Diversity of yeasts associated with Panax ginseng   总被引:1,自引:0,他引:1  
Biodiversity of yeasts was investigated in the ginseng cultivation field. Among 34 isolates tested in this study, 26 isolates belonged to the hymenomycetous yeast group. These 26 strains were classified into 12 species including four new-species candidates that did not have clear affiliation to any established species. Seven isolates among the remaining strains were classified into three ascomycetous yeast species, and one isolate was identified as a urediniomycetous yeast species.  相似文献

【目的】开展羊卓雍措水体可培养酵母菌多样性研究,探究影响其多样性的主要理化因子。【方法】采用膜过滤平置培养法分离纯化酵母菌,并结合rRNA ITS区域序列分析与经典分类法对酵母菌菌株进行鉴定。运用SPSS 20.0和CANOCO 5分析可培养酵母菌多样性及其与理化因子之间的关系。【结果】羊卓雍措水体可培养酵母菌为16个属25个种,优势属为Vishniacozyma,优势物种为Vishniacozyma victoriae。Pearson相关系数显示,pH、电导率、总溶解固体量、盐度与各样点可培养酵母菌种数和属数呈显著正相关;总磷与各样点及各区域可培养酵母菌属数呈显著负相关,与各样点种数呈显著负相关。冗余分析显示,pH和总磷是影响羊卓雍措水体可培养酵母菌分布的主要环境因子。【结论】羊卓雍措水体酵母菌资源比较丰富且存在明显的空间异质性,人类活动对酵母菌分布有较大影响。酵母菌分布与人类活动的关系值得进一步研究。  相似文献

Mechanisms underlying adaptation of various yeasts to salt stress were summarized. Stress response involves modulation of enzymatic activities and changes in gene expression. Elevated salinity of the environment can be regarded as a two-factor stress process that includes an osmotic component leading to loss of cellular turgor and a toxic component inhibiting a set of functions due to an increase in the intracellular concentration of Na+. Adaptation of yeast cells to these stress conditions obligatorily involves the accumulation of osmotically active compounds (mainly glycerol) to counterbalance an increased external osmotic pressure and the modification of plasma membrane transport systems to extrude Na+ from the cell.  相似文献

Mastitis is one of the most serious problems in the dairy cattle farms. The great majority of the cases are caused by bacteria, but lately there have been an increasing number of reports about cases of mycotic etiology. The objective of this work was to characterize the yeasts and yeast-like fungi associated with milk of cows with mastitis. Milk samples (n = 248) from a dairy belt situated around the region of Passo Fundo, hinterland of the state of Rio Grande do Sul, Southern Brazil, were analyzed. Aliquots of 0.1 ml of milk were inoculated on yeast extract-malta agar with chloramphenicol. After a period of incubation of 3-5 days at 22-25 degrees C, the counting of the morphologically distinct colonies was performed, as well as the isolation and identification through phenotypical and physiological criteria. It was possible to isolate 68 yeast species from 43 (17.3%) of the samples. The most frequent genera were Candida (37.9%), Pichia (19.1%), Cryptococcus (10.3%) and Rhodotorula (10.3%).  相似文献

为解析酱香型白酒酿造酒醅中酵母菌的菌群结构,获取酒醅中的主要酵母菌,采用高通量测序法分析酱香型白酒酒醅中酵母菌多样性及主要功能菌群,同时采用可培养分离方法获取酒醅中酵母菌活性菌株。从酱香型白酒下沙至五轮次酒醅中共检出59个属、129个种的酵母菌,分离得到酵母菌活性菌株41种,检测到的酵母菌种类与获得的酵母菌活菌在各香型白酒中最多。不同时期酒醅中的酵母菌种类和数量差异明显,其中下沙、造沙轮次以Pichia kudriavzevii为绝对优势酵母菌;一至五轮次随着轮次的递增,酒醅中优势酵母菌的种类增多,其中主要的优势酵母菌有Pichia kudriavzevii、Pichia manshurica、Zygosaccharomyces bailii、Saccharomyces cerevisiae、Candida apicola。酱香型白酒酒醅中蕴藏着极其丰富的酵母菌资源,对酵母菌菌群结构的解析有助于科学地认识酱香型白酒酿造过程中产酒与风味代谢机理,为发酵过程的调控提供一定依据。  相似文献

【背景】星云湖属云南高原湖泊,对其酵母菌的多样性研究和产类胡萝卜素菌株的筛选有助于开发利用星云湖酵母菌资源。【目的】研究云南星云湖酵母菌群落结构多样性,筛选和评价产类胡萝卜素活性菌株。【方法】采用膜过滤平置培养对星云湖水样进行酵母菌分离;应用26S rRNA D1/D2区域序列分析和形态特征及生理生化指标相结合,对分离获得的酵母菌进行鉴定;运用软件BIO-DAP分析酵母菌类群的多样性;采用酸热法提取类胡萝卜素,紫外分光光度计测定类胡萝卜素含量。【结果】从星云湖湖水中分离得到797株酵母菌,鉴定为18个属37个种。胶红酵母(Rhodotorula mucilaginosa)和Naganishia albida是优势种,分别占总菌株数的36.64%和28.86%;有28株酵母菌类胡萝卜素产量可达200μg/g细胞干重以上;有4株类胡萝卜素产量可达300μg/g细胞干重以上;粘红酵母(Rhodotorula glutinis) 2株、胶红酵母(Rhodotorula mucilaginosa)和Rhodosporidiobolus fluviale各1株。【结论】云南高原湖泊星云湖的酵母菌具有丰富的多样性,湖泊的营养化程度以及地理差异影响酵母菌群落的结构;红冬孢酵母属(Rhodosporidiobolus)和红酵母属(Rhodotorula)是星云湖类胡萝卜素产量可达300μg/g细胞干重以上的主要类群。有进一步开发研究的价值。  相似文献

采用过滤涂皿的方法对云南程海湖冬季湖水样品中的酵母菌进行分离,通过26S r DNA D1/D2区域序列分析并结合形态观察和生理生化测试对分离获得的菌株进行鉴定,同时采用胞外酶定性筛选培养基进行产酶活性筛选,分析冬季程海湖酵母菌的多样性及胞外酶活性。结果从程海湖中分离获得171株酵母菌,鉴定为14个属22个种和2个潜在的新分类单元;优势属是红冬孢酵母属Rhodosporidium、红酵母属Rhodotorula和隐球酵母属Cryptococcus;优势种是红冬孢酵母Rhodosporidium kratochvilovae和斯鲁菲亚红酵母Rhodotorula slooffiae;湖北半部的样点CH2的多样性指数(H′=1.5945)和丰富度指数(R=2.7576)均最高,均匀度指数较高的是湖北部的样点CH1(J=0.8531);样点之间种群差异大,相似性低。大多数酵母菌株具有产1种以上的胞外酶。  相似文献

A total of 198 yeasts were isolated from 140 samples collected from 7 mangrove forests in 4 provinces of Thailand, and were found to belong to 30 genera, 45 described species and at least 12 undescribed species based on their 26S rRNA (D1/D2 domain) gene sequence. The most prevalent species was Candida tropicalis, followed by Candida pseudolambica and Rhodosporidium paludigena. Lipid accumulation, as determined by Nile red staining, of the isolated yeasts revealed that 69 and 18 strains were positive and strongly positive, respectively, while quantitative analysis of the intracellular lipid accumulated in the latter indicated that 10 of these strains, Pseudozyma tsukubaensis (YWT7-2 and YWT7-3), Rhodotorula sphaerocarpa (YWW6-1 and SFL14-1SF), Saitozyma podzolica (YWT1-1, NS3-3 and NS10-2), Prototheca zopfii var. hydrocarbonea OMS6-1 and Prototheca sp. (YMTW3-1 and YMTS5-2), were oleaginous. In this study we found that under nitrogen depletion condition (155 C/N ratio) Pseudozyma tsukubaensis YWT7-2 accumulated the highest level of intracellular lipid at 32.4% (w/w, dry cell weight), with a broadly similar fatty acid composition to that in palm oil.  相似文献

【背景】西南高山葡萄酒产区的甘孜州产区,具有生产优质葡萄酒的自然禀赋。【目的】研究四川甘孜州葡萄酒产区真核微生物种类多样性、本土酿酒酵母遗传多样性,以及商业酵母对本土酵母多样性的影响。【方法】利用ITS高通量测序技术对赤霞珠接种发酵和自然发酵过程中的微生物进行多样性分析,并利用Interdelta指纹图谱分析法,对经过26S rRNA基因鉴定的野生酿酒酵母基因型进行分类。【结果】ITS测序结果显示,接种发酵和自然发酵各时期均注释到7个科7个属的酵母,通过Interdelta指纹图谱分析发现甘孜州产区的酿酒酵母共有5种基因型。该产区酿酒酵母的6株代表菌株与我国其他产区109株酿酒酵母的进化树分析结果显示,均与来自北京产区的酿酒酵母菌株亲缘关系更近。【结论】甘孜州葡萄酒子产区酵母资源丰富,表现出较高的微生物多样性和中等程度的本土酿酒酵母基因型多样性,为后续优良本土酵母菌株的筛选奠定基础。  相似文献

The killer properties of yeasts isolated from olive brines were examined in the absence and presence of sodium chloride in concentrations of up to 6% (wt/vol). An apparent enhancement of the killing action as the salt concentration increased, as well as changes in the spectra of activity against selected target strains, was observed in a few strains. Culture filtrates from killer strains grown at different NaCl concentrations (0, 3, or 6% [wt/vol]) were tested against sensitive yeasts cultivated under the same conditions. While the sensitivity of the target strain greatly increased in the presence of salt, no significant effect on toxin production was noticed.  相似文献

Although dissimilarities in cranial and post-cranial morphology among African pygmies groups have been recognized, comparative studies on skull morphology usually pull all pygmies together assuming that morphological characters are similar among them and different with respect to other populations. The main aim of this study is to compare cranial morphology between African pygmies and non-pygmies populations from Equatorial Africa derived from both the Eastern and the Western regions in order to test if the greatest morphological difference is obtained in the comparison between pygmies and non-pygmies. Thirty three-dimensional (3D) landmarks registered with Microscribe in four cranial samples (Western and Eastern pygmies and non-pygmies) were obtained. Multivariate analysis (generalized Procrustes analysis, Mahalanobis distances, multivariate regression) and complementary dimensions of size were evaluated with ANOVA and post hoc LSD. Results suggest that important cranial shape differentiation does occur between pygmies and non-pygmies but also between Eastern and Western populations and that size changes and allometries do not affect similarly Eastern and Western pygmies. Therefore, our findings raise serious doubt about the fact to consider African pygmies as a homogenous group in studies on skull morphology. Differences in cranial morphology among pygmies would suggest differentiation after divergence. Although not directly related to skull differentiation, the diversity among pygmies would probably suggest that the process responsible for reduced stature occurred after the split of the ancestors of modern Eastern and Western pygmies.  相似文献

Catalases from 16 different organisms including representatives from all three phylogenetic clades were purified and characterized to provide a comparative picture of their respective properties. Collectively the enzymes presented a diverse range of activities and properties. Specific activities ranged from 20,700 to 273,800 units per milligram of protein and maximal turnover rates ranged from 54,000 to 833,000 per second. The effective concentrations of common catalase inhibitors, cyanide, azide, hydroxylamine, aminotriazole, and mercaptoethanol, varied over a 100- to 1000-fold concentration range, and a broad range of sensitivities to heat inactivation was observed. Michaelis-Menten kinetics were approximately followed only at the low substrate concentrations. At high H(2)O(2) concentrations, inactivation of small-subunit enzymes resulted in lower velocities than what were predicted, whereas large-subunit enzymes had velocities higher than predicted. Kinetic constants such as K(m) and V(max) for catalases must be labeled as "apparent."  相似文献

Summary The effect of various NaCl concentrations on respiration and fermentation rates in cells with or without added glucose as exogenous substrate as well as on respiratory quotients was determined for Debaryomyces hansenii, Saccharomyces cerevisiae, Cryptococcus albidus, and Candida zeylanoides, all yeasts isolated from marine environment. A given strain had about the same respiratory and fermentatory intensity at 0% and 4% NaCl (w/v). A further increase considerably reduced the oxygen uptake or CO2-evolution. D. hansenii was the most NaCl tolerant yeast tested, giving about 10% activity still at a concentration of 24% NaCl, whether the activities of whole cells or cell homogenates were determined. For S. cerevisiae or Cr. albidus the respiratory activity was reduced to about the same degree at 16% NaCl for whole cells, at 12% NaCl for homogenates of Cr. albidus. A somewhat higher NaCl concentration was evidently tolerated for respiration and fermentation than for growth, very obvious in the case of C. zeylanoides.The minimum values for water activity (a w) permitting 10% respiration activity were higher when produced by electrolytes (NaCl, KCl, or Na2SO4), lower when due to sugars (metabolizable glucose or non-metabolizable lactose) and lowest when due to glycerol. The a w per se was evidently not solely decisive for the limitation of respiration activity.Attempts were made to assess an effect of high NaCl concentrations on the glucose uptake.The potassium content was higher in cells of the highly halotolerant D. hansenii than in those of the other yeasts and decreased with the increase in external, consequently in internal, Na+ concentration. The decrease in K+ content can presumably only proceed to a certain extent, below which the ability for growth and respiration was lost.  相似文献
