  • 4.4. Preincubation in the presence of ALA or PBG protected the enzymic activity of ALA-D, PBGase and deaminase against URO I-inactivation both under u.v. light and in the dark.
  • 5.5. Photoinactivating action of URO I would be mediated by reactive oxygen species generated by the excited porphyrin after its absorption of light. Dark inactivation, in aerobiosis, can also be partly mediated by amino acid oxidation, although to a lesser extent than that observed under u.v. light.

  • 2.2. Muscle homogenates expressed very low enzyme activities and the presence of ATP was necessary to detect AMP deaminase. The specific activity and substrate affinity of the purified enzyme were also very low.
  • 3.3. The purified prawn muscle AMP deaminase was contaminated by contractile proteins, one of the major contaminants being actin.

  • 1.1. To date, only a few authors have assayed the agglutinic activity of marine algae against fish erythrocytes, and in these cases, mainly against freshwater fish.
  • 2.2. For the first time, the hemagglutinic activity of 70 seaweeds (29 brown, 37 red and four green algae) against erythrocytes of 16 seaflsh species is reported.
  • 3.3. The presence of agglutinins was demonstrated in 100% of algae assayed, against at least one of the different types of erythrocytes tested.
  • 4.4. The results obtained confirm the presence of receptors for algae agglutinins on the surface of the erythrocytes of the fish studied.
  • 5.5. This could be useful in establishing the origins of fish populations, as these serological differences could distinguish between populations of cultivated and wild fish.

  • 1.1. The presence of gastrin-like immunoreactive materials in intestinal tissues of the earthworm Lumbricus terrestris has been demonstrated by means of radioimmunoassay technique.
  • 2.2. The greatest activity coincides with the areas of the intestines with high digestive enzyme activities. No gastrin-like activities were detected in nerve tissues.
  • 3.3. The results also indicate that earthworm gut tissues may contain different forms of gastrin or gastrin-like peptides.
  • 4.4. The absence of gastrin-like activity in earthworm nerve tissues may have significant phylogenetic implication for the proposed gastrin-cholecystokinin origin in neuronal elements of invertebrates.

  • 1.1. Sialyltransferase released into the medium during the incubation of rat jejunal slices in serum-free buffer, was susceptible to proteolytic degradation. Heat inactivated horse serum or its antiproteolytic heparin-binding fraction was found to be necessary in determining the activity of sialyltransferase released (Nadkarni et al., 1991).
  • 2.2. In the present study, we have shown that heat inactivated rat serum (HRS) or its antiproteolytic heparin-binding fraction (HBF) had a role in determining the sialyltransferase activity released during jejunal slice incubations.
  • 3.3. Galactosyltransferase was also released during incubations, but was not proteolytically degraded and the presence of HRS or HBF in incubations did not alter the levels of galactosyltransferase activity released.
  • 4.4. Trypsin activity in serum-free incubation medium was higher compared to medium containing HRS.
  • 5.5. Addition of serum-free medium obtained from 4 hr incubations of the jejunal slices, to medium obtained from parallel incubations done in the presence of HRS, caused inhibition of sialyl- but not galactosyltransferase activity.
  • 6.6. In jejunal homogenates stored at −20°C, sialyltransferase activity was decreased during 0–45 days of storage, whereas galactosyltransferase activity remained fairly stable for upto 56 days.
  • 7.7. Inclusion of HRS or HBF in homogenates resulted in higher sialyl- but not galactosyltransferase activity compared to serum-free homogenate samples.
  • 8.8. The results suggest that HRS or its antiproteolytic heparin-binding proteins have a role in determining the sialyltransferase activity released from the jejunal slices. In contrast galactosyltransferase released was not susceptible to proteolysis, and HRS or HBF was not required to express its activity.

  • 1.1. Photo-stimulation of larval adductor activity of Anodonta cygnea L. was investigated. Photosensitization takes place in the presence of different xanthene dyes such as eosin, erythrosin, rose bengal and phloxine.
  • 2.2. The sensitor-effect and energy-efficiency of the stain-fluorescence are correlated. The more effective stain is that which emits the lesser part of the absorbed light. This determines an activity arrangement: rose bengal >erythrosin⪢eosin.
  • 3.3. The reaction can be influenced by the temperature of dark-incubation and has a latency time. This suggests a dark-complex formation with biopolymers.
  • 4.4. It is probable that the photoreaction can take place without involving nervous elements.

  • 1.1. Three calcium-binding proteins have been purified from Ehrlich ascites tumor cells.
  • 2.2. They were identified by amino acid sequence analysis on selected fragments obtained by tryptic digestion.
  • 3.3. The proteins belong to the annexin family and were identified as annexins II, III and V.
  • 4.4. Antibodies raised against the proteins were used to examine for their presence in a number of murine tissues.

  • 1.1. A quick and simple procedure is described for purifying kallikrein from human whole saliva. The enzyme has been purified about 2700-fold with a yield of approx. 30%.
  • 2.2. The procedure is based on the immediate fractionation of saliva by ion exchange chromatography. This is followed by a combination of affinity and high performance liquid chromatography.
  • 3.3. The results indicate that another protein component binds to the enzyme at pH 8.0.
  • 4.4. The homogeneity of the enzyme has been demonstrated by gel electrophoresis in the absence as well as in the presence of sodium dodecylsulfate.
  • 5.5. A mol. wt of 40,100±1800 has been calculated from gel electrophores is experiments.
  • 6.6. Sedimentation equilibrium in an analytical ultracentrifuge gave a mol. wt of 39,700.
  • 7.7. The amino acid composition has been determined and it confirms that the enzyme has a low isoelectric point.
  • 8.8. The presence of tryptophan has been demonstrated by absorption and fluorescence spectroscopy.

  • 1.1. In a continuing investigation of phycocyanin-membrane surface interaction, fluorescence quenching experiments were performed with a mixture of two populations of fluorescence probe-encapsulated phospholipid bilayer vesicles in the presence and absence of phycocyanin.
  • 2.2. These membrane vesicles were prepared with 1,2-dimyristoyl phosphatidylcholine (DMPC), cholesterol and a probe molecule.
  • 3.3. A fluorophore was encapsulated in one population of membrane vesicles, while a quencher was encapsulated in another population of membrane vesicles.
  • 4.4. The result was compared with those of experiments in the presence of other biomolecules, including albumin, cytochrome c, hemoglobin, myoglobin or RNA.
  • 5.5. Interestingly, a one-third reduction of the fluorescence intensity was observed in the mixture of these two populations of membrane vesicles in phycocyanin's presence.
  • 6.6. In contrast, the other biomolecules caused no significant reduction in the fluorescence intensity.
  • 7.7. These findings were evidence of a phycocyanin-induced membrane perturbation.
  • 8.8. This was further demonstrated by a phycocyanin-induced change in the thermotropic behavior of DMPC vesicles, as measured by differential scanning microcalorimetry.
  • 9.9. Such a unique property of phycocyanin is believed to be associated with its known membrane surface-interacting character.
  • 10.10. A possible phycocyanin-modulated membrane-membrane interaction was discussed.

  • 1.1. Role of NADP-glutamate dehydrogenase in the depletion of citrate was analyzed using permeabilized yeast cells.
  • 2.2. Citrate was converted to 2-oxoglutarate, which was then metabolized to glutamate by NADP-glutamate dehydrogenase in the presence of ammonium ion.
  • 3.3. Formation of 2-oxoglutarate plus glutamate was in good agreement with the concentration of citrate decreased. Glutamate formation can be a good indicator of the depletion of citrate, because 70% of the citrate decreased was converted to glutamate.
  • 4.4. Glycolytic activity was closely correlated with the decrease in citrate under the in situ conditions.
  • 5.5. NADP-glutamate dehydrogenase increased in anaerobically grown yeast cells.
  • 6.6. An effective depletion of citrate by increased synthesis of NADP-glutamate dehydrogenase can explain the lowered mechanism of citrate causing glycolytic stimulation under the anaerobic growth conditions of yeast.
