Flowering of seedlings of Pharbitis nil, strains Violet andTendan, cultured in modified White's medium, was promoted bymedium dilution, the critical dark period being shortened byabout 15 min. Dilution of the N source alone was enough to causethe medium-dilution effect. Dilution of the culture medium duringthe day before and on the day of exposure to the dark-period(a total of two days) caused the maximum dilution effect. TheC and N contents of the cotyledons and of the shoot apices changedrapidly in response to medium dilution. In 1/2-strength White'smedium with 1/1,000 strength NO3 which was most effectivefor flower promotion, the C-N ratio was highest. In 1/2-strengthmodified White's medium, in which flowering was lowest withthe longest critical dark period, the C-N ratio was lowest.Thus, there is a close relation between flowering response andthe C-N ratio in cotyledons or shoot apices of Pharbitis nil. Gibberellin A3 (GA3) stimulated flowering when it was appliedto the shoot apex of seedlings of Pharbitis nil, dwarf strainKidachi; but, not when it was applied to the cotyledons. GA3applied to the plumule before or shortly after the start ofan inductive dark period promoted both flowering and shoot elongation;but, the later the time of application during the dark periodless the promotion of flowering, although marked promotion ofshoot elongation always took place. The variation with time in the response of flowering to GA3indicates that early floral processes at the apex are stimulatedby GA3, but that subsequent processes are insensitive to it.The early processes of floral stimulus produced by a 16 hr inductivedark period probably are completed within 20 hr at 28°Cafter the end of the dark period. At low temperatures, suchas 15 and 20°C, early floral processes continued for morethan 40 hr. When cotyledons were removed at various times, the export ofthe floral stimulus to the shoot apex was apparent within hoursof the generation of the floral stimulus in the cotyledons,which started with the passage of the critical 9-hr dark period. Etiolated cotyledons of cucumber (Cucumis sativus L.) were excisedand incubated for 16 days on agar in the absence or in the presenceof 6-benzylaminopurine (BA) (3 µM) or KCl (10 mM) undercontinuous light. Treatment with BA or KCl increased growthand greening of cotyledons, KCl being particularly active duringthe last few days of the incubation. The rate of photosynthetic electron transport reached a maximumafter 3 days, as did the chlorophyll content, and then it declinedby approximately 95%. An uncoupling from phosphorylation wasobserved in older cotyledons. BA and KCl increased the rateof electron flow per cotyledon, and KCl delayed the onset ofthe decline in this rate. PS I and PS II activities per chlorophyllunit were lower in BA-treated cotyledons, but KCl-treated cotyledonsbehaved like the controls. Fluorescence analysis indicated that the level of active chlorophyllsinvolved in the transfer of energy in PS II decreased between3 and 7 days under all conditions tested. In BA-treated cotyledons,the proportion of active chlorophylls was consistently lowerthan that in the control. Thus, it appears that, in the absenceof exogenous K+, cytokinin has only a low antisenescence effecton photosynthetic activity, and that K+ may enhance the photosyntheticelectron flow between plastoquinone and plastocyanin via thecytochrome b6-f complex. Maize (Zea mays L., hybrid Cargill 147) seedlings, grown inaseptic conditions, were inoculated with three strains of Azospirillumlipoferum (Al op 33, Al iaa 320, and ATCC 29708) or culturedin different concentrations of indol-3-acetic acid (IAA) orgibberellin A3 (GA3). After 48 h, root length, root surfacearea, root dry weight, and shoot dry weight were measured inall treatments. Gibberellin content was evaluated in selectedroots of control and Azospirillum inoculated seedlings by gaschromatography-mass spectrometry-selected ion monitoring withthe use of deuterated gibberellins as internal standards. Thethree strains of A. lipoferum, IAA (2 ng ml–1), and GA3(40 to 400 pg ml–1) significantly enhanced root growth.Improvement of root hair growth and density was obtained mainlywith A. lipoferum strain Al op 33 and GA3 40 pg ml–1.GA3 was identified by gas chromatography-mass spectrometry (byboth, full scan and selected ion monitoring) in a free acidfraction from roots of the seedlings inoculated with A. lipoferum.In the roots of the non inoculated seedlings GA3 was found afterhydrolysis of a fraction expected to contain glucosyl conjugates. Flowering of etiolated seedlings of Pharbitis nil resulted followinga single, brief red irradiation prior to an inductive dark period.Following this irradiation benzyladenine sprayed on the seedlingsenhanced flowering dramatically and this effect was maximalfor concentrations between 44 and 120µM. In the presenceof benzyladenine a brief (4 to 10 sec) low energy red irradiation(2.6 Wm–2) resulted in flowering and repeated far-redphotoreversal of this red promotion provided clear evidenceof the sole involvement of phytochrome. However, after suchbrief irradiations the critical dark period for flowering waslonger than is normally found in seedlings grown in light whichindicated that additional photoresponses might be importantin natural conditions. Three-day-old dark-grown soybean [Glycine max (L.) Merr.] seedlingswere transferred to 2 mM CaSO4 or 10–5 M dimethipin in2 nM CaSO4 and root-fed via liquid culture. Plants were placedin continuous darkness or in continuous white light (200 µE.m–2?s–11,PAR) at 25?C. Dimethipin inhibited root and shoot elongationin dark-grown plants after 24 h and 48 h, respectively. In thelight, root elongation was inhibited also after 24 h, but hypocotylelongation was not significantly affected. Extractable phenylalanineammonia-lyase (PAL) activity per axis in dimethipin-treateddark-grown axes was not generally affected but, in the lightdimethipin caused a significant decrease in PAL activity (24to 96 h). Total soluble hydroxyphenolics in axes were not affectedby dimethipin in light- or dark-grown plants. Anthocyanin andchlorophyll levels were lowered in hypocotyls of dimethipin-treatedplants after 48 to 96 h. Soluble protein in hypocotyls of light-or dark-grown seedlings was not substantially affected by dimethipin.Nitrate reductase (NR) activity (per organ) was generally notaffected by dimethipin in light-grown cotyledons, but in theroots of these seedlings, NR activity was significantly decreased.Proteolytic enzyme activity using three substrates (leucine-p-nitroanilide,LPNA; proline-p-nitroanilide, PPNA; and benzoylarginine-p-nitroanilide,BAPA) indicated little effect on enzyme activities per organin roots and hypocotyls. These data suggest that dimethipinat low concentrations can cause significant growth inhibitionin soybean seedlings grown in either light or darkness and thatfurthermore, extractable activities of some enzymes associatedwith nitrogen metabolism and secondary metabolism are alteredby this chemical. Time-courses of 14CO2-fixation and of enzyme activities involvedin photorespiration and photosynthesis were determined duringthe life span of cotyledons from sunflower seedlings (Helianthusannuus L.). Glycolate formation in vivo was estimated from theresults of combined labelling and inhibitor experiments. NADPH-glyceraldehyde-3-phosphatedehydrogenase, NADPH-glyoxylate reductase and chlorophyll werewell correlated with the time-course of 14CO2-fixation (photosynthesis).There was, however, a considerable discrepancy between the developmentalsequence of photosynthesis and that of both ribulose-l,5-bisphosphatecarboxylase and glycolate oxidase. Furthermore, time-coursesof glycolate oxidase activity in vitro and of glycolate formationin vivo differed significantly. Therefore, the use of glycolateoxidase as a marker for the activity of photorespiration ingreening sunflower cotyledons may be questionable. Results from14CO2-labelling experiments with cotyledons treated with theglycolate oxidase inhibitor 2-hydroxy butynoic acid suggestthat glycolate formation relative to CO2-fixation is reducedin senescent cotyledons. The effects of a range of applied nitrate (NO3) concentrations(0–20 mol m3) on germination and emergence percentageof Triticum aestivum L. cv. Otane were examined at 30, 60, 90and 120 mm sowing depths. Germination percentage was not affectedby either sowing depth or applied NO3 concentration whereasemergence percentage decreased with increased sowing depth regardlessof applied NO3 concentration. Nitrate did not affectemergence percentage at 30 mm sowing depth, but at 60 to 120mm depth, emergence percentage decreased sharply with an increasedapplied NO3 concentration of 0 to 1·0 mol m–3then decreased only slightly with further increases in appliedNO3 of about 5·0 mol m–3. Root and shoot growth, NO3 accumulation and nitrate reductaseactivity (NRA) of plants supplied with 0, 1·0 and 1·0mol m–3 NO3 at a sowing depth of 60 mm were measuredprior to emergence. The coleoptile of all seedlings opened withinthe substrate. Prior to emergence from the substrate, shootextension growth was unaffected by additional NO3 butshoot fr. wt. and dry wt. were both greater at 1·0 and1·0 mol m–3 NO3 than with zero NO3.Root dry wt. was unaffected by NO3. Nitrate concentrationand NRA in root and shoot were always low without NO3.At 1·0 and 10 mol m3 NO3, NO3 accumulatedin the root and shoot to concentrations substantially greaterthan that applied and caused the induction of NRA. Regardlessof the applied NO3 concentration, seedlings which failedto emerge still had substantial seed reserves one month afterplanting. Coleoptile length was substantially less for seedlingswhich did not emerge than for seedlings which emerged, but wasnot affected by NO3. It is proposed that (a) decreasedemergence percentage with increased sowing depth was due tothe emergence of leaf I from the coleoptile within the substrateand (b) decreased emergence percentage with additional NO3was due to the increased expansion of leaf 1 within the substrateresulting in greater folding and damage of the leaf. Partially purified homoserine dehydrogenase was prepared frompea seedlings. The optimum pH for this enzyme is approximately 5.4. The Kmvaluesfor ASA and TPNH are 4.6xl0–4Af and 7.7xl0–5M, respectively.This enzyme can also utilize DPNH but less effectively thanTPNH. In contrast with yeast homoserine dehydrogenase whichis insensitive to — SH reagents, the pea enzyme is inhibitedalmost completely by 10–4MPCMB and 10–5MHgCl2, theinhibition being removed by 10–2M thioglycolate. Homoserinedehydrogenase was found not only in decotylized seedlings, butalso in cotyledons. The significance of this enzyme in homoserine biosynthesis ingerminating pea seeds has been discussed. The long-day plant Lemna gibba G3 flowers normally in E medium(Hoagland-type medium plus 30 µM EDTA) but in 0.5 H mediumthere is no flowering. Ammonium is present in 0.5 H medium andis known to inhibit flowering in L. gibba G3, but even in NH4+-free0.5 H medium there is virtually no flowering under continuouslight. Increasing the phosphate concentration of the NH4+-free0.5 H medium from 1.15 ITIM to 12 or 16 mM results in substantialflowering. Decreasing the EDTA concentration from 850 µIMto 250 µM, or raising the nitrate concentration from 4mM to 12 mM, results in only a small increase in flowering.If the decrease in EDTA and increase in nitrate are combinedwith the increase in phosphate, however, the flowering responseis nearly as good as that obtained using E medium. Thus, withthese three changes the inhibitory effect of NH4+free 0.5 Hmedium for flowering in L. gibba G3 is almost completely reversed In the above studies flowering was not limited by daylength.When plants were grown on E medium under an 11 hour daylengthwhere flowering is limited by daylength, decreasing the phosphateconcentration in the medium reduced flowering, but increasingthe phosphate concentration in the medium did not stimulateflowering. Thus, when flowering is limited by daylength, highphosphate will not cause flowering, but a certain level of phosphateappears to be necessary for the expression of photoinductionunder long days. Mayoral, M. L., Plaut, Z. and Reinhold, L. 1985. Effect of sink-sourcemanipulations on the photosynthetic rate and carbohydrate contentof cucumber cotyledons.-J. exp. Bot. 36 1551–1558. The photosynthetic rate of cucumber cotyledons (Cucumis sativuscv. Dahla) reached a maximum value of 12 mg dm–2 h–1,10 d after emergence. In 12-d-old seedlings removal of one cotyledondoubled the CO2 fixation rate of the other, as observed 3 dafter treatment. When the primary leaf was removed, the photosyntheticrate of the cotyledons was decreased by 33%. At this stage ofgrowth elimination of the roots as a sink for assimilates bygirdling the hypocotyl affected neither the photosynthetic ratenor the carbohydrate content of the cotyledons. By contrast,in 18-d-old seedlings removal of the first leaf brought abouta 42% increase in the photosynthetic rate of the cotyledons.The simultaneous removal of the first leaf and one cotyledondoubled the rate of CO2 fixation of the remaining cotyledon.Girdling the hypocotyl lowered the photosynthetic rate of thecotyledons by 73%. In both 12- and 18-d-old seedlings a decreaseor increase in the sink-source ratio was correlated with anincrease or a decrease respectively in the carbohydrate contentof the cotyledons. The stomatal resistance of the cotyledonswas not affected by any of the treatments. Upland grasslands are a major component of natural vegetationwithin the UK. Such grasslands support slow growing relativelystable plant communities. The response of native montane grassspecies to elevated atmospheric carbon dioxide concentrationshas received little attention to date. Of such studies, mosthave only focused on short-term (days to weeks) responses, oftenunder favourable controlled environment conditions. In thisstudy Agrostis caplllaris L.5, Festuca vivipara L. and Poa alpinaL. were grown under semi-natural conditions in outdoor open-topchambers at either ambient (340µmol mol–1) or elevated(680µmol mol–1) concentrations of atmospheric carbondioxide (CO2 for periods from 79 to 189 d, with a nutrient availabilitysimilar to that of montane Agrostis-Fescue grassland in Snowdonia,N. Wales. Whole plant dry weight was increased for A. capillarisand P. alpina, but decreased for F. vivipara, at elevated CO2.Major components of relative growth rate (RGR) contributingto this change at elevated CO2 were transient changes in specificleaf area (SLA) and leaf area ratio (LAR). Despite changes ingrowth rate at 680 µmol mol–1 CO2, partitioningof dry weight between shoot and root in plants of A. capillarisand P. alpina was unaltered. There was a significant decreasein shoot relative to root growth at elevated CO2 in F. viviparawhich also showed marked discoloration of the leaves and increasedsenescence of the foliage. Gibberellin A1, (GA1), GA19, and GA20 in phloem exudates andcotyledons of seedlings of Pharbitis nil cv. Violet, grown underdifferent photoperiodic conditions, were qualitatively and semi-quantitativelyanalyzed by a combination of high performance-liquid chromatography(HPLC) and radioimmunoassays (RIA). The levels of GA19 and GA20were higher in cotyledons from plants grown under dark treatment(DT) conditons of 16 h-light/8 h-dark for 6 days followed by8 h-light/16 h-dark for 3 days than in those grown under continuouslight (CL) for 9 days. This relationship was also observed forthe GAs in phloem exudates, although the levels were much lowerthan in the cotyledons. When GAs were applied to the cotyledons,elongation of the epicotyl was promoted more by GA20 than byGA1 or GA19, especially under the CL treatment. The effect of brassinolide (BR) on the reproductive developmentof higher plants was investigated using three different experimentalsystem. Direct application of BR to the staminate inflorescenceof Luffa cylindrica induced bisexual and pistillate flowers,and eventually caused the inflorescence to develop into a shootthat was similar to the main shoot. However, the activity ofBR on this sex modification was inferior to that of N6-benzylaminopurine. BR slightly hastened the flowering of nonvernalized radish plants,Raphanus satiuus cv. Miyashige-sofuto, whereas no such effectwas observed in vernalized plants. GA3 promoted flowering bothin nonvernalized and vernalized plants. Salinity Reduces Water Use and Nitrate-N-use Efficiency of Citrus   总被引:1,自引:0,他引:1  
Five-month-old Cleopatra mandarin (Citrus reticulata Blanco)(CM) and Volkamer lemon (Citrus volkameriana Ten. and Pasq.)(VL) seedlings were grown in a glasshouse in 2·3-1 potsof Candler fine sand. Plants were irrigated with either non-saline(ECe = 0·23 dS m-1) or saline (6·13 dS m-1) waterusing 3:1 NaCl:CaCl2 solution over a 4-week period. A singleapplication of K15NO3 (19·64 atom % excess 15N) at 212mg N1-1, was substituted for a normal weekly fertilization after3 weeks and plants were harvested 7 d later. The transpirationrate, uptake of nitrogen, growth and nitrogen-use efficiency(NUE) on a dry weight basis (mg d. wt mg-1 N) of both specieswas reduced by salinity. Based on growth, water-use and chloride(Cl) accumulation in leaves, VL was more salt-sensitive thanCM, but 15N uptake was equally reduced by salinity in both species.Salinity reduced 15N uptake relatively more than shoot growthover the 7-d period, such that the 15NUE (mg d. wt µg-115N) of new shoot growth of both species increased. There wasno evidence of Cl antagonism of nitrate (NO3) uptake but totalplant 15NO3 uptake was positively correlated with whole planttranspiration in both species. The effects of several growth regulators and amino acids onin vitro organogenesis of Torenia fournieri Lind. were determinedusing internodal segments. Treatment with 2,4-D1 resulted innodular callus formation, while NAA and IAA induced roots constantlybut much less frequently shoot buds. Individually BA, zeatin,and 4-PU induced bud formation, but these shoot buds did notdevelop further. Formation of buds by cytokinin was influencedby a simultaneous application of NAA or 2,4-D, but not of IAA,its degree being reduced when BA was simultaneously appliedwith NAA or 2,4-D. When zeatin or kinetin was added with NAA,numerous roots were induced. The effects of various L-amino acids on in vitro organogenesiswere also investigated using the defined medium in which KNO3was a principal source of nitrogen. The formation of buds wasconsiderably stimulated by alanine and asparagine, and slightlyby glutamic acid in the medium containing both NAA and BA, inwhich bud formation was easily induced. On the other hand, allamino acids except for glutamic acid and aspartic acid inhibitedroom formation in this medium. Root formation was greatly stimulated by proline, alanine, glutamine,glutamic acid, and aspartic acid, and slightly by arginine andtryptophan in the medium containing NAA but no BA. Glutamicacid and aspartic acid also enhanced bud formation in this medium.  相似文献

Abscisic acid (ABA) at 1 x 10–4 M or 3 x 10–4 Mwas applied to the apical buds of Chenopodium rubrum plantsexposed to different photoperiodic treatments and showing differentpatterns of floral differentiation. Stimulation of growth inwidth of the apical meristem of the shoot and/or inhibitionof growth in length was obtained under all photoperiodic treatments.This change of growth pattern was followed by different effectson flowering. In non-induced plants grown under continuous light ABA stimulatedpericlinal divisions in the peripheral zone and the initiationof leaves as well as the growth in width of bud primordia. Inplants induced by two short days reduced growth of the meristemcoincided with ABA application. Longitudinal growth of the meristemwas inhibited in this case and only a temporary stimulationof inflorescence formation took place. In plants induced ata very early stage, ABA exerted a strong inhibitory effect onflowering. A permanent and reproducible stimulatory effect onflowering was obtained in plants induced by three sub-criticalphotoperiodic cycles if ABA was applied to apices released fromapical dominance. In this case formation of lateral organs andinternodes was promoted by ABA and was followed by stimulatedinflorescence formation. Gibberellic acid (GA2) at 1x 10–4M or 3 x 10–4 M brought about a similar effect on floweringas ABA, although the primary growth effect was different, i.e.GA2 stimulated longitudinal growth. The effects of ABA and GA2 on floral differentiation have beencompared with earlier results obtained from auxin and kinetinapplications. These growth hormones are believed to regulateflowering by changing cellular growth within the shoot apex.Depending on the actual state of the meristem identical growthresponses may result in different patterns of organogenesisand even in opposite effects on flowering. Seeds of apple cv. Golden Delicious were germinated and cultivatedin the greenhouse until the third leaf emerged. Respirationofgerminating seeds or photosynthesis of the first leaves wasmeasured by infra-red gas analysis and porometry, respectively.To study the role of phosphoenolpyruvate carboxylase (PEPC),the dominant carboxylase in the carbon economy, its CO2 refixationpotentialwas related to the amount of CO2 lost in respiration. With arange of 0.2 (dry seeds) to 18 (cotyledons) µmol CO2 h–1g–1 PEPC activity resembled or exceeded the amount ofC02 lost in respiration before the third leaf developed. Itis concludedthat PEPC largely contributes to economize the carbonmetabolism of apple seedlings before they become photosyntheticallycompetent. Siddiqi, M. Y. and Glass, A. D. M. 1987. Regulation of K+ influxin barley: Evidence for a direct control of influx by K+ concentrationof root cells.—J. exp. Bot. 38: 935–947. The kinetics of K+ (86Rb+) influx into intact roots of barley(Hordeum vulgare L. cv. Fergus) seedlings having different combinationsof root and shoot [K+], different growth rates and differentroot:shoot weight ratios were studied. K+ influx was stronglycorrelated with root [K+]; shoot [K+], growth rates, and root:shoot ratios appeared to have little effect on K+ influx. Adetailed study showed that both Vmax and Km for K+ influx wereaffected by root [K+] but not by shoot [K+]. We have suggestedthat factors such as growth rates and root: shoot ratio mayaffect K+ influx indirectly primarily via their influence onroot factors such as root [K+]. We have reiterated that othertypes of kinetic control, e.g. increased or decreased synthesisof ‘carrier systems’, may operate in addition todirect (allosteric?) control of K+ influx by root [K+]. Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration.
