Summary 1. Tetrodotoxin (TTX) has been widely used as a chemical tool for blocking Na+ channels. However, reports are accumulating that some Na+ channels are resistant to TTX in various tissues and in different animal species. Studying the sensitivity of Na+ channels to TTX may provide us with an insight into the evolution of Na+ channels.2. Na+ channels present in TTX-carrying animals such as pufferfish and some types of shellfish, frogs, salamanders, octopuses, etc., are resistant to TTX.3. Denervation converts TTX-sensitive Na+ channels to TTX-resistant ones in skeletal muscle cells, i.e., reverting-back phenomenon. Also, undifferentiated skeletal muscle cells contain TTX-resistant Na+ channels. Cardiac muscle cells and some types of smooth muscle cells are considerably insensitive to TTX.4. TTX-resistant Na+ channels have been found in cell bodies of many peripheral nervous system (PNS) neurons in both immature and mature animals. However, TTX-resistant Na+ channels have been reported in only a few types of central nervous system (CNS). Axons of PNS and CNS neurons are sensitive to TTX. However, some glial cells have TTX-resistant Na+ channels.5. Properties of TTX-sensitive and TTX-resistant Na+ channels are different. Like Ca2+ channels, TTX-resistant Na+ channels can be blocked by inorganic (Co2+, Mn2+, Ni2+, Cd2+, Zn2+, La3+) and organic (D-600) Ca2+ channel blockers. Usually, TTX-resistant Na+ channels show smaller single-channel conductance, slower kinetics, and a more positive current-voltage relation than TTX-sensitive ones.6. Molecular aspects of the TTX-resistant Na+ channel have been described. The structure of the channel has been revealed, and changing its amino acid(s) alters the sensitivity of the Na+ channel to TTX.7. TTX-sensitive Na+ channels seem to be used preferentially in differentiated cells and in higher animals instead of TTX-resistant Na+ channels for rapid and effective processing of information.8. Possible evolution courses for Na+ and Ca2+ channels are discussed with regard to ontogenesis and phylogenesis.  相似文献

We investigated the effects of pressure overload hypertrophy on inward sodium (I Na) and calcium currents (I Ca) in single left ventricular myocytes to determine whether changes in these current systems could account for the observed prolongation of the action potential. Hypertrophy was induced by pressure overload caused by banding of the abdominal aorta. Whole-cell patch clamp experiments were used to measure tetrodotoxin (TTX)-sensitive inward currents. The main findings were that I Ca density was unchanged whereas I Na density after stepping from –80 to –30 mV was decreased by 30% (–9.0 ± 1.16 pA pF–1 in control and –6.31 ± 0.67 pA pF–1 in hypertrophy, p < 0.05, n= 6). Steady-state activation/inactivation variables of I Na, determined by using double-pulse protocols, were similar in control and hypertrophied myocytes, whereas the time course of fast inactivation of I Na was slowed (p < 0.05) in hypertrophied myocytes. In addition, action potential clamp experiments were carried out in the absence and presence of TTX under conditions where only Ca2+ was likely to enter the cell via TTX-sensitive channels. We show for the first time that a TTX-sensitive inward current was present during the plateau phase of the action potential in hypertrophied but not control myocytes. The observed decrease in I Na density is likely to abbreviate rather than prolong the action potential. Delayed fast inactivation of Na+ channels was not sustained throughout the voltage pulse and may therefore merely counteract the effect of decreased I Na density so that net Na+ influx remains unaltered. Changes in the fast I Na do not therefore appear to contribute to lengthening of the action potential in this model of hypertrophy. However, the presence of a TTX-sensitive current during the plateau could potentially contribute to the prolongation of the action potential in hypertrophied cardiac muscle. Changes in membrane properties of chick embryonic hearts during development   总被引:13,自引:3,他引:10  
The electrophysiological properties of embryonic chick hearts (ventricles) change during development; the largest changes occur between days 2 and 8. Resting potential (Em) and peak overshoot potential (+E max) increase, respectively, from -35 mv and +11 mv at day 2 to -70 mv and +28 mv at days 12–21. Action potential duration does not change significantly. Maximum rate of rise of the action potential (+V max) increases from about 20 v/sec at days 2–3 to 150 v/sec at days 18–21; + V max of young cells is not greatly increased by applied hyperpolarizing current pulses. In resting Em vs. log [K+]o curves, the slope at high K+ is lower in young hearts (e.g. 30 mv/decade) than the 50–60 mv/decade obtained in old hearts, but the extrapolated [K+]i values (125–140 mM) are almost as high. Input resistance is much higher in young hearts (13 MΩ at day 2 vs. 4.5 MΩ at days 8–21), suggesting that the membrane resistivity (Rm) is higher. The ratio of permeabilities, P Na/P K, is high (about 0.2) in young hearts, due to a low P K, and decreases during ontogeny (to about 0.05). The low K+ conductance (g K) in young hearts accounts for the greater incidence of hyperpolarizing afterpotentials and pacemaker potentials, the lower sensitivity (with respect to loss of excitability) to elevation of [K+]o, and the higher chronaxie. Acetylcholine does not increase g K of young or old ventricular cells. The increase in (Na+, K+)-adenosine triphosphatase (ATPase) activity during development tends to compensate for the increase in g K. +E max and + V max are dependent on [Na+]o in both young and old hearts. However, the Na+ channels in young hearts (2–4 days) are slow, tetrodotoxin (TTX)-insensitive, and activated-inactivated at lower Em. In contrast, the Na+ channels of cells in older hearts (> 8 days) are fast and TTX-sensitive, but they revert back to slow channels when placed in culture.  相似文献

The use-dependent phasic blockage of sodium channels by tetrodotoxin (TTX) and saxitoxin (STX) was examined in frog nodes of Ranvier using trains of depolarizing pulses. The decline of the peak Na+ current from its initial value (I 0) before the train to a stationary value (I ) after the train was more pronounced at more negative holding potentials. The relationship betweenI /I 0 and holding potential was fitted by a sigmoid function which yielded values for the steepness of the voltage dependencies of around –15 mV for TTX and – 8 mV for STX. Similar values were obtained at toxin concentrations of 4 and 8 nM. The higher voltage sensitivity of STX versus TTX is interpreted in terms of the higher charge and the faster binding kinetics of STX. These differences also explain the frequency dependence of the decline of Na+ currents with STX (between 0.5 and 2 Hz) and the frequency independence with TTX. Variation of the pulse amplitude in a train of conditioning pulses revealed that the magnitude of the use-dependent actions of STX parallels the steady-state Na+ inactivation curveh . Inhibition of inactivation, by pre-treatment with chloramine-T, did not, however, abolish the use dependence. Instead, it introduced a change in the time constants of the decline of the Na+ currents and the magnitude became independent of the holding potential.  相似文献

Spontaneous beating of heart-cell aggregates from 4-day chick embryos was initially blocked by 10-5 g/ml tetrodotoxin (TTX). With continued exposure to the drug, the fraction of blocked aggregates decreased from about 80% at 15 min to about 25% at 2–3 h, at which time, beating aggregates had become desensitized to the toxin, showing no response to a fresh dose. Aggregates from 5-day hearts were more sensitive to TTX, but fewer became desensitized in its presence. Desensitization to TTX was not seen in 6- and 7-day aggregates. Inhibition of protein synthesis by cycloheximide did not affect beating or initial sensitivity to TTX of 4-day aggregates, but desensitization failed to occur. Before TTX, the mean value of maximal upstroke velocity (Vmax) of the action potentials in 4-day aggregates was 33 V/s. After desensitization Vmax was 12 V/s. Activity of desensitized aggregates in the presence of TTX was augmented by elevated calcium levels, and suppressed by presumed inhibitors of slow inward current (manganese, D600). Desensitization was reversible; upon removal of TTX 10-5 g/ml, aggregates regained their responsiveness to a fresh dose of the drug with a 2–3 h time-course similar to that of desensitization. This was prevented by continued exposure to TTX at concentrations as low as 10-8 g/ml. These data suggest that (a) desensitization involves a change in the mode of action-potential generating from one involving Na-specific, TTX-sensitive channels to one utilizing slower Mn-sensitive channels; (b) the process of desensitization occurs over a period of 2–3 h and is dependent upon the products of protein synthesis; and (c) desensitization is reversible after removal of TTX over a 2–3 h time-course similar to its onset.  相似文献

The effects of intracellular application of trypsin on the Cl current induced by hypotonic cell swelling (I Cl,swell) in human prostate cancer epithelial cells (LNCaP) was studied using the patch-clamp technique. In cells predialyzed with 1 mg/mL trypsin, I Cl,swell developed and diminished in response to the application and withdrawal of hypotonic solution about three times faster than that in control cells. In trypsin-infused cells, I Cl,swell also had about twofold higher current density and displayed considerably slowed voltage-dependent inactivation, which was quite pronounced in control cells at potentials above +60 mV. Trypsin-induced modification of I Cl,swell could be prevented by coinfusion of 10 mg/mL soybean trypsin inhibitor, suggesting that proteolytic cleavage of essential intracellular structural domains of the I Cl,swell-carrying volume-regulated anion channel (VRAC) was responsible for this functional modification. The effect of trypsin was not dependent on the presence of intracellular ATP. We conclude that VRACs, similarly to voltage-gated Na+, K+, and Cl channels, possess intracellular inactivation domain(s) subjected to proteolytic cleavage that may function in conformity with the classical ball-and-chain inactivation model.  相似文献

Summary Ouabain-resistant (OR), volume-or N-ethylmaleimide (NEM)-stimulated K+(Rb+)Cl fluxes were measured in low-K+ sheep red cells and found to be functionally separate but immunologically similar. In anisosmotic solutions both K+ effluxes and Rb+ influxes of NEM-treated and control cells were additive. In contrast to the NEM-stimulated K+Cl flux, metabolic depletion did not reduce K+Cl flux of normal or swollen cells. The anion preference of OR K+ efflux in NEM-treated cells was Br>Cl>HCO 3 =FI=NO 3 =CNS, and hence consistent with a reported Br>Cl>NO 3 sequence of the volume-dependent K+Cl transport. Alloimmune anti-Ll antibodies known to decrease passive K+ fluxes in low K+ cells reduced by 51% both volume-and NEM-stimulated, furosemidesensitive Rb+Cl fluxes suggesting their immunologic identity, a conclusion also supported by anti-L1 absorption studies. Since pretreatment with anti-L1 prevented the activation of Rb+ influx by NEM, and the impermeant glutathionmaleimide-I did not stimulate Rb+Cl influx, the NEM reactive SH groups must be located apart from the L1 antigen either within the membrane or on its cytoplasmic face. A model is proposed consisting of a K+Cl transport path(s) regulated by a protein with two functional subunits or domains; a chemically (C s) and a volume (V s)-stimulated domain, both interfacing with the L1 surface antigen. Summary Electrophysiological and pharmacological properties distinguished subtypes of adult mammalian dorsal root ganglion neurons (DRGn) in monolayer dissociated cell culture. By analogy of action potential waveform and duration, neurons with short duration (SDn) and long duration (LDn) action potentials resembled functionally distinct subtypes of DRGn in intact ganglia. Patch clamp and conventional intracellular recording techniques were combined here to elucidate differences in the ionic basis of excitability of subtypes of DRGn in vitro. Both SDn and LDn were quiescent at the resting potential. Action potentials of SDn were brief (< 2 msec), sensitive to tetrodotoxin (TTX, 5–10 nM), exhibited damped firing during long depolarizations, and did not respond to algesic agents applied by pressure ejection. Action potentials of LDn were 2–6 msec in duration, persisted in 30 µM TTX, and fired repetitively during depolarizing current pulses or exposure to algesic agents (e.g., capsaicin, histamine and bradykinin). Whole-cell recordings from freshly dissociated neurons revealed two inward sodium currents (INa; variable with changes in sodium but not calcium concentration in the superfusate) in various proportions: a rapidly activating and inactivating, TTX-sensitive current; and, a slower, TTX (30 M)-resistant INa. Large neurons, presumable SDn, had predominantly TTX-sensitive current and little TTX-resistant current. The predominent inward current of small neurons, presumably LDn, was TTX-resistant with a smaller TTX-sensitive component. By analogy to findings from intact ganglia, these results suggest that fundamentally different ionic currents controlling excitability of subtypes of DRGn in vitro may contribute to functional differences between subtyes of neurons in situ.  相似文献

Summary The effects of complete substitution of gluconate for mucosal and/or serosal medium Cl on transepithelial Na+ transport have been studied using toad urinary bladder. With mucosal gluconate, transepithelial potential difference (V T) decreased rapidly, transepithelial resistance (R T) increased, and calculated short-circuit current (I sc) decreased. CalculatedE Na was unaffected, indicating that the inhibition of Na+ transport was a consequence of a decreased apical membrane Na+ conductance. This conclusion was supported by the finding that a higher amiloride concentration was required to inhibit the residual transport. With serosal gluconateV T decreased,R T increased andI sc fell to a new steady-state value following an initial and variable transient increase in transport. Epithelial cells were shrunken markedly as judged histologically. CalculatedE Na fell substantially (from 130 to 68 mV on average). Ba2+ (3mm) reduced calculatedE Na in Cl Ringer's but not in gluconate Ringer's. With replacement of serosal Cl by acetate, transepithelial transport was stimulated, the decrease in cellular volume was prevented andE Na did not fall. Replacement of serosal isosmotic Cl medium by a hypo-osmotic gluconate medium (one-half normal) also prevented cell shrinkage and did not result in inhibition of Na+ transport. Thus the inhibition of Na+ transport can be correlated with changes in cell volume rather than with the change in Cl per se. Nystatin virtually abolished the resistance of the apical plasma membrane as judged by measurement of tissue capacitance. With K+ gluconate mucosa, Na+ gluconate serosa, calculated basolateral membrane resistance was much greater, estimated basolateral emf was much lower, and the Na+/K+ basolateral permeability ratio was much higher than with acetate media. It is concluded the decrease in cellular volume associated with substitution of serosal gluconate for Cl results in a loss of highly specific Ba2+-sensitive K+ conductance channels from the basolateral plasma membrane. It is possible that the number of Na+ pump sites in this membrane is also decreased.  相似文献

The presence of low levels (2–4 mM) of (NH4)2SO4 in sporulation medium results in a complete inhibition of ascus formation in Saccharomyces cerevisiae. Tests with other utilizable nitrogen sources and with the non-metabolizable ammonia analog, methylamine, indicate that the primary inhibitory effect is exerted by ammonium (NH4+) and not a metabolite of NH4+. Viability is also reduced but to a much lesser extent than sporulation. The levels of NH4+ that repress sporulation do not affect the rate of oxygen utilization or ATP levels of cells in sporulation medium. The period during which the cells are sensitive to NH4+ is relatively long, and they do not escape inhibition until the meiotic commitment stage. The most sensitive period is roughly coincident with the end of premeiotic DNA synthesis. NH4+ does not inhibit the initiation of premeiotic DNA synthesis, but DNA replication is arrested after initiation and continued incubation in the presence of NH4+ leads to massive DNA degradation. Commitment to recombination is not affected specifically by NH4+. Preliminary experiments with an NH4+-resistant mutant indicates that levels of NH4+ sensitivity are under genetic control.  相似文献

Summary Transbasal electrical potential (V b) and intraepithelial potassium chemical activity ((K+) i ) were measured in isolated midgut epithelium of tobacco hornworm (Manduca sexta) using double-barrelled glass microelectrodes. Values ofV b ranging from +8 to –48 mV (relative to blood side) were recorded. For all sites, (K+) i is within a few millivolts of electrochemical equilibrium with the blood side bathing solution. Sites more negative than –20 mV show relatively high sensitivity ofV b to changes in blood side K+ concentration: 43% of these sites can be marked successfully with iontophoresed Lucifer yellow CH dye and shown to represent epithelial cells of all three types present in the midgut. In about half of successful marks, dye-coupling of several adjacent cells is seen. Low potential sites — those withV b less negative than –20 mV —typically do not show high sensitivity ofVb to changes of external K+, but rather (K+) i rapidly approaches the K+ activity of blood side bathing solution. These sites can seldom be marked with Lucifer yellow (4% success). The mean (K+) i of the high potential sites is 95±29 (sd)mm under standard conditions, a value which is in accord with published values for the whole tissue.  相似文献

Summary Cultured HeLa cells behave as ideal osmometers when subjected to hyperosmolar media, and show no volume regulatory behavior. In hypoosmolar solutions, cell swelling is not as great as predicted, and this is due largely to a loss of intracellular KCl. In hyperosmolar solutions there is a stimulation of the ouabain-insensitive but loop diuretic-sensitive86Rb+ (K+) pathway. Analysis of the K+, Na+ and Cl dependency of this K+ flux pathway demonstrates that the increase is principally due to an increase in its maximal velocity (V max). The sensitivity of this pathway to diuretic inhibition is unchanged in hyperosmolar media. Diuretic-sensitive86Rb+ (K+) efflux stimulated by hypertonicity shows no marked dependence on external K+. The K+ loss observed in hypoosmolar media is distinct from the K+ transport pathway stimulated by hyperosmolar media on the basis of its sensitivity to furosemide and anion dependence.  相似文献

Evidence of a role for abscisic acid (ABA) in signalling conditions of water stress and promoting stomatal closure is convincing, but past studies have left few clues as to its molecular mechanism(s) of action; arguments centred on changes in H+-pump activity and membrane potential, especially, remain ambiguous without the fundamental support of a rigorous electrophysiological analysis. The present study explores the response to ABA of K+ channels at the membrane of intact guard cells ofVicia faba L. Membrane potentials were recorded before and during exposures to ABA, and whole-cell currents were measured at intervals throughout to quantitate the steady-state and time-dependent characteristics of the K+ channels. On adding 10 M ABA in the presence of 0.1, 3 or 10 mM extracellular K+, the free-running membrane potential (V m) shifted negative-going (–)4–7 mV in the first 5 min of exposure, with no consistent effect thereafter. Voltage-clamp measurements, however, revealed that the K+-channel current rose to between 1.84- and 3.41-fold of the controls in the steady-state with a mean halftime of 1.1 ± 0.1 min. Comparable changes in current return via the leak were also evident and accounted for the minimal response inV m. Calculated atV m, the K+ currents translated to an average 2.65-fold rise in K+ efflux with ABA. Abscisic acid was not observed to alter either K+-current activation or deactivation.These results are consistent with an ABA-evoked mobilization of K+ channels or channel conductance, rather than a direct effect of the phytohormone on K+-channel gating. The data discount notions that large swings in membrane voltage are a prerequisite to controlling guard-cell K+ flux. Instead, thev highlight a rise in membranecapacity for K+ flux, dependent on concerted modulations of K+-channel and leak currents, and sufficiently rapid to account generally for the onset of K+ loss from guard cells and stomatal closure in ABA.  相似文献

Summary The utility of the lipophilic anion thiocyanate (SCN+) as a probe for the indirect estimation of the cell membrane potential (V m ) in Ehrlich ascites tumor cells has been evaluated by comparison to direct electrophysiological measurements. SCN accumulation is consisten with first-order uptake into a single kinetically-identifiable cellular compartement, achieving steadystate distribution in 20–30 min at 22°C. The steady-state distribution ratio ([SCN] c /[SCN] e ) in physiological saline is 0.44±0.02. Treatment of the cells with proparanolol (0.13 mM), an activator of Ca2+ dependent K+ channels, reduces the steady-state distribution ratio to 0.19±0.02. Conversely, treatmetn with BACl2 (10 mM), an antagonist of the pathway, increases the SCN distribution ratio to 0.62±0.01. The equilibrium potentials (V SCN ) calculated under these conditions are virtually identical to direct electrophysiological measurements of theV m made under the same conditions. The effect of varing extracellular [K+]([K+] e ) in the presence of constant [Na+] e =100 mM has also been tested. In control cells, elevation of [K+] e from 6 to 60 mM reducesV SCN from –20.6±1.0 to –13.2±1.2 mV. Again, microelectrode measurements give excellent quantitative agreement. Propranolol increases the sensitivity of the cells to varying [K+] e , so that a 10-fold elevation reducesV SCN by approximately 31 mV. BaCl2 greatly reduces this reponse: a 10-fold elevation in [K+] e yielding only a 4-mV rediction inV SCN . It is concluded that the membrane potential of Ehrlich cells can be estimated accurately from SCN distribution measurements.  相似文献

Summary The mechanism of salt tolerance was studied using isolated internodal cells of the charophyteNitellopsis obtusa grown in fresh water. When 100 mM NaCl was added to artificial pond water (0.1 mM each of NaCl, KC1, CaCl2), no cell survived for more than one day. Within the first 30 minutes, membrane potential (Em) depolarized and membrane resistance (Rm) decreased markedly. Simultaneously, cytoplasmic Na+ increased and K+ decreased greatly. At steady state the increase in Na+ content was roughly equal to the decrease in K+ content. The Cl content of the cytoplasm did not change. These results suggest that Na+ enters the cytoplasm by exchange with cytoplasmic K+. Both the entry of Na+ and the exit of K+ are assumed to be passive and the latter being caused by membrane depolarization. Vacuolar K+, Na+, and Cl remained virtually constant, suggesting that rapid influx of Na+ from the cytoplasm did not occur.In 100 mM NaCl containing 10 mM CaCl2, membrane depolarization, membrane resistance decrease and changes in cytoplasmic [Na+] and [K+] did not occur, and cells survived for many days. When cells treated with 100 mM NaCl were transferred within 1 hour to 100 mM NaCl containing 10 mM CaCl2, Em decreased, Rm increased, cytoplasmic Na+ and K+ returned to their initial levels, and cells survived. Two possible mechanisms for the role of Ca2+ in salt tolerance inNitellopsis are discussed; one a reduction in plasmalemma permeability to Na+ and the other a stimulation of active Na+-extrusion.  相似文献

Summary Antibodies which were raised against highly purified membrane-bound (Na+–K+)-ATPase from the outer medulla of rat kidneys inhibit the (Na+–K+)-ATPase activity up to 95%. The antibody inhibition is reversible. The time course of enzyme inhibition and reactivation is biphasic in semilogarithmic plots.In the purified membrane-bound (Na+–K+)-ATPase negative cooperativity was observed (a) for the ATP dependence of the (Na+–K+)-ATPase activity (n=0.86), (b) for the ATP binding to the enzyme (n=0.58), and (c) for the ouabain inhibition of the (Na+–K+)-ATPase activity (n=0.77). By measuring the Na+ dependence of the (Na+–K+-ATPase reaction, a positive homotropic cooperativity (n=1.67) was found.As reactivation of the antibody-inhibited enzyme proceeds very slowly (t 0.5=5.2hr), it was possible to measure characteristics of the antibody-(Na+–K+)-ATPase complex: The antibodies exerted similar effects on the ATP dependence of the (Na+–K+)-ATPase reaction and on the ATP binding of the enzyme.V max of the (Na+–K+)-ATPase reaction and the number of ATP binding sites were reduced whileK 0.5 ATP for the (Na+–K+)-ATPase activity and for the ATP binding were increased by the antibodies. The Hill coefficients for the ATP binding and for the ATP dependence of the enzyme activity were not significantly altered by the antibodies. The antibodies increased theK 0.5 value for the Na+ stimulation of the (Na+–K+)-ATPase activity, but they did not alter the homotropic interactions between the Na+-binding sites. The negative cooperativity which was observed for the ouabain inhibition of the (Na+–K+)-ATPase activity was abolished by the antibodies.The data are tentatively explained by the following model: The antibodies bind to the (Na+–K+)-ATPase from the inner membrane side, reduce the ATP binding symmetrically at the ATP binding sites and reduce thereby also the (Na+–K+)-ATPase activity of the enzyme. The antibodies may inhibit the ATP binding by a direct interaction or by means of a conformational change at the ATP binding sites. This may possibly also lead to the alteration of the Na+ dependence of the (Na+–K+)-ATPase activity and to the observed alteration of the dose response to the ouabain inhibition.  相似文献

Sodium channels of human small-cell lung cancer (SCLC) cells were examined with whole-cell and single-channel patch clamp methods. In the tumor cells from SCLC cell line NCI-H146, the majority of the voltage-gated Na+ channels are only weakly tetrodotoxin (TTX)-sensitive (K d =215 mm). With the membrane potential maintained at –60 to –80 mV, these cells produced all-or-nothing action potentials in response to depolarizing current injection (>20 pA). Similar all-ornothing spikes were also observed with anodal break excitation. Removal of external Ca2+ did not affect the action potential production, whereas 5 m TTX or substitution of Na+ with choline abolished it. Action potentials elicited in the Ca2+-free condition were reversibly blocked by 4 mm MnCl2 due to the Mn2+-induced inhibition of voltage-dependent sodium currents (I Na). Therefore, Na+ channels, not Ca2+ channels, underlie the excitability of SCLC cells. Whole-cell I Na was maximal with step-depolarizing stimulations to 0 mV, and reversed at +45.2 mV, in accord with the predicted Nernst equilibrium potential for a Na+-selective channel. I Na evoked by depolarizing test potentials (–60 to +40 mV) exhibited a transient time course and activation/ inactivation kinetics typical of neuronal excitable membranes; the plot of the Hodgkin-Huxley parameters, m and h, also revealed biophysical similarity between SCLC and neuronal Na+ channels. The single channel current amplitude, as measured with the inside-out patch configuration, was 1.0 pA at –20 mV with a slope conductance of 12.1 pS. The autoantibodies implicated in the Lambert-Eaton myasthenic syndrome (LES), which are known to inhibit I Ca and I Na in bovine adrenal chromaffin cells, also significantly inhibited I Na in SCLC cells. These results indicate that (i) action potentials in human SCLC cells result from the regenerative increase in voltage-gated Na+ channel conductance; (ii) fundamental characteristics of SCLC Na+ channels are the same as the classical sodium channels found in a variety of excitable cells; and (iii) in some LES patients, SCLC Na+ channels are an additional target of the pathological IgG present in the patients' sera.Department of Biomedical EngineeringThis study was supported by National Institutes of Health grant NS18607 and a research grant from the Muscular Dystrophy Association. Summary Photoreceptor cells of the drone,Apismellifera , have a voltage-gated Na+ membrane conductance that can be blocked by tetrodotoxin (TTX) and generates an action potential on abrupt depolarization: an action potential is triggered by the rising phase of a receptor potential evoked by an intense light flash (Autrum and von Zwehl 1964; Baumann 1968). We measured the intracellular voltage response to a small (9%), brief (30 ms) decrease in light intensity from a background, and found that its amplitude was decreased by 1M TTX. The response amplitude was maximal when the background intensity depolarized the cell to –38 mV. With intensities depolarizing the cell membrane to –45 to –33 mV the average response amplitude was decreased by TTX from 1.2mV to 0.5mV. TTX is also known to decrease the voltage noise during steady illumination (Ferraro et al. 1983) but, despite this, the ratio of peak-to-peak signal to noise was, on average, decreased by TTX. Pathways of K+ movement across the erythrocyte membrane of frog Rana temporaria were studied using 86Rb as a tracer. The K+ influx was significantly blocked by 0.1 mmol·l-1 ouabain (by 30%) and 1 mmol·l-1 furosemide (by 56%) in the red cells incubated in saline at physiological K+ concentration (2.7 mmol·l-1). Ouabain and furosemide had an additive effect on K+ transport in frog red cells. The ouabain-sensitive and furosemide-sensitive components of K+ influx saturated as f(K+)e with apparent K m values for external K e + concentration of 0.96±0.11 and 4.6±0.5 mmol·l-1 and V max of 0.89±0.04 and 2.8±0.4 mmol·l cells-1·h-1, respectively. The residual ouabain-furosemide-resistant component was also a saturable function of K e + medium concentration. Total K+ influx was significantly reduced when frog erythrocytes were incubated in NO - 3 medium. Furosemide did not affect K+ transport in frog red cells in NO 3 - media. At the same K e + concentration the ouabain-furosemide-insensitive K+ influx in Cl- medium was significantly greater than that in NO - 3 medium. We found no inhibitory effect of 1 mmol·l-1 furosemide on Na+ influx in frog red cells in Cl- medium. K+ loss from the frog erythrocytes in a K+-free medium was significantly reduced (mean 58%) after replacement of Cl- with NO - 3 . Furosemide (0.5 mmol·l-1) did not produce any significant reduction in the K+ loss in both media. The Cl--dependent component of K+ loss from frog red cells was 5.7±1.2 mmol·l-1·h-1.
