Seasonal levels of LH, FSH, testosterone (T) and prolactin (PRL) were determined in plasma of six captive adult male pudu (Pudu puda) kept in Concepcion, Chile. Average PRL levels exhibited one peak (28 ng/ml) in December (summer); minimal levels (3 to 6 ng/ml) were detected between April and July. FSH concentrations remained at peak levels (54–63 ng/ml) from December until March; minimal values (25–33 ng/ml) were detected from April until October. T levels exhibited two, almost equal peaks; the first peak (2.8 ng/ml) was detected in March (rut) and the second one (2.7 ng/ml) in October (spring). Both T peaks were preceded by an earlier elevation of LH in February and July (both around 1.3 ng/ml). During the fall, only the alpha male exhibited a sharp peak of T (8.4 ng/ml), whereas in the spring five out of six bucks demonstrated an increase of T levels. Two peaks of LH and T and the 4 months of elevated FSH may be related to a long period of spermatogenesis observed in this species.  相似文献

The squirrel monkey (Saimiri boliviensis boliviensis) has a well-defined breeding season during which adult males undergo androgen-dependent morphological changes, with acquisition of active spermatogenesis. To assess the hormonal events of this annual cycle, blood samples were obtained weekly from ten adult males, and serum was assayed for testosterone (T), androstenedione (ΔA), and dehydroepiandrosterone (DHEA). A significant seasonal variation was noted in mean serum T (P < 0.02), ΔA (P < 0.02), and DHEA (P < 0.001) concentrations. Mean ΔA concentrations increased from a nonbreeding season nadir of 91.4 ± 12.9 ng/ml (mean ± standard error) to a prebreeding concentration of 139 ± 10.5 ng/ml and breeding season peak of 167.5 ± 15.4 ng/ml (P < 0.05). Mean DHEA concentrations increased from a nonbreeding season nadir of 8.3 ± 0.8 to a breeding season peak of 14.3 ± 1.2 (P < 0.001). Mean T levels in the nonbreeding (52.2 ± 11.6 ng/ ml) and prebreeding season (48.6 ± 7.4) were similar. However, T significantly increased during the breeding season to 103.5 ± 12.8 ng/ml (P < 0.05). Progressive changes in body weight and morphology paralleled the rise in serum ΔA levels. The pattern of peripheral serum androgen concentrations throughout the year would suggest annual activation of the hypothalamic-pituitary-adrenal and/or hypothalamic-pituitary-gonadal axes.  相似文献

Summary Lacertilian species display a remarkable diversity in the organization of the neural apparatus of their pineal organ (epiphysis cerebri). The occurrence of immunoreactive S-antigen and opsin was investigated in the retina and pineal organ of adult lizards, Uromastix hardwicki. In this species, numerous retinal photoreceptors displayed S-antigen-like immunoreactivity, whereas only very few pinealocytes were labeled. Immunoreactive opsin was found neither in retinal photoreceptors nor in pinealocytes. Electron microscopy showed that all pinealocytes of Uromastix hardwicki resemble modified pineal photoreceptors. A peculiar observation is the existence of a previously undescribed membrane system in the inner segments of these cells. It is evidently derived from the rough endoplasmic reticulum but consists of smooth membranes. The modified pineal photoreceptor cells of Uromastix hardwicki were never seen to establish synaptic contacts with somata or dendrites of intrapineal neurons, which are extremely rare. Vesiclecrowned ribbons are prominent in the basal processes of the receptor cells, facing the basal lamina or establishing receptor-receptor and receptor-interstitial type synaptoid contacts. Dense-core granules (60–250 nm in diameter) speak in favor of a secretory activity of the pinealocytes. Recently, increasing importance has been attached to the role of estrogens and their receptors in male reproduction, since they have been found to be abundant in the male reproductive tract. In the lizard, Podarcis sicula, a seasonal breeder, estrogens seem to be involved in the regulation of testicular activity. Particularly, it has been hypothesized that the block of spermatogenesis and the complete regression of the epididymis and other secondary sexual characters (SSCs) in autumn might be due to high estrogen levels. To investigate the role of estrogens in the reproductive process of male lizards, we utilized Fadrozole ((AI) [4-(5,6,7,8-tetrahydroimidazole [1,5-a] pyridin-5-yl)-benzonitrile monohydrochloride] (CGS 16949A)), a nonsteroidal inhibitor of aromatase, the enzyme involved in the aromatization of androgens to estrogens, evaluating its effects on plasma sex-hormone release, spermatogenesis and epididymis morphology. For this purpose, adult male lizards, captured during the autumnal recrudescence, were intraperitoneally injected with 0.5 microg and 5 microg/g/body weight of AI for 15 and 30 days. In the animals treated with the higher AI dose, estrogen levels decreased if compared to the control groups, whereas androgen levels increased. Furthermore, histologic sections of testes and epididymes showed that the 30-day treatment with AI-induced spermatogenesis resumption with release of sperms into the large lumen of the seminiferous tubules, and the epididymes appeared more developed with moderately secreting columnar canal cells. Therefore, it is proposed that failure of spermatogenesis in autumn might be due to high estrogen levels.  相似文献

With a view to elucidate seasonal variations in testicular spermatogenesis, quantitative analysis of spermatogenic cells was carried out in non-human primate species viz. rhesus (Macaca mulatta) and bonnet (M. radiata) monkeys during breeding (October-December) and non-breeding (May-June) seasons. The results revealed significant inhibition of testicular germ cell population during non-breeding compared with the breeding period in both the species. Quantitative determination of Sertoli cell-germ cell ratio showed a marked decrease in the number of type A-spermatogonia, spermatocytes (non-pachytene and pachytene) and spermatids (in steps 1-12 of spermiogenesis) in rhesus monkey during the non-breeding period. Bonnet monkeys exhibited the significant decline in the number of primary spermatocytes and spermatids during the non-breeding phase. In addition, average diameter of round seminiferous tubules and nuclear diameter of Leydig cells also decreased significantly in rhesus monkeys. However, bonnet monkeys did not show any significant change in nuclear diameter/morphology of Leydig cells, testicular tubular diameter and number of type A-spermatogoniae. Sertoli cell number did not show any significant change during both breeding and non-breeding periods in both the species. The results of this study indicate a prominent seasonal variation in testicular spermatogenic/Leydig cells in rhesus monkeys than those observed in bonnet monkeys.  相似文献

To determine the separate spermatogenic actions of FSH and testosterone, adult male lizards Hemidactylus flaviviridis with recrudescent testes were administered the non-steroidal antiandrogen flutamide either alone or in combination with FSH or testosterone, and the histology and histochemistry of the testes and ductus epididymides were studied. Flutamide-treated animals displayed a marked hypertrophy of Leydig cells. A few spermatids were also seen in testis of more than half the animals treated with flutamide. Flutamide also produced a significant increase of primary spermatocytes; no spermatids were observed in controls. A significant inhibition of spermatogenesis was noted in lizards treated either with testosterone alone or in combination with flutamide. Ovine FSH treatment caused a significant stimulation of spermatogenesis, as indicated by the increase of primary and secondary spermatocytes and the transformation of secondary spermatocytes into spermatids or, in a few cases, into spermatozoa. A considerable depletion of sudanophilic lipid and moderate delta 5-3 beta-hydroxysteroid dehydrogenase activity was noted in the Leydig cells of FSH-treated animals indicating enhanced steroidogenesis. Similar results were obtained when lizards were treated with flutamide + FSH. The effects of simultaneous treatment of flutamide with FSH or testosterone on ductus epididymidis revealed that flutamide markedly inhibited the epithelial cell height and lumen diameter with a loss of luminal content when compared to FSH or testosterone-treated lizards.  相似文献

Adult lizards (Sceloporus torquatus) were used to test whether seasonal differences in the effects of pinealectomy upon the locomotor activity rhythm exist. Animals were field collected and exposed to artificial light - dark cycles and constant temperature in winter and summer. Free running circadian rhythms under constant temperature and dim red light were monitored using infrared light-crossings. The effects of pinealectomy were assessed by analysing the circadian parameters of free running period and activity - rest ratio in constant darkness or light - dark cycles. Results obtained indicate that pinealectomy changes the free running period of locomotor activity rhythm, irrespective of season, while seasonal differences in activity-rest ratio were detected. Our findings support the hypothesis that seasonal regulation of circadian rhythms in lizards is accomplished, in part, via the output of the pineal gland.  相似文献

The intensity of sperm competition is often measured using the gonadosomatic index (testes/body weight). But sperm competition could be mediated more by size of the epididymis than by size of the testicles, and little information is available on the relationship between testicular and epididymal size. We found that both organs were positively correlated in size among male rhesus macaques. Body weight accounted for over 70% of the variance in testicle size and volumetric estimates of testicle size accurately reflected testicle weight. We conclude that methods for ascertaining testicle size are accurate, but the covariation in size between testicles and epididymis will hamper understanding of the physiological mechanisms involved in sperm competition in primates. © 1993 Wiley-Liss, Inc.  相似文献

Seasonal variation in fecal testosterone levels in free-ranging male Japanese macaques (Macaca fuscata) was examined with reference to their dominance rank and age class. Six adult (>or=7 years old, three higher-ranking and three lower-ranking) and four adolescent (5-6 years old, two higher-ranking and two lower-ranking) males were selected as target animals. Fecal samples of these males were collected during the first 3-5 days of each month and analyzed by the method developed by Barrett et al. [Primates 43:29-39, 2002 b]. Testosterone levels varied significantly across the 12 months, and were highest in the early and middle parts of the mating season (i.e., October and November). Higher-ranking adult males displayed a peak testosterone level in October, whereas lower-ranking adults had no clear peak in the mating season. Such a difference in testosterone peaks in males could provide higher-ranking males more opportunities to fertilize females at first ovulation in the mating season than lower-ranking males.  相似文献

Testosterone is strongly associated with the annual development of antlers in cervids, but endocrine research on wild, freely breeding ungulates is often done without repeated capture of known‐aged individuals. As a result, our knowledge on how testosterone fluctuates over the course of a lifetime and variation in lifetime patterns among individuals is limited. We investigated patterns of testosterone in a freely breeding population of white‐tailed deer (Odocoileus virginianus) in Alabama, USA, that breeds in January. Testosterone peaked during the height of the breeding season, despite this period occurring approximately 2 months later than in most temperate‐region white‐tailed deer populations. Age‐related differences in testosterone were only prevalent during the breeding season, with bucks ≥3.5 years old having greater testosterone (853 ng/dl ± 96 SE; p = 0.012) than bucks 1.5–2.5 years old (364 ng/dl ± 100 SE). Additionally, an individual''s testosterone level as a yearling was not positively associated with their lifetime maximum testosterone level (p = 0.583), and an individual''s mean testosterone level was positively associated with lifetime testosterone variation (p < 0.001). To our knowledge, our study is one of the first to assess how testosterone early in life might relate to individual testosterone later in life. We believe these data provide insight into lifetime hormonal patterns in cervids, and that these patterns may indicate intraspecific variation of lifetime reproductive strategies.  相似文献

The epididymis of the fan-throated lizard Sitana ponticeriana was examined with light and transmission electron microscopy to understand the cellular mechanisms of fabrication of secretion granules in epithelial principal cells, granule release into the lumen, and the fate of the dense structured granules after reaching the lumen. Principal cells of the ductus epididymis, except at the cauda, secrete electron-dense biphasic granules copiously, which decrease in abundance from the initial segment to corpus. The principal cell possesses a prominent Golgi apparatus and all versions of endoplasmic reticulum (ER), rough, smooth, and sparsely granulated. The material of the dense portion of the secretion granules, after processing at the Golgi apparatus, appears to accumulate in large ER cisternae in the supranuclear cytoplasm. It undergoes condensation when the cisternae become condensing vacuoles. Mitochondria appear to play a role in dense granule formation. The condensing vacuoles are displaced toward the apical cytoplasm when the material of the less dense portion is added to the condensing vacuoles at the Golgi area. Thus, the less dense and dense portions of the secretion granules are secreted and added to the condensing vacuoles separately. The composite granules are released into the lumen by exocytosis when the less dense portion merges with the luminal content, whereas the dense portion maintains its structured identity. The latter, initially measuring 1-2 microm in diameter, increases in size several times. It is inferred that these granules release their content gradually, resulting in the appearance of vacuoles, and suggesting that the granules have an insoluble matrix in which there is a sparingly soluble material. The substance leaching out of the granules appears to contribute to keeping the sperm quiescent and alive during storage in the male reproductive tract.  相似文献

Uromastix hardwicki was found more resistant to insulin as compared to other poikilotherms and this insulin resistance appears to be related to the preponderance of glucagon secreting alpha-cells in this species. The magnitude and the persistence of hypolgycemia caused by insulin are directly dependent upon the quantity of the dose. The higher doses (2000 and 3000 U/kg) resulted into shocks, but the lack of correlation between hypoglycemia and shocks has been attributed to the direct effect of insulin on the nervous system. Histologically, insulin induced degranulation of both the beta- and alpha-cells. The physiological action of insulin is apparently greater at higher ambient temperature. The ability of insulin to damage beta- and alpha-cells in Uromastix hardwicki further gathers evidence for the similarity of these cell types with those of mammals and other vertebrates in being the source of insulin and glucagon respectively, and in controlling the blood glucose homeostasis in this lizard.  相似文献

The metabolism of ascorbic acid, cholesterol, serum testosterone level and activities of 3Β and 17Β hydroxysteroid dehydrogenases were studied in testis and Cauda epididymis of prepubertal, pubertal and postpubertal (5, 15, 30, 45, 55 and 60 day old) rats. The data showed that serum testosterone levels and 3Β and 17Β hydroxysteroid dehydrogenases were increased with the age. The ascorbic acid metabolism was found to be stabilized in testis at day 30 being comparable with the adult, whereas a spurt in its metabolism occurred by day 45 and a significant depletion in ascorbic acid content in relation to the passage of the first wave of spermatozoa through cauda epididymis. The results of this study clearly elucidate that ascorbic acid is involved in metabolism of testis and epididymis in developing postnatal rats, in relation to the increasing demands for attaining a stable hormonal milieu, and the onset of puberty and the passage of the first wave of spermatozoa,via the formation of its free radical monodehydroascorbic acid and charge transfer complex mechanism  相似文献

Although different mechanisms exist to explain the presence of polymorphism in lizards, one model suggests that multiple morphotypes display the same level of fitness. Three male morphs (grey, yellow and orange) coexist in Sceloporus aeneus, a Mexican endemic oviparous lizard. Using a histological perspective, we test the hypothesis that spermatogenic output does not vary across morphotypes of Saeneus during its maximum testicular activity. Males of Saeneus (five grey, five yellow and five orange) were collected in Calimaya, Estado de México, Mexico. Snout-vent length (SVL), testis mass, diameter and epithelial heights for the seminiferous tubules and epididymis, and the number of layers of germ cells did not vary among morphs; moreover, according to principal component analysis, a high overlap among lateral colour morphs exists. Our results suggest strongly that the lateral colour morphs in Saeneus have the same spermatogenic output, and natural selection may be a stronger driving force than sexual selection within this species. Further studies into other lizard species with multiple morphotypes are required to determine whether the lack of variation in spermatogenic output observed in this endemic lizard is consistent across polymorphic species which will provide a greater understanding of the selective mechanisms acting on an individual’s fitness.  相似文献

The venom apparatus of Lapemis hardwicki , consisting of two functional fangs, their venom glands, and associated musculature, are described. The yield of venom per snake ranged from 2.4-5.2 mg. The LD50 of the crude venom varied from 0.7-1.4 mg/kg intravenously in mice. The toxicological, chemical and immunological properties of the venom are discussed.  相似文献

Characterizing leptin's structure and function in mammals has been the subject of thousands of studies since 1994. Recently, the study of leptin has expanded to include its distribution in non-mammalian taxa, and the role that leptin plays in the reproductive axis. We demonstrated in a previous study that Sceloporus undulatus, fence lizards (ectotherms), express a leptin-like protein. In the current study we quantified seasonal variation in this putative leptin among free-ranging fence lizards from two populations characterized by early and late reproductive maturation (after one or two years, respectively). Immunoblots were performed on whole blood samples to detect leptin and estimate its titer. Leptin titers were higher in the reproductive population of S. undulatus (early maturing: 2.5 ± 0.2 μg/mL; late-maturing 2.2 ± 0.3 μg/mL; mean ± 2 S.E.), but both populations showed the same seasonal pattern. Leptin titers were lowest in fall when fat stores are expected to be highest (spring: 2.6 ± 0.3 μg/mL; summer: 2.6 ± 0.3; μg/mL; fall: 1.8 ± 0.3 μg/mL), consistent with findings of seasonal variation in free-ranging mammals. Our data support previous work asserting that lizards express leptin and that it has a similar physiological function in endotherms and ectotherms. Our long-term goal is to use leptin to manipulate age at maturity and to test fundamental questions in the evolution of life-history strategies.  相似文献

To test whether the male could contribute to the birth peak seen in both wild and captive vervets, testicular volume and peripheral testosterone concentration were measured monthly in nine adults throughout a 14-month period. Volume was an average of 15% greater during the months of June to September, the period of natural breeding activity in the wild, than at other times of the year. Testosterone concentration rose throughout the time period but did not correlate with testicular volume. Quarterly biopsies of the right testis in animals either used for or withheld from breeding revealed the presence of spermatozoa and the existence of spermatogenesis throughout the year. There was no correlation of testicular volume with breeding status or biopsy. If these results from individually caged animals are representative of group-caged and wild animals, then such small seasonal changes are unlikely to affect year-round breeding. Therefore, a physiological contribution by the male to natural birth peaks seen in the wild and captivity is not readily apparent.  相似文献

The microanatomy of the testes and testicular ducts (rete testis, ductuli efferentes, ductus epididymis and ductus deferens) of Leiolepis ocellata (Agamidae) was investigated using light microscopy including histochemistry. Each testis contains seminiferous tubules and interstitial tissues. The former house spermatogenic cells (spermatogonia A & B, preleptotene, primary and secondary spermatocytes, spermatids (steps 1–8) and spermatozoa) and Sertoli cells, while the latter comprise peritubular and intersitial tissues. The rete testis is an anastomosing duct, having intratesticular and extratesticular portions. The proximal region of ductuli efferentes has wider outer ductal and luminal diameters than those of the distal region. The convoluted ductus epididymis is subdivided into four regions (initial segment, caput, corpus and cauda), based on the ductal diameter, epithelium characteristics and cell components. The ductus deferens has the greatest diameter and is divided into the ductal and ampulla ductus deferens. The ductal portion is subdivided into the proximal and distal regions, based on the epithelium types and ductal diameters. The ampulla ductus deferens is a fibromuscular tube, having numerous mucosal folds projecting into the lumen. Spermiophagy is detectable in the ductus epididymis and ductus deferens. The present results contribute to improved fundamental knowledge on the microanatomy of the reptilian reproductive system.  相似文献

There are no reported data on hormonal fluctuations in black‐handed spider monkey males. On previous research about the reproductive physiology of this monkey we have found that during the dry season females show ovulatory estrogen peaks and males present the best quality semen. As part of an ongoing research, in this study we assessed seasonal variations in the concentration of serum luteinizing hormone (LH) and testosterone (T) in three adult spider monkey males to corroborate the seasonal reproductive synchrony. At the same time sperm count and motility were evaluated to search for any correlation between those sperm parameters and hormonal concentrations. We took blood and semen samples (by electroejaculation) of anesthetized males throughout the rainy (June–September) and dry (October–May) months. Our results revealed that T and LH were higher throughout the dry season and there was a significant correlation between T concentration and sperm count. Although higher during the dry season, sperm motility tended to correlate with testosterone and LH levels. These results demonstrated that black‐handed spider monkeys have a tendency to show a seasonal pattern of reproduction being the dry season the most likely time to achieve fertilization. Am. J.
