The LIP2 isoenzyme gene from Candida rugosa has been completely synthesised and functionally expressed under the AOX1 promoter control in Pichia pastoris. The on-line monitoring and control of methanol, the key inducer carbon source in fed-batch cultures, has enhanced the yield product/biomass 7.8-fold and the productivity 12.8-fold compared to the best batch cultivation with the Pichia system and, 10-fold compared to the fed-batch cultivation process using the native C. rugosa strain.Nevertheless, the high ionic strength of culture broth favoured aggregation of Lip2, leading to total loss of lipolytic activity. After cultivation, a diaultrafiltration process was implemented to diminish ionic strength, allowing for the recovery of lipolytic activity in the diaultrafiltrate. The developed bioprocess resulted into a reproducible product in terms of quality and productivity.  相似文献

Seasonal variation in daily egg production rate of the planktonic calanoid copepod Acartia lilljeborgi Giesbrecht in relation to temperature, salinity and chlorophyll a concentration was studied in the Cananéia Lagoon estuarine system, from March 1995 to January 1996. Recently captured A. lilljeborgi adult females were individually incubated in bottles filled with surface water screened through a 40-m mesh, containing a natural assemblage of phytoplankton in the laboratory, at temperatures corresponding to ambient. Daily egg production rate ranged from 13.8±3.5 to 66.8± 15.1 eggs female–1 d–1 (mean ± 95% CL). The mean and maximum rates of daily egg production increased with temperature from 19.5 to 25.2 °C but then decreased with further increase in temperature at 28.4 through 29.1 °C, attaining the highest rates at approximately annual mean ambient water temperature (ca. 24–25 °C). The egg production rates increased linearly with chlorophyll a <40 m fraction. Hatching success varied from 68.6 to 91.9%. Cannibalism varied from 1.4±0.7 to 7.1±3.3 nauplii female–1 d–1 (mean ± 95% CL). These results suggest that water temperature and phytoplankton concentration are important factors affecting the egg production rate of A. lilljeborgi in the Cananéia Lagoon estuarine.  相似文献

To improve the productivity of Candida rugosa lipase (CRL) and alleviate respiration limitations during high cell-density fermentation, codon-optimized CRL LIP1, and Vitreoscilla hemoglobin (VHb) were co-expressed in Pichia pastoris. The activity of the recombinant strain that expressed LIP1 and VHb from dual promoters, named GS115/9Klip1FZvgb-lip1 #1, toward olive oil reached 620?U/mL, which was 1.69-fold greater than that of the recombinant strain GS115/9Klip1 #139 (365?U/mL) which only expressed LIP1 from a single promoter, and 1.37-fold greater than that of the recombinant strain GS115/9Klip1FZlip1 #39 (450?U/mL), which only expressed LIP1 from two promoters, in shaking flasks. With FM22 as the basic medium and methanol/D-sorbitol (1:1, v/v) as an inducer, the maximum activity of GS115/9Klip1FZvgb-lip1 #1 reached 7490?±?379.5?U/mL, which was 2.65-fold greater than that of GS115/9Klip1 #139 (2820?±?112?U/mL) and 1.82-fold greater than that of GS115/9Klip1FZlip1 #39 (4100?±?205?U/mL) in 10?L fermenters. The conversion ratio (C) and enantiomeric excess (ees) of racemic ibuprofen by immobilized CRL LIP1 reached 35.10 and 31.63%, respectively.  相似文献

All intertidal gastropods for which cold tolerance strategies have been assessed have been shown to be freeze tolerant. Thus, freeze tolerance is considered an adaptation to the intertidal environment. We investigated the cold tolerance strategies of three species of subtropical and temperate snails (Gastropoda: Littorinidae) to determine whether this group is phylogenetically constrained to freeze tolerance. We exposed dry acclimated and wet rehydrated snails to low temperatures to determine temperature of crystallisation (Tc), lower lethal temperature and LT50 and to examine the relationship between ice formation and mortality. Tc was lowest in dry Afrolittorina knysnaensis (–13.6±0.4 °C), followed by dry Echinolittorina natalensis (–10.9±0.2 °C) and wet A. knysnaensis (–10.2±0.2 °C). The Tc of both A. knysnaensis and E. natalensis increased with rehydration, whereas Tc of dry and wet Afrolittorina africana did not differ (–9.6±0.2 and –9.0±0.2 °C respectively). Wet snails of all species exhibited no or low survival of inoculative freezing, whereas dry individuals of A. knysnaensis could survive subzero temperatures above –8 °C when freezing was inoculated with ice. In the absence of external ice, Afrolittorina knysnaensis employs a freeze-avoidance strategy of cold tolerance, the first time this has been reported for an intertidal snail, indicating that there is no family-level phylogenetic constraint to freeze tolerance. Echinolittorina natalensis and A. africana both showed pre-freeze mortality and survival of some internal ice formation, but were not cold hardy in any strict sense.  相似文献

Summary Respiratory gas exchange and blood respiratory properties have been studied in the East-African tree frogChiromantis petersi. This frog is unusually xerophilous, occupies dry habitats and prefers body temperatures near 40°C and direct solar exposure. Total O2 uptake was low at 81 l O2·g–1·h–1±19.0 (SD) at 25°C increasing to 253.5 l O2·g–1·h–1±94.8 (SD) at 40°C giving aQ 10 value of 2.1. Skin O2 uptake at 25°C was 38.5% of total. The gas exchange ratio was 0.71 for whole body gas exchange, 0.61 for the lungs and 1.02 for the skin at 25°C.Blood O2 affinity was low with aP 50 of 47.5 mmHg at 25°C and pH 7.65. Then H-value at 25°C increased from 2.7 aroundP 50 to 5.0 at O2 saturations exceeding 70–80%. Surprisingly, blood O2 affinity was nearly insensitive to temperature expressed by a H value of ±1.0 kcal·mole between 25 and 40°C.The adaptive significance of the low O2 affinity, the increase ofn H with O2 saturation and the temperature insensitive O2-Hb binding is discussed in relation to the high and fluctuating body temperatures ofChiromantis.  相似文献

Summary The decreasing effect of -adrenergic blockade on skin resistance to vapor diffusion and the onset of cutaneous water evaporation in the pigeon (Columba livia) was investigated. Oral administration of 1, 2.3 and 5 mg propranolol to pigeons (268±53 g) initiated intensive trans-cutaneous water evaporation (CWE) up to 29.1 mg H2O·cm–2·h–1 in resting birds at 30°C air temperature (Ta), but had only a slight effect on CWE of birds exposed to 50 °C Ta.After 7 h of effective -adrenergic blockade (oral administration of 5 mg propranolol), skin and body temperature stabilized at 39.0±0.5 °C and 41.0±0.7 °C, compared to 40.2±0.8 °C and 41.9±0.6 °C in the control group, respectively. A slight hypothermia was accompanied by feather fluffing.Intradermal injection of 0.001, 0.01 and 0.12 mg propranolol also caused intensive CWE. Local -adrenergic blockade in relatively low blocker doses (0.001 and 0.01 mg propranolol) decreased skin resistance from a high value of 44.5 s·cm–1 to about 6.0 s·cm–1, and caused a sharp increase in CWE from a control value of about 4 to a high of 26.4 mg H2O·cm–2·h–1 during the first two hours of exposure to 30°C Ta.The possible role of -adrenergic blockade in regulation of trans-cutaneous water evaporation of latent heat dissipation is discussed.  相似文献

For the first time, growth throughout the life cycle of an annual fish, Cynolebias viarius, was assessed in the wild, within a temporary pool located in eastern Uruguay. Before pools dry out at the beginning of the warm season, C. viarius deposit eggs in the sediment. Embryos hatch when precipitation fills the pools again in March–April. During 1996, at biweekly or monthly intervals, environmental conditions were monitored and the length of 20 to 55 C. viarius measured. Pool size varied between 109 and 475m2, respectively. Water depth at its center reached between 10 (June) and 35cm (September). Water temperatures ranged from 6°C in June to 28.8°C in November. The water was slightly acidic (pH = 6.3 ± 0.2) and subsaturated with O2 (48–88%); conductivity averaged 258 ± 39Scm–1. On the first field trip after inundation (May 2), mean length of C. viarius was 9.9 ± 1.0mm. Maximum growth rate (0.66mm per day) was determined during the following two-week-interval and was associated with relatively high water temperatures (20–22°C). While fish length remained virtually unchanged during the colder winter months, C. viarius manifested growth, specially in weight, during the last third of the life cycle. Fish were sexually mature at 8 (67%) to 18 (100%) weeks of age. In the laboratory, specimens held at 25°C grew faster and reached sexual maturity at an earlier age (11 weeks) than at 15°C (10–16 weeks). Over the 5-months study period, mortality reached 37.5% at 15°C and 100% at 25°C. Results are compared to information available from other annual fishes.  相似文献

Diel and seasonal variations in abundance, population structure, biomass and production rate of the harpacticoid copepod Euterpina acutifrons were studied in the Cananéia Lagoon estuarine system, São Paulo, Brazil. Zooplankton samples were collected at 4-h intervals during multiple 24-h periods, from February 1995 to January 1996. Copepodites and adults of E. acutifrons were present in the plankton throughout the year (temperature, 18.6–29.4 °C; salinity, 4.5–33.0 psu; chlorophyll-a concentration, 1.32–20.42 g l–1). Abundance of E. acutifrons showed considerable diel variations. On most sampling dates, higher abundances were recorded at times when salinity was higher. Biomass varied from 0.044 ± 0.046 (daily mean ± SD) to 5.264±3.425 mg C m–3. The estimated production rates (minimum ± SD–maximum ± SD) were 0.034±0.035–4.95±3.25 (Ikeda-Motoda model), 0.035±0.036–5.123±3.347 (Huntley-Lopez model), and 0.016±0.017–2.101±1.372 mg C m–3 d–1 (Hirst-Sheader model).  相似文献

Summary Outwardly rectifying Cl channels in cultured human Jurkat T-lymphocytes were activated by excising a patch of membrane using the inside-out (i/o) patch-clamp configuration and holding at depolarized voltages for prolonged periods of time (1–6 min at +80 mV, 20°C). The single-channel current at +80 mV was 4.5 ± 0.3 pA and at –80 mV, it was 1.0 ± 0.4 pA. After activation, the probability of being open (P 0)for the lymphocyte channel was voltage independent. Activation of the Cl channel in lymphocytes was temperature dependent. Nineteen percent of i/o recordings from lymphocytes made at 20°C exhibited Cl channel activity. In contrast, 49% of recordings made at 30°C showed channel activity. The number of channels in an active patch was not significantly different at the two temperatures. Channel activation in excised, depolarized patches also occurred 20-fold faster at 30°C than at 20°C. There was no marked change in the single-channel conductance at 30°C. Open-channel conductance was blocked by 200 m indanyloxyacetic acid (IAA) or 1 mm SITS when applied to the intracellular side of the patch. Summary Acyl-CoA: lysophosphatidylcholine acyltransferase (LPCAT) (EC 2.3.1.23) activity was assayed in liver microsomes from rainbow trout,Salmo gairdneri, acclimated to 5°C and 20°C to assess its contribution to the temperature-induced restructuring of phospholipid acyl chain composition. The synthesis of phosphatidylcholine (PC) (from lyso-PC) was threefold the synthesis of phosphatidylethanolamine (PE) (from lyso-PE) under similar assay conditions. LPCAT activity (i) displayed an absolute requirement for lysophosphatidylcholine (LPC) and was enhanced by the presence of ATP, MgCl2 and CoA (which reduced the impact of endogenous acyl-CoA hydrolase activity by regenerating the acyl-CoA substrate) in the assay medium; (ii) remained linear with time up to 30 min; and (iii) increased linearly with microsomal protein concentration up to 0.2 mg/ml for the 20°C assay and 0.4 mg/ml for the 5°C assay. There was no difference in Km or Vmax values due to the acclimation history of the fish, but there were obvious differences due to assay temperature. The apparent Km values for LPC were 58.54±7.24 M and 12.26±2.14 M when assayed at 5°C and 20°C respectively; values for oleoyl-CoA were 9.11±0.78 M and 1.23±0.25 M under the same assay conditions. Activity was 1.99±0.31 nmol min–1 mg protein–1 when assayed at 5°C, and 3.8±0.45 nmol min–1 mg protein–1 when assayed at 20°C. These findings indicate that adjustments in the activity of LPCAT play no significant role in the temperature-induced restructuring of PC molecular species composition. Summary A high level ethanol-tolerant palm wineSaccharomyces yeast showed good adaptability (2.6±0.3 –16.4±0.7 days) in 5–30% unclarified molasses, and good viability in 10–30° Brix molasses-based media at 30°C for 3 days. Purification of the membrane-associated epoxide hydrolase from the yeast Rhodosporidium toruloides CBS 0349 to electrophoretic homogeneity was achieved in a single chromatographic step employing the affinity ligand adsorbent Mimetic Green. More than 68% of the total epoxide hydrolase activity present in the whole cells was recovered from the membrane fraction. The enzyme was purified 26-fold with respect to the solubilized membrane proteins and was obtained in a 90% yield. The purified epoxide hydrolase has an apparent monomeric molecular weight of 54 kDa, and a pI of 7.3. The enzyme was optimally active at 30–40 °C, and pH 7.3–8.5. The enzyme is highly glycosylated with a carbohydrate content >42%. The specific activity of the purified enzyme for (±)-1,2-epoxyoctane is 172 mol min–1 mg protein–1. The amino acid composition of the protein was determined. This is the first report of a yeast epoxide hydrolase purified to homogeneity in milligram amounts.  相似文献

Reproduction of the dominant Antarctic copepod Calanus propinquus was studied in February–April, 1989 aboard the R.V Dmitry Mendeleev during cruise N° 43 to the Weddell Sea. Single females were kept at 0 °C in the laboratory for 56 days with abundant food concentration (above 300 µg C l–1 of Platymonas viridis). Females released clutches at night at 2–3 day intervals. Most clutches contained from 10 to 40 eggs, mean 37.3 eggs female–1. Average carbon content of an egg was 0.37 ± 0.05 µg C. The maximum daily egg production rate of 30–50 eggs female–1 d–1 was observed for the first 3 days of the laboratory incubation, corresponding to 3.7–6.2% body C. The state of gonadal development of females showed the decline of the reproductive season in late February. The data suggest that egg laying in the region under study starts in December and lasts until March. The state of ovarian maturation, changes in vertical distribution and biochemical body composition of females suggest the possibility of two-year life cycle in C. propinquus in the southern Weddell Sea.  相似文献

The CO2 evolution of intact potato tubers (Solanum tuberosum, L., var. Bintje) was analyzed during a 10-day period of their warm (25 ± 2°C) or cold (5 ± 1°C) storage, to evaluate cold-stress effects on expression and activities of plant uncoupling mitochondrial protein (PUMP) and alternative oxidase (AOX). CO2 evolution rates were analyzed at 20°C, to reflect their possible capacities. The 20°C CO2 production declined from 13 to 8 mg kg–1 h–1 after 2 days of warm storage and then (after 3 to 7 days) decreased from 8 to 6.5 mg kg–1 h–1. In contrast, 20°C CO2 evolution did not change after the first day of cold storage, increased up to 14.5 mg kg–1 h–1 after 2 days, and decreased to about 12 mg kg–1 h–1 after 3 to 7 days of cold storage. Cold storage increased PUMP expression as detected by Western blots and led to elevated capacities of both PUMP (44%) and CN-resistant AOX (10 times), but not the cytochrome pathway. Since we found that cold storage led to about the same mitochondrial respiration of 40 nmol O2 min–1 mg–1 attributable to each of the respective proteins, we conclude that both AOX and PUMP equally contribute to adaptation of potato tubers to cold.  相似文献

Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation.  相似文献

The kinetics of glutaraldehyde inactivation of a protoplasmic (-fructofuranosidase) and an extracytoplasmic (acid phosphatase) enzyme inSaccharomyces rouxii cells were studied at pH 5.5 and 30°C. The effects of glutaraldehyde concentration (0.5–3%), pH value, and temperature were surveyed by varying the fixation conditions. Cells from 1- to 10-day cultures retained 50–75% of their acid phosphatase activity and 15–24% of their -fructofuranosidase activity after 1-h exposures to 0.5% glutaraldehyde. The surviving -fructofuranosidase activity remained physically cryptic and was revealed only after further membrane perturbation with ethyl acetate. This crypticity barrier disappeared after overnight incubation of the treated cells at 4°C, with or without added glutaraldehyde, during which time the enzyme was resistant to further inactivation. The velocity ratio for raffinose versus sucrose, as substrate, decreased in treated cells, and changes inV max andK m were indicative of frank destruction of some enzyme molecules as well as modification of survivors. A comparable set of changes was also generated by treating cell-free extract with glutaraldehyde. Glutaraldehyde (0.5%) killed all yeast cells at 30°C within 5 min; at 4°C survival rates were quite high—81% after 15 min and 65% after 1 h. The bearing of these examples of enzyme inactivation, permeability barrier abolition, and structural stabilization on the general problems of yeast cytochemistry is discussed.  相似文献

Moina mongolica, 1.0-1.4 mm long and 0.8 mm wide, is an Old World euryhaline species. This paper reviewed the recent advances on its autecology, reproductive biology, feeding ecology and perspective as live food for marine fish larviculture. Salinity tolerance of this species ranges from 0.4–1.4 to 65.2–75.4. Within 2–50 salinity, Moina mongolica can complete its life cycle through parthenogenesis. The optimum temperature is between 25 °C and 28 °C, while it tolerates high temperature between 34.4 °C and 36.0 °C and lower temperature between 3.2 °C and 5.4 °C. The non-toxic level of unionised ammonia (24 h LC50) for M. mongolica is <2.6 mg NH3–N l–1. Juvenile individuals filter 2.37 ml d–1 and feed 9.45×106 algal cells d–1, while mature individuals filter 9.45 ml d–1 and consume 4.94×106 algal cells d–1. At 28 °C, M. mongolica reaches sex maturity in 4 d and gives birth once a day afterward; females carry 7.3 eggs brood–1 and spawn 2.8 times during their lifetime. A variety of food can be used for M. mongolica culture including unicellular algae, yeast and manure, but the best feeding regime is the combination of Nannochloropsis oculata and horse manure. Moina mongolica reproduces parthenogenetically during most lifetime, but resting eggs can be induced at temperature (16 °C) combined with food density at 2000–5000 N. oculata ml–1. The tolerance to low dissolved oxygen (0.14–0.93 mg l–1) and high ammonia makes it suitable for mass production. Biochemical analyses showed that the content of eicospantanoic acid (20:53) in M. mongolica accounts for 12.7% of total fatty acids, which is higher than other live food such as Artemia nauplii and rotifers. This cladoceran has the characteristics of wide salinity adaptation, rapid reproduction and ease of mass culture. The review highlights its potential as live food for marine fish larvae.  相似文献

Production of the anti-listerial bacteriocin, pediocin, by lactic acid bacteria (LAB) transformed with the cloning vector pPC418 (Ped+, 9.1 kb) was influenced by composition of media and incubation temperature. Maximum pediocin production, tested against Listeria innocua, by electrotransformants of Lactococcus lactis ssp. lactis was measured in tryptone/lactose/yeast extract medium after 24 h growth at 30 °C, while incubation at 40 °C was optimum for Ped+ transformants of Streptococcus thermophilus and Enterococcus faecalis. The amount of pediocin produced by S. thermophilus in skim milk and cheese whey supplemented with 0.5% yeast extract was estimated as 51000 units ml–1 and 25000 units ml–1, respectively. Pediocin production remained essentially unchanged in reconstituted skim milk or whey media diluted up to 10-fold. The results demonstrate the capacity of recombinant strains of LAB to produce pediocin in a variety of growth media including skim milk and inexpensive cheese whey-based media, requiring minimum nutritional supplementation.  相似文献

In vitro cultures of Nephrolepis exaltata and Cordyline fruticosa were stored at 5°, 9° or 13°C, at a low irradiance (3–5 mol m–2 s–1) or in darkness. Prior to storage the cultures were subjected to 18°, 21°, 24° or 27°C and 15, 30 or 45 mol m–2 s–1 in a factorial combination.The optimal storage conditions for Nephrolepis were 9°C in complete darkness. These cultures were still transferable to a peat/perlite mixture at the end of the experimental period of 36 months.The optimal storage conditions for Cordyline were 13°C and a low light level (±3–5 mol m-2 s-1). When the pre-storage conditions were normal growth room conditions (24°C and 30 mol m-2 s-1), in vitro cultures could be stored for 18 months. With the most favourable pre-storage treatment (18°C and 15 mol m-2 s-1) some cultures still had green shoots after 36 months of storage, but did not survive transfer to peat/perlite.Pre-conditioning before storage was most favourable for Nephrolepis, and not that important, but still favourable, for Cordyline. There was an interaction between pre-storage temperature and pre-storage irradiance. To develop an effective fermentation system for producing Escherichia coliphytase AppA2, we expressed the enzyme in three inducible yeast systems: Saccharomyces cerevisiae (pYES2), Schizosaccharomyces pombe (pDS472a), and Pichia pastoris (pPICZ A), and one constitutive system: P. pastoris (pGAPZA). All four systems produced an extracellular functional AppA2 phytase with apparent molecular masses ranging from 51.5 to 56 kDa. During 8-day batch fermentation in shaking flasks, the inducible Pichia system produced the highest activity (272 units ml–1 medium), whereas the Schizo. pombesystem produced the lowest activity (2.8 units ml–1). The AppA2 phytase expressed in Schizo. pombehad 60–75% lower Kmfor sodium phytate and 28% higher heat-stability at 65 °C than that expressed in other three systems.
