15%) were positive for HEV RNA, 43 (19%) were seropositive for anti-HEV antibodies, and 10 (4%) had both markers. The number of young individuals positive for HEV RNA was larger than for older individuals, and the number of anti-HEV antibody positive individuals increased with age. The high throughput sequenced moose HEV genome was 35-60% identical to existing HEVs. Partial ORF1 sequences from 13 moose strains showed high similarity among them, forming a distinct monophyletic clade with a common ancestor to HEV genotype 1-6 group, which includes members known for zoonotic transmission.Conclusions
This study demonstrates a high frequency of HEV in moose in Sweden, with markers of current and past infection demonstrated in 30% of the animals. Moose is thus an important animal reservoir of HEV. The phylogenetic relationship demonstrated that the moose HEV belonged to the genotype 1-6 group, which includes strains that also infect humans, and therefore may signify a potential for zoonotic transmission of this HEV. 相似文献15.

Thirty new aryl-pyridazinone-substituted benzenesulphonylurea derivatives (I–XXX) were synthesized and evaluated for their anti-hyperglycaemic activity in glucose-fed hyperglycaemic normal rats. Twenty-three compounds (III–XI, XIV–XVII, XIX–XXIV, XXVI and XXVIII–XXX) showed more or comparable area under the curve (AUC) reduction percentage (ranging from 21.9% to 35.5%) as compared to the standard drug gliclazide (22.0%). On the basis of docking results, 18 compounds were screened for their in vitro ability to inhibit rat lens aldose reductase. Ten compounds (III–VI, XII, XVI–XVIII, XXI and XXVII) showed ARI activity with IC50 ranging from 34 to 242?μM. Out of these, two compounds IV and V showed best ARI activity which is comparable with that of quercetin. As a result, two compounds (IV and V) possessing significant dual action (anti-hyperglycaemic and aldose reductase inhibition) were identified and may be used as lead compounds for developing new drugs. 相似文献

Gallbladder cancer (GBC) is a fatal condition with dismal prognosis and aggressive local invasiveness; and with uncharacterised molecular pathology relating to non-specific therapeutic modalities. Given the importance of oxidative stress in chronic diseases and carcinogenesis, and the lacunae in literature regarding its role in gallbladder diseases, this study aimed to study the involvement of oxidative stress and deregulation in the base excision repair (BER) pathway in the pathogenesis of gallbladder diseases including GBC. This study involved patients from the North-East Indian population, where the numbers of reported cases are increasing rapidly and alarmingly. Oxidative stress, based on 8-OH-dG levels, was found to be significantly higher in gallbladder anomalies (cholelithiasis [CL] and cholecystitis [CS]) and GBC at the plasma and DNA level, and was associated with GBC severity. The expressions of key BER pathway genes were downregulated in gallbladder anomalies and GBC compared to controls, and in GBC compared to both non-neoplastic controls and gallbladder anomalies. Expression of XRCC1 and hOGG1 was significantly associated with both susceptibility and severity of GBC. The XRCC1 codon280 polymorphism was associated with disease susceptibility; and significantly higher oxidative stress was observed in hOGG1 genotypic variants. The genomes of GBC patients were found to be more hypermethylated compared to controls, with the promoters of XRCC1 and hOGG1 being hypermethylated and, therefore, being silenced. This study underlined the prognostic significance of the oxidative stress marker 8-OH-dG and BER pathway genes, especially hOGG1 and XRCC1, in gallbladder anomalies and GBC, as well as stated their potential for therapeutic targeting. 相似文献

In order to understand the pathway involved in the chemical enhancement of photosynthetic rate, sodium bisulfite (NaHSO3) and benzyladenine (BA), a growth regulator, were applied to strawberry plants. The influence of these compounds on gas exchange
and millisecond delayed light emission (ms-DLE) was investigated using 2-month-old plants. Results showed the net photosynthetic
rate (A) in leaves was promoted by both NaHSO3 and BA. Stomatal conductance (g) and transpiration rate (E) were significantly increased only by BA, while intercellular CO2 concentration (Ci) was significantly decreased by NaHSO3. The enhancement of A by NaHSO3 and BA was only a short-term effect, lasting approximately 5 days for NaHSO3 and 30 h for BA. Plants treated with NaHSO3, BA or NaHSO3 + BA, showed no significant fluctuations in carboxylation efficiency (CE), photorespiration (RP) or dark respiration (RD). These results suggest that the influences of NaHSO3 and BA on gas exchange particularly A, could be via different mechanisms: the enhancement of A by the application of low concentrations of NaHSO3 appears to be associated with increased cyclic electron flow, while BA enhancement of A is at least partially due to increased g and/or E. 相似文献

idea of metamorphosis in the plant kingdom as an ideal model to interpret connections of natural phenomena, in particular
as a conceptual frame for a natural system. Furthermore, he aimed to develop the philosophical statement of the metamorphosis,
in which he called for nature-philosophical conceptions in order to materialize his representation of plant “affinities,”
and of a kind of “ontogeny” of the whole plant kingdom. This paper demonstrates that, between speculative views and empirical
attempts, the extent to which Reichenbach actually belonged to a new “school” of thought, which left its mark on the history

In the present study, trikatu, an herbal compound was evaluated for its immunomodulatory and anti-inflammatory properties with reference to cell mediated immune responses (delayed type hypersensitivity reaction), humoral immune response (haemagglutination titer and plaque forming assay), macrophage phagocytic index, circulating immune complex and inflammatory mediators in rats. For comparison purposes, indomethacin was used as a reference drug for anti-inflammatory studies. The results obtained in our study showed a significant decrease in cell mediated immune responses, humoral immune responses (haemagglutination titre and plaque forming assay) and macrophage phagocytic index in trikatu treated rats (1000 mg/kg/b.wt.) compared to control animals implying its immunosuppressive property. In addition, significant anti-inflammatory effects were observed in trikatu treated adjuvant induced arthritic rats by a reduction in the levels of circulating immune complexes and inflammatory mediators (TNF-alpha and Interleukin-1beta). Thus, in conclusion, our data suggest that trikatu could be considered as a potential anti-inflammatory agent for treating autoimmune inflammatory disorders like rheumatoid arthritis with immunosuppressive property. 相似文献
