Summary Alpha-amylase production byBacillus licheniformis M27 in submerged fermentation was reduced from 480 to 30 units/ml when soluble starch concentration in medium was increased from 0.2 to 1.0%. In contrast, the enzyme production increased by 29 times even with 42 fold increase in the concentration of soluble starch and other starchy substrates in solid state fermentation system. The data establish regulation of the enzyme formation by enzyme end-product in submerged fermentation and ability of solid state fermentation to minimize it significantly. These features were not known earlier.  相似文献

Electrochemical energy as a source of reducing equivalent was applied to the cultures ofClostridum acetobutylicum to understand the effects of the pressure by reducing equivalent on anaerobic bacteria. The fermentation byC. acetobutylicum with methyl viologen and electrochemical energy produced more butanol (up to 26%) than the control culture, whilst less acetone (up to 25%) was produced. But no effect was observed on the growth of the culture. These results were indirectly supported byin vitro electrochemical reduction of NAD+ and artificial electron carriers.  相似文献

Summary For the protein upgrading of sugar-beet pulp in solid state fermentation byTrichoderma reesei andFusarium oxysporum, serveral conditions were studied to prepare an economical preculture for large scale process. Summary Acetone and butanol biosynthesis byClostridium acetobutylicum ATCC 824 was affected by lactose concentration and by agitation during the fermentation. At 1% and 3% lactose concentrations acid production predominated, while butanol production predominated at 5% lactose concentration. Summary The nature and amounts of by-products formed during conversion of sugar beets to ethanol byZ. mobilis in Conventional Submerged Fermentation (CSF) and Solid-State Fermentation (SSF) were investigated. It was found that the bacterium produced fewer by-products in SSF than CSF, and that by-products profile was different. The influence of fermentation temperature on synthesis of by-products in SSF was also studied. High fermentation temperature favoured sorbitol synthesis and low fermentation temperature the synthesis of levan. The best results were obtained at 35°C. An ethanol yield of up to 95% of the theoretical value with final ethanol concentration of 142 g/L were obtained.  相似文献

Summary The maximal concentration of ethanol produced during the fermentation of 320 g/l glucose bySaccharomyces bayanus was higher when the yeast cells were immobilized either by adsorption on celite or by entrapment in k-carrageenan beads (from 10.5% with free cells up to 14.5% and 13.1% (v/v) respectively). Summary During xylose fermentation byCandida shehatae ATCC 22984 with batch cell recycling, the volumetric ethanol fermentation rate increased two-fold, and the xylitol production rate increased three-fold as the cell density increased to ten-fold. In continuous fermentation with membrane-assisted cell recycle, the fermentation rates increased almost linearly with increasing agitation rates up to 300 rpm. The maximum continuous ethanol production rates obtained with 90 and 200 g L–1 xylose were respectively 2.4 and 4.4 g L–1h–1. The cell density was 65–70 g (dry wt) L–1. Ethanol yields ranged from 0.26 to 0.41 g g–1.  相似文献

Summary Xylose, the predominant sugar in red oak prehydrolysate, is fermented to ethanol byPichia stipitis CBS 5776. Toxic model compounds derived from red oak hemicelluloses, lignin, and extractives inhibited the fermentation. Treatment of the prehydrolysate with molecular sieve and mixed bed ion resins facilitated the ethanol fermentation giving about 10 g/l ethanol from 32 g/l initial xylose. Summary The effect of temperature on the solvent production byClostridium acetobutylicum has been studied in the range 25 to 40°C. It was found that the solvent yield decreased with increasing temperature; seemingly because of a reduction in acetone production. It appeared that the yield of the other major solvent, butanol, was not affected by the temperature. Considering total solvent yield and productivity only, the optimum fermentation temperature is 35°C.  相似文献

A resting cell system was used for the production of glycolipids byPseudomonas aeruginosa CFTR-6. In this, the growth phase was separated from the production phase to overcome the inhibition of glycolipid production by inorganic phosphate. It was shown that when the cells were transferred after the growth phase into a medium devoid of phosphate, glycolipid production was increased nearly twofold. The maximum glycolipid concentration was attained much more rapidly than the conventional batch fermentation system, thus increasing the productivity.  相似文献

Summary The fermentation of D-xylose byPachysolen tannophilus Y2460,Pichia stipitis Y7124,Kluyveromyces marxianus Y2415 andCandida shehatae Y12878 was investigated in aerobic, anaerobic and microaerophilic batch cultures. The aeration rate greatly influenced the fermentations; growth, rate of ethanol production and oxidation of ethanol are affected. Of the strains tested,Pichia stipitis appears superior; under anaerobic conditions it converts D-xylose (20 g/l) to ethanol with a yield of 0.40 g/l and it exhibits the highest ethanol specific productivity (3.5 g of ethanol per g dry cell per day) under microaerophilic conditions.  相似文献

Summary The influence of temperature, agitation speed, pH and biomass on the synthesis of 19 metabolites contributing to a strawberry aroma was followed over a 72 h fermentation of skim milk byPseudomonas fragi. Amongst the major odor-active metabolites were ethyl butyrate, ethyl hexanoate, ethyl 2-hexenoate, ethyl crotonate, ethyl isovalerate and ethyl 2-methyl hexanoate. Up to 17 ppm of some of these metabolites were detected at 60 h of fermentation, approximately 36 h after the beginning of the stationary growth phase. The production of most odor-active metabolites was higher at 11°C and 150 RPM than at 15, 20 or 25°C and 200 or 250 RPM. The development of off-aromas was observed at high temperatures and at high agitation speeds.  相似文献

Summary Enhanced rates of ethanol production byPachysolen tannophilus from D-xylose were obtained by performing the fermentation with recycled cells in suspension culture or immobilized in a Ca-alginate gel. Fermentation under these conditions did not require aeration. Summary Growth and ethanol production byPichia stipitis in media containing different nitrogen sources were compared. Ammonium sulphate (plus vitamins and trace elements) increased ethanol productivity 57% and ethanol-biomass yield 113%, compared to yeast extract. Consumption of ammoniu, amino-acids and total nitrogen was determined. Two ammonium effects, low uptake in complex media and fermentation stimulation in defined media are discussed in terms of ammonium- ammonia futile cycling and maintenance energy.  相似文献

Summary Quantitative differentiation of microbial activity and saprophytic colonisation in wheat straw substrate during SSF is described. Wheat straw contains an active indigenous microbial flora. In both untreated and heat pretreated substrates a peak of microbial respiratory activities occurs generally after 24 h of fermentation; addition of actidione and streptomycin has no marked inhibitory effect. Quantitative differences between endogenous microbial respiration and respiration byPleurotus during the initial four days of SSF are least in the heat pretreated substrates and greater when microbiological pretreatment is used. In treatments subjected to 48 h anaerobic fermentation at 50°C, saprophytic colonization similar to that observed in sterile substrates is obtained.  相似文献
