It was hypothesized that concentric and eccentric isokinetic muscle actions should yield detectable differences in the mechanomyograms, which may reflect properties of the contraction and relaxation phases of the muscles. A paired pattern classification technique was adapted to determine whether wavelet transformed mechanomyograms from the three superficial quadriceps muscles were different during maximal concentric and eccentric isokinetic muscle actions. Mechanomyograms for this study were recorded from eleven healthy men (mean ± SD age = 20.1 ± 1.1 yrs) who performed maximal concentric and eccentric isokinetic muscle actions of the dominant leg extensors at a velocity of 30° s?1. The results indicated that the paired pattern classification accurately classified the MMG intensity patterns in approximately 94% of the cases as being from a concentric or eccentric movement. Thus, it can be concluded that the differences in the intensity patterns recorded from concentric and eccentric muscle actions were significant. These findings indicated that the combined MMG wavelet analysis and pattern classification techniques could potentially be useful in situations where muscle activity during concentric muscle actions must be distinguished from that during eccentric muscle actions.  相似文献

Blood flow of the finger and the forearm were measured in five male subjects by venous occlusion plethysmography using mercury-in-Silastic strain gauges in either a cool-dry (COOL: 25 degrees C, 40% relative humidity), a hot-dry (WARM: 35 degrees C, 40% relative humidity), or a hot-wet (HOT: 35 degrees C, 80% relative humidity) environment. One hand or forearm was immersed in a water bath, the temperature (Tw) of which was raised every 10 min by steps of 2 degrees C until it reached 41 degrees or 43 degrees C. While the other hand or forearm was kept immersed in a water bath (Tw, 35 degrees C), blood flow in the heated side (BFw) was compared with the corresponding blood flow in the control side (BFc). Under WARM or HOT conditions, finger BFw was significantly lower than finger BFc at a Tw of 39-41 degrees C in the majority of subjects. When Tw was raised to 43 degrees C, however, finger BFw became higher than BFc in nearly half of the subjects. In the COOL state, finger BFw did not decrease but increased steadily when Tw increased from 37 degrees to 43 degrees C. In the forearm, BFw increased steadily with increasing Tw even in WARM-HOT environments. No such heat-induced vasoconstriction was observed in the forearm. From these results we conclude that in hyperthermic subjects, the rise in local temperature to above core temperature produces vasoconstriction in the fingers, an area where no thermal sweating takes place.  相似文献

The effect of position and number and of another intracellular location signal on the activity of the nuclear location signal was investigated. A minimal signal was inserted into several sites within the polypeptide chain of pyruvate kinase. The results observed suggest that a nuclear location signal can function at a variety of positions within a protein but that in some locations its activity is masked. Multiple copies of a partially defective signal were integrated into pyruvate kinase. The data suggest that multiple signals can cooperate to enhance nuclear accumulation. A nuclear location signal failed to function when inserted into polyomavirus middle T but was active in an identical variant lacking the carboxy-terminal hydrophobic tail. We conclude that while a minimal nuclear location signal is sufficient for nuclear localization, its activity is crucially dependent on the protein context within which it is present.  相似文献

An intramolecular electron-transfer process has previously been shown to take place between the Cys3--Cys26 radical-ion (RSSR-) produced pulse radiolytically and the Cu(II) ion in the blue single-copper protein, azurin [Farver, O. & Pecht, I. (1989) Proc. Natl Acad. Sci. USA 86, 6868-6972]. To further investigate the nature of this long-range electron transfer (LRET) proceeding within the protein matrix, we have now investigated it in two azurins where amino acids have been substituted by single-site mutation of the wild-type Pseudomonas aeruginosa azurin. In one mutated protein, a methionine residue (Met44) that is proximal to the copper coordination sphere has been replaced by a positively charged lysyl residue ([M44K]azurin), while in the second mutant, another residue neighbouring the Cu-coordination site (His35) has been replaced by a glutamine ([H35Q]azurin). Though both these substitutions are not in the microenvironment separating the electron donor and acceptor, they were expected to affect the LRET rate because of their effect on the redox potential of the copper site and thus on the driving force of the reaction, as well as on the reorganization energies of the copper site. The rate of intramolecular electron transfer from RSSR- to Cu(II) in the wild-type P. aeruginosa azurin (delta G degrees = -68.9 kJ/mol) has previously been determined to be 44 +/- 7 s-1 at 298 K, pH 7.0. The [M44K]azurin mutant (delta G degrees = -75.3 kJ/mol) was now found to react considerably faster (k = 134 +/- 12 s-1 at 298 K, pH 7.0) while the [H35Q]azurin mutant (delta G degrees = -65.4 kJ/mol) exhibits, within experimental error, the same specific rate (k = 52 +/- 11 s-1, 298 K, pH 7.0) as that of the wild-type azurin. From the temperature dependence of these LRET rates the following activation parameters were calculated: delta H++ = 37.9 +/- 1.3 kJ/mol and 47.2 +/- 0.7 kJ/mol and delta S++ = -86.5 +/- 5.8 J/mol.K and -46.4 +/- 4.4 J/mol.K for [H35Q]azurin and [M44K]azurin, respectively. Using the Marcus relation for intramolecular electron transfer and the above parameters we have determined the reorganization energy, lambda and electronic coupling factor, beta. The calculated values fit very well with a through-bond LRET mechanism.  相似文献

Previously reported results showed that the BmKK2's intron could be recognized and spliced in cultured HEK 293T cells. At the same time, a cryptic splicing site of BmKK2 gene was found in the second exon. Moreover, replacing BmKK2's intron with BmP03's intron (an artificial BmKK2-BmP03 mosaic gene) did not affect the intron's recognition and splicing, but increased the expression level of the toxin-GFP fusion protein (Cao et al., J Biochem Mol Toxicol 2006;20:1-6). In this investigation, the BmKK2's intron with 79 nucleotides length was artificially shifted from the 49th nt (the 17th Gly codon between the first base and the second base) to the 100th nt (the 34th Gly codon between the first base and the second base). Based on the constructed intron-splicing system, the results of RT-PCR and the western blotting analysis showed that the BmKK2's shifted-intron (named BmKK2-s) was not recognized and spliced correctly, but the cryptic splicing site of BmKK2 gene was still spliced in the second exon, which possibly indicated that locations of introns were very important to the recognition and splicing of introns, and splicing of introns was very much associated with the corresponding upstream and downstream exons. This result possibly provides evidence for splice-site recognition across the exons.  相似文献

Propulsive forces generated by swimmers hand/forearm, have been studied through experimental tests. However, there are serious doubts as to whether forces quantified in this way are accurate enough to be meaningful. In order to solve some experimental problems, some numerical techniques have been proposed using Computational Fluid Dynamics (CFD). The main purpose of the present work was threefold. First, disseminate the use of CFD as a new tool in swimming research. Second, apply the CFD method in the calculation of drag and lift coefficients resulting from the numerical resolution equations of the flow around the swimmers hand/forearm using the steady flow conditions. Third, evaluate the effect of hand/forearm acceleration on drag and lift coefficients. For these purposes three, two-dimensional (2D), models of a right male hand/forearm were studied. A frontal model (theta = 90 degrees, Phi = 90 degrees) and two lateral models, one with the thumb as leading edge (theta = 0 degrees, = 90 degrees), and the other with the small finger as the leading edge (theta = 0 degrees, Phi = 180 degrees). The governing system of equations considered was the incompressible Reynolds averaged Navier-Stokes equations with the standard k-epsilon model. The main results reported that, under the steady-state flow condition, the drag coefficient was the one that contributes more for propulsion, and was almost constant for the whole range of velocities, with a maximum value of 1.16 (Cd = 1.16). This is valid when the orientation of the hand/forearm is plane and the model is perpendicular to the direction of the flow. Under the hand /forearm acceleration condition, the measured values for propulsive forces calculation were approximately 22.5% (54.440 N) higher than the forces produced under the steady flow condition (44.428 N). By the results, pointed out, we can conclude that: (i) CFD can be considered an interesting new approach for hydrodynamic forces calculation on swimming, (ii) the acceleration of hand/forearm provides more propulsion to swimmers, confirming that some unsteady mechanism must be present in swimming propulsion.  相似文献

The fraction of crosstalk was examined from the surface EMG signals collected from digit- and wrist-dedicated flexors with a blind signal separation (BSS) algorithm. Six participants performed static power grip tasks in a neutral posture at four different exertion levels of 25%, 50%, 75%, and 100% MVC. The signals were collected from the flexor digitorum superficialis, flexor digitorum profundus, flexor carpi radialis, palmaris longus, and flexor carpi ulnaris using a bipolar electrode configuration. The percentage of root mean square (RMS) was used as an amplitude-based index of crosstalk by normalizing the signals including crosstalk to those excluding crosstalk by the BSS algorithm for each %MVC exertion. The peak R2 value of a cross-correlation function was also calculated as a correlation-based index of crosstalk for a group of forearm flexors by force level and algorithm application. The fraction of crosstalk ranged from 32% to 50% in the wrist-dedicated flexors and from 11% to 25% in the digit-dedicated flexors. Since surface EMG signals had such high levels of crosstalk, reduction methods like the BSS algorithm should be employed, as the BSS significantly reduced crosstalk in the forearm flexors 33% over all muscles and exertion levels. Thus, it is recommended that BSS be utilized to reduce crosstalk for the digit- and wrist-dedicated flexors during gripping tasks.  相似文献

The patterns of EMG activity in the biceps and triceps muscles were recorded during horizontal oscillatory movements of the forearm. Subjects showed increased frequency of oscillation as they voluntarily reduced movement amplitude. EMG burst duration was significantly correlated with wavelength of oscillation in every case. In almost half the cases burst intensity was also positively correlated with wavelength. Subjects seemed to be using one or both these methods to control amplitude. A model was developed in three stages which satisfactorily accounted for the data.  相似文献

Background  

Fluorescence microscopy is widely used to determine the subcellular location of proteins. Efforts to determine location on a proteome-wide basis create a need for automated methods to analyze the resulting images. Over the past ten years, the feasibility of using machine learning methods to recognize all major subcellular location patterns has been convincingly demonstrated, using diverse feature sets and classifiers. On a well-studied data set of 2D HeLa single-cell images, the best performance to date, 91.5%, was obtained by including a set of multiresolution features. This demonstrates the value of multiresolution approaches to this important problem.  相似文献
