A detailed comparison of green leaf phosphoenolpyruvate carboxylases from the C4-species Atriplex spongiosa and the C3-species Atriplex hastata revealed significant physical and kinetic differences. The two alloenzymes can be separated by anion exchange chromatography but have comparable molecular weights (350,000). Maximal velocity estimates were 38.0 and 1.48 micromoles per minute per milligram of chlorophyll for the carboxylases of A. spongiosa and A. hastata, respectively. Km phosphoenolpyruvate estimates were 0.49 and 0.08 mm for the C4A. spongiosa and C3A. hastata and the Km Mg estimates were 0.33 mm for the C4 species and 0.017 mm for the C3 species. The activity of the phosphoenolpyruvate carboxylase of A. spongiosa is more sensitive to chloride and phosphate than the phosphoenolpyruvate carboxylase of A. hastata, but both are equally sensitive to Mg chelating substances such as ATP, ADP, and citrate if assayed at their respective Km Mg values. A survey of the phosphoenolpyruvate carboxylases from 18 C4 and C3 species resulted in mean maximal velocity estimates of 29.0 ± 13.2 and 1.50 ± 0.57 micromoles per minute per milligram of chlorophyll for the C4 species and C3 species, respectively. Km phosphoenolpyruvate estimates were 0.59 ± 0.35 mm and 0.14 ± 0.07 mm for the C4 and C3, and Km Mg estimates were 0.50 ± 0.30 and 0.097 ± 0.057 mm for C4 and C3. All differences between means were significant at the 0.01 confidence level, supporting our hypothesis that the phosphoenolpyruvate carboxylase alloenzymes of C4 and C3 plants are functionally different and are associated with different photosynthetic roles. Both function in the photosynthetic production of C4 acids, the phosphoenolpyruvate carboxylase of C4 species largely producing malate or aspartate (or both) as a photosynthetic intermediate and the phosphoenolpyruvate carboxylase of C3 species producing malate or aspartate (or both) as a photosynthetic product.  相似文献

We investigated seasonal patterns of photosynthetic responses to CO2 concentrations in Spartina alterniflora Loisel, an aerenchymous halophyte grass, from a salt marsh of the Bay of Fundy (NB, Canada), and from plants grown from rhizome in controlled-environment chambers. From late May to August, CO2 compensation concentrations () of field-grown leaves varied between 2.5–10.7 cm3(CO2) m–3, with a mean of 5.4 cm3(CO2) m–3. From September onwards field leaves showed CO2 compensation concentrations from 6.6–21.1 cm3(CO2) m–3, with a mean of 13.1 cm3 m–3 well into the C3–C4 intermediate range. The seasonal variability in did not result from changing respiration, but rather from a sigmoidal response of net photosynthetic rate (P N) to applied CO2 concentration, found in all tested leaves but which became more pronounced late in the season. One explanation for the sigmoidal response of P N to external CO2 concentration could be internal delivery of CO2 from roots and rhizomes to bundle sheath cells via the aerenchyma, but the sigmoidal responses in S. alterniflora persisted out to the tips of leaves, while the aerenchyma extend only to mid-leaf. The sigmoidicity persisted when CO2 response curves were measured from low to high CO2, or from high to low CO2, and even when prolonged acclimation times were used at each CO2 concentration.  相似文献

A total cell recycling suspension perfusion reactor has been constructed for investigation of specific monoclonal antibody secretion rate of the 9.2.27 murine hybridoma under conditions of a very low growth rate. By rapidly recycling hybridoma cells using a thermostated tangential flow filter, 3.6 mg cell dry weight/cm3 could be maintained at growth rate of <0.05 max for over 250 h. Under these conditions, secretion of lactate, ammonia and l-alanine were directly related to the rate of l-glutamine supply. Monoclonal antibody accumulated in the reactor to levels in excess of 1400 g/ cm3. During intracellular polarization of identified sensory neurons of the leech by square pulses of hyperpolarizing current electrical parameters of the cell membranes were determined: input resistance of the neuron Rn, time constant of the membrane , the ratio between conductance of the cell processes and conductance of the soma , the resistance of the soma membrane rs, the input resistance of the axon r a , capacitance of the membrane Cs, and resistivity of the soma membrane Rs. The results obtained by the study of various types of neurons were subjected to statistical analysis and compared with each other. Significant differences for neurons of N- and T-types were found only between the values of , Cs, and Rs (P<0.01). These parameters also had the lowest coefficients of variation. The surface area of the soma of the neurons, calculated from the capacitance of the membrane (the specific capacitance of the membrane was taken as 1 µF/cm2) was 7–10 times (N-neurons) or 4–6 times (T-neurons) greater than the surface area of a sphere of the same diameter. The resistivity of the soma membrane Rs was 35.00 k·cm2 for cells of the N-type and 19.50 k·cm2 for T-neurons. The reasons for the relative stability of this parameter compared with the input resistance of the cell (coefficient of variation 22–7 and 53–31% respectively) are discussed. Rhodospirillum rubrum was grown continuously and photoheterotrophically under light limitation using a cylindrical photobioreactor in which the steady state biomass concentration was varied between 0.4 to 4 kg m–3 at a constant radiant incident flux of 100 W m–2. Kinetic and stoichiometric models for the growth are proposed. The biomass productivities, acetate consumption rate and the CO2 production rate can be quantitatively predicted to a high level of accuracy by the proposed model calculations. The quantity of proteins processed by an electrophoretic technique is proportional to the cross-sectional area of the gel. For preparative purifications, an increase in the cross-sectional area is desired, but the Joule heating phenomenon restricts such an increase. The governing heat equation is analyzed and simplified with reference to Counteracting Chromatographic Electrophoresis. The application of the method of weighted residuals yields a compact and accurate solution for the maximum temperature rise in the column which is suitable for design calculations. Pigment analysis was performed by means of normal phase HPLC on a number of bacteriochlorophyll a and b containing species of purple bacteria that contain a core antenna only. At least 99% of the bacteriochlorophyll in Rhodobacter sphaeroides R26, Rhodopseudomonas viridis and Thiocapsa pfennigii was esterified with phytol (BChl a p and BChl b p, respectively). Rhodospirillum rubrum contained only BChl a esterified with geranyl-geraniol (BChl a GG). Rhodospirillum sodomense and Rhodopseudomonas marina contained, in addition to BChl a p, small amounts of BChl a GG, and presumably also of BChl a esterified with dihydro and tetrahydro geranyl-geraniol (2,10,14-phytatrienol and probably 2,14-phytadienol). In all species bacteriopheophytin (BPhe) esterified with phytol was present. The BChl/BPhe ratio indicated that in these species a constant number of 25 ± 3 antenna BChls is present per reaction centre. This number supports a model in which the core antenna consists of 12 - heterodimers surrounding the reaction centre. Dry weight and Relative Growth Rate of Lemna gibba were significantly increased by CO2 enrichment up to 6000 l CO2 l–1. This high CO2 optimum for growth is probably due to the presence of nonfunctional stomata. The response to high CO2 was less or absent following four days growth in 2% O2. The Leaf Area Ratio decreased in response to CO2 enrichment as a result of an increase in dry weight per frond. Photosynthetic rate was increased by CO2 enrichment up to 1500 l CO2 l–1 during measurement, showing only small increases with further CO2 enrichment up to 5000 l CO2 l–1 at a photon flux density of 210 mol m–2 s–1 and small decreases at 2000 mol m–1 s–1. The actual rate of photosynthesis of those plants cultivated at high CO2 levels, however, was less than the air grown plants. The response of photosynthesis to O2 indicated that the enhancement of growth and photosynthesis by CO2 enrichment was a result of decreased photorespiration. Plants cultivated in low O2 produced abnormal morphological features and after a short time showed a reduction in growth.  相似文献

We determined for Vicia faba L the influence of nitrogen uptake and accumulation on the values of photon saturated net photosynthetic rate (P Nmax), quantum yield efficiency (), intercellular CO2 concentration (C i), and carboxylation efficiency (C e). As leaf nitrogen content (NL) increased, the converged onto a maximum asymptotic value of 0.0664±0.0049 mol(CO2) mol(quantum)–1. Also, as NL increased the C i value fell to an asymptotic minimum of 115.80±1.59 mol mol–1, and C e converged onto a maximum asymptotic value of 1.645±0.054 mol(CO2) m–2 s–1 Pa–1 and declined to zero at a NL-intercept equal to 0.596±0.096 g(N) m–2. fell to zero for an NL-intercept of 0.660±0.052 g(N) m–2. As NL increased, the value of P Nmax converged onto a maximum asymptotic value of 33.400±2.563 mol(CO2) m–2 s–1. P N fell to zero for an NL-intercept of 0.710±0.035 g(N) m–2. Under variable daily meteorological conditions the values for NL, specific leaf area (L), root mass fraction (Rf), P Nmax, and remained constant for a given N supply. A monotonic decline in the steady-state value of Rf occurred with increasing N supply. L increased with increasing N supply or with increasing NL.  相似文献

A vector system has been developed to express isoenzyme A1 of sweet potato peroxidase (POD) and was introduced into Saccharomyces cerevisiae. The system contains the signal sequence of Aspergillus oryzae -amylase to facilitate the extracellular secretion of peroxidase under the control of constitutive glyceraldehyde-3-phosphate dehydrogenase (GPD) promoter. In a batch culture using YNBDCA medium (yeast nitrogen base without amino acids 6.7 g l–1, Casamino acids 5 g l–1 and glucose 20 g l–1), the recombinant strain expressed the swpa1 gene giving a secretion yield of POD activity of ca. 90% of total expressed peroxidase. Supplementation with PMSF (0.05 mM) and Casamino acids (5 g/50 ml) increased extracellular POD activity to nearly 10 kU ml–1, equivalent to 1.5 kU g–1 cell dry wt. This is 9 fold higher than that obtained in medium without PMSF. From SDS-PAGE and native-PAGE analyses POD has an M r of 53 kDa.  相似文献

Summary The lysogenic cholera phage, Kappa is some ten to twenty folds more resistant to UV (254 nm) than are most of the T. phages ofE. coli, or the cholera phage PL 163/10, or the hostV. cholerae strain H218 Smr, the 37% (D 37) and 10% (D 10) survival doses being 255.8 J/m2 and 633.6 J/m2 respectively. The UV-irradiated Kappa phages could be photoreactivated in the hostV. cholerae strain H218 Smr to a maximum extent of 40%. The removal of the number of lethal hits per phage by the survival-enhancement treatment (photoreactivation) with time followed an exponential relation, the constant probability of removal of lethal hit per unit time being 2.8 × 10–2 min–1. The UV-irradiated phages could also be Weigle reactivated in the host strain H218 Smr by a small degree, the maximum reactivation factor (ratio of survivals in UV-irradiated and non-irradiated hosts) being 1.50.  相似文献

Effect of fruiting on carbon budgets of apple tree canopies   总被引:1,自引:0,他引:1  
Summary Carbon budgets were calculated from net photosynthesis and dark respiration measurements for canopies of field-grown, 3-year-old apple trees (Malus domestica Borkh.) with maximum leaf areas of 5.4 m2 in a temperature-controlled Perspex tree chamber, measured in situ over 2 years (July 1988 to October 1990) by computerized infrared gas analysis using a dedicated interface and software. Net photosynthesis (Pn) and carbon assimilation per leaf area peaked at respectively 8.3 and 7.7 mol CO2 m–2 s–1 in April. Net photosynthesis (Pn) and dark respiration (Rd) per tree peaked at 3.6 g CO2 tree–1 h–1 (Pn) and 1.2 g CO2 tree–1 h–1 (Rd), equivalent to 4.2 mol CO2 (Pn) and 1.4 mol CO2 (Rd) m–2 s–1 with maximum carbon gain per tree in August and maximum dark respiration per tree in October 1988 and 1989. In May 1990, a tree was deblossomed. Pn (per tree) of the fruiting apple tree canopy exceeded that of the non-fruiting tree by 2–2.5 fold from June to August 1990, attributed to reduced photorespiration (RI), and resulting in a 2-fold carbon gain of the fruiting over the non-fruiting tree. Dark respiration of the fruiting tree canopy progressively exceeded, with increasing sink strength of the fruit, by 51% (June–August), 1.4-fold (September) and 2-fold (October) that of the non-fruiting tree due to leaf (i. e. not fruit) respiration to provide energy (a) to produce and maintain the fruit on the tree and (b) thereafter to facilitate the later carbohydrate translocation into the woody perennial parts of the tree. The fruiting tree reached its optium carbon budget 2–4 weeks earlier (August) then the non-fruiting tree (September 1990). In the winter, the trunk respired 2–100 g CO2 month–1 tree–1. These data represent the first long-term examination of the effect of fruiting without fruit removal which shows increased dark respiration and with the increase progressing as the fruit developed.  相似文献

Eicosapentaenoic (EPA) and docosahexaenoic (DHA) acid productivities from chemostat cultures of an isolate of Isochrysis galbana have been studied. The productivities reached in the interval of dilution rates between 0.0295 h–1 and 0.0355 h–1 were 1.5mg·1–1·h–1 for lipids, 300 g·1–1·h–1 for EPA and 130g1·1–1·h–1 for DHA. Furthermore, light attenuation by mutual shading, and agitation speed influences on growth and fatty acid composition were analysed. A model relating steady-state dilution rates to internal average light intensity has been proposed, the parameter values of which obtained by non-linear regression were: maximum specific growth rate (max)=0.0426 h–1; the affinity of cells to light (Ik) = 10.92 W·m–2; the exponent (n) = 5.13; regression coefficient (r 2)=0.9999. Summary The function of the caecal bulb, and its adaptation to chronic high- or low-Na+ intake, was investigated by in vivo perfusion of anaesthetised birds. Effects of acute aldosterone injection (125 g·kg–1 body mass) were also measured.Evidence was found for primary active net absorption of Na+, inducing parallel Na-linked absorption of water and Cl and secretion of K+. Around 20–35% of total Cl absorption and K+ secretion were independent of Na+ fluxes, and these components appear to be driven by passive processes with apparent conductances of 6.3×10–3 (G Cl) and 1.1×10–3 (G K) S·cm–2.Acetate (40mM) stimulated Na+ fluxes (8.5–9.9 Eq·cm–2·h–1) and Na-linked water fluxes (27–44 l·cm–2·h–1). We investigated the properties of a galactosyltransferase (GalT) that is involved in the synthesis of -(14)-galactan side chains of pectins. A membrane preparation of etiolated 6-day-old soybean (Glycine max Merr.) hypocotyls transferred [14C]Gal from UDP-[14C]Gal into intact and partially hydrolyzed lupin -(14)-galactans of various chain lengths as exogenous acceptors, while activity to endogenous acceptors was negligible. Maximal activity occurred at pH 6.5 and 20–25°C in the presence of 25 mM Mn2+ and 0.75% Triton X-100. The transfer reaction onto the unmodified commercial pectic galactan (M r>150,000) from lupin we used was very low but increased when the M r of the galactan was reduced by partial acid hydrolysis. Among the partially hydrolyzed galactans, high-M r (average M r 60,000) -(14)-galactan was a more efficient acceptor [specific activity 2,000–3,000 pmol min–1 (mg protein)–1] than low-M r (average M r 10,000 and 5,000) polymers. Digestion of the radiolabeled product from high-M r galactan with endo--(14)-galactanase released mainly radioactive -(14)-galactobiose and Gal, indicating that the transfer of [14C]Gal occurred through -(14)-linkages. HPLC analysis showed that the enzyme also catalyzes incorporation of Gal into pyridylaminated (PA) -(14)-galactooligomers with degree of polymerization at least 5. Oscillatoria amphigranulata is a fast-growing (3 doublings/day) cyanobacterium isolated from sulfide hot springs in New Zealand. Photosynthesis, as measured by incorporation of [14C]-HCO 3 - , was initially inhibited by 0.3–1.5 mM sulfide at pH 7.9–8.1. However, conversion to sulfide-dependent anoxygenic photosynthesis occurred in about 2 h or less under light intensities of 3–14 klx. Under the stimulation of higher light intensity (8–14 klx) a partial recovery of oxygenic photosynthesis also occurred. It was concluded that oxygenic photosynthesis was responsible for 21–42% of the total incorporation at sulfide concentrations of 1.0–0.3 mM, respectively. This contribution was suppressed at 1.5 mM sulfide and not elicited under lower light intensities (3–7 klx). As judged by the inhibitory effect of 10 g/ml chloramphenicol protein synthesis was required for attainment of both anoxygenic photosynthesis and photosystem II recovery. Sulfide could not be replaced by thiosulfate, elemental sulfur or dithionite as electron donors in photosynthesis, but elemental sulfur could serve as the sole assimilatory source of sulfur. We investigated to what extent south-exposed leaves (E-leaves) of the evergreen ivy (Hedera helix L.) growing in the shadow of two deciduous trees suffered from photoinhibition of photosynthesis when leaf-shedding started in autumn. Since air temperatures drop concomitantly with increase in light levels, changes in photosynthetic parameters (apparent quantum yield, i and maximal photosynthetic capacity of O2 evolution, Pmax; chlorophyll-a fluorescence at room temperature) as well as pigment composition were compared with those in north-exposed leaves of the same clone (N-leaves; photosynthetic photon flux density PPFD< 100 mol · m–2 · s–2) and phenotypic sun leaves (S-leaves; PPFD up to 2000 mol · m–2 · s–1).In leaves exposed to drastic light changes during winter (E-leaves) strong photoinhibition of photosynthesis could be observed as soon as the incident PPFD increased in autumn. In contrast, in N-leaves the ratio of variable fluorescence to maximum fluorescence (FV/FMm) and i did not decline appreciably prior to severe frosts (up to -12° C) in January. At this time, i was reduced to a similar extent in all leaves, from about 0.073 mol O2 · mol–1 photons before stress to about 0.020. Changes in i were linearly correlated with changes in fv/fm (r = 0.955). The strong reduction in FV/FM on exposure to stress was caused by quenching in FM. The initial fluorescence (F0), however, was also quenched in all leaves. The diminished fluorescence yield was accompanied by an increase in zeaxanthin content. These effects indicate that winter stress in ivy primarily induces an increase in non-radiative energy-dissipation followed by photoinhibitory damage of PSII. Although a pronounced photooxidative bleaching of chloroplast pigments occurred in January (especially in E-leaves), photosynthetic parameters recovered completely in spring. Atmospheric depostion and stream discharge and solutes were measured for three years (September 1984 — August 1987) in two mixed conifer watersheds in Sequoia National Park, in the southern Sierra Nevada of California. The Log Creek watershed (50 ha, 2067–2397 m elev.) is drained by a perennial stream, while Tharp's Creek watershed (13 ha, 2067–2255 m elev.) contains an intermittent stream. Dominant trees in the area include Abies concolor (white fir), Sequoiadendron giganteum (giant sequoia), A. magnifica (red fir), and Pinus lambertiana (sugar pine). Bedrock is predominantly granite and granodiorite, and the soils are mostly Pachic Xerumbrepts. Over the three year period, sulfate (SO4 2–), nitrate (NO3 ), and chloride (Cl–1) were the major anions in bulk precipitation with volume-weighted average concentrations of 12.6, 12.3 and 10.0 eq/1, respectively. Annual inputs of NO3-N, NH4-N and SO4-S from wet deposition were about 60 to 75% of those reported from bulk deposition collectors. Discharge from the two watersheds occurs primarily during spring snowmelt. Solute exports from Log and Tharp's Creeks were dominated by HCO3 , Ca2+ and Na+, while H+, NO3 , NH4 + and PO4 3– outputs were relatively small. Solute concentrations were weakly correlated with instantaneous stream flow for all solutes (r2 <0.2) except HCO3 (Log Cr. r2 = 0.72; Tharp's Cr. r2 = 0.38), Na+ (Log Cr. r2 = 0.56; Tharp's Cr. r2 = 0.47), and silicate (Log Cr. r2 = 0.71; Tharp's Cr. r2 = 0.49). Mean annual atmospheric contributions of NO3-N (1.6 kg ha–1), NH4-N (1.7 kg ha–1), and SO4-S (1.8 kg ha–1), which are associated with acidic deposition, greatly exceed hydrologic losses. Annual watershed yields (expressed as eq ha–1) of HCO3 exceeded by factors of 2.5 to 37 the annual atmospheric deposition of H+.  相似文献

Hydraulic resistances to water flow have been determined in the cortex of hypocotyls of growing seedlings of soybean (Glycine max L. Merr. cv. Wayne). Data at the cell level (hydraulic conductivity, Lp; half-time of water exchange, T 1/2; elastic modulus, ; diffusivity for the cell-to-cell pathway, D c) were obtained by the pressure probe, diffusivities for the tissue (D t) by sorption experiments and the hydraulic conductivity of the entire cortex (Lpr) by a new pressure-perfusion technique. For cortical cells in the elongating and mature regions of the hypocotyls T 1/2=0.4–15.1 s, Lp=0.2·10-5–10.0·10-5 cm s-1 bar-1 and D c=0.1·10-6–5.5·10-6 cm2 s-1. Sorption kinetics yielded a tissue diffusivity D t=0.2·10-6–0.8·10-6 cm2 s-1. The sorption kinetics include both cell-wall and cell-to-cell pathways for water transport. By comparing D c and D t, it was concluded that during swelling or shrinking of the tissue and during growth a substantial amount of water moves from cell to cell. The pressure-perfusion technique imposed hydrostatic gradients across the cortex either by manipulating the hydrostatic pressure in the xylem of hypocotyl segments or by forcing water from outside into the xylem. In segments with intact cuticle, the hydraulic conductance of the radial path (Lpr) was a function of the rate of water flow and also of flow direction. In segments without cuticle, Lpr was large (Lpr=2·10-5–20·10-5 cm s-1 bar-1) and exceeded the corticla cell Lp. The results of the pressure-perfusion experiments are not compatible with a cell-to-cell transport and can only the explained by a preferred apoplasmic water movement. A tentative explanation for the differences found in the different types of experiments is that during hydrostatic perfusion the apoplasmic path dominates because of the high hydraulic conductivity of the cell wall or a preferred water movement by film flow in the intercellular space system. For shrinking and swelling experiments and during growth, the films are small and the cell-to-cell path dominates. This could lead to larger gradients in water potential in the tissue than expected from Lpr. It is suggested that the reason for the preference of the cell-to-cell path during swelling and growth is that the solute contribution to the driving force in the apoplast is small, and tensions normally present in the wall prevent sufficiently thick water films from forming. The solute contribution is not very effective because the reflection coefficient of the cell-wall material should be very small for small solutes.
