Two reliable systems for the cell-mediated passive transfer of adjuvant arthritis have been developed. Donor rats were sensitized with Mycobacterium butyricum in mineral oil. In the first system, intravenous injection of adjuvant-sensitized donor lymph node or spleen cells into adult-thymectomized, lethally irradiated, bone marrow cell-reconstituted syngeneic rats induced arthritis in the recipients. In the second system, adjuvant-sensitized donor lymph node or spleen cells were cultured in vitro with concanavalin A; these cells induced arthritis in normal recipients as well as in thymectomized, irradiated, bone marrow cell-reconstituted recipients. The passively transferred disease in both systems resembled classical adjuvant-induced arthritis clinically, radiographically, and histologically. Neither irradiated, adjuvant-sensitized donor cells nor cells from donors not injected with complete adjuvant could passively transfer arthritis.  相似文献

The response and subsequent recovery of mouse haemopoietic progenitor cells (spleen colony forming cells and agar colony forming cells) has been studied following two cytotoxic agents. Busulphan was administered to normal mice and vinblastine to mice where the progenitor cell proliferation rate had been increased by a period of continuous γ-irradiation. With both these agents there is a difference between the response of the spleen colony forming cells and the agar colony forming cells during the first five days. They then recover together, but much more slowly after busulphan than after vinblastine even though their proliferation rate is increased. The rate of progenitor cell recovery after busulphan is increased if the progenitor cells are depleted further by vinblastine. However, methotrexate, which severely depletes the peripheral blood count and bone marrow cellularity but not the progenitor cells, has no effect on the recovery following busulphan. These results suggest that following cytotoxic agents the agar colony forming cells (“committed” stem cells) are not self-maintaining but are dependent on a supply of cells from the pluripotential spleen colony forming cells. In addition it appears that the depletion of the progenitor cells of the bone marrow and not the depletion of the maturing cells, provides a stimulus for stem cell recovery.  相似文献

The responsiveness of bone marrow cells from tolerant donors was examined by reconstitution of lethally irradiated tolerogen-free recipients. In these animals, stem cells from tolerant donors gave rise to immunologically competent antigen sensitive B cells. The antibody produced by these cells could be detected by a sensitive plaque assay in liquid and by antigen elimination. The antibody was not demonstrable by an assay which only detected plaque forming antibody which was highly avid or was formed in large quantity per cell. In lethally irradiated animals, partially purified B cells from a tolerant animal could not cooperate with T cells from normal donors to reconstitute immunological responsiveness to immunogenic doses of the tolerance inducing antigen. We concluded that antigen sensitive B cells in the bone marrow become unresponsive following administration of tolerogenic forms of antigen. Responsiveness of the reconstituted recipient animals was due to the differentiation of donor stem cells and subsequent antibody production by their descendants. Earlier contradictory findings could be unified in terms of these observations and conclusions.  相似文献

Adult CBA mice were exposed to thymectomy, lethal irradiation, and protection by syngeneic bone marrow transplantation. In some experiments syngeneic bone marrow of donors, treated with hydrocortisone in a dose of 125 mg/kg for 3 days was used. The bone marrow of these donors contained cells with the Q-marker. Thymectomized and lethally irradiated animals subjected to the transplantation of syngeneic bone marrow from hydrocortisone-treated donors rejected the skin allotransplants, and the lymph node cells of these mice suppressed the endogenous colony-formation in the sublethally-irradiated hybrids (CBA X C57Bl/6) F1.  相似文献

The colony formation in spleen of lethally irradiated syngeneic or hybrid recipients was studied after transplantation of bone marrow cells, with or without macrophages from lymph nodules or from peritoneal cavity of mice, cells of macrophage-like cell line J-774, and monocytes from peripheral blood of healthy donors. The direction of stem cell differentiations in the presence of all the types of mononuclear phagocytes was seen to change from mainly erythroid to mainly myeloid one. The ratio of erythroid to myeloid colonies became equal to 0.5-0.9 instead of 2.0, when bone marrow cells were injected with equivalent quantity of mononuclear phagocytes. This new regulatory function of mononuclear phagocytes is discussed.  相似文献

Thymectomized, lethally irradiated mice reconstituted with normal bone marrow cells succumbed when challenged ip with rat Yoshida ascites sarcoma (YAS) cells 40 days after irradiation and reconstitution. In contrast, thymectomized irradiated mice reconstituted with bone marrow cells from YAS-immune donors rejected the subsequent tumor challenge. Pretreatment of the bone marrow cells from immune donors with anti-Thy 1.2 antiserum and complement completely abolished the transfer of anti-YAS resistance.Bone marrow cells from donors thymectomized 2 months before immunization enabled almost all recipients to reject YAS, but bone marrow cells from donors thymectomized 8 months before immunization protected only 50% of the recipients. Further analysis showed that mice thymectomized 8 months before immunization failed to generate anti-YAS antibody response, whereas the antibody response of mice thymectomized 2 months before immunization did not differ from that of non-thymectomized age-matched control mice. The data suggest that the immune reaction of mice against xenogeneic YAS requires long-lived T2 lymphocytes.  相似文献

It has been reported elsewhere that in addition to enhancing the expression of metallothionein genes, the previous injection of cadmium salts into sublethally X-irradiated mice increases by 10 times the number of endogenous spleen colonies. To understand the mechanism of the strong radioprotective cadmium effect donors and recipients were treated separately. It is shown that the survival of exogenous bone marrow colony-forming cells in lethally irradiated recipient remains at the control level independently of the donor cadmium treatment, whereas the injection of cadmium nitrate to recipient mice leads to the stimulation of colony formation by 1.7-1.8 times. The data allow to conclude that the cadmium effect on the survival of colony-forming hemopoietic murine cells after X-irradiation is not mediated by the enhanced expression of metallothionein genes.  相似文献

Using the spleen colony assay technique, it has been shown that busulphan (‘Myleran’) in a dose of 10-2 g/kg (1/2 LD50), causes a marked and prolonged depression (over 90%) in the number of colony forming units per femur (CFU/femur). This depression is apparent before there is any marked reduction of the total cell count per femur and is maximal 2–4 days after an intraperitoneal (i.p.) injection of the drug. It is then followed by a steady recovery, normal values being reached after about 20 days. In contrast, aminochlorambucil (2·5 x 10-3) g/kg = 1/2 LD50) although producing a rapid fall in the marrow cellularity has no discernable effect on the CFU/femur. If, however, a depression of CFUs is first induced by busulphan and, after allowing time for 50% recovery (13 days), aminochlorambucil is now given, a further severe depression of the CFUs/femur occurs resulting in a considerable prolongation of the neutropenia observed in the blood. The possible implications of this in the mode of action of these two drugs, and in the chemotherapy of leukaemia, are discussed.  相似文献

A method has been developed of decreasing immunologic activity of lymphocytes from spleen, lymph nodes, and bone marrow (freshly collected or frozen) with a high percentage of intact stem cells. In experiments in vivo on lethally irradiated mice, it was demonstrated that during combined transplantation to the recipients of preserved bone marrow from two genetically different donors, a rapid decrease or absence of the effect of inactivation of hemopoietic stem cells under the influence of allogeneic lymphocytes was observed in the mixed graft. When it is necessary to transplant large quantities of bone marrow from several genetically different donors, the use of cryopreserved bone marrow is preferable to freshly drawn marrow due to the higher proliferative activity and the decreased risk in the development of immunological reactions.  相似文献

Humoral factors affecting granulopoiesis have been detected by culturing bone marrow cells in intraperitoneal diffusion chambers. This study investigated the possibility that treatment with drugs alters the ability of cells to respond to these factors. Three days after treatment with 200 mg kg-1 cyclophosphamide, donor marrow cells were more responsive than normal cells to the factors produced in cyclophosphamide pretreated hosts. The optimum timing of host pretreatment for maximum colony stimulation also differed for cells from cyclophosphamide treated and untreated donors and these effects were found to depend on the dose of drug given to the animals.  相似文献

The data on the application of monoclonal antibodies (ICO-10) and rabbit complement for working the conditions of allogeneic bone marrow transplantation are presented in the paper. The treatment with monoclonal antibodies and bone marrow complement from BALB/c mice for 2 times prevented the development of transplant versus host reaction and completely protected lethally irradiated (CBA X X C57B1/6)FI mice-recipients from death. Thymus atrophy and the absence of T-cells in the peripheral blood was observed in these mice. The erythrocytes had markers characteristic of BALB/c and (CBA X C57B1/6)FI mice. Mouse splenocytes did not respond to the cells of donors and recipients in mixed lymphocyte culture reaction.  相似文献

Serum creatinine and urea changes as well as protein, erythrocytes, and leucocytes excretion with the urine in allogenic bone marrow recipients are discussed. An increase in serum creatinine and urea together with proteinuria and hematuria were noted in 1/3 of patients treated with busulphan and cyclophosphamide prior to bone marrow transplantation. In cases of bone marrow graft rejection or post-transplantation complications no abnormalities in the value of investigated parameters were noted, especially immediately before death. Possible causes of such renal function disorders are also discussed.  相似文献

When transplanting the bone marrow cells from adult C57BL mice to the lethally irradiated (CBA X C57BL) F1 hybrids of different age, the decrease of the colony forming activity of the stem haemopoietic cells was observed in the spleen of the older recipients, as compared with the 3 months old ones. The joint transplantation of the bone marrow and thymus cells resulted in both the cases in the stimulation of the growth of colonies. The number of endogenous colonies of haemopoietic cells arising in the spleen of animals following the sublethal irradiation was greater in younger hybrids. After the induction of the "transplant versus host" reaction by the lymph node or spleen cells from the CBA mice, the relative weight of spleen and regional lymph node, respectively, in the older recipients exceeded those in the younger ones.  相似文献

Normal bone marrow cells from Wistar Furth rats were competent to transfer immune responsiveness to bovine serum albumin to thymectomised, irradiated, syngeneic recipients. When the bone marrow cells were taken from donors thymectomised early in life they were incompetent, but competence was restored by addition of normal thymus cells. It was concluded that normal Wistar Furth bone marrow cells contain some thymus-derived cells. Thymus cells from tolerant donors were less effective in cooperation with bone marrow cells, however the thymus cells appeared less tolerant than their donors.  相似文献

THE ROLE OF BONE MARROW OF X-IRRADIATED MICE IN THYMIC RECOVERY   总被引:1,自引:0,他引:1  
The influence of the bone marrow on the repopulation of the thymus in X-irradiated mice has been investigated.
It was observed that the thymus and a certain population of bone marrow lymphocytic cells were repopulated in parallel in a cyclic fashion. This occurred either after a single exposure of mice to 400 R or after serial weekly X-ray treatments with 170 R. Lethally irradiated recipients which were grafted with bone marrow cells obtained 12-24 days after four weekly irradiations of donor mice with 170 R also exhibited a cyclic repopulation of both the thymus and the bone marrow lymphocytic population. In contrast, mice which were transplanted with bone marrow cells from unirradiated donors, containing an equal number of stem cells (CFU), exhibited a continuous rather than a cyclic recovery of both cell populations. the bone marrow stem cells of mice recovering from X-irradiation were found to have a decreased proliferative activity, since they produced significantly smaller spleen colonies in lethally irradiated recipients than marrow cells from unirradiated mice.
The results were interpreted as indicating that the bone marrow lymphocytic cells may act as thymic precursor cells and that thymic lymphopoiesis is dependent on the presence of such cells. Evidently, the production of lymphocytic cells will decrease when the stimulus for granulocyte production increases due to the limited proliferative activity of the surviving bone marrow stem cells after irradiation. This may result in a cyclic variation of the production of bone marrow lymphocytic cells and it follows that thymic lymphopoiesis will run parallel.  相似文献

RECENT progress in the definition of the DL-A antigens within a closely bred colony of beagles maintained at the Mary Imogene Basset Hospital (Cooperstown colony) has shown that these antigens are components of the principal system of histocompatibility (DL-A) in the dog1–3. Dausset and associates4 have described the segregation of DL-A antigens in 679 offspring of 151 consecutive matings within the Coopers-town colony. Through methods of backcross analysis, 23 DL-A haplotypes (that is, specificities determined by the DL-A region of one single chromosome) have been identified and the DL-A genotypes of 1,302 offspring of 517 matings in this colony have been defined4. This combined genetic and serological information has provided an opportunity to reassess the reported survival rates of allogeneic bone marrow transplantation in serologically matched littermate dogs5. We describe here the long term survival of bone marrow transplants and renal allografts obtained from the same donors in nine littermate and three non-littermate recipients selected on the basis of genetic and serological criteria of DL-A compatibility in the Cooperstown colony. The results are contrasted with the survival of twenty-two renal allografts performed in untreated recipients selected on the basis of similar criteria.  相似文献

Effect of transplantation of syngeneic bone marrow in the dose of 1 X 10(7) cell/ml on the state of pancreatic gland in lethally irradiated recipients has been studied at different stages of posttransplantation period for 3 months using 250 linear male rats G (CBA x C57B). Histological and biochemical investigation, conducted in dynamics, have shown that transplantation of native and cryopreserved bone marrow to lethally irradiated animals facilitates activation of compensatory-restoration processes manifesting themselves in mitotic division of glandular and epithelial cells, as well as optimizes exchange of carbohydrates in the irradiated organism.  相似文献

The authors analysed the capacity of various temperature fractions of RNA isolated from the spleen of donors of the bone marrow cells (of mice C57BL/6I) and recipients--hybrids (CBA X C57BL/6I) F1 to abolish the depression of colony formation in the nonsyngenous organism. In the administration of bone marrow cells of mice of parental genotype C57BL/6I of the irradiated recipients F1 there is observed a sharp depression of the number of colony forming units in the spleen F1. This depression can be eliminated by preliminary incubation of the bone marrow cells of mice of parental genotype with a 63 degrees fraction of the recipient's RNA. Preliminary inculation of the bone marrow cells of mice of parental genotype with 85 degrees and cytoplasmic fractions of recipient's RNA led to a partial restoration of colony formation only. The 45 degrees and 55 degrees RNA fractions of the recipient's RNA produced no restoring action. None of the temperature RNA fractions of the RNA of donor bone marrow cells were capable of abolishment of the colony formation depression in the nonsyngenous organism. It is supposed that restoration of the colony forming capacity in the nonsyngenous organism was connected with the activity of matrix RNA of the 63 degrees fraction obtained from the recipient's spleen.  相似文献

Delayed-type hypersensitivity (DH) to Listeria antigens was induced in inbred C3Hf/Umc mice by intravenous injection of a sublethal dose of viable Listeria monocytogenes. Bone marrow, spleen, and lymph node cells from the immune mice were capable of passive transfer of DH to syngeneic neonatally thymectomized or lethally (900 R) irradiated recipients. Immune thymus cells as well as immune serum were ineffective in transferring DH to irradiated animals. In vitro treatment with antitheta isoantibody (anti-θ) and complement abolished the capacity of spleen and bone marrow cells from immune donors to transfer DH to irradiated hosts, indicating the thymus dependency of this cell population. The results with bone marrow indicate the existence of a small, but biologically significant, thymus-dependent population in this tissue.  相似文献
