大绒鼠的分带核型研究   总被引:1,自引:0,他引:1  
本文采用G带、C带和银染核仁组织者(Ag-NORs)等技术,对大绒鼠(Eothe nomys miletus miletus)的核型进行观察分析。结果表明:2n=56,常染色体和性染色体皆为单臂染色体。X染色体的长度接近于No.1染色体,Y染色体的长度相当于14号染色体。G分带可鉴别每对染色体的特征,C-带核型中全部着丝点C带均显示不同程度的阳性。Y染色体整条呈阳性。Ag-NORs有5对,分别分布于1、2、6、14和27号染色体的着丝粒附近。通过核型分析,对大绒鼠的分类地位进行了初步探讨。  相似文献

八倍体小偃麦与天蓝偃麦草杂交F1染色体组构型   总被引:1,自引:0,他引:1  
首次获得麦草8号、麦草9号、远中2号八倍体小偃麦与天蓝偃麦草的属间杂种,杂交当代结实率为31.49%,39.28%和10.41%。杂种F1表现为两亲的中间型,植株高大、繁茂,穗长20~30 cm,小穗数25~30个,多年生,抗寒,在哈尔滨冬季无覆盖条件下可安全过冬。对F1植株进行减数分裂行为观察,结果发现,染色体配对不正常,单价体频率高,出现多价体。杂种F1减数分裂中期I染色体配对构型分别为:9.5Ⅰ+16.98Ⅱ+0.27Ⅲ、13.6Ⅰ+14.01Ⅱ+0.87Ⅲ、11.2Ⅰ+16.8Ⅱ+0.08Ⅲ。二价体数变动在13~18间、单价体数变动在11~17间、多价体变动在0.08~0.87间,二价体多数是棒状二价体,推测两亲有一对部分同源关系染色体组,其余为非同源染色体组,但有的染色体间有部分同源关系。小麦5B染色体上Ph基因可能受到E组染色体的抑制。  相似文献

蒙古冰草Actin基因片段的克隆及序列分析   总被引:2,自引:0,他引:2  
旨在利用同源序列法分离蒙古冰草(Agropyron mongolicum Keng)Actin基因同源片段,为研究其他基因在蒙古冰草中的表达和调控提供内标参照.根据禾本科植物小麦Actin基因(AB181991)的保守序列设计2对引物A4和A5,采用RT-PCR扩增蒙古冰草的Actin基因片段,分别得到656 bp和848 bp的片段,使用DNAman和DNAuser等分子生物学软件进行序列分析,将2个片段的重复序列合并后获得一段长度为962 bp的基因片段,编码237个氨基酸,将克隆的Actin基因片段命名为MwACT.该序列与其它植物Actin基因核苷酸序列的同源性均在80%以上,其中与小麦、大麦的同源性达到94%;与氨基酸序列的同源性均在90%以上.  相似文献

通过PCR克隆的方法,获得了分别来自二倍体长穗偃麦草的E基因组和四倍体长穗偃麦草的E_1基因组的4个高分子量麦谷蛋白亚基(HMW-GS)基因启动子的部分序列。序列分析表明,它们之间的同源性较高,两个x型亚基启动子序列之间只有1个碱基的差异,而两个y型亚基启动子序列完全相同,x和y型亚基启动子序列之间的长度和部分碱基位点都有差异。推测四倍体长穗偃麦草中的E_1基因组可能起源于二倍体的E基因组。与来自小麦族的A、B、D和G基因组部分亚基基因的启动子序列比较表明,小麦族的这一区域在进化上是相当保守的,不同基因组来源的序列同源性都在90%以上。经过对这些序列的聚类分析,表明长穗偃麦草的y型HMW-GS基因与其他亚基基因的进化关系较远,而x型亚基基因与一个来自小麦1B染色体的亚基基因关系最近。  相似文献

利用顺序基因组-重复序列原位杂交技术对1个来自中3不育系和普通小麦恢75杂种后代稳定株系H96276-2的染色体组成进行了分析。以中间偃麦草(Agropyronintermedium)基因组DNA为探针的荧光原位杂交结果表明,H96276-2的体细胞中有42条染色体,包括20对小麦染色体和1对小麦-中间偃麦草易位染色体,中间偃麦草染色体的易位片段位于1对小麦染色体的端部。进而用重复序列探针pSc119进行第2次荧光原位杂交,证明H96276-2中的中间偃麦草染色体易位片段位于小麦2B染色体的短臂上。  相似文献

将普通小麦“济南177”(Triticum oestivum cv.Jinan 177)原生质体(受体)和经紫外线照射的高冰草Agropyron elongatum)原生质体(供体)用PEG(聚乙二醇,polyethylene glycol)法诱导融合,获得外形偏向小麦的不对称体细胞杂种及后代。经过田间繁育,现已到F5代。对来源于同一个体细胞杂种克隆的不同株系(Ⅱ-2,8-l,Ⅱ-I-8)的F5代的根尖细胞染色体进行核型分析,并与其亲本比较,结果表明,杂种株系染色体的形态、数目在遗传上均趋于稳定。各杂种株系的染色体与亲本小麦“济南177”之间有多处显著的不同,杂种株系之间也存在差异,推测高冰草的染色体小片段可能进入了受体小麦的染色体中。  相似文献

Aims To investigate the effects of dew on plants, we conducted the experiment to determine the physiological characteristics and leaf structures of Leymus chinensis and Agropyron cristatum in response to increasing dew under drought stress.Methods Four treatments (no dew, three times dew and five times dew per week under drought stress, and well-watering) were designed to examine leaf relative water content, water potential, net photosynthetic rate, water use efficiency, biomass, and leaf structures of L. chinensis and A. cristatum. Important findings There was a significant increase in the relative water content and water potential by simulated dew increase for two plants species under drought stress (p < 0.05). For A. cristatum, simulated dew increase significantly enhanced the net photosynthetic rate, stomatal conductance, and transpiration rate of plants under drought stress (p < 0.05). On the other hand, there was no significant difference in the stomatal conductance and transpiration rate for L. chinensis among treatments. Simulated dew increase improved the aboveground biomass and root biomass of two species. The ratio of yellow leaves to the total leaves was decreased by simulated dew increase for two species. Dew increase also protected leaf structures against the drought stress, suggesting that the dew increase can slow down the death process of leaves resulted from drought stress. Therefore, the study demonstrated that dew increased the available water for the leaves of L. chinensis and A. cristatum grown in the drought stress and thus had positive effects on the photosynthesis, water physiology and plant development.  相似文献

1. In minesoil reclamation, the establishment of a sustainable plant cover requires the improvement of limiting conditions and the re-initiation of carbon (C) and nutrient cycling.
2. The approach used in this study for reclaiming an abandoned sandpit in Quebec, Canada, was based on a heavy organic amendment as an attempt to accelerate the reconstruction of a functional ecosystem.
3. The one-time intervention consisted of incorporating paper de-inking sludge into soil at two rates (0 and 105 dry t ha–1), supplemented with nitrogen (N) at three rates (3, 6 and 9 g kg–1 sludge) and phosphorus (P) at two rates (0·5 and 1·0 g kg–1 sludge) followed by seeding (mid-summer) of Agropyron elongatum (Host) Beauv. (tall wheatgrass).
4. Standing biomass increased in the presence of sludge after both the first and second full growing seasons. High N application rates further increased yield, more importantly in the second season. The high P rate improved grass establishment in all cases. Ground cover increased with time and doubled in the presence of sludge whereas it declined in the absence of sludge. Phosphorus and N uptake was improved consistently in the presence of sludge.
5. Sludge application resulted in improved water retention and cation exchange capacities, and an increase in pH and bulk density of sandpit minesoil, all of which may have accounted for the significant improvement in plant responses. Levels of soil C and N suggest that this reconstructed system is approaching sustainability.
6. Adequate N and P supplements will accentuate the positive influence of sludge on revegetation.  相似文献

通过盆栽实验, 对干旱胁迫下黄土高原地区冰草(Agropyron cristatum)叶片的抗坏血酸和谷胱甘肽合成及循环代谢相关酶及物质含量进行了研究。结果表明: 冰草可以通过增强叶片的抗坏血酸和谷胱甘肽合成及循环代谢酶: 抗坏血酸过氧化物酶、谷胱甘肽还原酶、脱氢抗坏血酸还原酶、单脱氢抗坏血酸还原酶、L-半乳糖酸-1, 4-内酯脱氢酶和γ-谷氨酰半胱氨酸合成酶活性, 维持植物体内抗坏血酸和谷胱甘肽水平及氧化还原状态, 从而抵御干旱造成的氧化胁迫。但叶片抗坏血酸和谷胱甘肽合成及循环代谢对不同水平干旱胁迫的响应, 随胁迫时间的延长而不同。在胁迫24天以前, 严重干旱下叶片的抗坏血酸和谷胱甘肽合成及循环代谢增强较显著; 在胁迫24天后, 由于该胁迫下植物所遭受的氧化胁迫较为严重, 叶片中上述6种酶的活性均呈降低趋势。而在中度干旱下叶片抗坏血酸和谷胱甘肽合成及循环代谢相关的6种酶在整个胁迫过程中均保持较高的活性。这说明, 冰草能够长时间有效地抵御中度干旱所造成的氧化胁迫, 但只能在一定时间范围内有效地抵御严重干旱所造成的氧化胁迫, 胁迫时间延长则会降低其抵御严重干旱的能力。  相似文献

Barleys with and without the Yd2 resistance factor, wheat alien addition stocks with other barley yellow dwarf virus (BYDV) resistance factors and true wheats were challenged with three Australian isolates of BYDV-RPV. Yd2 resistance was effective against two of the BYDV-RPV isolates and inoculated barleys which carry Yd2 did not develop BYD symptoms and shoot growth was not affected. However, barleys with Yd2 were susceptible to the third BYDV-RPV isolate. All barley lines inoculated with the third virus isolate developed typical BYD symptoms (yellowing), shoot growth was reduced compared to uninfected controls and virus titres determined by ELISA were high and similar in barleys with and without Yd2. In contrast, resistances from Thinopyrum intermedium and Agropyron pulcherrimum in wheat backgrounds were effective against all three BYDV-RPV isolates. Shoot growth of inoculated plants with either of these resistance factors did not differ from uninfected controls and virus titres determined by ELISA were very low.  相似文献

用PEG-6000模拟干旱胁迫处理蒙古冰草(Agropyron mongolicum Keng)幼苗,研究了渗透胁迫对蒙古冰草幼苗某些酶活性的影响。结果表明:在轻度干旱胁迫下,蒙古冰草幼苗可以通过提高酶活性来维持活性氧产生与清除之间的平衡,以减少干旱胁迫介导的氧化胁迫,使MDA含量、膜透性降低。在中度或重度干旱胁迫下,酶活性可能遭受破坏,即使酶活性得到一定程度的提高,也不能维持活性氧产生与清除之间的平衡,使植物产生氧化伤害。  相似文献

几种冰草属披碱草属牧草种群地上生物量形成规律的研究   总被引:1,自引:0,他引:1  
本文从种群生态的层次,研究了在宁夏盐池干草原向荒漠草原过渡地带冰草属、披碱草属6种牧草种群地上生物量的形成规律。结果表明,供试牧草种群高度生长有明显季节性。与其它5种牧草相比,俄罗斯野麦草生长发育早而快,其种群地上生物量及有效营养物量生长季内的变化曲线均呈单峰式,在乳熟期最大值分别为141.3和86.0g/m2。此外,本文还指出了牧草的最佳利用时期,并在牧草生产上提出一些建议。  相似文献

The chromosomal behaviour at meiosis in pollen mother cells of 33 sample materials from 4 species in Agropyron Gaerm. distributed over various regions in China were observed. We. found that B chromosomes were mainly-present in A. mongolicum Keng (2n=2x=14) and were absent in the tetraploid ,A. cristarum (L.) Gaertn. (2n=4x=28) and ,A. michnoi Roshev. (2n=4x=28). Of all A. desertorum (Fisch.) Schult. (2n=4x=28) populations, they were present in 40%. The pairing between even numbers of B chromosomes usually occured at diakinesis where as uneven numbers of B chromosomes at metaphase 1. B chromosomes at anaphase I separated at random. The Fagging Bs tended to reach two poles by chromatids through precocious division of Bs. The pairing and chiasma frequencies between A chromosomes were increased by the larger numbers of Bs. The relationship between B chromosomes and species ploidy, the homeology among Bs, etc were discussed.  相似文献

用不同浓度的NaCl溶液处理蒙古冰草幼苗后,测定了不同时间胁迫下植株地上部分鲜重和离子含量。结果表明:轻度盐胁迫对蒙古冰草幼苗的生长有一定的促进作用,而中度和重度盐胁迫对幼苗生长有明显的抑制作用。经分析认为,轻度盐胁迫下蒙古冰草可以将Na+等有害离子截流在根部,抑制其向地上部分的运输,而中度和重度盐胁迫下过多的Na+等有害离子运输到地上代谢活跃部分,限制了对K+、Ca2+等的吸收,造成植株的营养亏缺,从而引起植株的伤害。  相似文献

MYB转录因子是一类与植物抗逆相关的蛋白家族,通过激活植物抗逆应答基因的表达而提高其抗逆性。为挖掘蒙古冰草(Agropyron mongolicum)抗旱相关的MYB基因,本研究在已有转录组测序数据的基础上,用Plant TFDB数据库中的Prediction和Blast对MYB基因筛选及保守结构域分析,通过生物学方法对筛选出的MYB转录因子进行分析,预测其分子量和等电点等理化性质、基序、二级结构和三级结构;并用MEGA 7.0.21与已报道具有抗旱功能的29个MYB蛋白进行多序列比对分析并构建系统进化树。结果表明:蒙古冰草中筛选出9个MYB基因,蛋白大小在113~576 aa,分子量和等电点分别介于12.83~61.52 kD和4.68~9.80 kD之间,都为不稳定的亲水性蛋白;基序预测得到2个特征基序,最长的基序含50个氨基酸;α-螺旋和无规则卷曲是主要的二级结构,三级结构中都含有螺旋-转角-螺旋结构;进化树分析中,38个基因很好的聚为两类,其中Unigene 25843、Unigene 42380、Unigene 52355、Unigene 54016分别于AtMYB33、AtMYB61、Os MYB48-1、AtMYB20在同一分支上,高度同源,推测从蒙古冰草中筛选得到的MYB基因参与干旱胁迫应答,该研究为下一步蒙古冰草MYB基因功能的研究提供帮助。  相似文献

The role of reactive oxygen species (ROS) in the defense of nearly isogenic lines of common wheat (Triticum aestivum L., cv. Thatcher) with the genes of resistance to brown rust introgressed from Agropyron species was studied using light microscopy. This disease is induced by the fungus Puccinia triticina Erikss. The presence of superoxide anion in the sites of infection was detected with the dye nitro blue tetrazolium. In addition, we studied fungus development on plants treated with the inhibitor of Ca2+-channels, verapamil, disturbing penetration into the cells of Ca2+ required for ROS generation. During fungus development in the immune line with the Lr38 resistance gene (from A. intermedium (Host) Beuv.), oxidative burst developed at the sites of contacts of appressoria with stomata and exerted a fungicidic effect. When ROS generation was suppressed, the fungus developed haustoria in the mesophyll cells. In plants with the Lr19 gene (from A. elongatum (Host) Beuv.), only moderate amount of superoxide anion accumulated on the cell walls of stomatal guard cells and in the infection structures when the fungus penetrated into the substomatal cavity and in mesophyll cells. In plants with the Lr24 gene (from A. elongatum), superoxide anion was detected only around haustoria. Suppression of ROS generation in plants harboring the Lr19 and Lr24 genes did not affect fungus entrance into the substomatal cavity but facilitated penetration of haustoria into the mesophyll cells. At the same time, in the lines with the Lr1 gene (from T. aestivum), cytological examination did not detect O 2 ? accumulation in plant cells, whereas treatment with verapamil enhanced mycelium development. In all lines, the suppression of oxidative burst slowed the development of hypersensitive response.  相似文献

采用基因组原位杂交(GISH)检测和染色体组成分析方法,对大穗多花小麦新种质4844后代的15个株系进行遗传分析。结果发现,4844-12是1个稳定的异附加系,4844-2和4844-8是稳定的异代换系;对异代换系进行SSR分析表明,代换系中小麦的6D染色体被1对P染色体代换,说明这对冰草染色体与小麦6D染色体有部分同源关系,由此确定4844中的冰草染色体为6P;同时筛选出冰草6P染色体的4个SSR标记。  相似文献

在内蒙古自治区多伦县中科院植物所生态恢复试验站,选择优势种克氏针茅(Stipa krylovii) 和冰草(Agropyron cristatum),使用Li-6400便携式光合测定系统观测光合生理生态特征的生长季变化(DOY155-265)。结果表明:水分是本区域植物生长的最重要的限制因子。冰草与针茅受5cm和20cm表层土壤含水率的影响较大。水分是本区域植物生长的最重要的限制因子。冰草与针茅受5cm和20cm表层土壤含水率的影响较大。植物的光合速率和蒸腾速率受自身的气孔导度限制,光合速率和蒸腾速率随气孔导度的增大而增大,冰草这一特征较为明显,即冰草对环境响应更为敏感。作为优势种冰草和针茅的生态适应策略并不相同,针对当地现有环境条件,针茅更具生存优势。随着全球环境变化,干旱胁迫对草原生态系统产生了重大影响,使得这些地区成为响应环境变化的敏感区域,通过对半干旱温带草原优势种克氏针茅和冰草对光、水等环境资源的利用及之间的关系研究,有助于我们对全球环境变化提出本区域合适的应对策略。  相似文献

本文对普通小麦与长穗偃麦草(Elytrigia elongata=Agropyron elongatum.2n=70)杂交选育出来的5个八倍体小偃麦的染色体组进行了研究。通过八倍体小偃麦与普通小麦杂交,八倍体小偃麦相互间杂交,观察了杂种F_1花粉母细胞减数分裂行为。根据观察结果,讨论了长穗偃麦草染色体组的构成,认为长穗偃麦草的染色体组为E_1E_2F_2F_2N较为合适。在此基础上,确定了5个八倍体小偃麦的染色体组:7430为ABDE_1,68为ABDF_1,693为ABDF_1,7631为ABDF_2,784为ABDN。另外,还讨论了八倍体小偃麦染色组的重组问题。  相似文献
