The objectives of the present study were to determine the effects of resynchronization with GnRH on Day 21 after artificial insemination (AI) on pregnancy rate and losses of pregnancy in lactating dairy cows. Holstein cows (n=585) on two dairy farms were assigned to one of two treatments in a randomized complete block design. On Day 21 after a pre-enrollment AI, animals assigned to the resynchronization (RES) group received 100 microg of GnRH i.m., whereas animals in the control (CON) group received no treatment. All animals were examined ultrasonographically on Days 21 and 28 after AI, and blood samples were taken for progesterone measurement on Day 21. Pregnancy was diagnosed on Day 28 and reconfirmed 14 days later. Nonpregnant cows on Day 28 were inseminated using timed AI after the completion of the Ovsynch protocol 10 and 17 days after enrollment in the study for RES and CON groups, respectively. Progesterone concentration > or =2.35 ng/ml was used as an indicator of pregnancy on Day 21. For RES and CON cows, pregnancy rate at Days 21 (70.9% versus 73.0%, P<0.56), 28 (33.1% versus 33.6%; P<0.80) and 42 (27.0% versus 26.8%; P<0.98) after the pre-enrollment AI did not differ. Administration of GnRH on Day 21 after AI had no effect on pregnancy loss in RES and CON groups from days 21 to 28 (53.2% versus 53.5%; P<0.94) and days 28 to 42 (17.9%; P<0.74) after AI. Pregnancy rate after the resynchronization period was similar for both treatment groups. Resynchronization with GnRH given on Day 21 after AI for initiation of a timed AI protocol prior to pregnancy diagnosis does not affect pregnancy rate and pregnancy loss in lactating dairy cows.  相似文献

Objectives were to determine factors associated with conception rate (CR) and pregnancy loss (PL) in high producing lactating Holstein cows. In Study 1, CR was evaluated in 7633 artificial inseminations (AI) of 3161 dairy cows in two dairy farms. Pregnancy diagnosis was performed by palpation per rectum 39+/-3 days after AI. Environmental temperature was recorded at different intervals prior to and after AI. In Study 2, 1465 pregnancies from 1393 cows diagnosed at 31+/-3 days after AI by ultrasonography on three dairy farms were re-examined 14 days later to determine PL. Temperature > or =29 degrees C was considered to be heat stress (HS). Exposure to HS was defined as following: NH, no heat stress; HS1, exposure to at least 1 day of maximum temperature > or =29 degrees C and average daily maximum temperature (ADMT) <29 degrees C; and HS2, exposure to ADMT > or =29 degrees C. In Study 1, exposure of cows to HS1 and HS2 from 50 to 20 prior to AI was associated with reduced CR compared to cows not exposed to HS (28.8, 23.0, and 31.3%, respectively). Post-AI HS was not associated with CR. Cows inseminated following estrus detection or timed AI had similar CR. As the number of AI increased, CR decreased. Multiparous cows had lower CR than primiparous cows, and occurrence of milk fever and retained placenta was associated with decreased CR. In Study 2, PL was not associated with exposure to HS either prior to or after AI. Cows diagnosed with clinical mastitis experienced increased PL, but parity, number of AI, AI protocol, milk production, and days postpartum at AI were not associated with PL. In conclusion, CR was affected by HS prior to AI, parity, number of AI, and postparturient diseases, whereas PL was affected by clinical mastitis.  相似文献

Body condition may influence pregnancy rates to a timed insemination (Ovsynch/TAI) protocol and affect the economical performance of dairy farms. The objectives were to compare pregnancy rates using the Ovsynch/TAI protocol for the first service of lactating dairy cows with body condition scores < 2.5 (scale: 1 to 5, low BCS group) versus > or = 2.5 (control group) and to estimate the economic impact of the effect of body condition on reproductive performance. At 63 +/- 3 d post partum, cows were assigned to 2 experimental groups (low BCS = 81; control = 126), and were treated with GnRH at d 0 and with PGF2alpha 7 d later. At 48 h after PGF2alpha, cows received an injection of GnRH and were inseminated 16 h later. Pregnancy rates to the Ovsynch/TAI protocol were lower for the low BCS group than for the control group at 27 d (18.1 +/- 6.1% < 33.8 +/- 4.5%; P<0.02) and at 45 d (11.1 +/- 5.4% < 25.6 +/- 4.1%; P<0.02) after insemination. Economic analysis indicated that reducing the percentage of the herd in low body condition increases net revenues per cow per year. Body condition influenced pregnancy rates to the Ovsynch/TAI protocol.  相似文献

The objective of this study was to investigate the effect of supplementing previously treated anovulatory anestrous (AA) dairy cows with progesterone delivered intra-vaginally for 7 days, commencing 4 or 5 days after insemination, on first-service conception rate. A clinical trial, involving 990 AA dairy cows in 14 dairy herds, was conducted during the 2002/2003 breeding season. On Day -8, all cows were treated with a progesterone-containing intravaginal device (Cue-Mate). The devices were removed on Day -2; on Day -1, all cows were given 1mg of estradiol benzoate im. Cows in the control group (n = 499) received no further treatments. Cows in the treatment group (n = 491) that had been inseminated on Day 0 or 1 had a new device inserted (on Day 4 or 5), with removal of the device after 7 days. First-service conception rates for the control and treatment groups were not different (35.0% versus 36.7% respectively; P = 0.41). Furthermore, there was no difference in conception rates between cows that had devices inserted on Day 4 or 5 (31.3% versus 37.2% respectively; P = 0.45). In conclusion, supplementation of previously treated AA dairy cows with an intravaginal progesterone-releasing device for 7 days (commencing 4 or 5 days after insemination) did not significantly improve first-service conception rate.  相似文献

This study was undertaken to determine whether reproductive disorders previous to conception influence pregnancy attrition in dairy cows. Reproductive disorders were registered and pregnancy diagnoses were performed as a part of a reproductive health program at 9 commercial dairy herds in northeastern Spain. Data from 3022 diagnosed pregnant lactating cows were used. Pregnancy diagnosis by palpation per rectum was performed from 33 to 70 d post insemination. Pregnancy attrition was registered when the pregnancy diagnosis was negative at the second palpation carried out between 120 and 150 d following insemination. Data analysis was performed by multiple logistic regression methods. Pregnancy attrition rates were 2.6 and 1.8 times higher in cows with previous pyometra and retained placenta, respectively, than in cows without these disorders. No effect of endometritis, ovarian cysts and repeat breeding was shown on pregnancy attrition. Our results suggest that additional efforts to reduce the risk of retained placenta and pyometra should decrease the incidence of pregnancy attrition in dairy cows.  相似文献

A total of 334 first-service lactating cows in natural estrus were used in the study. Semen was deposited into the uterine body of 174 cows and deep into the uterine horn ipsilateral to the side of impending ovulation of 160 cows. In both groups, insemination was performed within the interval of 50 to 100 d postpartum at 8 to 15 h after estrus detection and after preovulatory follicle palpation. Pregnancy rates were determined by palpation per rectum 50 d post insemination. The pregnancy rate was higher (P < 0.05) for deep uterine horn insemination (70.62%) than for uterine body insemination (60.34%).  相似文献

The objective of this study was to determine the effect of GnRH (100 microg i.m.) treatment 5 and 15 days after timed insemination (TAI) on pregnancy rate and pregnancy loss in lactating dairy cows subjected to synchronization of ovulation. The study included 831 lactating dairy cows subjected to a Presynch-Ovsynch protocol for first service. On the day of TAI (Day 0), cows were randomly assigned to one of four experimental groups. Cows in Group 1 (n = 214) were treated with GnRH on Day 5; cows in Group 2 (n = 209) were treated with GnRH on Day 15; cows in Group 3 (n = 212) were treated with GnRH on both Day 5 and Day 15; cows in Group 4 (n = 196) were not treated. Pregnancy rate was evaluated at Day 27 and Day 45 after TAI. The interestrus interval and the proportion of cows diagnosed not pregnant based on expression of estrus and insemination before pregnancy diagnosis on Day 27 were determined. The results of this study are: (1) GnRH treatment on Day 5 or Day 15 did not increase pregnancy rate, or reduce pregnancy loss between Day 27 and Day 55 after TAI; (2) cows treated with GnRH on both Day 5 and Day 15 had a lower (P < 0.01) proportion of cows diagnosed not pregnant based on expression of estrus before ultrasonography on Day 27 (26.5%) compared to control cows (52.9%), and these cows had an extended (P = 0.05) interestrus interval (23.4 days vs. 21.5 days); and (3) GnRH treatment on both Day 5 and Day 15 after TAI reduced pregnancy rate on Day 27 (36.8% vs. 44.4% for control cows; P < 0.03) and Day 55 (28.3% vs. 36.2% for control cows; P < 0.01). Therefore, strategies to stimulate CL function using multiple doses of GnRH during the luteal phase need to consider potential negative effects.  相似文献

Conception rates (CR) are low in dairy cows and previous research suggests that this could be due to impaired early embryonic development. Therefore, we hypothesized that CR could be improved by embryo transfer (ET) compared with AI. During 365 days, 550 potential breedings were used from 243 lactating Holstein cows (average milk production, 35 kg/day). Cows had their ovulation synchronized (GnRH-7d-PGF(2alpha)-3d-GnRH) and they were randomly assigned for AI immediately after the second GnRH injection (Day 0) or for transfer of one embryo 7 days later. Circulating progesterone concentrations and follicular and luteal size were determined on Days 0 and 7. Pregnancy diagnosis was performed on Days 25 or 32 and pregnant cows were reevaluated on Days 60-66. Single-ovulating cows with synchronized ovarian status had similar CR on Days 25-32 with ET (n = 176; 40.3%) and AI (n = 160; 35.6%). Pregnancy loss between Days 25-32 and 60-66 also did not differ (P = 0.38) between ET (26.2%) and AI (18.6%). When single (n = 334) and multiple (n = 57) ovulators were compared, independent of treatment, multiple ovulators had greater (P < 0.001) circulating progesterone concentrations on Day 7 (2.7 ng/ml versus 1.9 ng/ml) and there was a tendency (P = 0.10) for a greater CR in multiple ovulators (50.9% versus 38.1%). However, there was no difference in CR between AI and ET cows with multiple ovulations (50.0% versus 51.7%). In single-ovulating cows, CR tended to be lower for AI than ET in cows ovulating smaller follicles (diameter < or = 15 mm; 23.7% versus 42.3%; P = 0.06) but not average-diameter follicles (16-19 mm; 41.2% versus 37.3%; P = 0.81) or larger (> or =20 mm; 34.3 versus 51.0%; P = 0.36) follicles. Thus, although ET did not improve overall CR in lactating cows, follicle diameter and number of ovulating follicles may determine success with these procedures.  相似文献

The objective was to investigate ovarian follicular development and hormone concentrations in previously inseminated cows with estrous cycles resynchronized with various resynchronization treatments. Lactating dairy cows were treated with a previously used intravaginal progesterone releasing device (IVD) for 7d (EB+IVD 7+EB, n=15) or 8d (EB+IVD 8+EB, n=16), starting 13d (Day 13) after a first estrus (Day 0) and AI. Estradiol benzoate (EB; 1mgim) was given at device insertion and 24h after removal. Other cows were given the same treatment as the EB+IVD 8+EB cows, but were not treated with EB at IVD insertion (IVD 8+EB, n=11). There were no differences (P>0.05) between EB+IVD 7+EB and EB+IVD 8+EB treatments for follicle dynamics and plasma progesterone concentrations during treatment. Based on a comparison between the IVD 8+EB treated cows and the pooled results of the EB+IVD 7+EB and EB+IVD 8+EB treated cows, EB at device insertion increased the number of follicular waves between Days 13 and 20 (mean+/-S.E.M.; 2.3+/-0.14 vs 2.7+/-0.10, P=0.033), delayed emergence of follicles that were dominant or emerging on Day 20 (17.2+/-0.36 vs 14.1+/-0.65d, P<0.001), reduced diameters of dominant or emerging follicles on Day 20 (9.0+/-0.58 vs 12.7+/-0.59, P<0.001), and reduced plasma progesterone concentrations by 0.85+/-0.44ng/mL (P=0.059) during treatment. Furthermore, comparison of the IVD 8+EB to the EB+IVD 8+EB treated cows demonstrated that treatment with EB at device insertion also reduced the diameter of ovulatory follicles (14.2+/-0.58 vs 19.0+/-0.71mm, P=0.001), delayed emergence of ovulatory follicles (17.0+/-0.32 vs 13.5+/-1.26, P=0.020), and reduced the interval from emergence to ovulation (7.0+/-0.32 vs 10.5+/-1.26d, P=0.020). We concluded that administration of EB altered ovarian follicular dynamics and tended to reduce plasma progesterone concentrations during treatment with an IVD that was used to resynchronize estrous cycles. However, use of a 7-d compared to an 8-day treatment with an IVD did not significantly affect follicle dynamics nor plasma progesterone concentrations during treatment.  相似文献

Thirty superovulated Friesian lactating cows were randomly assigned to two groups. Group I donors were inseminated with one unit of semen deposited into the uterine body at 8, 20 and 32 h after the onset of estrus. Group II donors were inseminated with one unit of semen deposited deep into the uterine horns at 15 h after the onset of estrus. In large herds, inseminators frequently thaw multiple straws of semen and prepare several insemination guns at once. The aim of this study was to measure the effect of breeding order, the order that thawed straws are used, on conception rate in commercial dairy herds. A single professional inseminator, utilizing semen from five suppliers, performed 2629 inseminations over 30 months. Up to four straws were thawed at one time with the restriction that straws were used within 20 min of thawing. Straws were thawed per supplier's recommendations, with 66.4% of the straws pocket thawed and the remainder warm-water thawed. Conception was determined by a pregnancy check at 42 days. Data were modeled by multiple logistic regression analysis, which included herd, breeding order, lactation number, times bred, month bred and year bred. Breeding order had no significant effect on the probability of conception: Cows 1-4 achieved adjusted conception rates of 0.36, 0.41, 0.37, and 0.39, respectively. Odds ratios (and 95% confidence intervals) were 1.00 (1.00-1.00), 1.22 (0.99-1.49), 1.04 (0.82-1.32), and 1.12 (0.86-1.45), respectively. Associated laboratory studies, which evaluated the effect of post-thaw holding time on percentage of sperm with progressive motility, found mean values at 20 min holding time to be largely unchanged from mean values at 5 min. Thawing temperatures were 6, and 35 degrees C and holding temperatures were 6, 22, and 35 degrees C. The length of the trial and the wide use of semen from multiple suppliers with differing thawing methods suggests that under similar conditions, a careful and experienced inseminator can thaw multiple straws of semen and prepare insemination guns sufficient to breed up to four cows within 20 min, without an adverse effect on conception.  相似文献

A total of 335 lactating dairy cows was used to determine the effect of oxytocin or PGF2a given 8 h after treatment with a luteolytic dosage of PGF2a on the percentage of cows exhibiting estrus within 7 d after treatment, and the pregnancy rate to a single insemination at this time. On the initial day of treatment (Day 0), cows with a palpable corpus luteum on the ovary were treated with 25 mg, im of PGF2a. At 8 h later, the cows were divided into 3 groups. Cows in Group 1 (n = 112) were treated with oxytocin (0.33 IU/kg bwt im); cows in Group 2 (n = 112) were treated with 25 mg, im of PGF2a; and cows in Group 3 (n = 111) served as the untreated controls. Cows in all 3 groups were continuously observed for estrus visually or by way of an activated heatmount detector within 7 d after treatment, and were inseminated within 12 h of the observed estrus.Plasma progesterone (P4) concentration was determined using radioimmunoassay on Day 0 and Day 2. Of the cows with P4 greater than 1 ng/ml on Day 0, the percentage of cows observed in estrus within 7 d after treatment was 75, 89 and 72% for cows in Group 1, Group 2 and Group 3, respectively. When all cows were evaluated, the percentage of cows observed in estrus within 7 d after treatment was 60, 70 and 55% for cows in Group 1, Group 2 and Group 3, respectively. In both instances, the value for cows in Group 2 was significantly higher than that for either cows in Group 1 or Group 3. The pregnancy rate for cows inseminated within 7 d was similar for cows in all 3 groups. The results of this study demonstrated that treatment of dairy cows with 2 luteolytic dosages of PGF2a at an 8-h interval resulted in more cows being observed in estrus within 7 d than with 1 treatment with PGF2a, or with oxytocin given at an 8-h interval after a luteolytic dosage of PGF2a.  相似文献

The effect of human chorionic gonadotropin (HCG) on corpus luteum (CL) function in lactating dairy cows and its subsequent effect on pregnancy rates achieved with artificial insemination was studied in two parts. In Part 1, four IM injections at intervals of seven days or two IM injections at a 14 day interval of HCG (10,000 IU) were given to two groups of six cows each, groups A and B respectively, beginning on days 9, 10 or 11 of the estrous cycle. Ten control cows were given 10 ml of isotonic saline solution on days 9, 10 or 11. Interestrous intervals were prolonged by an average of 8.1 and 3.4 days for groups A and B respectively over controls. In Part 2, 200 lactating dairy cows on nine farms were assigned, on an alternate basis at insemination, to control or treatment groups to study the effect of CL prolongation via HCG on pregnancy rate. A single injection of HCG (10,000 IU) was given 10, 11 or 12 days after a first or second insemination. Pregnancy rates for control and treated cows were similar (64 101 = 63% and 58 99 = 59% respectively). Interestrous intervals of treated nonpregnant cows were prolonged by approximately five days. Providing additional time for developing embryos to become established in utero by delaying luteolysis did not improve pregnancy rates achieved with artificial insemination in lactating dairy cows.  相似文献

A total of 585 repeat-breeder dairy cows was used to study the effect of GnRH treatment, either at or prior to insemination, on the pregnancy rate. The cows were divided into 6 treatment groups. Cows in Group 1 (n = 142) were observed in estrus, and 11 +/- 0.42 hours (mean +/- SEM) later they were given 100 ug, i.m. gonadotropin releasing hormone (GnRH) and were inseminated. Cows in Group 2 (n = 139) were observed in estrus and were inseminated 11.4 +/- 0.43 hours later. Cows in Group 3 (n = 33) were monitored for estrus with an activated heatmount detector but were not observed in estrus; they were inseminated 1.5 +/- 0.87 hours later and were given 100 ug, i.m. GnRH. Cows in Group 4 (n = 35) were not observed in estrus, but they did activate the heatmount detector and were inseminated 2.2 +/- 0.87 hours later. Cows in Group 5 (n = 107) were observed in estrus, given 100 ug, i.m. GnRH 2.0 +/- 0.40 hours later, and were inseminated 9 +/- 0.60 hours after GnRH treatment. Cows in Group 6 (n = 129) were observed in estrus and were inseminated 10 +/- 0.50 hours later. Pregnancy rates were analyzed by Chi-square. Interactions between pregnancy rate, treatment and time of insemination were evaluated using ANOVA and LSM (P < 0.05). There was no effect on pregnancy rate when GnRH was given at or prior to insemination. Cows inseminated on the basis of observed estrus had a higher pregnancy rate (P < 0.05) than cows inseminated on the observation of an activated heatmount detector. From the results of this study, it is concluded that treatment with GnRH at or prior to insemination did not improve the pregnancy rate of repeat-breeder dairy cows.  相似文献

The objective of this study was to compare fixed-time AI pregnancy rate in Angus crossbred beef cows inseminated with frozen-thawed or fresh-extended semen. Two ejaculates from each of two Angus bulls were collected by artificial vagina and pooled for each bull. The pooled semen from each bull was divided into two aliquots; Aliquot 1 was extended using Caprogen® (LIC, Hamilton, New Zealand) to a concentration of 3 × 106 sperm/straw and Aliquot 2 was extended using egg-yolk-glycerol extender to a concentration of 20 × 106 sperm/straw. Semen extended with Caprogen® was maintained at ambient temperature and semen extended with egg-yolk-glycerol extender was frozen and maintained at −196 °C until insemination. In each of two breeding seasons (Fall 2007 and Spring 2008), Angus-crossbeef cows (N = 1455) at 12 locations were randomly assigned within location to semen type [Fresh (N = 736) vs. Frozen (N = 719)] and sire [1 (N = 731) vs. 2 (N = 724)]. All cows were synchronized with 100 μg of GnRH im and a progesterone Controlled Internal Drug Release insert (CIDR) on Day 0, and on Day 7, 25 mg of PGF2α im and CIDR removal. All cows received 100 μg of GnRH im and were inseminated at a fixed-time on Day 10, 66 h after CIDR removal. Timed-AI pregnancy rates were influenced by season (P < 0.05), cows detected in estrus prior to and at AI (P < 0.001), and dam age (P < 0.01). Pregnancy rates were not affected by semen type (Fresh = 51.5% vs. Frozen = 50.4%; P = 0.66) and there were no significant interactions of semen type by estrus expression, semen type by sire, or semen type by season (P > 0.1). In conclusion, commercial beef cows inseminated with fresh-extended semen (3 × 106 sperm/straw) yielded comparable pregnancy rates to conventional frozen-thawed semen in a progesterone supplemented, CO-Synch fixed-time AI synchronization protocol and may provide an alternate to frozen semen for more efficient utilization of superior genetics.  相似文献

The objective was to compare the probability of pregnancy after fixed-time insemination in cows diagnosed as non-pregnant and re-inseminated following the Ovsynch protocol, with or without exogenous progesterone. Cows (n=415) used in this study originated from 25 farms. Upon diagnosis of non-pregnancy between 30 and 60 days after AI, cows were randomly assigned to receive either a progesterone releasing intravaginal device (PRID; n=208) or a placebo intravaginal device (PID; n=207). All cows received GnRH at enrollment (Day 0), PGF(2alpha) concurrent with intravaginal device removal 7 days later, GnRH on Day 9 and fixed-time insemination 16h later (Day 10). Cows observed in estrus prior to Day 7, had the device removed and were inseminated. Ovaries were examined by transrectal palpation at the time of enrollment and the prominent structures were assessed and recorded. Body condition score, lameness status, interval from previous insemination, and times bred at enrollment were recorded. At intravaginal device removal, the occurrence and intensity of vaginitis was determined according to the amount of debris on the device. Overall, the intravaginal device retention rate was 91%. A total of 5.2% of PID-treated cows and 2.9% of PRID-treated cows were detected in estrus within the 7 days treatment period. Pregnancy status was diagnosed between 30 and 56 days after insemination and all cows were followed for a minimum of 150 days after enrollment. Approximately 28% of cows had evidence of mild vaginitis in response to the intravaginal device, whereas 6% of cows had copious debris associated with the intravaginal device at removal. The probability of pregnancy after fixed-time insemination was 43.8% versus 34.9% in PRID-treated versus PID-treated animals. Exogenous progesterone provided through an intravaginal device to non-pregnant cows that had not displayed estrus improved the probability of pregnancy after fixed-time AI.  相似文献

The objective was to evaluate the effect of estrus occurrence (based on removal of tail-head marks) on ovarian responses and pregnancy per AI (P/AI; 30 d after AI) in suckled Bos indicus beef cows submitted to timed AI (TAI) protocols. Cows received an intravaginal device containing 1.0 g progesterone, and 2.0 mg estradiol benzoate im; 8 d later, the intravaginal device was removed, and they were given PGF (0.25 mg of cloprostenol sodium) and 300 IU of eCG, with TAI 48 to 52 h later. In Experiment 1, cows were assigned to receive one of three treatments: 1 mg of estradiol cypionate (ECP) im at progesterone (P4) device removal (N = 178); 10 μg of GnRH im at TAI (N = 190); or both treatments (N = 172). In cows given estradiol (ECP or ECP + GnRH), more displayed estrus (P = 0.002) and became pregnant (P < 0.0001) compared with those receiving only GnRH. In Experiment 2, the effect of the occurrence of estrus on ovarian responses was evaluated in cows (N = 53) synchronized using ECP at device removal. Cows that displayed estrus had a greater diameter of the largest follicle (LF) at device removal (P < 0.0001), a greater diameter at TAI (P < 0.0001), a greater ovulation rate (P = 0.02), a larger CL (P = 0.02), and a greater P4 concentration (P < 0.0001) than cows that did not display estrus. In Experiment 3, the effect of GnRH treatment on P/AI at TAI was evaluated in cows that received ECP at device removal, and either displayed, or did not display, estrus (N = 726). There was no estrus by GnRH interaction (P = 0.22); the P/AI was greater (P < 0.0001) in cows that displayed estrus (61.9%) than cows that did not display estrus (41.4%). However, GnRH did not improve (P = 0.81) P/AI (GnRH = 53.7% vs. no GnRH = 52.6%). In conclusion, exogenous estradiol at device removal increased both the proportion of suckled Bos indicus cows that displayed estrus and P/AI. Cows that displayed estrus had better ovarian responses (i.e., larger follicles at TAI, a greater ovulation rate, larger CL, and greater P4 concentrations) following an estradiol/P4-based synchronization protocol. Although occurrence of estrus improved pregnancy outcomes, GnRH at TAI did not improve P/AI in suckled Bos indicus cows treated with ECP, regardless of estrus occurrence.  相似文献

This is the very first report that suggests high pregnancy rates can be obtained with use of the Doublesynch protocol in anestrous dairy cows. Recently, a new synchronization method has been developed (Doublesynch) that resulted in synchronized ovulations both after the first and second gonadotropin-releasing hormone (GnRH) treatments. It was suggested that this protocol has the potential to increase the pregnancy rates in primiparous dairy cows. The aim of the current study was to confirm the success of the Doublesynch protocol and further to investigate the effect of this method on pregnancy rates in anestrous cows. Lactating primiparous Holstein (Bos taurus) cows (n = 165) between 60 and 172 d postpartum were monitored twice with 10-d intervals (on Days -10 and 0) by ultrasonography, and blood samples were collected. Cows were classified as anestrous if both blood samples had progesterone (P4) concentration <1 ng/mL and as cyclic if at least one of the two samples had P4 concentration ≥1 ng/mL. Cyclic cows were classified again as cyclic-high P4 (having an active corpus luteum) if the second blood samples had P4 concentrations ≥1 ng/mL and as cyclic-low P4 if P4 concentrations were <1 ng/mL on Day 0. Then, the cows classified as anestrous (n = 51), cyclic-high P4 (n = 63), or cyclic-low P4 (n = 51) were put into two treatment groups (Ovsynch or Doublesynch) randomly to establish six groups. Cows in the Ovsynch group were administered a GnRH (lecirelin 50 μg, im) on Day 0, PGF (Prostaglandin F2 alpha, D-cloprostenol 0.150 mg, im) on Day 7, and a second dose of GnRH 48 h later. Cows in the Doublesynch group were administered a PGF on Day 0, GnRH on Day 2, a second PGF on Day 9, and a second GnRH on Day 11. Timed artificial insemination (TAI) was performed 16 to 20 h after the second GnRH in both treatment groups. Pregnancy diagnosis was conducted (by ultrasonography) 45 ± 5 d after TAI. In anestrous cows and those with high and low progesterone concentration at treatment onset, Doublesynch treatment led to markedly increased pregnancy rates with respect to Ovsynch treatment (P < 0.05). On the overall analysis of data, it was revealed that the Doublesynch method increased pregnancy rates by 43 percentage units (29.8% vs. 72.8%, P < 0.0001) in relation to Ovsynch. Pregnancy rates of cows having small, medium, or large follicles at the day of second GnRH administration were similar in the Doublesynch group (70.4%, 85.2%, and 63.0%, respectively; P > 0.05), whereas pregnancy rates reduced dramatically as follicle size increased in the Ovsynch group, particularly in cows with follicles greater than 16 mm (45.5%, 28.1%, and 5.3%, respectively; P < 0.05). Our results confirm and support observations that the Doublesynch protocol increases the pregnancy rates in postpartum primiparous cows as reported previously. Our data also demonstrate that the Doublesynch method increases the pregnancy rates in anestrous cows. Thus, these data suggest that the Doublesynch protocol can be used to obtain satisfactory pregnancy rates after TAI in both anestrous and cycling primiparous dairy cows regardless of stage of estrous cycle.  相似文献

Fertility in relation to acetone concentration in milk and level of nutrition was studied in 38,624 lactations from 474 herds over a 3-year period. Herd-related data on nutrition were collected once each year. Milk acetone concentrations higher than 0.40 mM were deemed to be hyperketonaemic. The interval from calving to first service was about 5 days longer in cows with acetone concentrations >2.00 mM, while the interval to the last service was shortest at 0.40 to 1.00 mM. The risk for cystic ovaries was severely increased in first calving heifers with acetone concentrations >2.00 mM (odds ratio; 8.7). In herds with a high frequency of hyperketonaemic cows, primiparous cows had a 6-day longer period from calving to the first service and a 12-day longer period from calving to the last service. Increased feeding frequency of concentrate (2 vs 4 times/day) was related to shorter intervals from calving to first service and from calving to last service of 5 and 6 days, respectively, in mature cows. Increased total intake of energy in early lactation was related to shorter intervals from calving to last service in both primiparous and multiparous cows, 0.3 and 0.1 days per megajoule metabolizable energy, respectively. However, increased amounts of concentrate at calving in multiparous cows, and 15 days after calving in primiparous cows, were related to longer intervals from calving to last service and from calving to first service, respectively. The negative effect on these intervals was estimated to be approximately 2 days per kilogram of concentrate.  相似文献

Two experiments were performed to determine the effects of repeated administration of PGF2alpha in the immediate post partum period on the prevalence of clinical endometritis at 22 and 58 days post partum, and the probability of pregnancy at first insemination, in post partum dairy cows. In Experiment 1, 228 cows on day 7 post partum were used. Cows in Group 1 (n = 114) were treated twice with PGF2alpha (25 mg, im) 8 h apart on days 7 and 14 post partum, and only once on days 22 and 35 post partum. Cows in Group 2 (n = 114) served as untreated controls. Vaginoscopy and transrectal palpation of the genital tract were done on days 22 and 58 post partum. Cows in both groups were inseminated at estrus after a voluntary waiting period of 100 days. Pregnancy was determined by transrectal palpation between 45 and 50 days after insemination. Repeated administration of PGF2alpha in the immediate post partum period did not reduce the prevalence of clinical endometritis on days 22 or 58 post partum (65 versus 62% on day 22 and 28.3 versus 35.2% on day 58 in Groups 1 and 2, respectively). There was no significant difference in the probability of pregnancy at first insemination between cows in both groups. In Experiment 2, 418 cows on day 7 post partum were used. Cows in Group 1 (n = 209) were treated twice with PGF2alpha (25 mg, im) 8h apart on days 7 and 14 post partum, and only once on days 22 and 35 post partum. Cows in Group 2 (n = 209) served as untreated controls. Cows in both groups were subjected to the Presynch and Ovsynch protocols on days 49 and 75 post partum, respectively. Pregnancy was determined by transrectal ultrasonography between days 29 and 32 after timed insemination. There was no significant difference in the probability of pregnancy at first insemination between cows in both groups. In conclusion, repeated administration of PGF2alpha to cows in this herd had no effect on the prevalence of clinical endometritis at 22 and 58 days post partum, and that there was no effect on the probability of pregnancy after insemination at estrus among cows with a voluntary waiting period of > 100 days, or at timed AI at 85 days in milk when Presynch was employed.  相似文献
