Three pesticides have been studied for their genotoxicity by the use of assays in the plant Crepis capillaris, aimed at measuring chromosomal aberrations, micronuclei and sister chromosome exchange (SCE). The fungicides Rubigan 12 EC (fenarimol) and Rovral 25 Flo (iprodione) and the insecticide Omite 57 E (propargite) are all widely used nowadays. The aim of our study was to evaluate the genotoxic effects of these pesticides at concentrations corresponding to those applied in agricultural practice. In preliminary experiments we found that these concentrations do not influence cell proliferation and do not inhibit the growth of root meristems. In all experiments formulated commercial products were used. From the results we conclude that the three pesticides did not induce chromosomal aberrations as estimated by metaphase and anaphase analyses. They were also not capable to induce SCE. Rubigan did not induce micronucleus formation even at the highest concentration tested, but Omite and Rovral markedly increased micronucleus formation. The MN response depended on the sampling time and the concentration used, which showed a significant dose-response correlation (r=0.978, P<0.01 and r=0.941, P<0.01, respectively). A greater increase in micronucleus frequency was observed after Rovral treatment, where the highest concentration gave a response 8-10-fold above the negative control. Both pesticides induced high frequencies of lagging chromosomes, even after exposure to the lower test concentrations. The presence of lagging chromosomes is an indication of anti-microtubule activity of the pesticides tested. This effect was more strongly expressed after exposure to the two higher concentrations of Omite and Rovral. In this case a complete destruction of the mitotic spindle was observed, resulting in C-mitoses as well as in numerical aberrations-polyploidy and aneuploidy. The present findings suggest that Omite and Rovral at concentrations comparable to those used in practice can be regarded as potential aneugens.  相似文献

Sources of light beams such as white fluorescent light, are present in our daily life to meet the needs of life in the modern world. This study was conducted with the objective of determining the possible genotoxic, cytotoxic and aneugenic effects caused by this agent in different stages of the cell cycle (G0/early G1, S, and late G2), using different cytogenetic parameters (sister chromatid exchanges––SCE, chromosome aberrations––CA, and detection of aneugenic effects) in lymphocytes from temporary cultures of human peripheral blood. WFL showed a genotoxic effect in vitro, expressed by an increase in the frequency of SCE's, regardless of the cell cycle stage. However, no increase in the frequency of CAs was observed. In addition, disturbances in cell cycle kinetics and chromosomal segregation were also observed. Taken together, such data may contribute to a better understanding and a different management in the use of phototherapy for some pathological conditions.  相似文献

Summary From a callus culture which was started from explants of a single plant of Crepis capillaris, a line was isolated which could grow on auxin- and kinetin-free medium (in contrast to the other lines of the culture). The calluses of this habituated line grew on hormone-free medium as teratomas. Regenerated plants were obtained from the habituated line and from other lines of the culture as control. In the secondary callus cultures which originated from these plants, the subcultures belonging to the initially transformed line no longer had the ability to grow on hormone-free medium. Since this line was also a chromosome mutant and the same mutation was present in its regenerated plants and in the secondary culture line of them, the loss of the auxin-autotrophy cannot be explained in this case on the basis of a selection of cells with karyotypes different from those of the primary culture.  相似文献

The presence of a large number of pollutants, including mutagenic agents in the environment is a problem of a major concern. Rapid progress in plant biotechnology, especially in the development of cell transformation methods, including the production of transformed roots – ‘hairy roots’ – has opened new possibilities to use transformed root cultures in plant bioassays for the evaluation mutagenic effects of different agents. We have used Crepis capillaris hairy roots for evaluation of cytogenetic effects of mutagenic treatment. Effects of maleic acid hydrazide (MH) and X-ray treatment were analysed in chromosomal aberration, sister chromatid exchange (SCE) and TUNEL tests. Comparison of cytogenetic effects in hairy roots and roots of seedlings showed a much higher sensitivity of hairy roots, which makes them convenient material for monitoring DNA damage after mutagenic treatment.  相似文献

The replication sites and morphological steps of chromosomal condensation during S period in the nuclei of Crepis capillaris root tip cells have been studied with light and electron microscopic autoradiography. From light microscopic autoradiographic observations, the S period can be divided with three portions, early S, mid S, and late S period. Labelled nuclei for each portion of the S period have also been found by using electron microscopic autoradiography. With electron microscopic autoradiography it has been found that in early, mid, and late S period, the replication sites are distributed in the electron transparent regions, interspersed with dense chromatin masses of variable size which are distributed throughout the nucleus. The time-dependent behavior of the label indicates that when compared with either mid or early replicated DNA, a majority of this chromatin, which contains predominantly late replicated DNA, is the earliest chromatin to be organized into the condensed chromatin. They are organized into the condensed chromatin within 15 min after the termination of replication.  相似文献

Coal is a mixture of a variety of compounds containing mutagenic and carcinogenic polycyclic aromatic hydrocarbons. Exposure to coal is considered as an important non-cellular and cellular source of reactive oxygen species that can induce DNA damage. In addition, spontaneous combustion can occur in coal mining areas, further releasing compounds with detrimental effects on the environment. In this study the comet assay was used to investigate potential genotoxic effects of coal mining activities in peripheral blood cells of the wild rodents Rattus rattus and Mus musculus. The study was conducted in a coal mining area of the Municipio de Puerto Libertador, South West of the Departamento de Cordoba, Colombia. Animals from two areas in the coal mining zone and a control area located in the Municipio de Lorica were investigated. The results showed evidence that exposure to coal results in elevated primary DNA lesions in blood cells of rodents. Three different parameters for DNA damage were assessed, namely, DNA damage index, migration length and percentage damaged cells. All parameters showed statistically significantly higher values in mice and rats from the coal mining area in comparison to the animals from the control area. The parameter “DNA Damage Index” was found to be most sensitive and to best indicate a genotoxic hazard. Both species investigated were shown to be sensitive indicators of environmental genotoxicity caused by coal mining activities. In summary, our study constitutes the first investigation of potential genotoxic effects of open coal mining carried out in Puerto Libertador. The investigations provide a guide for measures to evaluate genotoxic hazards, thereby contributing to the development of appropriate measures and regulations for more careful operations during coal mining.  相似文献

研究白蝴蝶(Syngonium podophyllum)乙醇提取物、苍耳(Xanthium sibiricum)乙醇提取物、机油乳剂和0.3%印楝素乳油对桃蚜(Myzus persicae)和萝卜蚜(Lipaphis erysimi)的控制效果.室内四因子(1/2实施)二次正交回归旋转组合设计测试对有翅蚜的忌避作用,结果表明,对有翅桃蚜的主要忌避作用物为苍耳乙醇提取物,当苍耳提取物与白蝴蝶乙醇提取物混用,以及机油乳剂和印楝素混用时,对桃蚜有翅蚜的忌避效果提高;而对萝卜蚜有翅蚜的忌避作用主要受苍耳提取物和白蝴蝶提取物的影响.这些干扰作用均是非线性的.田间试验结果,单独使用白蝴蝶提取物对有翅成蚜有较强的驱避作用;对萝卜蚜自然种群的干扰控制作用以4种植物提取物和药剂混配效果最好,达95.7%;对桃蚜自然种群干扰控制效果最好的则是白蝴蝶提取物,控制效果达87%;同时,白蝴蝶提取物与印楝素乳油混配以及苍耳提取物与印楝素乳油混配,对两种蚜虫的控制效果均达80%以上.  相似文献

We report here results on forward mutation induction (recessive lethal mutations, RL) in Drosophila spermatozoa and spermatids by the three 1,2-alkyl-epoxides ethylene oxide (EO), propylene oxide (PO) and butylene oxide (BO), at doses ranging from 47 to 24,000 ppm h for EO, 375 to 48,000 ppm h for PO, and 24,000 to 91,200 ppm h for BO. The results indicate for EO mutation induction at doses 500-fold below the LD50. In crosses of mutagenized NER+ males with NER+ females, the 500-fold increase in EO dose from 47 ppm h to 24,000 ppm h resulted in no more than a 17-fold enhanced mutant frequency in spermatozoa. This flat dose–response relationship is primarily the result of efficient repair of EO-induced DNA adducts in the fertilized egg, as was evident from the up to 40-fold or 240-fold increased mutant frequencies above NER or NER+ background levels, respectively, in crosses with NER females. With decreasing dose, / ratios decreased from 9 to 14 at high doses down to ≈1 at the two lowest doses, indicating that a small fraction of premutagenic lesions induced by EO cannot be repaired by the NER system of Drosophila. Linear extrapolation from high to low EO exposure led to an underestimation of the mutation frequency actually observed at low doses. The pattern of EO-induced ring chromosome loss (CL) differed in two respects from that observed for forward mutations: (a) an increase in CL frequencies was observed only at the two highest EO exposure levels, and (b) inactivation of the NER pathway by the mus201 mutant had no measurable effect on the occurrence of CL. The absence of a potentiating effect of mus201 on EO-induced clastogenicity suggests the formation of clastogenic DNA lesions not causing point mutations, and which are not repaired by NER. Consistent with an inversed correlation of reactivities towards N7-guanine and chain length of 1,2-alkyl-epoxides, the relative mutagenic efficiencies of EO:PO:BO are 100:7.2:1.8 for the NER+ groups, and 100:20:0.7 in the absence of NER. Although in Drosophila germ cells EO is also more effective as a clastogen than PO, the difference (EO:PO=100:58) is much smaller than for recessive mutations. These results provide another argument that DNA lesions generating base substitutions as opposed to those causing clastogenic damage may not be the same for these agents.  相似文献

This study aimed to examine the antimutagenic and anticarcinogenic potential of Phyllanthus amarus Schum. et Thonn. using the bacterial preincubation mutation assay and an in-vivo alkaline elution method for DNA single-strand breaks in hamster liver cells. The aqueous extract of the entire plant showed an antimutagenic effect against induction by 2-aminofluorene (AF2), 2-aminoanthracene (2AA) and 4-nitroquinolone-1-oxide (4-NQO) in Salmonella typhimurium strains TA98 and TA100, and in Escherichia coli WP2 uvrA/pKM101. All the results were dose-dependent; however, inhibition of N-ethyl-N-nitrosoguanidine (ENNG)-induced mutagenesis was observed only with S. typhimurium TA100. The extract also exhibited activity against 2-nitrofluorene (2NF) and sodium azide-induced mutagenesis with S. typhimurium TA98 and TA100, respectively. Based on the alkaline elution method, the plant extract prevented in vivo DNA single-strand breaks caused by dimethylnitrosamine (DMN) in hamster liver cells. When the extract was administered 30 min prior to the administration of DMN, the elution rate constant decreased more than 2.5 times, compared to that of control. These results indicate that P. amarus possesses antimutagenic and antigenotoxic properties.  相似文献

Summary The mutagenic action of mitomycin C (MMC) on the chromosomes of two reconstructed karyotypes of barley was studied. MMC-induced chromatid aberrations were found to be distributed non-randomly along the chromosomes. The regions situated next to the secondary constrictions of chromosomes 6 and 7 appeared to be clearly pronounced aberration hot spots. In these segments, intercalary deletions and duplication-deletions were the most frequently induced aberration types. The comparative analysis of the frequency and localization of MMC-induced aberrations in the chromosomes of the two karyotype variants, which differ from each other by the position of the hot spot segments, provided new evidence about the influence of the segment transposition on the hot spot expressivity. The most remarkable finding obtained in the study is that the size of the segment involved in both intercalary deletions and duplication-deletions proved to be strongly affected by the structural peculiarity of the reconstructed chromosome. The possible reasons underlying this finding are discussed.  相似文献

应用蜘蛛血细胞微核试验和单细胞凝胶电泳试验研究了两种杀虫剂啶虫脒和毒死蜱对蜘蛛头胸部和腹部血细胞DNA的损伤作用。结果表明:在啶虫脒和毒死蜱各供试浓度作用下,对蜘蛛血细胞微核率有明显的影响,与对照组相比有显著性差异(p<0.05,p<0.01);且随着两种农药浓度升高,血细胞微核率显著增加,存在明显的剂量-效应关系(啶虫脒浓度与星豹蛛头胸部血细胞微核率相关系数r=0.9284,腹部为r=0.9071;毒死蜱与星豹蛛头胸部血细胞微核率相关系数r=0.9841,腹部为r=0.9793);啶虫脒和毒死蜱对星豹蛛血细胞DNA损伤有明显的剂量-效应关系(啶虫脒浓度与星豹蛛头胸部血细胞DNA损伤相关系数r=0.9838,腹部为r=0.9834;毒死蜱与星豹蛛头胸部血细胞DNA损伤相关系数r=0.9807,腹部为r=0.9659);且两种农药在同种农药同一浓度作用下,对星豹蛛腹部血细胞微核率和DNA损伤程度要明显大于头胸部。  相似文献

The clastogenic activity of the antineoplastic alkylating agent 3β-hydroxy-13-amino-13,17-seco-5-androstan-17-oic- 13,17-lactam-p-bis (2-chloroethyl) aminophenoxy acetic acid (NSC 294859) and its congeners was studied in human lymphocyte cultures in vitro. Cells were exposed to several concentrations of the drugs for 24 h. It was found that NSC 294859 reduces the mitotic index and causes chromosome- as well as chromatid-type aberrations in a dose-dependent way. From its congeners, the alkylating agent (p-bis (2-chloroethyl) aminophenoxy acetic acid) induces the same phenomena but to a lesser extent, while the modified steroid (3β-hydroxy-13-amino-13,17-seco-5-androstan-17-oic-13,17-lactam) causes cytogenetic damages at the control level. These results favour the assumption that the antitumor activity of NSC 294859 is mainly based on its cytogenetic effects.  相似文献

Genotoxic activation of hydrazine (HZ), two symmetrical dialkylhydrazines, namely, 1,2dimethylhydrazine and 1,2-diethylhydrazine (SDMH and SDEH), thiourea (TU) and ethylene thiourea (ETU) has been evaluated by means of the w/w+ somatic assay of Drosophila. Both low bioactivation insecticide-susceptible (IS) and high biotransformation insecticide-resistant (IR) strains were used. The combined application of insecticide-susceptible and insecticide-resistant strains should, in principle, detect somatic cell recombinagens in the Drosophila melanogaster in vivo w/w+ assay. The IS strain was more susceptible to toxicity induced by the test chemicals than the IR stocks. Its performance in the biotransformation of the chemicals tested was rather poor. TU was inactive in all strains. With the active compounds, spot frequencies increased approximately linearly with dose for each spot type. SDEH gave a strong positive result in all three female genotypes exposed. HZ, ETU and SDMH were overall weakly positive in the IR strain Haag-79 (HG-R). Interestingly, ETU was clearly positive in the IR Hikone-R (HK-R) strain. A comparison of the recombinagenic potencies between the active and the weakly positive compounds, and among strains, showed pronounced genotype-dependent differences between the low and the high bioactivation strains. The ability of Drosophila to express several procarcinogens in relation to insecticide-resistance after activation catalyzed by CYP450 enzymes is discussed.  相似文献

Hexavalent chromium (Cr(VI)) is a well-designated human lung carcinogen, with solubility playing an important role in its carcinogenic potential. Although it is known that particulate or water-insoluble Cr(VI) compounds are more potent than the soluble species of this metal, the mechanisms of action are not fully elucidated. In this study, we investigated the hypothesis that the difference in potency between particulate and soluble Cr(VI) is due to more chronic exposures with particulate chromate because it can deposit and persist in the lungs while soluble chromate is rapidly cleared. Chronic exposure to both insoluble lead chromate and soluble sodium chromate induced a concentration and time-dependent increase in intracellular Cr ion concentrations in cultured human lung fibroblasts. Intracellular Pb levels after chronic exposure to lead chromate increased in a concentration-dependent manner but did not increase with longer exposure times up to 72 h. We also investigated the effects of chronic exposure to Cr(VI) on clastogenicity and found that chronic exposure to lead chromate induces persistent or increasing chromosome damage. Specifically, exposure to 0.5 μg/cm2 lead chromate for 24, 48 and 72 h induced 23, 23 and 27% damaged metaphases, respectively. Contrary to lead chromate, the amount of chromosome damage after chronic exposure to sodium chromate decreased with time. For example, cells exposed to 1 μM sodium chromate for 24, 48 and 72 h induced 23, 13 and 17% damaged metaphases, respectively. Our data suggest a possible mechanism for the observed potency difference between soluble and insoluble Cr(VI) compounds is that chronic exposure to particulate Cr(VI) induces persistent chromosome damage and chromosome instability while chromosome damage is repaired with chronic exposure to soluble Cr(VI).  相似文献

Peripheral blood lymphocytes were tested in vitro for genotoxic effects of cadmium chloride. Whole blood samples of four healthy, non-smoking subjects were preincubated with CdCl2 in concentrations of 10(-4), 10(-3), and 5 . 10(-3) mol/L for three hours before the cells were assessed for DNA-damage using the single cell alkaline gel electrophoresis assay (comet assay) or cultivated for chromosomal aberrations (CA), sister chromatid exchanges (SCE), and the micronucleus (MN) test. The comet assay showed notable interindividual differences. The results of the cytogenetic tests showed an increase in the frequency of CA, MN, and SCE with CdCl2 in the treated cultures, yet none was able to show a correlation between concentrations of cadmium chloride and the frequency of damages. The MN slides were stained with Giemsa and with DNA fluorochrome 4', 6'-diamidino-2-phenylindole (DAPI). The frequency of MN in slides stained with DAPI was significantly higher than in those stained with Giemsa, which might be due to an underestimation of small micronuclei in Giemsa-stained slides.  相似文献

The resazurin assay utilising microtitre-plate, described by Drummond and Waigh in 2000, has been modified to achieve more accuracy in the determination of the minimum inhibitory concentration (MIC) values of natural products, including crude extracts, chromatographic fractions or purified compounds against various bacterial strains. This modified resazurin method is simple, sensitive, rapid, robust and reliable, and could be used successfully to assess antibacterial properties of natural products.  相似文献

Recent findings of acrylamide (AA) in many common foods have sparked renewed interest in assessing human health hazards. AA was evaluated by the International Agency for Research on Cancer as probably carcinogenic to humans. For this reason, the aim of this study is to evaluate the potential genotoxic effect of AA using chromosomal aberration analysis and micronucleus (MN) test in mouse bone-marrow cells and morphological sperm abnormalities. The result of the present work indicated that treatment with a single dose of 10, 20, or 30 mg/kg b.wt. of AA for 24 h and the repeated dose of 10 mg/kg b.wt. for 1and 2 weeks induced a statistically significant increase in the percentage of chromosomal aberrations and micronuclei in bone- marrow cells. These percentages reduced significantly in all groups treated with AA and the protective agent l-carnitine. Also the results indicated that the dose 10, 20 and 30 mg/kg b.wt. of AA induced a statistically significant percentage of morphological sperm abnormalities compared with the control group. Such effect reached its maximum (7.24 ± 0.61) with the highest tested dose which reduced to (4.02 ± 0.58) in the group treated with the same dose of AA and l-carnitine. In conclusion, the results confirm the protective role of LC against the mutagenicity of AA.  相似文献

Cytogenetic tests are effective tools for monitoring the health status of livestock and improving their genetic value. Cytogenetic screening allows for the detection of animals carrying chromosomal aberrations and to avoid using them as breeders. Progress in karyotype monitoring, with new molecular probes and automation, has greatly increased the productivity of this procedure. Several genotoxicity tests are available to detect the possible presence and effects of pollutants or drugs. Among these, the micronucleus test and the Comet assay are the most convenient in terms of costs and benefits. Finally, analysis of telomeres, the end of chromosomes and markers of genomic instability, may be developed into a new marker of stress and genetic value.  相似文献

Cotton (Gossypium spp.) plant growth is an important time-specific agronomic character that supports the development of squares, flowers, boll retention, and yield. With the use of a mixed linear model approach, we investigated 14 cotton chromosome substitution (CS-B) lines and their chromosome-specific F2 hybrids for genetic changes in plant growth that was measured during the primary flowering time under two environments. The changes in additive and dominance variances for plant height and number of mainstem nodes are reported, showing that additive effects for these two traits were a key genetic component after initial flowering occurred in the field. Time-specific genetic variance components were also detected where phenotypic values observed at time t were conditioned on the events occurring at time t − 1, demonstrating new genetic variations arising at several time intervals during plant growth. Results also revealed that plant height and number of nodes shared some common influence due to additive effects during plant development. With the comparative analyzes, chromosomes associated with the genetic changes in plant growth were detected. Therefore, these results should add new understanding of the genetics underlying these time-specific traits.
