Stability of Minute Virus of Mice to Chemical and Physical Agents   总被引:1,自引:0,他引:1       下载免费PDF全文
Minute virus of mice (MVM), a single-stranded deoxyribonucleic acid Parvovirus, was subjected to various inactivation procedures, including chemical disinfectants, heat, and ultraviolet radiation. MVM was found to be less stable than has been reported for other Parvoviruses. This virus was readily inactivated by a variety of chemical disinfectants, including alcohols, formaldehyde, glutaraldehyde, and chloroform. MVM was more sensitive to ultraviolet radiation than was Kilham's rat virus. MVM was more sensitive to heating at temperatures of 35 to 100 C than has been reported for other Parvoviruses. More than 95% of MVM infectivity was inactivated by heating (45, 60, or 100 C) for 60 min, acid (pH 2.0) treatment, or ultraviolet radiation treatment, although a small percentage (less than 2%) of the virus preparation was found to be resistant to these treatments. In addition, more than 99% of the infectivity of MVM was lost after storage at 4C for 10 weeks, although the virus was stable on storage in liquid nitrogen.  相似文献

Unpasteurized skim milk was storaged in a frozen state at ?7°C or ?20°C for up to several months. There was no increase of non casein and non protein nitrogens, but a slight increase of free tyrosine and a slight decrease of alkaline phosphatase activity were detected when storage period was prolonged. Destabilization occurred solely in caseinate complex, but non micellar casein appeared to be stable.

The contents of calcium and inorganic phosphate in the caseinate complex separated by ultracentrifugation were increased appreciably after frozen storage. The viscosity characteristics of frozen storaged skim milk was also investigated.

Caseinate complex was ultracentrifugally separated from skim milk before and after frozen storage, and then lyophilized. Skim milk itself was also lyophilized before and after frozen storage. Dispersibility was examined on the reconstituted suspension of the lyophilized samples.

The lyophilized sample from frozen storaged milk was much less dispersible than the lyophilized control sample prepared before frozen storage. However, when lyophilized samples were once resolved with reagents such as urea and potassium oxalate and then dialyzed against fresh milk, stable micelle resulted in both samples prepared before and after frozen storage.

Some reduction of dispersibility occurred during lyophilization and subsequent storage in a dried state in the caseinate complex prepared before frozen storage. This reduction was small when skim milk was lyophilized and stored.  相似文献

Infectious pancreatic necrosis virus (ATCC strain VR # 299) was lyophilized with different additives and stored at 4 C. Ampoules were assayed at 2 days, at 1, 3, and 6 months, and at 1, 3, and 4 years. Processing losses were least and subsequent maintenance of infectivity was best in skim milk, lactalbumin hydrolysate, and lactose. Quantitative data are given in tabular form.  相似文献

Most of the growth-supporting nutrients in fresh rumen fluid were found in the residue fraction. With one exception, no significant differences were found in total bacterial numbers either by roll tube or most-probable-number procedures when lyophilized rumen fluid residue was substituted for fresh rumen fluid. Lyophilized rumen fluid residue was stable for at least 5 months at room temperature. Rumen fluid supernatant from centrifugation at 1,000 x g had a mean density of 1.005 +/- 0.03 g/ml and contained 1.56% +/- 0.30% dry matter. On the basis of these values, 15.68 mg of lyophilized rumen fluid residue is equivalent to 1 ml of rumen fluid supernatant from centrifugation at 1,000 x g.  相似文献

The North American strain of viral hemorrhagic septicemia virus (NA-VHSV) could be recovered for up to 40 h in natural filtered seawater (27 ppt) with a 50% loss of infectivity after approximately 10 h at 15 degrees C. Addition of 10 ppb North Slope crude oil to the seawater had no effect on virus survival. However, when various concentrations of teleost ovarian fluid were added to seawater, virus could be recovered after 72 h at 0.01% ovarian fluid and after 96 h at 1.0%. When cell culture medium supplemented with 10% fetal bovine serum was added to the seawater, 100% of the virus could be recovered for the first 15 d and 60% of the virus remained after 36 d. These findings quantify NA-VHSV infectivity in natural seawater and demonstrate that ovarian fluid, which occurs naturally during spawning events, significantly prolongs the survival and infectivity of the virus. The extended stabilization of virus in culture medium supplemented with serum allows for low titer field samples to be collected and transported in an unfrozen state without significant loss of virus titer.  相似文献

Bacteriol. 90:132-135. 1965.-The infectivity of measles virus in water is rapidly destroyed at temperatures of 37 C and above. More than 50% of the infectivity is lost after 1 hr at 25 C, and almost 90% loss of infectivity occurs within 24 hr at 4 C. Magnesium chloride enhances the inactivation of the virus at all temperatures tested. Addition of either magnesium or sodium sulfate protects the virus against thermal inactivation. The stabilizing effect is demonstrable at temperatures ranging from 4 to 56 C, but is especially pronounced through 45 C. Prolonged storage (up to 6 weeks) of the virulent virus at 4 C in 1 m magnesium sulfate permits retention of substantial infectivity, whereas storage at 4 C in either water or 1 m magnesium chloride results in a loss of infectivity approximating 99% after 2 weeks. Magnesium chloride also enhances inactivation of the attenuated vaccine strain of measles virus. The attenuated virus, however, is strongly protected by magnesium sulfate against thermal inactivation, and retention of infectivity for long periods of time at 4 C seems feasible when the virus is kept in 1 m magnesium sulfate.  相似文献

Chikungunya virus infectivity was markedly stabilized in the presence of washed suspensions of human platelets but rapidly disappeared in similar preparations of rabbit platelets. Supernatant fluids collected from human platelets had some stabilizing effect on chikungunya virus over a 6-day incubation period at 37 degrees C. Rabbit platelet supernatant fluid had no virus-stabilizing effect, nor did it demonstrate any capacity to inactivate virus as compared to whole rabbit platelet preparations. Thin-section election microscopy demonstrated that chikungunya virus formed an associated with human platelets by becoming entrapped in platelet aggregates; during this process some of the platelets appeared to have undergone degranulation and lysis. Rabbit platelets exposed to chikungunya virus for 24 h demonstrated a considerable amount of platelet degranulation and lysis but virus was not visualized either in association with platelet membranes or within phagocytic vacuoles in the platelet cytoplasm. Human platelets, which appear to be more stable under these incubation conditions, may protect chikungunya virus infectivity from heat inactivation by surrounding viruses with large platelet aggregates whereas rabbit platelets, which appear to be more fragile, do not afford this type of protection. Thus, chikungunya virus in the presence of rabbit platelets may become inactivated by heat or may become bound irreversibly to membranes in such a fashion that infectivity assay and electron microscopy techniques may prove to be too insensitive for detection of virus.  相似文献

By means of precipitin tests, preparations of Type I polio virus have been found to contain at least three antigenically active components. The three precipitin bands have tentatively been correlated to the three distinct components observed in the ultracentrifuge. Type II also contains more than one antigen but the secondary bands are weaker. A lyophilized sample produced a precipitin pattern very similar to that formed by the untreated virus, in spite of the drastic changes in virus infectivity and particle size following this treatment. The Type III virus appeared to be antigenically homogeneous. No cross reactions could be observed between the different virus types and immune sera.  相似文献

Approximately 67% of infectivity is associated with the nucleus 8 hr after productive infection of HEp-2 cells with herpes simplex virus. Comparison of nuclear and cytoplasmic infectious virus and macromolecular aggregates labeled with (3)H-thymidine or with (3)H-choline revealed the following. (i) Cytoplasmic infectious virus and macromolecular aggregates banded in CsCl at a density corresponding to enveloped nucleocapsids. The virus was relatively stable; there was only 50% loss of infectivity and only 16% of the virions became disaggregated. (ii) Nuclear macromolecular aggregates banded in CsCl solution at a density corresponding to unenveloped nucleocapsids and, moreover, both the infectious virus and aggregates were highly unstable. (iii) In sucrose density gradients, the nuclear macromolecular aggregates and infectivity sedimented as a single band and migrated more slowly than the corresponding cytoplasmic material. (iv) The infectivity of nuclear and cytoplasmic virus is readily inactivated by digestion with phospholipase C and with pronase. We conclude the following. (i) Cytoplasmic virus consists of enveloped nucleocapsids. (ii) Nuclear virus consists of nucleocapsids covered with lipid or partially enveloped. (iii) The molecular integrity of viral lipids is essential for infectivity. (iv) The envelope protects the nucleocapsid and accelerates adsorption to cells; it is not, however, inherently essential for infectivity.  相似文献

Infective spores of three species of microsporidia were subjected to the lyophilization process by employing varying media as cryoprotectants. The infectivity of the lyophilized spores was then tested against a standard fresh spore preparation in the appropriate host insect. Spores of Octosporea muscaedomesticae served as an experimental model and were rendered noninfective in host Phormia regina (Calliphoridae: Diptera) after lyophilization with the following cryoprotective agents: skim milk (12%), ascorbic acid (5%) combined with thiourea (5%), glycerol (10%), mesoinositol (5%), and equine serum. Spores of O. muscaedomesticae lyophilized or vacuum-dried in 50% sucrose as well as in the hosts' tissues remained highly infective for as long as 2 years at a dose of 106 spores/fly and a trial length of 12 days. At a dose of 5 × 104 spores/fly there was a slight decrease in infectivity of the spores which had been lyophilized in the host's abdomen after a 2-year storage period compared with that of fresh, nonlyophilized spores. Naked spores of Nosema algerae suspended in 50% sucrose and lyophilized produced infection in 50% of the host population of Anopheles stephensi (Culicidae: Diptera) compared with 70% infection produced by fresh non-lyophilized spores. Spores of Nosema whitei lyophilized within its host larva Tribolium castaneum (Tenebrionidae: Coleoptera) remained 100% infective at a dose of 5 × 105 spores/gram diet. It is concluded that an aqueous solution of 50% sucrose and/or the host's tissues are excellent protectants for the cryogenic or vacuum-drying process of the above-named spores, and their protective function may apply also to other microsporidian species.  相似文献

为了研究优化冻干乙型脑炎减毒活疫苗的冻干工艺,运用正交试验法L934考察不同冻干参数对该疫苗成品质量的影响,以外观、残余水分、病毒滴度以及热稳定性为直观分析指标,并对病毒滴度进行方差分析。结果显示,主干燥时间和主干燥真空压力对病毒滴度的影响有显著统计学意义(P<0.05)。优化的冻干参数为预冻温度-35℃、时间2h;主干燥温度从-35℃升温至-10℃,再由-10℃升温至33℃,总耗时16h,真空压力0.220mbar;二次干燥的温度维持于30℃,真空压力为0.001mbar,终点测试压力无变化时结束。对冻干乙型脑炎减毒活疫苗冻干参数筛选优化后,可得到较佳的冻干曲线,经验证该曲线适用于生产。  相似文献

The objective of this study was to apply the pertinent findings from gamma inactivation of virus infectivity to the production of high quality diagnostic reagents. A Gammacell 220 (Atomic Energy of Canada, Ltd., Ottawa, Canada) was used to subject 38 viruses grown in either susceptible tissue cultures or embryonated chicken eggs to various doses of gamma radiation from a cobalt-60 source. The radiation required to reduce viral infectivity was 0.42 to 3.7 megarads (Mrad). The effect of gamma treatment on the antigenic reactivity of reagents for the complement fixation (CF), hemagglutination (HA) and neuraminadase assays was determined. Influenza antigens inactivated with 1.7 Mrad displayed comparable potency, sensitivity, specificity and stability to those inactivated by standard procedures with beta-propiolactone (BPL). Significant inactivation of influenza N1 and B neuraminidase occurred with greater than 2.4 Mrad radiation at temperatures above 4 degrees C. All 38 viruses were inactivated, and CF or HA antigens were prepared successfully. Antigenic potency remained stable with all antigens for 3 years and with 83% after 5 years storage. Influenza HA antigens evaluated after 9 years of storage demonstrated 86% stability. Gamma radiation is safer than chemical inactivation procedures and is reliable and effective replacement for BPL in preparing diagnostic reagents.  相似文献

An accelerated storage test was carried out on freeze-dried samples of five variants of tomato mosaic virus – from leaf material, plant sap and purified preparation with and without protectant, at 28, 37 and 45 °C. A model for longevity prediction of lyophilized tobamoviruses was developed. The preservation of tomato mosaic virus decreased to 10% after 0.7–2.8 years in unprotected variants and after 7.5–11 years in protected samples under real storage conditions. This article looks at storage factors influencing the stability of potential DNA calibration standards for use in quantitative polymerase chain reaction (PCR). Target sequences from the bacteria Campylobacter jejuni were cloned into a plasmid vector. Samples of these potential calibration standards were stored at +4, −20, and −80 °C as aqueous and lyophilized samples and were prepared as both single-use aliquots and multiple-use preparations. Results showed that the samples stored as single-use aqueous solutions at +4 °C and lyophilized samples stored at +4 and −20 °C were the most stable. Samples stored as frozen aqueous solutions at −20 °C were the least stable.  相似文献

Treatment of suspensions of vesicular stomatitis virus with Tween-ether results in a rapid and considerable loss of infectivity (ca. 4 logs in 2 min), but the residual infectivity is comparatively stable to further treatment with ether. The infectivity remaining after the short exposure to Tween-ether is not due to virus for the following reasons. (i) It is much less infective for tissue cultures than for mice, whereas the intact virion is equally infective for both hosts. (ii) The residual infectivity is much less stable than virus infectivity in both sucrose and tartrate gradients. (iii) Virus immune serum does not neutralize its activity. (iv) The infectivity is associated with material which sediments further in sucrose gradients and has a greater buoyant density in tartrate gradients than the virion. Experiments with (32)P-labeled virion showed that the infective substructure contains ribonucleic acid with the same sedimentation characteristics as that extracted from the virion. Electron microscopy shows that the infective component has the same overall bullet-like structure as the virion but lacks the outer envelope and fringe structure.  相似文献

Purification and preservation of potato virus X from leaf sap of tobacco plants before lyophilization was carried out by two methods: 1) precipitation by polyethylene glycol and ultracentifugation, and 2) precipitation by ammonium sulphate, chromatography on Sephadex G-50 and ultracentrifugation. The first method is preferable to the second because the final preparation contains more virus antigen. Both preparations were strongly infectious and maintained antigenic properties after lyophilization. To achieve a more gentle course of lyophilization of virus preparations, addition of urotropine and dextran T-10 to the virus suspension, purified by the precipitation by polyethylene glycol-6000, was examined. Addition of urotropine was proved unsatisfactory, because only antigenic properties were maintained after lyophilization while the infectivity disappeared. But we can recommend addition of dextran T-10 up to a concentration of 6% to the preparation of virus antigen before lyophilization. The course of lyophilization is much rapider, the lyophilized product can be very easily dissolved in water and is not hygroscopic. The product is strongly infectious and gives the serological precipitation reaction in a dilution four times that of X virus antigen lyophilized without addition of dextran T-10.  相似文献

Various stabilizers containing no animal-derived components were assessed for their efficacy in stabilizing live attenuated varicella virus under various storage temperatures. Stabilizers containing carrageenan, soy protein hydrolysates, or sucrose had a satisfactory performance to maintain infectivity of the cell-free virus when compared to control stabilizer containing animal-derived gelatin/gelatin hydrolysate.  相似文献

Conditions of Interferon processing were analyzed to select those that promote stability after freeze-drying. The effects of various preparative methods and treatment conditions were assessed by measuring the retention of biological activity by lyophilized interferon samples in two kinds of accelerated storage tests: the linear nonisothermal stability (LNS) test, a rapid method used for direct comparison of two or more preparations of interferon, and the multiple isothermal storage (MIS) test, a slow method requiring weeks to months to obtain data for the prediction of stability of a given preparation stored under various conditions.The most stable preparations of Newcastle-disease-virus-induced mouse L cell interferon were obtained using the following conditions: 1) perchlorate treatment to inactivate residual inducing virus, 2) nonspecific adsorption using zeolite for partial purification, 3) suspending medium of 0.5% bovine serum albumin in 0.1 m sodium phosphate buffer at pH 7, and 4) sublimation of ice in vacuo with a starting temperature of ?30 °C to a final residual moisture of about 3%. The final product, reference reagent G002-904-511, was stable throughout the course of the LNS test. From an extensive MIS test, this reference interferon was predicted to lose 1000 units of activity in 110 years at 4 °C and 1000 units in 100 days at 37 °C. After 6 years of storage at 37 °C when the predicted residual activity would be about 20% of the original potency, 35% of initial interferon activity remained, confirming the usefulness of the short-term predictive test.  相似文献
