Kinetic conductivity measurement, run as a microcomputer-controlled workstation with eight independent reaction cells, is shown here to be a simple, cost-effective method for assaying enzymes. The technique is demonstrated with enzymes that can be used as markers for the major subcellular compartments of hepatocytes as a one-day student laboratory class with great potential for variation and development. The method is virtually unknown in enzymology, but could be envisaged as standard apparatus for all biochemical laboratories and teaching classes.  相似文献

Spruce wood shavings from Picea abies were used for an adsorptive removal of both basic as well as acid dyes from waters. The sorption properties of the sorbents were modified with HCl, Na(2)CO(3) and Na(2)HPO(4). The treatment of the wood sorbents with alkaline carbonate solution as well as with phosphate solution increased the sorption ability for the basic dye (Methylene Blue), whereas the treatment with mineral acid decreased the sorption ability for Methylene Blue to some extent. The opposite is true for the sorption of the acid dye--Egacid Orange. The maximum sorption capacities estimated from the Langmuir-Freundlich isotherms ranged from 0.060 to 0.165 mmol g(-1) for Methylene Blue, and from 0.045 to 0.513 mmol g(-1) for Egacid Orange. The basic dye sorption decreased at low pH values in accordance with a presupposed ion-exchange mechanism of the sorption. The sorption of acid dye, on the other hand, decreased with increasing pH. The presence of inorganic salts as well as surfactants exhibited only minor effects on the dye sorption.  相似文献

The gluconic fragment of strophantin K oxidation by sodium metaperiodate yields a dialdehyde derivate conjugated with catalase. The conjugate obtained contains 11 molecules of cardiac glucoside. Adsorption and circular dichroism spectra of the native enzyme and its conjugate were compared and structural differences between both samples were revealed. The kinetics of ethanol oxidation into acetaldehyde by cumene hydroperoxide was studied at 30 degrees C in the phosphate buffer pH 6.6; this reaction was shown to proceed with the participation of catalase and its cat-str conjugate. The catalytic constants for catalase are 1.2-1.5 times as high as those for cat-str, whereas the Km values for both substrates for the conjugate as 1.5-2 times as high as those for catalase. Catalase modification by strophantin K increases the enzyme thermostability up to the isokinetic point of 40 degrees C; above this threshold the cat-str thermostability decreases as compared with the native enzyme. The thermodynamical activation parameters for catalase and cat-str inactivation were determined.  相似文献

Abstract— The activity profiles of the solubilized protein kinases from the microsomal and myelin fractions of bovine brain were examined by column chromatography and sucrose density gradient centrifugation. The main peak of adenosine 3',5'-monophosphate (cyclic AMP)-dependent activity with histone as substrate for each membrane enzyme was eluted with about 0.2 m -NaCl on a DEAE-cellulose column. A peak of activity stimulated with cyclic AMP was also eluted with about 0.1 m -NaCl for the microsomal enzyme. A peak with protamine and casein as substrate for the microsomal or myelin enzyme, respectively, was larger than that with histone as substrate for each enzyme. The first peak with histone as substrate on a DEAE–cellulose column appeared as two peaks on the Sepharose 6B column. The second peak with histone as substrate on DEAE–cellulose column was shown to be a holoenzyme consisting of regulatory and catalytic subunits. The holoenzyme and subunits were eluted at similar positions to each other between both membrane enzymes on Sepharose 6B column. The holoenzyme sedimented as two peaks of activity on sucrose density gradient centrifugation, both of which were stimulated with cyclic AMP. The preincubation of the holoenzyme with cyclic AMP resulted in shifting to a position of a smaller molecular size.
The results indicate the occurrence of multiple forms of protein kinases in membrane fractions of brain with respect to substrate specificity and physical property.  相似文献

The large, comprehensive vegetation database of Mecklenburg-Vorpommern/NE Germany with 51,328 relevés allowed us to study an entire regional flora of 133 non-native plants (NNP, immigration after 1492 AD) with regard to their preferences to all kinds of habitats and along different ecological gradients. For each relevé, we computed average Ellenberg indicator values (EIV) for temperature, light, moisture, reaction, nutrients and salt as well as plant strategy type weights. We partitioned the dataset into relevés with and without occurrences of NNP and compared them with respect to the relative frequencies of EIVs and strategy type weights. We identified deviations from random differences by testing against permuted indicator values. To account for bias in EIV between community types, NNP preferences were differentiated for 34 phytosociological classes. We tested significance of preferences for the group of NNP as a whole, as well as for single NNP species within the entire dataset, as well as differentiated by phytosociological classes and formations. NNP as a group prefer communities with high EIVs for temperature and nutrients and low EIVs for moisture. They avoid communities with low EIV for reaction and high EIV for salt. NNP prefer communities with high proportions of ruderal and low proportion of stress strategists. The differentiation by phytosociological classes reinforces the general trends for temperature, nutrients, moisture, R and S strategy types. Nevertheless, preferences of single species reveal that NNP are not a congruent group but show individualistic ecological preferences.  相似文献

The potential of liquid manure as sole nutrient source for cultivation of Ulva lactuca was investigated with the perspective of utilizing the produced biomass for feed and/or energy. Algae grown with manure demonstrated equal growth rates to algae grown with standard f/2-medium. The optimum manure concentration, expressed as ammonium concentration, was 25?μM. At these conditions, the biomass produced was potentially suitable for anaerobic digestion, due to a relative high carbon/nitrogen ratio (approximately 19). At higher manure concentrations, tissue concentrations of nitrogen, phosphorus, proteins, and amino acids increased, making the biomass less suitable for anaerobic digestion but potentially interesting as a feed supplement. Cultivating U. lactuca with manure as nutrient source has potential in terms of bioremediation as well as production of bioenergy and protein-feed.  相似文献

Mineral fillers have long been used in papermaking. The furnish of the writing and printing paper depends on using the fillers as internal or surface treatments. Talc, as an Egyptian filler material, was used as it is or after modification to be used as internal sizing for bleached rice straw pulp. Talc was modified chemically with phthalic anhydride and urea, as well as with using rosin size. Talc is chemically inert. The modification was carried out to change the nature of the native talc. The mechanical and optical properties for internal sizing papers were studied. The results indicated that modified talc with phthalic anhydride enhances the mechanical and optical properties for sized hand-sheets. It was clear that modification of the talc plays a role in improvement of the fiber–filler–fiber bond.  相似文献

1. Baboon ferrochelatase was purified to apparent homogeneity. 2. The pH optimum was 7.85 and the pI 5.3. 3. The estimated molecular weight was 205 K made up by two 50 + 60 K heterodimers. 4. The Km values for proto- and mesoporphyrin were 18.5 and 10.8 microM with iron as co-substrate. With cobalt as co-substrate the Km values were 34.5 and 10.4 microM, respectively. The mean Km value for iron was 2.2 microM while cobalt acted as a complete inhibitor. 5. Lead played a dual role that of both pseudo substrate and inhibitor. As shown by inhibitor kinetics, Pb acted as a two-step two-site parabolic competitive inhibitor. The mean Ki value at low Pb levels was 0.65 mM and at high levels 0.17 mM. 6. Substrate inhibition occurred above 36 microM for proto- and 44 microM for mesoporphyrin with iron as co-substrate. For iron, with mesoporphyrin as co-substrate it occurred above 29 microM.  相似文献

Nystatin (NYS), a polyene antifungal antibiotic, has been investigated in Langmuir monolayers alone and in mixtures with mammalian and fungi membrane sterols (cholesterol and ergosterol, respectively) as well as with a model phospholipid (DPPC). The interactions between film molecules have been examined both in a qualitative and quantitative way with the excess area per molecule (AExc), excess free energy of mixing (DeltaGExc) and the interaction parameter (alpha). The obtained results have been compared with those previously reported for another polyene antimycotic: amphotericin B (AmB) mixed with lipids. Higher affinity of NYS has been observed for ergosterol vs. cholesterol, however, the strongest attractions were found for its mixtures with DPPC. The obtained results have been verified with biological studies reported previously for both antibiotics (NYS and AmB). A thorough analysis of the Langmuir experiment results performed for both polyenes enabled us to conclude that the presence of DPPC can be considered as a key factor affecting their antifungal activity as well as their toxicity towards host cells.  相似文献

A new digital computer program has been developed for the seriation of anthropological data. The program facilitates and refines procedures in the application of certain matrix-ordering approaches, such as the Brainerd-Robinson technique. The new program has special advantages for obtaining the best ordering of a case that is inherent in the data, together with special provisions for experimentation. Some of the innovations consist of working with the whole matrix pattern from start to finish, more exhaustive row-column comparisons, provision for specifying different ordering criteria, consistent utilization of a chosen criterion for matrix evaluation as well as for row-column placement, and a means of lessening the effect of input-order bias. The program is designed to produce a close approximation of an ideal model in order to place data collections as accurately as possible on a continuum of similarity for purposes such as chronological ordering and the expression of intercultural relationships.  相似文献

The purpose of this study was to determine taste difference thresholds for monosodium glutamate (MSG) and sodium chloride (NaCl) in pigtail macaques (Macaca nemestrina) and spider monkeys (Ateles geoffroyi). Using a two-bottle preference test of brief duration, three animals of each species were presented with four different reference concentrations of 50, 100, 200, and 400 mM of a tastant and tested for their ability to discriminate these from lower concentrations of the same tastant. The just noticeable differences (JNDs), expressed as Weber ratios (DeltaI/I), were found to range from 0.1 to 0.5 for MSG and 0.2 to 0.45 for NaCl in the pigtail macaques, with a significant tendency for higher Weber ratios with higher reference concentrations. In the spider monkeys, JNDs ranged from 0.15 to 0.4 for MSG and 0.1 to 0.25 for NaCl, with Weber ratios staying fairly constant across the reference concentrations tested. Thus, the JNDs were found to be generally similar in both species and to be at least as low as those found in humans for MSG and NaCl, as well as those found in spider monkeys for sucrose. The results support the assumption that both pigtail macaques and spider monkeys may use differences in perceived intensity of MSG and NaCl as a criterion for food selection.  相似文献

The pathogenicity of four strains, O-1, O-2, O-3, and O-4, of Toxoplasma isolated in the form of oocyst from the feces of naturally infected cats was examined for such laboratory animals as mice, rats, rabbits, guinea pigs, and dogs, in comparison with that of the Beverley strain. Suspensions of seven graded doses of oocysts of each strain ranging from 1.0 X 10(-1) to 1.0 X 10(5) were inoculated orally into seven groups of five mice each. The O-1, O-2, and O-3 strains were as pathogenic for mice as the Beverley strain, but the O-4 strain was not so pathogenic as any other strain. Rats, rabbits, guinea pigs, and dogs were inoculated orally with around 1.0 X 10(5) oocysts. The four O strains were not so severly pathogenic for rats and rabbits as to cause death. The O-2 and O-3 strains showed strong pathogenicity for guinea pigs, almost all of which, when inoculated with them, died after manifesting severe clinical symptoms. The pathogenicity of the O-1 and O-2 strains showed essentially the same tendency for dogs as for any other animal. In inoculation with oocysts, as well as with proliferative forms or cysts, the same pathogenicity was not observed in different strains, even if the same species of host animals was used. On the contrary, the same pathogenicity was not always found even in one strain when a different species of animals was used.  相似文献

Details of breeding orangutans and gorillas at the Jersey Wildlife Preservation Trust are reviewed, with special reference to the diversity of managerial requirements and reproductive parameters for the two species of great apes represented. The importance of recording and publishing as much data as possible to provide important guidelines for others involved with the same or related species in captivity, as well as such information contributing directly to conservation in the wild, is underlined. The Jersey Trust's involvement with cooperative and coordinated breeding programs for the two species on both a national and international basis is reported, and prospects for future long-term management strategies are outlined.  相似文献

Based on batch cultivation data for the production of L-glutamic acid from glucose, a comparative evaluation was made for repeated batch cultivations using one and more fermentors. The problem was formulated as maximizing the productivity of metabolic product with the specified conversion with respect to the cell age and the volume fraction used as seed for the subsequent repeated batch cultivation. Simulations were carried out with the assumption of no lag in product formation for the cases where the total operation time was specified as 200 h with reproducible batch cultivation cycles. The product production was assumed to be solely a function of product concentration. The computation results show the advantage of using more than one fermentor from the viewpoints of productivity and conversion, which will apply in general to non-growth-associated product production with delay time. In particular, three fermentors are recommended for the production of L-glutamic acid chosen as an example in this article.  相似文献

The motivation for and process leading up to the publication of a new bull breeding soundness certification standard endorsed by the South African Veterinary Association is described. The veterinary certificate of bull breeding soundness and explanatory notes and minimum standards are shown. The first component of the certificate is a declaration by the veterinarian that the bull complies with the minimum standards set for examinations for the selected purpose, these being for use as a natural service sire, as a donor of semen for distribution, and for insurance purposes. This is followed by the details of the bull and owner, and a list of the recommended examinations and tests for the bull with provision for which were performed. Certificates are available in book form with the explanatory notes and minimum standards on the reverse, and a carbon copy which remains in the book.The clarity and ease of completion of the document are regarded as being positive features. Bulls are either classified as breeding sound or not, with no actual parameters indicated on the document and no certificate issued for those which do not meet the set criteria. Contact details of the parties involved are shown on the certificate to allow for communication as a means of avoiding disputes.  相似文献

The rodent is a particularly valuable model with which to test therapeutic interventions for aging, as rodent physiology is close enough to human physiology to give the findings relevance for human aging, and it is small enough to allow for use of statistically robust sample sizes. There are many rodent models to choose from, with advantages and disadvantages to each. The choice of model system, as well as other experimental design decisions such as diet and housing, is extremely important for the success of lifespan studies. These issues are discussed in this review of the use of the rodent model. The National Institute on Aging (NIA) Interventions Testing Program, which has grappled with all of these issues, is described.  相似文献

The construction of a modified perfusion chamber is presented, which can be used for a prolonged cultivation of mammalian cells and tissues, for observation of the behavior of living cells as well as for the study of different effects on these cells. The chamber is made as non-demountable of optical glass, with a diffusive barrier separating the pericellular zone from that with a perfusion medium. The scheme of the equipment for cultivation of cells and tissues in this diffusion chamber on controlling the composition of nutrient medium and gas phase is given.  相似文献

The common marmoset, Callithrix jacchus, can be infected with human varicella-zoster virus (VZV), both wild-type strain KMcC and attenuated vaccine strain Oka/Merck. Infection was accomplished with either whole-cell-associated or cell extract VZV by combined oral-nasal-conjunctival application and was characterized by substantial and persistent anti-VZV antibody responses. The infectivity of VZV for marmosets was destroyed by treatment of inocula with heat or UV light. Diluted inocula with as few as 40 PFU/ml were infectious for marmosets. The lungs were demonstrated to be a major site of viral replication; both the presence of viral antigens and signs of pneumonia were demonstrated in lung tissues. Four serial passages of VZV KMcC were carried out in C. jacchus by a process of in vitro isolation and culturing of VZV from infected lung tissue and reapplication of the cultured isolates to fresh animals. The isolated viruses were identified as VZV both serologically and by restriction endonuclease analyses. The C. jacchus infectivity model should prove useful for determining the efficacy of subunit and live recombinant VZV vaccines as well as for the study of zoster.  相似文献

A sensitive and specific radioimmunoassay has been developed for YGG. The tripeptide was previously derivatised with p-benzoquinone to prepare the immunogen and the 125I tracer as well as in samples submitted to the RIA. The sensitivity is about 1 nM as compared with 8000 nM for underivatised YGG. Measurable amounts of endogenous YGG immunoreactivity, co-eluting in HPLC with authentic YGG, were detected in mouse striatal extracts.  相似文献
