In this study, efficacy of seven different commercial disinfectant preparations was investigated against characteristic bacteria of a poultry slaughterhouse in Ankara (Turkey) by using paper disc-agar diffusion method and surface effectiveness test. According to paper disc-agar diffusion method, some disinfectants had wide efficacy against the test bacteria. The disinfectant effectiveness generally increased by the increasing of disinfectant concentration. However, some disinfectants were ineffective even though at their highest concentrations. The most effective disinfectant was B which contains QAC as active agent.Staphylococcus spp.,Staphyloccus aureus andEscherichia coli were the most sensitive bacteria to the disinfectants. Some pathogenic isolates, especiallySalmonella spp. andCampylobacter spp., were the most resistant ones to many of the disinfectants tested. The results of paper disc-agar diffusion method indicated the importance of characteristic bacterial strains of food plants as the test bacteria for disinfectant efficacy tests. Conversely of paper disc-agar diffusion method, all disinfectants were effective against the isolates by surface effectiveness test depending on exposure time. The disinfectants, except A and F, produced at least 3 log unit reduction during 5 min of exposure. However, all disinfectants at their lowest concentrations were effective against all tested bacteria during 15 and 30 min by this test.  相似文献

The incidence of candidiasis among immunocompromised patients and emergence of antimycotics resistant strains has increased significantly. The aims of this study were: to examine the in vitro activity of antimycotics and biocides against Candida clinical isolates; to detect cross-resistance of fungi to these preparations and to estimate whether disinfectants applied in hospital areas are active against clinical Candida isolates. In vitro susceptibility of 102 Candida isolates to eight antimycotics was examined by Etest and ATB Fungus. Sensitivity of these strains to four disinfectants and an antiseptic agent was tested according to EN 1275:2005. Amphotericin B, caspofungin and 5-fluorocytosine were the most effective antimycotics against all Candida isolates. Resistance to itraconazole and fluconazole was observed among C. krusei and C. glabrata. The MICs (Minimal Inhibitory Concentrations) for ketoconazole, voriconazole and posaconazole against Candida albicans ranged: 0.003 - >32 μg/ml and one strain was resistant to three agents tested. All analysed Candida strains were sensitive to biocides containing either chlorine, aldehyde, alcohol mixtures, glucoprotamin or chlorhexidine gluconate with isopropanol. Sensitivity to these agents was observed at concentrations lower than those concentrations recommended by manufacturers to achieve proper biocidal activity to those preparations. Our data suggest that these disinfectants can be effectively applied in clinical wards to prevent nosocomial Candida infections.  相似文献

As part of the simultaneous fermentation of both glucose and xylose to ethanol by a coculture process, compatibilities between xylose-fermenting yeasts and glucose-fermenting species were investigated. Among the Saccharomyces species tested, none inhibited growth of the xylose-fermenting yeasts. By contrast, many xylose-fermenting yeasts, among the 11 tested, exerted an inhibitory effect on growth of the selected Saccharomyces species. Killer character was demonstrated in three strains of Pichia stipitis. Such strains, despite their high fermentative performances, cannot be used to ferment D-xylose in association with the selected Saccharomyces species. From compatibility tests between xylose-fermenting yeasts and Saccharomyces species, pairs of microorganisms suitable for simultaneous xylose and glucose fermentations by coculture are proposed. Strains associated in the coculture process are distinguished by their resistance to mitochondrial inhibitors. The xylose-fermenting yeasts are able to grow on media containing erythromycin (1 g/L) or diuron (50 mg/L), whereas the Saccharomyces species are inhibited by these mitochondrial inhibitors.  相似文献

The focus on hygiene in the food industry has resulted in an increasing use of chemical disinfection and it has been speculated that this will impose a selective pressure and contribute to the emergence of disinfectant-resistant microorganisms. The frequency of strains with a low-level resistance to quaternary ammonium compounds (QAC) is relatively high for Listeria monocytogenes (10%), Staphylococcus spp. (13%) and Pseudomonas spp. (30%) and lower for lactic acid bacteria (1.5%) and coliforms (1%) isolated from food and food processing industry. In general, bacteria isolated after disinfection are more resistant and represent a potential food safety or food spoilage problem. Adaptation to disinfectants may be accompanied by cross-resistance to related disinfectants. We have recently found a genetic linkage between resistance to QAC and antibiotics in food associated staphylococci, and there is a growing concern about cross-resistance between antibiotics and disinfectants.Disinfectant resistance can in most cases be prevented by effective cleaning and disinfection procedures. More effort should be made to avoid build-up of resistance in food production environments.  相似文献

Nineteen out of 214 strains of microorganisms including moulds, bacteria, actinomycetes and yeasts, which were selected from the culture collection of our laboratory were found to produce the substances which flocculate Saccharomyces cerevisiae AJ4005 (baker’s yeast). Among them are Aspergillus sojae, Anixiella reticulata, Geotrichum candidum, Eupenicillium crustaceus, Circinella sydowi, Monascus anka, Sordaria fimicola, Pseudomonas fluorescens, Staphylococcus aureus, Corynebacterium brevicale, Brevibacterium insectiphilum, Streptomyces vinaceus.

The flocculants in the culture broth of these microorganisms precipitated readily by addition of acetone, and flocculated various microorganisms nonspecifically. The flocculant produced by Asp. sojae AJ7002 sedimented activated sludge well.  相似文献

This work constitutes a preliminary study in which the capacity of non-Saccharomyces yeasts isolated from ancient distilleries as bio-control agents against moulds and in the treatment of waste waters contaminated by heavy metals—i.e. bio-remediation—is shown. In the first control assays, antagonist effect between non-Saccharomyces yeasts, their extracts and supernatants against some moulds, analysing the plausible (not exhaustive) involved factors were qualitatively verified. In addition, two enzymatic degrading properties of cell wall plant polymers, quitinolitic and pectinolitic, were screened. Finally, their use as agents of bio-remediation of three heavy metals (cadmium, chromium and lead) was analysed semi-quantitatively. The results showed that all isolates belonging to Pichia species effectively inhibited all moulds assayed. Moreover, P. kudriavzevii is a good candidate for both bio-control and bio-remediation because it inhibited moulds and accumulated the major proportion of the three tested metals.  相似文献

Antimicrobial properties of olive leaf extract on some yeast were examined in this study. Fresh olive leaf extracts were prepared using various solvents (water, ethanol, acetone, ethyl acetate) in Soxhlet apparatus. Antimicrobial effects of these extacts were tested againstSaccharomyces cerevisiae ATCC 9763,Schizosaccharomyces pombe, Saccharomyces uvarum, Candida oleophila, Metschnikowia fructicola andKloeckera apiculata. The antifungal activities of these extracts were tested by the disc diffusion assay, minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC). All extracts showed various degrees of antifungal effects with 10–28 μg/ml MIC, 20–48 μg/ml MFC and 1.5–9.3 mm inhibitory zone values against yeasts utilised, except water. The results indicated that the tested yeasts were sensitive to acetone and ethyl acetate extracts. It was determined thatSaccharomyces cerevisiae ATCC 9763 was the most resistant among the yeasts.  相似文献

Lager beer is the most consumed alcoholic beverage in the world. Its production process is marked by a fermentation conducted at low (8 to 15°C) temperatures and by the use of Saccharomyces pastorianus, an interspecific hybrid between Saccharomyces cerevisiae and the cold-tolerant Saccharomyces eubayanus. Recent whole-genome-sequencing efforts revealed that the currently available lager yeasts belong to one of only two archetypes, “Saaz” and “Frohberg.” This limited genetic variation likely reflects that all lager yeasts descend from only two separate interspecific hybridization events, which may also explain the relatively limited aromatic diversity between the available lager beer yeasts compared to, for example, wine and ale beer yeasts. In this study, 31 novel interspecific yeast hybrids were developed, resulting from large-scale robot-assisted selection and breeding between carefully selected strains of S. cerevisiae (six strains) and S. eubayanus (two strains). Interestingly, many of the resulting hybrids showed a broader temperature tolerance than their parental strains and reference S. pastorianus yeasts. Moreover, they combined a high fermentation capacity with a desirable aroma profile in laboratory-scale lager beer fermentations, thereby successfully enriching the currently available lager yeast biodiversity. Pilot-scale trials further confirmed the industrial potential of these hybrids and identified one strain, hybrid H29, which combines a fast fermentation, high attenuation, and the production of a complex, desirable fruity aroma.  相似文献

Aims: Mycoplasma is minute bacteria that can be found ubiquitously in the environment and also in human, animal and plant tissues. In addition to their public health importance as aetiological agents of infections and possible association with certain cancers, mycoplasma is a major contamination concern in biotechnology. These bacterial cells are very small, can form biofilms and survive for extended periods of time when dried onto surfaces. Despite these concerns, there is little information concerning their resistance to currently used disinfection methods. The objective of this study was to evaluate commonly used biocidal treatments against three representative mycoplasma species. Methods and Results: Mycoplasma was dried onto stainless steel coupons and exposed to decontamination products. All strains survived drying and any significant viability loss because of the test method (including neutralization), as demonstrated by a ≤0·5 log10 for each tested species. The quaternary ammonium compound (QAC) tested presented poor efficacy, whereas 70% ethanol was fully efficient with complete inactivation after 5‐min exposure. Alkaline cleaner formulations presented increasing efficacy when tested at 0·2, 0·4 and 0·8% concentrations, with complete kill observed at 0·8% of two products tested. Decontamination with vaporized (gaseous) hydrogen peroxide (VHP) was very efficient at concentrations used for room and small enclosures decontamination (180–1200 ppm with various time exposures), as well as for device sterilization applications. Conclusions: Ethanol and alkaline detergent formulations were particularly efficient against mycoplasma, but a QAC formulation was not. VHP in room disinfection and device sterilization applications was effective against all mycoplasma species tested. Significance and Impact of the Study: Mycoplasma can provide resistance to environmental factors (such as drying) and disinfectants. Further studies are required to confirm the effectiveness of other disinfectants and the mechanisms of mycoplasma resistance.  相似文献

Methicillin-resistant Staphylococcus aureus (MRSA) originating from swine is concerning for public health, but an understanding of the emergence and persistence of MRSA in nursery herds is lacking. The aim of this study was to determine whether MRSA in nursery pigs is associated with particular herd-level parameters, including the use of antimicrobials, disinfectants, and heavy metals, which may be driving the selection and persistence of antimicrobial resistance. Nasal cultures for MRSA were completed for 390 pigs from 26 farms at the end of the suckling phase and again at 3 weeks postweaning. Herd-level information was collected, and a random subset of MRSA isolates was screened for resistance to zinc and quaternary ammonium compounds (QACs). Multivariate analysis revealed that in-feed concentrations of zinc (P < 0.001) and frequent disinfection of nursery pens (P < 0.001) are associated with MRSA shedding in nursery pigs. Furthermore, 62.5% (25/40) of MRSA isolates carried the zinc resistance gene czrC and demonstrated decreased susceptibility to zinc. All MRSA isolates carried at least 1 QAC resistance gene. The most common genotype was qacG qacH smr, which occurred in 32.5% (13/40) of isolates. Seven isolates (17.5%) demonstrated a significant tolerance to benzalkonium chloride, indicating a potential to survive commercial QAC exposure in the presence of organic matter. Overall, these findings indicate that high levels of in-feed zinc and QAC-based disinfectants are important drivers in the selection and persistence of MRSA in commercial swine herds, and these agents may be coselecting for other antimicrobial resistance genes.  相似文献

In vitro biodegradation experiments were done using some probiotic microorganisms. DifferentSaccharomyces cerevisiae, Lactobacilli andBacilli strains were tested for their ability to degrade Nivalenol (NIV), Deoxynivalenol (DON), Diacetoxyscirpenol (DAS), T2-Toxin and Ochratoxin A (OTA). The concentrations of selected mycotoxins were in the range of natural occurring toxin contaminations (1ppm for NIV and DON, 500ppb for DAS and T-2 and 50ppb for OTA). No alteration of concentrations could be registered for the tested trichothecenes. The best results could be achieved in experiments with OTA by up to 94% detoxification. Influence of toxins on colony forming unit of all tested microorganisms were recorded. Especially T-2 toxin and DAS have a slowing effect on growth of some strains.  相似文献

The increased prevalence of Clostridium difficile infection (CDI) has coincided with enhanced transmissibility and severity of disease, which is often linked to two distinct clonal lineages designated PCR-ribotype 027 and 017 responsible for CDI outbreaks in the USA, Europe and Asia. We assessed sporulation and susceptibility of three PCR-ribotypes; 012, 017 and 027 to four classes of disinfectants; chlorine releasing agents (CRAs), peroxygens, quaternary ammonium compounds (QAC) and biguanides. The 017 PCR-ribotype, showed the highest sporulation frequency under these test conditions. The oxidizing biocides and CRAs were the most efficacious in decontamination of C. difficile vegetative cells and spores, the efficacy of the CRAs were concentration dependent irrespective of PCR-ribotype. However, there were differences observed in the susceptibility of the PCR-ribotypes, independent of the concentrations tested for Virkon®, Newgenn®, Proceine 40® and Hibiscrub®. Whereas, for Steri7® and Biocleanse® the difference observed between the disinfectants were dependent on both PCR-ribotype and concentration. The oxidizing agent Perasafe® was consistently efficacious across all three PCR ribotypes at varying concentrations; with a consistent five Log10 reduction in spore titre. The PCR-ribotype and concentration dependent differences in the efficacy of the disinfectants in this study indicate that disinfectant choice is a factor for llimiting the survival and transmission of C. difficile spores in healthcare settings.  相似文献

The coffee fermentation is characterized by the presence of different microorganisms belonging to the groups of bacteria, fungi and yeast. The objectives of this work were to select pectinolytic microorganisms isolated from coffee fermentations and evaluate their performance on coffee pulp culture medium. The yeasts and bacteria isolates were evaluated for their activity of polygalacturonase (PG), pectin lyase (PL) and pectin methylesterase (PME) and metabolites production. Among 127 yeasts isolates and 189 bacterial isolates, 15 were pre-selected based on their ability to produce PL and organic compounds. These isolates were strains identified as Bacillus cereus, Bacillus megaterium, Bacillus subtilis, Candida parapsilosis, Pichia caribbica, Pichia guilliermondii and Saccharomyces cerevisiae. When cultivated in Coffee peel and pulp media in single culture or two by two mixed inocula, different behavior concerning to PME, PL and PG were found. The two principal components PC1 and PC2 accounted for 45.27 and 32.02 % of the total variance. UFLA CN727 and UFLA CN731 strains were grouped in the positive part of PC1 being characterized by 1,2-propanediol, hexanoic acid, decanoic acid, nonanoic acid and ethyl acetate. The UFLA CN448 and UFLA CN724 strains were grouped in the negative part of PC1 and were mainly characterized by guaiacol, butyric acid and citronellol. S. cerevisiae UFLACN727, P. guilliermondii UFLACN731 and C. parapsilosis UFLACN448 isolates are promising candidates to be tested in future studies as coffee starter cultures.  相似文献

Three different yeasts of the species Saccharomyces cerevisiae ferment ethyl alcohol to acetaldehyde aerobically and produce “flor” character in a wine medium with submerged culture techniques. The heavy metal resistance of yeasts isolated from sugary substrates such as orange, palm wine and pineapple and identified asSaccharomyces carlsbergensis andS. cerevisiae was studied. The yeast isolates were tested against different concentrations of cadmium, copper, manganese, silver and zinc salts ranging from 1 to 20 mmol/L. Local yeasts showed resistance to 3–15 mmol/L cadmium, 18–20 copper, 16–20 manganese, 1–9 silver and 16–19 for zinc. The significance of the results is discussed in relation to the effects of heavy metals on growth of microorganisms and selection of yeasts for the brewing industry in Nigeria.  相似文献

The concentrations and relative ratios of various aroma compounds produced by fermenting yeast cells are essential for the sensory quality of many fermented foods, including beer, bread, wine, and sake. Since the production of these aroma-active compounds varies highly among different yeast strains, careful selection of variants with optimal aromatic profiles is of crucial importance for a high-quality end product. This study evaluates the production of different aroma-active compounds in 301 different Saccharomyces cerevisiae, Saccharomyces paradoxus, and Saccharomyces pastorianus yeast strains. Our results show that the production of key aroma compounds like isoamyl acetate and ethyl acetate varies by an order of magnitude between natural yeasts, with the concentrations of some compounds showing significant positive correlation, whereas others vary independently. Targeted hybridization of some of the best aroma-producing strains yielded 46 intraspecific hybrids, of which some show a distinct heterosis (hybrid vigor) effect and produce up to 45% more isoamyl acetate than the best parental strains while retaining their overall fermentation performance. Together, our results demonstrate the potential of large-scale outbreeding to obtain superior industrial yeasts that are directly applicable for commercial use.  相似文献

Eight culture-collection yeast strains of various species and five newly isolated strains were tested for both growth in olive oil extraction effluents and fermentation of the sugars in the same media. The culture-collection yeast strains did not grow in an effluent containing 2·86% sugar (w/v), 8 g/litre phenolic substances, 4·58 g/litre titratable acidity and pH 4·96, whereas the newly isolated strains of Torulopsis sp. MK-1, Saccharomyces norbensis MC-1, S. oleaceus MC-2 and S. oleaginosus grew well and fermented the sugars. In the medium mentioned above, they produced alcohol in amounts of 1·63 to 1·38%, respectively. None of the yeasts grew in an olive oil extraction effluent vacuum-concentrated to over 13–14% of dry matter. The strain of T. sp. MK-1 showed a hogher stability.  相似文献

The active substances such as cardiotonics or toxics which were produced in the cultured broths or contained in the mycellia of microorganisms were examined by Ito’s assay method for cardiotonica using the embryonic heart of Japanese killi-fish.

There were some strains obtained from actinomycetes whose cultured broths or methanol mycelial extracts showed these activities, but fewer from moulds, yeasts and bacteria.  相似文献

Mixtures of yeasts were tested for theirability to control Penicillium expansum andBotrytis cinerea on Red Delicious apple fruits. The occurrence of synergistic or antagonisticinteractions between yeast strains in differentmixtures was also evaluated. Two strains ofRhodotorula (R. glutinis SL 1 and R. glutinisSL 30) and two strains of Cryptococcus (C. albidus SL 43 and C. laurentii SL 62) were selected fordeveloping yeasts mixtures.The R. glutinis SL 1–R. glutinis SL 30 mixtureexhibited a lower effectiveness than eachstrain alone, against both moulds. Othermixtures (R. glutinis SL 1–C. albidus SL 43 and R. glutinis SL 30–C. albidus SL 43) showedsynergism against P. expansum but not against B. cinerea. The R. glutinis SL 1–C. laurentii SL62 mixture was the only mixture which showedsynergism against gray mould. There was not anymixture, which showed high effectivenessagainst both moulds at the same time. Differentresults could be explained by the dynamics ofthe population of the yeasts.By using yeast mixtures, it was possible toimprove biocontrol without increasing theamount of antagonists applied. The synergismobserved could be useful in enhancingbiological control.  相似文献
