A simple model is used to investigate the relationship between clone number and patch number for each of two cell types in one and two-dimensional arrays. Considering the two cell types separately, this relationship is shown to vary with the relative proportions of the two cell types. If both cell types are considered, the total number of patches in a one-dimensional array reaches a maximum when the two cell types are present in equal proportions. Over the range considered for a two-dimensional array, the total number of patches is at a minimum when the two cell types are present in equal proportions because aggregation of patches is then maximal. The significance of these results is discussed in the light of previous interpretations of work with mouse aggregation chimaeras.  相似文献

Chimaeras between closely related and distant mammalian species were obtained during the last decade using the aggregation and injection methods. In the chimaeras between the closely related species Mus musculus--M. caroli, the cell clones of both parental species are equally represented; such chimaeras are fertile and their development does not differ from that of intraspecific chimaeras. But in the chimaeras between distant species mouse--rat or sheep--goat a selective predominance of cells with one genotype over those with another was noted. This leads to the total elimination of cell populations with one genotype, rat in particular in the mouse--rat chimaeras, or, possibly, to the sterility of the sheep--goat chimaeras.  相似文献

Mutant mice of Sl/Sld genotype are deficient in melanocytes, erythrocytes, mast cells and germ cells. Deficiency of melanocytes, erythrocytes and mast cells is not attributable to an intrinsic defect in their precursor cells but to a defect in the tissue environment that is necessary for migration, proliferation and/or differentiation. We investigated the mechanism of germ cell deficiency in male Sl/Sld mice by producing aggregation chimaeras from Sl/Sld and +/+ embryos. Chimaeric mice with apparent white stripes were obtained. Two of four such chimaeras were fertile and the phenotypes of resulting progenies showed that some Sl/Sld germ cells had differentiated into functioning sperms in the testis of the chimaeras. In cross sections of the testes of chimaeras, both differentiated and nondifferentiated tubules were observed. However, the proportions of type A spermatogonia to Sertoli cells in both types of tubules were comparable to the values observed in differentiated tubules of normal +/+ mice. We reconstructed the whole length of four tubules from serial sections. Differentiated and nondifferentiated segments alternated in a single tubule. The shortest differentiated segment contained about 180 Sertoli cells and the shortest nondifferentiated segment about 150 Sertoli cells. These results suggest that Sertoli cells of either Sl/Sld or +/+ genotype make discrete patches and that differentiation of type A spermatogonia does not occur in patches of Sl/Sld Sertoli cells.  相似文献

Spontaneous diploid-triploid chimaeras occur sporadically in various mammalian species including man, but so far have never been produced experimentally. In order to get a deeper insight into the developmental consequences of this anomaly, we have developed two procedures that enabled for the first time to produce routinely diploid-triploid embryos, foetuses, and animals in the mouse. These procedures are: (1) aggregation of cleaving diploid embryos with triploid embryos produced by suppression of the second polar body in zygotes, and (2) fusion of a haploid karyoplast with one blastomere of the two-cell diploid embryos. The first procedure yielded 23 living and 6 dead postimplantation embryos and foetuses (age: 8th-19th day) out of which 22 were chimaeric. In addition, three chimaeric neonates reached adulthood. Two animals were fertile, and one--an overt chimaera--was an infertile male. The rate of postimplantation development of aggregation chimaeras was normal or only slightly retarded, and with one exception the foetuses were morphologically normal. Generally, the highest contribution of the 3n component in extra-embryonic structures was noted in the yolk sac, and usually it was higher than its contribution to the organs of the body. Chimaerism was most often noted in the liver, the heart, the intestine, and the lungs. Participation of triploid cells to all tissues studied, both in the body and in extra-embryonic structures, appeared to decrease slightly as development progressed. The second procedure yielded 10 foetuses and 6 adults. Three foetuses were chimaeric. Six fertile adults were probably non-chimaeras: the triploid component was absent in the coat and in the blood.  相似文献

The retinal pigment epithelium (PE), with pigmentation as a cell-autonomous marker, was analyzed in three types of mice: congenic pigmented----albino chimaeras, X-inactivation mosaics (Cattanach's translocation), and mosaics homozygous for the pink-eyed unstable mutation, which contain rare fully pigmented cells. In 10 chimaeric and 34 X-inactivation eyes, the proportionate mix in the right and left eyes of an individual animal was similar, the mix was approximately constant in all parts of a given eye, average patch size was larger toward the periphery of the PE, and peripheral patches tended to be elongated in the radial dimension. In all 44 whole mounts from pink-eyed unstable mutants, patches of 1-12 pigmented cells, each representing a single clone, were scattered throughout the PE; they tended to be larger with increasing distance from the optic nerve head. The collective data are consistent with significant cell mixing prior to specification of the two eye fields, during early organ-forming stages, and during later development of the PE. The tendency of peripheral patches to orient radially reflects the edge-biased pattern of cell proliferation in the PE. Cell mixing appears to be more prominent posteriorly in the PE sheet; growth proceeds anteriorly for more generations.  相似文献

Spermatozoa of males from the inbred mouse strains, KE (albino) and CBA (agouti), are distinguishable by head shape and differ in quality: CBA spermatozoa show a lower percentage of abnormal heads and higher efficiency of fertilization. Aggregation chimaeras were produced to investigate whether these differences are intrinsic or extrinsic to spermatogenic cells. Among 24 overt chimaeras, 14 were males: 1 was sterile, 8 produced either KE or CBA spermatozoa, as recognized by shape and by progeny testing, and 5 (21%) were germ-line chimaeras. Both the level of abnormal sperm heads and the efficiency of fertilization of CBA and KE spermatozoa produced by chimaeric males (except two subfertile ones) were within the range characteristic for the respective strain. All 5 germ-line chimaeras, irrespective of their coat colour composition, produced about 98% KE and only 2% CBA spermatozoa, which indicated strong selection against CBA germ cells. However, mature CBA spermatozoa showed high competitive ability, because the proportion of agouti progeny (from the CBA component) sired by those males was significantly higher than the proportion of CBA spermatozoa, estimated from vaginal plug preparations after every mating. The fact that this difference was particularly striking for one chimaera with a preponderance of CBA somatic component may suggest some influences extrinsic to spermatogenic cells. We conclude from this study that sperm head shape, the level of sperm abnormalities and fertilizing capacity are determined largely autonomously by genes acting in the germ cells. The internal environment created by foreign somatic cells exerts only minor modifications, unless there has been deterioration beyond the range of 'normality'.  相似文献

Blastocysts obtained from mice differing in pigmentation (albino versus pigmented) and the isoforms of glucose phosphate isomerase (GPI 1A versus 1B) were electrofused and those containing a single chimaeric inner cell mass (ICM) were transferred to the uterus of pseudopregnant recipients. The pups were recovered on the 20(th) day by Caesarian section and fostered by females that had littered on the previous night or 24 h earlier. Altogether nine adult animals and two pups, which died soon after delivery, were available for GPI analysis. Between 9 and 13 organs/tissues were examined and the relative contribution of the GPI 1A and 1B isoforms was estimated using an electrophoretic GPI assay. Eight adult animals were overtly chimaeric and one was chimaeric in some internal tissues only. Eight mice were males: seven were fertile, one was infertile. The ninth adult mouse was a hermaphrodite. The fertile animals produced sperm of one genotype only, i.e. derived either from the albino or from the pigmented component. This is the first report showing that adult chimaeras can be produced from two combined blastocysts, provided that fusion of the adhering trophectoderm cells is first induced and the orientation of blastocysts enables the two ICMs to integrate into a single ICM. Our results suggest that in the preimplantation blastocyst, the organisation of the ICM remains labile thus making it possible for the fused blastocysts to establish new embryonic organisation and to develop into a single organism.  相似文献

In the trishanku (triA ) mutant of the social amoeba Dictyostelium discoideum, aggregates are smaller than usual and the spore mass is located mid-way up the stalk, not at the apex. We have monitored aggregate territory size, spore allocation and fruiting body morphology in chimaeric groups of (quasi-wild-type) Ax2 and triA cells. Developmental canalisation breaks down in chimaeras and leads to an increase in phenotypic variation. A minority of triA cells causes largely Ax2 aggregation streams to break up; the effect is not due to the counting factor. Most chimaeric fruiting bodies resemble those of Ax2 or triA . Others are double-deckers with a single stalk and two spore masses, one each at the terminus and midway along the stalk. The relative number of spores belonging to the two genotypes depends both on the mixing ratio and on the fruiting body morphology. In double-deckers formed from 1:1 chimaeras, the upper spore mass has more Ax2 spores, and the lower spore mass more triA spores, than expected. Thus, the traits under study depend partly on the cells’ own genotype and partly on the phenotypes, and so genotypes, of other cells: they are both autonomous and non-autonomous. These findings strengthen the parallels between multicellular development and behaviour in social groups. An analysis of aphakia (ak) gene expression in 16 day ak/ak C/C----+/+ c/c chimaeric embryos has shown, that ak gene, acting in developing lens, blocks lens cell differentiation and disturbs the formation by these cells of the extracellular matrix composing the lens capsule material. The dependence of capsule structure in chimaeras on the genotype of underlying cells indicates that lens cells are responsible for the formation of lens capsule.  相似文献

The aim of this study was to estimate how many blastomeres of the 4-cell mouse embryo contribute cells to the embryo proper and finally to the animal. To this end, 4-cell embryos of pigmented and albino genotypes were disaggregated and single blastomeres (henceforth called '1/4' or 'quarter' blastomeres) were reaggregated in the following combinations: one 'pigmented' blastomere + three 'albino' blastomeres or vice versa (henceforth called '1+3') and two pigmented blastomeres + two albino blastomeres (henceforth called '2+2'). The aggregations were cultured in vitro and transferred as blastocysts either to the oviduct or uterus of pseudopregnant females. Recipients were allowed to litter naturally, or the foetuses were removed by Caesarian section and raised by lactating foster mothers. Chimaerism was assessed on the basis of coat (adults) or eye pigmentation (dead neonates). Among 28 '1+3' animals, there were 13 chimaeric and 15 non-chimaeric individuals. The pigmentation of non-chimaeras was always concordant with the genotype of the three 1/4 blastomeres and not with the genotype of the single blastomere in the given aggregation. These results make rather unlikely the possibility that the mouse is built of cells derived either from one or all four 1/4 blastomeres. Both two remaining options (2 or 3 1/4 blastomeres) are conceivable but the observed ratio of chimaeras to non-chimaeras among '1+3' animals (13:15) fits better the assumption of two 1/4 blastomeres contributing cells to the animal body. This assumption finds additional support in the observation that among '2+2' animals there were non-chimaeras (5 out of 7) and these would not have been expected should three 1/4 blastomeres contribute cells to the mouse body.  相似文献

Embryonic stem (ES) cells have revolutionised our understanding of animal physiology. Analysis of chimaeric mice generated from these cells allows us to study the role of genes in development and function of the nervous system. The NMDA receptor, one of the two major ionotropic glutamate receptors, has been proposed to play fundamental roles in the survival, migration, differentiation, and activity-dependent maturation of neural cells. The NMDA receptor subunit 1 (NR1) gene is indispensable for receptor function, and knock-out mice die at birth, inhibiting the study of glutamate signalling in postnatal neurons. Homozygous NR1-/- ES cells were derived from matings of heterozygous mice under feeder-free conditions. Chimaeras were made by incorporating these ES cells into wild-type blastocysts and by the classical aggregation of morulae between wild-type and NR1-/- embryos. The resulting chimaeras survive and develop normally. NR1-/- neurons, identified by their lacZ label, were analysed and quantified in developing and adult brains with varying knock-out contributions in every single brain region. Specifically, postnatal ontogenesis of cerebellum and hippocampus was normal. Accordingly, in chimaeric mice, NMDA receptor-initiated signals are not required for the migration, differentiation, and survival of most types of neurons in the central nervous system, in a cell-autonomous way.  相似文献

Anomalous giant granules of beige (bg) mice have been used as a cell marker in the study of cell lineage of mast cells. Similar granules are known to exist in other tissues including kidney proximal tubules and liver parenchymal cells. In the present study, these granules were found to give yellow or orange autofluorescence when the tissue had been fixed with formaldehyde and embedded in paraffin. Thus, the granules can be used as a cell marker that can be visualized in serial paraffin sections without any specific histochemical staining. Chimaeric mice were produced by aggregation of 8-cell-stage embryos of beige (C57BL/6J-bgJ/bgJ) and A/J strains. The chimaeric liver showed beige cell patches with complicated shapes, although the patches frequently conformed to the shape of parenchymal cell cord or plate structures. In chimaeric kidney, beige cells formed coherent patches in the proximal tubules. The tubules were found to contain more than one clone. The patches frequently had long extended shapes suggesting growth of the clone along the tubule axis. Three-dimensional image reconstruction from the serial paraffin sections was carried out with the aid of a computer-assisted image analysis system, resulting in a clearer image of the patch shape.  相似文献

In a previous study of mouse tetraploid<-->diploid chimaeric blastocysts, tetraploid cells were found to be more abundant in the trophectoderm than the inner cell mass (ICM) and more abundant in the mural trophectoderm than the polar trophectoderm. This non-random allocation of tetraploid cells to different regions of the chimaeric blastocyst may contribute to the restricted tissue distribution seen in post-implantation stage tetraploid<-->diploid chimaeras. However, the tetraploid and diploid embryos that were aggregated together differed in several respects: the tetraploid embryos had fewer cells and these cells were bigger and differed in ploidy. Each of these factors might underlie a non-random allocation of tetraploid cells to the chimaeric blastocyst. A combination of micromanipulation and electrofusion was used to produce two series of chimaeras that distinguished between the effects of cell size and ploidy on the allocation of cells to different tissues in chimaeric blastocysts. When aggregated cells differed in cell size but not ploidy, the derivatives of the larger cell contributed significantly more to the mural trophectoderm and polar trophectoderm than the ICM. When aggregated cells differed in ploidy but not cell size, the tetraploid cells contributed significantly more to the mural trophectoderm than the ICM. In both experiments the contributions to the polar trophectoderm tended to be intermediate between those of the mural trophectoderm and ICM. These experiments show that both the larger size and increased ploidy of tetraploid cells could have contributed to the non-random cell distribution that was observed in a previous study of tetraploid<-->diploid chimaeric blastocysts.  相似文献

Bovine inner cell masses (ICMs) were isolated by immunosurgery at day 8 or 10, or by dissection at day 14, and combined with day-5.5 morulae. Aggregation was obtained between 89%, 62%, and 0% of the day-5:day-8, day-5:day-10, day-5:day-14 composites, respectively. Chromosome analysis of composites, respectively. Chromosome analysis of composites potentially carrying the 1/29 translocation as a chromosome marker and temporarily transferred to the bovine uterus for 8 days showed that chimaeric day-14 embryos can be obtained from day-5:day-8 aggregation. The definitive transfer of eight day-5:day-8 and 11 day-5:day-10 composites resulted in the birth of six and four calves, respectively; five of the six, but none of the four, were chimaeric. The five chimaeras showed mostly the ICM phenotype. The morphological differences between ICMs at different stages of development were examined by electron microscopy and related to the success of the aggregation technique. It is concluded that bovine embryonic cells can regulate for at least 3 days difference in development but not 5 days even though aggregation is still possible.  相似文献

Neural crest-derived melanoblasts are the progenitors of melanocytes, the pigment cells of the skin, hair and choroid. Previous studies of adult chimaeric mice carrying different coat colour markers have suggested that the total melanocyte population is derived from a small number of melanoblast progenitors, each of which generates a discrete unilateral transverse band of colour. This work also suggested minimal mixing of cells between clones. We have used two complementary approaches to assess the behaviour of migrating clones of melanoblasts directly in the developing embryo. First, we made aggregation chimaeras between transgenic Dct-lacZ and non-transgenic embryos, in which lacZ is a marker for melanoblasts. Second, we generated transgenic mice carrying a modified lacZ reporter construct containing a 289 base pair duplication (laacZ) under the control of the Dct promoter. The laacZ transgene is normally inactive, but reverts to wild-type lacZ at low frequency, labelling a cell and all of its progeny at random. Mosaic embryos containing labelled melanoblast clones were generated. In contrast to previous data, chimaeric and mosaic embryonic melanoblast patterns suggest that: (1) there is a large number of melanoblast progenitors; (2) there is a pool of melanoblasts in the cervical region; (3) different cell dispersion mechanisms may operate in the head and trunk regions; and (4) there is extensive axial mixing between clones.  相似文献

Mouse chimaeras were produced by aggregating eight-cell embryos from two different F2 matings, abbreviated to AF2 and BF2 respectively: (C57BL/ OIa.AKR-Gpi-1s a, c/Ws female × BALB/c male)F2 and (C57BL/Ws female × CBA/Ca male)F2. Quantitative electrophoresis of glucose phosphate isomerase (GPI-1) was used to estimate the proportions of the two cell populations in different tissues of the 12 day chimàeric conceptuses, with the % GPI-1A indicating the percentage of cells derived from the AF2 embryos. The % GPI-1A was found to be highly positively correlated within the primitive ectoderm lineage (between the fetus, amnion and yolk sac mesoderm) and within the primitive endoderm lineage (between the yolk sac endoderm and the parietal endoderm) but no correlation (either positive or negative) was seen between the two lineages. This confirms the results of a previous,study of chimaeras made between partially congenic strains and suggests the original conclusions have general validity. The % GPI-1A in the placenta was corrected for the expected contribution of maternal GPI-1, based on control experiments involving transfer of homozygous Gpi-1s b /Gpi-1s b embryos to the uteri of Gpi-1s a /Gpi-1s a pseudopregnant females. The corrected % GPI- lA in the placenta was positively correlated with that in each of the three primitive ectoderm derivatives. This suggests either (1) exchange of cells between the polar trophectoderm and the underlying part of the inner cell mass that forms the primitive ectoderm or (2) cells are incompletely mixed in the chimaeric blastocyst and patches of AF2 and BF2 cells straddle the boundary between the polar trophectoderm and the underlying primitive ectoderm. The second explanation does not imply the existence of shared developmental lineages between trophectoderm and primitive ectoderm in non-chimaeric embryos. Unlike that of any other tissue, the distribution of placental GPI-1A was U-shaped; in 17/28 placenta samples the proportion of the minor component was 10% or less. This suggests that the placental trophoblast is derived from a small number of coherenct clones of polar trophectoderm cells (either a small number of polar trophectoderm cells or a larger number if the two cell populations are not finely intermingled). Thus, although as a population the placentas of chimaeric conceptuses are balanced with respect to the % GPI-1A (mean close to 50%), individually most placentas are extremely unbalanced in their chimaeric composition (< 10% or > 90% GPI-IA). This non-random composition of the chimaeric placentas is in contrast to the widely held assumption that the distribution of cells in chimaeric conceptuses is normally random. Correspondence to: J.D. Analysis of aphakia (ak) gene expression in ak/ak C/C in equilibrium +/+ c/c experimental chimaeras has shown that the ak gene acts in the lens rudiment cells blocking it differentiation. In the lens of 12 day old ak/ak C/C in equilibrium +/+ c/c chimaeric embryos undifferentiated ak/ak cells were present among the normally differentiating fibres. In 14 and 18 day old chimaeric embryos and 20 day old chimaeric mice ak/ak cells are located under the lens epithelium and the capsule of posterior lens half. In the locations of ak/ak cells on the posterior lens surface capsule breaks resulted in the extrusion of lens material into the secondary eye cavity. In all studied chimaeric embryos the lens structure is more similar to that in the normal embryos, than in ak/ak embryos. This suggests that in the developing chimaeric lens ak/ak cells are sorted out as the development proceeds. The proliferation rate of +/+ cells appears to be higher than that of ak/ak cells.  相似文献

采用优势规则和随机规则为基础的两种尺度分析方法,对分类的TM数据进行了尺度变换分析。结果表明,随着尺度(粒度)增加,优势规则处理法使景观中优势类型的面积增加,非优势类型的面积减少,随机规则处理法使各景观类型的面积基本上保持不变,随尺度变大,整个景观和多数类型的最大斑块面积指标增加;最小斑块面积等于尺度大小的平方;平均斑块面积都增加;斑块数迅速减少,在优势规则系列中,多样性指标减小,而在随机规则处理中,基本没有变化,聚集度随尺度的增大而减小,但测量尺度固定的情况下,随图分辨率的提高而增大,随尺度的增加,Moran’s Ⅰ指标减小,景观类型在空间上趋于随机分布,但是测量尺度固定的情况下,随图分辨率的提高而增大,景观类型在空间上趋于聚集分布。  相似文献

Composite sheep embryos (N = 110) were produced by aggregation of blastomeres from 2-, 4- or 8-cell embryos. Each composite embryo consisted of equal numbers of blastomeres from 2-8 parent embryos, the total cell number ranging from one quarter of the normal cell number to 8 times the normal cell number. The embryos were embedded in agar and transferred to ligated sheep oviducts to allow development up to the early blastocyst stage. Of the 101 embryos subsequently recovered, 77 had formed normally organized blastocysts and 74 of these were transferred to 51 recipients. Thirty-eight recipients went to full term, producing a total of 53 lambs. Of the 48 lambs which survived to be blood typed at 60 days of age, 36 were judged to be chimaeric on the basis of their blood type and/or on the basis of external features. The proportion of chimaeras was larger amongst the lambs produced from composite embryos of the normal number of cells or more (25/26) than amongst lambs produced from composite embryos of less than the normal cell number (11/22).  相似文献
