Isolation and characterization of alpha-foetoprotein from foetal pigs

• 1.1. Porcine α-foetoprotein, APP, was isolated from serum of pig foetuses using preparative electrophoresis and irnmunosorbent techniques.
• 2.2. The purity of the isolated AFP was controlled by polyacrylamide gel electrophoresis, electro-immunoassay and by injection of isolated AFP into rabbits.
• 3.3. During the preparation of AFP from foetal pig serum an unexpected α₁-globulin appeared as a contaminating protein.
• 4.4. The molecular weight, isoelectric point and sedimentation coefficient were found to be 70,000, 4–61 and 4–56 respectively. No microheterogeneity as regards the physicochemical parameters investigated could be seen. The results were compared with those reported for some other mammals.

     

A peptide inhibitor for S-adenosyl-l-methionine-dependent transmethylation reactions: Purification and characterization

• 1.1. A proteinaceous inhibitor for S-adenosyl-l-methionine (AdoMet)-dependent transmethylation reactions has been purified to apparent homogeneity from rat liver cytosolic fraction.
• 2.2. The peptide was made up of 29 amino acid residues with a molecular weight of 2,584. Glycine accounted for 52% of the total amino acids.
• 3.3. Employing AdoMet: protein-carboxyl O-methyltransferase (Protein methylase II) and bovine serum γ-globulin as in vitro substrate, the mode of inhibition was found to be non-competitive with K_i value of 1.9 × 10⁻⁸ M.
• 4.4. When the inhibitor was present in the reaction mixture together with S-adenosyl-l-homocysteine (AdoHcy), which is a competitive inhibitor for AdoMet, the extent of inhibition exceeded that exerted by each individual inhibitor alone, suggesting that the sites of the inhibitors on the enzyme molecule are different.
• 5.5. Almost a stoichiometric relationship exists between the enzyme and the inhibitor molecule, the ratio being approx one.

     

Influence of environmental factors on the deep and skin temperature in the European bison,Bison bonasus (L.)

• 1.1. In 43 European bison divided into three groups (Group A, 3–8-month-old calves; Group B, 18-month-7-year-old young bison; Group C, 12–24-year-old bison) the rectal, humerus region and abdomen region temperatures were measured.
• 2.2. The experiments were carried out in winter months, from mid-December to mid-March.
• 3.3. The mean rectal temperatures changed from 38.55°C in calves to 38.15°C in the oldest bison.
• 4.4. The mean temperatures of the humerus region changed from 20.69°C in calves to 21.49°C in older bison.
• 5.5. The mean temperatures of the abdomen region changed from 20.79°C in calves to 22.17°C in older bison (Gr. B).
• 6.6. The cluster analysis divided the bison into four groups named hot, warm, cool and cold bison.
• 7.7. Only air temperature measured 2 m above the ground and snow cover influenced the integrated bison temperature. Age, sex and mass as well as some environmental factors had no influence.
• 8.8. Measurements made 1 to nearly 4hr after a bison's death showed a drop in rectal temperature and mostly increases in temperatures of the humerus and abdomen regions.

• 1.1. Serum from the Pacific hagfish,Eptatretus stouti,contains a complement-like protein (CLP).
• 2.2. CLP from unfractionated hagfish serum and from affinity-purified preparations binds to yeast cell surfaces.
• 3.3. Incubation with CLP enhances the phagocytosis of yeast by hagfish leukocytes.
• 4.4. CLP-mediated opsonization can be inhibited by anti-CLP antibody, EDTA, d(+)mannose and l(+)rhamnose.
• 5.5. Additional opsomic factors are also in hagfish serum.

     

Three digestive tract endocrine peptides and the circannual cycle of body weight in Spermophilus lateralis

• 1.1. Thirty-one male golden-mantled ground squirrels were divided into four physiological groups: low wt summer, medium wt summer, high wt summer and hibernation period. A second group of 10 females was divided into two groups: hibernation period at low T_b and hibernation period during a periodic arousal.
• 2.2. Blood serum, pancreas and antral stomach region were collected from each animal.
• 3.3. The serum was analysed by radioimmunoassay for pancreatic polypeptide immunoreactivity, the pancreas for pancreatic polypeptide and somatostatin immunoreactivity and the antral region of the stomach for gastrin immunoreactivity.
• 4.4. Significant between-stage differences (P < 0.05) were found in serum pancreatic polypeptide concentration and in pancreatic somatostatin content.

     

The binding curves in relation to protein concentration. The interaction of human serum albumin with phthalein dyes and azo-dyes

• 1.1. The binding curves of three phthalein dyes and two azo-dyes were measured in a concentration range of human serum albumin from 0.1 to 10%.
• 2.2. Contrary to other authors' findings, the courses of the binding curves appeared independent of protein concentration in all cases.
• 3.3. This discrepancy may be due to precision of the experimental techniques.

     

Purification and characterization of monkey albumin and proalbumin

• 1.1. Albumin purified from rhesus monkey (MSA) shows immunological cross-reactivity with human serum albumin (HSA) by RIA.
• 2.2. The amino-terminal sequence of MSA shows a high degree of homology to HSA.
• 3.3. Thirty minutes after injection of radioactive leucine directly into the portal vein, albumin was purified chemically from the liver, kidneys and serum.
• 4.4. At this time, 15% of the label was incorporated into liver homogenate protein.
• 5.5. A highly labelled immunoreactive albumin form was purified from liver to constant specific radioactivity and separated from tissue and serum albumin.
• 6.6. The specific radioactivity of this proalbumin was 36-times higher than the specific radioactivity of albumin in liver tissue.
• 7.7. These similarities to HSA suggest that this non human primate species can serve as a useful model of human albumin synthesis in vivo.

Induction of autoantibodies against spermatozoa by injection of allogeneic sperm in the Nile Tilapia,Oreochromis niloticus

• 1.1. The sperm-agglutinating factor (SAF) could be induced in the serum of male Nile tilapias, Oreochromis niloticus, by injection of allogeneic sperm.
• 2.2. Only one class of molecules was demonstrated to be SAF in the serum.
• 3.3. Analysis on purified SAF revealed it to be a tetrameric molecule of IgM with a mol. wt of 760kD.
• 4.4. Cross reaction of the IgM with sperm of other teleost species suggests that sperm-specific surface antigens may be in evolution.

Cloning and characterization of rabbit pancreatic colipase

• 1.1. Among the digestive enzymes synthesized by pancreas, lipase is the principle lipolytic enzyme which hydrolyses dietary glycerides.
• 2.2. For its action it requires a coenzyme, colipase.
• 3.3. The molecular mechanisms of the interaction of these two are not fully understood.
• 4.4. Further, molecular events that regulate and influence lipid absorption are ill denned.
• 5.5. The rabbit is the conventional animal model for the study of lipid absorption. We have undertaken the molecular cloning, and characterization of rabbit pancreatic colipase, the coenzyme for pancreatic lipase.
• 6.6. Colipase has been cloned from a gt 11 library of an adult rabbit pancreatic cDNA by probing with an oligonucleotide derived from human colipase sequence.
• 7.7. The total reading frame consists of 321 nucleotides coding for 90 amino acids of the functional protein and 17 nucleotides of the leader peptide.
• 8.8. Northern blot analysis revealed a distinct band around 0.5kb. Comparison with other species revealed an over all homology of 75% at the nucleotide level.
• 9.9. At the amino acid level highest conservation is observed at the lipase-binding region (AA 53–73).
• 10.10. Rabbit enzyme also retained the N-terminal pentapeptide of it preform.
• 11.11. The regions of homology and conservation may aid to define the sites of interaction of colipase with lipase.

     

The effect of Cryptocotyle lingua infection on the blood volume of Littorina littorea

• 1.1. Blood volumes of uninfeeted and parasitized individuals of L. littorea have been measured using inulin dilution.
• 2.2. Uninfected specimens have a blood volume of 64% and parasitized specimens a blood volume of 38% of the flesh weight.
• 3.3. Two different methods of inulin analysis give essentially similar results.

     

Domain structure of the 90-kDa stress protein: Heparin- and antibody-binding domain

• 1.1. To understand the physiological roles of the 90-kDa stress protein (HSP90), we investigated the heparin- and antibody-binding domains of the protein.
• 2.2. For heparin-binding sites, HSP90 was digested completely with trypsin, and the digests were applied to a heparin-Sepharose column and eluted with 1.0 M NaCl, followed by 8.0 M urea.
• 3.3. Each elutant was purified by a reverse-phase C₁₈ column.
• 4.4. Two peptides from the NaCl-eluted fraction and no peptide from the urea-eluted fraction were purified.
• 5.5. The purified peptides were sequenced by an automated peptide sequencer.
• 6.6. One of the heparin-binding sites was present between Leu-362 and Arg-365; another was present between Leu-645 and Lys-648.
• 7.7. These two peptides were basic and considerably hydrophilic.
• 8.8. For antibody-binding sites, HSP90 was mildly digested with trypsin, electrophoresed on SDS-polyacrylamide gels and transferred to PVDF membranes.
• 9.9. The four bound of the trypsin fragments could be sequenced with a peptide sequencer.
• 10.10. There was only one antibody-binding peptide, 38 kDa, starting from Pro-2. The others showed no cross-reactivity with the antibody and started from Leu-283.
• 11.11. Therefore, the epitopes of HSP90 are present between Pro-2 and Leu-282.
• 12.12. The heparin-binding sites are present from the middle region of the HSP90 molecule, and the antigen sites are at the N-terminal domain.

     

Investigation of phycocyanin-membrane interaction: Promotion of membrane interactions

• 1.1. In a continuing investigation of phycocyanin-membrane surface interaction, fluorescence quenching experiments were performed with a mixture of two populations of fluorescence probe-encapsulated phospholipid bilayer vesicles in the presence and absence of phycocyanin.
• 2.2. These membrane vesicles were prepared with 1,2-dimyristoyl phosphatidylcholine (DMPC), cholesterol and a probe molecule.
• 3.3. A fluorophore was encapsulated in one population of membrane vesicles, while a quencher was encapsulated in another population of membrane vesicles.
• 4.4. The result was compared with those of experiments in the presence of other biomolecules, including albumin, cytochrome c, hemoglobin, myoglobin or RNA.
• 5.5. Interestingly, a one-third reduction of the fluorescence intensity was observed in the mixture of these two populations of membrane vesicles in phycocyanin's presence.
• 6.6. In contrast, the other biomolecules caused no significant reduction in the fluorescence intensity.
• 7.7. These findings were evidence of a phycocyanin-induced membrane perturbation.
• 8.8. This was further demonstrated by a phycocyanin-induced change in the thermotropic behavior of DMPC vesicles, as measured by differential scanning microcalorimetry.
• 9.9. Such a unique property of phycocyanin is believed to be associated with its known membrane surface-interacting character.
• 10.10. A possible phycocyanin-modulated membrane-membrane interaction was discussed.

     

The lysozyme of the plaice Pleuronectes platessa L.

Lysozyme from the serum of the plaice, Pleuronectes platessa L., has been purified 78-fold with chitin coated cellulose.

• 1.2. Further purification on CM-cellulose yielded a single band on acrylamide electrophoresis, exhibiting lysozyme activity.
• 2.3. The quantitative amino acid composition of plaice serum lysozyme is reported.
• 3.4. The mol. wt is identical with hen egg white lysozyme.
• 4.5. A method is described for identifying fractions with lysozyme activity in polyacrylamide gels.

Seasonal changes of water,electrolytes and aldosterone levels in blood serum,brain and kidney of the Egyptian cobra “Naja haje haje”

 

• 1.The levels of water, Na, K, Ca and Mg in blood serum, brain and kidney and aldosterone level in blood of Naja haje haje were studied during the different phases of the annual cycle.
• 2.The water content in the tissues studied displayed only minor changes as the animals passed from one phase to the other.
• 3.A significant increase in Na was recorded in the brain during the different phases indicating a depressed sodium pump, whereas the blood Na level showed a significant decrease during hibernation.
• 4.K increased in blood serum, brain and kidney during hibernation, while a nonsignificant decrease was found in blood serum during arousal. The brain may act as a potassium reservoir.
• 5.An increase in Ca and Mg concentration was recorded in blood serum, brain and kidney during prehibernation and hibernation. The data suggested a homeostatic function in the transport and metabolism of these cations.
• 6.Aldosterone exhibited a highly significant decrease especially during hibernation. The aldosterone regulation of ionic composition is discussed.
• 7.Na/K and Ca/Mg ratios in the brain may explain the decreased excitability during winter torpor.
