Within 10 min after engorging on 10% sucrose, most females of Aedes aegypti do not seek a human blood meal, but remain quiescent and unresponsive to a human hand in a 1 ft3 cage. The duration of this inhibition in blood avidity varies greatly among individuals, but may last for 2 to 5 hr after drinking sucrose. There was no specific correlation between abdominal distension and blood avidity. When females engorged on varying concentrations of sucrose, it was found that as the concentration increased, fewer mosquitoes would take blood when it was offered after 1 hr. Only 25% of the females which engorged on 0·5 to 1 M sucrose took human blood at this time. As the concentration of sucrose increases, there is a marked decrease in spontaneous flight activity during the first hour after feeding. When unfed females are injected with 10% sucrose or trehalose, none of them took blood for 3 hr, whereas 50% of the saline-injected controls fed on blood.  相似文献

It has recently been recognized that flowers pollinated by generalist opportunistic nectarivores tend to have different nectar properties to those pollinated by specialist nectarivores (including both hummingbirds and specialist passerines). While renewed interest in specialist avian nectarivore sugar preferences and digestive physiology has helped explain the concentrated sucrose-dominated nectar of plants they feed on, there has been little progress in understanding why generalist or occasional nectar-feeding birds tend to be associated with flowers that have dilute hexose-dominated nectar. We examined sugar preferences and assimilation efficiencies over a range of concentrations, and concentration preferences, in Dark-capped Bulbuls Pycnonotus tricolor, one of the more common occasional avian nectarivores in southern Africa. Dark-capped Bulbuls showed significant preference for hexose sugar solutions, irrespective of concentration, when given a choice between hexose and sucrose solutions in equicaloric pair-wise choice tests conducted at five different concentrations (5–25%). This contrasts with results from specialist nectarivore groups which generally show a significant concentration-dependant switch in preference from hexose at low concentrations to sucrose at high concentrations for equicaloric solutions. In addition, Dark-capped Bulbuls showed an unusual lack of preference for solutions of higher sugar concentration when simultaneously offered four solutions varying in concentration from 10 to 25%. Dark-capped bulbuls also showed a unique effect of concentration on sugar assimilation efficiency, assimilating relatively more energy on 5% diets than on 25% diets. Although able to assimilate sucrose effectively, assimilation rates of hexose sugars were marginally higher. These results shed new light on pollination systems involving occasional nectarivores and, in particular, help to explain the prevalence of low concentration hexose-dominated nectars in flowers pollinated by these birds.  相似文献

We evaluated in mice their preference for different vegetable oils using the two-bottle choice test. Mice showed preference for all vegetable oils (at 1% concentration in emulsion) tested, including corn oil, canola oil, and mixed vegetable oil, compared to a control fluid without oil. Mice also significantly preferred highly palatable sucrose solutions (5-20%) in this test. One percent corn oil was selected to the same extent as were these sucrose solutions. Mixtures of 1% corn oil and several concentrations of sucrose were preferentially selected as compared to corn oil or sucrose alone. The concentration-response curves of oil-sucrose mixtures vs sucrose (0.5-20% concentrations) were similar to those of sucrose vs control solutions. These results suggest that 1% corn oil and sucrose at the concentrations tested did not affect each other's preference. Chronic intake of 20% sucrose solution, but not 100% corn oil, changed the preference for lower concentrations of each, and both induced remarkable increases in body weight of mice. In conclusion, we showed that mice preferred vegetable oils, and this preference compared favorably with highly palatable sucrose solutions and was not changed by chronic intake of corn oil.  相似文献

The study assessed the influence of sugar concentration (10, 20, 30, 50, 70, 100, 120 g l?1) on growth and ginsenoside biosynthesis in Panax quinquefolium hairy roots cultivated in shake flasks and a nutrient sprinkle bioreactor. The highest growth rate was achieved in medium containing 3–5 % sucrose. More than 70 g l?1 or less than 20 g l?1 sugar content in the medium induces significant inhibition of root growth when cultivated in shake flasks. The saponin content was determined using HPLC. The maximum yield (above 9 mg g?1 d.w.) of the sum of six examined ginsenosides (Rb1, Rb2, Rc, Rd, Re and Rg1) in hairy roots cultivated in shake flasks was obtained with 30 g l?1 sucrose in the medium. The sucrose concentration in the medium was found to correlate with saponin content in bioreactor-cultured specimens. A higher level of protopanaxadiol derivatives was found for lower (20 and 30 g l?1) sucrose concentrations; higher sucrose concentrations (50 and 70 g l?1) in the medium stimulated a higher level of Rg group saponins.  相似文献

In this study, we investigated the possibility of using the droplet-vitrification technique for cryopreserving nodal segments of in vitro plantlets of the endangered plant species Lithodora rosmarinifolia. Among the three vitrification solutions tested, only solutions B1, containing (w/v) 50 % glycerol and 50 % sucrose, and B3, containing 40 % glycerol and 40 % sucrose, were able to induce cryotolerance in nodal explants, resulting in intermediate survival and recovery after cryopreservation. A three-step vitrification protocol, including an additional dehydration treatment with half-strength vitrification solution for 30 min before the treatment with full-strength vitrification solution, did not lead to any improvement in survival and recovery compared with the two-step protocol. The optimal protocol was the following: preculture of nodal segments in liquid medium with 0.3 M sucrose for 16 h and 0.7 M sucrose for 5 h, treatment for 20 min in loading solution containing 1.9 M glycerol + 0.5 M sucrose, dehydration with vitrification solution B1 (glycerol 50.0 %, sucrose 50.0 %, w/v) for 60 min at room temperature, rapid cooling in minute droplets of vitrification solution, and rapid rewarming by immersion of nodal segments for 20 min in unloading solution containing 1.2 M sucrose. Under these conditions, 33 % recovery of cryopreserved nodal explants was achieved. Regrowth of cryopreserved samples was rapid and direct. These results indicate that long-term storage of L. rosmarinifolia by means of cryopreservation of nodal segments is possible, thereby contributing to securing the diversity of this rare and endangered plant species.  相似文献

Orchid is a major floral crop around the world and Dendrobium hybrids are considered to be one of the most popular orchids. In vitro germination of hybrid seeds is a common practice among orchid growers, however, in many cross pollinations the embryos may not develop to maturity, leading to poor seed germination. The effect of seed maturity and sucrose concentration were investigated via asymbiotic germination of nobile Dendrobium hybrids. Capsules were harvested from two hybrids (Den. Lucky Girl × Den. Second Love ‘Kirameki’ and Den. Lucky Girl × Den. Hamana Lake ‘Kumi’) and one selfing of Den. Second Love ‘Kirameki’ at 2, 3, 4, and 5 months after pollination and immature seeds were taken. Immature seeds from 3- to 5-month old capsules could be successfully germinated on Hyponex based medium. Immature seeds from 4-month old capsules showed greatest germination rate of tested treatments, whereas 3-month old immature seeds showed the least germination. After 6 weeks of in vitro culture, protocorms derived from embryos developed on every concentration of sucrose, but germination was greater at lower concentrations. Greater concentration of sucrose decreased normal-developed protocorms.  相似文献

This study investigated the toxicity and effectiveness of various cryoprotectant solutions to develop a cryopreservation technique by one-step fast cooling for gametophytic thalli of Porphyra yezoensis. Dimethyl sulfoxide (DMSO) was not harmful at concentrations up to 30 % after 30 s of immersion. However, an increase in concentration beyond 40 % was seriously detrimental, and the detrimental effect occurred at lower concentrations in the case of 60 s of immersion. Diglycerol was the least harmful among cryoprotectants tested, and its harmful effect was not found at concentrations up to 50 %, even after 60 s of immersion. The thalli loaded into a straw were directly plunged into liquid nitrogen after 30 s of immersion in various cryoprotectant solutions. When 5 % DMSO was added to diglycerol, high survival rates (more than 80 %) were achieved at diglycerol concentrations of 15–25 %, and the maximum survival was 94.6 % at diglycerol concentrations of 20 %. The thawed thalli restarted growth without any difference in comparison with unfrozen thalli.  相似文献

Whether nectarivores or frugivores place selective pressure on the plants they feed on, in terms of nectar or fruit traits, is much debated. Globally sugar preferences, concentration preference and digestive ability of avian nectarivores have been extensively researched. In contrast, relatively little is known about mammalian nectarivores or frugivores in terms of these, particularly Old World species. Consequently effect of sugar type and concentration on food preference in Wahlberg's epauletted fruit bat Epomophorus wahlbergi was investigated. Pair-wise choice tests were conducted using equicaloric hexose and sucrose solutions at five different concentrations (5%-25%). It was expected that they would prefer hexose sugars as these are dominant in available indigenous fruits. However, bats preferred hexoses only when offered dilute (5%) concentrations. From 10% to 25% they showed a decrease in volume intake. Their body mass was generally higher and similar after feeding during the night with the exception of 5% concentration where the mean body mass decreased. When E. wahlbergi were offered a range of sucrose or hexose solutions (10%-25%) respectively, they showed no concentration preference in terms of total volume consumed, nor energy intake. These findings suggest that these fruit bats do not appear to act as a selective pressure on sugar composition in Old World fruit. In fruit bats with high energy requirements, dietary flexibility may be an advantage when faced with seasonal and unpredictable fruit availability.  相似文献

Behavioural experimentation on Phormia regina shows that females previously fed only sucrose, and thus in a state of yeast preference, will ingest single-salt-sucrose mixtures in volumes similar to that of 10% yeast (Rachman et al., 1982). In particular, females will ingest NaCl 0.1 M in 0.2 M sucrose in volumes not statistically different from 10% yeast. In the experiments reported here, groups of females were given ad libitum access to both 0.2 M sucrose and NaCl 0.1 M in 0.2 M sucrose from days 0–4 after eclosion. Potassium and sodium salts were added to 0.2 M sucrose over a spectrum of concentrations and presented to the flies on day 6. The flies rejected all single-salt-sucrose mixtures offered. Even when the pre-test sodium salt was changed to a potassium salt during days 0–4, the flies still rejected the single-salt-sucrose mixtures offered subsequently. However, when sodium and potassium salts were mixed together in 0.2 M sucrose, females ingested the solutions in amounts similar to 10% yeast. Several different concentrations of the double-salt-sucrose solutions were ingested in volumes not statistically different from 10% yeast. Results support the underlying hypothesis that activity in the salt-sensitive chemosensory neurones is involved in protein preference.  相似文献

A novel fructosyltransferase (AoFT) capable of synthesizing sucrose 6-acetate (S6A) from sucrose and glucose 6-acetate has been purified to homogeneity from Aspergillus oryzae ZZ-01. Its molecular mass was ~50 kDa by SDS-PAGE; optimal activity was at 45 °C and it was stable from pH 4.5 to 7.5 with an optimum pH of 6. Mg2+, K+ (5 mM), propanol, toluene (50 %, v/v), Tween 20 or Triton X-100 (1 %, w/v) increased the transfructosylation activity by 20, 17, 17, 10, 25 and 20 %, respectively. An overall conversion of 32 % was achieved under optimal conditions over 24 h. This is the first report that the purified and characterized the fructosyltransferase from Aspergillus capable of synthesis of S6A from sucrose and glucose 6-acetate.  相似文献

Hamsters show a preference for Polycose, a mixture of starch-derived glucose polymers, that is as strong as their preference for sucrose. However, in the hamster, taste aversions to Polycose may be less easily acquired than taste aversions to sucrose and the qualitative aspects of Polycose are unknown in this species. In order to examine the taste of Polycose in the hamster, we utilized a taste-aversion protocol with two conditioning trials. Animals were trained to avoid one of three different conditioning stimuli: 50 mM sucrose, 100 mM Polycose and a mixture of 50 mM sucrose with 100 mM Polycose. Control animals were conditioned with deionized water. After the second conditioning trial, generalization testing began for the three conditioning stimuli plus 3 mM citric acid, 300 mM KCI and 30 mM NaCl. The results showed that aversions to Polycose, sucrose or the Polycose/sucrose mixture cross- generalized, demonstrating that Polycose and sucrose share a common taste percept in the hamster. None of the aversions generalized to NaCl, citric acid or KCI. In addition, comparisons among the patterns of taste generalizations indicated that the tastes of Polycose and sucrose also had distinct qualitative components. Finally, although the taste of 100 mM Polycose was more salient than the taste of 50 mM sucrose, the taste of sucrose could still be detected in a mixture with Polycose.   相似文献

Natural products are gaining tremendous importance in pharmaceutical industry and attention has been focused on the applications of in vitro technologies to enhance yield and productivity of such products. In this study, we investigated the accumulation of biomass and antioxidant secondary metabolites in response to different carbohydrate sources (sucrose, maltose, fructose and glucose) and sucrose concentrations (1, 3, 5, 7 and 9 %). Moreover, the effects of 3 % repeated sucrose feeding (day-12, -18 and -24) were also investigated. The results showed the superiority of disaccharides over monosaccharides for maximum biomass and secondary metabolites accumulation. Comparable profiles for maximum biomass were observed in response to sucrose and maltose and initial sucrose concentrations of 3 and 5 %. Maximum total phenolic and total flavonoid contents were displayed by cultures treated with sucrose and maltose; however, initial sucrose concentrations of 5 and 7 % were optimum for both classes of metabolites, respectively. Following 3 % extra sucrose feeding, cultures fed on day-24 (late-log phase) showed higher biomass, total phenolic and total flavonoid contents as compared to control cultures. Highest antioxidant activity was exhibited by maltose-treated cultures. Moreover, sucrose-treated cultures displayed positive correlation of antioxidant activity with total phenolics and total flavonoids production. This work describes the stimulatory role of disaccharides and sucrose feeding strategy for higher accumulation of phenolics and flavonoids, which could be potentially scaled up to bioreactor level for the bulk production of these metabolites in suspension cultures of A. absinthium.  相似文献

Collagen, the most abundant protein in mammals, is widely used for making biomaterials. Recently, organic solvents have been used to fabricate collagen-based biomaterials for biological applications. It is therefore necessary to understand the behavior of collagen in the presence of organic solvents at low (≤50 %, v/v) and high (≥90 %, v/v) concentrations. This study was conducted to examine how collagen reacts when exposed to low and high concentrations of ethanol, one of the solvents used to make collagen-based biomaterials. Solubility testing indicated that collagen remains in solution at low concentrations (≤50 %, v/v) of ethanol but precipitates (gel-like) thereafter, irrespective of the method of addition of ethanol (single shot or gradual addition); this behavior is different from that observed recently with acetonitrile. Collagen retains its triple helix in the presence of ethanol but becomes thermodynamically unstable, with substantially reduced melting temperature, with increasing concentration of ethanol. It was also found that the CD ellipticity at 222 nm, characteristic of the triple-helical structure, does not correlate with the thermal stability of collagen. Time-dependent experiments reveal that the collagen triple helix is kinetically stable in the presence of 0–40 % (v/v) ethanol at low temperature (5 °C) but highly unstable in the presence of ethanol at elevated temperature (~34 °C). These results indicate that when ethanol is used to process collagen-based biomaterials, such factors as temperature and duration should be done taking into account, to prevent extensive damage to the triple-helical structure of collagen .  相似文献

As chromium is widely used in many industries of which leather industries are the biggest consumers, wastes from tanneries pose a serious threat to the environment. Present research conducted greenhouse pot experiments on several plants using different solutions to find out their phytoextraction potential. Among the plants, Urtica dioica found to be very effective due to its higher uptake capacity for chromium. The analyses showed that with 500 mg/l solutions Cr concentration in roots was 38.9% higher (average 20 mg/kg) than plants fed with 200 mg/l solutions (phytostabilisation), while with 200 mg/l solutions more Cr was transported to leaves (phytoextraction), 10.52% higher (average 9.5 mg/kg) concentration than plants fed with 500 mg/l solutions, but also means that plants were less affected by toxicity. Zea mays showed high tolerance towards Cr with negligible concentration in leaves. Although average Cr concentration in Brassica napus is lower (25% and 21% for 200 mg/l and 500 mg/l solutions respectively) than Urtica dioica which still could account for phytoextraction, but due to its high vulnerability to insects its potentiality has downgraded. Among other elements, K being a component of the salt, played significant role for plants growth and survival—opening new avenue for phytoremediation of contaminated sites.  相似文献

The amounts of sugar solution consumed by Drosophila melanogaster flies were determined. Starved and desiccated flies of a wild type strain (QA) consume 7?9 × 10?2 λ of a 0.3 M sucrose solution per fly during the first hour and less later. They consume more of the 0.3 M sucrose solution than of the more diluted and the more concentrated solutions. In preference-aversion tests the flies discriminated between water and various sugar solutions, and between different sugar concentrations. Contrary to other fly species these flies did not prefer 0.05 M fructose over 0.05 M glucose. 0.3–0.5 M NaCl added to 0.1 M sucrose turned a preference over 0.01 sucrose into an aversion. A mutant, Lot-94, selected for its increased consumption of a 1 M NaCl solution was found to consume more of all test solutions. The amount of NaCl that had to be added to 0.1 M sucrose to turn the preference over 0.01 M sucrose by the mutant flies into aversion was not different from that found for the wild type flies.  相似文献

To improve micropropagation and acclimatization of turmeric (Curcuma longa L.), we evaluated the effects of media volume, plant density, macronutrient ion concentrations, cationic ratios [NH4 +]/[K+], and sucrose concentration. Multiplication was highest with low bud density. Yield of new plants was highest with high bud density, the most sucrose, the highest concentration of macronutrients, and the greatest volume of medium. However, maximum plant size required low-density, reduced sucrose and elimination of NH4 +. The largest plants grew quickest during greenhouse acclimatization when macronutrients were lowered to 20 mM. In a follow-up experiment, media volume was set at 40 mL with 5% sucrose with NH4 + reduced to 5 mM, and the effects of varying P, Ca, Mg, KNO3, and bud densities were assessed. The largest plants were produced at low density. More importantly, at high density the optimal concentrations of P, Ca, Mg, and KNO3 predicted plant size that was nearly equal to the maximum value from low-density cultures in the prior experiment. Growth of plants during greenhouse acclimatization was increased by modifications of in vitro medium with plants cultured with 3.32 mM P, 4.5 mM Mg, and 37 mM KNO3 predicted to grow most rapidly. The effect of starter fertilizer in the greenhouse mix was much less than the effects of P, Mg, and KNO3 in vitro. These results showed (1) optimal media formulae for different stages of micropropagation and (2) process-related factors such as plant density and media volume affected the optimal nutrient concentrations.  相似文献

Five yeast strains, Saccharomyces cerevisiae D8, M12, and S13; Hanseniaspora uvarum S6; and Issatchenkia orientalis KMBL5774, isolated from Korean grapes, were entrapped in Ca-alginate beads, which are non-toxic, simple to use, and economical. Ca-alginate beads containing yeast cells were soaked in protective solutions, such as skim milk, saccharides, polyols, and nitrogen compounds, before air-blast drying to improve the yeast survival rate and storage ability. The results showed that both entrapment in Ca-alginate beads and soaking in protective agents favorably affected the survival of all strains. The microenvironment formed by the beads and protective agents can protect the yeast cells from harsh environmental conditions, such as low water (below 10 %). All the yeast strains entrapped in Ca-alginate beads showed greater than 80 % survival and less than 11 % water content after air-blast drying at 37 °C for 5 h. In addition, air-blast dried cells of S. cerevisiae D8, M12, S13; H. uvarum S6; and I. orientalis KMBL5774 entrapped in 2 % Ca-alginate beads and soaked in protective agents (10 % skim milk containing 10 % sucrose, 10 % raffinose, 10 % trehalose, 10 % trehalose, and 10 % glucose, respectively) after air-blast drying at 37 °C for 5 h showed 90, 87, 92, 90, and 87 % viability, respectively. All dried entrapped yeast cells showed survival rates of at least 51 % after storage at 4 °C for 3 months.  相似文献

Effects of two different concentrations of arsenic (8.88 mg/l and 17.76 mg/l i.e., 25 and 50% of LC50 value) respectively and chromium (15.45 mg/l and 30.90 mg/l i.e., 25 and 50% of LC50 value respectively) were evaluated on digestive enzymes of Anabas testudineus for a period of 30 days under laboratory condition. Here As1 and As2, and Cr1 and Cr2 were referred to indicate the 25 and 50% of LC50 value of arsenic and chromium respectively. The result of different enzymes in different tissues was as follows: amylase and lipase activities showed highest in stomach, i.e., 466.81 and 283.33% respectively at 50% concentration of chromium; but moderate (272.78% at 25% of Cr and 264.65% at 50% of As) followed by lowest 219.07% at 25% As concentration. Protease activity at different concentrations of metals was in the order of As2 > As1 > Cr2 > Cr1. Protease activity showed highest in As2 or 50% concentration of As (397.92%) and lowest in 25% of Cr (316.39%). Lipase activity showed highest increment in intestine under As2 (409.23%) and lowest activity in Cr1 (137.84%). In intestine, the protease activity increased at As1 (288.99%) and the highest at As2 i.e., 367.39% compared to other concentrations. The significant changes (p > 0.05) in the enzyme activities in the different tissues like stomach and intestine in regard to different doses were investigated along with regression lines. So, it can be inferred that digestive enzyme activities of A. testudineus may be considered as the potential biomarkers for arsenic and chromium contamination in aquatic systems.  相似文献

A gene encoding Uninv, an invertase active in the neutral pH range, was isolated from a metagenomic library and the enzyme was biochemically characterized. The enzyme had an optimum pH of 6.5 and an optimum temperature of 50°C. It showed maximal ~37% sequence identity to all invertases identified so far. Uninv displayed typical Michaelis–Menten kinetics with substrate inhibition of sucrose, but it retained 50% activity at sucrose concentrations up to 1,000 mM. Uninv had no transfructosylating activity even at high sucrose concentrations. These properties will make this enzyme useful in the food industry.  相似文献

Growth of Spirulina sp. (MCRC-A0003), a cyanobacterium, was evaluated under different concentrations of carbon-dioxide (CO2) (4–50 %) in a closed glass photobioreactor. Although significant CO2 utilization by the cyanobacterial strain was observed up to 50 % concentration, complete utilization was observed only at 4, 10 and 20 % concentrations on 3rd, 6th and 8th day respectively. However, considerable reduction was witnessed in reactors containing 30–50 % CO2 only between 6th and 9th day. A corresponding increase in the biomass and primary metabolites like chlorophyll-a, carbohydrate and protein were observed. Biomass productivity of Spirulina in reactors sparged with 4, 10 and 20 % CO2 were 13.7, 43 and 44 % more than that in control reactor without CO2. While CO2 increased the levels of primary metabolites in the cyanobacterial cells, it was quite prominent in 10 % CO2 concentration with the chlorophyll-a, carbohydrate and protein contents were 64, 183 and 626 mg g?1 respectively. While 10 and 6.6 % increase were noticed in chlorophyll-a and protein, 17 % increase in carbohydrate levels was observed in Spirulina cells, which could be attributed to the conversion of CO2 to carbohydrate by the cyanobacterium.  相似文献
