Filariae are tissue-invasive nematodes that cause diseases such as elephantiasis and river blindness. The goal of this study was to characterize the role of histamine during Litomosoides sigmodontis infection of BALB/c mice, a murine model of filariasis. Time course studies demonstrated that while expression of histidine decarboxylase mRNA increases throughout 12 weeks of infection, serum levels of histamine exhibit two peaks—one 30 minutes after primary infection and one 8 weeks later. Interestingly, mice treated with fexofenadine, a histamine receptor 1 inhibitor, demonstrated significantly reduced worm burden in infected mice compared to untreated infected controls. Although fexofenadine-treated mice had decreased antigen-specific IgE levels as well as lower splenocyte IL-5 and IFNγ production, they exhibited a greater than fourfold rise in eosinophil numbers at the tissue site where adult L. sigmodontis worms reside. Fexofenadine-mediated clearance of L. sigmodontis worms was dependent on host eosinophils, as fexofenadine did not decrease worm burdens in eosinophil-deficient dblGATA mice. These findings suggest that histamine release induced by tissue invasive helminths may aid parasite survival by diminishing eosinophilic responses. Further, these results raise the possibility that combining H1 receptor inhibitors with current anthelmintics may improve treatment efficacy for filariae and other tissue-invasive helminths.  相似文献

The present study was carried out in order to obtain information on the mechanism of action of bithionol on Paragonimus westermani (Kerbert 1878). Bithionol stimulated lactic acid production of intact adult worms above the level of the control worms, while it inhibited oxygen consumption of intact adult worms in vitro. Bithionol treatment of adult worms in vivo decreased glycolytic and oxidative metabolism of homogenates of uterine eggs and adult worms.Bithionol inhibited lactic acid formation except when fructose 1, 6-diphosphate (FDP) was used as a substrate in the homogenates of adult worms, and it also inhibited oxygen consumption of homogenates of eggs and adult worms. Bithionol inhibited reduction of methylene blue when succinate was used as a substrate. Bithionol inhibited oxidation of reduced cytochrome c in the 1000g supernatant of homogenates of adult worms. Bithionol inhibited activity of the succinate oxidation in homogenates of adult worms.  相似文献

Balb/c mice were infected by transplanting 3, 5, 10, or 20 female adult Dipetalonema viteae under the dorsal skin. The microfilaremias resulting from infections with 3 or 5 adult worms were of lesser magnitude and of shorter duration than those produced in infections with 10 or 20 worms. Spleen cells taken from these mice at various intervals after infection were assayed in vitro for their ability to respond to phytohemagglutinin (PHA) or lipopolysaccharide (LPS). There was no depression in the response to LPS or to PHA in mice given infections of 3 or 5 D. viteae adult worms. In contrast, the response to PHA was significantly depressed in groups receiving 10 or 20 adult female worms 12 days after infection and by Day 25, the depression was severe. Thereafter the PHA responsiveness recovered gradually to reach control values on Day 60. In mice transplanted with 10 or 20 D. viteae adult worms there was no significant depression in the response to LPS at any time during the infection, but the response was increased slightly sporadically during infection. These results indicate that in mice, this infection causes an initial suppression in the function of PHA-sensitive T cells but has little effect on the B cells which respond to LPS. A factor present in serum taken on Day 25 from mice infected with 10 or 20 adult worms inhibited the proliferative response to PHA by spleen cells from normal mice. The recovery of PHA responsiveness in mice given the heavier infections coincided with death of the adult worms, but mitogen reactivity and microfilaremia were unrelated. Antigens from male or female worms induced cell division in spleen cells taken from infected mice after microfilaremia had ceased whether they were implanted with 3 or 10 adult worms.  相似文献

The present study was carried out in order to obtain information on the action of bithionol and menichlopholan on Clonorchis sinensis, Metagonimus takahashii and Paragonimus miyazakii. The drugs inhibited the motility of the newly excysted juveniles and adult worms in vitro. Moreover, the in vitro treatment slightly decreased lactate dehydrogenase activity in homogenates of adult worms. The drug treatments of adult worms in vitro also inhibited the activity of fumarate reductase, malic enzyme, phosphogluconate dehydrogenase and succinate dehydrogenase in the homogenate. In addition to this, these drugs caused morphological alterations in the ultrastructure of the tegument of the flukes.  相似文献

The bioactivity of an ethyl acetate extract of ginger (Zingiber officinale) towards Schistosoma mansoni adult pairs, both cultured in vitro and in vivo in laboratory mice, was investigated by monitoring worm mortality and fecundity. In vitro, a concentration of 200 mg l(-1) of extract killed almost all worms within 24 h. Male worms seemed more susceptible than female under these conditions. Cumulative egg output of surviving worm pairs in vitro was considerably reduced when exposed to the extract. For example, after 4 days of exposure to 50 mg l(-1), cumulative egg output was only 0.38 eggs per worm pair compared with 36.35 for untreated worms. In vivo efficacy of the extract was tested by oral and subcutaneous delivery of 150 mg kg(-1) followed by assessment of worm survival and fecundity. Neither delivery route produced any significant reduction in worm numbers compared with untreated controls. Worm fecundity was assessed in vivo by cumulative egg counts per liver at 55 days post infection with mice treated subcutaneously. Such infections showed egg levels in the liver of about 2000 eggs per worm pair in 55 days, in both treated and control mice, with no significant difference between the two groups. To ensure that density-dependent effects did not confound this analysis, a separate experiment demonstrated no such influence on egg output per worm pair, at intensities between 1 and 23 worms per mouse.  相似文献

This study was conducted to investigate the life history, morphology, and maturation of larval stages and adult worms of Fasciola gigantica in experimental mice. Lymnaea auricularia rubiginosa was used as the intermediate host, and Oryza sativa was used for encystment of the metacercariae, while Mus musculus was used as the definitive host for maturation study. Fresh eggs from the gall bladder of water buffaloes fully developed into embryonated ones and hatched out at days 11-12 after incubation at about 29ºC. Free-swimming miracidia rapidly penetrated into the snail host, and gradually developed into the next larval stages; sporocyst, redia, and daughter redia with cercariae. Fully-developed cercariae were separated from the redia and shed from the snails on day 39 post-infection (PI). Free-swimming cercariae were immediately allowed to adhere to rice plants, and capsules were constructed to protect metacercariae on rice plants. Juvenile worms were detected in intestines of mice at days 3 and 6 PI, but they were found in the bile duct from day 9 PI. Juvenile and adult flukes were recovered from 16 mice experimentally infected with metacercariae, with the average recovery rate of 35.8%. Sexually mature adult flukes were recovered from day 42 PI. It could be confirmed that experimentally encysted metacercariae could infect and develop to maturity in the experimental host. The present study reports for the first time the complete life history of F. gigantica by an experimental study in Thailand. The obtained information can be used as a guide for prevention, elimination, and treatment of F. gigantica at environment and in other hosts.  相似文献

Experiments were conducted to examine adult worm burdens, fecal egg output, and in vitro fecundity of Nematospiroides dubius in resistant LAF1 and susceptible CBA mice 12, 15, 18, and 21 days following primary and challenge infections. A strong correlation was obtained on the number of eggs produced by worms cultured in vitro and the egg production as assessed by fecal egg count. Worm counts, fecal egg counts, and in vitro fecundity were similar on all days studied following a primary infection in both mouse strains. However, after challenge infection, LAF1 mice showed lower worm burdens, fecal egg output, and in vitro egg production when compared to CBA mice. Although the egg production of surviving female worms from immune LAF1 mice was decreased, it never fell below a threshold of 100 eggs/day. The reduced fecundity may be a manifestation of a general anti-worm response rather than responses directed specifically at worm reproduction.  相似文献

Immature Schistosoma mansoni in mice are less susceptible to antimony therapy than adult worms. KSb tartrate inhibited phosphofructokinase (PFK) (EC 2.7.1.11) to a greater extent in extracts of 3-week-old worms than adults, and inhibited production of lactate in both immature and adult worms in vitro. In vivo, KSb tartrate was accumulated similarly by 3-week-old worms and by adults: measurements of hexosephosphate following drug treatment suggested similar inhibition of PFK in the two worm stages. If antimony acts by inhibition of PFK it is not clear why the young worms are more resistant to chemotherapy than adults.  相似文献

Similar proportions of the inoculum of Nematospiroides dubius larvae reached sexual maturity by 14 days after administration of 50–400 larvae but more adult worms had been expelled by day 63 after infection from those mice infected with 50 vs 400 larvae. There was a significant correlation between time and worm expulsion for all inoculum size groups except for mice given 400 larvae.In mice reinfected with 100 larvae, after termination of primary infections derived from 10 through 400 larvae, more worms from the challenging dose were recovered from mice given greater compared with those given smaller numbers of larvae at primary infection. The N. dubius population size after challenge infection was correlated positively both with number of larvae administered as the primary infection and with the resultant population size during that infection. The serum anti-N. dubius antibody titres after reinfection were higher in mice given 400 compared with those given fewer larvae at primary infection, and the fecundity and female to male sex ratio of the N. dubius populations decreased in proportion to these antibody titres.Protective immunity against challenge N. dubius infection, in mice which had been drenched free of adult worms established from 400 larvae for 5 down to 1 weeks before reinfection, increased from 45% (1 week) to 80% (5 weeks). There was a negative correlation between the population size of N. dubius during challenge infection and the duration between anthelmintic treatment and challenge infection.  相似文献

In mice, repeatedly infected orally with larvae of Nematospiroides dubius, resistance caused delay in the maturation of larvae, their investment in inflammatory nodules and, in sufficiently resistant animals, their death. The fecundity of adult worms was not affected by host resistance.Previously uninfected mice which had received adult worms by transplantation at operation produced only very low titres of reaginic antibody in comparison with mice infected with larvae by mouth.The migration of leucocytes from resistant mice was inhibited by a crude antigen derived from adult worms.In mice made passively immune by transfer of serum the entry of larvae into the wall of the intestine was delayed; there was no inflammatory response and the larvae did not die.In mice selectively depleted of thymus-derived lymphocytes no inflammatory response occurred and the maturation of larvae was not delayed in response to repeated infection.  相似文献

Mechanisms of parasite killing by eosinophils are widely studied and are often implicated in mediating resistance to parasitic infection, especially in conjunction with specific antibodies. Evidence for the eosinophil as an anti-parasite killer cell in vivo is limited and may not justify the belief that eosinophils engage and/or kill infective helminths. We reexamined this question in a mouse model of trichinosis in which antisera to eosinophils were previously used to show the requirement for eosinophils in resistance to this nematode. The current studies used mAb to IL-5 to suppress eosinophil levels in CF1 mice infected with Trichinella spiralis. In mice given a primary infection and injected with an isotype control mAb or left untreated, the medullary and peripheral blood eosinophil numbers peaked at 3 wk postinfection (PI) and returned to baseline levels by 4 wk PI. Peripheral blood eosinophil numbers in infected mice injected with anti-IL-5 were maintained at levels below those of uninfected normal mice through 4 wk of infection. Histologically, there was a prominent eosinophil accumulation in infected, untreated, or control-mAb-treated mice associated with nurse cell complexes containing infective juveniles in skeletal muscle at 3 and 4 wk PI. This was largely eliminated in mice treated with anti-IL-5 mAb. However, the number of muscle stage juvenile worms recovered 3 and 4 wk PI after acid pepsin digestion was unaffected by eosinophil depletion. Challenge infections, in which mice were infected at day 0 with 125 muscle stage worms and challenged at day 28 PI with 350 muscle stage worms, developed peak eosinophil numbers in bone marrow and peripheral blood 3 wk after primary infection and 2 wk after challenge infection in mice receiving either no treatment or control mAb. In challenged mice receiving anti-IL-5 mAb, medullary and peripheral blood eosinophil numbers remained at or below those of uninfected animals. Although all groups exhibited significant resistance measured as muscle stage worm burdens 56 days PI, eosinophil depletion did not affect resistance of muscle worm recovery. These results suggest that eosinophils are not essential in the control of T. spiralis in either primary or challenge infections of CF1 mice. This in vivo study illustrates the questionable value of in vitro killing assays to assign effector function to any single inflammatory cell type.  相似文献

International Journal for Parasitology14:121–126. Some possible mechanisms of thiabenzadole (TBZ) resistance have been investigated in Trichostrongylus colubriformis. Uptake of TBZ and 3-0-methyl glucose and changes in carbohydrate levels in a TBZ-resistant and a TBZ-susceptible strain of T. colubriformis were measured after in vitro incubation, in the absence or presence of TBZ. The rate of TBZ uptake in adult worms was greater in the resistant strain but the difference was not statistically significant. Final concentrations of TBZ in resistant worms were significantly greater than in the susceptible strain, but there was no difference in TBZ uptake by eggs of either strain. A mechanism of resistance based on decreased drug uptake by resistant parasites is thus unlikely. Resistant worms absorbed 3-0-methyl glucose faster than susceptible parasites, however, this rate was unchanged in either strain when the worms were exposed to TBZ and indicates that the drug does not affect glucose uptake. Levels of glucose, trehalose and glycogen fell during the incubation with the decrease being more pronounced in the resistant strain. When the worms were exposed to TBZ the same changes in carbohydrate levels occurred except for resistant worms maintained anaerobically where levels were higher, showing that TBZ does not affect the rate of carbohydrate utilization.  相似文献

In vitro activity of levamisole on the infective larvae, microfilariae and adult worms of Breinlia sergenti. International Journal for Parasitology4: 207–210. Levamisole shows in vitro activity against the infective larvae, microfilariae and adult worms of Breinlia sergenti. The polygraph studies using the adult worms indicate that levamisole causes an increase in the muscle tone; this action being dose related. The adult worms are more sensitive to the drug than the infective larvae and microfilariae. In vitro, levamisole is more potent compared with diethylcarbamazine against all the three stages of B. sergenti.  相似文献

International Journal for Parasitology12: 413–421. Incubation of Nematospiroides dubius larvae in serum in vitro induced 15% exsheathment after 3h. Larvae incubated in immune mouse serum at 37°C for 3h lost 20% of their infectivity for mice. Immune serum from donors given 1–7 concurrent or anthelmintic abbreviated infections all depressed larval infectivity to the same extent. Larvae incubated in immune sera were protected from the effects of passively transferred immune serum in mice following infection. The effects of incubation of larvae in immune serum were prolonged into the adult stages of the parasite and were seen as stunting of worms and a reduction in the male-female sex ratio of the parasites.  相似文献

The effects of concurrent primary infection of the rat with Eimeria nieschulzi and Trichinella spiralis on the number of oocysts of E. nieschulzi shed by the host and on the number, distribution, and fecundity of adult T. spiralis were analyzed. When rats were initially infected with E. nieschulzi followed 9 days later by infection with T. spiralis there occurred a significant decrease in the total numbers of adult worms in the small intestine, a significant shift in the position of these worms along the length of the small gut, a decrease in the fecundity of adult female worms, and a decrease in muscle parasitism when compared with rats infected with T. spiralis alone. When rats were initially infected with T. spiralis, followed 9 days later by infection with E. nieschulzi, there occurred a significant decrease in the numbers of oocysts shed over 24 hr on Days 7, 9, and 11 postinfection below that seen with rats infected only with Eimeria. These changes are discussed in terms of the enteropathophysiologic lesions and enteric inflammation known to occur during infections with these two parasites.  相似文献

The infectivity of Trichinella pseudospiralis infective larvae was reduced significantly following exposure to low pH or a combination of 1% pepsin at low pH compared to that for larvae isolated in phosphate-buffered saline (PBS) at pH 7.0. Reduction of host gastric pH by administration to mice of sodium bicarbonate solution in PBS was accompanied by an increase in the infectivity of larvae isolated in 1% pepsin/HCl (P/HCl) compared to that for worms inoculated into hosts given PBS alone. Fewer adult worms developing from larvae isolated in P/HCl became established in the host small bowel than was seen with larvae isolated in PBS; moreover, the fecundity in vitro of adult worms developing from P/HCl-isolated larvae was reduced below that for adults developing from larvae isolated from host muscle in PBS. More adult worms were recovered following infection of immune hosts with PBS-isolated larvae than were recovered from immune mice challenged with larvae isolated in P/HCl. Similar findings were observed in mice immunized by infection with Trichinella spiralis and challenged with T. pseudospiralis larvae isolated in either P/HCl or PBS. Immunization of mice with T. pseudospiralis larvae isolated by either method and challenged with larvae of T. spiralis resulted in recovery of similar percentages of the challenge inoculum.  相似文献

The effects of sublethal concentrations of chlorpyrifos, cyfluthrin, and hydramethylnon on adult longevity and fecundity of German cockroaches, Blattella germanica (L.), were investigated. Longevity of males declined linearly with increasing doses of insecticide. An LD50 of cyfluthrin decreased male longevity by 52%, whereas an LD50 of hydramethylnon reduced male longevity by 81%. Longevity of females increased linearly with increasing sublethal doses of chlorpyrifos, whereas all doses of both cyfluthrin and hydramethylnon decreased longevity of females. Fecundity increased linearly with increasing sublethal doses of chlorpyrifos. Number of oothecae formed, oothecae hatched, and number of offspring produced in each ootheca increased with increasing sublethal concentrations of chlorpyrifos. In contrast, fecundity declined with increasing sublethal concentrations of cyfluthrin and hydramethylnon. Formation of the first ootheca occurred approximately 8 d after mating for untreated females but generally longer with sublethal concentrations of all insecticides. The period between oothecae hatch and the formation of subsequent oothecae increased with successive oothecae in all treatments.  相似文献

Density-dependent factors frequently have been shown to regulate population parameters of free-living and parasitic helminths. To test the effects of various infection levels of Haemonchus contortus on fecundity and worm size, lambs were inoculated with 3,000, 10,000, and 30,000 infective larvae. Daily eggs per gram (epg) and daily total fecal production per lamb were monitored continuously. Worms were collected from abomasa on 6, 15, 22, and 30 days postinfection (PI). Female worms were smaller on each day in the high-dose group when compared to female worms from the low-dose group; males in the high-dose group were smaller from days 15 through 30 PI. The high- and medium-dose groups had higher mortality rates on day 30 PI, and fecundity (eggs/female/day) was 78% lower. Daily epg and daily total eggs/lamb/day were lower in the high-dose group. Fecundity and worm size were correlated with the log-transformed dose level but not with adult worm number. Early parasite and/or host responses apparently exert long-term negative effects on growth and reproduction relative to the size of the establishing population of H. contortus.  相似文献

Constant infusion of LH (400 micrograms NIH-S24) through an osmotic minipump inserted on Day 1 (oestrus) of the cycle in the hamster resulted in spontaneous superovulation (approximately equal to 29 ova) at the next expected oestrus, increased blood flow (P less than 0.001) to the ovary on Day 3, and slight depletion (0.1 greater than P greater than 0.05) of histamine in the ovary. Treatment with antihistamine (alpha-fluoromethylhistidine, an irreversible inhibitor of histidine decarboxylase, or cimetidine, an H2 blocker) by injections or by infusion using separate osmotic minipumps significantly (P less than 0.01) reduced the number of ova shed in the LH-treated hamsters. Infusion of LH with alpha-fluoromethylhistidine in the same osmotic minipump reduced the bioactivity of the LH. Infusion of antihistamine alone did not alter the normal number of ova shed. The results suggest that the LH-induced superovulation involves stimulation of histamine release; the histamine than may increase ovarian blood flow thus allowing more gonadotrophins to reach the ovary.  相似文献

Background

The near exclusive use of praziquantel (PZQ) for treatment of human schistosomiasis has raised concerns about the possible emergence of drug-resistant schistosomes.

Methodology/Principal Findings

We measured susceptibility to PZQ of isolates of Schistosoma mansoni obtained from patients from Kisumu, Kenya continuously exposed to infection as a consequence of their occupations as car washers or sand harvesters. We used a) an in vitro assay with miracidia, b) an in vivo assay targeting adult worms in mice and c) an in vitro assay targeting adult schistosomes perfused from mice. In the miracidia assay, in which miracidia from human patients were exposed to PZQ in vitro, reduced susceptibility was associated with previous treatment of the patient with PZQ. One isolate (“KCW”) that was less susceptible to PZQ and had been derived from a patient who had never fully cured despite multiple treatments was studied further. In an in vivo assay of adult worms, the KCW isolate was significantly less susceptible to PZQ than two other isolates from natural infections in Kenya and two lab-reared strains of S. mansoni. The in vitro adult assay, based on measuring length changes of adults following exposure to and recovery from PZQ, confirmed that the KCW isolate was less susceptible to PZQ than the other isolates tested. A sub-isolate of KCW maintained separately and tested after three years was susceptible to PZQ, indicative that the trait of reduced sensitivity could be lost if selection was not maintained.

Conclusions/Significance

Isolates of S. mansoni from some patients in Kisumu have lower susceptibility to PZQ, including one from a patient who was never fully cured after repeated rounds of treatment administered over several years. As use of PZQ continues, continued selection for worms with diminished susceptibility is possible, and the probability of emergence of resistance will increase as large reservoirs of untreated worms diminish. The potential for rapid emergence of resistance should be an important consideration of treatment programs.  相似文献
