Populus angustifolia, the narrowleaf cottonwood, is considered one of two native species of Populus section Tacamahaca restricted to western North America. Efforts to construct a definitive phylogeny of Populus spp. are complicated by ancient hybridization, but some phylogenetic analyses suggest P. angustifolia is more closely related to Asian congeners than to Populus trichocarpa, the other species of Populus section Tacamahaca in western North America. Because hosts and their obligate symbionts can display parallel phylogeographic patterns, we evaluated the possibility of a Beringian migration into North America by an Asian ancestor of P. angustifolia by determining the distributions, host preferences, and, in some cases, closest phylogenetic relatives of fungal leaf pathogens of P. angustifolia. Phyllactinia populi, a common foliar pathogen on Populus spp. in Asia but unknown on P. trichocarpa, was found on P. angustifolia in multiple sites. Mycosphaerella angustifoliorum, also unknown on P. trichocarpa, was commonly collected on P. angustifolia. Conversely, many common foliar pathogens of P. trichocarpa in western North America were not found on P. angustifolia; only Melampsora×columbiana and Drepanopeziza populi were common to both Populus species. Phylogenetic analyses revealed that M. angustifoliorum was not part of the diversification of Mycosphaerella on Populus that includes all other Mycosphaerella species on Populus in North America: Mycosphaerella populicola, Mycosphaerella populorum, M. sp. 1, and M. sp. 2. The latter two undescribed species represent a newly discovered diversification of M. populorum in western North America. Overall, the leaf pathogen community of P. angustifolia is consistent with a Beringian migration into North America by the ancestor of P. angustifolia.  相似文献

We isolated the phenolic glucoside salicortin from a Populus euramericana bark extract, and examined its ability to suppress inflammatory responses as well as the molecular mechanisms underlying these abilities, using lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Salicortin inhibited iNOS expression and the subsequent production of NO in a dose-dependent manner in the LPS-stimulated RAW 264.7 cells. Salicortin significantly suppressed LPS-induced signal cascades of NF-κB activation, such as IKK activation, IκBα phosphorylation and p65 phosphorylation in RAW 264.7 cells. In addition, salicortin inhibited the LPS-induced activation of JNK, but not ERK or p38 MAPK. Furthermore, salicortin significantly inhibited production of pro-inflammatory cytokines, such as TNF-α, IL-1β and IL-6 in the LPS-stimulated RAW 264.7 cells. These findings suggest that salicortin may show its anti-inflammatory activity by suppressing the LPS-induced expression of pro-inflammatory mediators through inhibition of NF-κB and JNK MAPK signaling cascades in macrophages. Lavandula angustifolia is a well known herbal medicine with a variety of useful properties, including its acaricidal effect. This experiment was carried out to study the bioacaricidal activity of L. angustifolia essential oil (EO) against engorged Rhipicephalus (Boophilus) annulatus (Acari; Ixodidae) females. For this purpose six serial concentrations (0.5, 1.0, 2.0, 4.0, 6.0 and 8.0% w/v) of L. angustifolia EO were used. There was considerable mortality in concentrations more than 4.0% although there were no differences between 6.0 and 8.0% in all measured criteria. The mortality rate 24 h after inoculation was 73.26 and 100% in groups treated with 4.0 and 8.0% EO, respectively. Lavender EO also reduced tick egg weight in a concentration-dependent manner. The amount of eggs produced varied from 0.12 g (at 0.5% EO) to 0.00 g (at 8.0% EO) but did not differ statistically from the control. L. angustifolia EO caused 100% failure in egg laying at 6.0 and 8.0% whereas this value in the control group was zero. A positive correlation between L. angustifolia EO concentration and tick control, assessed by relative mortality rate and egg-laying weight, was observed by the EO LC/EC50, which, when calculated using the Probit test, was 2.76-fold higher than the control. Lavender is a promising acaricidal against R. (B.) annulatusin vitro.  相似文献

Genomic studies have been used to identify genes underlying many important plant secondary metabolic pathways. However, genes for salicinoid phenolic glycosides (SPGs)—ecologically important compounds with significant commercial, cultural, and medicinal applications—remain largely undescribed. We used a linkage map derived from a full‐sib population of hybrid cottonwoods (Populus spp.) to search for quantitative trait loci (QTL) for the SPGs salicortin and HCH‐salicortin. SSR markers and primer sequences were used to anchor the map to the V3.0 P. trichocarpa genome. We discovered 21 QTL for the two traits, including a major QTL for HCH‐salicortin (R2 = .52) that colocated with a QTL for salicortin on chr12. Using the V3.0 Populus genome sequence, we identified 2,983 annotated genes and 1,480 genes of unknown function within our QTL intervals. We note ten candidate genes of interest, including a BAHD‐type acyltransferase that has been potentially linked to PopulusSPGs. Our results complement other recent studies in Populus with implications for gene discovery and the evolution of defensive chemistry in a model genus. To our knowledge, this is the first study to use a full‐sib mapping population to identify QTL intervals and gene lists associated with SPGs.  相似文献

The phenolic glycosides salicin and salicortin were found to influence larval growth and development rates and adult feeding preference of Phratora vulgatissima in laboratory feeding studies. Salicortin was more toxic to larvae than salicin, and none of the third instar larvae fed on Salix viminalis leaves amended with 1.52% (fresh mass) salicortin pupated. Condensed tannins (proanthocyanadins) did not affect larval performance. It was concluded that Salix burjatica resistance to willow beetle is due to the high levels of salicortin which occur in leaves of this species.  相似文献

Rapid identification of Populus L. species and hybrids can be achieved with relatively little effort through the use of primer extension-based single nucleotide polymorphism (SNP) genotyping assays. We present an optimized set of 36 SNP markers from 28 gene regions that diagnose eight poplar species (Populus angustifolia James, Populus balsamifera L., Populus deltoides Bartram, Populus fremontii Watson, Populus laurifolia Ledeb., Populus maximowiczii Henry, Populus nigra L., and Populus trichocarpa Torr. & Gray). A total of 700 DNA sequences from six Populus species (1–15 individuals per species) were used to construct the array. A set of flanking and probe oligonucleotides was developed and tested. The accuracy of the SNP assay was validated by genotyping 448 putatively “pure” individuals from 14 species of Populus. Overall, the SNP assay had a high success rate (97.6 %) and will prove useful for the identification of all Aigeiros Duby and Tacamahaca Spach. species and their early-generation hybrids within natural populations and breeding programs. Null alleles and intraspecific polymorphisms were detected for a few locus/species combinations in the Aigeiros and Tacamahaca sections. When we attempted to genotype aspens of the section Populus (Populus alba L., Populus grandidentata Michx., Populus tremula L., and Populus tremuloides Michx.), the success rate of the SNP array decreased by 13 %, demonstrating moderate cross-sectional transferability.  相似文献

Cottonwoods are well known as foundation riparian trees that support diverse communities and drive ecosystem processes. Although hybridization naturally occurs when the distributions of two or more cottonwood species overlap, few cottonwood hybrid zones have been genetically characterized. We use genetic and genomic analyses to characterize patterns of admixture and introgression for a newly described hybrid zone at the intersection of three species (Populus L. Salicaceae—Populus deltoides, Populus fremontii, and Populus angustifolia) in southwestern Colorado, USA. Analysis of nuclear and chloroplast microsatellite marker data detected substantial genetic variation among individuals, revealing that (1) hybridization is occurring between two, not three, species (P. deltoides and P. angustifolia); (2) gene flow is bidirectional; (3) hybrids are not abundant (admixture detected in only 34 of 270 trees), with most being early-generation F1 hybrids; (4) cytonuclear disequilibria exists and F1 hybrids tend to retain P. deltoides—like chloroplasts; and (5) roughly 30 % of the nuclear markers deviated from a neutral pattern of introgression, suggesting that selection may play a role in shaping the genetic structure of the hybrid zone in this region. Overall, our results show that despite strong selection maintaining species divergence, transfer of allelic variants across species boundaries can occur. Our study assesses the fine-scale genetic structure of hybridization between P. angustifolia and P. deltoides and lays the foundation for examining how geographic differences in hybrid zone dynamics arise and may influence subsequent ecological and evolutionary processes.  相似文献

Complete Populus genome sequences are available for the nucleus (P. trichocarpa; section Tacamahaca) and for chloroplasts (seven species), but not for mitochondria. Here, we provide the complete genome sequences of the chloroplast and the mitochondrion for the clones P. tremula W52 and P. tremula x P. alba 717-1B4 (section Populus). The organization of the chloroplast genomes of both Populus clones is described. A phylogenetic tree constructed from all available complete chloroplast DNA sequences of Populus was not congruent with the assignment of the related species to different Populus sections. In total, 3,024 variable nucleotide positions were identified among all compared Populus chloroplast DNA sequences. The 5-prime part of the LSC from trnH to atpA showed the highest frequency of variations. The variable positions included 163 positions with SNPs allowing for differentiating the two clones with P. tremula chloroplast genomes (W52, 717-1B4) from the other seven Populus individuals. These potential P. tremula-specific SNPs were displayed as a whole-plastome barcode on the P. tremula W52 chloroplast DNA sequence. Three of these SNPs and one InDel in the trnH-psbA linker were successfully validated by Sanger sequencing in an extended set of Populus individuals. The complete mitochondrial genome sequence of P. tremula is the first in the family of Salicaceae. The mitochondrial genomes of the two clones are 783,442 bp (W52) and 783,513 bp (717-1B4) in size, structurally very similar and organized as single circles. DNA sequence regions with high similarity to the W52 chloroplast sequence account for about 2% of the W52 mitochondrial genome. The mean SNP frequency was found to be nearly six fold higher in the chloroplast than in the mitochondrial genome when comparing 717-1B4 with W52. The availability of the genomic information of all three DNA-containing cell organelles will allow a holistic approach in poplar molecular breeding in the future.  相似文献

In continuation of the screening of South African seaweeds to identify potential candidates for the development of pharmaceutically active functional foods, we investigated the inhibitory effects of a crude 80 % methanol extract, solvent fractions and isolated compounds from the kelp Macrocystis angustifolia against enzymes involved in type 2 diabetes and dementia. Repeated column fractionation of the ethyl acetate fraction of the crude extract of M. angustifolia afforded two phenol derivatives identified by spectroscopic analyses (1D and 2D NMR): 4-(2-hydroxyethyl)phenol (tyrosol) (1) and 4-(1,2-dihydroxyethyl)phenol (2). These compounds were isolated from a marine alga for the first time. The ethyl acetate (IC50?=?14.08?±?1.21 μg mL?1) and butanol (IC50?=?77.94?±?11.69 μg mL?1) fractions exhibited potent inhibition against α-glucosidase and acetylcholinesterase (AChE) enzymes, respectively. Tyrosol (1) and its derivative, 4-(1,2-dihydroxyethyl)phenol (2), showed potent inhibition against both α-glucosidase and AChE enzymes. Based on in silico evaluation, these two compounds are anticipated to possess sufficient oral bioavailability in accordance to the Lipinski Rule of Five without any toxicity risk. Natural α-glucosidase and AChE inhibitors from M. angustifolia offer a novel approach to control type 2 diabetes and dementia.  相似文献

We exposed cuttings of two sympatric species of Sect. Tacamahaca Spach, Populus cathayana Rehder and Populus przewalskii Maximowicz, to two watering regimes in a greenhouse. In the semi-controlled environmental study, two watering treatments which were watered to 100 and 25% of field capacity were used, respectively. The effects of water deficit on early growth, biomass allocation and water use efficiency (WUE) were investigated. We found that there were significant interspecific differences in early growth, dry matter allocation and water use efficiency between two sympatric Populus species. Compared with P. cathayana, P. przewalskii showed higher shoot height, dry matter accumulation, number of leaves, total leaf area, fine root mass, fine root/total root ratio and water use efficiency under both well-watered and water-stressed treatments. On the other hand, P. przewalskii also showed higher root mass/foliage area ratio, root/shoot ratio and carbon isotope composition than P. cathayana under water-stressed treatment. The results suggested that there were different water-use strategies between two sympatric Populus species, P. przewalskii with higher drought tolerance may employ a conservative water-use strategy, whereas P. cathayana with lower drought tolerance may employ a prodigal water-use strategy. The findings confirm the existence of interspecific genetic differences in early growth, dry matter allocation and water use efficiency as affected by water stress, these variations in drought responses may be used as criteria for species selection and tree improvement.  相似文献

Condensation of (S,S)-1,2-cyclohexanediamine with 2 equiv. of 2-pyridine carboxaldehyde in toluene in the presence of molecular sieves at 70 °C gives N,N′-bis(pyridin-2-ylmethylene)-(S,S)-1,2-cyclohexanediamine (S,S-1) in 95% yield. Reduction of 1 with an excess of NaBH4 in MeOH at 50 °C gives N,N′-bis(pyridin-2-ylmethyl)-(S,S)-1,2-cyclohexanediamine (S,S-2) in 90% yield. Reaction of 1 or 2 with 1 equiv. of CuCl2 · 2H2O in methanol gives complexes [N-(pyridin-2-ylmethylene)-(S,S)-1,2-cyclohexanediamine]CuCl2 (3) and [Cu(S,S-2)(H2O)]Cl2 · H2O (4), respectively, in good yields. Complex 4 can further react with 1 equiv. of CuCl2 · 2H2O in methanol to give [Cu(S,S-2)][CuCl4] (5) in 75% yield. The rigidity of the ligand coupled with the steric effect of the free anion plays an important role in the formation of the helicates. Treatment of ligand S,S-1 with AgNO3 induces a polymer helicate {[Ag(S,S-1)][NO3]}n (6), while reaction of ligand 2 with AgPF6 or AgNO3 in methanol affords a mononuclear single helicate [Ag(S,S-2)][PF6] (7) or a dinuclear double helicate [Ag2(S,S-2)2][NO3]2 · 2CH3OH (8) in good yields, respectively. All compounds have been characterized by various spectroscopic data and elemental analyses. Compounds 1, 3-5, 7 and 8 have been further subjected to single-crystal X-ray diffraction analyses. The Cu(II) complexes do not show catalytic activity for allylation reaction, in contrast to Ag(I) complexes, but they do show catalytic activity for Henry reaction (nitroaldol reaction) that Ag(I) complexes do not.  相似文献

DUF579 (domain of unknown function 579) family proteins contain a DUF579 domain structure but vary greatly in their overall sequence similarity. Several DUF579 proteins have been found to play a role in cell wall biosynthesis in Arabidopsis, while DUF579 family genes have not yet been systematically investigated in Populus. In this study, the Populus DUF579 family proteins were found to be localized in different cell types and subcellular locations. The diverse expression patterns of the proteins indicate that they may perform different functions in Populus. Among the DUF579 family members, PtrDUF579-1 is found to be specifically expressed in vascular cambium zone cells where it is localized in the Golgi apparatus. Suppression of PtrDUF579-1 expression reduced plant height and stem diameter size. Cambium cell division and xylem tissue growth was inhibited while secondary cell wall formation was unchanged in PtrDUF579-1 suppressed plants. Cell walls analysis showed that the composition of the pectin fraction of the cambium cell wall was altered while other polysaccharides were not affected in PtrDUF579-1 suppressed plants. This observation suggest cambium expressed PtrDUF579-1 may affect cell wall biosynthesis and be involved in cambium cell proliferation in Populus. Overall, DUF579 family proteins play a diverse set of roles in Populus.  相似文献

Populus (Salicaceae) is one of the most economically and ecologically important genera of forest trees. The complex reticulate evolution and lack of highly variable orthologous single-copy DNA markers have posed difficulties in resolving the phylogeny of this genus. Based on a large data set of nuclear and plastid DNA sequences, we reconstructed robust phylogeny of Populus using parsimony, maximum likelihood and Bayesian inference methods. The resulting phylogenetic trees showed better resolution at both inter- and intra-sectional level than previous studies. The results revealed that (1) the plastid-based phylogenetic tree resulted in two main clades, suggesting an early divergence of the maternal progenitors of Populus; (2) three advanced sections (Populus, Aigeiros and Tacamahaca) are of hybrid origin; (3) species of the section Tacamahaca could be divided into two major groups based on plastid and nuclear DNA data, suggesting a polyphyletic nature of the section; and (4) many species proved to be of hybrid origin based on the incongruence between plastid and nuclear DNA trees. Reticulate evolution may have played a significant role in the evolution history of Populus by facilitating rapid adaptive radiations into different environments.  相似文献

Two new metabolites were obtained by microbial transformation of the triterpene nigranoic acid (3,4-secocyloarta-4 (28), 24 (Z)-diene-3,26-dioic acid), (1) in the culture of Trichoderma sp. JY-1, a fungus obtained from the branches of Kadsura angustifolia. Their structures were established as 15α, 16α-dihydroxy-3,4-secocyloarta-4 (28), 17 (20), 17 (E), 24 (E)-triene-3,26-dioic acid (2) and 16α, 20α-dihydroxy-18 (13  17β) abeo-3,4-secocyloarta-4 (28), 12 (13), 24 (Z)-triene-3,26-dioic acid (3) by analysis of NMR and MS data and by analogy with the data for the substrate nigranoic acid (1). Compound 2 was found to possess an unusual 17(20), 17 (E)-ene structure while compound 3 featured an unprecedented 18(13  17β)-abeo-secocyloarta skeleton. Additionally, compounds 13 showed weak anti-HIV activity with EC50 values of 10.5, 8.8 and 7.6 μg/mL, therapeutic index values (CC50/EC50) of 8.48, 9.12 and 10.1, respectively.  相似文献

New scandium and yttrium complexes ScL2(bpzmp)(THF) {L = Cl (1); L = CH2Si(CH3)3 (3)}, YL2(bpzmp) {L = Cl (2); L = OTf (5)} and Y(CH2SiMe3)2(bpzmp)(THF) (4) bearing the heteroscorpionate bpzmp ligand {bpzmp = (3,5-tBu2-2-phenoxo)bis(3,5-Me2-pyrazol-1-yl)methane} have been synthesized and characterized by means of NMR and Mass spectroscopy. The tridentate monoanionic ligand resulted κ3-coordinated to the metal via the oxygen and both the sp2 nitrogen atoms of the heterocycles, producing complexes in CS symmetry.The behavior of 1-4 in ethylene polymerization was investigated after proper activation with different activating agents. Complex 4, in combination with the Brönsted or Lewis acids [PhNMe2H][B(C6F5)4] or [Ph3C][B(C6F5)4], produced linear high molecular weight polyethylene in good yield.  相似文献

A series of novel asymmetric binuclear titanocenes linked with alkyl benzyl ethers p-[(C5H5TiCl2)C5H4CH2]C6H4O(CH2)n[C5H4(TiCl2C5H5)] (n = 2-5) (13-16) have been synthesized by treating p-(LiC5H4CH2)C6H4O(CH2)n(C5H4Li) (n = 2-5) (9-12) with C5H5TiCl3. The new complexes have been characterized by elemental analysis and NMR spectra. Their catalytic activity for ethylene polymerization was investigated in the presence of aluminoxane (MAO). The results show that 13-16 are efficient catalysts for producing polyethylene (PE) with a broad molecular weight distribution (MWD). Their catalytic activity is highly dependent on the length of the alkyl chain and the polymerization conditions. A longer alkyl chain increases the catalytic activity, whereas the molecular weight of the produced polyethylene decreases.  相似文献
