本文报道了不同孢子浓度下黄绿绿僵菌对褐飞虱和白背飞虱不同发育阶段的易感性和毒力的研究。实验设10.5孢子/mm2,116.7孢子/mm2和1027.1孢子/mm2三种孢子剂量,两种飞虱分为幼龄若虫(1龄和2龄若虫)、高龄若虫(3、4、5龄若虫)和成虫三个发育阶段。实验发现褐飞虱与白背飞虱的三个发育阶段对黄绿绿僵菌的不同浓度的孢子液有不同程度的易感性。黄绿绿僵菌对褐飞虱幼龄若虫的毒力指标LT50在三种孢子剂量下依次为>21、20.82和16.55;对高龄若虫的LT50在三种孢子剂量下依次为17.68、15.49和13.98;而对成虫的LT50在三种孢子剂量下依次为17.10、12.57和9.14。黄绿绿僵菌对白背飞虱幼龄若虫的毒力指标LT50在三种孢子剂量下依次为>21、17.29和13.13;对高龄若虫的LT50在三种孢子剂量下依次为16.94、15.02和13.03;而对白背飞虱成虫的LT50在三种孢子剂量下依次为12.78、10.16和7.64。二者的成虫的易感性比若虫的易感性强,高龄若虫的易感性比幼龄若虫的强。白背飞虱比褐飞虱对黄绿绿僵菌更加敏感。二者的死亡率随孢子浓度的增大而增大。  相似文献

Abstract: The potential of some isolates of Metarhizium anisopliae and Beauveria bassiana for use in the integrated management of Culex quinquefasciatus was evaluated. Metarhizium anisopliae isolate 1037 was selected in initial bioassays with a 50% lethal concentration (LC50) of 1.97 × 104 conidia/ml. This fungus caused higher mosquito larva mortality when applied as a conidial suspension to the surface of the water than as dry conidia, with a time to 50% lethal (LT50) of 1 day compared with 3.6 days for the dry conidial application. However, results with UV- and heat-inactivated conidia did not confirm a possible role of fungal toxins in causing mortality when ingested by C. quinquefasciatus larvae. Metarhizium anisopliae did not remain active for as long as the bacterium Bacillus sphaericus isolate 2362. At 24 h after application, the fungus-induced mortality on mosquito larvae was significantly lower than the mortality caused by the bacterium. By the second day, almost no activity by the fungus was observed. Results suggest that M. anisopliae isolate 1037 has potential for use in mosquito control programmes.  相似文献

The LT50 (lethal temperature) of first instar and adult stages of the peach-potato aphid Myzus persicae was lowered following long term acclimation at low temperatures.
First instars consistently showed greater cold hardiness than adult stages at each acclimation temperature, with the differential increasing as the temperature was lowered. When maintained at 5°C (the lowest acclimation regime) nymphs and adults had dLT508.3°C and 4.7°C respectively lower than those for non-acclimated individuals.
When 10°C acclimated adults were returned to 20°C, the acclimation effect was retained in full for 6 days but complete deacclimation occurred by day 10. In contrast the LT50 of their progeny increased gradually from the first day of adult deacclimation towards the level of the unacclimated control over a period of 10 days.
A change in cold hardiness was observed in first instars according to their position in the birth sequence. The LT50 of first-born nymphs (day 1 of reproduction) from 20°C parents was - 15.9°C rising to - 8.3°C by day 4 and remaining at this level until the end of the reproductive period.
The differential mortality between nymphs and adults observed in the laboratory was supported by the results of a field experiment. Adult aphids kept in clip-cages on a crop of oilseed rape showed greater mortality compared with those introduced as nymphs when the minimum temperature fell below -4°C for the first time in winter. At - 10°C mortality of aphids introduced as adults approached 100% whereas more than 50% of those introduced as nymphs were still alive at this temperature.  相似文献

Abstract.  1. When first instar nymphs and adults of the grain aphid Sitobion avenae (Fabricius) (Hemiptera: Aphidiae) were maintained in long-term cultures (>6 months) at 20 °C and 10 °C, the LT50 decreased from −8 and −8.8 °C to −16.0 and −13.5 °C, respectively.
2. When aphids from the 20 °C culture were transferred to 10 °C, there was a progressive increase in cold tolerance through three successive generations. Transfer of newly moulted pre-reproductive adults reared at 10 °C for three generations back to 20 °C resulted in a rapid loss of cold hardiness in their nymphal offspring.
3. In all generations reared at 10 °C, first born nymphs were more cold hardy than those born later in the birth sequence. The LT50 of nymphs produced on the first day of reproduction in the first, second and third generations maintained at 10 °C were −14.8, −17.0 and −16.6 °C, respectively. Thereafter, nymphal cold hardiness decreased over the subsequent 14 days of reproduction in each generation at 10 °C with mean LT50 values of −10.3, −12.6 and −14.8 °C, respectively. By contrast, the cold tolerance of first born nymphs of aphids reared continuously at 20 °C did not differ in comparison with later born siblings. The LT50 of adult aphids was also unaffected by ageing.

Abstract:  The bioactivities of destruxins (dtx), depsipeptides isolated from Metarhizium anisopliae , against Spodoptera litura were tested in laboratory. For contacting toxicities, dtx-E was more effective than dtx-A and dtx-B. The LC50s values of dtx-A, B and E were 197.98, 292.00 and 113.99 mg/l at 48 h after treatment, while the LT50s were 42.65, 59.45 and 23.68 h at 300 mg/l. In the experiment of antifeedant activity, dtx-A, dtx-B and dtx-E at five concentrations (200, 100, 50, 25 and 12.5 mg/l) were bioassayed. Destruxins in a dose-dependent manner gave an apparent antifeedant activity. Generally, dtx-A, over dtx-B and dtx-E had the significant (P < 0.05) larger choice and no-choice antifeedant indexes (CAIs and NCAIs). At the concentration of 200 mg/l, the CAIs or NCAIs of dtx-A, dtx-B and dtx-E were 96.78, 84.93 and 85.90 or 89.75, 62.42 and 72.28 respectively. Furthermore, the synergistic activity of crude destruxin (CD) for pathogenicity of Paecilomyces javanicus strain Pj01 was detected. The LC50s values of single Pj01 and the mixtures of Pj01 plus CD at 100 or 200 mg/l (Pj01-CD100 or Pj01-CD200) were respectively 474.63 × 105, and 197.45 × 105 or 113.11 × 105 spores/ml at the fifth day after treated. Meanwhile, Pj01, Pj01-CD100 and Pj01-CD200 gave the LT50s values of 6.99 day, 5.49 day and 4.21 day at 100 × 105 spores/ml. Clearly, dtx decreased the values of LC50 and LT50 of the strain Pj01.  相似文献

The interaction between two pathogens, the microsporidian Paranosema locustae Canning and the fungus Metarhizium anisopliae var. acridum Driver and Milner was studied under laboratory conditions in an attempt to develop an improved method of microbial control for the desert locust, Schistocerca gregaria Forsk?l. Fifth-instar locust nymphs, reared in the laboratory, were treated with various concentrations of one of the two pathogens or with both pathogens. The numbers of locusts killed were recorded each day and the production of pathogen spores within the dead locusts was assessed at the end (day 21) of each experiment. Locust nymphs treated with both P. locustae and M. anisopliae died sooner than nymphs infected with only one of the pathogens. At the lower concentrations of pathogen tested, the effects of the two pathogens were additive. At the higher concentrations the combined effects were synergistic. In terms of locust mortality, there was no evidence of any antagonistic effects between the two pathogens. However, the production of spores by P. locustae was reduced considerably when the host insects were infected also with M. anisopliae. For example, nymphs treated initially with P. locustae and then treated 3 and 10 days later with M. anisopliae produced 3-20 times and 2.5-8 times fewer spores, respectively, than nymphs treated only with P. locustae. Hence, in areas where M. anisopliae is applied, the natural persistence of P. locustae in the local grasshopper and locust populations may be diminished.  相似文献

【目的】硅可增强植物对多种植食性昆虫的抗性。本研究旨在了解硅处理对水稻叶鞘硅化程度及其对白背飞虱Sogatella furcifera刺吸和寄主选择行为的影响,以明确施硅增强水稻对白背飞虱的抗性的部分机制。【方法】采用扫描电镜观察施硅水稻的叶鞘硅化程度;利用刺吸电位技术记录白背飞虱若虫刺吸行为;采用笼罩法测定白背飞虱雌成虫栖息和产卵选择性。【结果】与不施硅的对照相比,硅处理(0.16和0.32 g SiO2/kg土壤)增加了水稻叶鞘硅细胞的数量,延长了白背飞虱若虫刺吸行为中非刺探波和路径波的总时间,缩短了取食韧皮部汁液的时间。在选择性测定中,与对照组相比,白背飞虱雌成虫对0.16和0.32 g SiO2/kg土壤处理的水稻的栖息率分别降低48.0%和67.4%,在其上的产卵量分别降低34.8%和46.1%。【结论】施硅增加水稻对白背飞虱的排驱性,阻碍白背飞虱的刺吸行为,因此有助于增强水稻对白背飞虱的抗性。  相似文献

Laboratory and net house assays of the entomopathogenic fungus, Beauveria bassiana CS-1, were conducted to examine the possibility of using this strain to control the diamondback moth (DBM), Plutella xylostella in Korea. Treatment of conidial suspensions at various concentrations indicated that corrected mortality at 1 × 108 conidia/ml was 86.23% with LT50 of 1.63 days under the laboratory conditions and 66.5% with LT50 of 3.61 days in the net house assay. Assessment of rigorous dose–response showed that at least 1800 conidia of the strain CS-1 should adhere to a single DBM larva to induce successful infection. These results indicated that the strain CS-1 has the capability to lower the DBM larval population rapidly as well as effectively.  相似文献

Cultured potato ( Solanum tuberosum L., cv. Red Pontiac) cells suspended in PEG 1000 solutions of 0.6 and O.S osmol exhibited significantly different freezing tolerance from the same cells when suspended in PEG 6000 solutions of the same osmolalities. Cells suspended in PEG 6000 showed cytorhysis instead of plasmolysis. Cells in 0.2 and 0.4 osmol PEG 1000 had LT50(1 of −2.5°C, but the LT50 decreased to −7.50C as the osmolality increased to 0.8 osmol. In PEG 6000 the LT50 remained at −2.50C for all osmolalities used, up to and including 0.8 osmol.
Released protoplasts suspended in 0.5 M sucrose had LT50 of −21.5°C, compared to −12°C for whole cells suspended in the same medium. These results lend credence to an involvement of the cell wall in freezing injury of cultured potato cells, and are interpreted in terms of the generation of a mechanical stress between cell wall and plasma membrane during the freeze-thaw cycle.  相似文献

Expression analysis of the genes involved in germination, conidiogenisis and pathogenesis of Metarhizium anisopliae during its saprophytic and pathogenic life stages can help plan strategies to increase its efficacy as a biological control agent. We quantified relative expression levels of the nitrogen response regulator gene (nrr1) and a G-protein regulator of genes involved in conidiogenesis (cag8), using an RT-qPCR assay. Comparisons were made between M. anisopliae var. anisopliae and M. anisopliae var. acridum during germination and conidiogenesis and at different stages of pathogenesis. The cag8 gene was repressed during germination and induced during conidial development and the pathogenic phase, and the nrr1 gene was induced during germination, conidiogenesis and the pathogenic phase. Both genes were more expressed in M. anisopliae var. anisopliae, demonstrating that different varieties of M. anisopliae differ in activation of genes linked to virulence for certain environments and hosts. This suggests that differences among these varieties in the ability to adapt could be attributed not only to specific genomic regions and genes, but also to differential gene expression in this fungus, modulating its ability to respond to environmental stimuli.  相似文献

A pre-infestation of the white-backed planthopper (WBPH), Sogatella furcifera Horváth, conferred resistance to bacterial blight caused by Xanthomonas oryzae pv. oryzae ( Xoo ) in rice ( Oryza sativa  L.) under both laboratory and field conditions. The infestation of another planthopper species, the brown planthopper (BPH) Nilaparvata lugens Stål, did not significantly reduce the incidence of bacterial blight symptoms. A large-scale screening using a rice DNA microarray and quantitative RT-PCR revealed that WBPH infestation caused the upregulation of more defence-related genes than did BPH infestation. Hydroperoxide lyase 2 ( OsHPL2 ), an enzyme for producing C6 volatiles, was upregulated by WBPH infestation, but not by BPH infestation. One C6 volatile, ( E )-2-hexenal, accumulated in rice after WBPH infestation, but not after BPH infestation. A direct application of ( E )-2-hexenal to a liquid culture of Xoo inhibited the growth of the bacterium. Furthermore, a vapour treatment of rice plants with ( E )-2-hexenal induced resistance to bacterial blight. OsHPL2 -overexpressing transgenic rice plants exhibited increased resistance to bacterial blight. Based on these data, we conclude that OsHPL2 and its derived ( E )-2-hexenal play some role in WBPH-induced resistance in rice.  相似文献

A mixture of lambda-cyhalothrin (lambda-cyhalothrin: chemical insecticide) and Metarhizium anisopliae ( flavoviride ) var. acridum Driver and Milner, an entomopathogenic fungus (bioinsecticide) was used for grasshopper control in Mali. An oil-based formulation of Metarhizium anisopliae ( flavoviride ) var. acridum Driver and Milner has been developed by LUBILOSA a collaborative project for locust and grasshoppers control. It takes 6 to 10 days for the biopesticide to kill the hosts, which is not a problem for larvae in fallows because they will die before reaching the farmers' fields. However, if crops are infested by adults, the farmers can not wait for 6 to 10 days. An experiment was conducted in Mali using a mixture of a biopesticide and chemical pesticide. The mixture of lambda-cyhalothrin (chemical insecticide) and Metarhizium anisopliae ( flavoviride ) var. acridum (biopesticide: oil-based entomopathogenic fungus spore suspension) was applied to nymphs of Sahelian grasshoppers, using ultra low volume (ULV) sprayers. Both the mixture and lambda-cyhalothrin alone gave quick mortality, with slightly higher mortality for the mixture. Mortality due to the Metarhizium treatments began 2 days after application and subsequently reached similar levels of mortality to the lambda-cyhalothrin mixture treatments. The efficacy of the mixture was greater than Metarhizium alone. The efficacy of lambda-cyhalothrin reached 80% on the day following application, but declined after 10 days, due probably to immigration of untreated grasshoppers.  相似文献

AIMS: To examine the ability of Agrobacterium to attach to Metarhizium anisopliae var. acridum strain CG423 under co-cultivation and to develop an Agrobacterium-mediated method of gene delivery into strain CG423, a promising agent for biological control of grasshoppers. METHODS AND RESULTS: The co-cultivation of Agrobacterium tumefaciens and M. anisopliae var. acridum was analysed under scanning electron microscopy. We observed that Agrobacterium attached to and formed aggregates around Metarhizium conidia and germ tubes. We also observed the occurrence of fibril-like structures connecting neighbouring bacterial-fungal cells. The Agrobacterium-mediated transformation was applied using two binary vectors carrying a benomyl resistance gene as a selection marker. The efficiency of transformation was up to 53 transformants per 10(5) target conidia. High mitotic stability of the transformants (89-97%) was demonstrated after five successive transfers on non-selective media. Molecular analysis revealed the occurrence of high frequency of gene conversion. CONCLUSIONS: In our study, we report that A. tumefaciens strain AGL-1 attaches to and genetically transforms the entomopathogenic fungus Metarhizium anisopliae var. acridum. SIGNIFICANCE AND IMPACT OF THE STUDY: We report for the first time, the attachment of Agrobacterium to fungal cells opening new avenues for the study of this essential step of the T-DNA transfer process. Considering the efficiency of the transformation protocol herein described, this is a useful tool for gene disruption in M. anisopliae var. acridum.  相似文献

Strains of the entomopathogens Beauveria bassiana (Balsamo) Vuillemin and Metarhizium anisopliae (Metschnikoff) Sorokin were screened for effectiveness against the Formosan subterranean termite, Coptotermes formosanus (Shiraki). Biological control methods are being considered to augment current integrated pest management schemes that involve chemical baits and nonrepellent termiticides. In this study Formosan subterranean termites selected from four colonies in the field were exposed in the laboratory to three strains of Beauveria bassiana and two strains of Metarhizium anisopliae . The exposed termites were then allowed to mingle with an equal number of unexposed termites from the same nest and mortality of the combined group was determined. One week after exposure, B. bassiana strains, ATCC 26037 and ATCC 90519, had caused significantly higher mortality than the two M. anisopliae strains, ESC 1 and II B and the remaining strain of B. bassiana , ATCC 90518. The LT50 measurement for strains 26037 and 90519 was comparable at 1.8 and 2.0 days, respectively. Strains ESC 1 and 90518 caused 50% mortality at 6.5 and 11.0 days, respectively. Termites exposed to strain II B and the control group of termites did not reach 50% mortality during the 21 day experimental period. Exposure of termites to all fungal strains resulted in mortality in excess of that seen in the control groups of unexposed termites.  相似文献

Time courses of formation of inositol 1,4,5-trisphosphate (IP3) were followed in the leaves of non-acclimated and cold (2°C)-acclimated winter oilseed rape ( Brassica napus L. var. oleifera ) plants, subjected to different freezing temperatures or to polyethylene glycol 8000 (PEG) and abscisic acid (ABA) treatments. Changes in water potential (Ψw) and in ABA level in the frost- and PEG-treated tissues were also determined. Results obtained indicate that temperatures sligthly higher than LT50 induced a transient and substantial increase in IP3 level, both in non-acclimated and cold-acclimated tissues. At comparable freezing temperature (–5°C) the response of cold-acclimated leaves was lower than that of non-acclimated ones. The PEG-depedent decrease in Ψw to –0.9 MPa or ABA (0.1 m M ) treatment gave rise to a transient increase in IP3 content in non-acclimated tissues only. Collectively, the data indicate that cold acclimation of plants may lead to lower cell responsiveness to the factors studied in terms of induction of IP3 formation. Changes in the IP3 content, observed in the present experiments, support our previous suggestion that non-killing freezing temperatures may induce the phosphoinositide pathway, both in non-acclimated and cold-acclimated tissues. Lowering of tissue water potential to some threshold value or a high exogenous ABA supply may mimic the freezing-dependent reaction in the non-acclimated leaves.  相似文献

Abstract:  The two-spotted stink bug, Perillus bioculatus (F) (Hem., Pentatomidae) is considered an important predator of Colorado potato beetle egg masses and small larvae. The susceptibility of P. bioculatus second instar nymphs to six isolates of Beauveria bassiana (Balsamo) Vuillemin, which are highly pathogenic to Colorado potato beetle, was tested in the laboratory. Five isolates were highly pathogenic and caused more than 77% mortality after 8 days at a concentration of 106 conidia/ml. However, isolate IPP46 showed low pathogenicity and caused only 11% mortality of nymphs. The pathogenicity of isolates was independent of their host, source or country of origin. Isolate IPP46 was tested at six different concentrations. A linear relationship between the concentration of B. bassiana and the mortality of nymphs was observed. The 50% lethal concentration (LC50) value was 3.4 × 107 conidia/ml. The LT50 decreased with increase of conidia concentration used. The present results suggest that isolate IPP46 has good potential as a biological control agent within an integrated pest management programme.  相似文献

Miscanthus , a perennial rhizomatous C4 grass, is a potential biomass crop in Europe, mainly because of its high yield potential and low demand for inputs. However, until recently only a single clone, M. × giganteus , was available for the extensive field trials performed across Europe and this showed poor overwintering in the first year after planting at some locations in Northern Europe. Therefore, field trials with five Miscanthus genotypes, including two acquisitions of Miscanthus × giganteus , one of M. sacchariflorus and two hybrids of M. sinensis were planted in early summer 1997 at four sites, in Sweden, Denmark, England and Germany. The field trials showed that better overwintering of newly established plants at a site was not apparently connected with size or early senescence. An artificial freezing test with rhizomes removed from the field in January 1998 showed that the lethal temperature at which 50% were killed (LT50) for M. × giganteus and M. sacchariflorus genotypes was −3.4 °C. However, LT50 in one of the M. sinensis hybrid genotypes tested was −6.5 °C and this genotype had the highest survival rates in the field in Sweden and Denmark. Although the carbohydrate content of rhizomes, osmotic potential of cell sap and mineral composition were not found to explain differences in frost tolerance adequately, moisture contents correlated with frost hardiness (LT50) in most cases. The results obtained form a basis for identifying suitable Miscanthus genotypes for biomass production in the differing climatic regions of Europe.  相似文献

Isogenic strains (with and without dsRNA) of the entomogenous fungi Metarhizium anisopliae var. acridum and Paecilomyces fumosoroseus were investigated for correlation between the presence of dsRNA and the production of cuticle-degrading proteases that play an important role in host parasitism, total secreted protein, and conidia production. Similar levels of cuticle-degrading subtilisin-like (Pr1) protease were observed for isogenic strains of M. anisopliae var. acridum after growth in medium supplemented with the cuticle of the grasshopper Rhammatocerus schistocercoides. Similarly, no statistical differences were observed for protease production, detected using the chromogenic substrate azocasein. For P. fumosoroseus isogenic strains, no significant differences in protease activity were observed after growth in the presence of either Euschistus heros or Nezara viridula (Hemiptera: Pentatomidae) cuticle. Similarly, no statistical differences were observed in virulence against E. heros. A comparison of mean conidia production showed a significantly higher production in the dsRNA-free isogenic strains of M. anisopliae var. acridum. Although, for most of the fungal phenotypes analysed, no overt effects were associated with the presence of these dsRNA infections, the reduction in conidia production by the isogenic strains of M. anisopliae var. acridum with dsRNA suggested that it may not be entirely accurate to describe these infections as latent.  相似文献

【目的】橙带蓝尺蛾Milionia basalis近年来在我国南方地区严重危害罗汉松Podocarpusmacrophyllus和竹柏Podocarpus nagi,扩散蔓延速度快,造成了很大的损失。本研究旨在评估金龟子绿僵菌Metarhizium anisopliae MaFZ-13菌株对橙带蓝尺蛾幼虫的致病力,探寻橙带蓝尺蛾生物防治新资源。【方法】通过绿僵菌MaFZ-13不同浓度(1.0×105~1.0×109孢子/mL)孢子悬浮液接种橙带蓝尺蛾幼虫,运用时间-剂量-死亡率(TDM)模型对生物测定结果进行分析,同时通过林间喷施1.0×108孢子/mL绿僵菌孢子悬浮液进行林间防治效果试验。【结果】橙带蓝尺蛾幼虫累计死亡率随着绿僵菌MaFZ-13孢子悬浮液孢子浓度的增加和接种时间的延长而增大。接种1.0×109孢子/mL孢子悬浮液8 d后以及接种1.0×108孢子/mL孢子悬浮液10 d后,幼虫死亡率均达到100%;接种1.0×107孢子/mL孢子悬浮液15 d后幼虫死亡率为95.6%。应用TDM模型对生测数据进行分析,结果表明所建模型通过Hosmer-Lemeshow拟合异质性检验,并由模型估算出了该菌株对橙带蓝尺蛾幼虫的致死剂量和致死时间。绿僵菌MaFZ-13菌株接种4, 5, 6和7 d后的半致死剂量(LC50)对数估计值分别为7.99, 7.12, 6.46和5.83。以1.0×107, 1.0×108和1.0×109孢子/mL的孢子浓度接种时,绿僵菌MaFZ-13菌株对橙带蓝尺蛾幼虫的LT50值分别为5.19,3.99和2.81 d。林间用1.0×108孢子/mL绿僵菌孢子悬浮液喷洒9 d后橙带蓝尺蛾幼虫死亡率可达到85.8%,防治效果为85.05%。【结论】绿僵菌MaFZ-13对橙带蓝尺蛾幼虫具有较强的致病力,林间防治效果良好,具有较好的应用潜力。  相似文献
