Uterine myoelectrical activity was recorded in seven pregnant sheep covering the period between 13 and 75 days post-coitum. Activity in the myometrium was present at day 13 and took the form of intermittent spikes of low amplitude. Bursts of spikes of irregular duration became noticeable between days 25 and 40 but most were not coordinated throughout the myometrium. Coordinated bursts of myoelectrical activity, which could be recorded at several sites simultaneously, first appeared between 40 and 65 days. These bursts had similar characteristics to the myoelectrical activity associated with uterine contractions during the last third of gestation. The myoelectrical activity showed a progressive increase in amplitude during the first half of gestation. There was no relationship between plasma progesterone levels and the increase in amplitude or appearance of coordinated bursts of uterine activity.  相似文献

The effect of botulinum toxin type A (BTX-A) on rat pyloric myoelectrical activity in vivo and the content and distribution of substance P (SP) in pylorus were investigated, respectively, with electromyography, radioimmunoassay, and immunohistochemistry. A pair of electrodes for recording pyloric myoelectrical activity and a guide cannula for drug injection were implanted into the pylorus. The changes of pyloric myoelectrical activity were recorded followed vehicle, 10, 20, and 40 U/kg body mass of BTX-A injection. Pyloric tissues were dissected for radioimmunoassay and immunohistochemistry after recording. The 3 dosages of BTX-A injections caused the reduction of slow wave of pyloric myoelectrical activity in amplitude but not in frequency and the diminishment of spike activity in amplitude and spike burst. The inhibitory effect of 20 U/kg BTX-A was significantly different from that of 10 U/kg (p<0.05), but not from the effect of 40 U/kg administration (p>0.05). After BTX-A intrasphincteric injection, SP content was reduced in the pylorus, and cell number of SP-immunoreactivity was decreased more in myenteric nerve plexus of circular muscle and in mucosa of pylori. In conclusion, BTX-A inhibits pyloric myoelectrical slow activity in amplitude and spike activity and weakens pyloric smooth muscle contractility depending on threshold of dose or concentration. BTX-A-induced inhibition of pyloric myoelectrical activity implies a mechanism of inhibiting SP release from the autonomic and enteric nervous terminals in the pylorus.  相似文献

Intravenous injection of small doses of E. coli endotoxin in sheep inhibited phasic contractions of forestomach and altered the myoelectrical activity of the antrum, duodenal bulb and jejunum. Previous intracerebroventricular administration of naloxone at a dose without effect when given intravenously (10 μg.kg?1) antagonized the endotoxin-induced inhibition of forestomach and alteration of the gastrointestinal myoelectrical activity.These findings suggest the involvement of a central opiate mechanism in the effects of E. coli endotoxin on gastro-intestinal motor activity.  相似文献

In conscious miniature pigs, with implanted electrodes in the wall of the antrum pylori, duodenum, jejunum, and ileum, the influence of IV infusions of CCK-8 (17.5 and 175 pM/kg/min) on gastrointestinal myoelectrical activity was measured. Although both doses under study induced a decrease in antral spike activity, only the higher dose resulted in an overall decrease in integrated myoelectrical activity. In the ileum both doses augmented spiking activity during the infusion, but inhibited electrical activity after the end of the infusion. No response was observed in the duodenum and jejunum. The experiments demonstrate the overall inhibitory effect of CCK-8 on antral electrical activity and its stimulatory influence on ileal smooth muscle.  相似文献

The presence of two types of fast myoelectrical activities, medium fast activity and fast activity, has been demonstrated previously in the electromyogram of colon in normal children and in the rat by the authors. An absence of medium fast activity in Hirschsprung's disease and in experimental aganglionosis of colon in the rat has also been described. In the present study the fast components of colonic myoelectrical activity were analysed during the procedures affecting ganglionic transmission. It was observed that ganglionic stimulants, such as balloon inflation, and intra-arterial injections of acetylcholine and small amounts of nicotine, increased the spike activity and the frequency of medium fast activity without affecting fast activity. The intra-arterial injections of ganglionic blocking agents, such as nicotine in large amounts and pentolinium tartrate, completely abolished the medium fast activity. These observations suggest that the ganglionic activity is responsible for the genesis of medium fast activity and that the absence of cholinergic ganglionic transmission is the most important single factor for the reported altered electromyogram pattern in aganglionosis.  相似文献

The effects of propranolol on the myoelectrical activity of the stomach, pylorus and duodenum were investigated in chronic experiments on rabbits. During the first phase of propranolol action a positive gastro-chronotropic effect was accompanied by a negative cardio-chronotropic effect. The gastro-chronotropic effect was 3 times higher than cardio-chronotropic one. The second phase of the propranolol effect manifested with periodical fluctuation of the gastroduodenal myoelectrical activity during stable bradycardia. Marked reactivity of the gastro-duodenum and periodical fluctuation of its activity are evidence of vital importance of the autoregulation mechanisms for gastroduodenal myoelectric activity.  相似文献

The influence of intravenous infusion (20 min) of carbachol (CAR) and isoproterenol (IPN) on myoelectrical activity of the uterus, vagina, and duodenum was studied in 6 conscious and unrestrained hens. IPN infusions (1.5, 7.5, and 15 micrograms/hen/min) were performed before oviposition, and CAR infusions (0.5, 1.5, and 5 micrograms/hen/min) were administered after oviposition. CAR infusions resulted in a significant stimulation of myoelectrical activity of the vagina (1.5 and 5 micrograms/min) and the duodenum (5 micrograms/min), and in a significant increase (45 +/- 6%) in heart rate (HR), as compared to control values. These effects were blocked by previous i.v. injections of atropine (0.4 mg/hen). IPN infusions induced a significant inhibition of myoelectrical activity of the middle (7.5 and 15 micrograms/min) and caudal (all doses) uterine segments, of the vagina (7.5 and 15 micrograms/min), and of the duodenum (15 micrograms/min), and a significant tachycardia (39 +/- 14%) as compared to control values. The inhibitory effects of IPN on all organs studied and the stimulatory action on HR were suppressed by propranolol (0.24 mg/hen). The sensitivity of the vagina to both drugs argues for a more active participation of this oviductal part in the mechanism of oviposition.  相似文献

The activation of the duodenum motor activity has been established. The changes in the motor activity were connected with trypsin and trypsin inhibitor activity of serum. Myoelectrical activity of the stomach was estimated in healthy Croatian subjects using the latest multi-channel percutaneous electrogastrograph. The aim of the study was to determine normal values of gastric myoelectrical activity for the population of Croatia. The study included 120 healthy subjects of both sexes, various age groups, body mass index values, and mode of lead placement. The measurement was performed 60 min before and 60 min after test meal. The following parameters of gastric myoelectrical activity were analyzed: dominant frequency (DF, c/min); dominant frequency within normal range (DFNR, %); coefficient of variation for dominant frequency (CVDF); dominant strength (DS, mV); postprandial increase intensity in dominant strength (PPIIDS; %); bradygastria (c/min, %); tachygastria (c/min, %); and arrhythmia. DF for the study group as a whole was around 3 cpm, at the normal range midpoint, and all other parameters were within the normal limits. On postprandial measurement, the rate of arrhythmias showed a significant decline. Age was found to influence DF, CVDF and arrhythmia in preprandial but not in postprandial period, whereas sex influenced DF, DS and bradygastria in preprandial period, and DF, CVDF, PPIIDS and tachygastria in postprandial period. The mode of lead placement had no impact on the electrogastrographic parameters observed. The values of gastric myoelectrical activity recorded in healthy Croatian subjects were within the normal range of the values defined by previous studies across Europe.  相似文献

Gastric myoelectrical and mechanical activity was recorded in miniature pigs using chronically implanted electrodes and strain gauge force transducers. Semiautomated methods were devised to obtain quantitative evaluations of the electrical and mechanical parameters measured in fasted and fed animals. The patterns of gastric myoelectrical activity in pigs were, on the whole, similar to the patterns described in dogs, including regular cyclic control activity and spike response activity associated with muscle contraction. However, several points were peculiar to the species studied: conduction velocity of pacesetter potentials increased only moderately in the antrum; tachygastria never occurred in the experiments; in response to a standard meal, the frequency of pacesetter potentials gradually increased; mechanical activity proceeded at its maximal force immediately after feeding and for a long period; no evidence of 'migrating electrical complexes' was found in the stomach during fasting. The 40-min period following administration of a test meal appeared especially suitable for pharmacological or physiological experiments in which inhibitory factors are to be tested on the stomach.  相似文献

In order to test our hypothesis that there is seasonal variation in digestion and absorption of dietary carbohydrate from the intestine, we previously determined the amount of unabsorbed carbohydrate after breakfast by the breath hydrogen test in the four seasons. In pursuing our hypothesis further, we also recorded gastric myoelectrical activity before and after the breakfasts. In the current report, we describe the seasonality of gastric myoelectrical activity. Twenty-six Japanese female subjects were studied in winter, spring, summer and autumn. The cutaneous electrogastrogram was analysed by spectral analysis to compute the pre- and post-prandial dominant slow wave frequency (DF), and percentage of the 2 - 4 cpm gastric slow wave (Normal %). Two-factor ANOVA indicated that there was no significant seasonal variation in DF and Normal %. These results indicate that seasonal variations in digestion and absorption of dietary carbohydrate are caused by factors other than gastric and small intestinal motility.  相似文献

Objective: To investigate the therapeutic potential of retrograde gastric electrical stimulation (RGES) for obesity in a rodent model of obesity. Research Methods and Procedures: The study was performed in 12 obese Zucker rats implanted with two pairs of gastric serosal electrodes, one pair for stimulation and the other for recording intrinsic gastric myoelectrical activity. It was composed of an acute study in three sessions to study the effect of RGES on intrinsic gastric myoelectrical activity and acute food intake and a chronic phase to study the short‐term effect of RGES on weight. RGES was performed through the distal stomach using long pulses at a frequency of tachygastria (known to induce gastric hypomotility). Results: RGES completely entrained intrinsic gastric myoelectrical activity and turned it into tachygastria at a certain strength. RGES reduced acute food intake compared with the control (p < 0.01). A 2‐week treatment of RGES resulted in a significant reduction in food intake (p = 0.002) and a significantly greater weight loss than sham stimulation (p = 0.004). Discussion: RGES at a tachygastrial frequency reduces food intake and results in weight loss in obese Zucker rats, and its effect is probably attributed to the introduction of tachygastria in the stomach.  相似文献

Intestinal bacteria, particularly those adhering to intestinal epithelial cells, are exposed to electric fields and currents generated by the muscular activity of the small intestine. This activity displays a regular pattern known as the myoelectrical migrating complex (MMC). In order to explore the possibility that these endogenous electric fields could affect bacterial growth, a digitised duodenal signal obtained via serosal electrodes from a healthy calf was recorded and then applied via platinum electrodes to Escherichia coli cultures. The culture tubes were placed within a Faraday shield, incubated at 37 degrees C with shaking, and stimulated by the electric current for 5 or 8 h. The growth of E. coli stimulated by the electric current was significantly altered compared to those of non-stimulated controls: after a period of intensive growth, inhibition of cell division was observed. This was not the case when the bacteria with lon mutation were used. Moreover, synchronic bacterial culture could not be achieved in the presence of the MMC-related electric field. These results suggest that the myoelectrical activity of the duodenum, through action on cell membrane, can affect cell division of intestinal bacteria.  相似文献

Purpose

A lack of fatigue-related muscle contractile property changes at time of perceived physical exhaustion and greater central than peripheral fatigue detected by twitch interpolation technique have recently been reported in cancer survivors with fatigue symptoms. Based on these observations, it was hypothesized that compared to healthy people, myoelectrical manifestation of fatigue in the performing muscles would be less significant in these individuals while sustaining a prolonged motor task to self-perceived exhaustion (SPE) since their central fatigue was more prominent. The purpose of this study was to test this hypothesis by examining electromyographic (EMG) signal changes during fatiguing muscle performance.

Methods

Twelve individuals who had advanced solid cancer and cancer-related fatigue (CRF), and 12 age- and gender-matched healthy controls performed a sustained elbow flexion at 30% maximal voluntary contraction till SPE. Amplitude and mean power frequency (MPF) of EMG signals of the biceps brachii, brachioradialis, and triceps brachii muscles were evaluated when the individuals experienced minimal, moderate, and severe fatigue.

Results

CRF patients perceived physical “exhaustion” significantly sooner than the controls. The myoelectrical manifestation of muscular fatigue assessed by EMG amplitude and MPF was less significant in CRF than controls. The lower MPF even at minimal fatigue stage in CRF may indicate pathophysiologic condition of the muscle.

Conclusions

CRF patients experience less myoelectrical manifestation of muscle fatigue than healthy individuals near the time of SPE. The data suggest that central nervous system fatigue plays a more important role in limiting endurance-type of motor performance in patients with CRF.  相似文献

The effect of i.v. infusions of bombesin and somatostatin, administered either separately or in combination, on migrating myoelectric complexes (MMCs) in the small intestine were studied in conscious, fasted rats. The myoelectrical activity was recorded by means of three bipolar electrodes chronically implanted into the duodenum and jejunum. Infusion of bombesin (0.5, 0.9 and 3 pmol . kg-1 . min-1) interrupted the MMC and induced irregular spiking activity similar to that observed on feeding. Only after the highest dose a consistent inhibition of the MMCs and a significant increase (P less than 0.05) of the spiking activity were achieved at all recording levels. Somatostatin (90 pmol . kg-1 . min-1) did not interrupt the MMC, but reduced significantly the incidence of the activity fronts and spiking activity of the MMCs (P less than 0.05). The effects of bombesin (3 pmol . kg-1 . min-1) on the MMC pattern were inhibited by simultaneous infusion of somatostatin (P less than 0.05). In a second series of experiments, using anesthetized rats, infusion of bombesin (0.5 and 3 pmol . kg-1 . min-1) increased the plasma concentration of neurotensin- gastrin-like immunoreactivities in a dose-dependent manner. The results show that bombesin alters the myoelectrical activity of the small intestine from a fasting to a fed pattern. Since the effect of bombesin was inhibited by the hormone release inhibitor somatostatin, it is suggested that the effect of bombesin on MMC may be secondary to the release of gastrointestinal peptides, such as neurotensin or gastrin.  相似文献

The aim of the study was to determine the impact of demographic and anthropometric parameters on the gastric myoelectrical activity characteristics in a healthy Croatian population. The influence of age, sex, body mass index (BMI) and menstrual cycle phase on the gastric myoelectrical activity characteristics was assessed. The study included 120 healthy subjects of both sexes (60 male and 60 female), divided into four age groups (18-35, 36-50, 51-65 and > 65 years) and three BMI groups (BMI < 25, 25-30 and > 30). Female subjects of reproductive age were divided into three groups according to menstrual cycle phase (day 1-3, day 4-8 and day 9-20 of menstruation). All study subjects underwent percutaneous electrogastrography (EGG) for 60 min before and 60 min after a test meal. The following parameters of the gastric myoelectrical activity were observed: dominant frequency (DF); dominant frequency within normal range (DFNR, %); coefficient of variation for dominant frequency (CVDF); dominant strength (DS. mV); postprandial increase intensity in dominant strength (PPIIDS, %); bradygastria (BG, c/min, %); tachygastria (TG, c/min, %); and arrhythmia (AR). Age was found to influence preprandial but not postprandial DFNR, CVDF and AR. Sex influenced preprandial DF, CVDF, DS and BG, and postprandial DF, CVDF, PPIIDS and TG. BMI exerted an impact on preprandial TG and AR, and postprandial DF, CVDF and AR. The phase of menstrual cycle influenced DF in preprandial period and none of EGG parameters in postprandial period.  相似文献

Ghrelin is a recently discovered peptide in the endocrine cells of the stomach, which may stimulate gastric motility via the vagal nerve pathway. However, the mechanism of ghrelin-induced changes in gastrointestinal motility has not been clearly defined. The purpose of this study was to investigate the pharmacological effects of ghrelin on gastric myoelectrical activity and gastric emptying in rats, and to investigate whether cholinergic activity is involved in the effects of ghrelin. The study was performed on Sprague-Dawley rats implanted with serosal electrodes for electrogastrographic recording. Gastric slow waves were recorded from fasting rats at baseline and after injection of saline, ghrelin, atropine, or atropine+ghrelin. Gastric emptying of non-caloric liquid was measured by the spectrophotometric method in conscious rats. Intravenous administration of rat ghrelin (20 microg/kg) increased not only dominant frequency, dominant power and regularity of the gastric slow wave but also the gastric emptying rate when compared with the control rats (P<0.01, P<0.05, P<0.05, P<0.001 respectively). These stimulatory actions of ghrelin on both gastric myoelectrical activity and gastric emptying were not fully eliminated by pretreatment with atropine sulphate. These results taken together suggest that ghrelin may play a physiological role in the enteric neurotransmission controlling gastric contractions in rats.  相似文献

Regional activities of phosphoinositide-specific phospholipase C (PLC) were measured after lateral fluid percussion (FP) brain injury in rats. The activity of PLC on phosphatidylinositol 4,5-bisphosphate (PIP2) in the rat cortex required calcium, and at 45 M concentration it increased PLC activity by about ten-fold. The activity of PLC was significantly increased in the cytosol fraction in the injured (left) cortex (IC) at 5 min, 30 min and 120 min after brain injury. However, in the same site, increases were observed in the membrane fraction only at 5 min after brain injury. In both the contralateral (right) cortex (CC) and ipsilateral hippocampus (IH), the activity of PLC was increased in the cytosol only at 5 min after brain injury. These results suggest that increased activity of PLC may contribute to increases in levels of cellular diacylglycerol and inositol trisphosphate in the IC (the greatest site of injury), and to a smaller extent in the IH and CC, after lateral FP brain injury. It is likely that this increased PLC activity is caused by alteration in either the levels or activities of one or more of its isozymes (PLC, PLC, and PLC) after FP brain injury.  相似文献

Our aims were to describe the myoelectrical activity in the single very thin layer of muscle of the canine gallbladder. The study was performed on 22 freshly removed canine gallbladders. Electrical activity was studied by the single sucrose-gap method and contractility of the tissue was measured simultaneously using a force transducer. The strips (15 x 1 mm) from different regions of gallbladder (fundus, corpus, neck) were cut in longitudinal, circular and oblique axes. The sucrose-gap apparatus together with connecting tubes, solutions and electrodes were kept at 37 degrees C and the initial tension applied to the tissue was set to 1 g. In 82.7% of recordings, spontaneous myoelectrical activity consisted of regular rhythmic changes in membrane potential similar to slow waves recorded in intestinal tissue. The overall mean frequency was 11.4 +/- 5.2 (mean +/- SD) cycles per min: 11.1 +/- 4.4 cycles per min in fundus, 11.9 +/- 6.2 cycles per min in corpus and 10.8 +/- 3.8 cycles per min in the neck of the gallbladder. In 84.2% of cases electrical activity correlated with mechanical activity and preceded it. No significant differences were seen between the electrical patterns in strips with different orientations or from the different regions of the gallbladder.  相似文献
