Excysted metacercariae of Echinostoma caproni were cultivated on the chick chorioallantoic membrane (CAM) maintained at 38.5 +/- 1 C and a relative humidity of 60-65%. Of 59 6-day-old embryos, each inoculated with 25 metacercariae, 29 (49.2%) were infected 2-12 days postinoculation. The total number of worms recovered from the infected eggs was 163 or 22.5% of the 725 inoculated metacercariae. Eggs contained from 1 to 12 (average 5.6) worms per CAM. Worm length increased rapidly from an average of 0.5 mm at 2 days to about 3.0 mm at 6 days postinoculation. Ovigerous worms first were seen on day 8 PI, but fluke eggs did not develop embryos. Worm development in ovo lagged about 1 day behind that of in vivo worms. One worm maintained for 17 days on 2 successive CAMs reached 6 mm in length, contained about 100 eggs in its uterus, and laid an additional 100 eggs on the CAM surface.  相似文献

Cercariae and schistosomula of Trichobilharzia ocellata were cultured to the organogeny stage in vitro in a medium based on Earle's saline (gassed with 5% CO2 in air) and containing lactalbumen hydrolysate and duck or chicken serum together with homologous red cells. Similar results were obtained irrespective of whether cercariae, schistosomula recovered from the skin of ducklings or chickens, or schistosomula recovered after cercariae had penetrated gelatine membranes were used to initiate cultures. A lipid coat developed around the body of each worm during the first 2–3 days in vitro, but subsequently it became dislodged. The worms ingested red cells, and the caeca became dark with haematin after a few days but by day 10 they were generally translucent. Maximum development was achieved at day 12; by this time males had attained a length of 2·1 mm and females 1·4 mm. Worms cultured for 7 and 9 days were injected into the leg veins of ducks and patent infections were established.  相似文献

Spermatozoa of the American oyster Crassostrea virginica were frozen to ?196 °C in concentrated Hanks' salt solution (2.6×) containing 8% dimethyl sulfoxide. About 0.2 ml of spermatozoa that were stored in liquid nitrogen for 68 days fertilized 91% of 65,600 eggs compared to fresh spermatozoa, which fertilized 92% of approximately the same number of eggs from the same females. Larvae resulting from fertilization of fresh eggs with 39-day-old cryopreserved spermatozoa appeared normal after 11 days.  相似文献

Of 50 white-throated woodrats (Neotoma albigula) collected from Socorro Co., New Mexico, 21 (42%) had eimerian oocysts in their feces when examined. Of the 21 Neotoma found positive for Eimeria, 19 (90%) harbored a single eimerian species at time of examination. Eimeria albigulae Levine, Ivens & Kruidenier, 1957, was found in 18 (86%), and E. ladronensis n. sp. was found in five (24%) infected woodrats. Sporulated oocysts of E. ladronensis are ellipsoidal, 19–25 × 13–15 (21.4 ± 1.3 × 14.1 ± 1.1) μm, have a smooth wall and one or two polar granules, but lack a micropyle and an oocyst residuum. Sporocysts are tapered at one end, 7–10 × 6–7 (8.5 ± 0.7 × 6.5 ± 0.3) μm, and have a Stieda body and sporocyst residuum, but no substieda body. Prepatent periods for E. albigulae and E. ladronensis n. sp. are 5–6 and 8–9 days, respectively; patent periods are 7–18 and approximately 11 days, respectively.  相似文献

The life cycle of I. canis Nemeséri, 1959 was studied in experimentally infected dogs. Freshly sporulated oocysts were ovoid and 34–40 × 28–32 μm. The endogenous stages were found directly beneath the epithelium of the distal portion of the small intestinal villi. Most of the endogenous stages were in the lower 1/3 of the small intestine, but occasionally they were found in other portions of the small intestine. Three asexual generations were present. First-generation schizonts were 16–38 × 11–23 μm and contained 4–24 merozoites; mature 1st-generation merozoites were 8–11 × 3–5 μm. First-generation schizogony lasted up to 7 days after inoculation. Second-generation schizonts were 12–18 × 8–13 μm and contained up to 12 merozoites which were 11–13 × 3–5 μm. Second-generation schizogony was present on postinoculation days 6 and 7. Third-generation schizonts were formed by nuclear division of 2nd-generation merozoites. Most 2nd-generation merozoites underwent nuclear division without leaving the parasitophorous vacuole of the 2nd-generation schizont. Mature 3rd-generation schizonts were 13–38 × 8–24 μm and contained 6–72 merozoites. Third-generation merozoites were 8–13 × 1–3 μm. Third-generation schizogony was present on days 6–8 after inoculation. Mature macrogametes were 22–29 × 14–23 μm. Mature microgametocytes were 20–38 × 14–26 μm. Gametes were present on postinoculation days 7–10. Oocysts were present in tissue sections on postinoculation days 8–10 and 12. The prepatent period was 9–11 days.  相似文献

A total of 1440 one-day-old female White Leghorn birds was used in a 2 × 4 × 2 factorial experiment to evaluate the inclusion of torula yeast (grown on sugar cane final molasses) in grower diets for 0–6 weeks of age (0 and 20%), for 7–23 weeks (0, 6, 12 and 17%) and in laying hens' diets (0 and 18%). In the period 0–6 weeks of age, final live weight and feed conversion (465 and 444 g and 2.70 and 2.82) obtained with 0 and 20%, respectively, differed significantly (P < 0.05) but this had no effect on the later performance of the birds. For 7–19 weeks of age, average live weight gain (1143, 1119, 1093 and 1021 g/bird) was reduced by increasing the percentage of torula yeast in the diet, but conversion (4.43, 4.47, 4.48 and 4.72) increased significantly only with 17% torula. Torula yeast levels used for weeks 7–23 had no influence on subsequent egg production. The inclusion of 18% yeast during the laying period did not affect egg production (250 and 247 eggs/bird/year) nor egg weight (58.4 and 58.1 g) but increased consumption from 37.4 to 38.8 kg/bird/year. The results obtained suggest the possibility of including 20%, 17% and 18% torula yeast in diets for White Leghorn birds for weeks 0–6, weeks 7–23 and for laying hens, respectively.  相似文献

Heliothis zea larval mortality time was established for topical applications and per os (feeding) with fungal conidia of Nomuraea rileyi. The 6-to 7-day-old larvae received per os dosages ranging from 6 × 106 to 9 × 109 and a topical dosage of ca. 1 × 109. These spore loads initiated fungal infections in larvae resulting in 1.5–100% accumulated mortality during the 30-day testing periods. The data indicated that regardless of the larval treatment method (topical or per os), or conidial dosage rates, the larval mortality time was never shorter than 5 days post-treatment, nor longer than 22 days. The highest percentage of larval deaths occurred from 7 to 9 days with a maximum mortality at 8 days.  相似文献

Peripheral eosinophilia is induced in Sprague-Dawley rats following infection with cercariae of Schistosoma mansoni. Beginning 3 weeks after infection, peripheral eosinophil levels rise above the baseline range; they reach peak values during the fifth week. Following a decline from peak values, peripheral eosinophil levels remain elevated and are observed to fluctuate for the next 5 months. The magnitudes of both the initial peak response at 5 weeks and the subsequent chronic level of peripheral eosinophils are dependent upon dose of cercariae. The initial peak response phase of peripheral eosinophilia coincides in time with the period of adult worm elimination (Weeks 4–6) in the schistosome-infected rat. Histological examination of the liver at 5 weeks after infection reveals eosinophil-rich inflammatory reactions associated with both live and dead worms residing in the portal blood vessels. Around live worms the inflammatory cells are localized in a perivascular arrangement; around dead worms these cells are in the vascular lumen in contact with destroyed worms. The chronic phase of peripheral eosinophilia is associated, in part, with inflammatory reactions surrounding eggs deposited in the liver by the few worm pairs which survive more than 6 weeks and remain within the liver. Histological examination during this period reveals granulomatous lesions within the liver surrounding eggs and dead worms. The granulomas are predominately monocytic (lymphocytes, macrophages) at 11 and 16 weeks. The initial peak response phase of peripheral eosinophilia appears to be a marker for tissue-localized reactions of eosinophils with worms. There are relationships between inflammatory reactions and survival of adult worms.  相似文献

In the northern Sea of Okhotsk, nekton and jellyfish consumed as many as 831 × 109 walleye pollock eggs per day in 2011. The nekton exerted the highest pressure, viz., 98.3% of the overall predation on pollock egg by aquatic animals. Of the entire quantity of consumed eggs, 55.9% were eaten by herring, 35.9% by walleye pollock, 6.5% by Sakhalin sole, and 1.7% by jellyfish. Among jellyfish, scyphomedusae Cyanea capillata and Chrysaora melonaster, as well as the hydromedusa Tima sachalinensis consumed the largest quantities of eggs. The total consumption of pollock egg by aquatic animals in 2011 was estimated at 42.4 × 1012, or 11.4% of the entire quantity of eggs that were spawned by walleye pollock in the waters of the northern part of the sea. The total amount of pollock eggs that were eaten by herring and pollock together for 51 days in 2011 amounted to 38.9 × 1012, which was 5.7 times as much as that in 2002. Thus, a significant growth of predation on pollock eggs by their main consumers, viz., herring and walleye pollock, was observed in 2011. This was caused by an increase in the populations of both species during the recent years and also by a higher concentration of pollock eggs.  相似文献

Coprological examination of the worm lizard Diplometopon zarudnyi Nikolskii revealed the presence of oöcysts of Choleoeimeria zarudnyi (Alyousif & Al-Shawa, 2003) n. comb. in five (17%) of the 30 lizards examined. Sporulated oöcysts were found in the faeces and the gallbladder contents. These are tetrasporocystic, ellipsoidal, 25–32 × 18–25 (mean 27 × 22) μm, with a smooth bi-layered wall. The dizoic sporocysts are ovoidal, 10–13 × 6–9 (mean 11 × 7) μm, with a granulated sporocyst residuum. Sporozoites are banana-shaped with an average size of 13 × 3 μm. Endogenous stages (meronts, gamonts and gametes) are confined to the gallbladder epithelium and the infected cells were hypertrophied. Based on the morphological features of the exogenous stages and the endogenous development of the present parasite, its generic affiliation is revised and Eimeria zarudnyi Alyousif & Al-Shawa, 2003 is transferred to the genus Choleoeimeria.  相似文献

An average of 11% of adult Schistosoma curassoni were recovered from 200 albino mice which had been infected subcutaneously with 150-250 cercariae. Worms were primarily found in the portal veins. The average number of intrauterine eggs per female during the first 100 days p.i. was 13 and the average number of eggs produced per female worm was 103 per day for the first 60 days post oviposition. The majority of eggs were recovered from the liver (98.3%). The oograms were determined until day 95 p.i. For screening of antischistosomal drugs it is recommended not to use infections older than 60 d.p.i.  相似文献

Vaccination against schistosomiasis with highly X-irradiated Schistosoma mansoni cercariae was studied in mice. The optimum dose of X radiation for the attenuation of cercariae was in the range of 24–48 krad. In selecting the optimum dose, lesions caused by migrating schistosomula in the lungs of the immunized host were considered. Cercariae exposed to 48 krad caused fewer lesions than those exposed to 24 krad but still effected a comparable worm reduction. The percentages of worm reduction in mice immunized with 48-krad X-irradiated cercariae increased with the number of immunizations up to the fifth immunization and then fluctuated in the sixth, seventh, and eighth days without increase. The optimum dose of immunizing cercariae was 500, and the optimum time interval for successive immunizations was 4 weeks. There was no significant difference in susceptibility to infection in the adult mice 161 to 694 days of age. The duration of acquired immunity in immunized mice is long, still evident 545 days from the last immunization. The present studies clearly showed that with the bioengineering method, the worm reduction in the immunized mice reached 91.1%, the effect of immunization was stronger in mice immunized with the highly X-irradiated cercariae than with the low X-irradiated cercariae, and X-irradiated cercariae were demonstrated to be a strong inducing agent for immunity in mice.  相似文献

Male/female pairs of adult milkweed bugs, Oncopeltus faciatus were offered reduced food rations (100 mg milkweed seeds/week, 50 mg seeds/week, and 25 mg seeds/week) at 232C, 14L : 10D photoperiod. The high rate of food consumption normally occurring during the teneral period (first 8 days posteclosion) and subsequent food consumption were reduced on both reduced rations. Initial fresh weight was lost by bugs on both reduced rations and their weekly fresh body weight tended to remain less than bugs on the 100 mg seeds/week ration.Normal onset of egg production (about 3 weeks posteclosion) was delayed 6 weeks and normal length of a female's life (13–14 weeks) was increased 7–8 weeks on the 25 mg seeds/week ration. Rate of egg production exhibited a linear decrease with decrease in food ration on both reduced rations due to an increased interclutch interval, while fresh egg weight, duration of egg stage, number of eggs/clutch and percentage of clutches containing fertile eggs were little affected.Gross egg production efficiency (efficiency of conversion of ingested food into eggs = dry mg egg production × 100/dry mg food ingested) was high on both the 100 and 50 mg seeds/week rations (27.9% and 21.1%, respectively) and low (7.7%) on the 25 mg seeds/week ration. These data are discussed in the context of the reciprocal interaction between nutrient intake and egg production.  相似文献

Donnelly F. A., Appleton C. C. and Schutte C. H. J. 1984. The influence of salinity on the cercariae of three species of Schistosoma. International Journal for Parasitology14: 13–21. The effect of salinity on the longevity and infectivity of cercariae of Schistosoma mattheei, Schistosoma haematobium and Schistosoma mansoni was determined. No significant differences in cercarial longevity occurred (p > 0.05) in low salinities (0–5.25%), whereas further increases in salinity resulted in progressive decreases in survival. In salinities ? 17.5%, cercariae were incapable of surviving for longer than 11 min. A maximum life-span of up to 122 h was recorded for some S. mattheei cercariae. Cercarial infectivity, as indicated by worm returns, was reduced progressively with increasing salinity up to a lethal limit of 10.5%. Differences in the salinity tolerance of the cercariae of the three species were discussed.  相似文献

Chemical composition of newborn larvae, muscle larvae and adult Trichinella spiralis. International Journal for Parasitology16: 455–460. The chemical composition of newborn larvae (NBL), muscle larvae (ML) and 4-day-old and 7-day-old adult Trichinella spiralis were compared. Total protein constituted 44.1% of the dry wt of ML, 36.9% of NBL and 30.3% of 7-day-old adult worms. Glycogen content was 16.1% of worm dry wt in ML and 7.8% in NBL and 7.2% of worm dry wt in adults. Trehalose content was 5.2% in NBL, 8.3% in ML and 8.7% in 7-day-old adult worms. Significantly lower levels of trehalose and glycogen were found in 4-day-old worms than were present in 7-day-old worms. Free glucose amounted to 0.1% of dry wt in ML, 0.37% in NBL and 0.87% in 7-day-old adults. RNA accounted for 3.5% of the dry wt of ML, 3.2% of the dry wt of 7-day-old adults and 1.1% of the dry wt of NBL. The DNA content of NBL was 0.23% of worm dry wt, in ML 0.48% of worm dry wt and 0.51% of worm dry wt in 7-day-old adults. The three parasite stages examined agreed closely with regard to types of amino acids in the free pool. Exceptions were as follows: NBL lacked taurine which both adults and ML contained; adults lacked methionine which both ML and NBL contained; and, trace amounts of cysteine were present in ML but absent from the other two stages.  相似文献

Three techniques were used to study post-metacercarial growth and development of chemically excysted metacercariae of Echinostoma revolutum on the chorioallantoic membrane of domestic chick embryos. The in ovo technique of Zwilling (1959) and the in vitro technique of Auerbach et al. (1974) provided for better worm recovery in chick embryos than the in ovo technique of Woodruff &; Goodpasture (1931). Regardless of the technique used, postmetacercarial development was obtained for this species, and 6- to 8-day-old chorioallantoic-wornis achieved sexual development to the coiled uterus stage comparable to worms grown in domestic chicks for 7 days. However, somatic growth of worms from the chorioallantois was stunted when compared to worms grown in chicks. Worms grown on the chorioallantois voided their excretory concretions and showed histochemical lipid staining identical to that seen in worms grown in chicks.  相似文献

Acanthoparyphium shinanense n. sp. (Digenea: Echinostomatidae) is described from chicks experimentally infected with the metacercariae encysted in 2 brackish water clam species, Ruditapes philippinarum and Coecella chinensis, in the Republic of Korea. The metacercariae were round to oval, armed with 23 collar spines, and 0.216 (0.203–0.226) mm in diameter. From 5 chicks experimentally infected each with 200 metacercariae, 34 juvenile (5-day-old worms) and 104 adult flukes (7-day-old worms) were harvested from their small intestines, with the average worm recovery rate of 13.8%. The adult flukes were 3.18 (2.89–3.55) mm long and 0.68 (0.61–0.85) mm wide, with an elongated, posteriorly tapering body, and a prominent head collar armed with 23 collar spines arranged in a single uninterrupted row. The posterior testis of A. shinanense was longitudinally elongated, which is similar to Acanthoparyphium spinulosum Johnston, 1917 but unique from the other closely related species, including Acanthoparyphium tyosenense Yamaguti, 1939, Acanthoparyphium kurogamo Yamaguti, 1939, and Acanthoparyphium marilae Yamaguti, 1934. The eggs of A. shinanense were larger than those of A. spinulosum, and the anterior extent of 2 lateral groups of vitellaria was slightly more limited in A. shinanense than in A. spinulosum. Molecular analysis of nuclear and mitochondrial genes revealed low homology with A. spinulosum from USA (96.1% in 5.8S rRNA) and Ukraine (97.9% in 28S rRNA), Acanthoparyphium n. sp. from USA (98.0% in 28S rRNA), and Acanthoparyphium sp. from Australia, Kuwait, and New Zealand. Biological characteristics, including its first intermediate host and natural definitive hosts, as well as its zoonotic capability, should be elucidated.  相似文献

This study was undertaken to assess the suitability of Danish Landrace/Yorkshire (L/Y) crossbred pigs as experimental hosts of a Chinese mainland strain of Schistosoma japonicum. Pigs were exposed to 200, 500 or 1000 cercariae and parasite burdens were determined by perfusion after either 8 or 11 weeks. All pigs became infected with onset of faecal egg excretion 6 to 7 weeks following exposure to cercariae. The pattern of faecal egg excretion differed markedly among the individual animals. Gross hepatic pathological lesions of varying degrees were noted in all of the pigs. Schistosome worm recoveries ranged from 1.5–23.4% of the cercarial exposure dose. Most schistosome eggs recovered from the tissues, expressed as eggs/g tissue, were found in the rectum (91%), caecum (3.1%) and liver (5.1%). The results show that Danish L/Y pigs may serve as appropriate experimental final hosts of the Chinese mainland strain of S. japonicum.  相似文献

Coccidian oöcysts recovered from the faeces of rough earth snakes Virginia striatula (Serpentes: Colubridae) were found to represent two previously unreported eimerians. Oöcysts of Eimeria desotoensis n. sp. were found in 5/32 (16%) of the snakes and were spherical to ellipsoidal, 18.4 × 17.2 (15–21.5 × 15–19.5) μm, with a thin, single-layered wall; their shape-index (length/width) was 1.07 (1.00–1.23). A micropyle and oöcyst residuum were absent; polar granule were present in 33% of the oöcysts. The sporocysts were ovoidal, 11.5 × 7.6 (10.5–13 × 7–8) μm, with a Stieda body; their shape-index was 1.51 (1.30–1.68). The sporocyst residuum was moderate in size and composed of a cluster of granules. Oöcysts of Eimeria hobartsmithi n. sp. were found in 2/32 (6%) of the snakes and were subspherical to ellipsoidal, 18.0 × 15.7 (16–20 × 15–17) μm, with a thin, single-layered wall; their shape-index was 1.15 (1.02–1.32). A micropyle, oöcyst residuum and polar granule were absent. The sporocysts were elongate, 13.2 × 6.3 (12–14.5 × 6–6.5) μm, with a Stieda body; their shape-index was 2.10 (1.88–2.34). A large sporocyst residuum was present in each sporocyst, often obscuring the sporozoites. In addition to the two new species, oöcysts of E. striatula Upton & McAllister, 1990 were observed in 38% of the snakes.  相似文献
