Electron microscopy revealed that Gene's organ in females of Argas walkerae Kaiser & Hoogstraal (Ixodida: Argasidae) is formed as a double-sac structure consisting of an outer epithelial and an inner cuticular sac. The latter emerges through the camerostomal aperture to the exterior in ovipositing ticks. The epithelial sac forms the corpus and the two blind-ending horns, which pass into the epithelium of the excretory duct of a gland at each side of Gene's organ and envelop the cuticular sac. Both excretory ducts open into the lumen between the epithelial and the cuticular sac. The cuticular sac is folded and consists of a fibrous endocuticula outwards towards the lumen between the epithelial and the cuticular sac and of a smooth epicuticula inwards. Parallel running grooves occur over the lateral epicuticular surface turning medially into cobble-stone pavement-like rises. Tubuli pass through the cuticular sac ending in pores on the epicuticular surface and open into the lumen between the epithelial and the cuticular sac. Muscle fibres pass through the epithelial sac at the horn tips and are inserted to the cuticular sac. In ovipositing females, the glands are fully developed and the lumen between the epithelial and the cuticular sac is filled with an amorphous mass.  相似文献

Scanning and transmission electron microscopical investigations revealed that Gené's organ in unfed and ovipositing females of Dermacentor reticulatus is formed as a double-sac-structure consisting of an outer epithelial and an inner cuticular sac. In ovipositing ticks the latter emerges through the camerostomal aperture to the exterior. Gené's organ in unfed ticks consists of a corpus, two posterior horns and a pair of undeveloped glands at each side, which differentiate in ovipositing ticks to compound, branched tubular glands with a main efferent duct for each gland opening into the lumen between the epithelial and the cuticular sac. Gené's organ of egg-laying females corresponds basically in morphology and structural organization to that of unfed ticks. Compared with unfed ticks, however, in ovipositing ticks the corpus and horns are longer and broader, the glands are fully developed and the cuticular sac is evertable. The epithelial sac as the outermost part of Gené's organ is continuous with the hypodermis of the basis capituli and the scutum, arises at the camerostomal aperture, forms the corpus and the two blind-ending horns, passes into the epithelium of the main excretory ducts of the glands and envelops the cuticular sac. The cuticular sac passes into the cuticle of the basis capituli and the scutum, arises at the camerostomal aperture, is folded, expands into the horn tips and consists inwards of a smooth epicuticula and outwards of a fibrous endocuticula. Muscles originating from the scutum pass caudomedially through the epithelial sac and are inserted into the cuticular sac. The entire surface of the maximally everted cuticular sac is covered with an amorphous mass. In cleaned samples, ledge-like structures appear on the lateral surface. These ledges turn into balloon-like structures which extend over the medial and dorsal surface. The entire surface including the balloon-like structures and the ledges are provided with numerous cribrate pits.  相似文献

The female reproductive system in Pergamasus mites consists of an unpaired vagina, vaginal duct, uterus, and ovary. Additionally, there are paired vaginal glands, as well as unpaired ventral and paired lateromedial glandular complexes. The vagina and vaginal duct are cuticle‐lined. In the dorsal wall of the vagina, this lining forms the endogynium which possesses a “sac” and two conspicuous “spherules” and is armed with “stipula” and other cuticular protrusions. The endogynium functions as a spermatheca, being a storing site for the spermatophore. The spherule procuticle is perforated by microvilli of underlying cells that are structurally very unusual. The lining of the vaginal duct forms numerous cuticular fibers directed toward the vagina. There is an external layer of muscles, supposedly functioning as a sphincter. The uterus is an organ in which the fertilized egg is stored for some time and starts embryonic development. Its wall is composed of glandular epithelial cells. The ovary consists of inner and outer parts. The former part is formed by a nutritive syncytium, whereas the latter contains growing oocytes. Two groups of glands connect with the genital tract. Paired vaginal glands are composed of glandular and secretion‐storing parts and open into the vagina. Paired lateromedial and unpaired ventral glandular complexes empty into the genital tract between the vaginal duct and uterus. The cement gland of Rhodnius prolixus is an epidermally derived tubular gland consisting of a distal synthetic region and a proximal muscular duct region. The synthetic region consists of numerous secretory units joined to a central chitinous duct via cuticular ductules. Proteinaceous secretion, synthesized by the goblet-shaped secretory cell, passes through the delicate cuticular lattice of a ductule-end apparatus and out through fine ductules to the central duct. Secretory cells are rich in rough endoplasmic reticulum and mitochondria. Light microscopy, SEM and TEM reveal the delicate lattice-like end apparatus structure, its formation and relationship to the secretory cell. The secretory cell associates via septate junctions with a tubular ductule cell that encloses a cuticle-lined ductule by forming an elaborate septate junction with itself. The ductules are continuous with the cuticle lining of the large central duct that conveys secretion to the proximal area. The proximal muscular duct has a corrugated cuticular lining, a thin epithelium rich in microtubules and thick longitudinal, striated muscles which contract during oviposition, forcing the secretion out. Histochemistry and electrophoresis reveal the secretion as proteinaceous.  相似文献

The ejaculatory duct of the migratory grasshopper (Melanoplus sanguinipes [Fabr.]) (Orthoptera : Acrididae) is divisible into 3 regions: upper ejaculatory duct (UED) into whose anterior end the accessory glands and vasa deferentia empty; the funnel characterized by its slit-like lumen; and the lower ejaculatory duct (LED). Anteriorly, the UED has a keyhole-shaped lumen surrounded by a thin intima and highly columnar epithelial cells whose most conspicuous feature is massive aggregations of microtubules. More posteriorly, the UED lumen differentiates into dorsal and ventral chambers, the former having a thick cuticular lining armed with spines. In the hindmost part of the UED, the ventral chamber expands to obliterate the dorsal chamber; its cuticular lining thickens, and conspicuous lateral evaginations develop. The thick cuticle includes 3 distinct layers and on its surface carries numerous spatulate processes. In this region, the epithelial cells develop numerous short microvilli beneath which are many mitochondria. As the funnel is reached, the intima becomes extremely thick, and the epithelial cells lack microvilli and most microtubules. Within the funnel, a new, very distinct form of cuticle appears, which is in “units”, each associated with an epithelial cell and having a rounded epicuticular cap. The new cuticle arises ventrally but rapidly spreads to encircle the entire lumen, at which point the LED is considered to begin. Beneath this new cuticle, the epithelial cells are columnar, have long microvilli, numerous mitochondria in the apical cytoplasm, and rough endoplasmic reticulum basally. Apically, adjacent cells are tightly apposed; however, prominent intercellular channels develop more basally. The ejaculatory duct's features are briefly discussed in terms of its role in spermatophore formation.  相似文献

The organization of the dwarf male of Bonellia viridis was studied by electron microscopy. The epidermis is formed by two types of epithelial cells: the majority are multiciliated cells; highly vacuolated, non-ciliated cells are less abundant. The body wall musculature consists of an outer circular, a diagonal, and a longitudinal layer. As a unique feature in coelomate spiralians it was found that the perikarya of all muscle cells are located internal to the entire contractile muscular layer. The muscles are solitary myocytes embedded in extracellular matrix. Masses of secretory and indifferent cells occur inside the muscles. Two types of secretory cells were distinguished. Both of them apparently undergo holocrine secretion. A complete lining of thin peritoneal cells delimits the body cavity. Also, the gut and sperm sac have a complete peritoneal lining. The coelomic lining of the gut is a single-layered myoepithelium, that of the sperm sac a pseudo-stratified myoepithelium. The vas deferens was seen to be ciliated. The entrance of the sperm sac is formed by a ciliated funnel that leads into the reservoir by means of a thin, ciliated canal. The existence of repeated transverse nerves and of four longitudinal nerve cords is described for the first time.  相似文献

The morphology and fine structure of the labial gland reservoirs in the subterranean termite Reticulitermes santonensis (Isoptera: Rhinotermitidae) was studied by light and transmission electron microscopy. The reservoir wall consists of a single epithelial cell layer and a cuticular intima. The reservoir ducts are formed by a flat epithelial matrix with cuticular ridges lining the duct lumen. Measurements of the ionic concentrations of reservoir fluids and haemolymph show that the osmolality of reservoir fluid ranges from 7 to 28 mosmol kg−1; the haemolymph osmotic pressure was 201 ± 31 mosmol kg. The reservoir lumen is effectively separated from the haemolymph compartment; a net water flow through the reservoir wall could not be induced in physiological experiments. Moreover, typical epithelial structures associated with a fluid transport against an osmotic gradient are lacking. Thus, our fine structural and physiological data support the view that a water transfer from the haemolymph through the reservoir wall into the reservoir lumen does not occur.  相似文献

The excretory duct of pyriform glands in Araneus diadematus is connected to the secretory sac through an intermediary cell ring. Apices of these cells bear thick, long microvilli and cytoplasmic extensions containing microtubules in bundles, some of which are derived from normal basal bodies. These finger-like extensions lie between the cuticular intima and the secretory product; they are thought to protect the intima and to initiate moulding of the silk thread. Structural features of the duct cells suggest that the latter play a role in the control of the water content of the silk glue which is restricted to the last portion of the duct where numerous nerve endings are inserted between cells. It is evident that duct structure and chemical and physical characteristics of silk are correlated in all spider silk glands.  相似文献

The cuticular lining of the stomodeum in orthopteroid insects usually bears cuticular structures, consisting of ridges, teeth, and spines, which are sometimes fairly complex. In some of these insects, particularly cockroaches and Orthoptera s. str., some of these structures have systematic value, mainly at the higher systematic levels. The cuticular lining of the stomodeum in Paulinia acuminata (De Geer) and Marellia remipes (Uvarov) (Orthoptera : Pauliniidae) (the only 2 genera included within the family) have been studied. Wide differences in the pattern of the stomodeal armature, and also in the shape, size and distribution of the spines in its different parts, are recorded. The proportions and limits of the crop and proventriculus, and the form and spatial distribution of spines and other characters, relative to the ridges, appear to be of taxonomic significance. They indicate that the 2 genera may not be closely related, as has been suggested, based on other evidence. While Marelliaappears to be related to the Acrididae, Pauliniaseems to be isolated from other acridoids, except for certain features in common with Proscopiidae and Eumastacidae.  相似文献

Vertebrate muscle Z-bands show zig-zag densities due to different sets of alpha-actinin cross-links between anti-parallel actin molecules. Their axial extent varies with muscle and fibre type: approximately 50 nm in fast and approximately 100 nm in cardiac and slow muscles, corresponding to the number of alpha-actinin cross-links present. Fish white (fast) muscle Z-bands have two sets of alpha-actinin links, mammalian slow muscle Z-bands have six. The modular structure of the approximately 3 MDa protein titin that spans from M-band to Z-band correlates with the axial structure of the sarcomere; it may form the template for myofibril assembly. The Z-band-located amino-terminal 80 kDa of titin includes 45 residue repeating modules (Z-repeats) that are expressed differentially; heart, slow and fast muscles have seven, four to six and two to four Z-repeats, respectively. Gautel et al. proposed a Z-band model in which each Z-repeat links to one level of alpha-actinin cross-links, requiring that the axial extent of a Z-repeat is the same as the axial separation of alpha-actinin layers, of which there are two in every actin crossover repeat. The span of a Z-repeat in vitro is estimated by Atkinson et al. to be 12 nm or less; much less than half the normal vertebrate muscle actin crossover length of 36 nm. Different actin-binding proteins can change this length; it is reduced markedly by cofilin binding, or can increase to 38.5 nm in the abnormally large nemaline myopathy Z-band. Here, we tested whether in normal vertebrate Z-bands there is a marked reduction in crossover repeat so that it matches twice the apparent Z-repeat length of 12 nm. We found that the measured periodicities in wide Z-bands in slow and cardiac muscles are all very similar, about 39 nm, just like the nemaline myopathy Z-bands. Hence, the 39 nm periodicity is an important conserved feature of Z-bands and either cannot be explained by titin Z-repeats as previously suggested or may correlate with two Z-repeats.  相似文献

Ultrastructural study of gonadal muscles in sea star, Asterina pectinifera, showed that myoepithelial cells were located only in the epithelial lining of the genital coelomic sinus. No myoepithelial cells were found in the visceral peritoneal epithelium or within connective tissue layer of the outer sac. Morphology of the myoepithelial cells in gonads of A. pectinifera varies during the reproductive cycle. During the gametogenic phase of the reproductive cycle, the myoepithelial cells get an elongated, spindle-like shape having a length of 20–30 μm. In prespawning gonads, many of the myoepithelial cells form cytoplasmic extensions of 3–5 μm in length, filled with myofilaments and penetrating into the underlying connective tissue of the outer sac or haemal sinus. Besides, myoepithelial cells, simultaneously anchored in the inner and outer sacs, were also observed. These changes result in development of more elaborated musculature and increase in contractility of the gonadal wall in prespawning gonads as compared to that during other stages of the reproductive cycle.  相似文献

The insect tracheal system is an air-filled branching network of internal tubing that functions to exchange respiratory gases between the tissues and the environment. The light and electron-micrographs presented in this study show tracheae in the process of moulting, captured from the metathoracic hopping femur of a juvenile third instar locust (Locusta migratoria). The images provide evidence for the detachment of the cuticular intima from the tracheal epithelial cells, the presence of moulting fluid between the new and old cuticle layers, and the withdrawal of the shed cuticular lining through larger upstream regions of the tracheal system during moulting. The micrographs also reveal that the cuticular intima of the fine terminal branches of the tracheal system is cast at ecdysis. Therefore, the hypothesis that tracheoles retain their cuticle lining at each moult may not apply to all insect species or developmental stages.  相似文献

Adult carabids that ingest particles of food have a muscular proventriculus lined internally with an intima that bears sclerotized projections. In experiments where six species of carabid were allowed to feed upon Drosophila, the average size of cuticular remains found in post-proventricular regions of the gut was generally significantly smaller than those in the crop. This reduction in size of cuticular particles is likely to be due to proventricular trituration rather than prolonged exposure to digestive secretions. There is no evidence that material is triturated by the proventriculus and returned to the crop, the main seat of digestion in carabids, so proventricular trituration is unlikely to enhance digestion. By crushing large, potentially abrasive, cuticular particles, the proventriculus may serve to protect midgut epithelial tissue.  相似文献

Genetic analysis of muscle specification, formation and function in model systems has provided valuable insight into human muscle physiology and disease. Studies in Drosophila have been particularly useful for discovering key genes involved in muscle specification, myoblast fusion, and sarcomere organization. The muscles of the Drosophila female reproductive system have received little attention despite extensive work on oogenesis. We have used newly available GFP protein trap lines to characterize of ovarian muscle morphology and sarcomere organization. The muscle cells surrounding the oviducts are multinuclear with highly organized sarcomeres typical of somatic muscles. In contrast, the two muscle layers of the ovary, which are derived from gonadal mesoderm, have a mesh-like morphology similar to gut visceral muscle. Protein traps in the Fasciclin 3 gene produced Fas3::GFP that localized in dots around the periphery of epithelial sheath cells, the muscle surrounding ovarioles. Surprisingly, the epithelial sheath cells each contain a single nucleus, indicating these cells do not undergo myoblast fusion during development. Consistent with this observation, we were able to use the Flp/FRT system to efficiently generate genetic mosaics in the epithelial sheath, suggesting these cells provide a new opportunity for clonal analysis of adult striated muscle.  相似文献

Electron micrographs of serial sections show that the male sexual apparatus of Scutellonema brachyurum includes two morphologically identical spicules. Each is composed of a swollen tubular head, crescentic shaft, and leaf-like blade with membranous velum expanded from the central trunk. The spicules are concave and grooved on the ventral side and convex on the dorsal side near the trunk. The trunk is continuous with the shaft and head. Nerve tissue occupies the core of the spicule and includes a dendritic process which gains access to the exterior via a small pore on the lateral side of the spicule tip. Three protractor and two retractor muscles are associated with each spicule. A sensory accessory piece connects with the tip of the gubernaculum and protrudes from the lower side of the opening of the spicular pouch; it protracts and retracts with the muscularized gubernaculum. The gubernaculum varies from bow-shaped in the distal part to boat-shaped in the mid region. A sac exits beneath the accessory piece as a buffer for its movement. A cuticular guiding bar originating from the dorsal wall of the spicular pouch has a tongue. The ventral surface of the tongue is sclerotized to separate the two spicules. It is mobile by muscles of the protractor gubernaculi, retractor gubernaculi, and seductor gubernaculi.  相似文献

Tropomodulin and tropomyosin are important components of sarcomeric thin filaments in striated muscles. Tropomyosin decorates the side of actin filaments and enhances tropomodulin capping at the pointed ends of the filaments. Their functional relationship has been extensively characterized in vitro, but in vivo and cellular studies in mammals are often complicated by the presence of functionally redundant isoforms. Here, we used the nematode Caenorhabditis elegans, which has a relatively simple composition of tropomodulin and tropomyosin genes, and demonstrated that tropomodulin (unc-94) and tropomyosin (lev-11) are mutually dependent on each other in their sarcomere localization and regulation of sarcomeric actin assembly. Mutation of tropomodulin caused sarcomere disorganization with formation of actin aggregates. However, the actin aggregation was suppressed when tropomyosin was depleted in the tropomodulin mutant. Tropomyosin was mislocalized to the actin aggregates in the tropomodulin mutants, while sarcomere localization of tropomodulin was lost when tropomyosin was depleted. These results indicate that tropomodulin and tropomyosin are interdependent in the regulation of organized sarcomeric assembly of actin filaments in vivo.  相似文献

The spermathecal complex of the bark beetle, Ips typographus, comprises the following elements: spermathecal duct, spermatheca and spermathecal gland. The spermathecal duct connects the vagina and the spermatheca and consists of a cuticular tube surrounded by an epithelial layer and circular muscles. The spermatheca is bottle-shaped and has a cuticle-lined lumen. Muscles are attached to both ends of the spermatheca. The spermathecal gland which is connected to the spermatheca possesses three cell types: glandular, hypodermal, and ductule. The glandular cells have different structural characteristics depending on the age and reproductive state of the females. After the emergence of the brood, two different kinds of secretory material are present in the glandular cells. There is evidence that one type of secretion is emitted during the first few days after brood emergence, while the other type accumulates to be secreted during later stages.  相似文献

The coxal glands of M. murismusculi consist of the proximal tubular portion (tubulus), the distal glandular sac and the terminal excretory duct. The tubulus comprises looped proximal and distal tubes that run in close association with each other. The cells of the proximal tube form numerous short protrusions that project into the neighbouring organs through the pores in their basal lamina. The sac is a distal part of the gland and so it cannot be considered as a homologue of the proximal filter sacculus of other arthropods. A large number of pinocytotic vesicles and lysosome-like bodies in the epithelial lining of the sac imply that the main functions of this organ may be the absorption of substances from the lumen of the gland and their subsequent intracellular transformation. In addition the sac of females was shown to produce dense secretory granules. The ultrastructural features of the glands are discussed and compared to other representatives of Acari.  相似文献

A laboratory colony of Oncopeltus fasciatus was found to be infected by Leptomonas oncopelti. The flagellates form a carpet attached to the cuticular intima of the rectal glands of adult bugs. The epithelial cells of these glands are characterized by infolded apical plasma membranes associated with mitochondria; the overlying cuticular intima shows endocuticular canals. The Leptomonas are attached by hemidesmosomes, located most often at the tip of the flagella. The Protozoa multiply by budding and the resultant straphangers cling to the parental flagellum. Adhesion of the flagellates to the cuticular lining is so strong that detaching flagellates carry with them the outer part of the epicuticle. Epicuticle repair presumably occurs through the endocuticular canals.  相似文献
