Photosynthesis in Chlorella pyrenoidosa is inhibited by iodo-acetic acid and iodo-acetamide, both of which attack the Blackman reaction. Since acetamide is without effect, the iodo-acetyl radical must be responsible. The study of the action of the acid is complicated by the fact that its ions penetrate slowly, if at all, so that negative results with this agent are without significance unless penetration can be established. The absorption spectrum of the cells is not affected by concentrations of iodo-acetamide which completely inhibit photosynthesis. This establishes that the chromophore groups of chlorophyll are not involved, and renders it unlikely that any other part of the molecule is. Inasmuch as cyanide likewise inhibits by way of the Blackman reaction, it would seem necessary to postulate that this complex can be attacked at two different loci, which may or may not be on the same molecule. The presence of the iodo-acetyl radical also gives rise to three other effects. (1) Concentrations (10–5 M or less) too small to inhibit photosynthesis may increase the rate by interacting with the photochemical complex. (2) Concentrations (ca. 10–4 M) which inhibit photosynthesis increase the rate of respiration. (3) Concentrations (10–3 M or more) higher than those required to inhibit photosynthesis inhibit respiration.  相似文献

Chlorella pyrenoidosa (van Niel's strain) was cultured in four environmental conditions: light-glucose, light-galactose, dark-glucose, and dark-galactose. The gradual change in structure was observed when the cultures were transferred from light to darkness (dark adaptation). Cells grown in light contained well-developed chloroplasts and the normal cytoplasmic sub-units. During the transition to the complete heterotrophic mode of nutrition, the chloroplast regressed to the proplastid stage with a gradual reduction in thylakoid number. A prolamellar body was noted in the most regressed proplastid condition and plastid storage material was observed in all stages. The cell wall thickened during the transition. Cells adapted to darkness on galactose exhibited abundant cytoplasmic storage material with osmiophilic properties.  相似文献

The effect of protein synthesis inhibition by cycloheximide on nucleolar RNA synthesis and processing has been studied in HeLa cells. Synthesis of 45S RNA precursor falls rapidly after administration of the drug. However, the nucleolar content of 45S RNA remains relatively constant for at least 1 hr because the time required for cleavage of the precursor molecule into its products is lengthened after treatment with cycloheximide. The efficiency of transformation of 45S RNA to 32S RNA remains constant with approximately one molecule of the 32S RNA produced for each cleavage of a molecule of 45S RNA. However, shortly after the cessation of protein synthesis the formation of 18S RNA becomes abortive. The amount of 32S RNA present in the nucleolus remains relatively constant. After long periods of protein synthesis inhibition the 28S RNA continues to be synthesized and exported to the cytoplasm but at a greatly reduced rate. When the protein synthesis inhibitor is removed, a prompt, although partial, recovery in the synthesis rate of 45S RNA occurs. Methylamine uptake in nitrogen-starved Chlorella pyrenoidosa Beij. follows Michaelis-Menten kinetics: maximum uptake is about 1.6 nmol μl?1· cells · min?1, half-saturation occurs at 4 μM methylamine, and the slope in the range where uptake is proportional to concentration is 0.4 nmol μl?1· min?1·μM?1. In cells grown in the presence of a non-limiting nitrogen concentration, methylamine uptake is directly proportional to concentration up to at least 0.5 mM, and the slope is 1/500 that for starved cells. Similar uptake kinetics have been reported for Penicillium chrysogenum and attributed to an inducible “ammonium permease.” Apparently, a similar permease occurs in algae.  相似文献

The unicellular green alga Coccomyxa, a component of the lichenPeltigera aphthosa, liberated about 7.2mµg biotin permg dry weight of cells into the culture medium during a growthperiod of 15–20 days. The corresponding figure for thefree-living alga Chlorella pyrenoidosa was 0.45mµg ofbiotin. Chromatographic analysis indicated that this was freebiotin and not a bound form of the vitamin. The biotin concentrationof rinsed Coccomyxa cells was 1.88mµg per mg dry weightof cells, of which less than 0.01mµg was extractable byhot water. Cells of Chlorella contained 0.16mµg of biotinper mg dry weight, of which 0.11mµg was extractable byhot water. The biotin content of Coccomyxa, which was about12 times that of Chlorella, is thus almost entirely in the boundform. Chlorella pyrenoidosa (Chick) was grown heterotrophically in batch culture on defined medium with glucose. Morphometric analysis of cells in the exponential growth phase showed that starch accounted for 57% of the volume of the chloroplast and 36% of the total cell volume. During the stationary growth phase, the amount of starch accounted for only 36% of the chloroplast volume and 13% of the total cell volume. This represented a 36% decrease in the amount of starch/cell between the exponential and stationary phase. Determination of starch as grams/cell using quantitative techniques on cell extracts showed a comparable decrease in the amount of starch during this same transition. Based on these results, morphometric techniques provided an accurate assay of starch and have the added advantage of visualization of cellular structures not available when quantitative techniques are used.  相似文献

Potassium is an important component information of polyphosphate bodies (PB) by Chlorella pyrenoidosa Chick. However, it was not detected in PB by X-ray energy dispersive microanalyses when the specimens were subjected to a standard preparation procedure for transmission electron microscopy. Intact cells were incinerated at 350 C on stainless steel grids coated with silicon monoxide. X-ray spectra from PB showed conspicuous peaks of energy counts in the Kα lines for phosphorus and potassium. It is proposed that potassium is a major cationic component of PB in C. pyrenoidosa grown in potassium sufficient medium.  相似文献

IN VIVO INHIBITION OF RAT BRAIN PROTEIN SYNTHESIS BY l-DOPA   总被引:3,自引:2,他引:1  
Abstract— A study has been made of the effect of a single intraperitoneal dose of l -DOPA on the in vivo metabolism of [14C]leucine and [14C]lysine by the brain, and on their uptake into brain protein. Administration of 500 mg DOPA/kg to 40-g rats raised the concentrations of several free amino acids; the only amino acid which underwent a statistically significant increment was alanine. Intracisternally-injected [U-14C]leucine was rapidly metabolized to other labelled compounds; DOPA administration did not influence significantly the rate of its metabolism. No similar metabolic change was observed after administering [U-14C]lysine intracisternally.
Incorporation of [14C]leucine and [14C]lysine into total brain protein was significantly reduced 45 min after DOPA administration. There was also depression of the uptake of labelled amino acid into a supernatant fraction, obtained by high speed centrifugation of the brain homogenate, and into brain microtubular protein (tubulin). Reduced amino-acid incorporation into brain proteins observed 45 min after l -DOPA injection coincided with extensive disaggregation of brain polyribosomes. At 120 min after DOPA treatment, disaggregation was no longer significant and there was a smaller depression in labelled amino aicd incorporation, which disappeared completely 240 min after l -DOPA injection. It is concluded that disaggregation of brain polysomes following DOPA treatment is an accurate reflection of a change in the intensity of brain protein synthesis in vivo.  相似文献

Abstract— Between 1 and 4 h after rats received a single injection of d-amphetamine (15 mg/kg)(when brain polysomes are known to be disaggregated), the in vivo incorporation of [14C]lysine into trichloroacetic acid-precipitable brain protein was reduced by 28–48%. Incorporation of the 14C label into the protein present in a 100,000 g supernatant extract of whole brain was similarly reduced (by 44%). Amphetamine administration suppressed protein synthesis in rat cerebral cortex, cerebellum, hypothalamus, striatum, and brainstem to an equivalent extent. The drug did not significantly affect lysine pool sizes measured in these brain regions; thus the reduced incorporation of labeled lysine was not the result of an isotope dilution effect. We therefore conclude that the brain polysome disaggregation resulting from amphetamine administration is associated with decreased in vivo synthesis of some brain proteins.  相似文献

蛋白核小球藻藻粉的PBS抽提液经硫酸铵二步分级沉淀 ,再经DEAE Sepharose和SephadexG 10 0层析 ,从中分离纯化得到蛋白核小球藻凝集素 (CPL)。经测定 ,该凝集素为单个亚基的蛋白质 ,相对亚基分子量为 1 4× 10 4 — 1 5× 10 4 ,分子中不含糖。在氨基酸组成中 ,苯丙氨酸 (Phe)的含量最高 ,其次是天冬氨酸 (Asp)和谷氨酸 (Glu) ,不含组氨酸 (His)。CPL能够凝集兔、绵羊及鸽子红细胞 ,其中对兔红细胞的凝集活性最大 ,最低浓度为 6 88μg/mL ,对鸡、鸭及人红细胞 (A型、O型及B型 )无凝集活性。卵黏蛋白和 7种单糖对CPL的凝血活性具有抑制作用。CPL具有很好的热稳定性 ,在 90℃处理 10min不失活。  相似文献

Chlorella pyrenoidosa Chick cells grown in liquid culture containing 5 × 10?3M sodium 2,4-dichloro-phenoxyacetic acid exhibited vesiculation of the plasma membrane, mitochondrial swelling with disorganization of cristae, disruption of the chloroplast lamellar system, and dictyosomes with irregularly swollen cisternae. Ultrastructural details previously not reported for species of Chlorella were also observed: (1) presence of a crystalline pyrenoid with an 80 A periodicity, (2) a nucleolus made up of ribosome-like particles, and (3) smaller ribosomes in the chloroplast and mitochondria than in the cytoplasm.  相似文献

A novel series of 6-methylpurine nucleoside derivatives with substitutions at 5′-position have been synthesised. These compounds bear a 5′-heterocycle such as triazole or a imidazole with a two carbon chain, and an ether, thio ether or amine. To extend the SAR study of 2-fluoroadenine and 6-methyl purine nucleosides, their corresponding α–linker nucleosides with l-xylose and l-lyxose were also synthesized. All of these compounds have been evaluated for their substrate activity with E. coli PNP.  相似文献

Marsilea vestita and M. drummondii were grown in sterile cultures to which concentrations of the protein synthesis inhibitors, 2-thiouracil (10 mg/liter) and 5-fluorouracil (1 mg/liter) had been added. When young sporelings are grown in a solution of thiouracil at optimum concentration, there is an inhibition of the rate of leaf formation, a retardation of the leaf heteroblastic series, and all leaves develop as land forms. When thiouracil is added to plants which are already producing typical adult, quadrifid leaves, the effects depend on whether the treated plants are water or land forms. Plants which are typically water forms convert to land forms. After treatment successive leaves develop typical sunken stomata on both leaf surfaces. The tissues of the rhizome, root and petiole are more compact and, in general, the cells of the plant have thicker walls. Vascular patterns are not changed, though the size of the rhizome, root and petiole may be reduced. Plants which are typically land forms are less affected than the water forms, but they show a small reduction in apex volume and an apparent reversion of the leaflet number from the typical quadrifid leaf to a trifid, bifid, or single lamina condition. In both land and water forms apical dominance may be broken by treatment with 10 mg/liter thiouracil or 1 mg/liter fluorouracil and numerous lateral branches develop. Higher concentrations (15–25 mg/liter of thiouracil) may result in abnormal development of lateral axillary buds, petiole bases and leaflets. The meristems of the plant are differentially sensitive to thiouracil; leaflet meristems are most sensitive, the root meristems are the least sensitive. It appears that a true reversion to juvenile leaf development need not occur even though protein synthesis and the volume of the apex are reduced. The development of the land or water form in Marsilea appears to depend on rate of growth. Hence inhibition of the growth of typical water forms, through inhibition of protein synthesis, causes a shift in development toward the morphology typical of land forms.  相似文献

Endogenous levels of indole-3-acetic acid were mesaured in synchronous cultures of Chlorella pyrenoidosa (TX-7-11-05). The cultures were synchronized by alternating light:dark periods of 15:9 hr at a temperature of 40 ± 1 C. After 2 synchronous cycles the cultures were exposed to a low light treatment of 350 ± 100 ft-c. The time to incipient cell division under these conditions was 6 hr and 15 min. Samples were taken at 3 sampling periods during the low light treatment period:low light 0 hr (LL0); low light 3 hr (LL3); and low light 6:15 hr (LL6:15). The algal extracts were analyzed by a fluorometric procedure which measured the indole-α-pyrone product formed by the action of the trifluoracetic acid-acetic anhydride reagent with IAA. The IAA levels increased gradually from the autospore stage (5.19 μg × 10?4/mg dry wt) to the adolescent stage (7.13 μg × 10?4/mg dry wt) and more rapidly when approaching the ripened adult stage (14.55 μg × 10?4/mg dry wt). The mean percentage increase from autospore to adolescent was 36.9%, and from adolescent to ripened adult 104.6%. The total percentage increase from autospore to adult was 180.3%. Levels of IAA were 2 times higher just prior to division than in the autospore stage.  相似文献

The temperature characteristic for the rate of O2 consumption by Chlorella pyrenoidosa suspended in Knop solution containing 1 per cent glucose was studied between 1° and 27°C. with the Warburg technic. The value of µ was found to be about 19,000 ±1,000 cal. There is some indication of a critical temperature at 20°C., with shift to a lower µ above this temperature. The effect of sudden changes in temperature on the rate of respiration and the variation of the latter with time at constant temperatures are discussed. It is concluded that the "normal" respiration (in absence of external glucose) does not appear in the determination of this temperature characteristic.  相似文献

Cordycepin is an analogue of adenosine lacking the 3'-OH. When incorporated into a growing RNA molecule, cordycepin prevents further elongation, thus producing a prematurely terminated RNA molecule. When HeLa cells are exposed to low concentrations of cordycepin, DNA and protein synthesis are unaffected during short exposure periods. The synthesis of completed ribosomal and ribosomal-precursor (45S) RNA is significantly depressed. Partially completed 45S ribosomal precursor molecules accumulate in the nucleolus. 18S ribosomal RNA can be cleaved from these incomplete precursors, while 32S ribosomal precursor cannot be produced from partially snythesized 45S molecules. The synthesis of transfer RNA is also reduced in the presence of cordycepin. The synthesis of the nuclear heterogeneous RNA species is unaffected by the drug while the cytoplasmic heterogeneous RNA is slightly reduced.  相似文献
