When water stress was imposed on detached leaves of two rice(Oryza sativa L.) cultivars, more ABA per unit fresh weightaccumulated in IR20, a small-leaved cultivar, than in 63–83,a large-leaved cultivar; the difference being up to threefold.In an F2 population of a cross between the two cultivars ABAaccumulation was found to be significantly negatively correlatedwith leaf fresh weight. This correlation persisted in the F3generation. Such a correlation was not evident, however, whena number of rice cultivars, which varied widely in leaf size,were examined. The difference in ABA accumulation between IR20 and 63–83was not accounted for by different spatial patterns of waterloss or ABA accumulation within a leaf, and cultivar differencesin ABA content were maintained both across, and at various positionsalong the leaf. No major differences in leaf anatomy were observed between thetwo cultivars. Differences found in leaf water relations characteristicswere few and generally minor. It therefore seems unlikely thatthese properties account for the difference between the cultivarsin the ability to accumulate ABA or for the correlation withleaf size. Plants accumulate abscisic acid (ABA) under droughted conditions. Genetic variation in the accumulation of ABA in detached and partially dehydrated leaves of rice has previously been reported, and this was found to be associated with variation in leaf size (smaller leaves made more ABA). Correlation analysis failed to distinguish clearly between a causal relationship between the two traits and close genetic linkage between loci controlling the traits. Here we present a detailed genetic analysis of ABA accumulation in detached and partially dehydrated rice leaves, using a population of F2 plants generated from the lowland × upland cross IR20 (high-ABA) × 63-83 (low-ABA) which was mapped with RFLP and AFLP markers. Several highly significant quantitative trait loci (QTLs) for ABA accumulation and leaf weight were identified. Only one of the minor QTLs for ABA accumulation (accounting for only 4% of the phenotypic variance) was coincident with any QTLs for leaf size such that the high-ABA allele was associated with smaller leaves. This analysis, therefore, showed that the association previously found between ABA accumulation and leaf size was probably largely due to genetic linkage and not to a direct effect of leaf size on ABA accumulation or vice versa. Because of the importance of ABA accumulation in regulating responses of plants to drought stress and the effects of plant size on the rate of development of stress, QTLs for drought-induced ABA accumulation, leaf size and tiller number were compared between rice and wheat. In particular, a possible location in rice was sought for a homoeologue of the major wheat vernalization responsive gene, Vrn1, as this gene is also associated with major effects on leaf size, tiller number and ABA accumulation in wheat. The likelihood of homoeologous loci regulating ABA accumulation, leaf size and tiller number in the two crops is discussed.  相似文献

Water stress was imposed by withholding water at an early vegetativestage from plants of two rice cultivars (IR20 and 63–83)grown in pots. As stress intensified the following sequenceof responses of the leaves was observed: (i) rise in abscisicacid (ABA) content, (ii) closure of stomata, (iii) initiationof leaf rolling. In both cultivars, turgor (p) declined linearly with total waterpotential () of the leaf. Bulk leaf ABA content increased linearlyas p declined, and attained twice the control (unstressed) levelfollowing a reduction in p of about 0.12 MPa. Stomatal conductance exhibited a sigmoidal relationship to p,declining abruptly when a particular ‘critical’p was reached (threshold response). The critical potentialsvaried considerably between experiments, but were closely correlatedwith control potentials and with the potentials at which ABAconcentration doubled relative to controls. Leaf rolling was initiated at s near to zero p. Increases inthe ratio of adaxial to abaxial conductance were associatedwith rolling. Variations in the above responses could be accounted for byvariations in the rate of stress development, which in termsof reduction ranged from 0.38 to 0.86 MPa day–1. Fastdrying rates resulted in: (a) reduced osmotic adjustment, (b)increased amounts of ABA in the leaf at a given level of orp, (c) an increase in the ABA concentration present at 50 percent stomatal closure, and (d) initiation of leaf rolling ata higher . Bot. 36: 1889–1899.Experiments were conducted to investigate whether abscisic acid(ABA) elicits the accumulation of solutes and lowering of osmotic(solute) potential (2) which occurs in leaves of pearl millet(Pennisetum americanum [L.] Leeke) exposed to water stress.When (?)–ABA was injected into the base of the shoot of15–d–old plants, 2 of the fifth leaf was reducedsignificantly below controls 27–72 h after treatment.The reductions, however, were small (< 0.10 MPa) and wereaccompanied by a significant inhibition of shoot growth. Incontrast, no significant reduction in 2 or in growth occurredwhen the hormone was introduced directly into leaf five viaits mid–rib. ABA concentrations in leaf five were highshortly after direct injection, but declined to control levelswithin 48 h. Injecting ABA into the shoot base resulted in lowerleaf five ABA concentrations. Hence, the ABA concentration inthe leaf was not the most critical factor for its effect on2. ABA also reduced 2 of shoots when applied to seedlings 48h or 72 h after sowing via the roots. As with older plants,the effects of ABA on 2 were small ( 0–2 MPa) and wereaccompanied by inhibition of shoot growth. A water stress treatmentand an ABA treatment were compared. Although both treatmentsresulted in a similar degree of growth inhibition, the stresstreatment was much more effective than was ABA in reducing 2. The characteristics of ABA-induced changes in the fluxes ofCO2 and water vapour from whole leaves of spring wheat (Triticumaestivum cv. Wembley) were examined. Aqueous solutions of ABAwere supplied via the transpiration stream to intact leavesof different ages mounted within a gas exchange cuvette. ABA caused a reduction in stomatal conductance (g) that wasproportional to the concentration in the solution fed to theleaf. For the maintenance of a reduction in g there was a requirementfor a continual supply of ABA. At concentrations greater than10–2 mol m–3 ABA reduced g by at least 50% of thecontrol value, while 1.0 mol m–3 closed stomata within2 h. Concentrations as low as 10–3 mol m–3 produceda 20% reduction in g. As leaves aged they became less responsiveto applied ABA. The possibility that the stomatal response may change aftera leaf has previously experienced a pulse of ABA was exploredby repeating the exposure of a leaf to 10–2 mol m–3ABA. The first pulse of ABA produced a greater reduction ing than a subsequent exposure the following day. This declinein response of g to ABA on repeated exposure was maintainedwith leaves of different ages. The characteristics of the stomatal response to ABA are discussedin the context of what is known about the location of receptorsfor the hormone. It seems likely that a failure to respond toABA that has previously accumulated in the guard cells shouldbe viewed by means of maximizing the sensitivity to the currentsupply of ABA. It is suggested that the smaller response ofthe stomata of older leaves to ABA makes them more susceptibleto water stress, so that they can act as sensors for decliningwater potentials to give early protection to younger, metabolicallyactive leaves. In rice, alday, wheat and tobacco (Nicotiana tabacum L. Samsunand Samsun NN) plants which contained large amounts of ABA,the transpiration rate decreased rapidly with 2 ppm SO2 fumigationand reached 20 to 65% of the initial level after 5- to 30-minexposure depending on their ABA contents. In the cases of broadbean and tobacco (N. glutinosa L.) with low ABA contents, therate slightly increased for 20 and 40 min, respectively, afterthe start of the fumigation and then decreased gradually. Thetranspiration rates of corn and sorghum, in spite of their extremelylow ABA contents, pronouncedly decreased with SO2 fumigationand reached 65 and 50%, respectively, of the initial levelsafter 40-min exposure. Foliar application of 0.04 N HC1 to N.tabacum L. Samsun NN leaves remarkably depressed the transpirationrate, while the application of 0.04 M Na2SO3 decreased the rateonly to the same level as water treatment. Foliar applicationof either HCl or Na2SO3 to N. glutinosa L. leaves exerted littlechange in the transpiration rate. When 10–4M ABA was appliedto broad bean leaves prior to HCl and Na2SO3 treatment, theirtranspiration rate was decreased by HCl, but not by Na2SO3 application.In sonicated epidermal strips peeled from broad bean leaves,Na2SO3 produced a slight increase in the stomatal aperture sizein the absence of ABA, but showed no effect in the presenceof ABA. The aperture size was identical in the pH range of 3.0to 7.0 in the incubation medium. In the presence of ABA in themedium, the aperture size was small in the acidic region ofpH with a minimal value at pH 4.0. ABA decreased the aperturesize at concentrations above 10–9 M at pH 4.0 and 10–6M at pH 7.0 in the medium. [2–14C] ABA uptake by epidermalstrips was large in the acidic region, especially at pH 4.0. Gibberellins GA1, GA4, GA8, GA9, GA19, GA20, GA29, GA44, GA81,indole-3-acetic acid (IAA) and abscisic acid (ABA) were identifiedin cambial region tissues of Eucalyptus globulus by comparingmass spectra and Kovats retention indices with those of authenticstandards. Using stable isotope labelled internal standardsGA19, GA20 and GA44 were quantified at levels of 2–7 ng(g fr wt)-1, other GAs were present at levels < 1 ng (g frwt)-1. Levels of IAA and ABA ranged from 417–1, 140 ng(g fr wt)-1 and 86–305 ng (g fr wt)-1 respectively. The possible roles of oxygen and carbon dioxide treatments inthe presence or absence of ethylene on tuber dormancy releasein potato (Solanum tuberosumL.) were examined. Using two gascompositions (I: 60% CO2–20% O2–20% N2and II: 20%CO2–40% O2–40% N2), the phase of tuber dormancyand previous storage temperature were demonstrated to be importantparameters for dormancy release by these gas mixtures. Gas Icaused decreased abscisic acid (ABA) levels within 24 h regardlessof previous storage temperature, although this effect was reversible.Exogenous C2H4, an effective dormancy release agent, also causeddecreased ABA levels within 24 h. It also enhanced dormancyrelease and further promoted ABA losses by gas I. Gas II treatmentled to slight reductions in ABA levels that were further decreasedby C2H4. Sprout length was modelled successfully by multipleregression analysis in terms of glucose and ABA levels withinthe apical eye tissues of Russet Burbank tubers immediatelyafter, and regardless of, previous gas treatments or storagetemperatures. Osmotic stress temporarilyreverses the inhibitions of photosynthesis and stomatal conductanceby abscisic acid—evidence that abscisic acid induces alocalized closure of stomata in intact, detached leaves.—J.exp. The influence of osmotic stress on whole leaf gas exchange wasmonitored in detached leaves of Glycine max supplied with anexogenous concentration (10–5 mol dm–3) of ±abscisicacid (ABA) sufficient to inhibit net photosynthesis and stomatalconductance by 60% and 70%, respectively, under a saturatingirradiance and normal air. Raising the osmotic (sorbitol) concentrationof the ABA solutions feeding leaves elicited rapid and synchronousreversals of the ABA-dependent inhibitions of net photosynthesisand conductance. These reversals reached a peak simultaneously,after which photosynthesis and conductance declined. The magnitudeof the transient stimulations at peak height was dependent uponthe sorbitol concentration of the ABA feeding solution, althoughthe time-course of the transients (half time, 4–6 min)was similar for the different osmotic concentrations applied.Irrespective of transient size the relative changes of photosynthesisand conductance were comparable; consequently the calculatedpartial pressure of CO2 in the substomatal space (Ci) remainedrelatively constant during the transient phase. In contrastto the ABA-treated leaves, elevating the osmotic concentrationof the distilled water supply feeding control leaves stimulatedconductance to a much greater relative extent than photosynthesis.The co-stimulations of photosynthesis and conductance inducedin ABA-treated leaves by osmotic shock were not due to a restrictionin the transpirational uptake of ABA and occurred irrespectiveof the source osmoticum applied. These data are consistent with the hypothesis that the ABA-dependentinhibition of photosynthesis at constant Ci is an artifact causedby the spatially heterogeneous closure of stomata in responseto ABA. Albino corn (Zea mays L.) seedlings lacking natural leaf pigmentswere obtained by germinating seeds treated with fluridone, aninhibitor of carotenoid biosynthesis. Basal rates of ethyleneproduction were less than 2.0 nl g–1 fr. wt h–1in both treated (albino) and untreated (normal) leaves but increasedby 10- to 20-fold in the presence of added ACC. ACC-dependentethylene production (ADEP) was inhibited by cobalt or cyanideions and stimulated by NaHCO3, CO2 and light. ADEP in both tissueswas stimulated by glucose, fructose, galactose and sucrose.The accumulation of respiratory CO2 did not account for thecarbohydrate response. The decline in the ADEP characteristicof albino leaf tissue was slowed by incubation in the presenceof sucrose. IAA and ABA stimulated ADEP in normal leaves butinhibited ADEP in albino leaves. Sucrose-stimulated ADEP wasinhibited in albino leaf tissue treated with IAA or ABA indicatinga possible role for the chloroplast in carbohydrate-facilitatedADEP. However, results from this study suggest that chloroplastsperform a function in the regulation of ethylene productionby leaf tissue that extends beyond merely influencing internallevels of CO2. In the absence of detectable ACC, EFE was responsiblefor the entire series of responses expressed in regulation ofethylene biosynthesis by corn seedling leaf tissue. Leaves from in vitro and greenhouse cultured plants of Malusdomestica (Borkh.) cv. Mark were subjected to 4 h of darkness;4 h of 1 M mannitol induced water stress; 1 h of 10–4M to 10–7 M cis-trans abscisic acid (ABA) treatment; 1h of 0.12% atmospheric CO2. Stomatal closure was determinedby microscopic examination of leaf imprints. In all treatments,less than 5% of the stomata from leaves of in vitro culturedplants were closed. The diameter of open stomata on leaves fromin vitro culture remained at 8 µm. In contrast, an averageof 96% of the stomata on leaves of greenhouse grown plants wereclosed after 4 h in darkness; 56% after 4 h of mannitol inducedwater stress; 90% after 1 h of 10–4 M ABA treatment; 61%after 1 h in an atmosphere of 0.12% CO2. Stomata of in vitroapple leaves did not seem to have a closure mechanism, but acquiredone during acclimatization to the greenhouse environment. Thelack of stomatal closure in in vitro plants was the main causeof rapid water loss during transfer to low relative humidity.  相似文献

The effects of a period of water stress (drought conditioning)on responses to a second (challenge) stress were examined inyoung vegetative rice (Oryza sativa L.) plants. Drought conditioningdid not affect the rate of subsequent stress development, nor,in a first experiment, did it influence relations between turgor(p) and total () leaf water potential. However, conditioningdid extend the range of p over which stomata remained open andsignificantly reduced the amount of ABA which accumulated inthe leaf at a given p. The change in stomatal behaviour (stomataladjustment) was quantitatively accounted for by the change inleaf ABA accumulation. The reduction in ABA accumulation due to conditioning did notinvolve a change in the potential capacity to produce ABA, asABA accumulation in partially dehydrated detached leaves wasnot reduced by conditioning. It is suggested that effects ofconditioning on leaf ABA content in the intact plant involvechanges in the rate of ABA export from the leaf. The Role of Endogenous Abscisic Acid in the Response of Plants to Stress   总被引:11,自引:1,他引:10  
When a continuous stream of warm air (38°C) was directedon to the leaves of dwarf bean seedlings they wilted and thengradually regained turgor. This process of adaptation was accompaniedby an increasing abscisic acid (ABA) level in the leaves andan increase in leaf resistance (RL). It is suggested that theleaf-water deficit induced by the warm-air treatment causedthe increase in ABA level and that the latter was responsiblefor stimulating stomatal closure, enabling the plants to regainfull turgor. A similar type of adaptation, brought about byan increased level of ABA in the leaves, is believed to occurin tomato, dwarf bean, and wheat plants when they are flooded.Predictably, in rice, a species adapted to a flooded environment,seedlings showed no increase in ABA level as a result of flooding. It is proposed that adaptation may involve the formation ofan equilibrium between ABA and its conjugate form (i. e. theglucose ester). The ABA-conjugate was observed to disperse slowlyfrom leaves recovering from a water deficit and therefore itmay act as a metabolic ‘back-stop’, enabling the‘free’ ABA level to remain high for a period evenwhen the leaves have regained turgor. Abscisic acid appears to be responsible for alleviating theeffects of water stress in plants, making it possible for plantsto pass through periods of stress with little harm.  相似文献

Epidermal strips from either well-watered or water-stressedplants of Commelina communis L. were subjected to a range ofABA concentrations (10–6–10–3 mol m–3)in the presence (330 parts 10–6 in air) or virtual absence(3 parts 10–6 in air) of CO2. The stomatal response toCO2 was greater in epidermis from water-stressed plants, althoughthere was a distinct CO2 response in epidermis from well-wateredplants. Additions of ABA via the incubation medium had littleeffect on the relative CO2 response. Stomata responded to ABAboth in the presence and virtual absence of CO2, but the relativeresponse to ABA was greatest in the high CO2 treatment. Whenwell-watered plants were sprayed with a 10–1 mol m–3ABA solution 1 d prior to use, the stomatal response of detachedepidermis to both CO2 and ABA was very similar to that of epidermisdetached from water-stressed leaves. It is hypothesized thata prolonged exposure to ABA is necessary before there is anymodification of the CO2 response of stomata.  相似文献

A nitrate reductase inactivating factor was found in extractsof leaf blades, leaf sheaths, and roots of rice seedlings. Thefactor was nondialyzable, precipitable with (NH4)2SO4, and heatlabile. The factor from rice roots inactivated NADH nitratereductase, FMNH2 nitrate reductase, and NADH cytochrome c reductasefrom rice shoots, but had no effect on the activities of NADHdiaphorase and nitrite reductase. The factors from rice shoots,rice roots, and maize roots inactivated NADH nitrate reductaseprepared from cultured rice cells. The factor from culturedrice cells also inactivated rice shoot NADH nitrate reductase. The activity of the inactivating factor showed a diurnal changein shoots of rice seedlings grown with NO3– medium, althoughthe fluctuation was not large compared to that of NADH nitratereductase activity. When the seedlings were placed in darkness,the activity of the factor did not change during 20 hr withNO3– medium. However, the activity of the factor fluctuatedwith NO3– -free medium in light; its activity startedto increase at the 8th hour after transfer. Fragments of maize leaves were incubated at controlled temperatureand irradiance either on distilled water or on one of threeconcentrations of cytokinin (10–1, 10–2 and 10–3mol m–3). The effects of zeatin or kinetin on stomatalaperture were determined by stripping abaxial epidermis fromthe fragments after incubation and immediately measuring stomatalapertures under the microscope. At each cytokinin concentrationleaf pieces were incubated at 5 or 350 µmol mol–1CO2 with or without ABA (10–1 mol m–3). At 5.0 µmolmol–1 CO2 increasing the concentrations of zeatin hada negligible effect upon stomatal aperture. When air containing350 umol mol–1 CO2 was bubbled through the incubationsolutions, apertures of stomata incubated on water were morethan halved. Increasing cytokinin concentrations reduced theeffect of CO2 on stomata and incubation on 10–1 mol m–3zeatin completely removed any CO2 response. Carbon dioxide and water vapour exchanges of the second leafof Zea mays in controlled environment cuvettes were measuredin an open gas-exchange system, during and following subjectionto low temperature stress. Photosynthetic CO2 assimilation (Fc)decreased markedly with decrease in leaf temperature so thatFc at 5 °C was c. 7% of Fc at 20 °C. Fc continued todecline if leaf temperature was maintained at 5 °C, andwhen returned to 20 °C the leaf could not regain its previousFc. This chill-induced reduction in the capacity of the leafto assimilate CO2 was proportional to the duration of the chilland increased with water vapour pressure deficit and photonflux density (In). Six hours at 5 °C decreased Fc on returnto 20 °C, relative to Fc prior to treatment, by c. 10% indarkness and by c. 50% in a photon flux density approachingfull-sunlight (Ip = 1.5 mmol m–2 s–1). The degreeof reduction in Fc following chill treatment showed an almostlinear dependence on both the length and temperature of thechill. Chill treatments resulted in a decrease in both stomataland mesophyll conductances. Examination of the responses ofFc to light and CO2 concentration suggested that chill damageto the capacity for CO2 assimilation resulted from effects onboth the light and CO2 limited processes within photosynthesis. Wheat (Triticum aestivum L.) embryos form in dynamically-regulatedovular environments. Our objectives were to improve developmentof cultured immature wheat embryos by simulating, in vitro,abscisic acid (ABA) levels and O2 tensions as found in wheatovules during zygotic embryogenesis. We characterized from intactwheat kernels embryo respiration, embryo morphology and embryoand endosperm + ABA levels at 13, 19 and 25 d post-anthesis(DPA). Young (13 DPA) embryos were then excised and culturedin vitro, where they were exposed to 0·2 or 2·Ommol m–3 ±ABA and 2.·1, 2·5 or 7·4mol m–3 (6, 7 and 21%, respectively) gaseous O2. At 6and 12 d in culture, + ABA levels, embryo respiration and embryomorphology were characterized by treatment. Thirteen-day-oldembryos from two different plant populations differed by 17-foldin initial ABA content. However, this difference did not affectprecocious germination in vitro, nor did it affect the amountof exogenous ABA required to reduce precocious germination by40%. In this respect, embryos from both populations were equallysensitive to exogenous ABA. Cavity sap O2 levels (2·1to 2·5 mol m–3) were much more effective in preventingprecocious germination of cultured embryos than were cavitysap levels of ABA (0·2 to 2·0 mmol m–3).The combination of physiological levels of both ABA and O2 largelynormalized DW accumulation and embryo morphology without alteringendogenous + ABA levels. Of the 560 leaf samples belonging to 259 species of green plantsexamined more than 50 per cent of the Angiosperms and 25 percent of the Pteridophytes and Gymnosperms revealed the presenceof N2-fixing micro-organisms in their phyllosphere. Plants particularlyremarkable in this respect are orchids and several other epiphytes,Scindapsus officinalis, Ficus and cucurbits. Most of the isolatesappear to be biotypes of Klebsiella pneumoniae. The more activestrains fixed more than 5 mg N g–1 glucose utilized andreduced more than 100 nmol C2H2 mg–1 cell d. w h–1. The efficacy of the phyllosphere N2-fixing isolates for N-nutritionof host plants was studied by spraying suspensions of the culturesgrown on N-free media on rice and wheat seedlings. In IR-26rice or Sonalika and Janak wheat grown on soil in wooden flatsor earthenware pots, 22 per cent of the 161 cultures studiedcaused increased height and about three-quarters of the culturesenhanced dry weight by more than 50 per cent; chlorophyll andN-contents were enhanced more than 50 per cent by about halfand two-thirds of the cultures respectively. In N-free sandculture 26 of the 50 promising strains doubled N-content, and30 doubled dry weight of the tested plants. In some cases dryweight, number of grains per panicle, and 1000 grain weightwere increased by 300, 70–83 and 126–158 per centrespectively; N-content of straw and seed was increased three-or fourfold. In several cases the beneficial effects were foundto match closely the performance of plants receiving ammoniumsulphate.
