Summary Cl transport in apical membrane vesicles derived from bovine tracheal epithelial cells was studied using the Cl-sensitive fluorescent indicator 6-methoxy-N-(3-sulfopropyl) quinolinium. With an inwardly directed 50 mM Cl gradient at 23°C, the initial rate of Cl entry (J Cl) was increased significantly from 0.32±0.12 nmol · sec–1 · mg protein–1 (mean±sem) to 0.50±0.07 nmol · sec–1 · mg protein–1 when membrane potential was changed from 0 to +60 mV with K/valinomycin. At 37°C, with membrane potential clamped at 0 mV, there was a 34±7% (n=5) decrease inJ Cl from a control value of 0.37±0.03 nmol · sec–1 · mg protein–1 upon addition of 0.2mm diphenylamine-2-carboxylate. The following did not alterJ Cl significantly (J Cl values gives as percent change from control): 50mm cis Na (–1±5%), 0.1mm furosemide (–3±4%), 0.1mm furosemide in the presence of 50mm cis Na (–5±2%), 0.1mm H2DIDS (–18±9%), a 1.5 pH unit inwardly directed H gradient (–7±7%), and 0.1mm H2DIDS in the presence of a 1.5 unit pH gradient (4±18%). With inward 50mm anion gradients, the initial rates of Br and I entry (J Br andJ 1, respectively) were not significantly different fromJ Cl.J Cl was a saturable function of Cl concentration with apparentK d of 24mm and apparentV max of 0.54 nmol · sec–1 · mg protein–1. Measurement of the temperature dependence ofJ Cl yielded an activation energy of 5.0 kcal/mol (16–37°C). These results demonstrate that Cl transport in tracheal apical membrane vesicles is voltage-dependent and inhibited by diphenylamine-2-carboxylate. There is no significant contribution from the Na/K/2Cl, Na/Cl, or Cl/OH(H) transporters. The conductive pathway does not discriminate between Cl, Br, and I and is saturable. The low activation energy supports a pore-type mechanism for the conductance.  相似文献

Summary The purpose of this study was to characterize the basolateral membrane of the S3 segment of the rabbit proximal tubule using conventional and ion-selective microelectrodes. When compared with results from S1 and S2 segments, S3 cells under control conditions have a more negative basolateral membrane potential (V bl=–69 mV), a higher relative potassium conductance (t K=0.6), lower intracellular Na+ activity (A Na=18.4mm), and higher intracellular K+ activity (A K=67.8mm). No evidence for a conductive sodium-dependent or sodium-independent HCO 3 pathway could be demonstrated. The basolateral Na–K pump is inhibited by 10–4 m ouabain and bath perfusion with a potassium-free (0-K) solution. 0-K perfusion results inA Na=64.8mm,A K=18.5mm, andV bl=–28 mV. Basolateral potassium channels are blocked by barium and by acidification of the bathing medium. The relative K+ conductance, as evaluated by increasing bath K+ to 17mm, is dependent upon the restingV bl in both S2 and S3 cells. In summary, the basolateral membrane of S3 cells contains a pump-leak system with similar properties to S1 and S2 proximal tubule cells. The absence of conductive bicarbonate pathways results in a hyperpolarized cell and larger Na+ and K+ gradients across the cell borders, which will influence the transport properties and intracellular ion activities in this tubule segment.  相似文献

Summary Sulphate uptake by rabbit ileal brush border membrane vesicles was stimulated by a transmembrane sodium gradient ([Na+] o >[Na+] i ), but not by a similar potassium gradient.35SO 4 2– influx (J oi SO4 ) from outside (o) to inside (i) these vesicles was a hyperbolic function of [SO 4 2– ] o and the affinity constant for anion transport was strongly influenced by [Na+] o (100mm Na+,K t SO4 =0.52mm SO 4 2– ; 10mm Na+,K t SO4 =4.32mm SO 4 2– ).J t SO4 was a sigmoidal function of [Na+] o at pH 7.4 for both low (0.2m) and high (4.0mm) [SO 4 2– ] o . The Na+-dependency ofJ t SO4 was examined at pH 6.0, 7.4, and 8.0 (same pH inside and outside). At pH 6.0 and 7.4 sigmoidal Na+-dependentJ t SO4 exhibited nonlinear Eadie-Hofstee plots indicative of a transport mechanism capable of binding a variable number of sodium ions over the [Na+] o range used. Hill plots of anion transport under these conditions displayed slopes near unity at low [Na+] o and slopes approximating 2.0 at higher cation concentrations. At pH 8.0, Na+-dependentJ t SO4 was hyperbolic and showed linear Eadie-Hofstee and Hill plots, the latter with a single slope near 1.0. When a H+ gradient was imposed across the vesicle wall (pH i =8.0, pH o =6.0), Na+-dependentJ t SO4 was hyperbolic and significantly increased at each [Na+] o over values observed using bilateral pH 8.0. In contrast, a H+ gradient oriented in the opposite direction (pH i =6.0, pH o =8.0) led to Na+-dependentJ t SO4 that was sigmoidal and significantly lower at each [Na+] o than values found using bilateral pH 6.0. Electrogenicity ofJ t SO4 at pH 8.0 for both high and low [Na+] o was demonstrated by using a valinomycin-induced transmembrane electrical potential difference. At pH 6.0, electrogenicJ t SO4 occurred only at low [Na+] o (5mm); anion transfer was electroneutral at 50mm Na+. A model is proposed for proton regulation of sodium sulphate cotransport where flux stoichiometry is controlled by [H+] i and sodium binding affinity is modified by [H+] o . Preliminary experiments with rabbit proximal tubular brush border membrane vesicles disclosed similarJ t SO4 kinetic properties and a common transport mechanism may occur in both tissues.  相似文献

Summary The Na+ requirement for active, electrogenic Cl absorption byAmphiuma small intestine was studied by tracer techniques and double-barreled Cl-sensitive microelectrodes. Addition of Cl to a Cl-free medium bathingin vitro intestinal segments produced a saturable (K m =5.4mm) increase in shortcircuit current (I sc) which was inhibitable by 1mm SITS. The selectivity sequence for the anion-evoked current was Cl=Br>SCN>NO 3 >F=I. Current evoked by Cl reached a maximum with increasing medium Na concentration (K m =12.4mm). Addition of Na+, as Na gluconate (10mm), to mucosal and serosal Na+-free media stimulated the Cl current and simultaneously increased the absorptive Cl flux (J ms Cl ) and net flux (J net Cl ) without changing the secretory Cl flux (J sm Cl ). Addition of Na+ only to the serosal fluid stimulatedJ ms Cl much more than Na+ addition only to the mucosal fluid in paired tissues. Serosal DIDS (1mm) blocked the stimulation. Serosal 10mm Tris gluconate or choline gluconate failed to stimulateJ ms Cl . Intracellular Cl activity (a Cl i ) in villus epithelial cells was above electrochemical equilibrium indicating active Cl uptake. Ouabain (1mm) eliminated Cl accumulation and reduced the mucosal membrane potential m over 2 to 3 hr. In contrast, SITS had no effect on Cl accumulation and hyperpolarized the mucosal membrane. Replacement of serosal Na+ with choline eliminated Cl accumulation while replacement of mucosal Na+ had no effect. In conclusion by two independent methods active electrogenic Cl absorption depends on serosal rather than mucosal Na+. It is concluded that Cl enters the cell via a primary (rheogenic) transport mechanism. At the serosal membrane the Na+ gradient most likely energizes H+ export and regulates mucosal Cl accumulation perhaps by influencing cell pH or HCO 3 concentration.  相似文献

Summary Intracellular potassium activity (a K i ) was measured in control conditions in mid-cortical rabbit proximal convoluted tubule using two methods: (i) by determination of the K+ equilibrium potential (E K) using Ba2+-induced variations in the basolateral membrane potential (V BL) during transepithelial current injections and (ii) with double-barrel K-selective microelectrodes. Using the first method, the meanV BL was –48.5±3.2 mV (n=16) and the meanE K was –78.4±4.1 mV corresponding to aa K i of 68.7mm. With K-selective microelectrodes,V BL was –36.6±1.1 mV (n=19),E K was –64.0±1.1 mV anda K i averaged 40.6±1.7mm. While these lastE K andV BL values are significantly lower than the corresponding values obtained with the first method (P<0.001 andP<0.01, respectively), the electrochemical driving force for K transport across the basolateral membrane ( K =V BLE K) is not significantly different for both techniques (30.1±3.3 mV for the first technique and 27.6±1.8 mV for ion-selective electrodes). This suggests an adequate functioning of the selective barrel but an underestimation ofV BL by the reference barrel of the double-barrel microelectrode. Such double-barrel microelectrodes were used to measure temporal changes ina K i and K in different experimental conditions where Na reabsorption rate (J Na) was reduced.a K i was shown to increase by 12.2±2.7 (n=5) and 14.1±4.4mm (n=5), respectively, whenJ Na was reduced by omitting in the luminal perfusate: (i) 5.5mm glucose and 6mm alanine and (ii) glucose, alanine, other Na-cotransported solutes and 110mm Na. In terms of the electrochemical driving force for K exit across the basolateral membrane, K, a decrease of 5.4±2.0 mV (P<0.05,n=5) was measured when glucose and alanine were omitted in the luminal perfusate while K remained unchanged whenJ Na was more severely reduced (mean change =–1.7±2.1 mV, NS,n=5). In the latter case, this means that the electrochemical driving force for K efflux across the basolateral membrane has not changed while both the active influx through the Na–K pump and the passive efflux in steady state are certainly reduced. If the main pathway for K transport is through the basolateral K conductance, this implies that this conductance must have decreased in the same proportion as that of the reduction in the Na–K pump activity.  相似文献

Summary The reaction of abdominal skins of the frog speciesRana temporaria on mucosal K+-containing solutions was studied in an Ussing-type chamber by recording transepithelial potential difference (PD), short-circuit current (SCC) and conductance (G). With Na-Ringer's as serosal medium, a linear correlation between PD and the logarithm of the mucosal K+-concentration ([K] o ) was obtained. The K+-dependent SCC saturated with increasing [K] o , and could quickly and reversibly be depressed by addition of Rb+, Cs+, and H+, Li+, Na+, and NH 4 + did not influence K+ current. A large scatter was obtained for kinetic parameters like the slope of the PD-log [K] o -line (18–36.5 mV/decade), the apparent Michaelis constant (13–200mm), and the maximal current of the saturable SCC (6–50 A·cm–2), as well as for the degree of inhibition by Cs+ ions. This seemed to be caused by a time-dependent change during long time exposure to high [K] o (more than 30 sec), thereby inducing a selectivity loss of K+-transporting structures, together with an increase in SCC andG and a decrease in PD. Short time exposure to K+-containing solutions showed a competitive inhibition of K+ current by Cs+ ions, and a Michaelis constant of 6.6mm for the inhibitory action of Cs+. Proton titration resulted in a decrease of K+ current at pH<3. An acidic membrane component (apparent dissociation constant 2.5×10–3 m) is virtually controlling K+ transfer. Reducing the transepithelial K+-concentration gradient by raising the serosal potassium concentration was accompanied by the disappearance of SCC and PD.  相似文献

Summary It has been suggested previously that La3+ can replace Na+ on various cotransport systems in renal brush border membranes. In the present study, we used rabbit renal brush border membrane vesicles to examine the specificity and kinetics of Ln3+/proline cotransport. Experiments were carried out under zero-trans, voltage clamped conditions using a rapid-mix/filtration technique. Initial experiments confirmed that La3+ produced the classical overshoot phenomenon. The initial rates of proline uptake relative to Na+ were Eu3+, Tb3+, Nd3+, Pr3+, Ho3+ (3.3)>Na+ (1.0)>La3+ (0.86) > choline+ (0.1). At a saturating salt concentration, uptake saturated with increasing proline concentration: theK t andJ max were 0.05mm and 17 pmol mg–1 sec–1 in Na+; and 0.28mm and 73 pmol mg–1 sec–1 in Tb3+. The higherJ max in Tb3+ indicates that the Tb3+-proline loaded carrier is more effective than the Na+-proline loaded carrier in overcoming some rate-limiting barriers in the transport process. Na+ activated proline uptake with a Hill coefficient of 1.6 and aK 0.5 of 21mm, while Tb3+ activated with a Hill coefficient of 0.88 and aK 0.5 of 28mm. The Hill coefficient for Na+ suggests two binding sites, whereas the Hill coefficient for Tb3+ may indicate negative cooperativity between the trivalent ligands at the binding sites. We conclude that lanthanides are able to substitute for Na+ on the brush border proline carrier and that the lanthanides may serve as useful probes for the ligand binding sites.  相似文献

Summary We have examined the effect of internal and external pH on Na+ transport across toad bladder membrane vesicles. Vesicles prepared and assayed with a recently modified procedure (Garty & Asher, 1985) exhibit large, rheogenic, amiloridesensitive fluxes. Of the total22Na uptake measured 0.5–2.0 min after introducing tracer, 80±4% (mean±se,n=9) is blocked by the diuretic with aK 1 of 2×10–8 m. Thus, this amiloridesensitive flux is mediated by the apical sodium-selective channels. Varying the internal (cytosolic) pH over the physiologic range 7.0–8.0 had no effect on sodium transport; this result suggests that variation of intracellular pHin vivo has no direct apical effect on modulating sodium uptake. On the other hand,22Na was directly and monotonically dependent on external pH. External acidification also reduced the amiloride-sensitive efflux across the walls of the vesicles. This inhibition of22Na efflux was noted at external Na+ concentrations of both 0.2 m and 53mm.These results are different from those reported with whole toad bladder. A number of possible bases for these differences are considered and discussed. We suggest that the natriferic response induced by mucosal acidification of whole toad urinary bladder appears to operate indirectly through one or more factors, presumably cytosolic, present in whole cells and absent from the vesicles.  相似文献

Summary The contribution of specific ions to the conductance and potential of the basolateral membrane of the rabbit urinary bladder has been studied with both conventional and ion-specific microelectrode techniques. In addition, the possibility of an electrogenic active transport process located at the basolateral membrane was studied using the polyene antibiotic nystatin. The effect of ion-specific microelectrode impalement damage on intracellular ion activities was examined and a criterion set for acceptance or rejection of intracellular activity measurements. Using this criterion, we found (K+)=72mm and (Cl)=15.8mm. Cl but not K+ was in electrochemical equilibrium across the basolateral membrane. The selective permeability of the basolateral membrane was measured using microelectrodes, and the data analyzed using the Goldman, Hodgkin-Katz equation. The sodium to potassium permeability ratio (P Na/P K) was 0.044, and the chloride to potassium permeability ratio (P Cl/P K) was 1.17. Since K+ was not in electrochemical equilibrium, intracellular (K+) is maintained by active metabolic processes, and the basolateral membrane potential is a diffusion potential with K+ and Cl the most permeable ions. After depolarizing the basolateral membrane with high serosal potassium bathing solutions and eliminating the apical membrane as a rate limiting step for ion movement using the polyene antibiotic nystatin, we found that the addition of equal aliquots of NaCl to both solutions caused the basolateral membrane potential to hyperpolarize by up to 20 mV (cell interior negative). This popential was reduced by 80% within 3 min of the addition of ouabain to the serosal solution. This hyperpolarization most probably represents a ouabain sensitive active transport process sensitive to intracellular Na+. An equivalent electrical circuit for Na+ transport across rabbit urinary bladder is derived, tested, and compared to previous results. This circuit is also used to predict the effects that microelectrode impalement damage will have on individual membrane potentials as well as time-dependent phenomena; e.g., effect of amiloride on apical and basolateral membrane potentials.  相似文献

Summary Patch-clamp techniques have been applied to characterize the channels in the basolateral membrane of resting (cimetidine-treated, nonacid secreting) oxyntic cells isolated from the gastric mucosa ofNecturus maculosa. In cell-attached patches with pipette solution containing 100mm KCl, four major classes of K+ channels can be distinguished on the basis of their kinetic behavior and conductance: (1) 40% of the patches contained either voltage-independent (a) or hyperpolarization-activated (b), inward-rectifying channels with short mean open times (16 msec fora, and 8 msec forb). Some channels showed subconductance levels. The maximal inward conductanceg max was 31±5 pS (n=13) and the reversal potentialE rev was atV p=–34±6 mV (n=9). (2) 10% of the patches contained depolarization-activated and inward-rectifying channels withg max=40 ±18 pS (n=3) andE rev was atV p=–31±5 mV (n=3). With hyperpolarization, the channels open in bursts with rapid flickerings within bursts. Addition of carbachol (1mm) to the bath solution in cell-attached patches increased the open probabilityP o of these channels. (3) 10% of the patches contained voltage-independent inward-rectifying channels withg max=21±3 pS (n=4) andE rev was atV p=–24±9 mV (n=4). These channels exhibited very high open probability (P o=0.9) and long mean open time (1.6 sec) at the resting potential. (4) 20% of the patches contained voltage-independent channels with limiting inward conductance of 26±2 pS (n=3) andE rev atV p=–33±3 mV (n=3). The channels opened in bursts consisting of sequential activation of multiple channels with very brief mean open times (10 msec). In addition, channels with conductances less than 6 pS were observed in 20% of the patches. In all nine experiments with K+ in the pipette solution replaced by Na+, unitary currents were outward, and inward currents were observed only for large hyperpolarizing potentials. This indicates that the channels are more selective for K+ over Na+ and Cl. A variety of K+ channels contributes to the basolateral K+ conductance of resting oxyntic cells.  相似文献

Summary We have measured the intracellular potassium activity, [K+]i and the mechanisms of transcellular K+ transport in reabsorptive sweat duct (RSD) using intracellular ion-sensitive microelectrodes (ISMEs). The mean value of [K+]i in RSD is 79.8±4.1mm (n=39). Under conditions of microperfusion, the [K+]i is above equilibrium across both the basolateral membrane, BLM (5.5 times) and the apical membrane, APM (7.8 times). The Na+/K+ pump inhibitor ouabain reduced [K+]i towards passive distribution across the BLM. However, the [K+]i is insensitive to the Na+/K+/2 Cl cotransport inhibitor bumetanide in the bath. Cl substitution in the lumen had no effect on [K+]i. In contrast, Cl substitution in the bath (basolateral side) depolarized BLM from –26.0±2.6 mV to –4.7*±2.4 mV (n=3;* indicates significant difference) and decreased [K+]i from 76.0±15.2mm to 57.7* ±12.7mm (n=3). Removal of K+ in the bath decreased [K+]i from 76.3±15.0mm to 32.3*±7.6mm (n=4) while depolarizing the BLM from –32.5±4.1 mV to –28.3*±3.0 mV (n=4). Raising the [K+] in the bath by 10-fold increased [K+]i from 81.7±9.0mm to 95.0*±13.5mm and depolarized the BLM from –25.7±2.4 mV to –21.3*±2.9 mV (n=4). The K+ conductance inhibitor, Ba2+, in the bath also increased [K+]i from 85.8±6.7mm to 107.0*±11.5mm (n=4) and depolarized BLM from –25.8±2.2 mV to –17.0*±3.1 mV (n=4). Amiloride at 10–6 m increased [K+]i from 77.5±18.8mm to 98.8*±21.6mm (n=4) and hyperpolarized both the BLM (from –35.5±2.6 mV to –47.8*±4.3 mV) and the APM (from –27.5±1.4 mV to –46.0* ±3.5 mV,n=4). However, amiloride at 10–4 m decreased [K+]i from 64.5±0.9mm to 36.0*±9.9mm and hyperpolarized both the BLM (from –24.7±1.4 mV to –43.5*±4.2 mV) and APM (from –18.3±0.9 mV to –43.5*±4.2 mV,n=6). In contrast to the observations at the BLM, substitution of K+ or application of Ba2+ in the lumen had no effect on the [K+]i or the electrical properties of RSD, indicating the absence of a K+ conductance in the APM. Our results indicate that (i) [K+]i is above equilibrium due to the Na+/K+ pump; (ii) only the BLM has a K+ conductance; (iii) [K+]i is subject to modulation by transport status; (iv) K+ is probably not involved in carrier-mediated ion transport across the cell membranes; and (v) the RSD does not secrete K+ into the lumen.  相似文献

Summary In order to permit future characterization and possible isolation of the Na+–H+ exchanger from the apical membrane of proximal tubular cells, studies were performed to solubilize and reconstitute this transporter. Rabbit brush border membranes were prepared by a magnesium aggregation method, solubilized with the detergent octyl glucoside, and reconstituted into artificial phospholipid vesicles. In the presence of a pH gradient (pHin 6.0, pHout 8.0), the uptake of 1mm 22Na+ into the proteoliposomes was five- to sevenfold higher than into liposomes. Amiloride (2mm) inhibited proton gradient-stimulated uptake of sodium by 50%. As compared to proton gradient conditions, the uptake of sodium was lower in the absence of a pH gradient but was significantly higher when the outside and inside pH was 6.0 than 8.0. TheK a for sodium in reconstituted proteoliposomes studied under pH gradient conditions was 4mm. The uptake of sodium in proteoliposomes prepared from heat-denatured membrane proteins was significantly decreased. These studies demonstrate that proteoliposomes prepared from octyl glucoside-solubilized brush border membrane proteins and asolectin exhibit proton gradient-stimulated, amiloride-inhibitable, electroneutral uptake of sodium. The ability to solubilize and reconstitute the Na+–H+ exchanger from the apical membrane of the proximal tubule will be of value in isolating and characterizing this transporter.  相似文献

Summary Sodium (22Na) transport was studied in a basolateral membrane vesicle preparation from rabbit parotid. Sodium uptake was markedly dependent on the presence of both K+ and Cl in the extravesicular medium, being reduced 5 times when K+ was replaced by a nonphysiologic cation and 10 times when Cl was replaced by a nonphysiologic anion. Sodium uptake was stimulated by gradients of either K+ or Cl (relative to nongradient conditions) and could be driven against a sodium concentration gradient by a KCl gradient. No effect of membrane potentials on KCl-dependent sodium flux could be detected, indicating that this is an electroneutral process. A KCl-dependent component of sodium flux could also be demonstrated under equuilibrium exchange conditions, indicating a direct effect of K+ and Cl on the sodium transport pathway. KCl-dependent sodium uptake exhibited a hyperbolic dependence on sodium concentration consistent with the existence of a single-transport system withK m =3.2mm at 80mm KCl and 23°C. Furosemide inhibited this transporter withK 0.5=2×10–4 m (23°C). When sodium uptake was measured as a function of potassium and chloride concentrations a hyperbolic dependence on [K] (Hill coefficient =1.31±0.07) were observed, consistent with a Na/K/Cl stoichiometry of 112. Taken together these data provide strong evidence for the electroneutral coupling of sodium and KCl movements in this preparation and strongly support the hypothesis that a Na+/K+/Cl cotransport system thought to be associated with transepithelial chloride and water movements in many exocrine glands is present in the parotid acinar basolateral membrane.  相似文献

Summary Cellular impalements were used in combination with standard transepithelial electrical measurements to evaluate some of the determinants of the spontaneous lumen-positive voltage,V e , which attends net Cl absorption,J Cl net , and to assess how ADH might augment bothJ Cl met andV e in the mouse medullary thick ascending limb of Henle microperfusedin vitro. Substituting luminal 5mm Ba++ for 5mm K+ resulted in a tenfold increase in the apical-to-basal membrane resistance ratio,R c /R bl , and increasing luminal K+ from 5 to 50mm in the presence of luminal 10–4 m furosemide resulted in a 53-mV depolarization of apical membrane voltage,V a . Thus K+ accounted for at least 85% of apical membrane conductance. Either with or without ADH. 10–4 m luminal furosemide reducedV e andJ Cl net to near zero values and hyperpolarized bothV a andV bl , the voltage across basolateral membranes; however, the depolarization ofV bl was greater in the presence than in the absence of hormone while the hormone had no significant effect on the depolarization ofV a , Thus ADH-dependent increases inV b were referable to greater depolarizations ofV bl in the presence of ADH than in the absence of ADH 68% of the furosemide-induced hyperpolarization ofV a was referable to a decrease in the K+ current across apical membranes, but, at a minimum, only 19% of the hyperpolarization ofV bl could be accounted for by a furosemide-induced reduction in basolateral membrane Cl current. Thus an increase in intracellular Cl activity may have contributed to the depolarization ofV bl during net Cl absorption, and the intracellular Cl activity was likely greater with ADH than without hormone. Since ADH increases apical K+ conductance and since the chemical driving force for electroneutral Na+,K+,2Cl cotransport from lumen to cell may have been less in the presence of ADH than in the absence of hormone, the cardinal effects of ADH may have been to increase the functional number of both Ba++-sensitive conductance K+ channels and electroneutral Na+,K+,2Cl cotransport units in apical plasma membranes.  相似文献

Summary Kinetic properties of Na+–Ca2+ exchange in a renal epithelial cell line (LLC-MK2) were assessed by measuring cytosolic free Ca2+ with fura-2 and45Ca2+ influx. Replacing external Na+ with K+ produced relatively small increases in free Ca2+ and45Ca2+ uptake unless the cells were incubated with ouabain. Ouabain markedly increased cell Na+ and strongly potentiated the effect of replacing external Na+ with K+ on free Ca2+ and45Ca2+ uptake.45Ca2+ influx in 140mm K+ or N-methyl-d-glucamine minus influx in 140mm Na+ was used to quantify Na+–Ca2+ exchange activity of Na+-loaded cells. The dependence of exchange on cell Na+ was sigmoidal; theK 0.5 was 26±3 mmol/liter cell water space, and the Hill coefficient was 3.1±0.2. The kinetic features of the dependence of exchange on cell Na+ partly account for the small increase in Ca2+ influx when all external Na+ is replaced by K+. Besides raising cell Na+ ouabain appears to activate the exchanger. Magnesium competitively inhibited exchange activity. The potency of Mg2+ was 8.2-fold lower with potassium instead of N-methyl-d-glucamine or choline as the replacement for external Na+. Potassium also increased theV max of exchange by 86% and had no effect on theK m for Ca2+. The exchanger does not cause detectable22Na+–Mg2+ exchange and does not appear to require K+ or transport86Rb+. Although exchange activity was plentiful in the epithelial cells from monkey kidney, others from amphibian, canine, opossum, and porcine kidney had no detectable exchange activity. All of the measured kinetic properties of Na+–Ca2+ exchange in the renal epithelial cells are very similar to those of the exchanger in rat aortic myocytes.  相似文献

Summary Regulation of urea transport by vasopressin in inner medullary collecting duct (IMCD) cells is thought to be important for the urinary concentrating mechanism. Isolated tubule perfusion studies suggest the existence of a saturable urea carrier. We have measured14C-urea efflux in IMCD cells which were freshly isolated and grown in primary culture. Cells were isolated from rat papilla by collagenase digestion and hypotonic shock. In suspended cells,14C-urea efflux (J urea from loaded cells was exponential with time constant 59±3 sec (sem,n=6, 23°C).J urea had an activation energy of 4.1 kcal/mole and was inhibited 42±7% by 0.25mm phloretin and 30–40% by the high affinity urea analogues dimethylurea and phenylurea.J urea was increased 40–60% by addition of vasopressin (10–8 m) or 8-bromo-cAMP (1mm); stimulatedJ urea was inhibited 55±8% by the kinase A inhibitor H-8. Phorbol esters and epidermal growth factor did not alterJ urea. IMCD cells grown in primary culture were homogeneous in appearance with>fivefold stimulation of cAMP by vasopressin. The exponential time constant for urea efflux was 610±20 sec (n=3).J urea was not altered by vasopressin, cAMP or phloretin. Another function of in vivo IMCD cells, vasopressin-dependent formation of endosomes containing water channels, was absent in the cultured cells. These results demonstrate presence of a urea transporter on suspended IMCD cells which is activated by cAMP and inhibited by phloretin and urea analogues. The urea transporter and its regulation by cAMP, and cAMP-dependent apical membrane endocytosis, are lost after growth in primary culture.  相似文献

Summary In intact ileal mucosa, uptake of SO4 across the brush border membrane requires the presence of Na and is saturable, withK1/2=1.3mm at 140mm Na (P.L. Smith, S.A. Orellana & M. Field, 1981.J. Membrane Biol. 63:199–206). The present study examines the substrate specificities and transport stoichiometry of the Na-dependent SO4 uptake process. The effects of variations in medium anion and cation composition on lumen-to-epithelium influx of SO4 (J me SO4 ) were determined under short-circuit conditions.J me SO4 is inhibited by thiosulfate, but not by phosphate, methylsulfate, vanadate or taurocholate. Cl is weakly inhibitory. Uptake of SO4 is poorly supported by Li, and is unaffected by K, indicating a specific dependence on Na. At low SO4 concentration (0.22mm),J me SO4 is a hyperbolic function of medium Na concentration; the corresponding Hill plot is linear with a slope of 1.0, suggesting a transport stoichiometry of 1 Na: 1 SO4. At high SO4 concentration (6.7mm), the Na-dependent SO4 velocity curve is sigmoidal and yields a Hill plot which is again linear but has a slope of 1.56, suggesting transport of more than 1 Na per SO4. SO4 uptake in presence of Na exhibits a dependence on medium pH. At 0.22mm SO4 and 140mm Na,J me SO4 was doubled by lowering pH from 7.4 to 6.8. However, at 6.7mm SO4 and 140mm Na, changing pH had no effect onJ me SO4 over the range 6.8 to 8.5. The pH dependence ofJ me SO4 at 6.7mm SO4 was restored when medium Na was lowered to 3mm, suggesting that pH sensitivity is a function of the concentration of preformed NaSO 4 ion pair. The results suggest that SO4 influx across the ileal brush border occurs by electroneutral Na+/NaSO 4 or Na+/H+/SO 4 2– cotransport, the former being favored by high concentrations of Na and SO4.  相似文献

Summary Na, K-ATPase function was studied in order to evaluate the mechanism of increased colonic Na+ transport during early postnatal development. The maximum Na+-pumping activity that was represented by the equivalent short-circuit current after addition of nystatin (I sc N ) did not change during postnatal life or after adrenalectomy performed in 16-day-old rats.I sc N was entirely inhibited by ouabain; the inhibitory constant was 0.1mm in 10-day-old (young) and 0.4mm in 90-day-old (adult) rats. The affinity of the Na, K pump for Na+ was higher in young (11mm) than in adult animals (19mm). The Na, K-ATPase activity (measured after unmasking of latent activity by treatment with sodium dodecylsulfate) increased during development and was also not influenced by adrenalectomy of 16-day-old rats. The inhibitory constant for ouabain (K I ) was not changed during development (0.1–0.3mm). Specific [3H]ouabain binding to isolated colonocytes increased during development (19 and 82 pmol/mg protein), the dissociation constant (K D ) was 8 and 21 m in young and adult rats, respectively. The Na+ turnover rate per single Na, K pump, which was calculated fromI sc N and estimated density of binding sites per cm2 of tissue was 500 in adult and 6400 Na+/min·site in young rats. These data indicate that the very high Na+ transport during early postnatal life reflects an elevated turnover rate and increased affinity for Na+ of a single isoform of the Na, K pump. The development of Na+ extrusion across the basolateral membrane is not directly regulated by corticosteroids.  相似文献

Summary The initial mechanisms of injury to the proximal tubule following exposure to nephrotoxic heavy metals are not well established. We studied the immediate effects of silver (Ag+) on K+ transport and respiration with extracellular K+ and O2 electrodes in suspensions of renal cortical tubules. Addition of silver nitrate (AgNO3) to tubules suspended in bicarbonate Ringer's solution caused a rapid, dose-dependent net K+ efflux (K m =10–4 m,V max=379 nmol K+/min/mg protein) which was not inhibited by furosemide, barium chloride, quinine, tetraethylammonium, or tolbutamide. An increase in the ouabain-sensitive oxygen consumption rate (QO2) (13.9±1.1 to 25.7±4.4 nmol O2/min/mg,P<0.001), was observed 19 sec after the K+ efflux induced by AgNO3 (10–4 m), suggesting a delayed increase in Na+ entry into the cell. Ouabain-insensitive QO2, nystatin-stimulated QO2, and CCCP-uncoupled QO2 were not significantly affected, indicating preserved function of the Na+, K+-ATPase and mitochondria. External addition of the thiol reagents dithiothreitol (1mm) and reduced glutathione (1mm) prevented and/or immediately reversed the effects on K+ transport and QO2. We conclude that Ag+ causes early changes in the permeability of the cell membrane to K+ and then to Na+ at concentrations that do not limit Na+, K+-ATPase activity or mitochondrial function. These alterations are likely the result of a reversible interaction of Ag+ with sulfhydryl groups of cell membrane proteins and may represent initial cytotoxic effects common to other sulfhydryl-reactive heavy metals on the proximal tubule.  相似文献

Summary Human red cells were prepared with various cellular Na+ and K+ concentrations at a constant sum of 156mm. At maximal activation of the K+ conductance,g K(Ca), the net efflux of K+ was determined as a function of the cellular Na+ and K+ concentrations and the membrane potential,V m , at a fixed [K+]ex of 3.5mm.V m was only varied from (V m E K)25 mV and upwards, that is, outside the range of potentials with a steep inward rectifying voltage dependence (Stampe & Vestergaard-Bogind, 1988).g K(Ca) as a function of cellular Na+ and K+ concentrations atV m =–40, 0 and 40 mV indicated a competitive, voltage-dependent block of the outward current conductance by cellular Na+. Since the present Ca2+-activated K+ channels have been shown to be of the multi-ion type, the experimental data from each set of Na+ and K+ concentrations were fitted separately to a Boltzmann-type equation, assuming that the outward current conductance in the absence of cellular Na+ is independent of voltage. The equivalent valence determined in this way was a function of the cellular Na+ concentration increasing from 0.5 to 1.5 as this concentration increased from 11 to 101mm. Data from a previous study of voltage dependence as a function of the degree of Ca2+ activation of the channel could be accounted for in this way as well. It is therefore suggested that the voltage dependence ofg K(Ca) for outward currents at (V m E K)>25 25 mV reflects a voltage-dependent Na+ block of the Ca2+-activated K+ channels.  相似文献
