Exercise increases metabolic rate and the production of reactive oxygen species (ROS) but also elevates protein turnover. ROS cause damage to macromolecules (e.g., proteins) and thereby contribute to aging. Protein turnover removes and replaces damaged proteins. The balance between these two responses may underlie beneficial effects of physical activity on aging. Effects of lifelong exercise on antioxidant enzyme activities and fractional synthesis rate of protein (FSRP) were examined at various ages (2-26 mo) in heart, liver, and muscle of mice that had been selectively bred for high wheel-running activity, housed with (S+) or without (S-) a running wheel, and their random-bred controls (C+) housed with running wheels. FSRP decreased with age and increased in muscle of young, but not old, activity-selected mice. Enzyme activity of superoxide dismutase and glutathione peroxidase decreased with age and showed a peak at 10 mo of age in liver. Selection for wheel-running activity did not affect antioxidant enzyme activity. Daily energy expenditure correlated positively with antioxidant levels in liver. This might indicate that oxidative stress (ROS production) increases with metabolic rate, driving upregulation of antioxidant enzymes. Alternatively, the elevated energy expenditure may reflect the energetic cost of elevated protection, consistent with the disposable-soma hypothesis and with other studies showing positive links between energy expenditure and life span. Long-term elevations in voluntary exercise did not result in elevations in antioxidant enzyme activities or protein synthesis rates. 相似文献

The effect of exercise on oxidant stress and on alterations in antioxidant defense in elderly has been investigated extensively. However, the impact of regularly performed long-term physical activity starting from adulthood and prolonged up to the old age is not yet clear. We have investigated the changes in the activities of antioxidant enzymes - superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) - and lipid peroxidation in various tissues of rats which had performed (old-trained) or had not performed (old-control) regular swimming exercise for one year. These animals were compared with young-sedentary rats. Increased lipid peroxidation was observed with ageing in all tissues (heart, liver, kidney, striated muscle) and swimming had no additional effect on this elevation of lipid peroxidation. Heart and striated muscle SOD activites, and striated muscle CAT activity increased as a consequence of ageing, whereas kidney and liver CAT activities, as well as GPx activities in kidney, liver, lung and heart were significantly decreased compared to young controls. Lung and heart SOD, liver CAT activities as well as GPx activities in liver, lung and heart were increased significantly in rats which performed exercise during ageing, compared to the old-control group. These findings suggest that lifelong exercise can improve the antioxidant defense in many tissues without constituting any additional oxidant stress. 相似文献

To identify the effects of a tower-climbing exercise on bone marrow heme biosynthesis and hematological status in rats given glucocorticoid-injections as a model of aging, 29 male Sprague-Dawley rats, 10 weeks of age, were assigned to three groups: a saline control (C, n = 9), a glucocorticoid-sedentary (GS, n = 10) group, and a glucocorticoid-exercise (GE, n = 10) group. The GS and GE groups were given 2 mg/kg prednisolone daily, and the C group was given 2 ml of saline daily, subcutaneously. Each group was meal-fed commercial rat chow isoenergetically and given free access to water for 8 weeks. The GE group were allowed to climb a 200-cm tower to drink water from a bottle set at the top of it. Weight gain during the 8-week experimental period was greater in the C group than in the GS and GE groups. The gastrocnemius and tibialis anterior muscles were heavier in the C group than in other groups. The hematological parameters were not influenced by glucocorticoid administration with or without climbing exercise. Bone marrow delta-aminolevulinic acid dehydratase activity was significantly higher in the GE group than in the C group. These results suggest that 8 weeks of climbing exercise increases heme biosynthesis without alteration of hematological status in rats given glucocorticoid-injections. Resistance exercise may be a preventive therapy for iron-deficiency anemia associated with aging. 相似文献

The relationship between voluntary distance running and antioxidant capacity was studied in rats after three weeks voluntary running. Hydroxyl radical level, reduced glutathione level, activities of glutathione reductase and superoxide dismutase were measured in plasma, liver, brain, soleus and gastrocnemius white muscle. Hydroxyl radical level of liver negatively correlated with the running distance (r=-0.616, P<0.001). The reduced glutathione levels of liver and brain increased depending on the running distance and the correlation was confirmed between them in liver (r=0.638, P<0.01) and brain (r=0.766, P<0.001). The hydroxyl radical level in liver positively correlated with the activities of glutathione reductase (r=0.464, P<0.05) and superoxide dismutase (r=0.549, P<0.05). A significant positive correlation was detected between the hydroxyl radical level and superoxide dismutase activity in brain (r=0.488, P<0.05). These results demonstrate that physical activity correlates well with glutathione level and anti-oxidant enzyme activities in liver, suggesting a close relation between physical activity and induction of antioxidant systems. 相似文献

Diabetes mellitus is associated with diabetic impairment of testicular function, ultimately leading to reduced fertility.
Its etiology may involve oxidative damage by reactive oxygen substances, and protection against this damage can be offered
by antioxidant supplementation. The aim of this study was to investigate the effects of intraperitoneal administration of
vitamin C and E, selenium (Se), and vitamin E plus Se (COM) on concentrations of lipid peroxide (as malondialdehyde; MDA),
reduced glutathione (GSH), and vitamin E concentrations and glutathione peroxidase (GSH-Px) activity in the testes of rats
with diabetes induced by streptozotocin (STZ). Sixty groups were used (10 animals each) and these animals were initially allocated
to two groups: control group and diabetic group. The diabetic group was subdivided into five groups as follows: diabetic control
(DC), vitamin E, Se, COM, and vitamin C. Animals in the DC group and vitamin C, vitamin E, Se, and COM groups were made diabetic
by the injection of STZ on 4 d after an injection of vitamins C and E, Se, and COM. Those vitamins and Se were also administered
for 21 consecutive days. The MDA, vitamin E, GSH levels, and GSH-Px activities in testes were determined.
Although the vitamin E concentration was higher in the control than in the DC group, the MDA levels were found to be lower
in the control than in the DC group. The MDA levels in the testes samples of vitamin C, vitamin E, Se, and COM groups were
lower than the DC group. However, GSH-Px activity and GSH levels in the testes were not significantly different between the
control and DC groups. Vitamin E concentrations in the vitamin C, vitamin E, Se, and COM groups and GSH levels and GSH-Px
activities in the Se, COM, and vitamin C groups were higher than either the control or DC group.
The results indicate that reactive oxygen substances may be involved in possible testicular complications in diabetes of rats.
Administration of vitamins C and E and Se reduced the testicular lipid peroxidation; these vitamins and Se had significant
protective effects on testes of rats against oxidative damage in diabetes.

Several animal models have been developed to study the pathogenesis of hypertension. Deoxycorticosterone acetate (DOCA) salt induced hypertensive rats are adrenal models used to mimic human Conn's syndrome. Because previous studies showed a beneficial effect of chronic exercise (swimming) on the development of arterial hypertension in spontaneously hypertensive rats (which appears similar to human essential hypertension), we decided to evaluate the effects of swimming on DOCA-salt induced hypertension and liver antioxidant status. Therefore, the aim of this experiment was to study whether the swim training would improve hypertension and liver antioxidant status in DOCA-salt rats. DOCA-salt rats and control Sprague-Dawley rats were trained to swim 1 h/day, 5 days/week for 6 weeks and were sacrificed 48 h after the last exercise period. Systolic blood pressure was recorded before the sacrifice, and liver antioxidant status was evaluated in hepatic homogenates after the sacrifice. Swim exercise did not decrease systolic blood pressure in control and DOCA-salt rats but induced changes in liver activities of antioxidant enzymes, showing that exercise provoked liver oxidative stress in control and DOCA-salt rats. In comparison with our previous studies using spontaneously hypertensive rats, we conclude that the beneficial effects of chronic exercise on systolic blood pressure in rats are dependent on strain and the type of experimental hypertension. 相似文献

Although testosterone was declared a controlled substance in 1990, the potential for steroid dependence is largely unexplored. The present study used food-induced drinking with oral testosterone self-administration in hamsters to determine (1) the dose-response for testosterone reward, (2) links between testosterone self-administration and voluntary exercise, and (3) factors predicting individual differences in androgen intake. Testosterone (1-4 mg/ml) was presented in aqueous solution 3 h/day for 35 days, with and without food. At 3 mg/ml, testosterone maintained fluid intake, even without food (3.0 +/- 0.2 ml/3h). At 4 mg/ml, fluid intake declined to 2.4 +/- 0.3 ml/3h in the presence of food. However, no dose-response relationship between testosterone self-administration and reward was observed. To test the interaction of testosterone and exercise, males drinking testosterone (RUN + T) or vehicle received a running wheel. Additional males self-administered testosterone without exercise. Testosterone intake correlated positively with exercise and negatively with body weight in RUN + T males. Experiment 3 determined the relationship between testosterone self-administration and mating or saccharin preference. There were no statistically significant correlations. However, testosterone self-administration increased mating behavior. These data demonstrate that testosterone is a mild reinforcer. Although preference for androgens is not predicted by mating or saccharin intake, testosterone intake is linked with voluntary exercise. 相似文献

The purpose of the present study was to investigate the effect of short-term endurance training on plasma total antioxidant status (TAS) and on in vitro platelet aggregation and ATP release. Blood samples were collected from the abdominal aorta of rats following short-term treadmill exercise (25 m/min, 0 % grade, 30 min) for three consecutive days, as well as in non-exercised control group. Platelet aggregation and platelet ATP release were evaluated by impedance and bioluminescence techniques, respectively. Plasma TAS was measured spectrophotometrically. Plasma TAS was higher and ADP-induced platelet ATP release was lower in the short-term training group with respect to the control group (p<0.001). Significant negative correlation (r = -0.56, p<0.05) was found between plasma TAS and ADP-induced platelet ATP release. Neither ADP- and collagen-induced maximum aggregation rate nor collagen-induced platelet ATP release were significantly different between the groups. According to these results, short-term training caused an alteration in platelet functions limited to the secretion response, which may be related to the oxidant/antioxidant balance changes favoring the antioxidants. The improved plasma total antioxidant capacity was possibly sufficient to prevent exercise-induced oxidative damage, and the adaptive response of platelets might be associated with enhanced antioxidant status. 相似文献

Recently we reported that ferric reducing ability of plasma (FRAP) assay, as an index of total antioxidant activity, increases
in growing rats in response to high dose of vitamin K. In this study, it was found that acetaminophen (APAP) can cause elevation
in FRAP in suckling and adult rats. This study was initiated to assess the contribution of individual antioxidant factors
on elevation in FRAP. A surge in FRAP, 1 h after high dose APAP (250 or 450 mg/kg BW) administration was recorded in both
young as well as adults. Whereas, low dose drug (25 mg/kg) failed to alter FRAP in both the age groups. Time-course studies
show that drug-dependent elevation in FRAP begin rapidly, reaching a maximum at 1 h (> 500%). Increased FRAP was associated
with a marked increase (∼14-fold) in plasma bilirubin, 6 h after drug administration at 450 mg/kg only in suckling rats. Similarly,
APAP-related increase in superoxide dismutase activity in erythrocytes was limited to young rats of both the age groups. Other
factors measured during this period viz., plasma uric acid, bilirubin and total protein together with catalase activity of
erythrocytes remained unchanged in treated rats. Under these circumstances, APAP-related depletion in liver glutathione was
almost similar in both the age groups. During a 12 h study, the concentration of lipid peroxidation products, in liver of
treated groups remained within the levels of respective controls. The endpoint hepatotoxic effects of APAP was almost similar
in both the age groups, suggesting that like adults, immature rats can cope with toxic effects of APAP owing to their drug-dependent

This study investigated the effects of epigallocatechin gallate (EGCG) on the growth performance and antioxidant capacity of 35-d-old broilers exposed to heat stress. Broilers, 14 d of age, were divided into four groups with six replicates per group (eight chickens/replicate). Thermoneutral group (Group TN) was fed the basal diet and maintained at 28°C for 24 h/d. The heat-stressed groups were housed at 35°C for 12 h/d and 28°C for 12 h/d and fed the basal diet supplemented with EGCG at 0, 300 and 600 mg/kg diet (Groups HS0, HS 300 and HS600, respectively). Compared with Group TN, heat-stressed groups showed significantly reduced gain, feed intake and serum total protein and glucose levels; inhibited serum alkaline phosphatase activities; and increased serum levels of uric acid, cholesterol and triglycerides and the activity of serum creatine kinase, lactate dehydrogenase and aspartate aminotransferase (p < 0.05). Compared with Group HS0, Group HS600 exhibited an increased gain and feed intake; and normalised blood parameters and enzyme activities. Compared with Group TN, the expression of antioxidant-related liver proteins was decreased in Group HS0 and increased in Groups HS300 and HS600 (p < 0.05). The results suggest that EGCG can improve the growth performance and alleviate the oxidant damage by modulating the antioxidant properties of broilers. 相似文献

In order to test for possible sex differences in endurance capacity, groups of young, physically active women (n = 6) and men (n = 7) performed bicycle ergometer exercise at 80% and 90% of their maximal oxygen uptakes (VO2 max). The groups were matched for age and physical activity habits. At 80% VO2 max the women performed significantly longer (P less than 0.05), 53.8 +/- 12.7 min vs 36.8 +/- 12.2 min, respectively (means +/- SD). Mid-exercise and terminal respiratory exchange ratio (R) values were significantly lower in women, suggesting a later occurrence of muscle glycogen depletion as a factor in their enhanced endurance. At 90% VO2 max the endurance times were similar for men and women, 21.2 +/- 10.3 min and 22.0 +/- 5.0 min, respectively. The blood lactate levels reached in these experiments were only marginally lower (mean differences 1.5 to 2 mmol X l-1) than those obtained at VO2 max, suggesting high lactate levels as a factor in exhaustion. The changes in body weight during the 80% experiments and the degree of hemoconcentration were not significantly different between men and women. 相似文献

The antioxidant activities of the thymoquinone-rich fraction (TQRF) extracted from Nigella sativa and its bioactive compound, thymoquinone (TQ), in rats with induced hypercholesterolemia were investigated. Rats were fed a semipurified diet supplemented with 1% (w/w) cholesterol and were treated with TQRF and TQ at dosages ranging from 0.5 to 1.5 g/kg and 20 to 100 mg/kg body wt, respectively, for 8 weeks. The hydroxyl radical (OH·)-scavenging activity of plasma samples collected from experimental rats was measured by electron spin resonance. The GenomeLab Genetic Analysis System was used to study the molecular mechanism that mediates the antioxidative properties of TQRF and TQ. Plasma total cholesterol and low-density-lipoprotein cholesterol levels were significantly decreased in the TQRF- and TQ-treated rats compared to untreated rats. Feeding rats a 1% cholesterol diet for 8 weeks resulted in a significant decrease in plasma antioxidant capacity, as measured by the capacity to scavenge hydroxyl radicals. However, rats treated with TQRF and TQ at various doses showed significant inhibitory activity toward the formation of OH· compared to untreated rats. Upon examination of liver RNA expression levels, treatment with TQRF and TQ caused the up-regulation of the superoxide dismutase 1 (SOD1), catalase, and glutathione peroxidase 2 (GPX) genes compared to untreated rats (P < 0.05). In support of this, liver antioxidant enzyme levels, including SOD1 and GPX, were also apparently increased in the TQRF- and TQ-treated rats compared to untreated rats (P < 0.05). In conclusion, TQRF and TQ effectively improved the plasma and liver antioxidant capacity and enhanced the expression of liver antioxidant genes of hypercholesterolemic rats. 相似文献

Nasturtium officinale R. Br. (Brassicaceae) has been used as a home remedy by the people of south eastern (SE) region of Iran as a medicinal plant. This therapeutical application has been attributed to Nasturtium officinale (N. officinale) antioxidant capacity which is mostly tested by means of cell-free assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP). In addition, the antioxidant effect of N. officinale extract has been investigated in hypercholesterolaemic rats in vivo. The results revealed that the extract has notable scavenging activity against DPPH radicals as well as potent reducing power in FRAP assay. Intragastric administration of N. officinale (500 mg/kg body weight per day) to groups of hypercholesterolaemic rats for 30 days lowered their blood total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol (LDL-C) levels by 37, 44 and 48%, respectively. However, the blood high density lipoprotein cholesterol (HDL-C) levels in the same treated rats increased by 16%. To evaluate the mechanism(s) of action, we studied the antioxidative potential of N. officinale extract in terms of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione reductase (GR) activities and also the level of reduced glutathione (GSH) in the liver tissues. In addition, hepatic tissue malondialdehyde level (MDA, an index of lipid peroxidation) was also determined. Under hypercholesterolaemic condition, hepatic MDA was increased. Moreover, our data indicated GSH depletion along with significant reduction in the activities of CAT and SOD in rats fed high-fat diet rats. On the other hand, significant elevation in the activities of GPx and GR were seen in the same group of rats. Treatment of hypercholesterolaemic rats with N. officinale extract significantly increased the GSH level along with enhanced CAT and SOD activities in liver tissues. Furthermore, N. officinale extract significantly decreased hepatic MDA as well as GPx and GR activities in plant-treated rats. Based on our data, it can be concluded that N. officinale has a high hypolipidaemic activity and this may be attributed to its antioxidative potential. 相似文献

There are large individual differences in the success rates of exercise intervention programs aimed at the prevention and treatment of obesity-related disorders. In the present study, we tested the hypothesis that differences in coping style may impact the success rates of these intervention programs. We tested insulin responses before and after voluntary wheel running in both passive (insulin resistant) Roman Low Avoidance (RLA) and proactive (insulin sensitive) Roman High Avoidance (RHA) rats using intravenous glucose tolerance tests (IVGTTs). To control for a potential difference between voluntary and forced exercise, we also included RLA and RHA rats that were subjected to forced running. We found the following: 1) when given the opportunity to run voluntarily in a running wheel, passive RLA rats run more than proactively than RHA rats; 2) voluntary exercise leads to a normalization of insulin responses during an IVGTTs in RLA rats; and 3) there were no behavioral and physiological differences in efficacy between voluntary and forced running. We conclude that exercise, both forced and voluntary, is a successful lifestyle intervention for the treatment of hyperinsulinemia, especially in individuals with a passive coping style. 相似文献
