[背景] 氨氮浓度会明显影响亚硝化单胞菌的活性,但氨氮浓度对吸附态亚硝化单胞菌菌种的抑制动力学尚缺乏研究。[目的] 研究氨氮浓度对3种吸附态亚硝化单胞菌(Nitrosomonas eutropha CZ-4、Nitrosomonas halophila C-19和Nitrosomonas europaea SH-3)的影响。[方法] 以碳酸钙作为吸附基质,设定氨氮浓度为25-1 000 mg/L,测定3种亚硝化单胞菌(N.eutropha CZ-4、N. halophila C-19和N. europaea SH-3)的亚硝氮积累速率与最大比生长速率,并通过Edwares2模型建立氨氧化的抑制动力学方程。[结果] N. halophila C-19在初始氨氮浓度为50-100 mg/L时的亚硝氮积累最快,N. europaea SH-3的亚硝氮积累则在初始氨氮浓度为50-200 mg/L时最快,而N. eutropha CZ-4则适于在初始氨氮浓度为50-400 mg/L时积累亚硝氮;N. eutropha CZ-4的最大比生长速率出现在初始氨氮浓度为50-400 mg/L时,明显高于N. halophila C-19(25-100 mg/L),而N. europaea SH-3的生长速度在初始氨氮浓度为50-800 mg/L区间内无显著差异;N. europaea SH-3的KI(922.76 mg/L)显著高于N. eutropha CZ-4(597.88 mg/L),而CZ-4的KI又显著高于N. halophila C-19(186.24 mg/L),N. europaea SH-3的Km(72.06 mg/L)显著高于N. halophila C-19(23.23 mg/L)。[结论] 3种吸附态亚硝化单胞菌的生长和氨氧化对氨氮浓度变化的响应存在明显差异,对于认识不同亚硝化单胞菌在不同氨氮浓度污水中的功能并开发相应的工程技术具有重要意义。  相似文献

Aims:  To monitor emissions of NH3 and N2O during composting and link these to ammonia oxidation rates and the community structure of ammonia oxidizing bacteria (AOB).
Methods and Results:  A laboratory-scale compost reactor treating organic household waste was run for 2 months. NH3 emissions peaked when pH started to increase. Small amounts of N2O and CH4 were also produced. In total, 16% and less than 1% of the initial N was lost as NH3-N and N2O-N respectively. The potential ammonia oxidation rate, determined by a chlorate inhibition assay, increased fourfold during the first 9 days and then remained high. Initially, both Nitrosospira and Nitrosomonas populations were detected using DGGE analysis of AOB specific 16S rRNA fragments. Only Nitrosomonas europaea was detected under thermophilic conditions, but Nitrosospira populations re-established during the cooling phase.
Conclusions:  Thermophilic conditions favoured high potential ammonia oxidation rates, suggesting that ammonia oxidation contributed to reduced NH3 emissions. Small but significant amounts of N2O were emitted during the thermophilic phase. The significance of different AOBs detected in the compost for ammonia oxidation is not clear.
Significance and Impact of Study:  This study shows that ammonia oxidation occurs at high temperature composting and therefore most likely reduces NH3 emissions.  相似文献

Energy metabolism and the production of ammonia in hybridoma cell culture and its inhibitory effects on cell growth are reviewed. The interactive roles of glucose and glutamine metabolism affect the rate of production of ammonia, and these interactions are described. It is shown that growth inhibition usually occurs between 2–4 mM ammonia although some cell lines have been shown to adapt to much higher concentrations, particularly in continuous culture. In batch cultures cell growth appears to be particularly susceptible to increased ammonia concentrations during the early stages of growth; ammonia increased the rate of cell death in the late stage of batch growth. The specific productivity of monoclonal antibodies is much less sensitive to the released ammonia than is growth; lower volumetric productivities relate to the lower viable cell concentrations which are achieved at the high ammonia levels. Techniques to prevent ammonia accumulation or remove ammonia selectively have been relatively unsuccessful to date.  相似文献

三江平原典型小叶章湿地土壤氨挥发特征及影响因素   总被引:4,自引:0,他引:4  
采用通气法对三江平原典型草甸小叶章湿地和沼泽化草甸小叶章湿地土壤的氨挥发进行了原位测定,并对其主要影响因素进行了分析。结果表明,二者的氨挥发速率在生长季内的变化趋势基本一致,7月中旬前出现两次挥发高峰和一次低值,之后整体呈严格单调下降趋势,后者的氨挥发速率较高,平均为前者的1.35±0.53倍;二者累计氨挥发量的变化趋势也基本一致,7月中旬前增加迅速,且值比较接近;之后增加缓慢,但其值发生明显分异,表现为后者大于前者;生长季内,典型草甸小叶章湿地土壤的氨挥发总量为6.35 kg N.hm-2,而沼泽化草甸小叶章湿地则为6.87 kg N.hm-2,二者之比为1∶1.08;氮素物质基础不是影响二者氨挥发过程的重要限制因素,大气温度及其所引起的其它温度波动是影响氨挥发速率变化的重要因素;降水及土壤水分波动与散失是引起氨挥发速率局部波动的重要原因;土壤pH和质地是导致氨挥发速率普遍较低的根本原因;而各种因素综合作用的结果则是引起二者氨挥发速率和氨挥发量变化及差异的主要原因。  相似文献

Laboratory experiments have shown appreciable losses of ammonia after injection of anhydrous ammonia into dry and wet soils. In this study losses of ammonia injected into a moist (tension 10 kPa), dry (tension 160 kPa) and a wet (tension 1.6 kPa) sandy loam were measured under field conditions using wind tunnels. Losses were insignificant from a moist soil. However losses from a dry and a wet soil were 20% and 50% of injected ammonia, respectively. From the dry soil, losses of gaseous ammonia took place within the first hours after injection, which indicates a rapid transport through cracks and voids. From the wet soil, 20% of the injected ammonia was lost more gradually between 6 h and 6 d. This indicates that upward movement of water due to evaporation may be the cause of these ammonia losses which proceeded for longer periods.  相似文献

Rats were fed a diet containing ammonium for up to 6 months. High ammonia levels were attained in brain. The amount of polymerized tubulin in microtubules increased, while the amount of free tubulin remained unchanged. Polymerization of tubulin from brain of ammonium fed rats (30 min, 37°C) was approximately 60% of control. Depolymerization of the microtubules was also affected and took approximately 3 times longer than in controls. These results indicate that both assembly and disassembly of tubulin in brain are impaired by high ammonia levels. Interestingly, the amount of microtubule-associated proteins was not affected.  相似文献

Suspension cultured cells of a blast-resistant rice genotype (Oryza sativa L. cv. Gigante Vercelli) were treated with cell wall hydrolysates prepared from the fungal pathogen Magnaporthe oryzae. As a consequence, a complex pattern of phenylalanine ammonia lyase time course specific activity levels was evident. Ion-exchange chromatographic fractionation of crude extracts suggested that the early (6 h) and the late (48-72 h after elicitation) increase of activity relied upon the sequential induction of two different isoenzymes. The relative expression levels of 11 genes putatively coding for a phenylalanine ammonia lyase were measured by semi-quantitative capillary gel electrophoresis of RT-PCR products. Two genes were indeed found to be induced by treatments with the hydrolysate, and data were validated by real-time PCR. Conversely, only the early-responsive enzyme form was observed following elicitation in a blast-sensitive rice genotype (cv. Vialone nano). Therefore, the late-responsive isoform may represent a candidate gene to select for decreased sensitivity to blast.  相似文献

Partial nitrification has proven to be an economic way for treatment of industrial N-rich effluent, reducing oxygen and external COD requirements during nitrification/denitrification process. One of the key issues of this system is the intermediate nitrite accumulation stability. This work presents a control strategy and a modeling tool for maintaining nitrite build-up. Partial nitrification process has been carried out in a sequencing batch reactor at 30 degrees C, maintaining strong changing ammonia concentration in the reactor (sequencing feed). Stable nitrite accumulation has been obtained with the help of an on-line oxygen uptake rate (OUR)-based control system, with removal rate of 2 kg NH4 (+)-N x m(-3)/day and 90%-95% of conversion of ammonium into nitrite. A mathematical model, identified through the occurring biological reactions, is proposed to optimize the process (preventing nitrate production). Most of the kinetic parameters have been estimated from specific respirometric tests on biomass and validated on pilot-scale experiments of one-cycle duration. Comparison of dynamic data at different pH confirms that NH3 and NO2- should be considered as the true substrate of nitritation and nitratation, respectively. The proposed model represents major features: the inhibition of ammonia-oxidizing bacteria by its substrate (NH3) and product (HNO2), the inhibition of nitrite-oxidizing bacteria by free ammonia (NH3), the INFluence of pH. It appears that the model correctly describes the short-term dynamics of nitrogenous compounds in SBR, when both ammonia oxidizers and nitrite oxidizers are present and active in the reactor. The model proposed represents a useful tool for process design and optimization.  相似文献

Increasing usage of nitrogen fertilizer for food production has resulted in severely environmental problems of nutrients enrichment. This study aimed to examine the response of ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) to a long-term nitrogen fertilization in Tibetan alpine meadow. The abundance and composition of both AOB and AOA were assessed using quantitative real-time PCR, cloning and sequencing techniques based on amoA gene under different fertilization gradient (0, 30, 60, 90, and 120 g m?2 year?1). Our results showed that, abundances of AOA amoA genes (ranging from 1.48 × 109 to 2.00 × 109 copies per gram of dry soil) were significantly higher than those of AOB amoA genes (1.25 × 107 to 2.62 × 108 copies per gram of dry soil) under fertilization scenario. The abundance of AOB amoA genes increased with increasing nitrogen fertilization, whereas fertilization had little effect on AOA abundance. Sequences of clone libraries of the different treatments revealed that AOB communities were dominated by representatives of Cluster 4, constituting 48.94–64.44% in each clone library. Sequences of Clusters 9, 1 and 2 were prevalent in soils under higher fertilization. All archaeal amoA sequences recovered were affiliated with the soil/sediment clade and marine sediment clade, and no significant difference was observed on the community structure among different fertilization treatments. Variations in the AOB community structure and abundance were linked to ammonium-N and soil pH induced by different fertilization treatments. These results showed that the abundance and structure of the AOB community respond to the fertilization gradient, not AOA.  相似文献

In the sequence of events leading from ammonia to N2 during the process of biotransformation of inorganic nitrogen compounds, the weakest link, with respect to our knowledge and understanding of the organisms involved, is nitrification. In particular, this is true for the oxidation of ammonia to nitrite. The enzymes have not been thoroughly studied, and the enzymatic mechanisms have not been identified. Almost any biochemical and physiological aspect studied proved to be controversial, and major ecological questions still remain unanswered. The effect of phosphate (PO4 +3) and pH in regulating nitrate (NO3) and ammonia (NH3 +) uptake by phytoplankton was investigated in two Oklahoma lakes using 15N tracers. Addition of PO4 +3 above ambient concentrations had a negligible effect on the rate of uptake of NO3 or NH3 +. Manipulation of pH of lake water had little effect on uptake of either NO3 or NH3 +. A correlation analysis suggested that NO3 is not used by phytoplankton when NH3 + concentrations exceed about 210 µg NH3 +-N(1)–1.  相似文献

自然界中氮的生物地球化学循环主要由微生物驱动,由固氮作用、硝化作用、反硝化作用和氨化作用来完成。过去数十年间,随着异养硝化、厌氧氨氧化和古菌氨氧化作用的发现,人们对环境中氮素循环认识逐步深入,提出了多种脱氮途径新假说。对湖泊生态系统中氮素的输入、输出及其在水体、沉积物和水土界面的迁移转化过程进行了概括,对湖泊生态系统中反硝化和厌氧氨氧化脱氮机理及脱氮效率的最新研究进展进行了探讨,并对以后的氮素循环研究进行了展望。  相似文献

A continuous nitrification reactor treating saline wastewater was operated for almost 1 year under low dissolved oxygen (DO) levels (0.15-0.5 mg/L) and high nitrogen loadings (0.26-0.52 kg-N/(m(3) day)) in four phases. The diversity and abundance of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) were analyzed by cloning, terminal restriction fragment length polymorphism (T-RFLP) and quantitative polymerase chain reaction (qPCR). The results showed that there were only one dominant AOA species and one dominant AOB species in the reactor in all of the four experimental phases. The amoA gene of the dominant AOA only had a similarity of 89.3% with the cultured AOA species Nitrosopumilus maritimus SCM1. All of the AOB species detected in the reactor belong to Nitrosomonas genus and it was found that the AOB populations changed with the ammonium loadings and DO levels. The abundance of AOB in the reactor was ~40 times larger than that of AOA, and the ratio of AOB to AOA increased significantly up to ~2,000 to ~4,000 with the increase of ammonium loading, indicating that AOB are much more competitive than AOA in high ammonium environments and probably AOA play a less important role than AOB in the nitrification reactors.  相似文献

Oxidation of [2,3-14C]succinate in the intramitochondrial Krebs cycle was used as a probe to investigate the effect of ammonia on protein incorporation and Krebs cycle oxidation of succinate carbons in isolated rat hepatocytes. At low concentrations of ammonium chloride (0.1 to 0.5 mM) a slight increase in14CO2 formation from [2,3-14C]succinate was observed, however, the stimulatory effect of insulin was significantly reduced. Insulin failed to cause any stimulation of succinate carbons incorporation into hepatocyte protein in the presence of ammonium chloride. Addition of ammonium chloride also depressed the movement of tracer carbons into the gluconeogenesis pathway. The activity of the amphibolic amino acid pool was significantly enhanced by ammonia. The data presented in this paper lend strong support to the Krebs-cycle depletion theory of hepatic coma. They also suggest that reduced mitochondrial Krebs cycle activity caused by increased amphibolic depletion of substrates results in loss of insulin sensitivity in ammonia toxicity.Special issue dedicated to Dr. Santiago Grisolia.  相似文献

采用通气法测定了北方冬小麦/夏玉米轮作体系田间土壤的原位氨挥发。结果表明,与冬小麦施用基肥相比,夏玉米追肥后土壤的氨挥发速率很快升高,但军发高峰期持续时间较短,最大氨挥发速率亦低于冬小麦,冬小麦拨节期追肥,氨挥发速率低且呈波动变化,未出现高峰值,从整个生长季节来看,冬小麦不施氮和每公顷施氮120、240、360kg时的累计挥发量分别为4.4、6.9、13.0、38.4kgN/hm^2,夏玉米为8.4、15.1、20.0、26.1kgN/hm^2。按我国北方冬小麦/夏玉米播种面积1864.4万hm^2计,每年由氨挥发向大气排放的氨素达23.8-120.2万t,其中17.2-96.4万t来自氮肥,相当于氮肥投入的2.1%-9.5%。  相似文献

The effect of antioxidants on ammonia induced CNS and renal pathobiology in 10 sheep infested by L. Cuprina larvae was investigated. The condition produces severe dermatitis, proliferation of macro-phages and hyperammonaemia, and free radicals may therefore be involved in the pathogenesis. Five of the sheep (treated group) were given daily intramuscular (im) injections of 2 g sodium ascorbate, 5.9 g dl-alpha tocopherol (11 days) and 3 g desferrioxamine mesylate (6 days) with 70 mg oral butylated-hydroxyanisole (11 days). The treatment prevented rises in jugular ammonia, creatinine, urea, sodium and pH, and decreases in water intake, urine output and glucose. The findings showed that antioxidants prevented ammonia induced CNS and renal pathobiology and suggest that free radicals contribute to the pathogenesis of the condition.  相似文献
