The degradation of diesel fuel, B20 blend and biodiesel in liquid cultures by a seven-member bacterial consortium was compared under conditions with full aeration or with limited aeration with nitrate added as main electron acceptor. Community dynamics was assessed employing real-time PCR and the ddCt method for relative quantification. Biodegradation rates increased with increasing biodiesel content, but were significantly reduced under conditions with nitrate. Despite large variations in biodegradation rates, magnitude changes in population numbers were typically observed only from zero to one order, regardless the type of fuel and electron acceptor. Only Comamonadaceae and Variovorax sp. distinctly preferred aerobic conditions, and during aerobic growth showed suppression as fuel contained more biodiesel. Thus, the consortium is relatively stable and most of the degraders can shift their metabolism from hydrocarbons to biodiesel. The stability of the consortium is of interest in the context of biodiesel-mediated biodegradation of petroleum hydrocarbons.  相似文献

Leaks and spillages during the extraction, transport and storage of petroleum and its derivatives may result in environmental contamination. Biodiesel is an alternative energy source that can contribute to a reduction in environmental pollution. The aim of the present work was to evaluate biodegradation of diesel, biodiesel, and a 20% biodiesel-diesel mixture in oxisols from southern Brazil, using two bioremediation strategies: natural attenuation and bioaugmentation/biostimulation. Fuel biodegradation was monitored over 60 days by dehydrogenase activity, CO2 evolution and gas chromatography. The bacterial inoculum employed for bioaugmentation/biostimulation consisted of Bacillus megaterium, Bacillus pumilus, Pseudomonas aeruginosa, and Stenotrophomonas maltophilia and PCR-DGGE using 16S RNAr primers showed that some members of this consortium survived in the soil after 60 days. The biodegradation of pure biodiesel was higher for bioaugmentation/biostimulation than for natural attenuation, suggesting that the addition of the microbial consortium, together with adjustment of the macronutrient ratio, increased biodiesel degradation. The results of dehydrogenase and respiratory activity, together with GC analysis, suggested that the presence of biodiesel may, by stimulating general microbial degradative metabolism, increase the biodegradation of petroleum diesel. The microbial community was altered by both treatments, with natural attenuation producing a lower diversity index than the amended soil. The bioaugmentation/biostimulation strategy was showed to have a high potential for cleaning up soils contaminated with diesel and biodiesel blends.  相似文献

Fast development of ionic liquids as gaining more and more attention valuable chemicals will undoubtedly lead to environmental pollution. New formulations and application of ionic liquids may result in contamination in the presence of hydrophobic compounds, such as petroleum mixtures. We hypothesize that in the presence of diesel fuel low-water-soluble ionic liquids may become more toxic to hydrocarbon-degrading microorganisms. In this study the influence of 1-alkoxymethyl-2-methyl-5-hydroxypyridinium chloride homologues (side-chain length from C3 to C18) on biodegradation of diesel fuel by a bacterial consortium was investigated. Whereas test performed for the consortium cultivated on disodium succinate showed that toxicity of the investigated ionic liquids decreased with increase in side-chain length, only higher homologues (C8–C18) caused a decrease in diesel fuel biodegradation. As a result of exposure to toxic compounds also modification in cell surface hydrophobicity was observed (MATH). Disulphine blue active substances method was employed to determine partitioning index of ionic liquids between water and diesel fuel phase, which varied from 1.1 to 51% for C3 and C18 homologues, respectively. We conclude that in the presence of hydrocarbons acting as a solvent, the increased bioavailability of hydrophobic homologues is responsible for the decrease in biodegradation efficiency of diesel fuel.  相似文献

In this study, the combustion characteristics and emissions of two different petroleum diesel fuels (No. 1 and No. 2) and biodiesel from soybean oil were compared. The tests were performed at steady state conditions in a four-cylinder turbocharged DI diesel engine at full load at 1400-rpm engine speed. The experimental results compared with No. 2 diesel fuel showed that biodiesel provided significant reductions in PM, CO, and unburned HC, the NO(x) increased by 11.2%. Biodiesel had a 13.8% increase in brake-specific fuel consumption due to its lower heating value. However, using No. 1 diesel fuel gave better emission results, NO(x) and brake-specific fuel consumption reduced by 16.1% and 1.2%, respectively. The values of the principal combustion characteristics of the biodiesel were obtained between two petroleum diesel fuels. The results indicated that biodiesel may be blended with No. 1 diesel fuel to be used without any modification on the engine.  相似文献

Abstract Successful stimulation of N2 fixation and petroleum hydrocarbon degradation in indigenous microbial consortia may decrease exogenous N requirements and reduce environmental impacts of bioremediation following petroleum pollution. This study explored the biodegradation of petroleum pollution by indigenous N2 fixing marine microbial consortia. Particulate organic carbon (POC) in the form of ground, sterile corn-slash (post-harvest leaves and stems) was added to diesel fuel amended coastal water samples to stimulate biodegradation of petroleum hydrocarbons by native microorganisms capable of supplying a portion of their own N. It was hypothesized that addition of POC to petroleum amended water samples from N-limited coastal waters would promote the growth of N2 fixing consortia and enhance biodegradation of petroleum. Manipulative experiments were conducted using samples from coastal waters (marinas and less polluted control site) to determine the effects of POC amendment on biodegradation of petroleum pollution by native microbial consortia. Structure and function of the microbial consortia were determined by measurement of N2 fixation (acetylene reduction), hydrocarbon biodegradation (14C hexadecane mineralization), bacterial biomass (AODC), number of hydrocarbon degrading bacteria (MPN), and bacterial productivity (3H-thymidine incorporation). Throughout this study there was a consistent enhancement of petroleum hydrocarbon degradation in response to the addition of POC. Stimulation of diesel fuel biodegradation following the addition of POC was likely attributable to increases in bacterial N2 fixation, diesel fuel bioavailability, bacterial biomass, and metabolic activity. Toxicity of the bulk phase water did not appear to be a factor affecting biodegradation of diesel fuel following POC addition. These results indicate that the addition of POC to diesel-fuel-polluted systems stimulated indigenous N2 fixing microbial consortia to degrade petroleum hydrocarbons. This paper describes results obtained from CI engine performance running on dual fuel mode at fixed engine speed and four loads, varying the mixing system and pilot fuel quality, associated with fuel composition and cetane number. The experiments were carried out on a power generation diesel engine at 1500 m above sea level, with simulated biogas (60% CH4–40% CO2) as primary fuel, and diesel and palm oil biodiesel as pilot fuels. Dual fuel engine performance using a naturally aspirated mixing system and diesel as pilot fuel was compared with engine performance attained with a supercharged mixing system and biodiesel as pilot fuel. For all loads evaluated, was possible to achieve full diesel substitution using biogas and biodiesel as power sources. Using the supercharged mixing system combined with biodiesel as pilot fuel, thermal efficiency and substitution of pilot fuel were increased, whereas methane and carbon monoxide emissions were reduced.  相似文献

【目的】实施近海柴油污染的生物治理。【方法】以柴油为唯一碳源,从深圳港口海域富集筛选柴油降解菌;采用复配、正交试验等方式构建混合菌群;通过单因素试验研究环境因素对菌群降解柴油的影响;使用气相色谱-氢火焰检测器(GC-FID)分析降解前后柴油各组分的变化;通过生理生化试验和16S rRNA基因序列分析对菌株进行鉴定。【结果】获得了16株柴油降解菌,7 d内对柴油的降解率最高达40.8%;选择菌株C1-8、C2-10、C3-13构建了混合菌群CQ1,投加量分别为0.5%、2.0%和1.0%,CQ1对柴油去除率比单菌提高了10%以上;CQ1的最适环境条件为:温度30 °C、pH 7.6、摇床转速220 r/min、柴油浓度20 g/L,优化后9 d内对柴油去除率达60%以上;GC-FID结果显示,菌群CQ1可降解大部分C11?C27的正构烷烃,对C21?C27的烷烃降解可达100%。经鉴定,菌株C1-8、C2-10和C3-13分别为微杆菌(Microbacterium sp.)、剑菌(Ensifer sp.)和变异棒杆菌(Corynebacterium variabile)。【结论】CQ1在近海柴油污染的生物修复中具有良好的应用前景。  相似文献

Water is the most common choice of absorption medium selected in many gasification systems. Because of poor solubility of tar in water, hydrophobic absorbents (diesel fuel, biodiesel fuel, vegetable oil, and engine oil) were studied on their absorption efficiency of biomass tar and compared with water. The results showed that only 31.8% of gravimetric tar was removed by the water scrubber, whereas the highest removal of gravimetric tar was obtained by a vegetable oil scrubber with a removal efficiency of 60.4%. When focusing on light PAH tar removal, the absorption efficiency can be ranked in the following order; diesel fuel > vegetable oil > biodiesel fuel > engine oil > water. On the other hand, an increase in gravimetric tar was observed for diesel fuel and biodiesel fuel scrubbers because of their easy evaporation. Therefore, the vegetable oil is recommended as the best absorbent to be used in gasification systems.  相似文献

The biodiesel processor was developed for the production of biodiesel from non-edible oil of jatropha and karanj. The newly developed biodiesel processor is suitable for farmers in village level biodiesel production. The biodiesel processor was capable of producing 15 kg biodiesel per batch in 1.5 h at reaction temperature of 60°C. The biodiesel was produced from raw jatropha and karanj oil, and its blends with diesel were tested for power generation in a 7.5-kVA diesel engine generator set. The fuel properties, namely, kinematic viscosity and specific gravity, were found within the limits of Bureau of Indian Standards specifications. The overall efficiency of the generator for 4,500 W loading condition of jatropha- and karanj-biodiesel-blended fuel were recorded in the range of 21–23% and 24–27%, respectively. The overall efficiency of the generator for 6,000 W loading conditions was improved for jatropha and karanj biodiesel blends and were found in the range of 31–33% and 33–39%, respectively. Biodiesel blends B80 and pure biodiesel of karanj produced more power, and maximum overall efficiency was recorded as compared with diesel fueled generator. The overall efficiency on jatropha-biodiesel-blended fuel were found less than the diesel-fueled generator. The biodiesel processor based on alkali-catalyzed transesterification process can be used for quality biodiesel production from edible and non-edible vegetable oils. This processor can be integrated with rural energy system for domestic and small-scale industrial unit for biodiesel production.  相似文献

Different bioremediation techniques (natural attenuation, biostimulation and bioaugmentation) in contaminated soils with two oily sludge concentrations (1.5% and 6.0%) in open and closed microcosms systems were assessed during 90 days. The results showed that the highest biodegradation rates were obtained in contaminated soils with 6% in closed microcosms. Addition of microbial consortium and nutrients in different concentrations demonstrated higher biodegradation rate of total petroleum hydrocarbons (TPH) than those of the natural attenuation treatment. Soils treated in closed microcosms showed highest removal rate (84.1 ± 0.9%) when contaminated at 6% and bacterial consortium and nutrients in low amounts were added. In open microcosms, the soil contaminated at 6% using biostimulation with the highest amounts of nutrients (C:N:P of 100:10:1) presented the highest degradation rate (78.7 ± 1.3%). These results demonstrate that the application of microbial consortium and nutrients favored biodegradation of TPH present in oily sludge, indicating their potential applications for treatment of the soils impacted with this important hazardous waste.  相似文献

The purpose of the present study was to investigate possible methods to enhance the rate of biodegradation of oil sludge from crude oil tank bottom, thus reducing the time usually required for bioremediation. Enhancement of biodegradation was achieved through bioaugmentation and biostimulation. About 10% and 20% sludge contaminated sterile and non-sterile soil samples were treated with bacterial consortium (BC), rhamnolipid biosurfactant (RL) and nitrogen, phosphorus and potassium (NPK) solution. Maximum n-alkane degradation occurred in the 10% sludge contaminated soil samples. The effects of treatment carried out with the non-sterile soil samples were more pronounced than in the sterile soils. Maximum degradation was achieved after the 56th day of treatment. n-Alkanes in the range of nC8-nC11 were degraded completely followed by nC12-nC21, nC22-nC31 and nC32-nC40 with percentage degradations of 100%, 83-98%, 80-85% and 57-73% respectively. Statistical analysis using analysis of variance and Duncan's multiple range test revealed that the level of amendments, incubation time and combination of amendments significantly influenced bacterial growth, protein concentration and surface tension at a 1% probability level. All tested additives BC, NPK and RL had significant positive effects on the bioremediation of n-alkane in petroleum sludge.  相似文献

Two different biodiesel fuels, obtained from waste cooking oils with different previous uses, were tested in a DI diesel commercial engine either pure or in 30% and 70% v/v blends with a reference diesel fuel. Tests were performed under a set of engine operating conditions corresponding to typical road conditions. Although the engine efficiency was not significantly affected, an increase in fuel consumption with the biodiesel concentration was observed. This increase was proportional to the decrease in the heating value. The main objective of the work was to study the effect of biodiesel blends on particulate emissions, measured in terms of mass, optical effect (smoke opacity) and size distributions. A sharp decrease was observed in both smoke and particulate matter emissions as the biodiesel concentration was increased. The mean particle size was also reduced with the biodiesel concentration, but no significant increases were found in the range of the smallest particles. No important differences in emissions were found between the two tested biodiesel fuels.  相似文献

The biodegradation of oil products in the environment is often limited by their low water solubility and dissolution rate. Rhamnolipids produced by Pseudomonas aeruginosa AT10 were investigated for their potential to enhance bioavailability and hence the biodegradation of crude oil by a microbial consortium in liquid medium. The characterization of the rhamnolipids produced by strain AT10 showed the effectiveness of emulsification of complex mixtures. The addition of rhamnolipids accelerates the biodegradation of total petroleum hydrocarbons from 32% to 61% at 10 days of incubation. Nevertheless, the enhancement of biosurfactant addition was more noticeable in the case of the group of isoprenoids from the aliphatic fraction and the alkylated polycyclic aromatic hydrocarbons (PHAS) from the aromatic fraction. The biodegradation of some targeted isoprenoids increased from 16% to 70% and for some alkylated PAHs from 9% to 44%.  相似文献

The biodegradation of rapeseed oil methyl ester (RME) in pure and in mixtures with diesel fuel was investigated. Higher ratio of diesel fuel in the mixture resulted in higher count of bacteria. Fungal growth was advanced by higher RME contents. The growth of microorganisms gained from soil was strongest in B 20 (20 vol.% biodiesel and 80 vol.% diesel fuel) mixtures followed by B 5 (5 vol.% biodiesel and 95 vol.% diesel fuel) mixtures and pure RME. The formation of free fatty acids (FFA) in the RME sample was measured according to DIN EN 14214. The content of FFA in inoculated RME samples rose from 0.08 mass% to 0.344 mass% at the beginning. The oxidation stability of inoculated samples of B 20, B 5 and pure RME decreased faster than the oxidation stability of blank samples. An optical evaluation showed the formation of turbidity. Partly, the formation of sediment was observed, especially in B 20 and B 5 samples.  相似文献

The biodegradation of No. 2 diesel fuel under anaerobic conditions was investigated using sediments collected from wetlands of Barataria-Terrebonne estuary in Louisiana. The results indicated enhanced biodegradation of diesel fuel under sulfate-reducing, nitrate-reducing, methanogenic, and mixed electron acceptor conditions. However, the rate of diesel degradation was the highest under mixed electron acceptor conditions followed in order by sulfate-reducing, methanogenic, and nitrate-reducing conditions. Under mixed electron acceptor condition, 99% removal of diesel fuel was achieved within 510 days, while under sulfate-reducing condition 62% degradation of diesel fuel was observed for the same period. Diesel fuel was also degraded to a smaller extent in the culture condition where electron acceptors were not supplemented (natural attenuation condition). This study showed evidence for enhanced diesel fuel metabolism in a mixed microbial population system similar to any contaminated field site, where a heterogeneous microbial population exists.  相似文献

We studied the spray characteristics of inedible oil using experimental and simulation methods. Spray penetration, spray cone angle and spray tip speed were measured at different biodiesel ratios in a constant volume vessel with wide visualization and high back pressure, using a high-speed camera. The characteristics of biodiesel spray were simulated under the same conditions using Star-CD software. The experimental results showed that, as the ratio of biodiesel in the blends increased, spray penetration and spray speed increased, but the spray cone angle decreased. Throughout the spray injection period, the region at 0.05–0.475S (spray tip penetration) was a key area affecting spray cone angle. From 0.8 ms after injection, the spray penetration deviation ratios started to increase with increasing biodiesel blend ratios. Simulation results showed similar macroscopic spray characteristics to the experimental results for jatropha oil. The results also showed that the Sauter mean diameter of blend fuels was greater than that of diesel, and spray was more concentrated, due to the higher viscosity and surface tension of the biodiesel, compared with conventional diesel fuel. The macroscopic and microscopic spray properties of blended fuels containing 5%, 10% and 20% biodiesel were similar to diesel.  相似文献

A microbial consortium which rapidly mineralized the environmentally persistent pollutant benzo[a]pyrene was recovered from soil. The consortium cometabolically converted [7-(14)C]benzo[a]pyrene to (14)CO(2) when it was grown on diesel fuel, and the extent of benzo[a]pyrene mineralization was dependent on both diesel fuel and benzo[a]pyrene concentrations. Addition of diesel fuel at concentrations ranging from 0.007 to 0.2% (wt/vol) stimulated the mineralization of 10 mg of benzo[a]pyrene per liter 33 to 65% during a 2-week incubation period. When the benzo[a]pyrene concentration was 10 to 100 mg liter(-1) and the diesel fuel concentration was 0.1% (wt/vol), an inoculum containing 1 mg of cell protein per liter (small inoculum) resulted in mineralization of up to 17.2 mg of benzo[a]pyrene per liter in 16 days. This corresponded to 35% of the added radiolabel when the concentration of benzo[a]pyrene was 50 mg liter(-1). A radiocarbon mass balance analysis recovered 25% of the added benzo[a]pyrene solubilized in the culture suspension prior to mineralization. Populations growing on diesel fuel most likely promoted emulsification of benzo[a]pyrene through the production of surface-active compounds. The consortium was also analyzed by PCR-denaturing gradient gel electrophoresis of 16S rRNA gene fragments, and 12 dominant bands, representing different sequence types, were detected during a 19-day incubation period. The onset of benzo[a]pyrene mineralization was compared to changes in the consortium community structure and was found to correlate with the emergence of at least four sequence types. DNA from 10 sequence types were successfully purified and sequenced, and that data revealed that eight of the consortium members were related to the class Proteobacteria but that the consortium also included members which were related to the genera Mycobacterium and Sphingobacterium.  相似文献

In this study, the ability of Lysinibacillus sphaericus to degrade aromatic hydrocarbons as well as complex hydrocarbon mixtures, such as diesel oil and oily sludge, was evaluated. L. sphaericus was able to grow when toluene, naphthalene, or phenanthrene were used as a sole carbon source in minimal salt medium. Removal efficiencies of up to 95% were found for C10-C28 hydrocarbons in the biodegradation assays of diesel oil. The biodegradation of oily sludge was evaluated in landfarming-like experiments in the open air and in completely covered containers in the field. After 50 days of treatment, the removal efficiency of total petroleum hydrocarbons in open-air and closed assays was of 84.1% and 60.1%, respectively. Furthermore, L. sphaericus was able to degrade volatile hydrocarbons (benzene, toluene, ethylbenzene, and phenol) in the headspace of closed containers, preventing the emission of these compounds to the atmosphere. L. sphaericus was herein proposed as a promising candidate to be used in bioremediation strategies of petroleum hydrocarbons.  相似文献

This field study investigated the colonization process of soil contaminated with different petroleum products (petrol, diesel fuel, spent engine oil; dose: 6000 mg of fuel·kg?1 dry mass [d.m.] of soil) by epigeic and edaphic invertebrates during the progress of natural bioremediation and bioremediation enhanced using selected microorganisms (ZB-01 biopreparation). Epigeic fauna was captured using pitfall traps. Occurrence of edaphic fauna in soil samples as well as total petroleum hydrocarbon contents (TPH) were also investigated. Results showed that inoculation with ZB-01 biocenosis allowed the degradation of petroleum derivatives in the soil contaminated with diesel fuel and engine oil, with 82.3% and 75.4% efficiency, respectively. Applying bioremediation to all contaminated soils accelerated the process of recolonization by edaphic invertebrates. However, the 28-month period was too short to observe full population recovery in soils contaminated with diesel fuel and engine oil. Microbe-enhanced bioremediation accelerated recolonization by epigeic invertebrates on soil contaminated with diesel fuel, whereas it exerted inhibitory effect on recolonization of soil contaminated with engine oil (especially by Collembola). The observed discrepancies in the rates of recolonization for soils contaminated with petrol and diesel fuel that were still noted at the stage of no longer different TPH levels justify the idea to include the survey of edaphic faunal density as one of the parameters in the ecological risk assessment of various bioremediation techniques.  相似文献

Diesel fuel pollution in coastal waters, resulting from recreational boating and commercial shipping operations, is common and can adversely affect marine biota. The purpose of this study was to examine the effect of additions of particulate organic carbon (POC) in the form of naturally-occurring marsh grass (Spartina alterniflora), inorganic nutrients (nitrogen and phosphorus), inert particles, and dissolved organic carbon (DOC) on diesel fuel biodegradation and to attempt to formulate an effective bioremedial treatment for small diesel fuel spills in marine waters. Various combinations of treatments were added to water samples from a coastal marina to stimulate diesel fuel biodegradation. Diesel fuel was added in concentrations approximating those found in a spill and biodegradation of straight chain aliphatic constituents was estimated by measuring mineralization of 14C hexadecane added to diesel fuel. All treatments that included POC showed stimulation of biodegradation. However, the addition of inert particles (glass fiber filters and nylon screening) caused no stimulation of biodegradation. The addition of nitrogen and phosphorus alone did not result in stimulation of biodegradation, but nitrogen and Spartina (although not phosphorus and Spartina) did result in stimulation above that of Spartina alone. Maximum biodegradation rates were obtained by the addition of the Spartina POC, ammonium, and phosphate. The addition of mannitol, a labile DOC source with POC and phosphate resulted in a decrease in diesel fuel biodegradation as compared to POC and phosphate alone. The seasonal pattern of diesel fuel biodegradation showed a maximum in the summer and a minimum in the winter. Therefore, of the treatments tested, the most effective for bioremediation of diesel fuel in marine waters is the addition of POC, nitrogen, and phosphorus.  相似文献
