Wild strains of fruit flies (Diptera: Tephritidae) placed into laboratory rearing conditions are subjected to selection pressures caused by the diet, cages, density of flies, and other factors. Selection that changes mating behavior of the strain may result in less effective males released in sterile insect programs. Tests were performed to examine the effects of protein in diet and adult interactions on egg production and mating during sexual maturation of the Mexican fruit fly (Anastrepha ludens Loew) in laboratory cages. Flies were offspring of wild flies collected from Chiapas or Nuevo Leon, Mexico, and reared on Valencia oranges. Experiments demonstrated effects of yeast hydrolysate protein in adult diet and pairing with males on production of mature and immature eggs, numbers of females producing eggs, and mating with females aged 15 d. Addition of protein to 4% fructose in the adult diet approximately tripled mature egg production in females maintained for the total maturation period with an equal number of males. Females that matured without males produced approximately 33% more-mature eggs when fed protein than those fed no protein. Total egg production of females matured without males and fed sugar only or sugar with protein was more than twice that of females matured with males. Tests to examine the effects of male and female diet separately on female egg production showed slightly higher egg production in females fed protein, or females paired with males fed protein, but these differences were not significant. The most definitive effects were that combining wild strain females and males in cages during maturation reduced egg production. This effect was greatest when flies were not fed protein.  相似文献

The inability to properly balance energy intake and expenditure with nutrient supply forms the basis for some of today's most pressing health issues, including diabetes and obesity. Mechanisms of nutrient homeostasis may also lie at the root of dietary restriction, a manipulation whereby reduced nutrient availability extends lifespan and ameliorates age-related deteriorations in many species. The traditional belief that the most important aspect of the diet is its energetic (i.e. caloric) content is currently under scrutiny. Hypotheses that focus on diet composition and highlight more subtle characteristics are beginning to emerge. Using Drosophila melanogaster , we asked whether diet composition alone, independent of its caloric content, was sufficient to impact behavior, physiology, and lifespan. We found that providing flies with a yeast-rich diet produced lean, reproductively competent animals with reduced feeding rates. Excess dietary sugar, on the other hand, promoted obesity, which was magnified during aging. Addition of dietary yeast often limited or reversed the phenotypic changes associated with increased dietary sugar and vice versa, and dietary imbalance was associated with reduced lifespan. Our data reveal that diet composition, alone and in combination with overall caloric intake, modulates lifespan, consumption, and fat deposition in flies, and they provide a useful foundation for dissecting the underlying genetic mechanisms that link specific nutrients with important aspects of general health and longevity.  相似文献

Recent studies in fruit flies have imposed dietary restriction (DR) by diluting yeast and have reported increased lifespan as the yeast-to-sugar ratio decreased. In this study, the effects of DR on the lifespan of Bactrocera dorsalis were investigated using constant-feeding diets with different yeast:sugar ratios and an intermittent-feeding diet in which flies ate every sixth day. Antioxidant enzyme activities and the malondialdehyde concentration were also measured in virgin females under constant-feeding DR protocols to investigate their relationships with lifespan. The results showed that B. dorsalis lifespan was significantly extended by DR, and carbohydrate-enriched diet may be important for lifespan-extension. Female flies lived significantly longer than males at all dietary levels under both feeding regimes, indicating no interaction between diet and sex in determining lifespan. Antioxidant enzyme activities increased with the amount of yeast increased in the diets (0–4.76%) between starvation and DR treatments, indicating that the antioxidants may have influences in determining lifespan in B. dorsalis under starvation and DR treatments. However, antioxidants cannot keep up with increased oxidative damage induced by the high yeast diet (25%). These results revealed that the extension of lifespan by DR is evolutionarily conserved in B. dorsalis and that yeast:sugar ratios significantly modulate lifespan in this species.  相似文献

The nutritional requirements of Drosophila have mostly been studied for development and reproduction, but the minimal requirements for adult male and female flies for lifespan have not been established. Following development on a complete diet, we find substantial sex difference in the basic nutritional requirement of adult flies for full length of life. Relative to females, males require less of each nutrient, and for some nutrients that are essential for development, adult males have no requirement at all for lifespan. The most extreme (and surprising) sex differences were that chronic cholesterol and vitamin deficiencies had no effect on the lifespan of adult males, but they greatly decreased lifespan in females. Female oogenesis rather than chromosomal karyotype and mating status is the key cause of this gender difference in life‐sustaining nutritional requirements. These data are important to the way we understand the mechanisms by which diet modifies lifespan.  相似文献

It is established that glucose restriction extends yeast chronological and replicative lifespan, but little is known about the influence of amino acids on yeast lifespan, although some amino acids were reported to delay aging in rodents. Here we show that amino acid composition greatly alters yeast chronological lifespan. We found that non-essential amino acids (to yeast) methionine and glutamic acid had the most significant impact on yeast chronological lifespan extension, restriction of methionine and/or increase of glutamic acid led to longevity that was not the result of low acetic acid production and acidification in aging media. Remarkably, low methionine, high glutamic acid and glucose restriction additively and independently extended yeast lifespan, which could not be further extended by buffering the medium (pH 6.0). Our preliminary findings using yeasts with gene deletion demonstrate that glutamic acid addition, methionine and glucose restriction prompt yeast longevity through distinct mechanisms. This study may help to fill a gap in yeast model for the fast developing view that nutrient balance is a critical factor to extend lifespan.  相似文献

It is generally believed that butterflies (and other holometabolous insects) rely primarily on reserves accumulated during the larval stage for reproduction, whereas the carbohydrate-rich adult diet is thought to mainly cover energy requirements. In at least some species though, realization of the full reproductive potential is extensively affected by post-eclosion nutrition. While the importance of carbohydrates is fairly well understood, the role of adult-derived amino acids and micronutrients is controversial and largely unknown, respectively. We here focus on the effects of different adult diets on female reproduction in the tropical, fruit-feeding butterfly Bicyclus anynana (Nymphalidae). Carbohydrates were the most important adult-derived nutrients affecting reproduction. Adding amino acids, vitamins or minerals to sucrose-based solutions did not yield a reproductive output equivalent to that of fruit-fed females, which showed the highest performance throughout. This suggests that either not yet identified compounds of fruit substantially contribute to reproduction, or that resource congruence (the use of nutrient types in a specified ratio) rather than any specific nutrient component is of key importance. Apart from adult income, realized fecundity depended on egg size and longevity, with the former dominating when dietary quality was low, but the latter when quality was high. Thus, the egg size-number trade-off seems to be affected by female nutrition.  相似文献

Six experiments on growing rats were conducted to measure the nutritive value of diets based on cereals (wheat and barley, 1:1) and ethanol-grown yeast. Optimum protein provision in isonitrogenous diets (11.3% crude protein) determined on the basis of live weight gain, feed efficiency and protein efficiency ratio was found to be 60 and 40% for cereal and yeast protein, respectively, corresponding to a diet containing 84.5% cereals and 13.2% yeast (Experiments 1 and 2). The addition of single amino acids (l-lysine, 0.05–0.15%; dl-methionine, 0.1–0.3%) or their combinations (l-lysine + dl-methionine, 0.05 + 0.1%; 0.1 + 0.1%; 0.05 + 0.2%; and 0.1 + 0.2%) to the diet containing 13.2% yeast had no significant effect on either growth rate or feed efficiency (Experiments 3 and 4). A non-fortified diet alone proved to be comparable in nutritive value to a standard diet containing high-quality protein components.The diet with yeast providing 20% and cereals providing 80% of the total protein (5.55% yeast, 92.15% cereals) with combined supplement of amino acids (dl-methionine 0.2% + l-lysine 0.2% + l-isoleucine 0.1%; Experiment 5) promoted growth rate and feed efficiency similar to the standard diet, in spite of the fact that the experimental diet protein-content was about 3% lower. In a separate experiment (Experiment 6), similar results were achieved with the supplement of l-lysine alone (0.15% of the diet).As suggested by the results of model experiments on rats, the nutritive value of diets based on cereals and ethanol-grown yeast, possibly supplemented with amino acids, is comparable with that of diets containing animal protein components.  相似文献

Life‐history parameters and the fitness of tephritid flies are closely linked to diet. Studies of locomotor behaviour can provide insights to these links, although little is known about how locomotor behaviour is influenced by diet. In the present study, video recordings of Queensland fruit flies Bactrocera tryoni Froggatt (Diptera: Tephritidae) (‘Q‐flies’) that are maintained individually in cages are used to determine how diet affects the activity patterns (flight, walking, grooming, inactivity) of males and females at ages ranging from 4 to 30 days. The frequency and total duration of activities over 10‐min trials are affected by diet, age and sex. Supplementation of diet with hydrolysed yeast results in a higher frequency and duration of flight in flies of all ages and both sexes. The effect of diet on other activities varies with age. Q‐flies fed sugar only increase walking frequency steadily from 4 to 30 days post‐eclosion, whereas flies fed sugar + yeast have higher walking frequencies at 4 and 10 days than flies fed sugar only, although they then exhibit a sharp decline at 30 days post‐eclosion. The frequency and duration of inactivity remain consistent in flies fed sugar + yeast, whereas flies fed sugar only exhibit a marked increase in inactivity from 4 to 30 days post‐eclosion. Compared with older flies, 4 day‐old Q‐flies fed sugar only spend considerably more time grooming. The potential of activity monitoring as a quality control test for flies that are mass‐reared for use in sterile insect technique programmes is discussed.  相似文献

Nutrition is commonly a powerful determinant of sexual performance in insects, and recent studies have found this to be the case in Queensland fruit flies (Tephritidae: Bactrocera tryoni Froggatt; 'Q-flies'); male Q-flies allowed to self-regulate intake of yeast hydrolysate, a rich source of amino acids and vitamins used in most mass-rearing programmes (protein) and sucrose (carbohydrate), had greatly enhanced sexual performance compared with males provided only sucrose. While some yeast hydrolysate is clearly beneficial for the sexual performance of adult male Q-flies, the questions of what proportion of yeast hydrolysate in the diet is sufficient to yield full benefits, or is too much, have not yet been addressed. To address these questions, the present study assessed sexual performance and longevity of adult male Q-flies maintained on diets containing various proportions of yeast hydrolysate and sucrose. Male Q-flies maintained as adults on dry mixtures containing 9%, 17% or 25% yeast hydrolysate had mating probability, mating latency, copula duration and longevity similar to those provided yeast hydrolysate and sucrose in separate dishes and allowed to self-regulate intake. As in previous studies, while longevity was unaffected we found a marked reduction in sexual performance when the flies were completely denied access to yeast hydrolysate, and the few that did mate had relatively short copulations. At the other extreme, flies receiving diets with high levels of yeast hydrolysate (50%, 75%, 83% and 91%) suffered marked reductions both in longevity and in mating performance.  相似文献

The New World screwworm, Cochliomyia hominivorax Coquerel, is mass reared for screwworm eradication initiatives that use the sterile insect technique. New methods for rearing have helped to reduce the cost of the eradication program. We examined the effect and interaction of three temperatures (24.5, 29.5 and 34.5 degrees C), two diets (2% spray-dried blood plus 0.05% vitamins and corn syrup carrageenan) and three population densities (300, 400, and 500 flies/cage) on egg production, egg hatch, number of observable fertilized eggs, mortality (male and female) and ovarian development. The three population densities did not affect any of the parameters monitored. Using the protein diet increased egg production at all temperatures. Diet did not affect egg hatch or female mortality. Male mortality was significantly greater when fed the protein diet and reared at 24.5 degrees C and 34.5 degrees C. Egg hatch was significantly less when the flies were reared at 34.5 degrees C. When exposed to high temperatures (37 degrees C and 40 degrees C) egg production, egg hatch, fertility and mortality were adversely affected. At the higher temperatures, yolk did not adequately form during oogenesis. When compared to the normal rearing photoperiod (12 L:12 D), short photoperiod (1 L:23 D) increased egg production, egg hatch and fertility but lowered mortality.  相似文献

Reduced mechanistic target of rapamycin (mTOR) signalling extends lifespan in yeast, nematodes, fruit flies and mice, highlighting a physiological pathway that could modulate aging in evolutionarily divergent organisms. This signalling system is also hypothesized to play a central role in lifespan extension via dietary restriction. By collating data from 48 available published studies examining lifespan with reduced mTOR signalling, we show that reduced mTOR signalling provides similar increases in median lifespan across species, with genetic mTOR manipulations consistently providing greater life extension than pharmacological treatment with rapamycin. In contrast to the consistency in changes in median lifespan, however, the demographic causes for life extension are highly species specific. Reduced mTOR signalling extends lifespan in nematodes by strongly reducing the degree to which mortality rates increase with age (aging rate). By contrast, life extension in mice and yeast occurs largely by pushing back the onset of aging, but not altering the shape of the mortality curve once aging starts. Importantly, in mice, the altered pattern of mortality induced by reduced mTOR signalling is different to that induced by dietary restriction, which reduces the rate of aging. Effects of mTOR signalling were also sex dependent, but only within mice, and not within flies, thus again species specific. An alleviation of age‐associated mortality is not a shared feature of reduced mTOR signalling across model organisms and does not replicate the established age‐related survival benefits of dietary restriction.  相似文献

Caloric restriction (CR) has been widely accepted as a mechanism explaining increased lifespan (LS) in organisms subjected to dietary restriction (DR), but recent studies investigating the role of nutrients have challenged the role of CR in extending longevity. Fuelling this debate is the difficulty in experimentally disentangling CR and nutrient effects due to compensatory feeding (CF) behaviour. We quantified CF by measuring the volume of solution imbibed and determined how calories and nutrients influenced LS and fecundity in unmated females of the Queensland fruit fly, Bactocera tryoni (Diptera: Tephritidae). We restricted flies to one of 28 diets varying in carbohydrate:protein (C:P) ratios and concentrations. On imbalanced diets, flies overcame dietary dilutions, consuming similar caloric intakes for most dilutions. The response surface for LS revealed that increasing C:P ratio while keeping calories constant extended LS, with the maximum LS along C:P ratio of 21:1. In general, LS was reduced as caloric intake decreased. Lifetime egg production was maximized at a C:P ratio of 3:1. When given a choice of separate sucrose and yeast solutions, each at one of five concentrations (yielding 25 choice treatments), flies regulated their nutrient intake to match C:P ratio of 3:1. Our results (i) demonstrate that CF can overcome dietary dilutions; (ii) reveal difficulties with methods presenting fixed amounts of liquid diet; (iii) illustrate the need to measure intake to account for CF in DR studies and (iv) highlight nutrients rather than CR as a dominant influence on LS.  相似文献

Improvements were made in the meridic larval diet for Sitophilus oryzae by replacing the minerals and vitamins supplied by dietary brewer's yeast and wheat germ with mineral and vitamin mixtures. The effects of different concentrations of individual vitamins were studied with the improved diet containing 20% casein. In later tests the dietary casein was replaced with a mixture of 15 amino acids. The results indicated that these larvae, which contain an associated bacteria-like micro-organism, required thiamine, nicotinic acid, pyridoxine, folic acid, and biotin in the diet. No requirement for riboflavin or pantothenic acid could be demonstrated on either the 20% casein diet or the amino acid diet possibly due to contamination of the cornstarch with these two vitamins. In addition, the larvae did not require choline or inositol for the growth of one larval generation.The asymbiotic larvae of S. granarius failed to develop on the improved casein diet indirectly implicating the symbiotes in a nutritional rôle; however, the symbiotes present in S. oryzae apparently do not provide B vitamins. Larvae of S. oryzae failed to develop when the concentration of casein was reduced to 10% while growth was maintained with a 10% concentration of the amino acid mixture. Casein is not an optimal source of amino acids for this species.  相似文献

Commercially raised feeder insects used to feed captive insectivores are a good source of many nutrients but are deficient in several key nutrients. Current methods used to supplement insects include dusting and gut‐loading. Here, we report on the nutrient composition of four species of commercially raised feeder insects fed a special diet to enhance their nutrient content. Crickets, mealworms, superworms, and waxworms were analyzed for moisture, crude protein, fat, ash, acid detergent fiber, total dietary fiber, minerals, amino acids, fatty acids, vitamins, taurine, carotenoids, inositol, and cholesterol. All four species contained enhanced levels of vitamin E and omega 3 fatty acids when compared to previously published data for these species. Crickets, superworms, and mealworms contained β‐carotene although using standard conversion factors only crickets and superworms would likely contain sufficient vitamin A activity for most species of insectivores. Waxworms did not contain any detectable β‐carotene but did contain zeaxanthin which they likely converted from dietary β‐carotene. All four species contained significant amounts of both inositol and cholesterol. Like previous reports all insects were a poor source of calcium and only superworms contained vitamin D above the limit of detection. When compared to the nutrient requirements as established by the NRC for growing rats or poultry, these species were good sources of most other nutrients although the high fat and low moisture content of both waxworms and superworms means when corrected for energy density these two species were deficient in more nutrients than crickets or mealworms. These data show the value of modifying the diet of commercially available insects as they are growing to enhance their nutrient content. They also suggest that for most insectivores properly supplemented lower fat insects such as crickets, or smaller mealworms should form the bulk of the diet. Invasive animals depend on finding a balanced nutritional intake to colonize, survive, and reproduce in new environments. This can be especially challenging during situations of fluctuating cold temperatures and food scarcity, but phenotypic plasticity may offer an adaptive advantage during these periods. We examined how lifespan, fecundity, pre‐oviposition periods, and body nutrient contents were affected by dietary protein and carbohydrate (P:C) ratios at variable low temperatures in two morphs (winter morphs WM and summer morphs SM) of an invasive fly, Drosophila suzukii. The experimental conditions simulated early spring after overwintering and autumn, crucial periods for survival. At lower temperatures, post‐overwintering WM lived longer on carbohydrate‐only diets and had higher fecundity on low‐protein diets, but there was no difference in lifespan or fecundity among diets for SM. As temperatures increased, low‐protein diets resulted in higher fecundity without compromising lifespan, while high‐protein diets reduced lifespan and fecundity for both WM and SM. Both SM and WM receiving high‐protein diets had lower sugar, lipid, and glycogen (but similar protein) body contents compared to flies receiving low‐protein and carbohydrate‐only diets. This suggests that flies spend energy excreting excess dietary protein, thereby affecting lifespan and fecundity. Despite having to recover from nutrient depletion after an overwintering period, WM exhibited longer lifespan and higher fecundity than SM in favorable diets and temperatures. WM exposed to favorable low‐protein diet had higher body sugar, lipid, and protein body contents than SM, which is possibly linked to better performance. Although protein is essential for oogenesis, WM and SM flies receiving low‐protein diets did not have shorter pre‐oviposition periods compared to flies on carbohydrate‐only diets. Finding adequate carbohydrate sources to compensate protein intake is essential for the successful persistence of D. suzukii WM and SM populations during suboptimal temperatures.  相似文献

A variety of insects are commonly fed to captive insectivores but detailed nutritional analyses are only available for the most commonly fed species. Soldier fly larvae, Turkestan cockroach nymphs, tebo worms, and adult house flies were analyzed for moisture, protein, fat, ash, acid detergent fiber, neutral detergent fiber, minerals, amino acids, fatty acids, vitamins, and selected carotenoids. The acid detergent fiber was analyzed for amino acids to estimate chitin content. Nutrient content varied widely between the four insect species. Ranges for the macronutrients were as follows: moisture (60.2–74.8%), crude protein (15.5–19.7%), crude fat (1.9%–29.4%), acid detergent fiber (1.4–3.0%), neutral detergent fiber (2.6–3.8%), and ash (0.8–3.5%). Energy content ranged from a low of 918 kcal/kg for house flies to 2,977 kcal/kg for tebo worms. The chitin content of these four species ranged from 6.7 to 21.0 mg/kg. The nutrients most likely to be deficient when these species of insects are used as food for insectivores are vitamin A, vitamin D, calcium, vitamin E, thiamine, iodine, and vitamin B12. The number of nutrients deficient vs. the NRC requirements for rats on an energy basis by insect species was as follows: soldier fly larvae (3), tebo worms (15), Turkestan cockroach nymphs (5), and adult house flies (6). These data are valuable in helping assess the nutrient intake of captive insectivores and in developing gut‐loading diets to improve the nutrient intake of captive insectivores. Adult dietary yeast modulates mortality rate and reproduction of the Mediterranean fruit fly, Ceratatis capitata. In the medfly, a sugar-only diet leads to low mortality rates and reduced reproduction; addition of dietary yeast increases both mortality and egg laying. In Drosophila melanogaster low availability of dietary yeast is known to increase life span and reduce the rate of reproduction. Despite these similarities, because of differences in experimental design it remains unclear whether a common physiological mechanism modulates the effect of diet on survival. Here, we investigate how mortality rate and reproduction in D. melanogaster respond to the treatment regime used to study the medfly: no-yeast versus full diet. We find that adult medfly and D. melanogaster have opposite responses to the absence of yeast: D. melanogaster have high mortality when on no-yeast diet; when switched to full diet, D. melanogaster reduce mortality rates to the level presented by females continuously maintained on yeast. This reduction in mortality is accompanied by increased fecundity. These patterns are observed in all tested wildtype stocks, but flies made sterile by mutation in the gene oo18 RNA-binding protein (orb) lack this response. D. melanogaster, unlike medflies, appear to require adult dietary yeast to maintain maximal survival, and the capacity to assimilate yeast for somatic processes is one wildtype function of the gene orb.  相似文献

Senescence is shaped by age-dependent trade-offs between fitness components. Because males and females invest different resources in reproduction, the trade-offs behind age-dependent reproductive effort should be resolved differently in the sexes. In this study, we assess the effects of diet (high carbohydrate and low protein vs. equal carbohydrate and protein) and mating (once mated vs. virgin) on lifespan and age-dependent mortality in male and female field crickets (Teleogryllus commodus), and on male calling effort. Females always had higher actuarial ageing rates than males, and we found a clear lifespan cost of mating in females. Mated males, however, lived longer than virgin males, possibly because virgins call more than mated males. The fastest age-dependent increases in mortality were among mated males on the high-carbohydrate diet. Males on a high-carbohydrate diet showed a faster increase in calling effort earlier in life, and a more pronounced pattern of senescence once they reached this peak than did males on a diet with equal amounts of protein and carbohydrates. Our results provide evidence that the cost of mating in this cricket species is both diet and sex-dependent, and that the underlying causes of sex differences in life-history traits such as lifespan and senescence can be complex.  相似文献

Dietary restriction extends lifespan in a wide variety of animals, including Drosophila, but its relationship to functional and cognitive aging is unclear. Here, we study the effects of dietary yeast content on fly performance in an aversive learning task (association between odor and mechanical shock). Learning performance declined at old age, but 50‐day‐old dietary‐restricted flies learned as poorly as equal‐aged flies maintained on yeast‐rich diet, even though the former lived on average 9 days (14%) longer. Furthermore, at the middle age of 21 days, flies on low‐yeast diets showed poorer short‐term (5 min) memory than flies on rich diet. In contrast, dietary restriction enhanced 60‐min memory of young (5 days old) flies. Thus, while dietary restriction had complex effects on learning performance in young to middle‐aged flies, it did not attenuate aging‐related decline of aversive learning performance. These results are consistent with the hypothesis that, in Drosophila, dietary restriction reduces mortality and thus leads to lifespan extension, but does not affect the rate with which somatic damage relevant for cognitive performance accumulates with age.  相似文献

Fruit‐feeding butterflies are among the longest lived Lepidoptera. While the use of pollen‐derived amino acids by Heliconius butterflies has been interpreted as important for the evolution of extended lifespans, very little is known about the life‐history consequences of frugivory. This issue is addressed by investigating effects of four adult diets (sugar, sugar with amino acids, banana, and moistened banana) on lifespan and reproduction in the fruit‐feeding butterfly Charaxes fulvescens Aurivillius (Lepidoptera: Nymphalidae). Female butterflies were collected from Kibale National Park, Uganda, and kept individually in cages near their natural habitat and data were collected on lifespan, oviposition, and hatching of eggs. Lifespan in captivity was longer for the sugar and the amino acid cohort, than for the banana cohorts. The longitudinal pattern of oviposition was erratic, with many days without oviposition and few periods with high numbers of eggs laid. Butterflies typically did not lay eggs during their 1st week in captivity and the length of the period between capture and first reproduction was significantly shorter for butterflies fed moistened banana. The length of the reproduction period (first reproduction–last reproduction in captivity) and the reproduction rate (total number of eggs/length of the reproduction period) did not differ significantly between the diet treatments. Those fed with amino acid and moistened banana had significantly higher egg hatchability than those fed with sugar and banana. We found no evidence for a lifespan cost of reproduction. Our results show that (1) female C. fulvescens can use amino acids in their diet for laying fertile eggs, (2) more wing‐wear does correlate with lower survival in captivity (indicating aging in the wild), but not with intensity of reproduction (providing no evidence for reproductive aging), and (3) fruit‐feeding butterflies may be dietary restricted in the field.  相似文献
