To investigate the effect of fasting and refeeding on the body mass, thermogenesis and serum leptin in Brandt's voles, the changes in body and body fat mass, resting metabolic rate (RMR), mitochondrial cytochrome c oxidase (COX) activity in liver and brown adipose tissue (BAT), uncoupling protein 1 (UCP1) content of BAT, serum leptin level and post-fasting food intake were monitored and measured.  相似文献

  • 1.1. Fluorescence and electron microscopy were used to visualize differences between avian adipose tissue (AAT) collected from clavicular and abdominal regions of the great tit, the willow tit, the house sparrow and the Japanese quail, and interscapular brown adipose tissue (BAT) obtained from the Djungarian dwarf hamster.
  • 2.2. Multilocular fat cells were found in AAT. The prerequisite for multilocularity, however, was not simply winter acclimatization [short photophase 4L:20D and low ambient temperature (< −20°C in January in Oulu)] or cold-acclimation (−25°C). Multilocular adipocytes were found during autumn and in unacclimated control birds as well. Mitochondria in the AAT were fewer and about one-sixth the length of those in BAT. This finding was associated with low cytochrome oxidase (COX) activity in the tissue homogenate and isolated mitochondrial fraction of the AAT (< 5.2% of that in BAT).
  • 3.3. Catecholamine fluorescence was seen only around arteries in the AAT. Signs of sympathetic parenchymal innervation were found neither in winter- nor in cold-acclimated birds, but typically, sympathetic nerve fibers forming a basket-like network around every cell were seen in the brown fat of the hamster.
  • 4.4. Our results show that AAT in the adult birds resembles white adipose tissue more than brown. Multilocularity of adipocytes may improve lipolysis to deliver fatty acids for muscle fuel of shivering or NST.

  • 1.1. The biochemical characteristics of homogenates and mitochondria isolated from the outer and inner layers of the ventricular myocardium of carp were studied.
  • 2.2. The homogenate prepared from the inner layer exhibited higher activity of cytochrome oxidase than that from the outer layer. No difference was found in the activity of cytochrome oxidase between mitochondria from the inner and outer layers. Difference spectra of cytochromes also showed that their content in mitochondria of both layers is similar and that the higher oxidative capacity of the spongious layer is due to a higher content of mitochondria.
  • 3.3. In comparison with rat heart a higher content of cyt aa3 and a lower content of Cyt b and cyt cc1 were found in carp heart mitochondria.
  • 4.4. In comparison with rat heart, carp heart mitochondrial enzymes were more sensitive to freezing-thawing and to detergent action.

  • 1.1. We have compared the primary structure and the predicted secondary structure of subunit I (COI) of cytochrome oxidase with those of other integral redox enzymes which contain membrane-buried iron centres.
  • 2.2. Some striking analogies have been found between the deduced transmembrane folding for COI and the known three-dimensional structure of the photosynthetic reaction centre of Rhodobacter sphaeroides.
  • 3.3. These structural analogies are paralleled by a fundamental functional analogy between these two redox systems, since they both oxidize reduced cytochrome c at the positive side of the membrane, transferring electrons to membrane-buried metal centres.
  • 4.4. A statistical evaluation has been performed on the amino acid composition of the peptides containing known histidine ligands of the membrane-buried iron in cytochrome b of the bc 1 complex and in the bacterial reaction centre.
  • 5.5. This evaluation was then applied to the peptides which contain conserved histidines in subunit I of cytochrome oxidase, subunit that is known to bind both haem a and a3, indicating which of these histidines are the most likely ligands of the membrane-buried iron of the a-haems.
  • 6.6. A sequence homology has been found between the known oxygen binding site and the haem binding peptide in cyt P450 and two peptides which are conserved in all the sequences of COI, thus indicatingt the possible oxygen catalytic site of cytochrome oxidase.

  • 1.1. Mineral balance was studied in meadow voles (Microtus pennsylvanicus) maintained in the laboratory.
  • 2.2. Urine and fecal Na+ contents of voles on low-Na+ diets were comparable to those reported for other herbivore species, but urine and fecal K levels were higher.
  • 3.3. Voles approached Na+ balance (input = output) on diets with Na+ content as low as 56 ppm.
  • 4.4. There was not a clearcut hypertrophy of the adrenal-gland zona glomerulosa in voles maintained on low-Na+ diets.
  • 5.5. Plasma K content and bone water content were higher in voles maintained on high-Na + vegetation diets, suggesting expansion of extracellular fluid volume.

  • 1.1. An apparent effect of insulin administration on enlargement of interscapular brown adipose tissue (BAT) was found in heat-exposed rats, but not in warm-adapted or cold-acclimated rats.
  • 2.2. BAT extracts from the heat-acclimated/insulin-treated (HI) rats notably increased the capillary growth in an in vitro angiogenesis model in which microvascular fragments and myofibroblastic (Mf) cells isolated from lipid tissues were grown in co-culture, although a direct effect of insulin was not high.
  • 3.3. BAT extracts from the HI rats stimulated the production of endothelial cell growth factor and collagen by Mf cells.
  • 4.4. It is probable that an increased angiogenic activity contributes to the capillary growth and tissue growth in BAT of HI rats.

  • 1.1. Adenylate cyclase activity was determined in membranes of white and brown adipose tissue (WAT and BAT, respectively) from rats fed a high-energy diet (EXP group) vs those fed a nutritionally balanced one (CON group).
  • 2.2. The isoproterenol- and guanine nucleotide-induced adenylate cyclase activity in WAT membranes of EXP rats was lower than that in CON rats.
  • 3.3. Relative adenylate cyclase activity in like treated BAT membranes was higher in EXP than in CON rats.
  • 4.4. It is concluded that feeding high-energy diets to rats induces similar post-receptor modifications of adenylate cyclase as found in genetic obese rodents.

  • 1.1. One adult male, eight pups (including two full term foetuses) and nine adult female harbour seals (Phoca vitulina) were analysed for indices of mixed function oxidase (MFO) activity.
  • 2.2. MFO activity was present in liver samples, but was at or below detection limits in samples of kidney, lung and pancreas.
  • 3.3. Hepatic ethoxyresorufin O-de-ethylase and benzo[a]pyrene hydroxylase activities were similar to those reported in other seals and in other mammals.

  • 1.1. Galactose oxidase is known to catalyze the oxidation of the C-6 hydroxymethyl group of galactose to an aldehyde group.
  • 2.2. When the products of a galactose oxidase-catalase treatment of raffinose were examined by gel filtration and ion exchange chromatography, we found that in addition to the expected 6'-aldehydoraffinose, two other components were present.
  • 3.3. One component was neutral and had an elution volume close to that of maltopentaose and on treatment with sodium borohydride or hypoiodite, this component was converted to raffinose or 6'-carboxyraffinose, respectively.
  • 4.4. Fast atom bombardment mass spectroscopy in the negative mode indicated that the major molecular ion had an M/Z of 1003.
  • 5.5. These data are consistent with this component being a dimer of 6'-aldehydoraffmose.

  • 1.1. The effects of prostaglandin (PG) E1, and I2 analogs (OP-41483 and OP-2507) on the Superoxide generation of human neutrophil NADPH oxidase (EC 1.6.99.6) in both whole-cell and cell-free systems were investigated.
  • 2.2. In a whole-cell system, OP-2507 inhibited the Superoxide generation by neutrophils exposed to phorbol myristate acetate concentration-dependently through its superoxide-scavenging action.
  • 3.3. The concentration of the drug required for 50% inhibition of the oxidase (IC50) was 21 μM.
  • 4.4. In a cell-free system, however, the drug in concentrations of < 100 μM did not inhibit the activation of NADPH oxidase by sodium dodecyl sulfate because of its inactivation by the detergent.
  • 5.5. Although PGE1 and OP-41483 did not inhibit the Superoxide production by stimulated neutrophils in a whole-cell system, they both inhibited the activation of NADPH oxidase in a cell-free system concentration-dependently, with IC50 values of 44 and 170 μM, respectively.
  • 6.6. In addition, in the cell-free system, the Km value for NADPH of the oxidase was unchanged by PGE1.
  • 7.7. The results suggest that the PGI2 analog, OP-2507, is a possible superoxide-scavenger and that PGE1 inhibits the NADPH oxidase activation by sodium dodecyl sulfate in a cell-free system concentration-dependently.

  • 1.1. In a continuing investigation of phycocyanin-membrane surface interaction, fluorescence quenching experiments were performed with a mixture of two populations of fluorescence probe-encapsulated phospholipid bilayer vesicles in the presence and absence of phycocyanin.
  • 2.2. These membrane vesicles were prepared with 1,2-dimyristoyl phosphatidylcholine (DMPC), cholesterol and a probe molecule.
  • 3.3. A fluorophore was encapsulated in one population of membrane vesicles, while a quencher was encapsulated in another population of membrane vesicles.
  • 4.4. The result was compared with those of experiments in the presence of other biomolecules, including albumin, cytochrome c, hemoglobin, myoglobin or RNA.
  • 5.5. Interestingly, a one-third reduction of the fluorescence intensity was observed in the mixture of these two populations of membrane vesicles in phycocyanin's presence.
  • 6.6. In contrast, the other biomolecules caused no significant reduction in the fluorescence intensity.
  • 7.7. These findings were evidence of a phycocyanin-induced membrane perturbation.
  • 8.8. This was further demonstrated by a phycocyanin-induced change in the thermotropic behavior of DMPC vesicles, as measured by differential scanning microcalorimetry.
  • 9.9. Such a unique property of phycocyanin is believed to be associated with its known membrane surface-interacting character.
  • 10.10. A possible phycocyanin-modulated membrane-membrane interaction was discussed.

  • 1.1. Isolated mitochondria from the whole southern armyworm larvae, Spodoptera eridania, show all of the characteristics of mammalian liver mitochondria, except for target site sensitivity to cyanide.
  • 2.2. The armyworm larval mitochondria are 17 times less sensitive to cyanide when compared to rat liver mitochondria and cannot be completely inhibited with extremely large doses.
  • 3.3. These data suggest the presence in the southern armyworm of either a cyanide-insensitive cytochrome oxidase, or the elaboration of cyanide-insensitive oxidative pathway reminiscent of an alternative oxidative pathway that is known to coexist in plants alongside the cyanide-sensitive pathway.

  • 1.1. Common carp (Cyprinus carpio) exposed to experimental temperatures of 12, 18, 24, 30 or 36°C for a 4-week period were used to investigate the effect of temperature acclimation on the frequency of opercular movement (FOM), growth and cytochrome c oxidase (CCO) activity in heart, liver and muscle.
  • 2.2. An exponential relationship between FOM and temperature after the first week (1010 =1.76) disappeared after the second week.
  • 3.3. The initially high FOM at temperatures of 30 or 36°C and the low FOM at 18 or 12°C changed over 4 weeks to approach the FOM of fish at 24°C.
  • 4.4. This change in the relationship of FOM to temperature from highly dependent to independent appeared to be thermal compensation.
  • 5.5. Heart and liver CCO activities were significantly affected by temperature, with the lowest activity at the approximate optimum temperature for growth, 24°C.
  • 6.6. Highest CCO activities for heart and liver occurred at both the highest and lowest temperatures.
  • 7.7. Among the three tissues, heart CCO activity was generally the highest and most affected by acclimation temperature.
  • 8.8. Muscle tissue had the lowest CCO activity and was unaffected by temperature.
  • 9.9. The high CCO activity at a cold acclimation of temperature 12°C was probably due to thermal compensation and the high activity at 36°C may have been a result of thermal stress.

  • 1.1. The hepatic d-aspartate oxidase activity was found to be higher in female ddY and ICR mice than in their male counterparts. On the contrary, the free d-aspartate content in the liver was lower in female mice than in male mice, suggesting that d-aspartate is actually metabolized by d-aspartate oxidase in vivo.
  • 2.2. Oral administration of d-aspartate to the animals increased the hepatic d-aspartate oxidase activity 2–3 fold in both genders without any significant difference in the rate of the increase between the genders.
  • 3.3. Several peroxisomal enzyme activities other than d-aspartate oxidase examined were not affected by this treatment.
  • 4.4. Experiments in vitro suggested that the increase in the d-aspartate activity might be explained in part by stabilization of the enzyme by d-aspartate.
  • 5.5. The administration of clofibrate, a peroxisome proliferator, to male mice, increased the hepatic d-aspartate oxidase activity with a significant simultaneous decrease of d-aspartate content in the liver, in agreement with a possible role of the enzyme n vivo.
  • 6.6. On the other hand, the administration of clofibrate or dehydroepiandrosterone to female mice decreased the d-aspartate oxidase activity.
  • 7.7. The peroxisome proliferators were suggested to act to eliminate the gender difference of hepatic d-aspartate oxidase activity in mice.

  • 1.1. Treatment of isolated rat liver mitochondria with methyl methacrylate (MM) produced membrane disruption as evidenced by the release of citrate synthase, and changes in the ultrastructure of mitochondria.
  • 2.2. At concentration 0.1%, MM uncoupled oxidative phosphorylation as evidenced by stimulation of state 4 respiration supported either by pyruvate plus malate or succinate (+rotenone) and ATP-ase activity in intact mitochondria.
  • 3.3. At concentration 1% MM stimulated ATP-ase activity in intact mitochondria and succinate (+rotenone) oxidation at state 4 and was without effect on this substrate oxidation at state 3.
  • 4.4. MM inhibited pyruvate plus malate oxidation either at state 3 or in the presence of uncoupling agents.
  • 5.5. MM inhibited the NADH oxidase of electron transport particles at a concentration which failed to inhibit either succinic oxidase or the NADH-ferricyanide reductase activity.
  • 6.6. The data presented suggest that in the isolated mitochondria MM inhibits NADH oxidation in the vicinity of the rotenone sensitive site of complex I.
  • 7.7. The general conclusion is that MM may block an electron transport and to uncouple oxidative phosphorylation in rat liver mitochondria. The overall in vitro effect would be to prevent ATP synthesis which could result in cell death under in vivo conditions.

  • 1.1. 3,3',4,4'-tetrachlorobiphenyl (PCB 77), but not hexachlorobenzene, induced liver micro-somal cytochrome P-450 (Cyt P-450), ethoxycoumarin-O-deethylase (ECOD) and ethoxyresorufin-O-deethylase (EROD) in rainbow trout. Maximum induction was observed in a PCB 77 injected group of fish (1.0mg/kg, i.p. injection) 13 days after the injections being 2, 10 and 50 times the values of non-induced fish, respectively.
  • 2.2. The apparent Km value of ethoxyresorufin of this induced group of fish differed only slightly from that of non-induced fish. The apparent Vmax value (EROD) was 50 times higher.
  • 3.3. Freezing small pieces of liver in liquid nitrogen did not produce cytochrome P-420.
  • 4.4. Fluorimetric and spectrophotometric measurements of EROD correlated.
