A field experiment was conducted to examine the effect of the arbuscular mycorrhizal fungus Glomus macrocarpum and salinity on growth of Sesbania aegyptiaca and S. grandiflora. In the salt-stressed soil, mycorrhizal root colonisation and sporulation was significantly higher in AM-inoculated than in uninoculated plants. Mycorrhizal seedlings had significantly higher root and shoot dry biomass production than non-mycorrhizal seedlings grown in saline soil. The content of chlorophyll was greater in the leaves of mycorrhiza-inoculated as compared to uninoculated seedlings. The number of nodules was significantly higher in mycorrhizal than non-mycorrhizal plants. Mycorrhizal seedling tissue had significantly increased concentrations of P, N and Mg but lower Na concentration than non-mycorrhizal seedlings. Under salinity stress conditions both Sesbania sp. showed a high degree of dependence on mycorrhizae, increasing with the age of the plants. The reduction in Na uptake together with a concomitant increase in P, N and Mg absorption and high chlorophyll content in mycorrhizal plants may be important salt-alleviating mechanisms for plants growing in saline soil.  相似文献

Arbuscular mycorrhizal (AM) fungi influence the expression of defence-related genes in roots and can cause systemic resistance in plants probably due to the induced expression of specific defence proteins. Among the different groups of defence proteins, plant food allergens were identified. We hypothesized that tomato-allergic patients differently react to tomatoes derived from plants inoculated or not by mycorrhizal fungi. To test this, two tomato genotypes, wild-type 76R and a nearly isogenic mycorrhizal mutant RMC, were inoculated with the AM fungus Glomus mosseae or not under conditions similar to horticultural practice. Under such conditions, the AM fungus showed only a very low colonisation rate, but still was able to increase shoot growth of the wild-type 76R. Nearly no colonisation was observed in the mutant RMC, and shoot development was also not affected. Root fresh weights were diminished in AM-inoculated plants of both genotypes compared to the corresponding controls. No mycorrhizal effects were observed on the biomass and the concentration of phosphate and nitrogen in fruits. Real-time quantitative polymerase chain reaction analysis revealed that six among eight genes encoding for putative allergens showed a significant induced RNA accumulation in fruits of AM-colonised plants. However, human skin reactivity tests using mixed samples of tomato fruits from the AM-inoculated and control plants showed no differences. Our data indicate that AM colonisation under conditions close to horticultural practice can induce the expression of allergen-encoding genes in fruits, but this does not lead necessarily to a higher allergenic potential.  相似文献

A pot experiment was conducted to examine the effect of arbuscular mycorrhizal fungus, Glomus fasciculatum, and salinity on the growth of Acacia nilotica. Plants were grown in soil under different salinity levels (1.2, 4.0, 6.5, and 9.5 dS m−1). In saline soil, mycorrhizal colonization was higher at 1.2, 4.0, and 6.5 dS m−1 salinity levels in AM-inoculated plants, which decreased as salinity levels further increased (9.5 dS m−1). Mycorrhizal plants maintained greater root and shoot biomass at all salinity levels compared to nonmycorrhizal plants. AM-inoculated plants had higher P, Zn, and Cu concentrations than uninoculated plants. In mycorrhizal plants, nutrient concentrations decreased with the increasing levels of salinity, but were higher than those of the nonmycorrhizal plants. Mycorrhizal plants had greater Na concentration at low salinity levels (1.2, 4.0 dS m−1), which lowered as salinity levels increased (6.5, 9.5 dS m−1), whereas Na concentration increased in control plants. Mycorrhizal plants accumulated a higher concentration of K at all salinity levels. Unlike Na, the uptake of K increased in shoot tissues of mycorrhizal plants with the increasing levels of salinity. Our results indicate that mycorrhizal fungus alleviates deleterious effects of saline soils on plant growth that could be primarily related to improved P nutrition. The improved K/Na ratios in root and shoot tissues of mycorrhizal plants may help in protecting disruption of K-mediated enzymatic processes under salt stress conditions.  相似文献

Plant hormones, including auxins, might be signals during the establishment of an arbuscular mycorrhizal (AM) symbiosis. Here, we report on the concentrations of three auxins native to nasturtium ( Tropaeolum majus L.) during early AM development. Indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), and phenylacetic acid (PAA) were previously identified as endogenous compounds in this species by full-scan gas chromatography–mass spectrometry. All auxinic compounds were influenced by AM colonization but showed completely different patterns. At very early stage, free IAA and IBA were lower in infected than in control roots, whereas PAA concentration was higher in infected roots than in controls. At later stages, PAA was reduced in colonized roots, whereas, especially, IBA was increased in colonized roots compared with controls. Measurement of total auxins confirmed a complex regulation pattern for the three compounds. In hyphae of Glomus intraradices , none of the auxins was detectable. Biosynthesis of the three auxins was measured using heavy labeled isotopes as precursors in control and AM-inoculated roots. While not much difference was found in the IAA labeling pattern between controls and AM-inoculated roots at both time points, IBA synthesis was slightly higher in AM-inoculated roots. Double labeling experiments showed that two distinct pathways, a tryptophan-dependent and a tryptophan-independent biosynthetic pathway contribute to the synthesis of IAA in T. majus roots. Because T. majus is difficult to genetically manipulate, we have used tobacco plants transformed with the auxin-inducible promoter GH3 fused to the β-glucuronidase (GUS) reporter gene to investigate whether AM structures would co-localize to cells harboring the auxin-inducible promoter. Although the GUS activity increased significantly in AM-inoculated roots, there was no obvious correlation between GH3::GUS expression and fungal structures.  相似文献

A pot-culture experiment was carried out to investigate the effect of arbuscular mycorrhizal (AM) fungus (Glomus macrocarpum Tul. and Tul.) on plant growth and Cd2+uptake by Apium graveolens L. in soil with different levels of Cd2+. Mycorrhizal (M) and non-mycorrhizal (NM) plants were grown in soil with 0, 5, 10, 40 and 80 Cd2+ mg kg−1soil. The infectivity of the fungus was not affected by the presence of Cd2+ in the soil. M plants showed better growth and less Cd2+ toxicity symptoms. Cd2+ root : shoot ratio was higher in M plants than in NM plants. These differences were more evident at highest Cd2+ level (80 mg kg−1 soil). Chlorophyll a and chlorophyll b concentrations were significantly higher in AM-inoculated celery leaves. The dilution effect due to increased biomass, immobilization of Cd2+ in root and enhanced P-uptake in M plants may be related to attenuation of Cd2+toxicity in celery.  相似文献

Summary T-DNA circularization is one of the molecular events specifically induced in agrobacterial cells upon their infection of dicotyledonous plant cells. We developed a seedling co-cultivation procedure to determine whether or not monocotyledonous plants have the ability to induce T-DNA circularization and vir gene expression. Co-cultivation of Agrobacterium tumefaciens with seedlings of dicotyledonous plants showed that the circularization event takes place efficiently. The exudates and extracts of the seedlings also effectively induced T-DNA circularization and vir gene expression, indicating that dicotyledonous seedlings contain diffusible factors capable of inducing these molecular events. In contrast, neither T-DNA circularization nor vir gene expression was detectable when Agrobacterium was incubated with seedlings of monocotyledonous plants. Supplementing with acetosyringone, a known inducer of vir gene expression and T-DNA circularization, resulted in the induction of circularization during co-cultivation with monocotyledonous seedlings. These results indicate that the seedlings of monocotyledonous plants have no detectable amounts of diffusible inducers, unlike dicotyledonous seedlings. Therefore, it is unlikely that the vir genes are expressed in Agrobacterium inoculated in monocotyledonous plants. This may be one of the blocks in tumorigenesis of monocotyledonous plants by Agrobacterium.  相似文献

Production of Jatropha curcas as a biodiesel feedstock on marginal lands is growing rapidly. Biomass production on these lands is limited. Hence, the objective of this study was to evaluate the effect of arbuscular mycorrhiza (AM) fungi and salinity (0.1, 0.2, 0.3, 0.4, and 0.5% NaCl) on (1) seedling growth, leaf relative water content (RWC), lipid peroxidation, solute accumulation (proline and sugars), and photosynthetic pigments (Chl a and b) of Jatropha; (2) mycorrhizal colonization (%) and mycorrhizal dependency (MD) of Jatropha; and (3) glomalin content (Bradford reactive soil protein) in soil. Increased soil salinity significantly (P < 0.05) decreased AM root colonization (r 2 = 0.98) of AM-inoculated plants and decreased survival (r 2 = 0.93) and growth (shoot length, r 2 = 0.89; tap root length, r 2 = 0.93; shoot diameter, r 2 = 0.99; shoot dry weight, r 2 = 0.92; and root dry weight, r 2 = 0.92) of non-AM-inoculated Jatropha. Under salt stress, AM-inoculated Jatropha plants had greater dry weight of shoots and roots, better leaf water status, less leaf membrane damage (low lipid peroxidation activity), higher solute (proline and sugars), and higher leaf chlorophyll concentrations than non-AM-inoculated plants. The mycorrhizal dependency (MD) of Jatropha increased from 12.13 to 20.84% with salinity (0–0.4% NaCl). Root AM colonization (%) and glomalin content in soil were negatively correlated with salinity (P < 0.05, r = −0.95). We conclude that inoculation with AM fungi lessens the deleterious effect of salt stress on seedling growth parameters under salt levels up to 0.5% NaCl (electrical conductivity of 7.2 dS m−1). Inoculation of Jatropha seedlings with AM fungi can promote the establishment of Jatropha under NaCl-induced stress.  相似文献

The Nicotiana tabacum transgenic plants expressing a Cucurbita pepo antisense PHYA RNA were obtained. The seedlings of transgenic tobacco with reduced phytochrome A (PHYA) content displayed decreased sensitivity to continuous broad-band far-red radiation (λ > 680 nm). Under far-red irradiance transgenic seedlings showed less elongation of the hypocotyls, more rapid plastid development, more chlorophyll accumulation, less repression of lightdependent NADPH:protochlorophyllide oxidoreductase than wild-type plants that was in accordance with PHYA control of plant development. Dynamics of the far-red radiation dependent changes in low temperature chlorophyll fluorescence spectra for the transgenic and wild-type seedlings were consistent with the more rapid formation of photosynthetic apparatus in the seedlings with reduced PHYA.  相似文献

Plants respond to feeding by herbivorous insects by producing volatile organic chemicals, which mediate interactions between herbivores and plants. Yet, few studies investigated whether such plant responses to herbivory differ between historical host and novel plants. Here, we investigated whether herbivory by the pine weevil Hylobius abietis causes a release of volatile organic chemicals from a novel tree Pinus brutia and compared the relative amounts of volatiles released from herbivore's historical hosts and P. brutia. We collected volatiles emitted from P. brutia seedlings that were either subjected to feeding by H. abietis or no feeding. Our results indicated that feeding increased emission of volatile compounds, composed of monoterpenes and sesquiterpenes, and that the emission was several fold higher in the damaged seedlings than in undamaged seedlings. In particular, emission of monoterpenes and sesquiterpenes increased by 4.4‐and 10‐fold in the damaged plants, respectively. Strikingly, individual monoterpenes and sesquiterpenes showed much greater dissimilarity between damaged and undamaged seedlings. Furthermore, several minor monoterpenes showed negative relationships with the weevil gnawed area. We discussed these results with the results of previous studies focused on historical host plants of H. abietis and hypothesized the ecological relevance and importance of our results pertaining relevance to the plant–herbivory interactions.  相似文献

Herbivore‐induced changes in plants have been widely viewed as defensive responses against further insect attack. However, changes in plants as a consequence of herbivore feeding can elicit various responses in herbivores; these are variable, context dependent, and often unpredictable. In this laboratory study, the responses of Thrips tabaci Lindeman (Thysanoptera: Thripidae) to volatiles emitted by intact and herbivore‐damaged or mechanically damaged cotton seedlings [Gossypium hirsutum L. (Malvaceae)] were investigated in dual‐choice olfactometer assays. Thrips tabaci showed increased attraction to seedlings subject to foliar mechanical damage and those with foliar damage inflicted by conspecifics or Tetranychus urticae Koch (Acari: Tetranychidae), upon which it preys. However, T. tabaci did not discriminate between intact seedlings and those with foliar damage inflicted by Helicoverpa armigera Hübner (Lepidoptera: Noctuidae), two other species of thrips, Frankliniella schultzei Trybom and Frankliniella occidentalis Pergrande (Thysanoptera: Thripidae), or those with root damage inflicted by Tenebrio molitor L. (Coleoptera: Tenebrionidae). Attraction of T. tabaci was also affected by herbivore density on damaged plants. That is, seedlings damaged by higher densities of T. urticae or T. tabaci were more attractive than seedlings damaged by lower densities of the corresponding arthropod. Although attracted to plants damaged by conspecifics or T. urticae, T. tabaci showed greater attraction to seedlings damaged by T. urticae than to seedlings damaged by conspecifics. Results are discussed in the context of the responses of F. schultzei and F. occidentalis to herbivore‐induced cotton seedlings, highlighting the complexity, variability, and unpredictability of the responses of even closely related species of insects to plants under herbivore attack.  相似文献

Summary Seedlings of the Caesalpinoids Hymenaea courbaril, H. parvifolia and Copaifera venezuelana, emergent trees of Amazonian rainforest canopies, and of the Araucarian conifers Agathis microstachya and A. robusta, important elements in tropical Australian rainforests, were grown at 6% (shade) and 100% full sunlight (sun) in glasshouses. All species produced more leaves in full sunlight than in shade and leaves of sun plants contained more nitrogen and less chlorophyll per unit leaf area, and had a higher specific leaf weight than leaves of shade plants. The photosynthetic response curves as a function of photon flux density for leaves of shade-grown seedlings showed lower compensation points, higher quantum yields and lower respiration rates per unit leaf area than those of sun-grown seedlings. However, except for A. robusta, photosynthetic acclimation between sun and shade was not observed; the light saturated rates of assimilation were not significantly different. Intercellular CO2 partial pressure was similar in leaves of sun and shade-grown plants, and assimilation was limited more by intrinsic mesophyll factors than by stomata. Comparison of assimilation as a function of intercellular CO2 partial pressure in sun- and shade-grown Agathis spp. showed a higher initial slope in leaves of sun plants, which was correlated with higher leaf nitrogen content. Assimilation was reduced at high transpiration rates and substantial photoinhibition was observed when seedlings were transferred from shade to sun. However, after transfer, newly formed leaves in A. robusta showed the same light responses as leaves of sun-grown seedlings. These observations on the limited potential for acclimation to high light in leaves of seedlings of rainforest trees are discussed in relation to regeneration following formation of gaps in the canopy.  相似文献

Maize chlorotic mottle virus (MCMV) is one of the co‐infection pathogens that cause corn (maize) lethal necrosis, but the transmission mechanism of MCMV is not yet clear. In order to determine the ability of western flower thrips (Frankliniella occidentalis; WFT) to transmit MCMV, imported maize seeds from Thailand were germinated in an insect‐free greenhouse and the seedlings were tested for the transmission by WFT of chlorotic mottle virus disease. The thrips (WFT), starved for 48 h then allowed to feed for 30 min on maize plants infected with MCMV or asymptomatic maize plants, were transferred to healthy seedlings. After 35 days, the seedlings with WFT from diseased maize plants showed chlorotic mottle symptoms, whereas seedlings with WFT from asymptomatic maize plants remained healthy. A single band of 711 bp was amplified by RT‐PCR using primers MCMV‐F/MCMV‐R from the MCMV‐infected plants and WFT collected from the diseased plants. Sequencing of the amplified product and further sequence comparison indicated that the two viruses from both sources showed 99% similarity of nucleotides and they should be regarded as identical. In addition, isometric particles c. 30 nm in diameter, characteristic of MCMV, were found in the WFT samples from diseased maize plants. Thus, it is concluded that WFT transmits MCMV. Our findings suggest that corn lethal necrosis disease can be controlled or minimized by the eradication of WFT from the field or greenhouses.  相似文献

Internode extension-growth responses to neighbouring plants and to red to far-red ratios (R:FR) were investigated in wild-type (WT) and aurea (au)-mutant seedlings of tomato grown under natural radiation. The genomic location of the au mutant is not known, but one of its consequences is the reduced phytochrome level. In WT seedlings, internode growth was promoted by the presence of non-shading neighbours reflecting far-red light (FR), the shade of a tall canopy, FR provided as a supplement during the photoperiod, and FR pulses either provided at the end of the day or delayed into the dark period. Supplementary FR during the photoperiod also promoted growth in herbicide-treated partially bleached WT seedlings. The au mutant showed higher background extension-growth rates, but only responded to the most severe treatments: deep shade light and very low R:FR at the end of the day, i.e. au-mutant seedlings were less sensitive than WT seedlings to R:FR signals. Wild-type seedlings were transferred from the glasshouse to a growth room and exposed to white light with two levels of phytochrome-absorbable radiation but similar phytochrome photoequilibria and radiation for photosynthesis. The plants exposed to the lowest level showed a transient increase of internode extension growth rate and a simultaneous reduction of response to FR pulses, i.e. reproduced some of the features of au-mutant seedlings. Phytochrome itself could set the degree of response to Pfr during neighbour detection.  相似文献

The objective of this study was to investigate the effects of arbuscular mycorrhizal fungus (AMF) inoculation on growth and drought tolerance of Poncirus trifoliata seedlings. The seedlings were inoculated with or without Glomus mosseae before exposure to a short-term (3 days) water depletion, and relevant physiological and biochemical parameters (plant height, chlorophyll content, relative water content, activity of antioxidant enzymes) and expression patterns of several stress-responsive genes were examined. Inoculation with G. mosseae led to growth promotion of the seedlings, as revealed by larger plant height and higher relative water and chlorophyll contents. When subjected to drought treatment, the AMF-inoculated (AM) plants showed better tolerance than the nonmycorrhizal (NAM) plants. Under drought, the AM plants exhibited higher level of proline and activity of two antioxidant enzymes, superoxide dismutase (SOD) and peroxidase (POD). In addition, mRNA abundance of four genes involved in reactive oxygen species homeostasis and oxidative stress battling was higher in the AM plants when compared with the NAM plants. These results indicate that AMF inoculation stimulated growth and enhanced drought tolerance of the seedlings, which may be due to activation of an arsenal of physiological, biochemical and molecular alterations.  相似文献

Agrobacterium tumefaciens harbouring the Ti plasmid incites crown gall tumor on dicotyledonous species. Upon infection of these plants, T-DNA in the Ti plasmid is transferred by unknown mechanisms to plant cells to be integrated into nuclear DNA. WhenAgrobacterium is incubated with protoplasts or seedlings of dicotyledonous plants, circulation of T-DNA and expression ofvir (virulence) genes on the Ti plasmid are induced. The circularization event is efficiently induced by mesophyll protoplasts of tobacco which are highly competent for transformation by the T-DNA, and is also induced by diffusible phenolic compounds excreted from the protoplasts. The circularization and formation of crown gall both require the expression of thevirD locus, one of the induciblevir genes. These results suggest that the circularization of T-DNA reflects one of steps of the T-DNA transfer during formation of crown gall. In contrast to dicotyledonous plants, monocotyledonous plants are thought to be unresponsive to infection byAgrobacterium. We showed that monocotyledonous plants do not excrete diffusible inducers for the expression ofvir genes, while they contain a novel type of a signal substance(s). This inducer is not detected in the exudates of seedlings of monocotyledonous plants, but is found in the extracts from the seedlings, and also those from the seeds, bran and germ of wheat and oats. This finding suggests that T-DNA processing, and possibly its transfer, should take place whenAgrobacterium invades seedlings and seeds of monocotyledonous plants. 以根结线虫侵染的文冠果一年生苗木为材料,观察分析苗木根系的形态和显微结构,植株生长发育以及主要矿质养分在苗木中的分布特性,并分析不同矿质元素与线虫侵染的关联关系,以揭示根结线虫对文冠果苗木生长发育的影响机制。结果表明:(1)受根结线虫侵染的文冠果苗木根系形成根结,依据根结发生程度分为0(对照,正常植株)、2、3、4级;与对照相比,具有根结的文冠果苗木根系解剖结构特征主要表现为皮层相对较厚,木质部排列扭曲,导管较少,射线数目较少且分布不均匀;韧皮细胞内含物明显较多;多个巨细胞及细胞空腔主要存在于韧皮部。(2)具根结的苗木株高和地径较对照均增加,且株高增幅达显著水平(P<0.05)。(3)具根结苗木根、茎、叶部位的N、P含量均较对照下降,根部降幅达显著水平(P<0.05),其中具2~4级根结的根部N含量分别较对照显著下降7.8%、16.0%和29.5%,P含量分别显著下降15.6%、7.1%和43.3%;根部Fe含量、Zn含量显著上升,2~4级根结根系中Fe含量较对照分别增加1.56倍、0.81倍和3倍,Zn含量分别增加1.11倍、1.56倍和1.78倍;具3和4级根结的苗木根系中K含量较对照显著增加(P<0.05),具2和4级根结的苗木叶片中K含量较对照分别显著下降61.5%和47.0%。(4)苗木根部、茎部N元素含量对于根结线虫侵染的响应最明显,且其含量随侵染程度的增大而降低,叶部Mg元素含量与根结线虫侵染率、K元素含量与整株苗木生物量均呈显著负相关关系(P<0.05)。该研究为根结线虫对文冠果苗木养分吸收利用及运转的影响提供了一定的理论依据。  相似文献

Cuscuta is a stem holoparasitic plant without leaves or roots, parasitizing various types of host plants and causing major problems for certain crops. Cuscuta is known as a generalist and, thus, must have unique parasite strategies to cope with different host plants. For elucidating metabolic responses and mechanisms of parasitization, metabolomic approaches using GC/MS were applied. We compared five stages of Cuscuta japonica: early stage seedlings, with far red light (FR) cue, with contact signal, haustorium induced seedlings by both signals and adult plant parasites on host plants. Sugars, amino acids, organic acids, nucleic acids, and polyols were identified from the polar phase fraction. The apical part contained metabolite profiles different from the haustorium induced part or the basal part. Amino acid and some organic acids were up-regulated for haustorium induction but decreased after parasitization. After attachment to different host plants, metabolite profiles of Cuscuta japonica changed dramatically due to the absorption of specific host plant metabolites such as pinitol. Cuscuta seedlings attached to pinitol rich host plants contained more pinitol and showed different profiles from those attached to plants having less or lacking pinitol.  相似文献

Barley (Hordeum vulgare), corn (Zea mays), bean (Phaseolus vulgaris), and radish (Raphanus sativus) seedlings were continuously irradiated under a lighting device for 5–10 d at an increased ultraviolet (UV)-B fluence rate. In their growth parameters, composition, and leaf surface, these four species responded differently to the increased UV-B exposure. Bean seedlings suffered the most serious effects, radish and barley less, and corn was hardly influenced at all. In all plant species, the fresh weight, the leaf area, the amounts of chlorophylls, carotenoids and the galactolipids of the chloroplasts were reduced. The lipid content of the corn and bean seedlings also diminished. But all the irradiated plants showed a rise in their protein content compared to the control plants. The content of flavonoids increased in barley and radish seedlings by about 50%. The effects on growth parameters and composition were more extensive with increasing UV-B fluence rates, at least as shown in the case of barley seedlings. The fresh weights fell proportionally with the chlorophylls and carotenoids. In contrast, the flavonoid content of barley leaves rose parallel to the increasing UV-B fluence rates and reached 180% of the value in the control plants with the highest UV-B fluence rate. Scorching appeared regularly in the form of bronze leaf discoloration at the highest UV-B fluence rates.
