The pulping of wheat straw with dimethyl formamide was studied in order to investigate the effects of the cooking variables (temperature (190 degrees C, 200 degrees C, and 210 degrees C) and time (120 min, 150 min, and 180 min) and organic solvent ratio (30%, 50%, and 70%) dimethyl formamide (DMF+water) value) on the degradation of cellulose and degree of polymerization (DP) of organosolv pulp. The SCAN viscosity was applied to estimating the extent of cellulose degradation produced by cooking condition and then, it was compared with Kraft pulp at equal Kappa number. Response of pulp and handsheets properties to the process variables were analyzed using statistical software (MINITAB 14). The process variables (cooking temperature and cooking time) must be set at low variables with high DMF ratio in order to ensure a high yield and high SCAN viscosity. Also, pulps with high mechanical properties can be acceptably obtained at 210 degrees C for 150 min with 50% DMF. Generally, the cooking temperature was a significant factor while the cooking time and DMF ratio had a smaller role. By the comparison of Kraft and organosolv pulp, it can be resulted that DMF basically had improvement role on reducing of cellulose degradation by reason of high SCAN viscosity of organosolv pulp than Kraft pulp under equal kappa number and, scanning electron microscopy (SEM) of obtained pulp. Consequently, the protective action of organic solvent on non-cellulosic polysaccharides of wheat straw against degradation under Kraft pulping conditions was pointed as a main reason of the fairly high yield of organosolv pulps.  相似文献

Ultrasound enhanced glucose release from corn in ethanol plants   总被引:2,自引:0,他引:2  
This work evaluated the use of high power ultrasonic energy to treat corn slurry in dry corn milling ethanol plants to enhance liquefaction and saccharification for ethanol production. Corn slurry samples obtained before and after jet cooking were subjected to ultrasonic pretreatment for 20 and 40 s at amplitudes of vibration ranging from 180 to 299 microm(pp) (peak to peak amplitude in microm). The resulting samples were then exposed to enzymes (alpha-amylase and glucoamylase) to convert cornstarch into glucose. A comparison of scanning electron micrographs of raw and sonicated samples showed the development of micropores and the disruption of cell walls in corn mash. The corn particle size declined nearly 20-fold following ultrasonic treatment at high power settings. The glucose release rate from sonicated samples increased as much as threefold compared to the control group. The efficiency of ultrasound exceeded 100% in terms of energy gain from the sugar released over the ultrasonic energy supplied. Enzymatic activity was enhanced when the corn slurry was sonicated with simultaneous addition of enzymes. This finding suggests that the ultrasonic energy did not degrade or denature the enzymes during the pretreatment.  相似文献

High temperature extrusion cooking of rye was used as a pretreatment for ethanol fermentation, and yeasts and bacteria were compared for their fermentation rates. Extrusion cooking caused, on average, a 7.5% increase in ethanol yield in comparison to autoclaved samples. The best results were achieved for grain with a moisture of 21-23% which was extruded at temperatures of 160-180 °C. Extrusion decreased the relative viscosity of rye grain water extracts, so it was possible to mash it without α-amylase. The efficiency of fermentation of extruded rye without Termamyl was equal to that of autoclaved and traditionally mashed rye (using α-amylase). The rate of fermentation of extruded rye grain by Zymomonas was higher during the first stage, but the final ethanol yield was similar for the bacterium and the yeast. Though both microorganisms gave good quality distillates, the concentration of compounds other than ethanol achieved from extruded rye mashes, which were fermented by Z. mobilis, was five times lower than for yeasts.  相似文献

The nectivorous ant Camponotus mus shows a broad size variation within the worker caste. Large ants can ingest faster and larger loads than small ones. Differences in physiological abilities in fluid ingestion due to the insect size could be related to differences in decision-making according to ant size during nectar foraging. Sucrose solutions of different levels of sugar concentration (30% or 60%w/w), viscosity (high or low) or flow rate (ad libitum or 1microl/min) were offered in combination to analyse the behavioural responses to each of these properties separately. Differences were found depending on ant body size and the property compared. A regulated flow produced smaller crop loads for medium and large ants compared to the same solution given ad libitum. All foragers remained longer times feeding at the regulated flow source but larger ants often made longer interruptions. When sugar concentration was constant but viscosity was high, only large ants increased feeding time. Constant viscosity with different sugar concentration determined longer feeding time and bigger loads for the most concentrated solution for small but not for large ants. Small ants reached similar crop loads in a variety of conditions while large ants did not. These differences could be evidence of a possible specialization for nectar foraging based on ant body size.  相似文献

Nectar is an essential resource for bumblebees and many other flower-visiting insects. The main constituents of nectar are sugars, which vary in both composition and concentration between plant species. We assessed the influence of sugar concentration, sugar solution viscosity and sugar solution composition on the imbibition and energy intake rate of bumblebees, Bombus impatiens Cresson (Hymenoptera: Apidae). To do this, we measured their rate of solution intake for 49 different sugar solution treatments, which varied in both sugar composition and concentration. In general, the imbibition rates of bumblebees were found to increase with increasing sugar concentration, probably due to their preference for high sugar concentrations, up to a concentration of 27% (w/w), at which point solutions reached a threshold viscosity of approximately 1.5–1.6 mPa.s. Above this threshold, the increasing viscosity of the solutions physically inhibited the imbibition rates of bees, and imbibition rate began to decrease as the concentration increased. Nevertheless, bumblebee energy intake rate increased with increasing concentration up to about 42–56%. Although we found that sugar solution composition had an impact on both imbibition and energy intake rate, its effect was not as straightforward as that of sugar concentration and viscosity.  相似文献

以‘富士’苹果叶片为材料, 采用刺伤接种法, 比较了苹果树腐烂病菌强致病菌LXS080601和弱致病菌LXS081501侵染对寄主体内丙二醛(MDA)含量、渗透调节物质及防御酶活性的影响。结果表明, 两菌株侵染后, 叶片MDA、蛋白质和可溶性糖含量均增加, 接种LXS080601叶片MDA含量快速上升, 最大增幅为141.15%, 而接种LXS081501叶片的MDA含量变化较小, 增幅仅为1.16%-16.24%, 但后者可溶性糖和蛋白质含量增幅分别高达158.12%和113.57%, 显著高于接种LXS080601的处理。同时, 两菌株均能诱导叶片内4种防御酶活性的升高, 但LXS081501诱导的苯丙氨酸解氨酶(PAL)、过氧化物酶(POD)、过氧化氢酶(CAT)和多酚氧化酶(PPO)活性均显著高于LXS080601, 说明不同致病力腐烂病菌对寄主防御酶活性的影响存在显著差异。  相似文献

The use of ultrasound pretreatment to enhance liquefaction and saccharification of cassava chips was investigated. Cassava chip slurry samples were subjected to sonication for 10-40 s at three power levels of low (2 W/mL), medium (5 W/mL), and high (8 W/mL). The samples were simultaneously exposed to enzymes to convert starch into glucose. The cassava particle size declined nearly 40-fold following ultrasonic pretreatment at high power input. Scanning electron micrographs of both unsonicated (control) and sonicated samples showed disruption of fibrous material in cassava chips but did not affect the granular structure of starch. Reducing sugar release improved in direct proportion to the power input and sonication time. The reducing sugar increase was as much as 180% with respect to the control groups. The slurry samples with enzyme addition during sonication resulted in better reducing sugar release than the samples with enzyme addition after sonication. The heat generated during sonication below starch gelatinization temperature apparently had no effect on the reducing sugar release. The reducing sugar yield and energy efficiency of ultrasound pretreated samples increased with total solids (TS) contents. The highest reducing sugar yield of 22 g/100 g of sample and efficiency of 323% were obtained for cassava slurry with 25% TS at high power. The reducing sugar yield at the completion of reaction (R(infinity)) were over twofold higher compared to the control groups. The integration of ultrasound into a cassava-based ethanol plant may significantly improve the overall ethanol yield.  相似文献

通过测定不同发育时期肉苁蓉和寄主梭梭体内主要糖类物质含量和蔗糖代谢相关酶活性,以研究寄生植物与寄主植物的糖代谢及其关系。结果表明:未寄生肉苁蓉的梭梭以积累葡萄糖为主,而寄生肉苁蓉的梭梭在夏季休眠期以积累葡萄糖为主,进入秋季旺盛生长期时以积累蔗糖为主。肉苁蓉的糖分积累与梭梭不同,己糖含量约占可溶性总糖的62.45%,而蔗糖仅为可溶性总糖的4.98%,故肉苁蓉为己糖积累型。寄主梭梭同化枝内蔗糖磷酸合成酶活性较转化酶活性和蔗糖合成酶活性低,其中寄生肉苁蓉的梭梭的分解酶类活性高于未寄生肉苁蓉的梭梭。肉苁蓉体内转化酶活性较低,而蔗糖合成酶和蔗糖磷酸合成酶活性较高,且蔗糖合成酶活性高于蔗糖磷酸合成酶活性,表现为肉苁蓉中的分解酶类活性高于合成酶类活性,较高的分解酶类活性促进了蔗糖的分解,从而促进了糖分由寄主梭梭向肉苁蓉的不断转移。总体来看,肉苁蓉和寄主梭梭体内糖分的代谢主要以蔗糖合成酶为主,其它2种酶为辅协同参与调控。  相似文献

Carbohydrate accumulation is common in frost-resistant plants, and many enzymes participate in this process. The sugar content and expression levels of metabolic enzymes related to sugar biosynthesis in response to drops in temperature were measured in two cultivars of winter wheat (Triticum aestivum) with different cold tolerances. The results indicate that the two cultivars examined, Dongnongdongmai 1 and Jimai 22, accumulated high levels of carbohydrate before November 4 (above 0°C), and that accumulation decreased as temperatures fell. However, this decrease was more modest in Dongnongdongmai 1, which had a higher sugar content. Sucrose and fructose were the main soluble sugars, indicating an important role in freezing tolerance. Gene expression studies revealed that expression of the genes encoding chloroplastic enzymes was significantly upregulated in the tillering nodes. Expression upregulation of TaSS and TaTPT may be helpful for sugar accumulation before November 4.  相似文献

A positive relationship between feeding duration and meal size of avian nectarivores has often been assumed in earlier studies. We investigated whether feeding duration can be used as a surrogate for the amount of sugar solution ingested by white-bellied sunbirds, Cinnyris (Nectarinia) talatala. Feeding durations of sunbirds consuming three sucrose concentrations (10%, 20%, and 40% w/w) were measured using an infrared photodetection system, and the amounts consumed were recorded simultaneously by weighing the feeder throughout the experiment. For all three diet concentrations, a positive relationship was found between the time spent feeding per 30 min and the mass consumed. Therefore, feeding duration is demonstrated to be an index of the amount ingested on a particular sugar concentration. The rate of ingestion, however, depended on the sugar concentration, with the highest rate at the lowest concentration of 10% and the lowest rate at the 40% concentration. Less total time was spent feeding on the 20% solution than on the 10% solution, but time increased on the 40% diet because of viscosity effects. There appeared to be a weak relationship between feeding patterns and sex, but this was not significant, probably because of interindividual variation.  相似文献

黄原胶在采油工程中有重要的应用,但其难降解性质给采油工程带来很多问题。从塔里木油田胡杨木根部样品中分离得到1株黄原胶降解菌BIT-BJ001,对其发酵条件的研究表明,此黄原胶降解菌最适培养条件为:黄原胶0.3%,酵母粉0.5%,Na+浓度0.8%,Mg2+浓度0.8%,初始pH值为10,温度60℃。菌种BIT-BJ001降解黄原胶的能力与发酵时间、发酵液中还原糖浓度有关,发酵96 h,黄原胶降粘率达到最高,发酵液中还原糖浓度过高,将抑制菌株对黄原胶的降解。  相似文献

The effects of Tween 80 on the enzymatic hydrolysis of newspaper were tested. By monitoring sugar production it was found that the surfactant (0.1%) increased the rate and extent of cellulose saccharification. Consequently, the rate of enzyme usage in the hydrolysis reactor was improved by 33%. In addition, in the presence of surfactant the recovery of enzymes was higher. Analysis of the enzyme solution showed that with Tween 80 present larger fractions of enzyme remained in solution throughout hydrolysis. Thus, it appears that the surfactant hindered the immobilization of the enzymes on the substrates by reducing the strength of adsorption.  相似文献

The pasting and gel textural properties of corn starch in syrup at different concentrations were investigated by Rapid Visco Analyzer (RVA) and Texture profile analysis (TPA) tests. The results showed that the pasting temperatures of corn starch greatly increased, especially at higher sugar concentration. Increasing concentration of syrup caused an increase in peak, trough and final viscosity of corn starch. Peak viscosity and the disintegration rate of starch increased in the following order: fructose syrup> maltose syrup> glucose syrup. Increasing syrup concentration to 13%, 25% and 50% resulted in a lower retrogradation rate than the control. When the maltose syrup concentration increased to 50%, the retrogradation rate decreased to 14.30% from 33.38%. The highest hardness was observed when the syrup concentration was 25%. There was a particular low hardness when the concentration of syrup was 50%. The springiness of starch gels in syrup was similar at different concentrations.  相似文献

Self-diffusion coefficients for both components are reported for the highly concentrated aqueous solutions of some disaccharides and fructose as a function of temperature and concentration. These data are complemented by viscosity measurements. The disaccharides studied are sucrose, alpha,alpha-trehalose, allosucrose, and leucrose. Up to a sugar concentration of approximately 30% w/w the viscosity and the self diffusion coefficients of the four disaccharides are identical within experimental error for a given concentration and temperature. Water diffusion shows no differences in the four systems studied under these conditions. At higher concentrations significant differences are observed that become more pronounced with increasing temperature. Analysis of the data by the VTF equation yields the result that at a given concentration the self diffusion coefficients of the sugar Dc and the viscosity eta are described by identical ideal glass transition temperatures T0, while the diffusion of the water D(W) molecule decouples from these properties. Brewer's spent grains (BSG), the voluminous residue after mashing, contains on dry weight basis about 40–50 % polysaccharides. For the recovery of soluble carbohydrates from BSG different physical, thermal and enzymatic treatments were used to solubilize the polysaccharides in BSG. Heating by microwave radiation to 160 °C in the presence of 0.1 M HCl released 35 % of the material in the form of reducing sugar, indicating that about 80 % of the polysaccharides were hydrolyzed. Nevertheless, 0.1 M acetic acid will even solubilize 30 % of the material as oligosaccharides on a prolonged incubation when pretreating the spent grains by extrusion cooking. A combination of the enzymes Ceremix Plus MG and CelluPract AL 70 achieved an almost 25 % release of saccharides after 4 hrs of incubation at 50 °C. A combination of extrusion cooking and enzymatic hydrolysis seems to be a very promising procedure.  相似文献

The influence of agitation and aeration on growth and on production of glucose oxidase of Asp. niger has been studied. It was found that both rate of growth and glucose oxidase production was higher at an agitation speed of 700 rpm than at 460 rpm. Further increase in speed of agitation resulted in neither a higher rate of growth nor a higher glucose oxidase activity. Total glucose oxidase activity was highest in a medium containing 5% sugar (at an agitation speed of 700 rpm) and did not get higher when the sugar concentration of the medium was increased to 7%. When pure oxygen was bubbled through the culture the rate of growth of the culture (in the linear phase) was 95 mg. mycelial dry wt./100 ml./hr., and only 61 mg. when air was applied. The glucose oxidase activity of oxygenated culture was double the activity of aerated culture. Viscosity of the homogenized culture became higher with higher concentration of mycelia. The viscosity of oxygenated culture was found to be lower than that of aerated culture.  相似文献

Cracked corn was irradiated with gamma rays at 0-100 Mrad and the effects of the irradiation on sugar yield, susceptibility to enzymatic hydrolysis of starch, yeast growth, and alcohol production were studied. Gamma irradiation at 50 Mrad or greater produced a considerable amount of reducing sugar but little glucose. At lower dosages, gamma irradiation significantly increased the susceptibility of corn starch to enzymatic hydrolysis, but dosages of 50 Mrad or greater decomposed the starch molecules as indicated by the reduction in iodine uptake. About 12.5% reducing sugar was produced by amylase treatment of uncooked, irradiated corn. This amount exceeded the level of sugar produced from cooked (gelatinized) corn by the same enzyme treatment. The yeast numbers in submerged cultivation were lower on a corn substrate that was irradiated at 50 Mrad or greater compared to that on an unirradiated control. About the same level of alcohol was produced on uncooked, irradiated (10(5)-10(6) rad) corn as from cooked (121 degrees C for 30 min) corn. Therefore, the conventional cooking process for gelatinization of starch prior to its saccharification can be eliminated by irradiation. Irradiation also eliminated the necessity of sterilization of the medium and reduced the viscosity of high levels of substrate in the fermentation broth.  相似文献

The effects of sodium hexametaphosphate concentration, incubation temperature, and pH on the extraction of polysaccharide from soybean okara (soybean curd waste) were examined. The results suggest that the soybean polysaccharides were almost completely extracted with 50 times their volume of a 2% hexametaphosphate solution at 100°C for 2h, avoiding protein contamination. The viscosity, molecular weight, and sugar composition of the polysaccharides were compared with those of the commercially available samples. No differences were observed in the molecular weight or neutral sugar composition. However, it was found that the galacturonate distribution in the molecules of the two polysaccharides was different. The viscosity of the polysaccharide solution was changed by adding small amount of salt.  相似文献

本文探讨龙柚果实发育过程中果肉糖积累与蔗糖代谢相关酶活性的变化。结果表明,在龙柚果实发育过程中,3种可溶性糖含量同步上升,在果实膨大期和成熟期,以蔗糖积累为主。在龙柚糖积累过程中,蔗糖合成酶(SS)和蔗糖磷酸合成酶(SPS)活性较高;而蔗糖中性转化酶(NI)活性则随着蔗糖的积累而降低。  相似文献

The control of crop emptying in foraging honeybees was investigated in individuals trained to collect defined amounts of sugar solutions. Following feeding, they were dissected after fixed periods of time in order to measure crop content and haemolymph sugar titers. Between feeding and dissection, the metabolic rate of every investigated forager was measured using open-flow respirometry, so as to assess the effects of both food quality (concentration, molarity and viscosity of the fed sugar solution) and food quantity on the transport rate through the proventriculus. The sugar transport rate through the proventriculus was observed to be mainly dependent on the metabolic expenditure of the individual. Bee foragers were able to precisely adjust the sugar transport rate to their metabolic rates, but under certain conditions, an excess of sugars was transported through the proventriculus, more than needed to cover the bee’s energetic demands. This excess depended on the nutritive value and quantity of the fed sugar solution, and on the time after feeding. It did not depend on the metabolic rate of the bee, the molarity, or the viscosity of the fed sugar solution. As long as the bees did not exhaust their crop contents, the haemolymph sugar titers were unaffected by this excess amount transported, by the time after feeding, the concentration and the viscosity of the fed sugar solution. For all feeding conditions assayed, the haemolymph trehalose titer remained constant, while the titers of other haemolymph sugars varied. It is suggested that the trehalose concentration in the haemolymph is regulated in honeybees, and that it represents the controlled variable in the feedback loop responsible for the transport rate through the proventriculus.  相似文献
