Lymphomyeloid organs of two common species of Antarctic fish, Trematomus nicolai and Chionodraco hamatus, were studied with the aim of analysing some morphological aspects of these organs in relation to adaptation to low environmental temperature. The thymuses of T. nicolai and C. hamatus were flattened, incompletely lobated, with numerous Hassall-like bodies, which were mainly located in the central part of the organ in C. hamatus. In T. nicolai, thymocytes, erythroid and reticular epithelial cells filled the organ. In C. hamatus, the thymocytes intermingled with reticular epithelial cells were often close to groups of melano-macrophages. In both species, the thymus did not show distinct compartmentalisation; however, the thymocytes had significantly different sizes in the outer and inner portions of the thymus. The head kidney of both species was completely filled by haematopoietic tissue, highly vascularised and mainly lymphopoietic in T. nicolai, while both erythropoietic and lymphopoietic in C. hamatus. The spleen appeared mainly erythropoietic in T. nicolai and mainly lymphopoietic in C. hamatus. Solitary melano-macrophages in T. nicolai were close to numerous small vascular ellipsoids where erythroid and lymphoid cells were intermingled without the formation of red and white pulp areas. In C. hamatus, large lymphoid areas were organised around the capillaries. The possible adaptation of lymphoid organs to the low temperature of polar water is discussed. Summary The age composition of Chionodraco hamatus samples caught by fixed nets in Terra Nova Bay during the Austral summer 1987/88 was determined by examination of thin otolith sections. Individual ages were estimated by counting the number of annuli seen in the sections, since we postulated that annuli were laid down yearly. Fish were estimated to be 5–10 years old. Although our apparent ages were not confirmed by other independent estimates, our data seem to compare well with the values reported for other Antarctic fishes. Females in our samples were on average larger than males of the same age and grow somewhat faster, at least over much of the sampled size range.  相似文献

Artedidraconids (Plunderfishes) are small benthic notothenioid fishes of the Antarctic and South Georgia shelf and slope. The family Artedidraconidae is monophyletic; however, the relationships within the family have remained poorly explored until now, and based on a small sample of the genus Artedidraco. The present study focuses on the interrelationships among the artedidraconid genera and the phylogeny of the genus Artedidraco. 2,353 base pairs from 77 specimens were sequenced from the partial mitochondrial cytochrome oxidase I gene and cytochrome b gene, the partial mitochondrial control region and the partial nuclear rhodopsin retrogene. The genus Artedidraco is not monophyletic, confirming the preliminary relationships found by Derome et al. (Mol Phylogenet Evol 24:139–152, 2002): Pogonophryne, Dolloidraco and Histiodraco are well embedded within the genus Artedidraco. From Artedidraco skottsbergi and A. loennbergi to A. orianae and A. mirus, the tree shows that there is an increasing number of upper lateral line tubular scales and decreasing number of disc-shaped scales. There is also a trend toward a decrease in the number of epipleural ribs and an increase in number of pleural ribs along the tree.  相似文献

We studied the genome of an antarctic ice fish, Chionodraco hamatus, in order to detect highly repetitive DNAs that may play a role in heterochromatinization processes and sex chromosome differentiation. We used two different experimental approaches. Hybridization of a Bkm probe to genomic DNA showed slight differences between the two sexes. Using restriction enzymes, a Bgl II satellite (pIF) was isolated. In situ hybridization revealed a preferential localization of pIF on the centromeres and the telomeres of most chromosomes, as well as an interstitial band on the long arms of the neo-Y sex chromosome, where probably the hypothetical fusion took place. Dot-blot experiments showed that pIF is still present in species belonging to different families of the same suborder. Though preliminary, our results suggest a conservative nature of this DNA which might have played a definite functional role in the genome of these polar fishes.  相似文献

The white-blooded Antarctic icefish is the only known vertebrate lacking oxygen-transporting haemoglobins. Fish skin mucus, as the first line of defence against pathogens, can reflect fish welfare. In this study, we analysed the skin mucus proteome profiles of the two Antarctic fish species, the white-blooded Antarctic icefish, Chionodraco hamatus, and the red-blooded Antarctic fish, Notothenia coriiceps, unfolding the different proteins by liquid chromatography coupled with tandem mass spectrometry isobaric tags for relative and absolute quantitation (iTRAQ) technology. Of the 4444 totally identified proteins, 227 differentially expressed proteins (DEPs) were found in the comparison between C. hamatus and N. coriiceps, of which 121 were upregulated and 106 were downregulated in the icefish. In the Kyoto Encyclopedia of Genes and Genomes pathway annotation, we found two pathways “Legionellosis” and “Complement and coagulation cascades” were significantly enriched, among of which innate immune candidate proteins such as C3, CASP1, ASC, F3 and C9 were significantly upregulated, suggesting their important roles in C. hamatus immune system. Additionally, the DEP protein–protein interaction network analysis and “Response to stress” GO category provided candidate biomarkers for deep understanding of the distinct immune response of the two Antarctic fish underlying the cold adaptation.  相似文献

Amino acid sequences of α- and β-chains of human hemoglobin and of hemoglobins of coelacanth and 24 teleost fish species, including 11 antarctic and two temperate Notothenioidei, were analyzed using maximum parsimony. Trees were derived for the α- and β-chains separately and for tandemly arranged sequences, using the human and coelacanth sequences as outgroups in all analyses. The topologies of the trees of the α-and β-chains are highly congruent and indicate a specific pattern of gene duplications and gene expression of teleost hemoglobins which has not yet been investigated into more detail. The Notothenioid fish generally contain a single major hemoglobin and often a second minor component. The α- and β-chains of the major components form a monophyletic group in all investigated trees, with the nonantarctic Pseudaphritis as their sister taxon. The minor chains also are a monophyletic group and form an unresolved cluster with the major chains and the hemoglobins of tuna and red gurnard. The carbonic anhydrase (CA) family of zinc metalloenzymes includes many known isozymes that have different subcellular distributions. The study described here focuses on identification of the structural features that define low-temperature adaptation in a Chionodraco hamatus protein, both for the reaction center, at an atomic level, and for the tertiary structure of the protein. To this aim, an x-ray absorption near-edge spectroscopy/Minuit x-ray absorption near-edge spectroscopy analysis of the reaction center was undertaken for both a structurally characterized human CAII and CA of C. hamatus. Higher structural levels were analyzed by sequence comparison and homology modeling. To establish whether the structural insights acquired in fish CAs are general, theoretical models were generated by homology modeling for three temperate-climate-adapted fish CAs. The measured structural differences between the two proteins are discussed in terms of the differences in the electrostatic potential between human CAII and CA of C. hamatus. We conclude that modulation of the interaction between the catalytic water molecule and the zinc ion could depend on the effect of the electrostatic potential distribution.  相似文献

Clades that have undergone episodes of rapid cladogenesis are challenging from a phylogenetic point of view. They are generally characterised by short or missing internal branches in phylogenetic trees and by conflicting topologies among individual gene trees. This may be the case of the subfamily Trematominae, a group of marine teleosts of coastal Antarctic waters, which is considered to have passed through a period of rapid diversification. Despite much phylogenetic attention, the relationships among Trematominae species remain unclear. In contrast to previous studies that were mostly based on concatenated datasets of mitochondrial and/or single nuclear loci, we applied various single-locus and multilocus phylogenetic approaches to sequences from 11 loci (eight nuclear) and we also used several methods to assess the hypothesis of a radiation event in Trematominae evolution. Diversification rate analyses support the hypothesis of a period of rapid diversification during Trematominae history and only a few nodes in the hypothetical species tree were consistently resolved with various phylogenetic methods. We detected significant discrepancies among trees from individual genes of these species, most probably resulting from incomplete lineage sorting, suggesting that concatenation of loci is not the most appropriate way to investigate Trematominae species interrelationships. These data also provide information about the possible effects of historic climate changes on the diversification rate of this group of fish.  相似文献

Feeding habits of early life stages of the channichthyid Chionodraco hamatus were investigated on samples collected in the western Ross Sea in early summer of 1996 and 2004. The stomach content analysis was carried out on larval and postlarval specimens ranging from 14 to 39 mm SL. Unlike larvae and juveniles of other channichthyids, which elsewhere largely rely on early life stages of Antarctic krill or ice krill, no euphausiids were found in the stomach contents of C. hamatus, except for a single large individual of Thysanoessa macrura. In both years, dietary composition consisted almost exclusively of notothenioid fish larvae. Early larvae of Pleuragramma antarcticum overwhelmingly dominated the diet in terms of abundance, biomass and frequency of occurrence, accounting for 98.4% of the index of relative importance (IRI). Other fish larvae consumed occasionally in small amount were Trematomus lepidorhinus, T. scotti and C. hamatus itself. As a result, the feeding strategy of C. hamatus was considerably shifted toward specialization, relying on relatively few taxa of prey, each of them showing a high prey-specific abundance. Comparing both dietary composition and feeding strategy of C. hamatus in the two different sampling years, several differences were observed, probably due to different environmental conditions, mainly linked to a significant delay of ice retreat and formation of the Ross Sea polynya verified in the 2003–2004 summer season.  相似文献

Species that live in extreme conditions have specially adapted physiology and tissue/organ organisation. The adaptation of lymphoid organs to low temperatures in polar species could be an original field of study, indicating how the immune system works under extreme conditions. In fishes, the head kidney is a key organ for immunity and here the cytology of this organ is studied in two common Antarctic species: Trematomus bernacchii and Chionodraco hamatus. Ultrastructural analysis revealed heterogeneity of epithelial cells, with reticular cells, subcapsular- and perivascular-limiting cells. Differences in the size and morphology of epithelial cells were observed between the polar species and warm water species of fish. Intermingled with epithelial cell leucocytes, such as lymphocytes, thrombocytes and macrophages, had comparable morphology in both species, contrary to sharp differences observed in the morphology of erythrocytes and granulocytes. The functional adaptation of the head kidney to the low temperatures of polar water is discussed.  相似文献

We report the presence of Tc1 transposon-like sequences in the Antarctic ice-fish Chionodraco hamatus, belonging to the Notothenioidei. The complete DNA sequence of these transposon-like elements is reduced in length compared to other Tc1 transposons, but it appears to share significant structural similarities with them. It contains a degenerate open reading frame, whose inferred 264 amino acid sequence shares sequence similarity with the 'aspartic acid, aspartic acid (35) glutamic acid' family of transposases, particularly those from Caenorhabditis species (sp.) and Drosophila sp. Southern blot analysis and polymerase chain reaction amplification indicate that Tc1 transposon-like sequences are present in other notothenioid species, though their amount can vary in the different lineages.  相似文献

The teleost suborder Notothenioidei is restricted to the Southern Ocean and has been described as a species flock spanning the whole of it. Within the suborder, the subfamily Trematominae is important for coastal Antarctic ecosystems. The eleven Trematomus species occupy a large range of ecological niches. The genus is monophyletic if the genus Pagothenia (two additional species) and Cryothenia amphitreta, also nested within it, are included. Although the Trematominae have received much interest, the relationships among these fourteen species are still unclear. Several recent studies have tried to resolve these interrelationships; however no complete and clear picture has emerged, probably because of the use of a low number of insufficiently variable markers. The only common results places T. scotti as the sister-group of the rest of the subfamily and T. loennbergi close to T. lepidorhinus. We use here more variable markers. Four nuclear markers, two of which are new, and a mitochondrial marker for the biggest trematomine sampling ever gathered (14 species, 78 specimens). We found that several nuclear haplotypes are shared by several species (mostly in very closely related species). The haplotype patterns coupled with the cytogenetics of the subfamily suggest that a phenomenon of incomplete lineage sorting (ILS) is likely to be at play. Using a calibration linked to fossil evidence, we evaluate the relative ages of each clade within the Trematominae to assess the proximity of the speciation events to one another. The main trematomine diversification was recent and sudden.  相似文献

Summary The development of the thymus was examined in different stages of Harpagifer sp. from Signy Island (South Orkney Islands; 60°43S, 45°38W). The thymus was typical, both in position and structural development, of that observed in warmer-water teleosts. The infiltration of the thymic epithelia was not observed until 4 weeks post-hatch. Full development of the lymphoid organs was not achieved until the juvenile stage. Although an increased infiltration of the thymus, by sub-epithelial connective tissues and epithelial mucous cells, occurred in the juvenile and adult stages, there was no evidence of an advanced stage of thymic regression or involution in the adult Harpagifer. Thus a suppressive influence of the low temperature environment, on the onset and degree of thymic development and involution, was indicated in this species.The signy Island population of Harpagifer has been given the species name H. antarcticus (Prof. J.C. Chionodracine (Cnd) is a 22-residue peptide of the piscidin family expressed in the gills of the Chionodraco hamatus as protection from bacterial infections. Here, we report the effects of synthetic Cnd on both Psychrobacter sp. TAD1 and Escherichia coli bacteria, as well as membrane models. We found that Cnd perforates the inner and outer membranes of Psychrobacter sp. TAD1, making discrete pores that cause the cellular content to leak out. Membrane disruption studies using intrinsic and extrinsic fluorescence spectroscopy revealed that Cnd behaves similarly to other piscidins, with comparable membrane partition coefficients. Membrane accessibility assays and structural studies using NMR in detergent micelles show that Cnd adopts a canonical topology of antimicrobial helical peptides, with the hydrophobic face toward the lipid environment and the hydrophilic face toward the bulk solvent. The analysis of Cnd free energy of binding to vesicles with different lipid contents indicates a preference for charged phospholipids and a more marked binding to native E. coli extracts. Taken with previous studies on piscidin-like peptides, we conclude that Cnd first adsorbs to the membrane, and then forms pores together with membrane fragmentation. Since Cnd has only marginal hemolytic activity, it constitutes a good template for developing new antimicrobial agents.  相似文献

The anatomical distribution of atrial natriuretic peptide (ANP)-immunoreactive structures and the autoradiographic localization of ANP binding sites were studied in the brain of the Antarctic fish, Chionodraco hamatus. ANP-containing elements were colocated with ANP binding sites in the dorsal medial and lateral subdivisions of the telencephalon, prethalamic nuclear complex, and in the nucleus of the medial longitudinal fasciculus of the mesencephalon. However, mismatching was observed in other brain regions, particularly at mesencephalic and metencephalic levels. In the pituitary, ANP immunoreactivity occurred only in the pars distalis, whereas ANP binding sites were localized in the whole pituitary. In this paper we describe the occurrence of ANP immunoreactivity and ANP binding sites in the brain and pituitary of an Antarctic fish. In particular, in the cerebellum and pituitary of C. hamatus, ANP binding sites are distributed in corresponding brain regions of dipnoans, amphibians and mammals. The immunocytochemical and histoautoradiographic data suggest that ANP acts as neuromodulator in the brain of C. hamatus. Moreover, the presence of ANP-like substances in tanycytes lining the diencephalic ventricle suggests a chemosensorial role for such liquor-contacting cells and a possible modulatory effect of ANP on the osmoregulation of the cerebrospinal fluid. The unfertilized egg envelope of medaka ( Oryzias latipes ) consists of two major groups of subunits, ZI-1,2 (74–76 kDa) and ZI-3 (49kDa). During egg envelope hardening after egg activation, both subunit groups decreased in amount, new protein bands of 57–65, 110 and 125 kDa appeared and, finally, no bands were detectable on sodium dodecylsulfate-polyacrylamide gel electrophoresis. The 110 and 125 kDa bands are intermediates formed by polymerization of such subunit groups. In contrast, treatment with iodoacetamide, an inhibitor of polymerization, revealed that the 57–65 kDa intermediates originated from ZI-1,2 by limited hydrolysis. ZI-1,2 comprises at least three distinct proteins of quite similar structure with their N -termini undetectable by Edman degradation, while the 57–65 kDa intermediates also consist of at least three proteins with the same N -terminal amino acid sequence: DGKPSNPQQPQVPQYPSK-. This fact strongly suggests a participation of a protease in the conversion of ZI-1,2 into 57–65 kDa proteins. EDTA and 1,10-phenanthrolinium inhibited the conversion and both Ca2+ and Zn2+ recovered the inhibition. These results suggest that the assumed protease is a metalloprotease.  相似文献
