 Although Pinaceae and Betulaceae have been reported to contain Glomus–type root endophytes, its ecological importance and the conditions influencing this symbiosis are poorly understood. Seedlings of Abies lasiocarpa, Alnus rubra, Pinus contorta, Pinus ponderosa, Pseudotsuga menziesii, and Tsuga heterophylla were inoculated with Glomus intraradices to determine the vesicular-arbuscular mycorrhizae (VAM) development and responsiveness of these hosts. The role of companion VAM host plants on mycorrhizal colonization and nutrient uptake by Pseudotsuga menziesii was also examined by growing seedlings of Pseudotsuga menziesii in dual culture with VAM hosts Thuja plicata or Calamagrostis rubescens. After 8 weeks, no seedlings were colonized. At 16 weeks, 8 of 17 Thuja plicata seedlings grown with Pseudotsuga menziesii and all 18 inoculated Thuja plicata seedlings grown alone were colonized with vesicles and hyphae. Two of 17 inoculated Pseudotsuga menziesii seedlings grown in dual culture with Thuja plicata were colonized with abundant vesicles and hyphae. No ectomycorrhizal seedlings grown in monoculture were colonized. At 9 months, all 10 Calamagrostis rubescens and all 10 inoculated Pseudotsuga menziesii seedlings grown in dual culture were colonized by vesicles and hyphae. Two of 10 inoculated Pseudotsuga menziesiiand 1 of 10 inoculated Pinus ponderosa seedlings grown in monoculture were similarly colonized. The mean phosphorus content in the needles of colonized Pseudotsuga menziesii seedlings grown with Calamagrostis rubescens was about twice as high as in noncolonized Pseudotsuga menziesiiseedlings grown with Calamagrostis rubescens. Tissue nitrogen did not differ between these treatments. The results show that Glomus intraradices colonization of Pinaceae is most successful when a VAM host is present, although some vesicular colonization of Pinaceae occurred in the absence of a VAM host. In a study of host-parasite interrelationship and the mechanism of parasitization, ribonuclease activity was determined in Cuscuta reflexa Roxb, and in infected and control host plants of Lantana camara L. In the haustoria-bearing region of Cuscuta , the concave half of the vine bearing the haustorial site, always showed significantly higher RNase activity than the convex half, irrespective of the differing enzyme activity of the parasite tissue growing on different hosts — Brassica campestris L., Helianthus animus L., Lantana camara L., Medicago saliva L. and Solatium nigrum L. The uninfected host branch of L. camara showed the maximum specific RNase activity in the apical region which decreased toward the base, while the infected host branch showed minimum specific RNase activity in the apical region, gradually increasing towards the infected region.  相似文献

 Vesicular-arbuscular mycorrhizal (VAM) colonization can alter transpiration of host leaves, but scientists remain unclear about the mechanisms involved. We tested whether intact root systems were required to observe effects of root colonization by Glomus intraradices on leaf transpiration, or whether some VAM influence resided in leaves even after they were detached from root systems. We measured the transpiration of detached leaves of VAM and nonmycorrhizal plants exposed to different levels of several substances known to influence stomata locally or act in whole-plant regulation of transpiration: abscisic acid, calcium, phosphorus, and hydrogen ions. In rose, some VAM influence on transpiration resided in leaves, even after they had been separated from their root systems. However, removing leaves from their root systems eliminated the VAM influence on stomatal behavior of cowpeas. Accepted: 22 June 1998  相似文献

The genetic basis of vascular differentiation and function isrelatively poorly understood, partly due to the difficulty ofscreening for mutants defective in internal vascular tissues.Here we present an approach based on a predicted increase invascular-related gene expression in response to an auxin transportinhibitor-induced vascular overgrowth. We used microarray analysesto identify 336 genes that were up-regulated 2-fold in shoottissues of Arabidopsis thaliana showing vascular overgrowth.Promoter–marker gene fusions revealed that 38 out of 40genes with 4-fold up-regulation in vascular overgrowth tissueshad vascular-related expression in transgenic Arabidopsis plants.Obtained expression patterns included cambial tissues and differentiatingxylem, phloem and fibers. A total of 15 genes were found tohave vascular-specific expression patterns in the leaves and/orinflorescence stems. This study provides empirical evidenceof the efficiency of the approach and describes for the firsttime the in situ expression patterns of the majority of theassessed genes.  相似文献

Cells capable of photosynthesis in the parasitic angiosperm Cuscuta reflexa Roxb. (dodder) are highly localized. Immunolocalization of ribulose-1,5 bisphosphate carboxylase-oxygenase (Rubisco) and autofluorescence of chlorophyll in transverse sections of stems showed that they were largely restricted to a band of cells adjacent to the vascular bundles, consequently, the concentrations of Rubisco and chlorophyll were low per unit area or fresh weight. When 14CO2 was supplied to stem segments of C. reflexa it preferentially accumulated in these cells adjacent to the vasculature. Although the conductance for CO2 movement to the cells containing chlorophyll and Rubisco was very low, both the light reactions and dark reactions of photosynthesis appeared to be functional. De-epoxidation of the xanthophyll-cycle pigments after exposure to high light, and the chlorophyll fluorescence parameters, photochemical quenching (qP), non-photochemical quenching (NPQ) and the quantum efficiency of photosystem II (φPSII) responded normally to changes in photon flux density, indicating functional light-driven electron transport. The response of CO2 exchange to photon flux density followed a typical hyperbolic curve, and positive rates of CO2 fixation occurred when external CO2 was increased to 5%. We propose that CO2 for carbon assimilation is derived from internally respired CO2 and that this layer of photosynthetic cells makes a positive contribution to the carbon budget of C. reflexa. Summary The mechanism of parasitism of Cuscuta, especially the absorption of nutrients from its host, is not clear. As it might be connected with the function of plant hormones, the endogenous levels of all hormone groups in the parasite, Cuscuta reflexa, and its host, Vicia faba, were investigated. Since the content of auxins, gibberellins and cytokinins is higher in the host than in the parasite, there is no indication that any of these phytohormones is involved in the absorption of nutrients by Cuscuta. However, the content of growth inhibitors, especially free abscisic acid, is much higher in Cuscuta than in the host. There is a gradient of abscisic acid with a maximum in the basal, haustoria-bearing stem region in which the transfer of nutrients from host to parasite occurs. The high content of abscisic acid within the parasite may be a causal connection with the parasitic absorption of nutrients from host sieve tubes.This publication is respectfully dedicated to the academician Prof. Dr. A. The present investigation examines (1) whether the external VAM mycelium survives winter freezing to act as a source of inoculum in the spring, and (2) whether soil disturbance reduces the infectivity of the external VAM mycelium following freezing of the soil. Sealed pouches of fine nylon mesh were placed in pots containing soil inoculated with a Glomus species. The mesh was impervious to roots but not to hyphae. Following two 3-week growth cycles of maize in the pots, the pouches were transplanted to the field. Pouches were removed from the field once during the 4 months when the soil was frozen, and once after spring thaw. Measurements were made of VAM spore density, hyphal length and viability in the pouches. Bioassays for infectivity were conducted on all pouches. Some VAM hyphae survived freezing and remained infective following winter freezing, in the absence of plant roots. Soil disturbance did not reduce the infectivity of hyphae following exposure to freezing temperatures. We observed a change in the distribution of viable cytoplasm within hyphae over winter, which we hypothesize represents an adaptation allowing hyphae to survive freezing temperatures. We suggest that the effect of disturbance on hyphal infectivity may be related to this seasonal change in the distribution of hyphal viability.  相似文献

Cytokinins in the different regions of Cuscuta reflexa Roxb. vine have been extracted and assayed using the Xanthium cotyledon bioassay with N6-benzylaminopurine as the reference cytokinin. Maximum cytokinin level was found in the haustoria-bearing region with the gradient decreasing towards the apex. The cytokinin content of the apex was less than 1% of that in the whole haustorial region. Interestingly, the concave half of the vine contained about forty times more cytokinin in the haustorial region than the corresponding convex half. The high concentration of cytokinin or cytokinin-like substances in the haustoria-bearing site in Cuscuta might be due either to a high RNase activity in the region or translocation from the host to parasite or both.  相似文献

A genetic map covering 615 cM in 12 linkage groups was assembled based on 92 RFLP and AFLP markers segregating in a population of 107 doubled haploid lines (DH lines) of Brassica oleracea. The DH-line population was obtained through microspore culture from the of two homozygous parents: DH-line Bi derived from the cabbage landrace Bindsachsener, and DH-line Gr from broccoli cv ‘Greenia’. Sixty-five percent of the loci, and in some cases complete linkage groups, displayed distorted segregation ratios, a frequency much higher than that observed in populations of the same species. DH-line Bi was resistant to clubroot, which is caused by a Dutch field isolate of Plasmodiophora brassicae. Resistance in the DH-line population was determined in two ways: by assigning symptom grades to each plant, and by measuring the fresh weights of the healthy and affected parts of the root system of each plant. Using a multiple QTL mapping approach to analyze the fresh weight data, we found two loci for clubroot resistance; these were designated pb-3 and pb-4. The additive effects of these loci were responsible for 68% of the difference between the parents and for 60% of the genetic variance among DH-line means. Also, indications for the presence of two additional, minor QTLs were found. Analysis of symptom grades revealed the two QTLs pb-3 and pb-4, as well as one of the two minor QTLs indicated by analysis of the fresh weight data. The plant cytoskeleton has been implicated in a variety of morphogenetic events in higher plants. Most of this work, however, has concentrated on epidermal cells or primary tissues. We have investigated the cortical microtubular (CMT) and microfilament (MF) components of the cytoskeleton in a secondary tissue  –  active vascular cambium of Aesculus hippocastanum L. (horse-chestnut)  –  and followed the changes in these components during the early stages of differentiation of fusiform cambial derivatives to axial elements of the secondary vascular system. A correlative approach was used employing indirect immunofluorescence microscopy of α-tubulin on 6 μm sections, and transmission electron microscopy of 60 nm sections. The study has demonstrated a rearrangement of the CMT cytoskeleton, from random to helical, as fusiform vascular cambial cells begin to differentiate as secondary phloem vascular tissue. A similar CMT rearrangement is seen as fusiform cambial cells begin to differentiate as secondary xylem fibres. This rearrangement is interpreted as evidence of determination of cambial derivatives towards vascular development. Axially-oriented MF bundles are present in fusiform cambial cells and their axial orientation is retained in the vascular derivatives at early stages of their development even though the CMTs have become rearranged. Understanding the genetic mechanisms for the phenotypic plasticity and developmental instability of a quantitative trait has important implications for breeding and evolution. Two clonally replicated plantations of two 3-generation inbred pedigrees derived from the highly divergent species Populus trichocarpa and P. deltoides were used to examine the genetic control of macro- and micro-environmental sensitivities and their genetic relationships with the trait mean across two contrasting environments. For all stem-growth traits studied, the trait mean had a higher broad-sense heritability (H2) level than macroenvironmental sensitivity, both with much higher values than microenvironmental sensitivity. Genetic correlation analyses indicated that the trait mean was more or less independent of macro- or micro-environmental sensitivity in stem height. Thus, for this trait, the genetic difference in response to the two environments might be mainly due to epistasis between some regulatory loci for plasticity and loci for trait mean. However, for basal area and volume index, pleiotropic loci might be more important for their genetic differences between the two environments. No evidence was found to support Lerner’s (1954) homeostasis theory in which macro- or micro-environmental sensitivity is the inverse function of heterozygosity. Nutrient availability limits plant production acrossa wide range of terrestrial ecosystems. In this studyof a Californian annual grassland community, theinfluence of arbuscular mycorrhizal (AM) associationson plant nutrient acquisition was investigated usingfactorial combinations of nitrogen (N), phosphorus (P)and benomyl fungicide. N additions resulted in asignificant increase in shoot biomass demonstratingthat plant productivity in these soils was N-limited. The effect of P additions consistently depended onfungicide treatment. In the absence of benomyl, shootP accumulation was unaltered by P addition. In thepresence of benomyl, P addition significantlyincreased shoot P accumulation and was associated witha consistent trend towards enhanced shoot biomass.The induction of P-deficiency with benomylapplication suggests that effective P acquisition byAM may contribute to the strong N-limitation ofproduction observed in many terrestrial ecosystems.  相似文献

We report here the RFLP mapping of quantitative trait loci (QTLs) which affect some important agronomic traits in cultivated rice. An anther culture-derived doubled-haploid (DH) population was established from a cross between indica and japonica rice varieties. A molecular linkage map comprising 137 markers was constructed based on this population which covered the rice genome at intervals of 14.8 cM on average. The linkage map was used to locate QTLs for such important agronomic traits as heading date, plant height, number of spikelets per panicle, number of grains per panicle, 1 000-grain weight and the percentage of seed set, by interval mapping. Evidence of genotype-by-environment interaction was found by comparing QTL maps of the same population grown in three diverse environments. A total of 22 QTLs for six agronomic traits was detected which were significant in at least one environment, but only seven were significant in all three environments; seven were significant in two environments and eight could only be detected in a single environment. However, QTLs-by-environment interaction was trait dependent. QTLs for spikelets and grains per panicle were common across environments while traits like heading date and plant height were more sensitive to environment. To study the relationships between different species of the Einkorn group, 408 accessions of Triticum monococcum, T. boeoticum, T. boeoticum ssp. thauodar and T. urartu were analyzed electrophoretically for their protein composition at the Gli-1 and Gli-2 loci. In all the species the range of allelic variation at the loci examined is remarkable. The gliadin patterns of T. monococcum and T. boeoticum were very similar to one another but differed substantially from those of T. urartu. Several accessions of T. boeoticum and T. monococcum were shown to share the same alleles at the Gli-1 and Gli-2 loci, confirming the recent nomenclature that considers these wheats as different subspecies of the same species, T. monococcum. The gliadin composition of T. urartu resembled that of the A genome of polyploid wheats more than did T. boeoticum or T. monococcum, supporting the hypothesis that T. urartu, rather than T. boeoticum, is the donor of the A genome in cultivated wheats. Because of their high degree of polymorphism the gliadin markers may help in selecting breeding parents from diploid wheat germ plasm collections and can be used both to search for valuable genes linked to the gliadin-coding loci and to monitor the transfer of alien genes into cultivated polyploid wheats. The objective of the current investigation was to develop a reliable method to obtain vesicular arbuscular mycorrhizae (VAM) in micropropagated plantlets and to determine their influence on growth. An in vitro system for culturing the VA mycorrhizal fungus Glomus intraradices with Ri T-DNA-transformed carrot roots or nontransformed tomato roots was used in this study as a potential active source of inoculum for the colonization of micropropagated plantlets. After root induction, micropropagated plantlets grown on cellulose plugs (sorbarod) were placed in contact with the primary mycorrhizae in growth chambers enriched with 5000 ppm CO2 and fed with a minimal medium. After 20 days of tripartite culture, all plantlets placed in contact with the primary symbiosis were colonized by the VAM fungus. As inoculum source, 30-day-old VA mycorrhizal transformed carrot roots had a substantially higher infection potential than 5-, 10-or 20-day-old VAM. Colonized plantlets had more extensive root systems and better shoot growth than control plants. The VAM symbiosis reduced the plantlet osmotic potential. This response may be a useful pre-adaptation for plantlets during transfer to the acclimatization stage.  相似文献

Transgenic potato (Solanum tuberosum L.) plants were constructed with a Petunia inflata-derived cDNA encoding a pectin methyl esterase (PME; EC 3.1.1.11) in sense orientation under the control of the cauliflower mosaic virus 35S promoter. The PME activity was elevated in leaves and tubers of the transgenic lines but slightly reduced in apical segments of stems from mature plants. Stem segments from the base of juvenile PME-overexpressing plants did not differ in PME activity from the control, whereas in apical parts PME was less active than in the wild-type. During the early stages of development stems of these trangenic plants elongated more rapidly than those of the wild-type. Further evidence that overexpression of a plant-derived PME has an impact on plant development is based on modifications of tuber yield, which was reduced in the transgenic lines. Cell walls from transgenic tubers showed significant differences in their cation-binding properties in comparison with the wild-type. In particular, cell walls displayed increased affinity for sodium and calcium, while potassium binding was constant. Furthermore, the total ion content of transgenic potatoes was modified. Indications of PME-mediated differences in the distribution of ions in transgenic plants were also obtained by monitoring relaxations of the membrane potential of roots subsequent to changes in the ionic composition of the bathing solution. Summary. The regulation of vascular smooth muscle cells (VSMCs) function by taurine has been a subject of increasing interest and investigation, and taurine is taken up into cells through a specific transporter system, the taurine transporter (TAUT). In the present study, we examined the expression of TAUT in VSMCs and the kinetic parameters of the uptake process of TAUT in VSMCs. RT-PCR and western blot demonstrated that the mRNA and protein of TAUT was expressed in VSMCs in vitro. Immunohistochemistry using antibody for TAUT revealed the expression of this protein in rat thoracic aorta. The maximal [3H]taurine uptake rate in VSMCs was 37.75 ± 3.13 pmol/min per mg of protein, with a K m value of 5.42 ± 0.81 μM. Thus, VSMCs are able to express a functional taurine transporter. The regulation and detailed function of taurine and TAUT in VSMCs remain unclear, but our findings suggest a functional role for them in VSMCs metabolism.  相似文献
