Background and Aims

Seagrasses are marine, flowering plants with a hydrophilous pollination strategy. In these plants, successful mating requires dispersal of filamentous pollen grains through the water column to receptive stigmas. Approximately 40 % of seagrass species are monoecious, and therefore little pollen movement is required if inbreeding is tolerated. Outcrossing in these species is further impacted by clonality, which is variable, but can be extensive in large, dense meadows. Despite this, little is known about the interaction between clonal structure, genetic diversity and mating systems in hydrophilous taxa.

Methods

Polymorphic microsatellite DNA markers were used to characterize genetic diversity, clonal structure, mating system and realized pollen dispersal in two meadows of the temperate, monoecious seagrass, Posidonia australis, in Cockburn Sound, Western Australia.

Key Results

Within the two sampled meadows, genetic diversity was moderate among the maternal shoots (R = 0·45 and 0·64) and extremely high in the embryos (R = 0·93–0·97). Both meadows exhibited a highly clumping (or phalanx) structure among clones, with spatial autocorrelation analysis showing significant genetic structure among shoots and embryos up to 10–15 m. Outcrossing rates were not significantly different from one. Pollen dispersal distances inferred from paternity assignment averaged 30·8 and 26·8 m, which was larger than the mean clone size (12·8 and 13·8 m).

Conclusions

These results suggest highly effective movement of pollen in the water column. Despite strong clonal structure and moderate genetic diversity within meadows, hydrophilous pollination is an effective vector for completely outcrossed offspring. The different localized water conditions at each site (highly exposed conditions vs. weak directional flow) appear to have little influence on the success and pattern of successful pollination in the two meadows.

The high prevalence of dioecy in marine angiosperms or seagrasses (>50% of all species) is thought to enforce cross‐fertilization. However, seagrasses are clonal plants, and they may still be subject to sibling‐mating or bi‐parental inbreeding if the genetic neighborhood is smaller than the size of the genets. We tested this by determining the genetic neighborhoods of the dioecious seagrass Thalassia testudinum at two sites (Back‐Reef and Mid‐Lagoon) in Puerto Morelos Reef Lagoon, Mexico, by measuring dispersal of pollen and seeds in situ, and by fine‐scale spatial autocorrelation analysis with eight polymorphic microsatellite DNA markers. Prevalence of inbreeding was verified by estimating pairwise kinship coefficients; and by analysing the genotypes of seedlings grown from seeds in mesocosms. Average dispersal of pollen was 0.3–1.6 m (max. 4.8 m) and of seeds was 0.3–0.4 m (max. 1.8 m), resulting in a neighborhood area of 7.4 m2 (range 3.4–11.4 m2) at Back‐Reef and 1.9 (range 1.87–1.92 m2) at Mid‐Lagoon. Neighborhood area (Na) derived from spatial autocorrelation was 0.1–20.5 m2 at Back‐Reef and 0.1–16.9 m2 at Mid‐Lagoon. Maximal extensions of the genets, in 19 × 30 m plots, were 19.2 m (median 7.5 m) and 10.8 m (median 4.8 m) at Back‐Reef and Mid‐Lagoon. There was no indication of deficit or excess of heterozygotes nor were coefficients of inbreeding (FIS) significant. The seedlings did not show statistically significant deficit of heterozygotes (except for 1 locus at Back‐Reef). Contrary to our expectations, we did not find evidence of bi‐parental inbreeding in this dioecious seagrass with large genets but small genetic neighborhoods. Proposed mechanisms to avoid bi‐parental inbreeding are possible selection against homozygotes during fecundation or ovule development. Additionally, the genets grew highly dispersed (aggregation index Ac was 0.09 and 0.10 for Back‐Reef and Mid‐Lagoon, respectively); such highly dispersed guerrilla‐like clonal growth form likely increases the probability of crossing between different potentially unrelated genets.

Seagrass meadows are in decline globally. Although numerous experimental methods have been implemented to restore meadows, few have been successful in the long term. Poor decisions on the sourcing of transplants from donor sites, including poor genetic integration and/or low genetic diversity, may impact on restoration success. However, despite evidence to suggest a positive association between genetic diversity and ecological resilience, there is usually little or no input from genetic data to inform on the genetic management of ecological restoration. Cockburn Sound has seen a 77% decline in seagrass cover since 1967. A transplant trial was conducted between 2004 and 2008 with sprigs of Posidonia australis being planted into a bare sand area. Survival was monitored annually, and in 2012, we compared genetic diversity in this transplant area with the original donor site. Genetic diversity in the restored meadow was very high and comparable to the donor site, with no genetic differentiation detected. The high level of genetic diversity and choice of site may have played an important role in the success of this restoration trial. The observed natural recruits around the site after establishment of transplants suggest that local restoration efforts may improve seafloor habitat and facilitate natural expansion of the meadow.

Acrocomia aculeata is a perennial, fruit-producing palm tree, native to tropical forests. Its fruits have spurred interest because of their significant potential for use in the cosmetic industry and as feedstock for biofuel. In the present study, the genetic structure and mating system in Acrocomia aculeata were analyzed, using eight nuclear micro-satellite loci and samples from São Paulo and Minas Gerais states, Brazil. By means of Bayesian analysis, these populations were clustered into two or three groups. A high multilocus outcrossing rate suggests that outcrosses were predominant, although a certain degree of biparental inbreeding also occurred. Thus, although monoecious and self-compatible, there is every indication that A. aculeata bears a mixed reproductive system, with a predominance of outcrossing. Given the genetic structure revealed hereby, future conservation strategies and germplasm collecting should be focussed on sampling and preserving individuals from different clusters.

Posidonia oceanica Delile is an endemic of the Mediterranean Sea. P. oceanica is one of the few marine higher plants of the Mediterranean and plays a significant role in coastal ecosystems of the Mediterranean Sea. In the past, a number of phytochemical investigations with different focus were carried out. The results of these investigations are summarized and appraised here. A total of 51 natural products was reported from P. oceanica, including phenols, phenylmethane derivatives, phenylethane derivatives, phenylpropane derivatives and their esters, chalkones, flavonols, 5α-cholestanes, and cholest-5-enes. Many of the compounds reported for P. oceanica were, however, not detected by appropriate phytochemical methods and some most probably represent artifacts and are no genuine natural products.

Differences among populations in the intensity of sexual selection resulting from distinct genetic mating systems can lead to divergent morphological evolution and speciation. However, little is known about how genetic mating systems vary between populations and what factors may contribute to this variation. In this study, we compare the genetic mating systems of two geographically distinct populations of the dusky pipefish (Syngnathus floridae), a species characterized by polygynandry and male pregnancy, from the Atlantic Coast of Virginia and the Gulf Coast of Florida. Our results revealed significant interpopulation variation in mating and reproductive success. Estimates of the opportunity for selection (I), the opportunity for sexual selection (I(s)) and the Bateman gradient (beta(ss)) were higher among males in the Florida population than in the Virginia population, suggesting that sexual selection on males is stronger in the Florida population. The Virginia population is larger and denser than the Florida population, suggesting that population demographics may be one of many causal factors shaping interpopulational mating patterns. This study also provides evidence that the adult sex ratio, operational sex ratio, population density and genetic mating system of S. floridae may be temporally stable over timescales of a month in the Florida population. Overall, our results show that this species is a good model for the study of mating system variation in nature and that Bateman's principles may be a useful technique for the quantitative comparison of mating systems between populations.

Comparative studies of related plant species indicate that evolutionary shifts in mating systems are accompanied by changes in reproductive attributes such as flower size, floral morphology, and pollen/ovule ratio. Recent theoretical work suggests that patterns of investment in reproduction should also change with the mating system. In a glasshouse study, we investigated the extent to which mating system differences among populations of Eichhornia paniculata (Pontederiaceae) were correlated with changes in allocation to male and female function, floral display, and the regulation of investment in reproduction through fruit and ovule abortion. Significant differences in the amount of biomass allocated to reproductive structures were evident among six populations of E. paniculata. As predicted by sex allocation theory, the proportion of dry weight allocated to male function decreased with the outcrossing rate of populations. Six of the eight attributes used to characterize floral display also differed significantly among populations. However, with the exception of two attributes describing the number of flowers produced by inflorescences, these were not correlated with outcrossing rate. Levels of fruit and ovule abortion were determined in two populations with contrasting mating systems under different nutrient and pollination treatments. Virtually all fruits initiated by plants from a self-fertilizing population were matured, while the amount of fruit abortion in an outcrossing population increased with flower production. Ovule abortion was low in both populations. Our results demonstrate that the evolution of self-fertilization in E. paniculata is associated with changes in investment to reproduction that normally distinguish selfing and outcrossing species.

? Premise of the study: Microsatellite primers were developed for the seagrass Enhalus acoroides to investigate genetic variation and identify clonal structure. ? Methods and Results: Four polymorphic loci and 32 monomorphic loci were developed in E. acoroides. Two to four alleles per locus were observed at the polymorphic loci across 60 individuals of two E. acoroides populations. The observed and expected heterozygosities within populations ranged from 0.100 to 0.5667 and from 0.0977 to 0.5079, respectively. ? Conclusions: Our study revealed very low polymorphism in E. acoroides, even at the polymorphic loci. Nevertheless, these primers are a useful tool to study genetic variation, clonal structure, and mating system.

Acacia sciophanes is an extremely rare and Critically Endangered species known from two small populations separated by less than 7 km. Specifically we aimed to investigate whether rarity in A. sciophanes is associated with decreased levels of genetic variation and increased levels of selfing by comparing patterns of genetic variation and mating system parameters with its widespread and common sister species A. anfractuosa. Fourteen polymorphic allozyme loci were used to assess genetic diversity with four of these used in the estimation of mating system parameters. At the species level A. sciophanes has lower allelic richness, polymorphism, observed heterozygosity and gene diversity than A. anfractuosa and significantly lower levels of gene diversity at the population level. Both species have a mixed mating system but the largest population of A. sciophanes has lower levels of outcrossing, higher correlated paternity and increased bi-parental inbreeding compared with two A. anfractuosa populations. However, both correlated paternity and bi-parental inbreeding appear to be at least partly influenced by population size regardless of the species. We suggest that A. sciophanes is likely to be an intrinsically rare species and that in particular the lower levels of genetic diversity and increased selfing are a feature of a species that has the ability to persist in a few localised small populations. Despite recent extensive habitat destruction our comparative study provided no clear evidence that such events have contributed to the lower genetic diversity and increased selfing in A. sciophanes and we believe its ability to exist in small populations may not only be an important factor in its survival as a rare species but also indicates that it may be less susceptible to the impacts of habitat loss and fragmentation. The key to this species conservation will be the maintenance of suitable habitat, particularly through improved fire regimes and control of invasive weeds, that will allow the two small populations to continue to persist in extremely restricted areas of remnant vegetation.

Posidonia oceanica is an endemic seagrass species in the Mediterranean Sea. In order to assess levels of genetic structure in this species, the microsatellite polymorphism was analysed from meadows collected in several localities, along the coasts of the Tyrrhenian Sea (Mediterranean Sea). The existence of single population units and the recruitment of seedlings collected in some localities were investigated. Moreover, genetic structure at different spatial scales and biogeographic relationships among populations were also assessed. Our analysis showed the existence of clear patterns of genetic structure in P. oceanica in the area considered in the analysis. P. oceanica, in fact, is present in separate meadows that represent discrete populations, characterized by low genetic diversity. Comparable levels of genetic variability between mature meadows and seedlings were found. Patterns of genetic relatedness among populations seem to be in accord with direction of dominant current flux in the whole area, separating South Tyrrhenian from North Tyrrhenian populations. Moderate levels of gene flow between populations and genetic substructure within populations, together with the finding of the limited role of sexual reproduction in increasing genetic variability, should be a cause for concern for the persistence of this essential resource in the Mediterranean basin.

We conducted DNA fingerprinting analyses to ascertain the mating system and population genetic structure of the palila, an endangered Hawaiian honeycreeper, which occupies a fragmented range on the Mauna Kea volcano of the island of Hawai'i. DNA fingerprinting of twelve complete families from the Pu'u La'au population revealed no evidence of extrapair fertilization or intraspecific brood parasitism. Band-sharing coefficients from fingerprints produced with two probes revealed that the large Pu'u La'au population on the southwest slope of Mauna Kea, and a smaller, geographically separate population on the east slope (at Kanakaleonui) had relatively high and virtually identical levels of minisatellite variability (mean S of 0.27 for each population based on combined data of M13 and Jeffreys 33.15 probes). The two populations also had nearly identical allele frequencies based on their mean corrected similarity, Sij, of 0.98. These data suggest that the two populations have not been fragmented long and/or have sufficient current gene flow to ameliorate any affects of genetic drift. We conclude that present levels of inbreeding are low within both populations, and that proposed translocations of individuals from Pu'u La'au to Kanakaleonui appear appropriate from a genetic standpoint.

Posidonia oceanica, a seagrass endemic to the Mediterranean forms extended and extremely persistent meadows. It is a clonal plant with an apparently irregular pattern of flowering events. An extensive bibliographic review allowed the reconstruction of past flowering events of this species around the Mediterranean, with a high degree of confidence for the last 30 years. The data series on annual flowering prevalence (FP, flowering records per total records) and flowering intensity (FI, fraction of flowering shoots) produced have been compared with four series on Sea Surface annual Temperature maxima (SSTmax) obtained for the NW Mediterranean (averaged from the local data series of l'Estartit and Villefranche: 1957–2005) and for the Eastern, Western basin and the whole Mediterranean sea (extracted from NCEP Reynolds interpolated SST maps: 1982–2005). Significant warming trends are detected in the Mediterranean SSTmax series, at a rate of (mean+SE) 0.04±0.01°C yr−1 (R2=0.24, P<0.01, N=24 years), in the Eastern basin series (0.06±0.01°C yr−1, R2=0.43, P<0.001, N=24 years) and in the long SSTmax series of the NW Mediterranean (0.02±0.01 C yr−1, R2=0.12, P<0.02, N=49 years). The magnitudes of the SSTmax anomalies around the absolute warming trend do not increase with time in any SSTmax series. Peaks of FP and FI in the Mediterranean seem to occur each 9–11 years, and coincide with peaks of annual SSTmax. Annual FP and FI increase with the residuals of annual SSTmax warming trend in all Mediterranean basins (FPMED: R2=0.27, P<0.01, N=23; FPNW: R2=0.34, P<0.01, N=31; FPE: R2=0.20; P<0.10, N=23). An outstanding event of P. oceanica flowering across the Mediterranean has been registered in Autumn 2003; 1 month after the highest annual SSTmax recorded in the series. The hypothesis of flowering induction by thermal stress as the possible cause of this relationship is discussed, as well as the potential use of P. oceanica flowering record as early indicator of biological change induced by global sea warming in Mediterranean marine ecosystems.

Summary Assortative mating by size is a common mating pattern that can be generated by several different behavioural mechanisms, with different evolutionary implications. Assortative mating is typically associated with sexual selection and has been regarded as an attribute of populations, species, mating systems or even higher order taxa. In most animal groups, however, appropriate analyses of assortative mating at these different levels are lacking and the causes and forms of assortative mating are poorly understood. Here, we analyse 45 different population level estimates of assortative mating and non-random mating by size in seven confamiliar species of water striders that share a common mating system. A hierarchical comparative analysis shows that virtually all the variance within the clade occurs among samples within species. We then employ meta-analysis to estimate the overall strength of assortative mating, to determine the form of assortative mating and to further assess potential differences among species as well as the probable causes of assortative mating in this group of insects. We found overall weak but highly significant positive assortative mating. We show that analyses of the degree of heteroscedasticity in plots of male versus female size are critical, since the evolutionary implications of true and apparent assortative mating differ widely and conclude that the positive assortative mating observed in water striders was of the true rather than the apparent form. Further, within samples, mating individuals were significantly larger than non-mating individuals in both males and females. All of these non-random mating patterns were consistent among species and we conclude that weak positive assortative mating by size is a general characteristic of those water strider species that share this mating system. We use our results to illustrate the importance of distinguishing between different forms of assortative mating, to discriminate between various behavioural causes of assortative mating and to assess potential sources of interpopulational variance in estimates of assortative mating. Finally, we discuss the value of using meta-analytic techniques for detecting overall patterns in multiple studies of non-random mating.

Androdioecy was first described by Darwin in his seminal work on barnacle diversity; he identified males and hermaphrodites in the same reproductive population. Today, we realize that many androdioecious plants and animals share astonishing similarities, particularly with regard to their evolutionary history and mating system. Notably, these species were ancestrally dioecious, and their mating system has the following characteristics: hermaphrodites self‐fertilize frequently, males are more successful in large mating groups, and males have a mating advantage. A male mating advantage makes androdioecy more likely to persist over evolutionary times. Androdioecious barnacles, however, appear to persist as an outlier with a different evolutionary trajectory: they originate from hermaphroditic species. Although sexual systems of androdioecious barnacles are known, no information on the mating system of androdioecious barnacles is available. This study assessed the mating system of the androdioecious barnacle Chelonibia testudinaria. In contrast to other androdioecious species, C. testudinaria does not self‐fertilize, males do not have a mating advantage over hermaphrodites, and the average mating group is quite small, averaging only three individuals. Mating success is increased by proximity to the mate and penis length. Taken together, the mating system of C. testudinaria is unusual in comparison with other androdioecious plants and animals, and the lack of a male mating advantage suggests that the mating system alone does not provide an explanation for the maintenance of androdioecy in this species. Instead, we propose that sex‐specific life history equalizes male and hermaphroditic overall fitness.

Male and female brown planthoppers, Nilaparvata lugens(Stål) (Homoptera: Delphacidae), exchange substrate-transmitted signals prior to mating. The pulse repetition frequency of the male song is known to be involved in mate recognition and also to vary among geographical populations. Here the variability of male signals, female signals, and female preferences has been examined within a population. Female preference variation has been partitioned into variation in mean preference and variation in the window of preference of individuals. The genetic component of variation has been examined using isofemale lines. Male signal variation was limited (CV=8%) and was mainly within individuals. Female signal variation was greater (CV=15%). Female mean preference varied little (CV=10%) and was closely matched to the male signal mean, but the preference window was wide (> 4 male signal standard deviations on average) and variable (CV=56%). There was evidence for genetic variation only for preference window. These results are discussed in relation to theories of signal system evolution.

An understanding of the process of submarine pollination should provide insight into the evolutionary and reproductive ecology of the marine angiosperms (seagrasses). The flow around the reproductive organs of the seagrass Zostera marina L. (Potamogetonales) was, therefore, examined in a flow chamber. The phenological emergence of flowers during (1) pollen capture and (2) pollen release, and by fruit during (3) seed release, led to a reduction in flow rate toward the inflorescence. This change in flow due to floral emergence was associated with a 50% increase in the fluid shear stress [tau = (2.2 _ 0.3) x 10-3 Pa for an immature flower vs. tau = (3.1 _ 0.5) x 10-3 Pa for a receptive flower]. The Reynolds number (Re) and fluid shear stress around inflorescences and infructescences were comparable, indicating a dynamic similarity in the processes of pollen capture and fruit dehiscence [Re = 47 _ 5, tau = (1.6 _ 0.3) x 10-3 Pa for inflorescences; Re = 38 _ 5, tau = (1.3 _ 0.1) x 10-3 Pa for infructescences]. These results indicate that the emergence of reproductive organs leads to changes in fluid shear stress, which will affect the release, transport, and capture of particles including pollen. Theoretical considerations of these observations using aerosol-filtration theory suggest that pollen capture in Z. marina occurs through direct interception of pollen by stigmas.

The Haute Island mouflon (Ovis aries) population is isolated on one small (6.5 km2) island of the remote Kerguelen archipelago. Given a promiscuous mating system, a cyclic demography and a strong female-biased sex ratio after population crashes, we expected a low effective population size (Ne). We estimated Ne using demographic and temporal genetic approaches based on genetic information at 25 microsatellite loci from 62 and 58 mouflons sampled in 1988 and 2003, respectively. Genetic Ne estimates were higher than expected, varying between 104 and 250 depending on the methods used. Both demographic and genetic approaches show the Haute Island Ne is buffered against population crashes. The unexpectedly high Ne likely results from the cyclic winter crashes that allow young males to reproduce, limiting the variance of male reproductive success. Based on individual-based simulations, we suggest that despite a strongly female-biased sex ratio, the effects of the mating system on the effective population size more closely resemble random mating or weak polygyny.
