Mycoplasma mobile cells glide on solid surfaces such as glass with a fast and continuous motion in the direction of the membrane protrusion (head-like structure) at one cell pole. To examine its cell-surface movement, a latex bead was attached to a cell and behavior in gliding was monitored. The bead was carried without movement relative to the cell body, suggesting that the cell does not roll around the cell axis and the surface movement is limited to a small area. A small percentage of cells showed an elongated head-like structure in an old batch culture. The head-like structure moved forward, sometimes leaving the cell body in one position, resulting in a stretching of this head-like structure. These results indicate that the head-like structure drags the cell body, leading us to conclude that the force for gliding is generated at the head-like structure.  相似文献

Several mycoplasma species are known to glide in the direction of the membrane protrusion (head-like structure), but the mechanism underlying this movement is entirely unknown. To identify proteins involved in the gliding mechanism, protein fractions of Mycoplasma mobile were analyzed for 10 gliding mutants isolated previously. One large protein (Gli349) was observed to be missing in a mutant m13 deficient in hemadsorption and glass binding. The predicted amino acid sequence indicated a 348,758-Da protein that was truncated at amino acid residue 1257 in the mutant. Immunofluorescence microscopy with a monoclonal antibody showed that Gli349 is localized at the head-like protrusion's base, which we designated the cell neck, and immunoelectron microscopy established that the Gli349 molecules are distributed all around this neck. The number of Gli349 molecules on a cell was estimated by immunoblot analysis to be 450 +/- 200. The antibody inhibited both the hemadsorption and glass binding of M. mobile. When the antibody was used to treat gliding mycoplasmas, the gliding speed and the extent of glass binding were inhibited to similar extents depending on the concentration of the antibody. This suggested that the Gli349 molecule is involved not only in glass binding for gliding but also in movement. To explain the present results, a model for the mechanical cycle of gliding is discussed.  相似文献

A new sulphur-containing natural alkaloid named microbiaeratin (1a) was isolated, together with the known microbiaeratinin (2, bacillamide) from the culture filtrate of Microbispora aerata strain IMBAS-11A. The organism was isolated from penguin excrements collected on the Antarctic Livingston Island. The structure was elucidated by one- and two-dimensional NMR experiments and mass spectrometric investigations.  相似文献

Little is known about the higher-order structure of prespliceosomal A complexes, in which pairing of the pre-mRNA's splice sites occurs. Here, human A complexes were isolated under physiological conditions by double-affinity selection. Purified complexes contained stoichiometric amounts of U1, U2 and pre-mRNA, and crosslinking studies indicated that these form concomitant base pairing interactions with one another. A complexes contained nearly all U1 and U2 proteins plus approximately 50 non-snRNP proteins. Unexpectedly, proteins of the hPrp19/CDC5 complex were also detected, even when A complexes were formed in the absence of U4/U6 snRNPs, demonstrating that they associate independent of the tri-snRNP. Double-affinity purification yielded structurally homogeneous A complexes as evidenced by electron microscopy, and allowed for the first time the generation of a three-dimensional structure. A complexes possess an asymmetric shape (approximately 260 x 200 x 195 angstroms) and contain a main body with various protruding elements, including a head-like domain and foot-like protrusions. Complexes isolated here are well suited for in vitro assembly studies to determine factor requirements for the A to B complex transition.  相似文献

An ethanol utilizing yeast, strain E-6, was isolated from leaves of a radish (Raphanus sativus). The taxonomic study of the organism showed it belongs to the genus Candida. When the organism was grown on ethanol, about 200 mg/liter of oily drops was formed on the surface of the culture. They were suggested to be a mixture of higher fatty acids by gas-chromatographical analysis. The main component of the acids was isolated as methyl ester by thin-layer chromatography. The chemical structure was examined by means of infrared, nuclear magnetic resonance, mass, and elementary analyses. The oxidation products of the component with permanganate-periodate and with nitric acid were determined. It was proved that the said main component was 15-hydroxy-9,12-octadecadienoic acid.  相似文献

The prevalence of Listeria monocytogenes in the environment of dairy farms was surveyed from December 1993 to June 1994 in one city of Hokkaido. L. monocytogenes was isolated from 3 out of 5 farms investigated. Serovar 4b organism was isolated from the brain stem of a cow from one farm which was clinically diagnosed as having listeriosis. The same serovar of L. monocytogenes was also isolated from the rectal contents of a healthy cow, straw on the floor, straw in the barn, and silage scattered around the silo from the same farm. At another farm, with no reported cases of bovine listeriosis, serovar 1/2 organism was isolated from the same types of samples as the above mentioned farm except from straw on the floor. The difference in the isolation rates of the organism from straw on the floor between the two farms (22%: 5/23 vs 0%: 0/24) is considered to be caused by the different feeding methods of silage between the two farms.  相似文献

Clostridium perfringens (a versatile pathogenic bacterium) secretes enterotoxins (the deltatoxin, virulent factor) and causes food borne gastroenteritis and gasgangrene. The organism was isolated and characterized from improperly cooked meat and poultry samples. The isolated organism showed multiple drug resistance indicating that the treatment is challenging. Hence, there is need for improved therapeutic agents. The rational design of improved therapeutics requires the crystal structure for the toxin. However, the structure for the toxin is not yet available in its native form. Thus, we modeled the toxin structure using α- hemolysin of Staphylococcus aureus (PDB: 3M4D chain A) as template. The docking of the toxin with the herbal extract curcumin (1,7-bis(4-hydroxy-3- methoxyphenyl)hepta-1,6-diene-3,5-dione) showed a binding energy of -8.6 Kcal/mol, in comparison to the known antibiotic Linezolid with binding energy of -6.1 Kcal/mol. This data finds application in the design and development of novel compounds against the deltatoxin from Clostridium perfringens.  相似文献

Aureobasidium pullulans (de Bary) Arnaud was isolated from different natural materials plant blossoms in particular. Elements of vegetative multiplication, structure of colonies and cultures in liquid media were analyzed in detail, leading to construction of the life cycle of this organism. Morphological polymorphism was found to be combined with the production of melanin and the polysaccharide pullulan. Morphological analysis served for a directed selection for studies of physiological properties of this organism and its practical application.  相似文献

A spoilage organism isolated from turbid beer is described. The bacterium was gram negative, catalase negative, strictly anaerobic, and rod shaped, having flagella only on one side of the cell. The main metabolic product was propionic acid. In addition acetic, succinic, and lactic acids and acetoin were formed. Malonate inhibited the production of propionic acid by the strain studied and by both Pectinatus and Propionibacterium strains. The guanine-plus-cytosine content of deoxyribonucleic acid was 36 mol%. Differences between this strain and Pectinatus strains were 2 to 5 percentage points. Immunofluorescent staining and gel diffusion precipitin tests revealed that the antigenic structure differed from those of Pectinatus strains. The isolated organism can, despite some differences, be regarded as belonging to the genus Pectinatus.  相似文献

This paper summarizes some structural characteristics of Potato virus X (PVX), the flexuous filamentous plant potexvirus. A model of PVX coat protein (CP) tertiary structure in the virion proposed on the basis of tritium planigraphy combined with predictions of the protein tertiary structure is described. A possible role of glycosylation and phosphorylation in the CP structure and function is discussed. Two forms of PVX virion disassembly are discussed: (i) the virion co-translational disassembly after PVX CP in situ phosphorylation and (ii) disassembly of PVX triggered by different factors after linear destabilization of the virion by binding of the PVX-coded movement protein (TGBp1) to one end of the polar CP-helix. Special emphasis was placed on a translational activation of encapsidated PVX RNA and rapid disassembly of TGBp1-PVX complexes into free RNA and CP. The results of experiments on the PVX CP repolymerization and PVX reconstitution are considered. In particular, the products assembled from PVX RNA, CP and TGBp1 were examined. Single-tailed particles were found with a helical, head-like structure consisting of helically arranged CP subunits located at the 5'-tail of RNA; the TGBp1 was bound to the end of the head. Translatable 'RNA-CP-TGBp1' complexes may represent the transport form of the PVX infection.  相似文献

A new antifungal antibiotic named acmycin was isolated from a soil streptomycete. Detailed comparative taxonomic studies showed that the organism differed from three related species of streptomycetes. The organism was referred to asStreptomyces sp. AC2. This study provides new information on the Fe uptake system capable of supporting growth of the organism. Pseudomonas fluorescens isolated from the rhizosphere of barley, a gramineous plant, produced a siderophore under iron-limiting conditions. Its chemical structure was identified as pyochelin, on the basis of 1H and 13C NMR data of a stable methyl ester derivative. The same iron-limiting conditions induced a new set of outer membrane proteins (75 and 55 kDa), consistent with a siderophore-mediated iron-uptake system.  相似文献

A hyphomyceteous fungus producing light-colored ramoconidia was isolated from midge (Clinodiplosis sp.) in southern China. In this study, this fungus was compared with 16 morphologically similar Cladosporium-like genera, but could not be assigned to any of them due to its distinct morphological characteristics, namely absence of sympodial conidiogenous cells and presence of swollen head-like conidiogenous regions. Moreover, on the basis of phylogenetic analyses of 18S ribosomal DNA (rDNA), 5.8S rDNA, and 28S rDNA, as well as genes for the RNA polymerase II subunit and the translation elongation factor 1-alpha, this fungus was identified as a member of the Ustilaginomycetes in Basidiomycota. Therefore, Capitulocladosporium clinodiplosidis gen. et sp. nov. is proposed to accommodate the new taxon, and its morphology is described and illustrated herein. The new genus is placed in Ustilaginomycetes but without further specification on order and family level (incertae sedis), since its teleomorph and lifestyle are currently unclear.  相似文献

Abstract The primary structure of the 16S rDNA of Acidothermus cellulolyticus , a cellulolytic, asporogenous rod isolated from a hot spring, was determined by automated sequence analysis. This Gram-variable organism is not related to members of other aerobic thermophilic genera found in similar environments, such as Thermus and Thermomicrobium , but clusters within the phylogenetically defined radiation of the actinomycetes where it represents an individual subline of descent.  相似文献

The gorgonian Paramuricea clavata is a benthic organism often included in conservation management plans since it creates complexity in the ecosystems and is extremely vulnerable to disturbances. Eight microsatellite markers isolated from an enriched genomic library were characterized in a total of 50 individuals from two north-western Mediterranean populations. All loci were polymorphic and the number of alleles per locus ranged between 2 and 14. Significant genetic differentiation was observed between populations. The polymorphic markers presently isolated will allow for the assessment of the spatial genetic structure between and within populations of this umbrella species of the Mediterranean Sea.  相似文献

The ability to hydrolyze collagen was studied in 262 cultures of spore-forming aerobic bacteria isolated from the soil, human and animal organism, from insects and also of collection strains. 53% of the studied cultures possessed the collagenolytic activity. 砂鼠利什曼原虫(Leishmania gerbilli)存在于我国西北甘肃某地的砂土鼠或大砂鼠(Rhombomys opimus)体内,是中国特有的种类,王捷等60年代分离培养成功。对L.gerbilli的前鞭毛体的亚显微结构的观察表明:1.质膜下微管间距约为50nm,比别的利什曼原虫的宽;2.鞭毛基部极少观察到基板(basal plate);3.线粒体发达,从动基体所在部位伸向虫体各个部位,内有复杂的结构;4.在通常情况下,L.gerbilli的动基体呈棒状,但当细胞膨胀后则无论在光镜或电镜下动基体均呈扁环结构;5.虫体后部有发达的复片层结构。这一结构由一系列的膜卷绕成,其意义不明。  相似文献

Abstract A virulent bacteriophage, øSK311, was isolated from Staphylococcus carnosus , an organism used as a starter culture for the production of dry sausage. Electron microscopic studies revealed that this bacteriophage showed some morphological similarities with the Escherichia coli phages T4 and λ. The host range of øSK311 extends over 9 different staphylococcal species. A phage-resistant mutant of S. carnosus could be isolated. Cells of this mutant exhibited a capsule-like structure. The DNA of øSK311 possesses a G + C content of 31.4 mol% and appears to be highly modified.  相似文献

Bacteria isolated from aviation turbine fuel (AVTUR) grew rapidly using the jet fuel as the sole source of carbon and energy. One organism, identified as a Pseudomonas putida survived in the fuel phase for periods of up to one month. This organism grew on and oxidized a wide range of alkanes, with optimum growth occurring on octane. The mechanism of alkane metabolism in this organism was found to be ω-oxidation followed by β-oxidation.  相似文献
