The infection of roots by the pathogenic Oomycete Pythium aphanidermatuminvolves interactions between the fungal zoospores and rootsurface mucilage polysaccharides. After initial recognitionat the root surface the zoospores are triggered to encyst duringwhich adhesive glycoproteins are secreted followed by a fibrillarcyst wall. In this paper a simple in vitro assay has been usedto assess the ability of a variety of macromolecules to inducezoospore encystment. Mucilage polysaccharides of the cress rootsurface trigger encystment. Whole mucilage was fractionatedby gel filtration and a fraction low in uronic acid, containing5% fucose, was shown to be more effective in triggering encystmentthan a uronic acid-rich fraction. Encystment can also be inducedby commercial pectin. The lectin Con A, and PA1, one of a rangeof monoclonal antibodies specific for zoospore surface antigens,also triggered encystment. In Western blotting experiments PA1recognizes protein epitopes of a 75 kDa surface antigen. Theresults suggest that at least one mechanism of zoospore triggeringmay involve a specific zoospore surface receptor. The production of resting cysts in four species of dinoflagellates(Scrippsiella trochoidea, Ensiculifera sp., Alexandrium lusitanicumand Lingulodinium polyedra) was studied in response to severalenvironmental factors of ecological importance (nitrate, phosphate,iron, copper and cyanocobalamin deficiencies, high concentrationsof copper, turbulence, darkness plus concentration. as wellas various media biologically conditioned by dinoflagellates)using unialgal cultures and enrichments of natural populations.Some nutritional deficiencies, mainly phosphorus or nitrogen(in this order), are the most effective inducers of encystment.Among the other deficiencies tested, only iron deficiency wasimportant, affecting only A.lusitanicum. In some cases, biologicalconditioning produced considerable encystment reductions, makingit an important means of competition between species. We suggestthat encystment may be induced in these neritic species by deficienciesin compounds that act as indicators of changes in the hydrographicconditions to which the particular species are adapted.  相似文献

A mathematical model of the energy budget of the coped Acartiatonsa is proposed. It takes into account the processes of ingestion(encounter, capture, ingestion sensu stricto), digestion (assimilation,gut transit and egestion), catabolic expenses and biomass production.In order to represent the potential effects of small-scale turbulenceon the whole physiological processes of a copepod, some processsubmodels already published are combined. A major assumptionof the model is a satiety effect resulting from midgut filling,which leads to a decrease in the feeding activity. The modelpermits the simulation of the short-term dynamics of ingestionunder different food and turbulence conditions, as well as anintegrated physiological balance over 24 h. The model is validatedthrough comparison with data at both scales Simulations showthat turbulence increases ingestion rates and gut contents,and causes a decrease in gut passage time and assimilation efficiency.As a consequence, the dependent physiological processes areaffected differently by turbulence, which preferentially increasesegestion and egg-production rates Simulated daily ingestionrates of A.tonsa, for suspension feeding on. Thalassiosira weissflogiiand for ambush predation on Strombidium sulcatum, are in goodagreement wth the available experimental observations. The concurrentdirect effect of turbulence on the copepod's metabolism, dueto increased escape reactions, is also simulated. Results ofthe model show that a switch from suspension feeding on diatomsin calm condition to ambush predation on ciliates in turbulentconditions, might allow A.tonsa to maintain its gross growthefficiency at the same level. It is suggested that a dynamicrepresentation of processes occurring over a time scale of afew seconds is necessary to obtain, once integrated over 24h, the correct simulation of the effect of microscale turbulenceon ingestion and the related physiological processes.  相似文献

The investigation was carried out at one station in Korsfjorden,a typical deep silled fjord of western Norway. During 14 cruisesfrom 4 February to 30 June 1977 ATP, chlorophll a, phaeopigmenta, and in situ 14C-assimilation were measured in the net (>30µm), nano and ultraplankton (<5 µm). Sampleswere collected from five light depths within euphotic zone.The impact of hydrographical conditions and light regime onthe bloom dynamics was also studied. In the periods 4 February-7 March and 13 April-30 June, ultraplankton contributed >60%to the total primary production while net and nanoplankton dominatedfrom 7 March to 13 April. The diatoms Skeletonema costatum,Chaetoceros compressits and C. debilis, and Rhizosolenia hebetatavar. semispina made up the main part of the biomass on 21 March,28 March and 4 April respectively. A shade adapted diatom societywas located at the top of the nutricline in late June with S.costatum, Chaetoceros spp., and Thalassiosira spp. as the dominantspecies. The highest assimilation number of eight for the netplankton and four for the ultraplankton were found at the depthof 32% light intensity on 28 March and 24 May respectively.Linear relationships were found between chlorophyll a and ATPfor the different size fractions with regression slopes rangingfrom 4.3 to 5.8. The total primary production for the periodof investigation was calculated to 74 g C m–2. Light regimeand water column stability were decisive factors for the outburstof the first diatom bloom in late March. Grazing on net planktondiatoms increased during late March-early April. Changes inthe longshore wind-stress component were found to be essentialfor the understanding of the bloom dynamics.  相似文献

对东海原甲藻和中肋骨条藻按照起始Chl-a比1∶5、1∶1和5∶1进行了f/2条件下的共培养实验,以探讨这两种藻的种间竞争特征。实验结果表明在共培养体系中,中肋骨条藻完全占优势,而东海原甲藻的生长受到明显的抑制。应用Lotka-Volterra种间竞争模型对共培养实验进行模拟的结果表明,东海原甲藻与中肋骨条藻的种间竞争结果与初始密度配比无关,中肋骨条藻总会竞争胜过东海原甲藻。为了探讨他感作用对东海原甲藻和中肋骨条藻种间竞争的影响,采用了中肋骨条藻的无藻细胞滤液来进行培养实验。实验结果显示,中肋骨条藻滤液对东海原甲藻及其本身的生长均无明显影响,这表明他感作用并非中肋骨条藻获得优势的主要竞争方式。  相似文献

Striga hermonthica is a root hemiparasitic angiosperm nativeto the African semi-arid tropics. It is a major weed of C4 cerealsbut locally it is also an important weed of the C3 plant, rice[Oryza sativa). Infected rice plants produced 17% and 42% ofthe total biomass of uninfected plants when grown at two differentammonium nitrate concentrations, 1 and 3 mol m–3, respectively.S. hermonthica prevented grain production at both concentrationsof nitrogen. At the lower concentration no heads were produced.At the higher concentration head weight was only 6% of uninfectedcontrols. S. hermonthica also altered the partitioning of drymatter between plant parts, such that shoot growth was reducedto a greater extent than root growth. As a consequence the root-to-shootratio of infected plants was approximately five times greaterthan that of uninfected control plants. Light saturated ratesof photosynthesis In infected plants were 56% and 70% of thoseof uninfected controls, at low and high nitrogen, respectively.Infection also led to lower values of stomatal conductance althoughthe substom-atal CO2 concentration was unaffected. Analysisof the response of photosynthesis to substomatal CO2 concentration(A/CI curves) demonstrated that lower rates of photosynthesiscould not be solely attributed to lower stomatal conductances.Lower initial slopes and asymptotic rates suggest that bothcarboxylation and processes controlling regeneration of ribulose-1,5-bisphosphate are reduced by infection. The data are discussedwith respect to the influence of S. hermonthica on the growthand photosynthesis of C4 hosts, where in contrast to the situationwith rice, nitrogen feeding results in a marked alleviationof the effects of the parasite on the host. Tardigrades have two forms of dormancy, namely cryptobiosis and encystment. The encystment is a form of diapause known for a limited number of species of tardigrades and still little studied. To increase the knowledge on encystment, two species of eutardigrades from Italy have been considered: the moss-dwelling Amphibolus volubilis (Eohypsibiidae), and the limnic Dactylobiotus parthenogeneticus (Murrayidae). Cysts have been collected in nature, or induced under laboratory conditions. In the latter case, it was possible to follow the several steps of encystment processes. Two different types of cyst (“type 1” and “type 2”) have been found in A. volubilis, while in D. parthenogeneticus only one type has been found. In general, the ovoid-shaped cysts are constituted by a series of cuticles surrounding the animals and resemble an onion or a Matrioshka Russian doll. In all three types of cyst, the encystment processes show both common and peculiar traits. Encystment begins with the discharging of the sclerified parts of the buccal-pharyngeal apparatus, as in the molting process, but without the loss of the old cuticle. Then, two or three new cuticles are serially synthesized, according to cyst type. In A. volubilis, the ultrastructure of these new cuticular involucra is similar to that of non-encysted animal cuticles, while in D. parthenogeneticus the ultrastructure of the new cuticular involucra differs from that of non-encysted animal cuticle. A modified buccal-pharyngeal apparatus has been observed both in “type 2” cyst of A. volubilis and in the D. parthenogeneticus cyst.  相似文献

Free‐living amoebae are ubiquitous protozoa commonly found in water. Among them, Acanthamoeba and Vermamoeba (formerly Hartmannella) are the most represented genera. In case of stress, such as nutrient deprivation or osmotic stress, these amoebae initiate a differentiation process, named encystment. It leads to the cyst form, which is a resistant form enabling amoebae to survive in harsh conditions and resist disinfection treatments. Encystment has been thoroughly described in Acanthamoeba but poorly in Vermamoeba. Our study was aimed to follow the encystment/excystment processes by microscopic observations. We show that encystment is quite rapid, as mature cysts were obtained in 9 h, and that cyst wall is composed of two layers. A video shows that a locomotive form is likely involved in clustering cysts together during encystment. As for Acanthamoeba, autophagy is likely active during this process. Specific vesicles, possibly involved in ribophagy, were observed within the cytoplasm. Remarkably, mitochondria rearranged around the nucleus within the cyst, suggesting high needs in energy. Unlike Acanthamoeba and Naegleria, no ostioles were observed in the cyst wall suggesting that excystment is original. During excystment, large vesicles, likely filled with hydrolases, were found in close proximity to cyst wall and digest it. Trophozoite moves inside its cyst wall before exiting during excystment. In conclusion, Vermamoeba encystment/excystment displays original trends as compare to Acanthamoeba.  相似文献

A sediment trap study was conducted at Daya Bay, South ChinaSea, to investigate the relationships between encystment andpopulation dynamics of Scrippsiella trochoidea from December1999 to January 2001. A dense bloom of S. trochoidea occurredduring the study period from August to September 2000, withthe maximum cell number of 3.18 x 104 cells mL–1.Two morphotypes of cysts, one with a thick calcareous wall (calcifiedcyst) and another without the obvious calcareous cover (non-calcifiedcyst), were observed during this investigation. The morphologicaland excystment characteristics of these two cyst types werestudied as well. Mass encystments of S. trochoidea, with themaximum of 3.05 x 105 cysts m–2 d–1for calcified cyst, and 1.54 x 107 cysts m–2 d–1for non-calcified cyst, coincided with the maximum abundanceof the vegetative cells. Encystment caused the transfer of atotal of 2.24–4.49 x 108 cells m–2 vegetativecells from the water column to the sea bottom during the bloomand resulted in a considerable loss of the bloom population.High assemblages of cysts of S. trochoidea were detected inthe surface sediments as well. This rich ‘seed bed’in the surface sediments caused by the high efficiency of encystmentafter blooms acting as a benthic reservoir for future vegetativepopulation, together with the short dormant period (15–26days) and high germination rate (50–90%), may explainthe repeated occurrence of S. trochoidea blooms in Daya Bay.  相似文献

Nitrate utilization has been characterized in nitrogen-deficient cells of the marine diatom Skeletonema costatum. In order to separate nitrate uptake from nitrate reduction, nitrate reductase activity was suppressed with tungstate. Neither nitrite nor the presence of amino acids in the external medium or darkness affects nitrate uptake kinetics. Ammonium strongly inhibits carrier-mediated nitrate uptake, without affecting diffusion transfer. A model is proposed for the uptake and assimilation of nitrate in S. costatum and their regulation by ammonium ions.  相似文献

Summary The importance of the surface structure and chemistry in zoospores and cysts of oomycetes is briefly reviewed and the organelle systems associated with encystment described. The surface structure and chemistry of primary and secondary zoospores and cysts ofSaprolegnia diclina (a representative saprophytic species) andS. parasitica (a representative salmonid fish pathogen) were explored using the lectins concanavilin A (Con A) and wheat germ agglutinin (WGA) and monoclonal antibodies (MAbs) raised against a mixed zoospore and cyst suspension ofS. parasitica. The binding of lectins and antibodies to spores was determined using immunofluorescence microscopy with fluorescein isothiocyanate-labelled probes and with electron microscopy with gold-conjugated probes applied to spore suspensions post-fixation. In both species Con A, which is specific for glucose and mannose sugars, bound to both the surface of primary and secondary zoospores (the surface glycocalyx) and their cyst coats and readily induced zoospore encystment. The binding to the cysts appeared to be mainly associated with the matrix material released from the primary and secondary encystment vesicles and which appeared to diminish with time. No binding to germ tube walls was observed with this lectin. The MAb labelling showed a generally similar binding pattern to the primary and secondary cysts to that observed with Con A, although the binding to zoospores was more variable. Primary zoospores bound the antibodies but secondary zoospores appeared less reactive. It is suggested that the MAbs share a common epitope with one or more of the Con A-binding components. In both species WGA, which is specific for amongst other things the sugar N-acetyl glucosamine, bound to localised apical patches on the primary zoospores. This lectin also binds to the ventral groove region of secondary zoospores ofS. diclina, which were induced to encyst by this lectin. In contrast secondary zoospores ofS. parasitica were not induced to encyst by the addition of WGA and showed a patchy dorsal binding with this lectin. WGA also binds to both the inner wall of discharged primary cysts and the young germ tube walls of both species. The Conductance model is a simple mechanistic model used topredict the growth of species in monoculture or mixtures fromparameter values derived from plants grown in isolation. Incontrast to many mechanistic models that require extensive parameterization,the Conductance model is able to capture the growth of a broadrange of species using a few simplified assumptions regardingplant growth and easily derived species-specific parameter values.We examine the assumptions within the Conductance model thattotal leaf area per plant is proportional to total plant weight,and that an isolated plant has a projected crown zone area thatis proportional to the 2/3 power of its weight. Power ratherthan linear relations were found between weight and leaf areafor Brassica oleracea, Daucus carota, Matricaria inodora, Solanumnigrum,Stellaria media , Trifolium repens and Veronica persica.For all seven species, the value of the power was less thanunity. All species also exhibited a power relation between crownzone area and weight, with the slope of this relation beingless than 2/3 for B. oleracea, D. carota and S. media. Althoughmorphology type accounted for some of the variation in the parametervalues relating to light interception, there were considerabledifferences between species within upright or prostrate foliagespecies groups. The Conductance model was used to predict yieldsof B. oleracea, S. nigrum and V. persica grown in both monocultureand binary weed-crop mixtures over a range of temporal and spatialscales. After calibrating the model to non-competing plants,the model was used to predict growth of the weed and crop speciesin contrasting densities and stand types. In some crop-weedcombinations, predicted crop and weed weights were within 17%of observed values, with no systematic deviations. To clarify the way in which the light available for growth affectsrespiration in leaves of sun and shade plants, we examined therespiratory properties of mature leaves of Spinacia oleraceaL., a sun species, and of Alocasia macrorrhiza (L.) G. Don.,a shade species, that had been grown at various irradiances.In leaves of S. oleracea, the respiratory rates, on a dry massbasis, decreased with time during the night, and the higherwas the growth irradiance during the day, the higher was therespiratory rate. The marked decreases in the respiratory rateduring the night were accompanied by decreases in the concentrationof carbohydrates in the leaves. By contrast, the respiratoryrates of leaves of A. macrorrhiza were virtually constant throughoutthe night and the absolute rates were lower than those of S.oleracea even though the absolute value of the concentrationof carbohydrates and its decrease at night resembled to thosein S. oleracea. The maximum activities of respiratory enzymeswere also similar to those in S. oleracea. However, the leavesof A. macrorrhiza contained less soluble protein than thoseof S. oleracea. These results suggest that, in S. oleracea,the concentration of carbohydrates might determine the respiratoryrate while such is not the case in A. macrorrhiza. Physiol. 82(2): 667-677, 1997.The purpose of this study was to examine extensions of a modelof hypoxic ventilatory decline (HVD) in humans. In the original model (model I) devised by R. Painter, S. Khamnei, and P. Robbins(J. Appl. Physiol. 74: 2007-2015, 1993), HVD is modeledentirely by a modulation of peripheral chemoreflex sensitivity. In thefirst extension (model II), a more complicated dynamic is usedfor the change in peripheral chemoreflex sensitivity. In the secondextension (model III), HVD is modeled as a combination ofboth the mechanism of Painter et al. and a component that isindependent of peripheral chemoreflex sensitivity. In all cases, aparallel noise structure was incorporated to describe the stochasticproperties of the ventilatory behavior to remove the correlation of theresiduals. Data came from six subjects from a study by D. A. Bascom, J. J. Pandit, I. D. Clement, and P. A. Robbins (Respir. Physiol.88: 299-312, 1992). For model II, there was a significantimprovement in fit for two out of six subjects. The reasons for thiswere not entirely clear. For model III, the fit was againsignificantly improved in two subjects, but in this case the subjectswere those who had the most marked undershoot and recovery ofventilation at the relief of hypoxia. In these two subjects, thechemoreflex-independent component contributed ~50% to total HVD.In the other four subjects, the chemoreflex-independent componentcontributed ~10% to total HVD. It is concluded that in somesubjects, but not in others, there may be a component of HVD thatis independent of peripheral chemoreflex sensitivity.

It was shown in the previous paper of this series that the Mexicanspecies Solanum bulbocastanum, S. cardiophyllum subsp. cardiophyllumand subsp. ehrenbergii, S. michoacanum and S. pinnatisectummay be differentiated to some extent by the double diffusiontechnique, and more fully by immuno-electrophoretic analysis.Above all, the relative electrophoretic mobilities of certaincomponents, as expressed by the index have been found to be very constant for each species, with the apparent exception of S. bulbocastanum. A detailed investigation of S. bulbocastanum has now shown thatalthough the many clones of this species appeared similar whenexamined by double diffusion, differences were exposed by immuno-electrophoreticanalysis. In particular, the values fell into two discrete groups, 0.15 to 0.23 and 0.30 to 0.46. Theplants in these groups had been collected from Oaxaca and Guatemala,and from Central Mexico, respectively. Most of the former grouphad been identified as S. bulbocastanum subsp. partitum, andall the latter group as subsp. bulbocastanum and subsp. dolichophyllum.Not only was the wide variation in values of index resolved into two meaningful groups, but it was also found thatall lines within any one collection had almost identical values. The index value for S. cardiophyllum subsp.cardiophyllum is 1.0, and for subsp. ehrenbergii is 0.7. The F1 hybrids between either ofthese subspecies and S. bulbocastanum had fairly uniform valuesclose to the calculated means of the parental values. The shapesof the leaves of these hybrids were also uniform and more orless intermediate between those of the parents in each case. The F2 hybrids between S. bulbocastanum and S. pinnatisectum,and also the triple hybrids between these two species and eitherS. cardiophyllum or S. michoacanum, showed a wide range of valuesof index and of leaf shapes, with partial segregation towards the parental types. The serological andmorphological characters segregated independently. S. X sambucinum was found to have a wide variation in serologicaland morphological characters. This variation was consistentwith the theory that S. X sambucinum is a natural hybrid betweenS. pinnatisectum and S. cardiophyllum subsp. ehrenbergii. Within the groups of species investigated, the relative electrophoreticmobilities of certain antigens as displayed by immuno-electrophoreticanalysis were found to vary significantly in different species,in different subspecies of the same species, and in specieshybrids.  相似文献

We found that the gradient of a host-specific attractant, cochliophilin A (5-hydroxy-6,7-methylenedioxyflavone) isolated from the roots of spinach triggered encystment followed by germination of zoospores of Aphanomyces cochlioidesat a concentration less than micromolar order. This compound did not affect the growth and reproduction of this phytopathogen up to 10–6 M concentration in the culture medium. We also observed that mastoparan, an activator of heterotrimeric G-protein could inhibit the motility of zoospores and then strikingly effect encystment followed by 60–80% germination of cysts. Concomitant application of cochliophilin A and mastoparan showed stronger encystment followed by 100% germination of cysts. In addition, we have observed that chemicals interfering with phospholipase C activity (neomycin) and Ca2+ influx/release (EGTA and loperamide) suppress cochliophilin A or mastoparan induced encystment and germination. These results suggest that G-protein mediated signal transduction mechanism may be involved in the differentiation of the A. cochlioides zoospores. This is the first report on the differentiation of oomycete zoospores initiated by a host-specific plant signal or a G-protein activator.  相似文献

I tested several hypotheses regarding the antipredator behaviorsof ground skinks (Scincella lateralis) based on a simple-optimalitymodel for avoiding predation. Field studies demonstrated thatS. lateralis with lower body temperatures fled earlier in predatoryevents when the predator was farther away than did warmer animal.This may reflect a compensatory shift in behavior related todecreased sprint speeds observed at lower temperatures. Thesedifferences in maximum sprint speeds at five substrate temperatureswere not linked to morphological characteristics such as mass,snout-vent length, or tail length. Body temperatures of S. lateraliswere highly correlated with substrate temperatures in the field,and these environmental temperatures correlated with the dailyand seasonal activity schedules of S. lateralis. The thermoregulatorystrategy of ground skinks closely parallels theoretical predictionsfor forest-dwelling lizards based on optimum performance curves.In the laboratory, S. lateralis did not alter flight responsesbased on the availability of cover objects or tail conditionof the lizards. Escape behaviors of males and females did notdiffer. Likewise, S. lateralis. showed no differences in responseto differently sized predator models. Differences were foundbetween the reactions of adult and juvenile skinks to an approachingpredator model. The germination behaviour of S. nigrum L. and S. physalifoliumRusby var. nitidibacatum (Bitter) Edmonds is compared, basedon temperature requirements during imbibition. Three seed lotsof S. nigrum had base temperatures (Tb) between 7.5C and 10C,showing a lower Tb when the period of freezing days, duringwhich each population was collected, was reduced. S. physalifoliumhas a higher value for Tb at constant temperatures (21C) thatcan be interpreted as a dormancy constraint. This constraintis released by alternating temperatures at amplitudes exceeding5C and with the high temperature above 21C by apparently reducingTb to 12.5C. This implies that for S. physaiifolium temperature has a dualeffect on germination. It is the driving force for changes indormancy, but germination also depends on the temperature. Therole of temperature for S. nigrum is simpler: each populationcollected showed differences in the thermal time required forgermination that could be related to the temperature regimenof the original environment. Bot. 38: 433–441. S. anglica is one of the few C4 species which occurs naturallyin cool temperate zones. It is known to attain photosyntheticrates which equal or exceed those of C3 grasses over the temperaturerange typical of the spring and summer in cool temperate climates.This study examines whether S. anglica can also attain comparablegrowth rates at these temperatures. Seedlings of S. anglicaand L. perenne cv. S23 were grown in controlled environmentsat 10,15,20 and 25 °C. Quantitative growth analysis wasconducted by taking frequent harvests to determine the progressionsof leaf area and plant weight of individual plants with time.Quadratic regressions were found to describe these progressionswell. Instantaneous derived growth parameters were calculatedfrom the fitted regressions. Both absolute and relative growthrates of S. anglica were significantly lower than for L. perenne,this being largely attributable to a lower ratio of leaf areaproduction per unit of plant dry weight. Although the amountof dry matter invested into leaves was similar, the leaf areaper unit of leaf dry weight was lower in S. anglica. In comparisonto L. perenne, the rate of dry matter accumulation per unitof leaf area (ULR) was higher in S. anglica at 25 °C andinitially equal at 10 °C. Prolonged exposure to 10 °Csteadily reduced ULR in S. anglica which approached zero at80 d. Although growth in S. anglica is reduced more by low temperaturethan it is in L. perenne, by comparison to other C4 speciesthe assimilatory capacity of S. anglica is more tolerant oflow temperature exposure. Observations, both experimental and microscopic, indicate thatzoospores of Rhizophydium planktonicum Canter emend., can remainalive, but rarely become adherent upon cells of Asterionellaformosa Hassall under conditions of very low light or in completedarkness. The behaviour patterns of zoospores towards host cells underdarkened conditions were compared with those which took placeunder well illuminated conditions leading to the settlementof zoospores on host cells. The differences noted may help toexplain the lack of zoospores found upon inadequately illuminateddiatom cells. Some evidence suggests that young zoospores lack the abilityto adhere to host cells. After the encystment of zoospores uponAsterionella cells in the light, their further growth and developmentcan continue in darkness.
