When investigating resistance to herbivorous insects in transgenic plants, it is primarily the damaging stage of the insect that is targeted. In our study, we showed that small effects of the transgenic plant on the pest performance might lead to a clear effect on the pest population level. Pollen beetle, Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae), larvae were reared on three transgenic lines of Brassica napus L. (Brassicaceae) expressing pea (Pisum sativum L.) lectin at 0.2, 0.6, and 0.7% of total soluble protein in anthers, and then monitored until the adult stage. Two lines without the gene construct and consequently containing no pea lectin were used as controls. The egg viability, larval weight, development time, and survival rate were recorded. In a separate experiment, the development time from egg to adult, adult weight, and winter survival of pollen beetles developed on intact plants were recorded. Survival of larvae to adult stage was significantly lower on the transgenic plant lines, independent of lectin level. Survival during hibernation was only 2.4% and was not affected by plant line. Combining previously published results from adult feeding and oviposition assays with the new data on larval development we summarize the effect of the transgenic oilseed rape on the pollen beetle, from oviposition in spring to overwintered adults of the new generation. Net reproductive rate was then shown to be reduced by half on the transgenic oilseed rape compared to the control plants.  相似文献

Pollen beetles, Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae), are major pests in oilseed rape (OSR), Brassica napus L. (Brassicaceae). Among the predator species in the generalist predator complex present in OSR fields, wolf spiders (Araneae: Lycosidae) are found on the ground and cobweb spiders (Araneae: Theridiidae) build webs in the foliage. Here we study the incidence of predation of pollen beetles by these two spider groups using DNA‐based molecular analysis. Wolf spiders of the genus Pardosa and the cobweb spider, Theridion impressum L. Koch, were each collected in three winter OSR fields over a period of about 3 weeks. Pollen beetle densities as well as the occurrence of predators and alternative prey were monitored. In total, 13.8% of the collected Pardosa spp. tested positive for pollen beetle DNA in the PCR analyses, whereas 51.7%T. impressum were positive. The likelihood of detecting pollen beetle DNA in the gut contents of both spider groups was positively related to pollen beetle larval density. The implications of these results for conservation biological control and future studies of food webs in OSR are discussed.  相似文献

Pollen beetles (Meligethes aeneus) and cabbage seed weevils (Ceutorhynchus assimilis) are major pests of oilseed rape (Brassica napus) throughout Europe. In field cage experiments in both winter and spring rape, honey bees (Apis mellifera) effectively transported the entomopathogenic fungus Metarhizium anisopliae to the flowers, causing infection and mortality of both adult and larval pollen beetles, as well as of adult seed weevils. External conidiation was observed on many of the dead pest insects. Although some external conidiation also occurred on dead honey bees, reduction in honey bee colony size during the experiments appeared unrelated to the fungus. The potential of this technique for integration into pest management strategies for the crop, particularly in association with a trap crop, is discussed.  相似文献

Abstract. The behavioural response of adults of the pollen beetle, Meligethes aeneus F. (Coleoptera: Nitidulidae), to the odour of one of their host-plants, oilseed rape ( Brassica napus L.), was observed in the laboratory using a fourarmed airflow olfactometer, and under semi-natural conditions using a markrelease–recapture technique. Oilseed rape leaf and flower odour was attractive to pollen beetles in the olfactometer and they dispersed upwind from the centre of a circular array (radius 20 m) of yellow water traps, baited with extracts of oilseed rape leaves or flowers. The dispersal of pollen beetles in the absence of oilseed rape odour was predominantly downwind and crosswind. The percentage recapture of pollen beetles was significantly greater in traps baited with rape flower extracts than leaf extracts. Pollen beetles can use odour-mediated upwind anemotaxis to locate oilseed rape plants from distances of at least 20 m over open ground.  相似文献

Abstract.  1. Pollen is considered to be an important dietary component for many species of flower-feeding herbivores. Its influence on oviposition site selection by the pollen beetle Meligethes aeneus , and on the development of its larvae was investigated.
2. The effects of pollen presence and absence on adult, egg, and larval incidence in the field, and on larval development in the laboratory were compared through the use of Synergy, a composite hybrid oilseed rape Brassica napus variety comprising male-fertile (with pollen) and male-sterile (without pollen) plants.
3. In the field, adult females were more abundant on male-fertile plants during flowering, and a greater proportion of male-fertile than male-sterile buds were accepted for oviposition. These data indicate a possible role of pollen in oviposition site selection by female pollen beetles.
4. The numbers of first instar larvae on the two plant lines did not differ; however, more second instars were found on male-fertile than on male-sterile flowers. This suggests a greater larval survival on male-fertile plants, possibly due to the more readily available food resources and better nutrition afforded by the presence of pollen.
5. Laboratory experiments confirmed that a diet which included pollen improved survival to adulthood and resulted in heavier pupae and adults; however, pollen was not obligatory for larval survival and development.
6. The pollen beetle, previously thought to be an obligate pollen feeder, is therefore more generalist in its requirements for development. These findings may relate to the nutritional and behavioural ecology of other flower-feeding herbivores.  相似文献

The response of the pollen beetle Meligethes aeneus Fab. (Coleoptera, Nitidulidae) to the volatiles of undamaged plants and conspecifics was tested in a Y-tube-olfactometer-bioassay. Beetles that had hibernated preferred significantly the volatiles emitted by their most important host plant, oilseed-rape (Brassica napus L. ‘Lorar’) in the early bud-stage. However, the odour emitted by rye (Secale cereale L.), tomato plants (Solanum lycopersicum L.), and yarrow (Achillea millefolium L.) were also attractive when tested against the corresponding growing-medium. Dock plants (Rumex obtusifolius L.) and touch-me-not (Impatiens parviflora L.) possessed no attractive properties. When tested against each other, oilseed-rape was preferred significantly by M. aeneus above all other plants, with the exception of tomato. The results indicate that M. aeneus is able to locate its host plant by olfactory stimuli in the early bud-stage, i.e. in that stage in which the infestation begins in the field and when the typical yellow colour and floral scent of oilseed-rape are absent. Female pollen beetles avoided significantly the volatiles emitted by female conspecifics and an ether extract of conspecifics of mixed sex, while volatiles from males had no significant effect on females. Furthermore, males showed no preferences when responding to conspecific odour in the olfactometer. These results suggest the existence of an epideictic pheromone for M. aeneus.  相似文献

Proteinase inhibitors (PIs), lectin and patatin purified from potato tubers were tested in no‐choice feeding assays with pollen beetle larvae (Meligethes spp.). The idea was to search for resistance factors possible to introduce into oilseed rape (Brassica napus) by genetic engineering. The larval diet was prepared by soaking oilseed rape anthers in protein solutions of known concentrations. Potato lectin was the most potent in that it was the only of these proteins that reduced both larval survival and growth rate, while cysteine, aspartic and metallo PIs and patatin only reduced larval growth rate. Serine PIs had no significant effect on larval performance. Subsequently, the effect of potato lectin was compared to that of lectins from other food or feed crops, resulting in the following mortality‐ based ranking of activity: Con A from jackbean > wheat germ lectin > potato lectin > peanut lectin. In choice tests, larvae did not discriminate between Con A‐ and control‐diets. These results suggest that the effect of Con A on larvae is toxic, not deterrent. Adult response was stronger to Con A than to potato lectin in no‐choice tests, just as it was in larvae. However, adult survival rate and weight was not affected by Con A but the lectin significantly reduced adult feeding as well as oviposition rates. A resistance factor that suppresses adult feeding on flower buds is important for reduced impact of the pollen beetle on the Brassica oilseed crop.  相似文献

Pollen beetles (Meligethes spp.; Coleoptera: Nitiduliae) are a major pest of oilseed rape, Brassica napus L. (Brassicaceae) in northern Europe. Phradis interstitialis Thomson, P. morionellus Holmgr., and Tersilochus heterocerus Thomson (Hymenoptera: Ichneumonidae) are among the most frequent pollen beetle parasitoids. These three species differ in temporal occurrence, as well as in preferred host stage. The behavioural responses of female parasitoids to odours from oilseed rape at bud and flowering stage were evaluated in two‐choice experiments. The role of visual stimuli was examined by combining green and yellow colours with odour stimuli. All three species were attracted to odours from the bud stage of oilseed rape. Tersilochus heterocerus was attracted to odours of flowering rape, but the two Phradis species avoided the flower odours. However, when the odours of flowering rape were combined with yellow, and odours of the bud stage were combined with green, P. interstitialis was equally attracted to both stimuli, and T. heterocerus showed an increased preference for flower odours, while no effect of colours could be found in P. morionellus. The observed differences in responses between the parasitoids may reflect differences in their biology and may be involved in the niche segregation of these often coexisting species. The volatile blends released from the two phenological stages were identified and compared. Clearly, odours can be reliable cues for differentiating between oilseed rape in the bud and flowering stage. Of 20 identified compounds, 18 were released at a significantly higher rate from flowering plants. The terpenes sabinene, myrcene, limonene, and (E,E)‐α‐farnesene were the dominant volatiles in the bud and flower headspace. A group of aromatic compounds including benzaldehyde, methyl benzoate, and phenyl acetaldehyde were mainly released from flowering rape.  相似文献

1 The spatio‐temporal distributions of predatory carabid beetles (Coleoptera: Carabidae) and their potential prey, the larvae of three coleopterous pests, Meligethes aeneus (Fabricius) and Ceutorhynchus spp. [Ceutorhynchus pallidactylus (Marsham), the cabbage stem weevil, and Ceutorhynchus assimilis (Paykull), the cabbage seed weevil], were studied within a crop of winter oilseed rape. The distributions of Collembola were recorded as potential alternative prey. Insect distributions were analysed and compared using Spatial Analysis by Distance Indices. 2 Mature larvae of the pests dropped from the crop canopy to the soil for pupation in temporal succession from May to early July. Their distributions within the crop were irregular and differed with species. 3 Adults of seven species or genera of carabid were abundant and active within the crop during May and June: Nebria brevicollis (Fabricius), Anchomenus dorsalis (Pontoppidan), Loricera pilicornis (Fabricius), Amara similata (Gyllenhal), Asaphidion spp., Pterostichus madidus (Fabricius) and Pterostichus melanarius (Illiger). 4 During May, N. brevicollis was spatially associated with peak numbers of M. aeneus larvae and with Collembola. Anchomenus dorsalis was spatially associated with Ceutorhynchus spp. larvae during two peaks in the abundance of the latter in early and late June. Nebria brevicollis and A. dorsalis coincided in both time and space with larvae of the three coleopterous pests when they were most vulnerable to predation by epigeal predators and are therefore good candidates for conservation biocontrol. 5 The importance of carabid beeetles in the natural enemy complex in winter oilseed rape and their potential for biocontrol of spring and summer pests are discussed in relation to husbandry practices for the crop and its adjacent areas which could be manipulated to promote carabid survival for integrated pest management.  相似文献

New control strategies for insect pests of arable agriculture are needed to reduce current dependence on synthetic insecticides, the use of which is unsustainable. We investigated the potential of a simple control strategy to protect spring‐sown oilseed rape, Brassica napus L. (Brassicaceae), from two major inflorescence pests: the pollen beetle, Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae), and the seed weevil, Ceutorhynchus assimilis (Paykull) (Coleoptera: Curculionidae), through exploitation of their host plant preferences. The strategy comprised, for the main crop, Starlight [an oilseed rape cultivar with relatively low proportions of alkenyl glucosinolates in the leaves (thereby releasing lower levels of attractive isothiocyanates than conventional cultivars)] and turnip rape, Brassica rapa (L.) (Brassicaceae), as a trap crop. We tested the system in laboratory, polytunnel semifield arena, and field experiments. The odours of Starlight were less attractive in olfactometer tests to both pests than those from a conventional cultivar, Canyon, and the plants were less heavily colonized in both polytunnel and field experiments. Turnip rape showed good potential as a trap crop for oilseed rape pests, particularly the pollen beetle as its odour was more attractive to both pests than that of oilseed rape. Polytunnel and field experiments showed the importance of relative growth stage in the system. As turnip rape flowers earlier than oilseed rape, beetles would be maintained on turnip rape past the damage‐susceptible growth stage of oilseed rape. The development of a pest control regime based on this strategy is discussed.  相似文献

The use of semiochemicals for the manipulation of the pollen beetle, Meliethes aeneus (Fabricius) (Coleoptera: Nitidulidae), is being investigated for potential incorporation into a push‐pull strategy for this pest, which damages oilseed rape, Brassica napus L. (Brassicaceae), throughout Europe. Previous laboratory behavioural studies using volatiles from non‐host plants showed that M. aeneus is repelled by the odour of lavender, Lavendula angustifolia Mill. (Lamiaceae), essential oil. This article reports on semi‐field and field trials to investigate this behaviour under more realistic conditions. Semi‐field experiments were conducted to assess the relative importance of olfaction at different points in host location behaviour by M. aeneus. The results showed that oilseed rape plants treated with lavender odour were less colonised by M. aeneus in comparison with an untreated control, but that the treatment effect was much reduced if the lavender odour was applied after colonisation. The field experiment demonstrated that lavender odour caused a significant reduction in the number of adult M. aeneus infesting the oilseed rape plants in the treatment plots compared to the control plots. Overall, these findings are very encouraging for the future development of a push‐pull pest control system.  相似文献

The spatio‐temporal distribution of Psylliodes chrysocephala (L.) (Coleoptera: Chrysomelidae), a pest of oilseed rape (Brassica napus) (L.) (Cruciferae) and its potential predators, carabid beetles, within a crop of winter oilseed rape is described. The distribution of Collembola, a potential alternative food source for the predators, is also investigated. Insects were collected from spatially referenced sampling points across the crop and the counts mapped, analysed, and the degree of spatial association between the distributions determined using Spatial Analysis by Distance IndicEs (SADIE). Immigration into the crop by adult P. chrysocephala occurred from two edges and resulted in a non‐uniform distribution of the pest within the crop. Infestation of rape plants by P. chrysocephala larvae was greatest within the central area of the crop. Significant spatial association between adult female P. chrysocephala and the larval infestation of plants occurred throughout October. Three carabid species were active and abundant during peak pest immigration into the crop, viz., Trechus quadristriatus (Schrank) (Coleoptera: Carabidae), Pterostichus madidus (Fabricius) (Coleoptera: Carabidae), and Nebria brevicollis (Fabricius) (Coleoptera: Carabidae). Two of these species, T. quadristriatus and P. madidus, showed significant spatial association with the larvae of P. chrysocephala during October. All three carabid species showed a significant spatial association with Collembola during mid‐September, indicating that the latter may be an important food source for carabids during this period. In laboratory feeding experiments, only T. quadristriatus consumed the eggs of P. chrysocephala suggesting that, in the adult stage, this species may be the most important of the naturally occurring carabids as a predator of P. chrysocephala in the field. Adult T. quadristriatus may be a valuable component of an Integrated Pest Management strategy for winter oilseed rape, and the conservation of this species could be beneficial.  相似文献

The use of semiochemicals for manipulation of the pollen beetle Meligethes aeneus (Fabricius) (Coleoptera: Nitidulidae) is being investigated for potential incorporation into a push‐pull control strategy for this pest, which damages oilseed rape, Brassica napus L. (Brassicaceae), throughout Europe. The response of M. aeneus to non‐host plant volatiles was investigated in laboratory assays to establish whether they have any effect on host plant location behaviour. Two approaches were used. First a novel, moving‐air bioassay using air funnels was developed to compare the response of M. aeneus to several non‐host plant essential oils. The beetles avoided the host plant flowers in the presence of non‐host volatiles, suggesting that M. aeneus uses olfactory cues in host location and/or acceptance. The results were expressed as ‘repellency values’ in order to compare the effects of the different oils tested. Lavender (Lavendula angustifolia Miller) (Lamiaceae) essential oil gave the highest repellency value. In addition, a four‐arm olfactometer was used to investigate olfactory responses, as this technique eliminated the influence of host plant visual and contact cues. The attraction to host plant volatiles was reduced by the addition of non‐host plant volatiles, but in addition to masking the host plant volatiles, the non‐host volatiles were avoided when these were presented alone. This is encouraging for the potential use of non‐host plants within a push‐pull strategy to reduce the pest colonisation of crops. Further testing in more realistic semi‐field and field trials is underway.  相似文献

1 Host‐plant‐odour‐induced upwind anemotaxis is accepted as the main mechanism by which herbivorous insects find their host plant within an heterogenous environment, but field data supporting this hypothesis are scarce. 2 The flight directions of the pollen beetle Meligethes aeneus to and from a plot of winter oilseed rape and the direction of the wind were recorded concurrently. The beetles were sampled by eight double‐sided window traps encircling the plot. Distal sides of the traps, relative to the plot, sampled the beetles as they flew towards the plot, whereas the proximal sides of the traps sampled them as they flew away from the plot. Paired data on daily catch of beetles in distal or proximal traps and the volume of air impacting each trap were compared. 3 Correlations between daily catch of M. aeneus into distal traps and trap wind volume were negative, indicating that flights by both overwintered‐ and new‐generation insects towards the plot were upwind. 4 Correlations between daily catch of M. aeneus into proximal traps and trap wind volume varied with generation. Catch of overwintered‐generation M. aeneus was negatively correlated with trap wind volume, indicating that flight was upwind. Proximal trap catch of the new‐generation M. aeneus was weakly/modestly positively correlated with trap wind volume, indicating that flights were downwind or crosswind. 5 Understanding the effect of wind direction on flight of M. aeneus holds potential for improving the forecasting of their arrival and spatial distribution on the crop for integrated pest management.  相似文献

The diamondback moth (DBM), Plutella xylostella (L.) (Lep., Plutellidae), is one of the most important pests of oilseed rape, Brassica napus L., in the world. In this study, resistance of oilseed rape cultivars to DBM was evaluated in fields of Tehran based on injury rate of the cultivars. To evaluate cultivar resistance, 19 cultivars of oilseed rape were planted in randomised complete-block design with three replications. Total experiments were conducted in experimental field of Shahed University. Results showed that DBM had activity on the plants during season and peak of the injury occurred on 27 May. Results indicated that the total of oilseed rape cultivar was spotty. Statistical analysis showed that there was significant difference among determined characteristics in all cultivars. The highest and lowest numbers of holes in the leaf were observed in Midas, Karaj3, Zarfam and Hyola420, Hyola308 cultivars, respectively, in sampling of the end growth. Opera and Midas cultivars have the highest percentage of leaf infestation and the lowest percentage of leaf infestation was observed in Hyola420 and Hyola308. Also, Karaj3 and Zarfam cultivars have the highest percentage of meristem infestation and the lowest percentage of meristem infestation was observed in Licard and Hyola420 cultivars. Based on peak of the injury rate and importance of meristem injury, Hyola420 and Hyola308 cultivars were relatively resistant to DBM, and Karaj3 and Zarfam were relatively sensitive to DBM.  相似文献

对海南省儋州地区荔枝园中荔枝蝽Tessaratoma papillosa(Drury)种群动态及空间分布进行了系统调查与分析。结果表明,在2005—2006与2008—2009年期间,荔枝蝽的产卵高峰期分别为4月与2月,若虫高峰期为3月,成虫高峰期分别为4月及7月;分析表明荔枝蝽各虫态在荔枝园中均为聚集分布,且成虫个体间相互排斥。  相似文献

The spatio-temporal distribution of Ceutorhynchus assimilis Payk. (Coleoptera: Curculionidae) adults caught in a rectangular grid of flight traps in a crop of winter oilseed rape (Brassica napus L.) was mapped and was analysed using Spatial Analysis by Distance IndicEs (SADIE). Their distribution was compared to that of their larvae and that of their parasitoid Trichomalus perfectus (Walker) (Hymenoptera: Pteromalidae) in pods. The distribution of immigrating C. assimilis adults was consistent with their arrival at the crop boundaries and movement within the crop towards its centre. Adult C. assimilis were aggregated at all times, invasion being on two fronts, leading to the formation of two major clusters within the crop. Large areas of the crop remained relatively unpopulated. During the emigration phase, numbers declined simultaneously in all parts of the crop. The distributions of adult and larval C. assimilis and of larval T. perfectus were spatially associated. The distribution of the parasitoid did not show a density dependent relationship with that of its host. We discuss the movements of insects which underlie their population distributions, the value of integrating spatial information into improved management strategies for C. assimilis and the potential for the spatial targeting of insecticides to reduce the amount applied and to conserve T. perfectus.  相似文献

2007-2009年对黑龙江大豆蚜田间种群动态的调查结果表明,6月中旬大豆蚜Aphis glycines Matsumura开始在田间出现,3~5周后田间有蚜株率达到100%。7月未至8月初蚜量达到高峰期,9月中下旬在田间逐渐消失。大豆蚜有翅蚜与无翅蚜的发生动态基本一致,有翅蚜高峰期有时会稍有提前。2007年大豆蚜发生时期早、基数大、增长周期长导致高峰期蚜量显著高于2008年和2009年。大豆蚜田间分布聚集度指标I﹥0、m*/m﹥1、Ca﹥0、C﹥1,整个生长季种群都表现为聚集分布。建立的Iwao回归关系方程(m*=7.14758+1.11200m)表明种群分布的基本成分为个体群,个体之间表现为相互吸引。  相似文献
